Abstract:Cryopreservation, one of the most effective preservation methods, is essential for maintaining the safety and quality of food. However, there is no denying the fact that the quality of muscle food deteriorates as a result of the unavoidable production of ice. Advancements in cryoregulatory materials and techniques have effectively mitigated the adverse impacts of ice, thereby enhancing the standard of freezing preservation. The first part of this overview explains how ice forms, including the theoretical foundations of nucleation, growth, and recrystallization as well as the key influencing factors that affect each process. Subsequently, the impact of ice formation on the eating quality and nutritional value of muscle food is delineated. A systematic explanation of cutting-edge strategies based on nucleation intervention, growth control, and recrystallization inhibition is offered. These methods include antifreeze proteins, ice-nucleating proteins, antifreeze peptides, natural deep eutectic solvents, polysaccharides, amino acids, and their derivatives. Furthermore, advanced physical techniques such as electrostatic fields, magnetic fields, acoustic fields, liquid nitrogen, and supercooling preservation techniques are expounded upon, which effectively hinder the formation of ice crystals during cryopreservation. The paper outlines the difficulties and potential directions in ice inhibition for effective cryopreservation.
Study on the Effect of Ultrasonic Assisted Freezing on the Ice Crystals and Water Changes in Penaeus chinensis

Abstract:The formation of ice crystals and the change of moisture of Penaeus chinensis treated by ultrasonic assisted freezing(UF),immersion freezing(IF)or air refrigerator freezing(RF),and the effect of different exposure time were investigated. The result showed that the time to pass through the zone of maximum crystallization was 130 s for UF,showing an obviously higher rate in comparison with IF or RF.Wherein,the minimal tissue destruction was observed for samples treated by UF with the minimum diameter of crystal of 46.81 μm. Furthermore, Compared to the other groups, samples treated by UF had a higher water holding capacity than others about 7.19 %-19.50 % and a lower dehydration than others about 1.30 %-2.38 %.The result of Pearson analysis indicated that there was an extremely significant correlation between ice crystals area and moisture change for all the groups,and a significant correlation between the roundness and moisture change was also noticed for samples treated by UF,while no correlation was observed for those treated by IF or RF.

Abstract:Freezing of natural biomaterials results in the formation of ice crystals and the generation of hypertonicity, both of which are deleterious to biomaterials. Although the cryopreservation of cells, tissues, and even organs has been achieved empirically by vitrification using cryoprotectants, the underlying mechanisms are poorly understood. To better understand the crystallization and vitrification processes of cryoprotected cells, onion epidermal cells immersed in dimethyl sulfoxide (DMSO) solutions with different concentrations were employed as platforms. The crystallization and vitrification dynamic processes of the individual and multiple monolayer cells were recorded using a high-speed microscope camera and the forms of the intracellular and extracellular ices were further confirmed by corresponding Raman spectra. The effects of DMSO concentration and cooling/warming rate on both processes were investigated and the findings were of an important significance to improve the understanding of the mechanisms of intracellular ice formation in individual cells and the ice propagation between adjacent cells. It is expected to provide a theoretical basis for cryopreservation by vitrification and point toward a new pathway for developing cryopreservation protocols.
Preliminary Study on the Freeze-Drying of Human Bone Marrow-Derived Mesenchymal Stem Cells

Abstract:Long-term preservation and easy transportation of human bone marrow-derived mesenchymal stem cells (hBM-MSCs) will facilitate their application in medical treatment and bioengineering. A pilot study on the freeze-drying of hBM-MSCs was carried out. hBM-MSCs were loaded with trehalose. The glass transition temperature of the freeze-drying suspension was measured to provide information for the cooling and primary drying experiment. After freeze-drying, various rehydration processes were tested. The highest recovery rate of hBM-MSCs was (69.33±13.08)%. Possible methods to improve freeze-drying outcomes are discussed. In conclusion, the present study has laid a foundation for the freeze-drying hBM-MSCs.

