Abstract:OBJECTIVE:To investigate the polymorphism of HLA class II alleles in the Han population in Hubei province.METHODS:The alleles of DRB1*(n=168), DQB1*(n=160) and DPB1*(n=93) were typed by using the polymerase chain reaction/sequence specific primer(PCR/SSP) and polymerase chain reaction/restriction fragment length polymorphism (PCR/RFLP) techniques.RESULTS:39 HLA-DRB1, 15 DQB1 and 17 DPB1 alleles were found. Alleles with higher frequencies are DRB1*0901 (GF=14%), *1501(GF=11.3%), *0301(GF=7.1%), *0803(GF=4.8%); DQB1*0301(GF=18.8%), *0303(GF=18.4%), *0201(GF=10%), *0302(GF=8.4%); DPB1*0501(GF=31.2%), *0401(GF=15.1%), *0201(GF=14%), *0402(GF=11.8%), respectively. As compared with Caucasians, the Han people in Hubei province have higher gene frequencies of DRB1*0901, *1001, *0803; DQB1*0303, *0502 and DPB1*0501, *0202; and lower gene frequencies of DRB1*0301, *0401, *1301; DQB1*0201, *0603 and DPB1*0401, suggesting that Hubei Hans have their own layout of HLA class II allele frequencies.CONCLUSION:These data may serve as normal reference values for the Han population in south China.

Abstract:DRB genes encode proteins that play an important role in the immune response, and their expressional regulation is crucial to the immune reaction. Sequence variation at the regulatory region can directly affect the gene expression level. The aim of the present study was to use Chinese samples to investigate the variation in the regulation region of the human leukocyte antigen (HLA)-DRB–expressed genes. Seventy- one single nucleotide polymorphisms (SNPs) were found in the four HLA-DRB–expressed genes. By comparing these data with SNPs in the U.S. National Center for Biotechnology Information dbSNP database, 69 SNPs (97.2%) were found to be novel. In addition, two genetic variations of insertion-deletion polymorphisms were discovered within the regulatory region of HLA-DRB1 gene. These polymorphisms can be used as resources of markers for association studies of complex diseases, for assessment of individual predisposition to diseases, and as research markers for population genetics and evolution.

Abstract:Objective This study is aimed to investigate the association of human leukocyte antigen (HLA)-DRB1 with rheumatoid arthritis (RA) in Chinese Han population.Methods A total of 281 Chinese Han patients with RA and 202 healthy controls were recruited.DNA was extracted from PBMC and HLA typing was performed by sequence based typing and PCR-Sequence Specific Primer.The frequency of HLADRB1 was compared between patients and controls using x2 test with continuity correction.Results The susceptible HLA-DRB1 alleles were * 0101,* 0102,*0404,* 0405,and * 0410 which belonged to QRRAA.DRRAA and DERAA were protective alleles.At genotypic level,The association of S3P and S3D was detected.However,the protective effect of S3D was shown to be in a recessive mode.Conclusion Our results have shown that there are racial differences in RA susceptibility between Chinese Han population and Caucasians.

Abstract:This study aimed to clarify the association of HLA Class I and II with dcSSc and lcSSc in Thais. HLA typing for 11 gene loci (Class I: HLA-A, B and C, and Class II [HLA-DR, DP and DQ]) was carried out using the Next Generation DNA Sequencing method (three fields) in 92 Thai patients with systemic sclerosis (55 dcSSc, 37 lcSSc) and 135 healthy controls (HCs). The distribution of HLA alleles in patients with dcSSc and lcSSc was compared. When compared with HCs, the AF of A*24:02:01, A*24:07:01, B*27:04:01 and B*27:06 showed an increasing trend in lcSSc patients without statistical significance. DRB1*15:02:01, DRB5*01:02:01, DQA1*01:01:01, DQB1*05:01:24, DPA1*02:01:01 and DPB1*13:01:01 increased significantly in dcSSc patients. DQB1*05:01:24 and DPB1*13:01:01 also increased significantly in lcSSc patients, but less significantly than in dcSSc patients. The association of DPB1*05:01:01 with lcSSc was significantly protective. HLA-A*24:02:01, B*27:06 and C*03:04:01 formed a three-locus haplotype that also constituted an eight-locus haplotype with DRB1*15:02:01, DQA1*01:01:01, DQB1*05:01:24, DPA1*02:01:01 and DPB1*13:01:01. There was a possibility that HLA Class I would play a role in the pathogenesis of lcSSc, while Class II played more of a role in the dcSSc in Thai patients.

