Abstract:The effects of metastable energy level and peptide sequence on the fragmentation patterns of singly charged peptide ions dissociated in collisions with metastable rare gas atoms were studied. Fragmentation spectra of singly charged peptide ions were shown to be more structure-informative and very different from those obtained in low-energy collision-induced dissociation. Unusual odd-electron radical a- and x-ions were observed. Several fragment ions corresponding to a side-chain loss were also observed, which allowed differentiation between leucine and isoleucine. The fragmentation mechanism depends on electronic energy transferred during interaction with metastable gas atoms and proceeds either via Penning ionization with formation of radical odd-electron doubly charged molecular cation or via high-energy excitation of internal degrees of freedom of the peptide cation.

Abstract:Introduction: Understanding the sex determination mechanisms in birds has great significance for the biological sciences and production in the poultry industry. Sex determination in chickens is a complex process that involves fate decisions of supporting cells such as granulosa or Sertoli cells. However, a systematic understanding of the genetic regulation and cell commitment process underlying sex determination in chickens is still lacking. Objectives: We aimed to dissect the molecular characteristics associated with sex determination in the gonads of chicken embryos. Methods: Single-nucleus RNA-seq (snRNA-seq) and ATAC-seq (snATAC-seq) analysis were conducted on the gonads of female and male chickens at embryonic day 3.5 (E3.5), E4.5, and E5.5. Results: Here, we provided a time-course transcriptional and chromatin accessible profiling of gonads during chicken sex determination at single-cell resolution. We uncovered differences in cell composition and developmental trajectories between female and male gonads and found that the divergence of transcription and accessibility in gonadal cells first emerged at E5.5. Furthermore, we revealed key cell-type-specific transcription factors (TFs) and regulatory networks that drive lineage commitment. Sex determination signaling pathways, dominated by BMP signaling, are preferentially activated in males during gonadal development. Further pseudotime analysis of the supporting cells indicated that granulosa cells were regulated mainly by the TEAD gene family and that Sertoli cells were driven by the DMRT1 regulons. Cross-species analysis suggested high conservation of both cell types and cell-lineage-specific TFs across the six vertebrates. Conclusions: Overall, our study will contribute to accelerating the development of sex manipulation technology in the poultry industry and the application of chickens as a unique model for studying cell fate decisions.

Abstract:The ability to detect and remove male chicks pre-hatch would be a big step forward to the egg-laying and related industries. The current practice of culling male chicks post-hatch creates a major ethical dilemma for many countries. Hatching out and growing male layer chicks is not a sustainable option for farmers. A genetic based in ovo sex selection application would effectively negate the need to cull or grow out male chickens and would contribute to a more sustainable industry with a view to future food security. Recent advancements in avian gene technology allow specific marking of the sex-determining chromosome in chickens so that the males can be identified before hatching and removed before incubation. This provides a simple solution to meet a pressing need for the industry and a leading opportunity for the adoption of biotechnology in animal agriculture.

Abstract:Japanese agricultural scientist Toyama Kametaro's report about the Mendelian inheritance of silkworm cocoon color in Studies on the Hybridology of Insects (1906) spurred changes in Japanese silk production and thrust Toyama and his work into a scholarly exchange with American entomologist Vernon Kellogg. Toyama's work, based on research conducted in Japan and Siam, came under international scrutiny at a time when analyses of inheritance flourished after the "rediscovery" of Mendel's laws of heredity in 1900. The hybrid silkworm studies in Asia attracted the attention of Kellogg, who was concerned with how experimental biology would be used to study the causes of natural selection. He challenged Toyama's conclusions that Mendelism alone could explain the inheritance patterns of silkworm characters such as cocoon color because they had been subject to hundreds of years of artificial selection, or breeding. This examination of the intersection of Japanese sericulture and American entomology probes how practical differences in scientific interests, societal responsibilities, and silkworm materiality were negotiated throughout the processes of legitimating Mendelian genetics on opposite sides of the Pacific. The ways in which Toyama and Kellogg assigned importance to certain silkworm properties show how conflicting intellectual orientations arose in studies of the same organism. Contestation about Mendelism took place not just on a theoretical level, but the debate was fashioned through each scientist's rationale about the categorization of silkworm breeds and races and what counted as "natural". This further mediated the acceptability of the silkworm not as an experimental organism, but as an appropriately "natural" insect with which to demonstrate laws of inheritance. All these shed light on the challenges that came along with the use of agricultural animals to convincingly articulate new biological principles.

