Abstract:In this study, we developed (W1/O/W2) double emulsions encapsulated by calcium-alginate hydrogel beads system (ACGs) for intestinal-targeted delivery of probiotics (Lactobacillus reuteri). Firstly, the carboxymethyl konjac glucomannan-chitosan (CMKGM-CS) nanogels were successfully fabricated by EDC/NHS initiated crosslinking, as concluded from the TEM images, FTIR spectra, XRD, etc. Then, double emulsions were prepared and encapsulated with various concentrations of alginate to form ACGs hydrogel beads. In vitro probiotic release experiments showed the lyophilized ACG-2 and ACG-3 hydrogel beads had a sustained release in simulated intestinal fluid (SIF), the viability of cells exceeded 107 CFU/mL at 6 h. The lyophilized ACG-3 hydrogel beads exhibited the viable release amount of 8.4 × 107 CFU/mL after storage for 90 d at 4 °C. Besides, the alginate concentration in the ACGs hydrogel beads influences the swelling behavior and structure of hydrogel beads by affecting the hydrogen bonds between alginate and CMKGM-CS, thereby mediating the release of probiotics.
Enhanced Viability of Probiotics (pediococcus Pentosaceus Li05) by Encapsulation in Microgels Doped with Inorganic Nanoparticles

Abstract:The major reduction in probiotic viability that occurs during food storage and gastrointestinal transit limits their potential health benefits. Microencapsulation is one of the most effective methods to protect probiotics from various harsh conditions. In this study, a model probiotic (Pediococcus pentosaceus Li05) was encapsulated in an alginate-gelatin microgels in the absence and presence of magnesium oxide (MgO) nanoparticles (NPs). The morphology and surface properties of the encapsulation systems were characterized by transmission electron microscopy (TEM) and atomic force microscopy (AFM), which showed that both the probiotics and NPs were successfully incorporated into spherical microgels. The viability of the probiotic was evaluated after exposure to different conditions: long-term storage in an aerobic environment; heat treatment; and gastrointestinal transit. Encapsulation of the probiotics significantly enhanced their viability under these different conditions. Probiotics encapsulated in MgO-loaded microgels were more stable than free bacterial cells or those encapsulated in microgels alone: less than 2 log(10) CFU reduction after 40 min incubation in gastric fluids versus 5 log(10) CFU reduction in the first 10 min for free cells. The SEM images indicated that the NPs may lead to enhanced probiotic viability by filling pores inside the microgels, which may have inhibited the ability of oxygen and hydrogen ions to access the probiotics. Moreover, the MgO NPs neutralized the hydrogen ions in the gastric fluids, thereby reducing acid-induced degradation of the probiotics. These results demonstrate that MgO-loaded microgels may be a promising encapsulation and delivery system for improving the efficacy of orally administered probiotics by protecting them from the harsh conditions during storage and in the gastrointestinal tract.

Abstract:In this study, we developed a novel delivery system using carboxymethyl konjac glucomannan-chitosan (CMKGM-CS) nanogels stabilized single and double emulsion incorporated into alginate hydrogel as microcapsule matrix for intestinal-targeted delivery of probiotics. Through in vitro experiments, it was demonstrated that alginate hydrogel provided favorable biocompatible growth conditions for the proliferation of Lactobacillus reuteri (LR). The alginate hydrogel containing single (ASE) or double emulsions (ACG) enhanced the resistance of LR to various adverse environments. Simulated gastrointestinal digestion experiments revealed that the survivability of LR in free, CON, ASE and ACG group decreased by 6.45 log CFU/g, 4.21 log CFU/g, 1.26 log CFU/g and 0.65 log CFU/g, respectively. In vivo studies conducted in mice showed that ACG maintained its integrity during passage through the stomach and released the probiotics in the targeted intestinal area, whereas the pure alginate hydrogels (CON) were prematurely released in the gastrointestinal tract. Moreover, the viable counts of ACG in different intestinal segments (jejunum, ileum, cecum, and colon) were increased by 1.11, 1.42, 1.68, and 1.89 log CFU/g, respectively, after 72 h of oral administration compared to the CON group. This research contributed valuable insights into the development of an effective microbial delivery system with potential applications in the biopharmaceutical and food industries.