Study of the Association of HLA-DR-DQ Haplotypes with Rheumatoid Arthritis

Abstract:Objective To investigate the associations of HLA-DR-DQ haplotypes with rheumatoid arthritis(RA) in the Han nationality Chinese population. Method PCR-RFLP was used to determine DRB1, DRB3, DRB5, DQA1 and DRB1 alleles in 35 RA patients and 100 healthy controls. Results The frequency of DRB1*0405-DQA1*0301-DQB1*0401 haplotype was significantly increased in RA patients as compared to healthy controls(14% vs 4.5%, RR=3.97,P0.01), The disease was more severe in patients with former haplotype than in patients without, whereas the haplotype of DRB1*1501-DRB5*0101-DQA1*0102-DQB1*0602 was decreased in RA patients as compared with controls(4.3% and 12% in RA and healthy, respectively, P0.01). Conclusion The DRB1*0405-DQA1*0301-DQB1*0401 haplotype was associated with the pathogenesis and severity of RA, and DRB1*1501-DRB5*0101-DQA1*0102-DQB1*0602 haplotype might play a protective role in RA susceptibility.

Abstract:Objective To evaluate the association of genetic polymorphism of HLA-DQ and HLA-DR-DQ linkage with Rheumatiod arthritis in Chinese Han population.Methods Odds ratios(OR) of HLA-DQ and HLA-DR alleles distributions in RA patients against healthy controls were analyzed.All the relevant studies were identified.The Meta-analysis was applied for investigating heterogeneity among individual studies and summarizing across studies.Results Four studies enroll 348 RA patients and 162 controls.HLA-DQA1*0301(OR=1.820,P<0.01)and DQB1*0401(OR=4.807,P<0.01) are associated with RA in Chinese Han population.DR4-DQA1*0301(OR=8.437,P<0.05) and DR4-DQB1*0401(OR=3.215,P<0.01) show their linkage in these patients.However,HLA-DQA1*0102(OR=0.352,P<0.05)、DQB1*0602(OR=0.404,P<0.01) and DQB1*0604(OR=0,P=0.05) play a protection role in RA in the same population.Conclusions HLA-DQA1*0301,DQB1*0401 and DR4-DQA1*0301 are susceptible,while HLA-DQA1*0102,DQB1*0602 and DQB1*0604 are protective to RA in Chinese Han population.DR4-DQA1*0301 and DR4-DQB1*0401 show their linkage in RA in the same population.
Association of HLA-DRB1 locus with treatment response to abatacept or TNF inhibitors in patients with seropositive rheumatoid arthritis

Abstract:The strongest genetic risk factor for rheumatoid arthritis (RA) has been known as HLA-DRB1 based on amino acid positions 11, 71, and 74. This study analyzed the association between specific HLA-DRB1 locus and treatment response to abatacept or TNF inhibitors (TNFi) in patients with seropositive RA. A total of 374 Korean RA patients were treated with abatacept (n = 110) or TNFi (n = 264). Associations between HLA-DRB1 and treatment response after 6 months were analyzed using multivariable logistic regression. Seropositive RA patients with HLA-DRB1 shared epitope (SE) had a favorable response to abatacept (OR = 3.67, P = 0.067) and an inversely associated response to TNFi (OR 0.57, P = 0.058) based on EULAR response criteria, but the difference was not statistically significant in comparison to those without SE. In analyses using amino acid positions of HLA-DRB1, a significant association was found between valine at amino acid position 11 of SE and good response to abatacept (OR = 6.46, P = 5.4 × 10 –3 ). The VRA haplotype also showed a good response to abatacept (OR = 4.56, P = 0.013), but not to TNFi. Our results suggest that treatment response to abatacept or TNFi may differ depending on HLA-DRB1 locus in seropositive RA, providing valuable insights for selecting optimal therapy.