Abstract:Aquaculture is one of the most efficient modes of animal protein production and plays an important role in global food security. Aquaculture animals exhibit extraordinarily diverse sexual phenotypes and underlying mechanisms, providing an ideal system to perform sex determination research, one of the important areas in life science. Moreover, sex is also one of the most valuable traits because sexual dimorphism in growth, size, and other economic characteristics commonly exist in aquaculture animals. Here, we synthesize current knowledge of sex determination mechanisms, sex chromosome evolution, reproduction strategies, and sexual dimorphism, and also review several approaches for sex control in aquaculture animals, including artificial gynogenesis, application of sex-specific or sex chromosome-linked markers, artificial sex reversal, as well as gene editing. We anticipate that better understanding of sex determination mechanisms and innovation of sex control approaches will facilitate sustainable development of aquaculture.

Abstract:Although the basic principles controlling the sex of mammalian offspring have been known for a relatively long time, recent application of certain modern cellular methodologies has led to development of a flow cytometric system capable of differentiating and separating living X- and Y-chromosome-bearing sperm in amounts suitable for AI and therefore, commercialization of this sexing technology. After a very long history of unsuccessful attempts to differentiate between mammalian sperm that produce males from those that produce females, a breakthrough came in 1981 when it was demonstrated that precise DNA content could be measured. Although these initial measurements of DNA content killed the sperm in the process, they led to the ultimate development of a sperm sorting system that was capable, not only of differentiating between live X- and Y-sperm, but of sorting them into relatively pure X- and Y-sperm populations without obvious cellular damage. Initial efforts to predetermine the sex of mammalian offspring in 1989 required surgical insemination, but later enhancements provided sex-sorted sperm in quantities suitable for use with IVF. Subsequent advances in flow sorting provided minimal numbers of sperm sufficient for use in AI. It was not until the flow cytometric sorting system was improved greatly and successful cryopreservation of sex-sorted bull sperm was developed that efficacious approaches to commercialization of sexed semen could be implemented worldwide in cattle. A number of companies now offer sex-sorted bovine sperm. Innovative approaches by a diverse group of scientists along with advances in computer science, biophysics, cell biology, instrumentation, and applied reproductive physiology provided the basis for commercializing sexed semen in cattle.
Effect of Aromatase Inhibition on Gonadal Differentiation and Sexual Behavior of Chickens

Abstract:In the present study aromatase inhibitor was injected into air sac to reveal function of estrogen in differentiation of Hyline chicken gonads and sexual behavior.After sexual maturation,the sexual behavior and serum steroid levels were checked.The results showed that the treatment of aromatase induced masculinization of the secondary sexual characteristics and mating behavior in the sex reversed female chickens.Histological examination manifested different degree of gonadal reversal:(1)completely reversed hens had right testis and left ovotestis.In both gonads there were seminiferous tubules filling with germ cells,but the diameter of the tubules were less than that of the normal cock.(2)incompletely reversed hens had regressed right gonad and intact left ovary.The serum E 2 levels were in such an order as:incompletely reversed hennormal hencompletely reversed hennormal cock,and the serum T levels as:completely reversed henincompletely reversed hennormal cocknormal hen.Frequency of male mating behavior correlated positively with ratio of serum T/E 2.The above results indicated that inhibition of estrogen synthesis before sexual differentiation induced masculinization of gonadal structure,steroid secretion and mating behavior,and the serum T and E 2 levels and their ratio determined the frequency and strength of male mating behavior of the sex reversed female chickens.
Overview of chicken embryo genes related to sex differentiation

Abstract:Sex determination in chickens at an early embryonic stage has been a longstanding challenge in poultry production due to the unique ZZ:ZW sex chromosome system and various influencing factors. This review has summarized the genes related to the sex differentiation of chicken early embryos (mainly Dmrt1 , Sox9 , Amh , Cyp19a1 , Foxl2 , Tle4z1 , Jun , Hintw , Ube2i , Spin1z , Hmgcs1 , Foxd1 , Tox3 , Ddx4 , cHemgn and Serpinb11 in this article), and has found that these contributions enhance our understanding of the genetic basis of sex determination in chickens, while identifying potential gene targets for future research. This knowledge may inform and guide the development of sex screening technologies for hatching eggs and support advancements in gene-editing approaches for chicken embryos. Moreover, these insights offer hope for enhancing animal welfare and promoting conservation efforts in poultry production.