Abstract:ABSTRACT The rapid development in mobile computing has brought context sensing and information available for music recommendation as well. We reviewed 19 experimental contextual music systems and found that while the context factors of location, time, activity, and identity are adopted in a wide variety of ways, the systems mainly rely on common UI solutions for interacting with these factors. Specifically, context factors could be employed to offer explanations, transparency, and visualizations of music recommendations in more explorative ways, providing novel user experiences. Based on our review, we provide implications for design and research on future media discovery systems, which we believe can realize the great potential of context-aware content services.
Cooling dynamics of droplets exposed to solid surface freezing and vitrification

Abstract:Solid surface freezing or vitrification (SSF/SSV) can be done by depositing droplets of a sample, e.g., cells in a preservation solution, onto a pre-cooled metal surface. It is used to achieve higher cooling rates and concomitant higher cryosurvival rates compared to immersion of samples into liquid nitrogen. In this study, numerical simulations of SSF/SSV were conducted by modeling the cooling dynamics of droplets of cryoprotective agent (CPA) solutions. It was assumed that deposited droplets attain a cylindrical bottom part and half-ellipsoidal shaped upper part. Material properties for heat transfer simulations including density, heat capacity and thermal conductivity were obtained from the literature and extrapolated using polynomial fitting. The impact of CPA type, i.e., glycerol (GLY) and dimethyl sulfoxide (DMSO), CPA concentration, and droplet size on the cooling dynamics was simulated at different CPA mass fractions at temperatures ranging from -196 to 25 °C. Simulations show that glycerol solutions cool faster compared to DMSO solutions, and cooling rates increase with decreasing CPA concentration. However, we note that material property data for GLY and DMSO solutions were obtained in different temperature and concentration ranges under different conditions, which complicated making an accurate comparison. Experimental studies show that samples that freeze have a delayed cooling response early on, whereas equilibration times are similar compared to samples that vitrify. Finally, as proof of concept, droplets of human red blood cells (RBCs) were cryopreserved using SSV/SSF comparing the effect of GLY and DMSO on cryopreservation outcome. At 20% (w/w), similar hemolysis rates were found for GLY and DMSO, whereas at 40%, GLY outperformed DMSO.

Abstract:Recently, we proposed a low-glycerol cryoprotectant formulation (consisting of 0.4 M trehalose and 5% glycerol) for cryopreservation of human red blood cells (RBCs), which greatly reduced the concentration of glycerol, minimized intracellular ice damage, and achieved high recovery. Although this study was successful in cellular experiments, the nonequilibrium phase transition behaviors of the cryoprotective agent solution have not been systematically analyzed. Therefore, it is essential to provide reliable thermodynamic data to substantiate the viability of this cryopreservation technique. In this study, the phase change behaviors and thermal properties of typical trehalose and/or glycerol solutions quenched in liquid nitrogen were investigated using differential scanning calorimetry and cryomicroscopy. It was found that the glass transition temperatures of both the trehalose aqueous solution (<1.0 M) and glycerol aqueous solution (<40% w/v) did not vary apparently with the concentration at low concentrations, while they increased significantly with increasing concentration at high concentrations. Moreover, it was revealed that the inhibitory effect of trehalose on ice growth was affected by glycerol. We further found that the addition of low concentrations of glycerol facilitates the partial glass transition of trehalose solutions at low concentrations. The results of this work provide reliable thermodynamic data to support the cryopreservation of human RBCs with unusually low concentrations of glycerol.