Abstract:Biopolymer microgels (hydrogel beads) can be used to improve probiotic stability during food storage and gastrointestinal passage. In this study, the impact of including natural polymeric and colloidal fillers on the viability of Lactobacillus casei encapsulated in calcium alginate microgels was examined. The polymeric filler was polysaccharides isolated from Choerospondias axillaris fruit peel (CP) and the colloidal filler was oil-in-water emulsion stabilized by CP. Results showed that addition of CP or CP coated lipid droplets significantly increased the antioxidant activity but reduced the apparent viscosity of the alginate solutions. Encapsulation of the probiotics in pure calcium alginate microgels reduced their viability (≈ 0 log CFU g−1 after 24 h). The inclusion of the polymeric or colloidal fillers within the microgels eliminated this negative effect. Moreover, they enhanced probiotic viability under simulated gastrointestinal tract conditions (> 7.0 log CFU g−1). The calcium alginate microgels containing CP had the highest probiotic viability (7.42 ± 0.08 log CFU g−1) after 20 days storage. Our results indicated that simple calcium alginate microgels are not effective at protecting Lactobacillus casei during storage or gastrointestinal passage. However, incorporation of suitable polymeric or colloidal fillers can significantly enhance their viability.

Abstract:To improve the viability of Lactobacillus plantarum (P) during digestion and storage, the probiotics were encapsulated by alginate (ALG) and alginate-gelatin (ALG-GE) hydrogels beads. ALG-P-GE showed much better physicochemical properties than ALG-P. The scanning electron microscopy (SEM) results validated the incor-poration of bacterial cells into the beads. ALG-P-GE exhibited good encapsulation efficiency of 97.7 %, and the storage and thermal stability of probiotic were increased by 15 % and 8 %, respectively, when comparing with ALG-P. ALG-P-GE beads could protect the probiotics from inactivation in simulated gastric fluid and then release it in simulated intestinal fluid. The protective mechanism of ALG-GE for probiotics was further studied by fourier transform infrared spectroscopy (FTIR), X-ray diffraction (XRD) and found that ALG and GE can form gel network through hydrogen bonding and electrostatic interactions. In the mimic beverage systems, ALG-P-GE beads could protect the encapsulated probiotics and increase its viability. The storage, thermal, and digestion stability of encapsulated probiotic were significantly increased and showed high viability in the mimic beverage systems. ALG-P-GE beads have great potential for the protection and delivery of probiotics in food systems.
Probiotic Encapsulation: Bead Design Improves Bacterial Performance during In Vitro Digestion

Abstract:The stability and release properties of all bioactive capsules are strongly related to the composition of the wall material. This study aimed to evaluate the effect of the wall materials during the encapsulation process by ionotropic gelation on the viability of Lactobacillus fermentum K73, a lactic acid bacterium that has hypocholesterolemia probiotic potential. A response surface methodology experimental design was performed to improve bacterial survival during the synthesis process and under simulated gastrointestinal conditions by tuning the wall material composition (gelatin 25% w/v, sweet whey 8% v/v, and sodium alginate 1.5% w/v). An optimal mixture formulation determined that the optimal mixture must contain a volume ratio of 0.39/0.61 v/v sweet whey and sodium alginate, respectively, without gelatin, with a final bacterial concentration of 9.20 log10 CFU/mL. The mean particle diameter was 1.6 ± 0.2 mm, and the experimental encapsulation yield was 95 ± 3%. The INFOGEST model was used to evaluate the survival of probiotic beads in gastrointestinal tract conditions. Upon exposure to in the vitro conditions of oral, gastric, and intestinal phases, the encapsulated cells of L. fermentum decreased only by 0.32, 0.48, and 1.53 log10 CFU/mL, respectively, by employing the optimized formulation, thereby improving the survival of probiotic bacteria during both the encapsulation process and under gastrointestinal conditions compared to free cells. Beads were characterized using SEM and ATR-FTIR techniques.