Abstract:In the present study aromatase inhibitor (AI) was injected into air sac of chicken embryos to reveal thefunction of estrogen in the sexual behaviors and the development of the medial preoptic nucleus (POM) in the hypothalamus.After sexual maturation, sexual behaviors and serum steroid levels were checked. The results showed that the treatment ofAI induced masculinization of the mating behaviors in the sex-reversed hens. Estradiol (E2) and testosterone (T) levelschanged, and the frequency of masculine mating behaviors was correlated positively with the ratio of serum T/E2. POM cellsof 1-day-old female and male chicken had no obvious difference, but the cell size became larger after AI treatment. Therewere no obvious changes of the morphology of the POM cells in adult sex-reversed hens compared with the control, but thecell density and the cell size decreased greatly. These results indicated that inhibition of aromatase activity during theembryonic period caused the serum sex steroid and the sexual behaviors towards the male and the frequency of male matingbehaviors were correlated positively with the ratio of serum T/E2. The POM structure of 1-day-old chickens did not existsexual difference, while cell density and cell size of the adult chickens changed.

Abstract:Background: Sex determination and differentiation are complex and delicate processes. In female chickens, the process of sex differentiation is sensitive and prone to be affected by the administration of aromatase inhibitors, which result in chicken sex reversal and infertility. However, the molecular mechanisms underlying sex differentiation and infertility in chicken sex reversal remain unclear. Therefore, we established a sex-reversed chicken flock by injecting an aromatase inhibitor, fadrozole, and constructed relatively high-resolution profiles of the gene expression and chromatin accessibility of embryonic gonads. Results: We revealed that fadrozole affected the transcriptional activities of several genes, such as DMRT1, SOX9, FOXL2, and CYP19A1, related to sex determination and differentiation, and the expression of a set of gonadal development-related genes, such as FGFR3 and TOX3, by regulating nearby open chromatin regions in sex-reversed chicken embryos. After sexual maturity, the sex-reversed chickens were confirmed to be infertile, and the possible causes of this infertility were further investigated. We found that the structure of the gonads and sperm were greatly deformed, and we identified several promising genes related to spermatogenesis and infertility, such as SPEF2, DNAI1, and TACR3, through RNA-seq. Conclusions: This study provides clear insights into the exploration of potential molecular basis underlying sex differentiation and infertility in sex-reversed chickens and lays a foundation for further research into the sex development of birds.

Abstract:The present experiment was designed to investigate a possibility of the in vitro cultured somatic cells to produce the germ line interspecific chimera in bird. The frozen-thawed fibroblast-like cells derived from a male 9-day embryo of Indian peafowl were cultured and treated with polyethylene glycol(PEG, mw8000). The treated cells were injected into the blastoderm of unincubated fertile White Leghorn eggs(at stage X). The manipulated eggs were incubated until hatching. A total of 108 White Leghorn eggs were manipulated with the injection of about 800-1600 peafowl cells for each egg. The chimerism was assesed by observation of the plumage and DNA analysis. To do DNA assay, peafowl mitochondrial cytochrome b (cyt b) gene carrying species-specific DNA loci with 418 bp fragments was adopted as a genetic marker. Of 108 manipulated eggs, only four hatched and two of them revealed partially pigmented feather mosaicism, denoting the existence of peafowl-specific cyt b gene in the feather pulp. In addition, the peafowl-contributed mosaic tissues were also found in the muscle, testis and ovary of dead embryos and one chick. The present results demonstrate that the cultured peafowl fibroblast-like cells, which were treated with PEG before the injection into the blastoderm of White Leghorn eggs, differentiated and integrated into the recipient tissues during embryonic development. This experiment also suggests that intersepcific chimera birds may be produced by using this method in the future.