Abstract:OBJECTIVE To conduct a meta-analysis of 11 randomized controlled trials comparing enteral nutritional support supplemented with key nutrients versus standard enteral nutritional support to determine effects on morbidity and mortality rates and hospital stay. BACKGROUND DATA Recent studies have shown that malnutrition occurs in up to 30% of patients undergoing gastrointestinal surgery, resulting in an increased risk of postoperative complications and death. With the realization that key nutrients can modulate inflammatory, metabolic, and immune processes, enteral nutritional regimens (supplemented with large amounts of key nutrients) have been developed for clinical use. METHODS Eleven prospective, randomized controlled trials evaluating 1009 patients treated with combinations of key nutrients (Impact, Immun-Aid) were evaluated. Outcome measures examined were the incidences of pneumonia, infectious complications, and death, and length of hospital stay. Meta-analyses were undertaken to obtain the odds ratio and 95% confidence interval for incidences of infectious complications, pneumonia, and death, and the weighted mean difference and 95% confidence interval for length of hospital stay. RESULTS The provision of nutritional support supplemented with key nutrients to patients with critical illness resulted in a decrease in infectious complications when compared with patients receiving standard nutritional support and a significant reduction in overall hospital stay. Similar results were documented in patients with gastrointestinal cancer. However, there were no differences between patient groups for either pneumonia or death. CONCLUSIONS This meta-analysis has demonstrated that nutritional support supplemented with key nutrients results in a significant reduction in the risk of developing infectious complications and reduces the overall hospital stay in patients with critical illness and in patients with gastrointestinal cancer. However, there is no effect on death. These data have important implications for the management of such patients.
Quantifying the Growth Rate and Morphology of Ice Crystals Growing in Cryoprotectants Via High-Speed Camera and Cryo-Microscope

Abstract:Recently, several significant progresses have been made on the studies of extracellular and intracellular ice formation based on high-speed camera and cryo-microscope. This experimental methodology could accurately capture rapid formation process of ice crystals at micro-scale. In this paper, we are dedicated to quantify and comparatively investigate the growth rate and morphology of ice crystals growing in DMSO, sucrose and trehalose, respectively via high-speed camera and cryomicroscope. Several impact factors such as the concentration of cryoprotectants and the cooling rate have been investigated. The results indicate that the species and concentration of cryoprotectants and the cooling rate could significantly affect the growth rate and morphology of ice crystals. DMSO is better than trehalose and sucrose as cryoprotectant because of its molecular structure. This work may enhance current understanding of the factors for ice crystals formation and help optimize the cryopreservation process in the near future.
Acoustic Droplet Vitrification Method for High-Efficiency Preservation of Rare Cells

Abstract:Cryopreservation is a key step for current translational medicine including reproductive medicine, regenerative medicine, and cell therapy. However, it is challenging to preserve rare cells for practical applications due to the difficulty in handling low numbers of cells as well as the lack of highly efficient and biocompatible preservation protocols. Here, we developed an acoustic droplet vitrification method for high-efficiency handling and preservation of rare cells. By employing an acoustic droplet ejection device, we can encapsulate rare cells into water-in-air droplets with a volume from ∼pL to ∼nL and deposit these cell-containing droplets into a droplet array onto a substrate. By incorporating a cooling system into the droplet array substrate, we can vitrify hundreds to thousands of rare cells at an ultrafast speed (about ∼2 s) based on the high surface to volume ratio of the droplets. By optimizing this method with three different cell lines (a human lung cancer cell line, A549 cells, a human liver cell line, L02 cells, and a mouse embryonic fibroblast cell line, 3T3-L1 cells), we developed an effective protocol with excellent cell viability (e.g., >85% for days, >70% for months), proliferation, and adhesion. Abstract:The micro- and nano-scale freezing has been a focus in many fields of scientific research and engineering practices, especially in those involved with cryobiology physics. Although a series of theoretical models and experimental techniques have been available to tackle the tough issues that associate with the freezing phase change process, the topic to quantify the ice crystal formation and its subsequent effects on the biological materials were not sufficiently addressed at all, especially when facing the newly emerging area of nano-cryobiology technology. Not only the interactions between the ice front and the cell, but also the heat and solute balance during freezing produce impact on the output of a cryosurgery or cryopreservation. There is currently a strong lack of the generalized methods which are indispensable to grasp the physical pictures from micro or nanoscale aspect and thus help achieve an intensive understanding of the mechanisms of cell damage during process of ice crystallization. This review article is dedicated to present an overview on the theoretical and experimental strategies potentially appropriate for quantifying the ice crystal formation in cryobiology area. Attentions will be focused on several major theoretical directions such as the thermodynamic theory, the kinetic theory, and the density functional theory etc., concerning the micro or nano phase change and the ice nucleation activities. Interpretation of the effects of external conditions such as freezing rate, interfacial effect, size effects, electric field and restricted space etc. on the ice growth will be given. Numerical simulation method and experimental techniques will also be outlined. The review does not intend to give an exhaustive knowledge of all the themes, but rather to provide a most basic background for further probing the emerging nano-cryobiology field and thus promote innovation in future research and practices of cryobiology.
Characterizing cellular membrane partitioning of DMSO using low-temperature Raman spectroscopy