Abstract:The relationship of plasma levels of high density lipoprotein (HDL) cholesterol, apolipoproteins A-I and A-II (the major apolipoproteins in HDL), low density lipoprotein (LDL) cholesterol, triglyceride, and glucose to microangiopathy was evaluated in 49 insulin-dependent diabetic subjects. Although the HDL cholesterol/LDL cholesterol ratio (a risk determinant for macroangiopathy) was lower in women with proteinuria, no other relationships between HDL cholesterol or the A apolipoproteins and renal microangiography were found. The only independent association between HDL and retinal microangiopathy was found in women, where an inverse correlation was found between the apo A-I/apo A-II ratio and the number of microaneurysms (rs = −0.561, P < 0.05). Men showed strong relationships of glucose, triglyceride, cholesterol, and LDL cholesterol to renal microangiopathy whereas women, in general, had stronger correlations of these variables with retinal microangiopathy. Thus, several alterations in lipoprotein cholesterol distribution and HDL composition are associated with diabetic microangiopathy. In addition, differences between sexes suggest that previously undescribed hormonal factors may influence the severity of this process.

Abstract:Biopolymer microgels were designed to encapsulate and protect probiotics during storage and passage through the upper human gastrointestinal tract (GIT), but then release them in the colon. An anaerobic probiotic strain (Bifidobacterium pseudocatenulatum G7; BPG7) was encapsulated within calcium alginate microgels. In some cases, either colloidal antacid (CaCO3) or colloidal antacid and nanoemulsion (nE) lipid droplets were co-encapsulated with the probiotics. These additives were used to control the porosity and/or internal pH of the microgels. Initially, the viability of free and encapsulated probiotics was evaluated when they.Encapsulation greatly improved the stability of the probiotics, with the effectiveness depending on the formulation: antacid-microgels > control microgels > antacid-nE-microgels. These effects were mainly attributed to the ability of the colloidal CaCO3 particles to inhibit molecular diffusion processes. The ability of the microgels to protect the probiotics under simulated gastrointestinal tract (GIT) conditions was then assessed. The free probiotics were completely inactivated when exposed to gastric and small intestine conditions. In contrast, the antacid- and antacid-nE-microgels were highly effective at improving the viability of the probiotics during passage through the upper GIT. Moreover, these microgels were fermented under simulated colonic conditions, thereby releasing the probiotics. These results suggest that antacid-loaded calcium alginate microgels can be used for the encapsulation, protection, and colonic delivery of probiotics. Nevertheless, further studies are required using in vivo models.

Abstract:Natural hydrogels such as alginate are hopeful gadgets for cellular encapsulation, drug delivery, and many others because of their properties, such as biodegradable and non-toxic for the environment. Probiotics, as intestinal microbiota, have numerous benefits for the host's health. On the other hand, the survival of probiotics is critical, especially in the food and medicine industry, and probiotics should endure disagreeable status that occurs in passing through the gastrointestinal system. Encapsulation can be applied to protect probiotics but increase their bio-accessibility, so the survival rate of bacteria and their transportation to different body parts. In this review, the literature on delivery systems results focuses on combining alginates with other biopolymers to produce hydrogels that enclose probiotics, only increasing their encapsulation performance and survival compared to alginates in gastrointestinal simulation conditions collected. All the tastes and limitations, along with the benefits of microencapsulation of probiotics using hydrogels, can lead to the emergence of biopolymers, new ways of protecting probiotics, and ultimately producing products that are safe for consumers.