Association of human leukocyte antigen alleles with chronic lung diseases in rheumatoid arthritis

Abstract:Objectives: Chronic lung diseases including interstitial lung disease and airway disease (AD) occur in RA patients. Interstitial lung disease and AD in RA are extra-articular manifestations that influence the prognosis quoad vitam of RA. Studies on associations of HLA alleles with RA have been carried out, and shared epitopes of several alleles are reported to be associated with RA susceptibility. Few association studies in RA subpopulations with chronic lung diseases have been conducted. The aim of the study was to identify HLA alleles predisposing to RA phenotypes including the presence of AD. Methods: Associations of HLA-DRB1 and DQB1 alleles with chronic lung diseases in RA were analysed. Results: A positive association was found between the DR4 serological group and resistance to usual interstitial pneumonia [P = 0.0250, odds ratio (OR) 0.62, 95% CI: 0.41, 0.93]. The DR2 serological group was associated with susceptibility to usual interstitial pneumonia (P = 0.0036, OR = 1.86, 95% CI: 1.23, 2.81). An association was found for shared epitopes alleles with bronchiolitic AD (P = 0.0040, OR = 2.06, 95% CI: 1.24, 3.41). DQB1*03:01 was associated with bronchiectatic AD (P = 0.0021, corrected P-value (Pc) = 0.0315, OR = 1.99, 95% CI: 1.30, 3.06), as well as with emphysema (P = 0.0007, Pc = 0.0104, OR = 2.43, 95% CI: 1.49, 3.95). In combined analysis, a predisposing association of DQB1*03:01 (P = 1.94 ×10(-5), Pc = 0.0003, OR = 2.16, 95% CI: 1.53, 3.06) and a negative association of DQB1*03:02 (P = 0.0008, Pc = 0.0117, OR = 0.33, 95% CI: 0.17, 0.67) with bronchiectatic AD or emphysema were observed in RA. Conclusion: The present study identified an association of HLA-DQB1*03:01 with predisposition to, and DQB1*03:02 with resistance to, bronchiectatic AD or emphysema in RA.

Abstract:The dollarization of the domestic banking system represents a source of vulnerability for emerging market countries, and a debate has emerged over whether the exchange rate regime has an impact on dollarization. This paper argues that the regime is far less important than the literature has previously claimed. Unofficial dollarization results from a lack of faith in the domestic currency, which ultimately stems from the belief that the government will not follow policies that promote long-run currency stability. Empirical results indicate that improved government quality reduces unofficial dollarization, while the exchange rate regime plays no direct role in promoting dollarization.

Abstract:Objective The strong genetic contribution of the major histocompatibility complex (MHC) region to rheumatoid arthritis (RA) has been generally attributed to human leukocyte antigen (HLA)-DRB1. However, due to the high polymorphisms and linkage disequilibrium within MHC, it is difficult to define novel and/or independent genetic risks using conventional HLA genotyping or chip-based microarray technology. This study aimed to identify novel RA risk variants by performing deep sequencing for MHC. Methods We first conducted target sequencing for the entire MHC region in 357 anticitrullinated protein antibodies (ACPA)-positive patients with RA and 1001 healthy controls, and then performed HLA typing in an independent case-control cohort consisting of 1415 samples for validation. All study subjects were Han Chinese. Genetic associations for RA susceptibility and severity were analysed. Comparative modelling was constructed to predict potential functions for the newly discovered RA association variants. Results HLA-DQ alpha 1:160D conferred the strongest and independent susceptibility to ACPA-positive RA (p=6.16x10(-36), OR=2.29). DR beta 1:37N had an independent protective effect (p=5.81x10(-16), OR=0.49). As predicted by comparative modelling, the negatively charged DQ alpha 1:160D stabilises the dimer of dimers, thus may lead to an increased T cell activation. The negatively charged DR beta 1:37N encoding alleles preferentially bind with epitope P9 arginine, thus may result in a decreased RA susceptibility. Conclusions We provide the first evidence that HLA-DQ alpha 1:160D, instead of HLA-DRB1*0405, is the strongest and independent genetic risk for ACPA-positive RA in Han Chinese. Our study also illustrates the value of deep sequencing for fine-mapping disease risk variants in the MHC region.
The HLA‐DRB1 Shared Epitope is Not Associated with Antibodies Against Cyclic Citrullinated Peptide in Chinese Patients with Rheumatoid Arthritis