Abstract:Additives that help cells survive the stresses of freezing and thawing are known as cryoprotective agents (CPAs). Two different types of CPAs have been identified: penetrating and non-penetrating. Common penetrating CPAs include dimethylsulfoxide (DMSO) and glycerol. The location of a CPA (intracelluar or extracellular) is important for understanding the molecular mechanisms of action for the agent. Low-temperature Raman spectroscopy is a label-free method of detecting the location of CPAs at low temperature with high spatial resolution and chemical specificity. To this end, cells cryopreserved in DMSO using a variety of cooling rates and DMSO concentrations and imaged using Raman spectroscopy were analyzed using automated image analysis to determine the partitioning ratio (concentration of DMSO outside/concentration of DMSO inside the cell). The partitioning ratio was roughly 1 for Jurkat cells frozen at 1°C/min in varying concentrations of DMSO with the exception of 1% DMSO which had a partitioning ratio of 0.2. The partitioning ratio increased from 1 to 1.3 as the cooling rate increased from 1°C to 5°C/min. Different cell types, specifically sensory neurons cells and human induced pluripotent stem cells, exhibited differences in partitioning ratio when frozen in 10% DMSO and 1°C/min suggesting that differences in freezing response may result from differences in solute partitioning. The presence of intracellular ice changed the distribution of DMSO inside the cell and also the partitioning ratio.

Abstract:Oocyte vitrification has become a widely adopted method in clinical practice. However, the solidification behavior and its impact on oocytes during the ultrarapid cooling process remain poorly understood. In this study, we established a system and methodology to observe crystallization behavior in oocytes during quench cooling and warming. Subsequently, the threshold concentration of cryoprotective agents (CPAs) required for oocyte vitrification was determined through a visualization method. The results demonstrated that the ice front could not be observed in the image sequence when using 16.5% DMSO +16.5% EG during high-speed quench cooling (2821.58°C/min). Finally, oocytes were encapsulated with an antifreezing hydrogel (7.5% EG +7.5% DMSO +0.5% alginate) and subjected to high-speed quench cooling. No ice crystals appeared in the antifreezing hydrogel-encapsulated oocytes at a low concentration of osmotic CPA (2.4 M). This research opens up new possibilities for oocyte vitrification with a reduced concentration of CPA.