Preparation and characterization of pectin-alginate-based microbeads reinforced by nano montmorillonite filler for probiotics encapsulation: Improving viability and colonic colonization

Abstract:Hydrogel microbeads can be used to enhance the stability of probiotics during gastrointestinal delivery and storage. In this study, the pectin-alginate hydrogel was enhanced by adding montmorillonite filler to produce microbeads for encapsulating Lactobacillus kefiranofaciens (LK). Results showed that the viscosity of biopolymer solutions with 1 % (PAMT1) and 3 % (PAMT3) montmorillonite addition was suitable for producing regular-shaped microbeads. A layered cross-linked network was formed on the surface of PAMT3 microbeads through electrostatic interaction between pectin-alginate and montmorillonite filler, and the surrounding LK with adsorbed montmorillonite was encapsulated inside the microbeads. PAMT3 microbeads reduced the loss of viability of LK when passing through the gastric acid environment, and facilitated the slow release of LK in the intestine and colonic colonization. The maximum decrease in viability among all filler groups was 1.21 log CFU/g after two weeks of storage, while PAMT3 freeze-drying microbeads only decreased by 0.46 log CFU/g, indicating that the gel layer synergized with the adsorbed layer to provide dual protection for probiotics. Therefore, filler-reinforced microbeads are a promising bulk encapsulation carrier with great potential for the protection and delivery of probiotics and can be developed as food additives for dairy products.
The Effect of Encapsulating a Prebiotic-Based Biopolymer Delivery System for Enhanced Probiotic Survival

Abstract:Orally delivered probiotics must survive transit through harsh environments during gastrointestinal (GI) digestion and be delivered and released into the target site. The aim of this work was to evaluate the survivability and delivery of gel-encapsulated Lactobacillus rhamnosus GG (LGG) to the colon. New hybrid symbiotic beads alginate/prebiotic pullulan/probiotic LGG were obtained by the extrusion method. The average size of the developed beads was 3401 μm (wet), 921 μm (dry) and the bacterial titer was 109 CFU/g. The morphology of the beads was studied by a scanning electron microscope, demonstrating the structure of the bacterial cellulose shell and loading with probiotics. For the first time, we propose adding an enzymatic extract of feces to an artificial colon fluid, which mimics the total hydrolytic activity of the intestinal microbiota. The beads can be digested by fecalase with cellulase activity, indicating intestinal release. The encapsulation of LGG significantly enhanced their viability under simulated GI conditions. However, the beads, in combination with the prebiotic, provided greater protection of bacteria, enhancing their survival and even increasing cell numbers in the capsules. These data suggest the promising prospects of coencapsulation as an innovative delivery method based on the inclusion of probiotic bacteria in a symbiotic matrix.

Abstract:This research aimed to evaluate the encapsulation of the probiotic strain, Streptococcus thermophilus, in hydrogels employing sodium alginate (SA) with κ-carrageenan (κC) in gelation baths with varying salt concentrations (CaCl2 and KCl) followed by freeze-drying. The experimentation was conducted at varying levels of κC (0-0.5 % w/v) and SA (2-4 %). Freeze-dried hydrogels were evaluated based on encapsulation efficiency and loss of viability and further characterised. The study could successfully establish an encapsulation efficiency of 87.814 % and a viability loss of 1.201 log CFU·g-1 for the optimised samples. The SEM micrographs of the optimised Ca-alginate/κC hydrogels exhibited a much denser network with fewer pores. The influence of SA/κC in the beads was confirmed by FTIR and DSC, where distinct peak shifts were observed, which indicated the presence of κC and SA polymers. The probiotic survival under simulated gastrointestinal tract (GIT) conditions, performed in accordance with the INFOGEST protocol, indicated that the optimised Ca-alginate/κC beads had a lower rate of release in the gastric phase and a much higher rate of survival and release in the intestinal phase than the control sample. The swelling behaviour of beads varied due to varying pH in both gastric and intestinal phases, and the κC in the optimised beads affected the swelling ratio significantly.