Abstract:Objective: Rheumatoid arthritis (RA) is a destructive autoimmune polyarthritis that has been associated with a group of human leucocyte antigen (HLA)-DRB1 alleles that share a common amino-acid sequence at residues 70-74 called the shared epitope (SE). Recently, anti-cyclic citrullinated peptide (CCP) antibodies have also been reported to be associated with HLA-DR4 and have gained wide acceptance as early diagnostic markers for RA in Caucasian patients. The current study was performed to investigate whether the association between the SE (HLA-DRB1*0401/04/05/10) and anti-CCP antibodies is also present in Chinese Han patients with RA.Methods: One hundred and four RA patients and 122 healthy controls were recruited. HLA-DR4 was detected by the sequence-specific primer polymerase chain reaction (SSP-PCR) phototyping method. Anti-CCP antibodies and immunoglobulin M rheumatoid factor (IgM-RF) were measured by enzyme-linked immunosorbent assay (ELISA) and laser nephelometry, respectively.Results: Of the Chinese patients with RA, 76.5% exhibited anti-CCP antibodies compared with none of the controls (76.5% vs. 0%, p<0.001). The prevalence of the SE was significantly higher in patients with RA compared with controls [p=0.010, odds ratio (OR)=2.42, 95% confidence interval (CI)=1.16-5.07]. Among the HLA-DR4 alleles, the presence of HLA-DRB1*0401 was significantly higher in RA patients than in controls (p=0.0118, OR=9.68, 95% CI=1.13-448.8). In our study we found that the SE was not associated with production of anti-CCP antibodies (p=0.2899, OR= 1.920, 95% CI=0.52-8.89).Conclusions: The prevalence of the SE is significantly lower in Chinese RA patients, as compared with previous reports of a study using a Caucasian cohort, indicating that distinct genetic risk factors might be associated with anti-CCP antibody production. These data emphasized the complexity of the genetic effects of the major histocompatibility complex on the RA phenotype.

Abstract:OBJECTIVE:To investigate the relationship between HLA-DR4/1 subtypes and T cell response to influenza virus hemagglutinin (HA) as well as anti-HA308-317 antibodies in rheumatoid arthritis (RA). METHODS:The peripheral blood mononuclear cells (PBMCs) from 70 RA patients were cultured with HA308-317 for 5 days. T cell proliferative responses to HA308-317 were evaluated by [3H] thymidine incorporation assay. ELISA was used to detect the antibodies to HA308-317. HLA-DR4/1 subtypes were analyzed by PCR with sequence-specific primers. RESULTS:Twenty-seven of the 70 RA patients (38.6%) were positive for HLA-DR4/1 subtypes and 43 patients (61.4%) were negative for HLA-DR4/1 subtypes. In the HLA-DR4/1 positive group, T cell proliferative response to HA308-317 was observed in 17 (62.9%) RA patients and anti-HA308-317 antibodies were present in 15 RA patients (55.6%). In the HLA-DR4/1 negative group, T cell proliferative response to HA308-317 was observed in 10 (23.3%) patients and anti-HA308-317 antibodies were present in 25 (58.1% ) patients. T cell proliferative response to HA308-317 in the HLA-DR4/1 positive group was higher than in the negative group (P<0.01). However, there was no significant difference in production of antibodies to HA308-317 between the HLA-DR4/1 positive and negative groups (P>0.05). CONCLUSION:There is a relationship between HLA-DR4/1 subtypes and T cell response to HA308-317. Antibodies to HA308-317 are not associated with HLA-DR4/1 phenotype. HA308-317 peptide may be involved in the pathogenesis of RA through activating HLA-DR4/1 specific T cells.

Abstract:The association between Human Leukocyte Antigen (HLA) class II and rheumatoid arthritis (RA) has been extensively studied, but few reported DR-DQ haplotype. Here we investigated the association of HLA-DRB1, DQA1, DQB1, and DR-DQ haplotypes with RA susceptibility and with anti-CCP antibodies in 281 RA patients and 297 control in Han population. High-resolution genotyping were performed. The HLA-DRB1 shared epitope (SE)-encoding allele *0405 displayed the most significant RA association (P = 1.35x10(-6)). The grouped DRB1 SE alleles showed great association with RA (P = 3.88x10(-13)). The DRB1 DRRAA alleles displayed significant protective effects (P = 0.021). The SE-dependent DR-DQ haplotype SE-DQ3/4/5 remained strong association with both anti-CCP -positive (P = 3.71x10(-13)) and -negative RA (P = 3.89x10(-5)). Our study revealed that SE alleles and its haplotypes SE-DQ3/4/5 were highly associated with RA susceptibility in Han population. The SE-DQ3/4/5 haplotypes were associated with both anti-CCP positive RA and -negative RA.
A Variant of Hla-Dr4 Determines Susceptibility to Rheumatoid-Arthritis in A Subset of Israeli Jews