Characterizing Intracellular Ice Formation of Lymphoblasts Using Low-Temperature Raman Spectroscopy

Abstract:Raman microspectroscopy was used to quantify freezing response of cells to various cooling rates and solution compositions. The distribution pattern of cytochrome c in individual cells was used as a measure of cell viability in the frozen state and this metric agreed well with the population-averaged viability and trypan blue staining experiments. Raman imaging of cells demonstrated that intracellular ice formation (IIF) was common and did not necessarily result in cell death. The amount of intracellular ice as well as ice crystal size played a role in determining whether or not ice inside the cell was a lethal event. Intracellular ice crystals were colocated to the sections of cell membrane in close proximity to extracellular ice. Increasing the distance between extracellular ice and cell membrane decreased the incidence of IIF. Reducing the effective stiffness of the cell membrane by disrupting the actin cytoskeleton using cytochalasin D increased the amount of IIF. Strong intracellular osmotic gradients were observed when IIF was present. These observations support the hypothesis that interactions between the cell membrane and extracellular ice result in IIF. Raman spectromicroscopy provides a powerful tool for observing IIF and understanding its role in cell death during freezing, and enables the development, to our knowledge, of new and improved cell preservation protocols.
Premelting-directed interfacial design of nanoshell-coated cells in directional freezing and thawing

Abstract:Freezing interface interacting with soft cells is a core issue in cryopreservation. However, most of existing cryopreservation approaches face challenges such as complex processing and poor controllability. Herein we report a new, nanotechnology-based method to modulate interfacial interactions between ice and cells via the premelting theory. Through the interfacial design by controllably modifying biocompatible nanoshells on the surface of cells, the effective Hamaker constant between cells and ice can be modified. The thickness of premelted films between cells and ice is further regulated, and directional migration of coated cells occurs in a thermal gradient to achieve directional thawing of coated cells. The decreased mortality of freezing coated cells suggests the premelting modulation in freezing and thawing can effectively protect cells from mechanical damage of ice formation during freezing and ice recrystallization during thawing.

Abstract:Rapid and uniform rewarming is critical to cryopreservation. Current rapid rewarming methods require complex physical field application devices (such as lasers or radio frequencies) and the addition of nanoparticles as heating media. These complex devices and nanoparticles limit the promotion of the rapid rewarming method and pose potential biosafety concerns. In this work, a joule heating-based rapid electric heating chip (EHC) was designed for cryopreservation. Uniform and rapid rewarming of biological samples in different volumes can be achieved through simple operations. EHC loaded with 0.28 mL of CPA solution can achieve a rewarming rate of 3.2 × 105 °C/min (2.8 mL with 2.3 × 103 °C/min), approximately 2 orders of magnitude greater than the rewarming rates observed with an equal capacity straw when combined with laser nanowarming or magnetic induction heating. In addition, the degree of supercooling can be significantly reduced without manual nucleation during the cooling of the EHC. Subsequently, the results of cryopreservation validation of cells and spheroids showed that the cell viability and spheroid structural integrity were significantly improved after cryopreservation. The viability of human lung adenocarcinoma (A549) cells postcryopreservation was 97.2%, which was significantly higher than 93% in the cryogenic vials (CV) group. Similar results were seen in human mesenchymal stem cells (MSCs), with 93.18% cell survival in the EHC group, significantly higher than 86.83% in the CV group, and cells in the EHC group were also significantly better than those in the CV group for further apoptosis and necrosis assays. This work provides an efficient rewarming protocol for the cryopreservation of biological samples, significantly improving the quantity and quality of cells and spheroids postcryopreservation.

Abstract:The storage of living cells is the major challenge for cell research and cell treatment. Here, we introduced a novel supramolecular gel cryopreservation system which was prepared in the microchannel, and the supramolecular gel (BDTC) was self-assembled by gelator Boc-O-dodecyl-L-tyrosine (BDT). This cryopreservation system could obviously minimize the cell injury because the BDTC supramolecular gel had a more compact three-dimensional network structure when the BDT gelator self-assembled in the confined space of microchannel. This compact structure could confine the growth of the ice crystal, reduce the change rate of cell volumes and osmotic shock, decrease the freezing point of the cryopreservation system, and possess better protection capability. Furthermore, the results of functionality assessments showed that the thawed cells could grow and proliferate well and remain the same growth trend of the fresh cells after the RSC96 cells flowed out from the microchannel. This novel method has potential to be used for the cryopreservation of cells, cell therapy, and tissue engineering.
Biological Chip Technology to Quickly Batch Select Optimum Cryopreservation Procedure

