Abstract:Herein, we present an electrochemical desulfurative protocol for the formation of alkyl boronic esters from thioethers. The paired electrolytic transformation utilizes HBpin as coupling partner and proceeds with inert electrodes. The transformation features mild conditions, broad substrate scope and excellent functional group tolerance, as illustrated by late-stage functionalization of pharmaceutical compounds and natural products. Furthermore, the protocol is scalable, successfully producing gram quantities of borylated product.

Abstract:The use of alkylthio electrophiles in synthesis has remained elusive because of the lack of a suitable reagent that is practical and of excellent enantioselectivity and appropriate reactivity. In this work we introduce a novel alkylthio reagent based on the 2-mercapto-5-methyl-1,3,4-thiadiazole (MMTD) fragment for direct alkylsulfenylation of ketones and aldehydes. It can be readily prepared by the oxidative coupling between thiadiazole and other alkylthio reagents and be combined with chiral primary aminocatalysis. This protocol provides facile access to diverse α-alkylthio quaternary carbon centers with good stereoselectivities.

Abstract:Thioethers are critical in the fields of pharmaceuticals and organic synthesis, but most of the methods for synthesis alkyl thioethers employ foul-smelling thiols as starting materials or generate them as by-products. Additionally, most thiols are air-sensitive and are easily oxidized to produce disulfides under atmospheric conditions; thus, a novel method for synthesizing thioethers is necessary. This paper reports a simple, effective, green method for synthesizing dialkyl or alkyl aryl thioether derivatives using odorless, stable, low-cost ROCS2K as a thiol surrogate. This transformation offers a broad substrate scope and good functional group tolerance with excellent selectivity. The reaction likely proceeds via xanthate intermediates, which can be readily generated via the nucleophilic substitution of alkyl halides or aryl halides with ROCS2K under transition-metal-free and base-free conditions.

Abstract:Organic sulfides play a crucial role in drug discovery and organic synthesis. The construction of C-S bonds usually requires strong basic conditions, transition metal catalysis, or stoichiometric oxidizing /reducing agents. Herein, we report an electrochemically driven C-S radical-radical cross coupling for the synthesis of benzylic thioethers. This approach is applicable to various benzyl halides and disulfides, providing corresponding benzylic thioethers in moderate to excellent yields. The reaction can be conducted at room temperature in air without the need for transition metals, external reducing agents, or sacrificial anodes, making it an environmentally friendly, cost-effective, and mild synthetic strategy.

Abstract:The formation of aryl C-S bonds is an important chemical transformation because aryl sulfides are valuable building blocks for the synthesis of biologically and pharmaceutically active molecules and organic materials. Aryl sulfides have traditionally been synthesized through the transition-metal-catalyzed cross-coupling of aryl halides with thiols. However, the aryl halides used are usually bromides and iodides; readily available, low-cost aryl chlorides often not reactive enough. Furthermore, the deactivation of transition-metal catalysts by thiols has forced chemists to use high catalyst loadings, specially designed ligands, high temperatures, and/or strong bases, thus leading to high costs and the incompatibility of some functional groups. Herein, we describe a simple and efficient visible-light photoredox arylation of thiols with aryl halides at room temperature. More importantly, various aryl chlorides are also effective arylation reagents under the present conditions.

Abstract:Abstract Organic sulfides form the core scaffold of a wide range of pharmaceuticals, natural products, and materials, and serve as key intermediates in synthesis. Prior methods to organic sulfides require the use of transition metal (TM) catalysts, prefunctionalized or chelating group-containing substrates, and elevated reaction temperatures. A general TM-free C–H thiolation protocol using readily accessible sulfur source is highly desirable. Herein, we disclose a direct C(sp)–, C(sp2)–, and C(sp3)–H thiolation reaction using β-sulfinylesters as the versatile sulfur source. The key step of this protocol is chemoselective C–S bond cleavage of the sulfonium salts that is in situ formed from the corresponding (hetero)arenes, alkenes, alkynes, and 1,3-dicarboxyl compounds with β-sulfinylesters. The successful capture of acrylate byproduct supports a retro-Michael reaction mechanism. This method is expected to be used widely because of several advantageous aspects including TM-free, mild reaction conditions, and broad substrate scope including drug molecules.
Counteranion-Controlled Chemodivergent Transfer-Hydrothiolation/Carbothiolation Utilizing Thioethers as Bifunctional Reagents