Abstract:HLA-DR4 is associated with risk for developing rheumatoid arthritis (RA) in most populations. In Israeli Jews, in whom the Dw10 subtype of DR4 predominates, no association of RA with DR4 has been found. The inability to detect an association could be due to the high frequency of DR4-Dw10. We used DNA typing with amplification by the polymerase chain reaction and dot-blotting with allele-specific oligonucleotides to determine DR4 variants in 131 Jewish RA patients living in Israel and 134 controls. In both Ashkenazi Jews and non-Ashkenazi Jews, the rare variant Dw15 (previously identified in Japanese populations and in Japanese patients with RA) was found to be the main allele associated with the risk of developing RA (relative risk = 9.2, corrected P less than 0.001). However, this low-frequency allele could be responsible for susceptibility in only 11.5% of the patients. Susceptibility for rheumatoid factor-positive RA was associated with Dw4 and Dw15; the risk for rheumatoid factor-negative RA was associated only with Dw14. The distribution of the HLA-DQ alleles associated with DR4 showed that more than half of the RA patients with Dw15 also had HLA-DQw2. The frequencies of DQw7 and DQw8 were not different in RA patients compared with controls. The results suggest that, as in other populations, susceptibility for the development of RA in Israeli Jews is associated with DRB1 locus alleles of the DR4 group.

Abstract:The relative distributions of 480 DR2-related DR,DQ haplotypes have been determined in Australian Aborigines, Papua New Guinean Highlanders, coastal Melanesians, Micronesians, Polynesians, Javanese, and Southern and Northern Chinese. Using sequence-specific oligonucleotides (SSOs) for hybridization of polymerase chain reaction (PCR) products from DRB1, DRB5, DQA1, and DQB1 genes, 15 different DR2-related haplotypes were identified. The predominant DR2 haplotype in Oceania involved a novel combination of DRB1*1502,DRB5*0101 alleles; this haplotype occurred sporadically in Java, but not in China. In Southern China, the most frequent DR2 haplotype involved the unusual arrangement DRB1*1602,DRB5*0101; alternatively, DRB1*1602 was associated with a new DRB5 SSO pattern. This study has important implications for molecular HLA-typing protocols that assume particular DRB1, DRB5 or DR,DQ linkage relationships. Further, the novel DRB1,DRB5 haplotype in Oceania suggests that the mixed lymphocyte culture (MLC) determinants Dw2 and Dw12 are discriminated by codon 86 at the DRB1 locus.
Novel genetic variants of HLA gene associated with Thai Behcet's disease (BD) patients using next generation sequencing technology

Abstract:Behçet's disease (BD) manifests as an autoimmune disorder featuring recurrent ulcers and multi-organ involvement, influenced by genetic factors associated with both HLA and non- HLA genes, including TNF-α and ERAP1 . The study investigated the susceptible alleles of both Class I and II molecules of the HLA gene in 56 Thai BD patients and 192 healthy controls through next-generation sequencing using a PacBio kit. The study assessed 56 BD patients, primarily females (58.9%), revealing diverse manifestations including ocular (41.1%), vascular (35.7%), skin (55.4%), CNS (5.4%), and GI system (10.7%) involvement. This study found associations between BD and HLA-A*26:01:01 (OR 3.285, 95% CI 1.135–9.504, P -value 0.028), HLA-B*39:01:01 (OR 6.176, 95% CI 1.428–26.712, P -value 0.015), HLA-B*51:01:01 (OR 3.033, 95% CI 1.135–8.103, P -value 0.027), HLA-B*51:01:02 (OR 6.176, 95% CI 1.428–26.712, P -value 0.015), HLA-C*14:02:01 (OR 3.485, 95% CI 1.339–9.065, P -value 0.01), HLA-DRB1*14:54:01 (OR 1.924, 95% CI 1.051–3.522, P -value 0.034), and HLA-DQB1*05:03:01 (OR 3.00, 95% CI 1.323–6.798, P -value 0.008). However, after Bonferroni correction none of these alleles were found to be associated with BD. In haplotype analysis, we found a strong linkage disequilibrium in HLA-B*51:01:01 , HLA-C*14:02:01 ( P -value 0.0, Pc-value 0.02). Regarding the phenotype, a significant association was found between HLA-DRB1*14:54:01 (OR 11.67, 95% CI 2.86–47.57, P -value 0.001) and BD with ocular involvement, apart from this, no distinct phenotype- HLA association was documented. In summary, our study identifies specific HLA associations in BD. Although limited by a small sample size, we acknowledge the need for further investigation into HLA relationships with CNS, GI, and neurological phenotypes in the Thai population.