Abstract:In the practices of cryobiology, selection of an optimum freeze/thawing program and an idealistic cryoprotective agent often requires rather tedious, time consuming and repetitive tests. Integrating the functions of sample preparation and viability detection, the concept of biochip technology was introduced to the field of cryopreservation, aiming at quickly finding an optimum freezing and thawing program. Prototype devices were fabricated and corresponding experimental tests were performed. It was shown that microflow-channel chip could not offer a high quality solution distribution. As an alternative, the spot-dropping chip proved to be an excellent way to load the sample quickly and reliably. Infrared thermal mapping on such a chip showed that it had a rather uniform heat transfer boundary. Applying the spot-dropping chip combined with the thermoelectric cooling device, the final output of cryopreservation of multiple samples was tested, and the optimal freeze/thawing program as well as the potentially best concentration of the cryoprotective agent was found by analyzing the results. Further, application of this technique to measure the thermo-physical properties of the cryo-protective agent was also investigated. The study demonstrated that a biochip with integrated automatic loading and inspection units opens the possibility of a massive optimization of the complex cryopreservation program in a quicker and more economical way.
Quantitative investigation on the effects of ice crystal size on freeze-drying: The primary drying step

Abstract:The size of ice crystals formed in the freezing process has a significant impact on the subsequent primary drying process. However, the effects have not been fully understood. The purpose of this study was to quantitatively investigate the effects of ice crystal size on water vapor concentration, temperature, and primary drying time. A mathematical model considering the effects of ice crystals size was proposed, and it was solved by Comsol Multiphysics. Results show that there is less water vapor in the products with larger ice crystals. Temperature is found to decrease initially, and larger ice crystals lead to a higher temperature in the dried zone while the lower temperature in the frozen zone. The primary drying process proceeds faster when the ice crystal size is large, and the decrement of primary drying time gets smaller when further increasing the size of ice crystals. This work could provide help for the optimization of freeze-drying.

Profiling the Morphology Effects of MoS2 Nanomaterials on Ice Suppression and Rapid Rewarming for Cryopreservation

Abstract:With the booming development of nanotechnology, an increasing number of nanomaterials and engineering strategies are being used to explore the effective inhibition of ice crystals, thereby reducing ice damage to biological samples and improving the efficiency of cryopreservation. However, the ice-inhibiting effect of nanomaterial shapes and their corresponding photothermal behavior have rarely been studied in depth in cell cryopreservation applications. Here, molybdenum disulfide (MoS2) nanomaterials with different morphologies [nanosheets (NSs), nanoflowers (FLs), and nanomicrospheres (MSs)] were controllably synthesized by a hydrothermal method to profile the influence of MoS2 morphology on ice suppression and rapid rewarming. The prepared MoS2 nanomaterials exhibited excellent capabilities for regulating the nucleation temperature and recrystallization size of ice crystals, which varied with morphology (MSs > FLs > NSs). Unique surface morphology, more functional groups on the surface that could form hydrogen bonds with ice/water molecules, and the adsorption-inhibition mechanism contributed to the excellent ice regulation performance. Surprisingly, the photothermal effect of MoS2 MSs was also the best as the temperature could sharply rise from 24 to 82 degrees C under 808 nm laser irradiation for 1 min. Finally, using MoS2 MSs as a cryoprotectant (CPA) to cryopreserve A549 cells, a cell survival rate of 85.3% was still achieved even when MoS2 MSs replaced 90% of dimethyl sulfoxide in the CPA solution. Our work is the first to correlate the shape variations of nanomaterials with their regulation of ice crystal behavior during the freezing-thawing process and photothermal properties, synergistically improving the efficiency of cell cryopreservation. This study provides guidance for the practical design of nanomaterials for collaborative deicing.
