Abstract:Interaction forces of adhesive proteins employed by cyprid larvae of Amphibalanus amphitrite for temporary attachment during surface exploration in marine fouling were studied by AFM force spectroscopy using chemically modified, reactive colloidal probes. The proteins were covalently attached to the surfaces of the probes by incubation in the protein deposits (footprints) left behind at the surface by the cyprids. This covalent coupling enabled robust and reproducible probing of adhesion of the attachment proteins to model surfaces with variable hydrophilicity. Three model monolayer surfaces were designed and prepared that exhibited different wettabilities derived from variations in the monolayer chemical composition. The morphology and size of cyprid protein deposits was imaged by AFM. The deposits showed larger area of spreading on more hydrophobic surfaces, whereas the overall volume of the secreted proteins exhibited no significant variation. Notable difference in adhesion forces was found among the surfaces by force spectroscopy, with substantially higher values measured on the hydrophobic surface (21 ± 2 nN) than that measured on the more hydrophilic surface (7.2 ± 1 nN). The same surfaces were also tested in laboratory essays. Rather surprisingly, no significant differences were found in values of fractional cyprid settlement among the surfaces studied, indicating that variations of surface wettability and adhesion strength of settlement proteins may be insufficient to explain settlement trends.
Protein-mediated bioadhesion in marine organisms: A review

Abstract:Protein-mediated bioadhesion is one of the crucial physiological processes in marine organisms, by which they can firmly adhere to underwater substrates. Most marine adhesive organisms are biofoulers, causing negative effects on marine ecosystems and huge economic losses to aquaculture and maritime industries. Furthermore, adhesive proteins in these organisms are promising bionic candidates for high-performance artificial materials with great application value. In-depth understanding of the bioadhesion in marine ecosystems is of dual significance for resolving biofouling issue and developing marine bionic products. Here, we review the research progress of protein-mediated bioadhesion in marine organisms. The adhesion processes such as protein biosynthesis and secretion are similar among organisms, but the detailed features such as compositions, structures, and molecular functions of adhesive proteins are distinct. Hydroxylation, glycosylation, and phosphorylation are important post-translational modifications during the processes of adhesion. The contents of some amino acids such as glycine, tyrosine and cysteine involved in underwater adhesion are significantly higher, which is a sequence feature of barnacle cement and mussel foot proteins. The amyloid structures and conserved domains/motifs such as EGF and vWFA distributed in adhesive proteins are involved in the underwater adhesion. In addition, the oxidative cross-linking also plays an important role in marine bioadhesion. Overall, the unique and common features identified for the protein-mediated bioadhesion in diverse marine organisms here provide background information and essential reference for characterizing marine adhesive proteins and associated functional domains, formulating antifouling strategies, and developing novel biomimetic adhesives.

Abstract:Marine mussels are able to firmly affix on various wet surfaces by the overproduction of special mussel foot proteins (mfps). Abundant fundamental studies have been conducted to understand the molecular basis of mussel adhesion, where the catecholic amino acid, l-3,4-dihydroxyphenylalanine (DOPA) has been found to play the major role. These studies continue to inspire the engineering of novel adhesives and coatings with improved underwater performances. Despite the fact that the recent advances of adhesives and coatings inspired by mussel adhesive proteins have been intensively reviewed in literature, the fundamental biochemical and biophysical studies on the origin of the strong and versatile wet adhesion have not been fully covered. In this review, we show how the force measurements at the molecular level by surface force apparatus (SFA) and single molecule atomic force microscopy (AFM) can be used to reveal the direct link between DOPA and the wet adhesion strength of mussel proteins. We highlight a few important technical details that are critical to the successful experimental design. We also summarize many new insights going beyond DOPA adhesion, such as the surface environment and protein sequence dependent synergistic and cooperative binding. We also provide a perspective on a few uncharted but outstanding questions for future studies. A comprehensive understanding on mussel adhesion will be beneficial to the design of novel synthetic wet adhesives for various biomedical applications.
Deciphering protein-mediated underwater adhesion in an invasive biofouling ascidian: Discovery, validation, and functional mechanism of an interfacial protein