Abstract:The pursuit of catalysis capable of forging C(sp3)-S bonds is extremely desired because these bonds have substantial importance in pharmaceuticals, functional materials, and organic synthesis. Hydrothiolation and carbothiolation of feedstock alkenes are among the most straightforward and prominent approaches to C(sp3)-S bond formations. However, hydrothiolation of unactivated alkenes typically proceeds in an anti-Markovnikov fashion, and carbothiolation relied on three-component coupling strategies using highly reactive pre-functionalized electrophilic sulfur sources and nucleophilic carbon sources. While carbothiolation of alkenes with a single, bifunctional reagent represents a concise and streamlined synthetic strategy for -alkyl organosulfur compounds, it has not been reported. Herein, by strategically using heterolytic cleavage of C(sp3)−S bond, we report a chemodivergent transfer-hydrothiolation and carbothiolation of alkenes with thioethers serving as bifunctional reagents. These catalytic systems can control the cleavage and reformation of strong C(sp3)−S -bonds. The chemo-control is achieved through careful selection of the counteranion associated with the Rh-centre. Counteranions with relatively strong basicity and coordinating ability, such as TfO-, promote an unusual Markovnikov transfer-thiol-ene reaction. Conversely, less basic and non-coordinating counteranions, such as BF4-, enable an unprecedented thioether-ene reaction, directly adding thioethers across alkenes. Mechanistic and computational studies elucidated that the coordination or non-coordination of counteranions on Rh can alter the basicity of thiolate, resulting in chemodivergence.

Abstract:An electrochemical three-component reaction involving elemental sulfur is disclosed for achieving a metal-free, oxidant-free synthesis of thioesters in a high atom-economical, step-economical and chemoselective manner. A mechanistic investigation indicates that the use of elemental sulfur to trap acyl radical derived from radical umpolung of α-keto acid with an electrochemical design can efficiently generate a carbonyl thiyl radical, which can further be captured by diazoalkane to afford various thioesters.

Abstract:Sulfoxide is an important structural motif widely found in medicine, materials, chiral catalysis, and many other fields. The development of green and sustainable synthetic methods for their efficient preparation is of great significance. Herein, we have unraveled an electrochemical β-chlorosulfoxidation of alkenes, using the readily available thiols and hydrochloride as the limiting agents. This protocol features a simple operation, wide substrate scope, and good functionality compatibility. Notably, the thus obtained β-chlorosulfoxides have been shown to be versatile building blocks for the preparation of diverse functionalized β-sulfoxides.

Abstract:An intermolecular reductive C-C coupling of electrophilic alkyl radicals and alkenes has been developed. Thiols were used as both hydrogen-donating reagents and alkyl radical precursors in the presence of triethyl phosphite and radical initiator. A wide range of alkenes, including styrenes, and aliphatic olefins were well tolerated in this transformation. Mechanistic studies indicated that a phosphite promoted radical desulfurization of thiols to access electrophilic alkyl radicals and a radical chain propagation process may be involved in this transformation.

Abstract:Through effective regulation of active species in photocatalytic process, the oxidation or C(sp3)‐S bond cleavage upgrading of widespread aryl alkyl sulfides under mild conditions was successfully achieved by using a pyridinium photocatalyst. Benefiting from the excellent redox ability of the photocatalyst, the electron transfer between the pyridinium molecule and the substrate, molecular oxygen, or counter‐anion effectively promotes the conversion and upgrading of the substrate thioether. Among them, the efficient generation of reactive oxygen species (ROS) enables the highly selective oxidation of sulfides to sulfoxides under visible light and air atmosphere. More importantly, the chlorine radical (Cl•) generated by electron transfer, reported for the first time, contributes to the cleavage of C(sp3)‐S bonds, achieving the transformation of aryl alkyl sulfides to disulfides. By harnessing the superior photocatalytic ability of pyridinium molecules, this work not only achieves the highly selective conversion of thioether by taming the active species in the photocatalytic process, but also sheds light on the untapped potential of chlorine radicals in the field of C(sp3)‐S bond activation and cleavage.