Abstract:Advances in perinatal care have improved the chances for survival of extremely low birthweight (<800 grams) and gestational age (<26 weeks) infants. A review of the world literature reveals that among regional populations, survival at 23 weeks' gestation ranges from 2 to 35%, at 24 weeks' gestation 17 to 62% and at 25 weeks' gestation 35 to 72%. These wide variations may be accounted for by differences in population descriptors, in the criteria used for starting or withdrawing treatment, in the reported duration of survival and differences in care. Major neonatal morbidity increases with decreasing gestational age and birthweight. At 23 weeks' gestation, chronic lung disease occurs in 57 to 86% of survivors, at 24 weeks in 33 to 89% and at 25 weeks' gestation in 16 to 71% of survivors. The rates of severe cerebral ultrasound abnormality range from 10 to 83% at 23 weeks' gestation, 9 to 64% at 24 weeks and 7 to 22% at 25 weeks' gestation Of 77 survivors at 23 weeks' gestation, 26 (34%) have severe disability (defined as subnormal cognitive function, cerebral palsy, blindness and/or deafness). At 24 weeks' gestation, the rates of severe neurodevelopmental disability range from 22 to 45%, and at 25 weeks' gestation 12 to 35%. When compared with children born prior to the 1990s, the rates of neurodevelopmental disability have, in general, remained unchanged. We conclude that, with current methods of care, the limits of viability have been reached. The continuing toll of major neonatal morbidity and neurodevelopmental handicap are of serious concern.

Abstract:Probiotics are used in food products, dietary supplements and pharmaceutical products because they may provide health-promoting effects in humans. To be efficacious, probiotics need to be viable at sufficient abundance within the large intestine. However, many commercial products containing probiotics suffer from a substantial loss of bacterial viability during shelf storage and during gastrointestinal transit. In this study, a probiotic (Lactobacillus salivarious Li01) was incorporated into either alginate or alginate-gelatin microgels. The morphology of the microgels was characterized by scanning electron microscopy, which indicated that they had a spherical shape and that almost all of the bacterial cells were encapsulated inside them. Probiotic viability was determined under aerobic conditions, heat treatment, and simulated gastrointestinal conditions. Encapsulation significantly enhanced the viability of the probiotic during aerobic storage. The microgels maintained their structures under simulated gastric conditions, but either eroded or swelled under simulated small intestine conditions. The alginate-gelatin microgels were the most effective at protecting the bacteria under simulated gastrointestinal conditions when compared to the alginate microgels and non-encapsulated bacteria. In conclusion, alginate-gelatin microgels have great potential for the protection and delivery of probiotics in food, supplement, and pharmaceutical products.

Abstract:The purpose of this study was to prepare sodium alginate (SA)/pectin (PE) hydrogel microspheres using the extrusion method to encapsulate Lactobacillus bulgaricus. Microscopic observation showed that the beads were spherical with a smooth and uniform surface. For microspheres with a diameter range of 140-156 μm, the encapsulation efficiency reached 85.67%. After simulating saliva, gastric juice, and intestinal juice, the activity of the microcapsules was estimated to be 5.78 × 104 log colony forming unit (CFU)/mL. These data show that the use of SA and PE encapsulated probiotics exhibit enhanced viability. In addition, double-layer beads containing probiotic microspheres and yogurt were prepared, and physical and chemical analysis was performed using scanning electron microscopy, Fourier-transform infrared spectroscopy, and differential scanning calorimetry. Texture and sensory property analysis revealed that the beads had good elasticity, chewiness, and high commercial value. Collectively, these findings indicate that SA and PE can be used for the encapsulation, protection, and gastrointestinal delivery of probiotics. Moreover, these microcapsules exhibit good stability in vitro and improve yogurt characteristics by increasing the survival rate of encapsulated probiotics, thus demonstrating their commercial application potential.