Abstract:Studies on inheritance of a number of traits, peculiarities of the karyotype, and chromosome mapping in chicken are reviewed. Prominent among these are studies on behavioral genetics, which are performed at different levels and in different aspects: neuromuscular apparatus and entire organism, behavior and population structure, and behavior and individual fecundity. Creation of lines that differed in neural reactivity provided a model for study of systemic control of cytogenetic processes. On the basis of data on genetic control of feather color, a hypothesis was proposed concerning the mechanism of action of the eumelanin suppressor gene. In the Department of Genetics and Breeding of St. Petersburg State University, research was reformed on the relationships between hereditary and non-hereditary variability, as well as the determination and realization of the genotype reaction norm. In the framework of this research, a study on the variability of chicken growth was performed. A single short-term cooling of chickens during the first week of life resulted in a prolonged (up to eight-week) alteration of growth rate. The following structural genes were mapped in chicken mitotic chromosomes by the method of in situ hybridization: the gene for the transferrin receptor, the beta-globin gene cluster, the homologues of the human ZFY and SRY gene families, the viral oncogene v-fos, and a TG-rich minisatellite.
Possibilities for genetic manipulation of sex ratio in livestock

Abstract:Reports are reviewed in which genetic differences for sex ratio of offspring were found among breeds, breed crosses, strains, lines, line crosses, families or individuals. Although reports in which genetic variation could not be identified outnumber those in which it could, important genetic variation does exist in at least some populations. The direct or correlated response of sex ratio to selection is reviewed. Again, results are equivocal, but there is ample evidence that sex ratio will respond to selection in at least some populations of laboratory organisms. Segregation distortion in the mouse and in Drosophila is examined. Segregation distortion occurs when a heterozygous male mated to a homozygous female does not produce the two offspring types in equal proportions and the disparity cannot be accounted for by sampling or differential mortality. Although the most well-known systems have no effect (in the mouse) or only a moderate effect (in Drosophila) on sex ratio, systems in which sex ratio is subject to segregation distortion are known to exist in insects and could exist in mammals. Finally, the possibility is discussed that XX/XY chromosome chimaeric bulls, born co-twin to a heifer, might produce a preponderance of heifer progeny. A few reports suggest this might be the case, but other research is not in agreement.

Abstract:Despite the notable progress made in recent years, the understanding of the genetic control of gonadal sex differentiation and asymmetrical ovariogenesis in chicken during embryonic development remains incomplete. This study aimed to identify potential key genes and speculate about the mechanisms associated with ovary and testis development via an analysis of the results of PacBio and Illumina transcriptome sequencing of embryonic chicken gonads at the initiation of sexual differentiation (E4.5, E5.5, and E6.5). PacBio sequencing detected 328 and 233 significantly up-regulated transcript isoforms in females and males at E4.5, respectively. Illumina sequencing detected 95, 296 and 445 DEGs at E4.5, E5.5, and E6.5, respectively. Moreover, both sexes showed asymmetrical expression in gonads, and more DEGs were detected on the left side. There were 12 DEGs involved in cell proliferation shared between males and females in the left gonads. GO analysis suggested that coagulation pathways may be involved in the degradation of the right gonad in females and that blood oxygen transport pathways may be involved in preventing the degradation of the right gonad in males. These results provide a comprehensive expression profile of chicken embryo gonads at the initiation of sexual differentiation, which can serve as a theoretical basis for further understanding the mechanism of bird sex determination and its evolutionary process.

Abstract:The determination of sex in mammals is established and controlled by various complex mechanisms. In contrast, sex control in poultry remains an unresolved issue. In this study, RNA-sequencing was conducted for male gonads and ovarian tissues in chicken embryos of up to 18.5 days to identify metabolic factors influencing male and female sex differentiation, as well as gonadal development. Our results reveal that PKM2, a critical glycolysis-related protein, plays a significant role in chicken sex differentiation via PPARG, a crucial hormone gene. We propose that our discoveries bolster the notion that glycolysis and oxidative phosphorylation function as antecedent contributors to sexual phenotypic development and preservation.
Morphological variation of tail bone among two chicken breeds and their F1 progeny