Abstract:The highly polymorphic Human Leukocyte Antigen system encompasses different loci that have been studied in transplantation as well as diseases and population associated research. This study is the first and largest of its kind to describe the distribution of HLA-A, -B and -C alleles in Lebanon. Respectively, 1994, 1309 and 1163 Lebanese individuals referred for HLA typing and possible bone marrow/kidney donation were tested for HLA-A, HLA-B and HLA-C alleles using the polymerase chain reaction/Sequence specific priming (PCR-SSP) method. Our data were compared to that of several populations with interesting and common findings shared with the Moroccan, Jordanian, Tunisian, Omani, Korean, Chinese, Japanese, Peruan, Bulgarian, Irish, Polish, Spanish, Swiss, American, African and Brazilian populations. The following data concerning the Lebanese population will help future investigators to study the relation of HLA-A, -B and -C alleles with common diseases in Lebanon and will add to the available international literature. This new data will serve as a major reference report in the region.
Can Ferumoxytol be Used as a Contrast Agent to Differentiate Between Acute and Chronic Inflammatory Kidney Disease?: Feasibility Study in a Rat Model

Abstract:ObjectivesFerumoxytol, an intravenous iron supplement, can be used in off-label mode as a contrast agent in magnetic resonance imaging. The aim of this study was to assess whether ferumoxytol can be used as a marker of inflammation in animal models of acute and chronic inflammatory kidney diseases. Material and MethodsThe institutional animal care committee approved this study. A total of 18 rats were examined: 6 healthy Sprague Dawley rats as a control group; 6 rats with polycystic kidney disease (PKD) as a model for chronic inflammatory disease; Thy-1, an antibody triggering glomerulonephritis, was injected in 6 rats as a model for acute inflammation. Each rat was examined directly before and 24 hours after intravenous administration of ferumoxytol at a dose of 30 mg Fe/kg body weight. T2* times of renal tissue were approximated using a multiecho sequence. Changes in relative T2* times and T2 signal intensity after ferumoxytol injection were calculated. ResultsStatistically significant differences between the 3 groups were found: the T2* times of both, Thy-1 and PKD rats were statistically significant different compared with the control group (T2* time ratio after/before: Thy-1, 0.21; PKD, 0.19, control, 0.28; P = 0.002). The highest T2 signal loss in the renal cortex was observed in the Thy-1 rats (T2 signal intensity ratio after/before: Thy-1, 0.49; PKD, 0.79; control, 0.78; P = 0.0005). ConclusionsFerumoxytol-enhanced magnetic resonance imaging allows detection and differentiation of acute and chronic inflammatory kidney disease based on different patterns of parenchymal ferumoxytol depositions. Ferumoxytol thus might help to differentiate between different types of inflammation in various kidney diseases.

Abstract:OBJECTIVE:To investigate whether susceptibility or resistance to allergic rhinitis associated with HLA-DRB alleles.METHOD:Using PCR-SSP (sequence-specific primer polymerase chain reaction), we tested the frequency distribution of HLA-DRB alleles in 41 patients with allergic rhinitis (AR) and 41 healthy controls from Beijing China.RESULT:The frequence of HLA-DRB1 * 0301.2 and HLA-DRB4 * 0101 was lower in AR than in controls (2.44% vs 17.07%, P < 0.05; 29.27% vs 51.22%, P < 0.05).CONCLUSION:HLA-DRB1 * 0301.2 and HLA-DRB4 * 0101 alleles might confer protection against AR.