Abstract:Summary Alginate‐based encapsulation is an effective probiotics delivery system in the intestine. However, due to calcium ions and gastric juice damaging to cell wall/membrane integrity during freeze‐drying and gastrointestinal ingestion, lots of cell loss their viability. This study, taking Lactiplantibacillus plantarum grx16 as an example, indicated that co‐encapsulation of cryoprotectants and porous starch within calcium alginate capsules could combinedly improve viability of probiotics encapsulated. Compared with calcium alginate‐protectant capsule (CAC‐Pro) (50.67%), combination of porous starch increased the encapsulation rate of PS0.8‐CAC‐Pro capsules to 82.68%. Meanwhile, the death rate constants of PS0.8‐CAC‐Pro capsules decreased by 31.3% (‐20 °C), 13.5% (4 °C), and 7.5% (25 °C) respectively. The survival rate of PS0.8‐CAC‐Pro capsules was 81.94% after continuous simulated gastrointestinal digestion, which was significantly higher than that of calcium alginate capsules (69.46%) and CAC‐Pro capsules (76.92%). This study provided an effective approach for modifying alginate capsule capsules to reduce the loss of probiotics viability.

Automatic Extraction of Axioms for Planning

Abstract: Axioms can be used to model derived predicates in domain-independent planning models. Formulating models which use axioms can sometimes result in problems with much smaller search spaces than the original model. We propose a method for automatically extracting a particular class of axioms from standard STRIPS PDDL models. More specifically, we identify operators whose effects become irrelevant given some other operator, and generate axioms that capture this relationship. We show that this algorithm can be used to successfully extract axioms from standard IPC benchmark instances, and show that the extracted axioms can be used to significantly improve the performance of satisficing planners.

Abstract:Prebiotics-based encapsulation aids in improving the structure of microbeads and the survivability of probiotics. The current study focused on the exploration of a prebiotic-based encapsulation system (alginate-inulin) to improve the viability of probiotics under in vitro and carrier food. Probiotic ( L. acidophilus ) was encapsulated by the ionotropic gelation method. Microbeads with inulin inclusion were found to be compact and smooth with the highest encapsulation efficiency (98.87%) among the rest of the treatments. Alginate-inulin-based microbeads showed the highest count (8.41log CFU) as compared to other treatment as well free cells under simulated gastrointestinal conditions. Furthermore, alginate-inulin encapsulation maintained recommended (10 7 –10 8 log CFU/ml) probiotic viability in carrier food throughout the storage period. Probiotic encapsulation aids in controlling the post-acidification of the carrier product (yogurt). The results of this study indicated that the alginate-inulin-based encapsulation system has promising potential to ensure the therapeutic number of probiotics in vitro as well in carrier foods.

Abstract:Lactobacillus rhamnosus GG (LGG) and Lactobacillus acidophilus NCFM (LNCFM) were encapsulated in alginate microgel particles (microbeads) by a novel dual aerosols method. The encapsulated probiotics in microbead gel matrix were further stabilized in maltodextrin solids by either spray or freeze-drying to form probiotic microcapsule powders. The free cells of probiotics were also sprayed and freeze-dried in maltodextrin only without microgel encapsulation. After rehydration of microgel-encapsulated powder, gel particles regained their shape. There was no difference in the loss of viability between encapsulated and unencapsulated probiotics during spray drying or freeze-drying. For LNCFM, spray-dried bacteria with or without gel encapsulation exhibited less death (3.03 and 3.07 log CFU/g reduction, respectively) than those of freeze-dried bacteria (4.36 and 4.89 log CFU/g reduction, respectively) after 6 months storage at 4 °C. The same trend was also observed in spray-dried LGG without gel encapsulation which showed 5.87 log CFU/g reduction in viability after 6 months at 4 °C; however, freeze-dried LGG without gel encapsulation exhibited a rapid reduction in viability of 5.91 log CFU/g within just 2 months. Gel-encapsulated LGG which was freeze-dried exhibited less death (3.32 log CFU/g reduction) after 6 months at 4 °C. This work shows that spray drying results in improved subsequent probiotic survivability compared to freeze-drying and that alginate gel encapsulation can improve the survivability following freeze-drying in a probiotic-dependent manner.