Abstract:Fancy breeds of Japanese indigenous chicken display extensive morphological diversity. There are marked differences in tail and bone traits between Tosa‐jidori and Minohikichabo breeds. Using parental breeds and their F1 hybrid, significant differences on strain and sex were revealed in morphometric parameters of pygostyle, tail feather number, and body weight. Interestingly, different patterns of genetic inheritance in the F1 hybrid were observed at different locations, even within the same pygostyle bone. Fancy breeds of Japanese indigenous chicken display extensive morphological diversity, particularly in tail feathers. Although marked differences in tail and bone traits have been reported between Tosa‐jidori (wild type) and Minohikichabo (rich type) breeds, little is known about the pattern of genetic inheritance in cross experiments. Therefore, this study aimed to investigate the strain and sex effects, and inheritance patterns, in the morphometric variation of pygostyle bones among Tosa‐jidori, Minohikichabo, and their F1 hybrids. Five morphological traits, angle of the apex of the pygostyle, pygostyle length, margo cranialis length, tail feather number, and body weight, were evaluated at the adult stage. A significant strain difference was detected in all traits, whereas significant sex differences were observed in only three traits, but not in the angle of the apex of the pygostyle and tail feather number. In F1 hybrids, the angle of the apex of the pygostyle was significantly different to that of Tosa‐jidori but not that of Minohikichabo, whereas the pygostyle length and tail number of F1 hybrids were significantly different from those of Minohikichabo but not those of Tosa‐jidori. A significant heterosis effect was found in the margo cranialis length and body weight. All five traits showed nonadditive inheritance patterns but varied in each trait between partial dominance (angle of the apex of pygostyle), full dominance (pygostyle length and tail feather number), and over‐dominance (margo cranialis length and body weight). Interestingly, different patterns of genetic inheritance in the F1 hybrid were observed at different locations, even within the same pygostyle bone. Using the Japanese indigenous chicken model, these results provide a substantial step toward understanding the genetic architecture of morphology in chickens.

Abstract:The sex determination and control of poultry is a key problem in production and scientific research despite few studies on regulatory factors, especially transcription factors in sex determination. In the early stage of this study, high-throughput sequencing was used to screen the differentially expressed gene JUN in male and female embryonic stem cells (ESCs) and primordial germ cells (PGCs). The qRT-PCR discovered that the JUN gene significantly increased from embryonic days (E) 2.5 later in chicken embryo development, and the female gonad expression was much higher than that of the male after E14.5. Lentivirus shRNA-JUN, shRNA-Smad2 interference, and OE-JUN overexpression vectors were successfully constructed. After interfering with JUN in vivo, male characteristics appeared in ZW embryonic gonads at E18.5. Meanwhile, the male-specific genes DMRT1 and Sox9 were upregulated, the female-specific genes FOXL2, ESR1, and CYP19A1 were downregulated, and the estradiol in the gonads was significantly decreased. The situation was reversed after the overexpression of JUN, ZZ chicken embryo developed into female sexual characteristics. The double luciferase report has found that the Smad2 promoter activity was significantly upregulated after interference with JUN, and significantly increased after the deletion of the JUN binding site. After the injection of the Smad2-shRNA vector into the blood vessel in vivo, it was discovered that DMRT1 and Sox9 of ZW embryos at E18.5 were downregulated, FOXL2 and CYP19A1 were significantly upregulated, and the gonads show femininity. In conclusion, this study proves that JUN is a key regulator in the process of chicken female sex differentiation, which can inhibit the transcription of Smad2 and promote the synthesis of estradiol, and participate in the process of chicken sex differentiation. This study lays a foundation for the analysis of the molecular mechanism of chicken sex determination and the development of poultry sex control technology.

Abstract:Cephapirin, a new semisynthetic cephalosporin antibiotic for parenteral use was evaluated in vitro and in patients with severe bacterial infections. Strains of Staphylococcus aureus, Group A streptococci, Diplococcus pneumoniae, Proteus mirabilis, Escherichia coli, Klebsiella pneumoniae, pseudomonas, and salmonella were tested in vitro and were found to have the same degree of sensitivity to cephapirin as to cephalothin, with the exception of Group A streptococci and pneumococci which appeared more sensitive to cephapirin. Seven of 10 patients treated were cured clinically and bacteriologically, and in one instance the outcome was indeterminate. Side effects were minimal. One patient developed mild eosinophilia after high dose therapy for six weeks. There was little pain and no phlebitis at the injection or infusion sites. There was no evidence of renal toxicity even after prolonged therapy at high dosage. Serum and urine concentrations of cephapirin were similar to those achieved with equivalent doses of currently available parenteral cephalosporins.