Abstract:A new cooperative nickel reductive catalysis and N,N-dimethylformamide-mediated strategy for umpolung C-S radical reductive cross coupling of S-(trifluoromethyl)arylsulfonothioates with alkyl halides to produce alkyl aryl thioethers is described. This reaction features excellent selectivity, wide functionality tolerance, broad substrate scope, and facile late-stage modification of biologically relevant molecules. Mechanistic studies recognize initial generation of an amidyl radical anion via thermoinduced reduction of DMF with Sn, followed by umpolung reduction and single electron transfer of the nucleophilic sulfonyl moiety to form a sulphydryl radical and engage the Ni0/NiI/NiIII/NiI catalytic cycle.

Abstract:The introduction of a trifluoromethyl group into sulfur-containing compounds would significantly alter the biochemical properties of the molecules. Reductive coupling has proven to be an exceptionally powerful strategy to construct C-C and C-X bonds. There are some reports of accessing various alpha-functionalized trifluoroalkanes via C-C bond forming reductive coupling, but C-X reductive coupling is still out of coverage. Herein, we report a concise and efficient Ni-catalyzed reductive C-S coupling, which enables access to a series of alpha-CF3 functionalized thioethers in good to excellent yields under mild conditions, with a broad substrate scope and excellent functional group compatibility. Additionally, this protocol provides a new approach for subsequent trifluoromethylation modification of sulfur-containing organic structures.

Abstract:Alkynyl sulfides are prevalent in bioactive molecules and functional materials. A variety of methods for the construction of alkynyl sulfides have been developed, however, the general and practical synthetic strategy to afford alkynyl sulfides from readily available starting materials remains non-trivial. Herein, we report that the widely accessible 1,2,3-thiadiazoles as robust alkynylthiolate anions, which couple with C(sp 1-3 )-electrophiles to construct corresponding alkynyl sulfides in the presence of metal catalyst or under metal free conditions (79 examples with average 68% yield). The alkynylthiolation reaction with aryl iodides and bromoalkynes is experimentally easy to conduct under mild conditions in the presence of palladium or copper catalysts; the alkynylthiolation with 1 o -, 2 o -alkyl electrophiles and difluorocarbene precursor are performed under metal free conditions. Gram-scale reactions and diverse synthetic transformations convincingly demonstrate the synthetic potential of this method.

Abstract:Organic thioethers play an important role in the discovery of drugs and natural products. However, the green synthesis of organic sulfide compounds remains a challenging task. The convenient and efficient synthesis of 5-alkoxy-3-halo-4-methylthio-2(5H)-furanones from DMSO is performed via the mediation of 1,3-dibromo-5,5-dimethylhydantoin (DBDMH), affording a facile route for the sulfur-functionalization of 3,4-dihalo-2(5H)-furanones under transition metal-free conditions. This new approach has demonstrated the functionalization of non-aromatic Csp2-X-type halides with unique structures containing C-X, C-O, C=O and C=C bonds. Compared with traditional synthesis methods using transition metal catalysts with ligands, this reaction has many advantages, such as the lower temperature, the shorter reaction time, the wide substrate range and good functional group tolerance. Notably, DMSO plays multiple roles, and is simultaneously used as an odorless methylthiolating reagent and safe solvent.

Abstract:An electrochemical protocol for the synthesis of sulfursulfoxonium ylides and 1,3-dithioketals by reacting sulfoxonium ylides with disulfides has been developed under simple and mild conditions. By changing the solubility of the raw materials and the dielectric parameters of the electrolyte, sulfurization enabled a selective dehydrogenation of C-S and the construction of 1,3-dithioketals. The transformation is an ideal approach to prepare organosulfur reagents with a broad functional group tolerance as well as high selectivity, which leads to vicinal difunctionalized organosulfur compounds.
