Abstract:Rationale: Motile cilia are essential for airway clearance and when genetically impaired result in chronic airway infections that characterize the disease primary ciliary dyskinesia (PCD).Given the complex structure of motile cilia, it is not surprising that mutations in nearly 50 proteins result in PCD.Assembly proteins are a class of proteins found in the cytoplasm that when mutant result in absent ciliary axonemal dynein motors.Based on studies in single cell and non-mammalian model organisms, a group of about 12 proteins called dynein axonemal assembly factors (DNAAFs) are thought to interact within discrete foci in the cytoplasm.The DNAAFs are proposed to orchestrate the construction of dynein motor complexes prior to transport to the cilia.Little is known about where and how this process occurs, and less is known about the process in human airway cells.Methods: Immunofluorescent microscopy was used to localize complexes of assembly proteins in normal human lung tissue.Protein-protein interaction and stoichiometry were determined by fluorescence fluctuation spectroscopy (FFS) of fluorescently-tagged assembly proteins expressed in HEK293T cells.Results: Immunolocalization of a group of assembly proteins known to be mutant in PCD (DNAAF2, DNAAF5, SPAG1) were expressed primarily in ciliated airway cells.Cells showed similar patterns of cytoplasmic distribution, featuring a single perinuclear punctum in some cells, with smaller puncta distributed throughout the cytoplasm.This pattern is different from the appearance in model organisms such as Xenopus which feature several puncta within the cytoplasm.Co-chaperone protein RUVBL1, required for common cellular protein handling, showed an expression pattern similar to the DNAAFs.Client dynein protein DNAI1 was identified only within the perinuclear puncta during airway cell differentiation, and had a lower cytoplasmic signal in mature cells.FFS showed that the three DNAAFs examined exist in large multimers, consistent with a scaffold-like function for dynein motor protein assembly.Expression of human PCD mutant DNAAF2, DNAAF5, and SPAG1 in HEK293 cells resulted in large cytoplasmic collections, which extended uniquely from a perinuclear punctum.Conclusions: These observations suggest DNAAFs colocalize in perinuclear puncta in large multimers.Their location differs in human airway cells from that described in model systems.The localization of the mutant proteins responsible for PCD provides fundamental information in understanding normal and mutant dynein protein assembly for the cilia function.Mutant protein localization establishes key information for testing currently conceived therapeutic strategies, such as complementation of the mutant protein with a normal one.