Abstract:Discovering macromolecules and understanding the associated mechanisms involved in underwater adhesion are essential for both studying the fundamental ecology of benthos in aquatic ecosystems and developing biomimetic adhesive materials in industries. Here, we employed quantitative proteomics to assess protein expression variations during the development of the distinct adhesive structure - stolon in the model fouling ascidian, Ciona robusta. We found 16 adhesive protein candidates with increased expression in the stolon, with ascidian adhesive protein 1 (AAP1) being particularly rich in adhesion-related signal peptides, amino acids, and functional domains. Western blot and immunolocalization analyses confirmed the prominent AAP1 signals in the mantle, tunic, stolon, and adhesive footprints, indicating the interfacial role of this protein. Surface coating and atomic force microscopy experiments verified AAP1's adhesion to diverse materials, likely through the specific electrostatic and hydrophobic amino acid interactions with various substrates. In addition, molecular docking calculations indicated the AAP1's potential for cross-linking via hydrogen bonds and salt bridges among Von Willebrand factor type A domains, enhancing its adhesion capability. Altogether, the newly discovered interfacial protein responsible for permanent underwater adhesion, along with the elucidated adhesion mechanisms, are expected to contribute to the development of biomimetic adhesive materials and anti-fouling strategies. STATEMENT OF SIGNIFICANCE: Discovering macromolecules and studying their associated mechanisms involved in underwater adhesion are essential for understanding the fundamental ecology of benthos in aquatic ecosystems and developing innovative bionic adhesive materials in various industries. Using multidisciplinary analytical methods, we identified an interfacial protein - Ascidian Adhesive Protein 1 (AAP1) from the model marine fouling ascidian, Ciona robusta. The interfacial functions of AAP1 are achieved by electrostatic and hydrophobic interactions, and the Von Willebrand factor type A domain-based cross-linking likely enhances AAP1's interfacial adhesion. The identification and validation of the interfacial functions of AAP1, combined with the elucidation of adhesion mechanisms, present a promising target for the development of biomimetic adhesive materials and the formulation of effective anti-fouling strategies.

Abstract:Mussels can strongly adhere to hydrophilic minerals in sea habitats by secreting adhesive proteins. The adhesion ability of these proteins is often attributed to the presence of Dopa derived from posttranslational modification of Tyr, whereas the contribution of structural feature is overlooked. It remains largely unknown how adhesive proteins overcome the surface-bound water layer to establish underwater adhesion. Here, we use molecular dynamics simulations to probe the conformations of adhesive protein Pvfp-5. and its salt-tolerant underwater adhesion on superhydrophilic mica. Dopa and positively charged basic residues form pairs, in this intrinsically disordered protein, and these residue pairs can lead to firm surface binding. Our simulations further suggest that the unmodified Tyr shows similar functions on surface adhesion by forming pairing structure with a positively charged residue. We confirm the presence of these residue pairs and verify the strong binding ability of unmodified proteins using nuclear magnetic resonance spectroscopy and lap shear tests.

Abstract:Calcium ion (Ca 2+ ) is involved in the protein-mediated larval adhesion of fouling ascidians, yet the effects of environmental Ca 2+ on larval adhesion remain largely unexplored. Here, the larvae of fouling ascidian C. robusta were exposed to different concentrations of Ca 2+ . Exposures to low-concentration (0 mM and 5 mM) and high-concentration (20 mM and 40 mM) Ca 2+ significantly decreased the adhesion rate of larvae, which was primarily attributed to the decreases in adhesive structure length and curvature. Changes in the expressions of genes encoding adhesion-, microvilli-, muscle contraction-, and collagen-related proteins provided a molecular-level explanation for adhesion rate reduction. Additionally, larvae likely prioritized their energy towards immunomodulation in response to Ca 2+ stresses, ultimately leading to adhesion reduction. These findings advance our understanding of the influencing mechanisms of environmental Ca 2+ on larval adhesion, which are expected to provide references for the development of precise antifouling strategies against ascidians and other fouling species.

Abstract:Developing adhesives that can function underwater remains a major challenge for bioengineering, yet many marine creatures, exemplified as mussels and barnacles, have evolved their unique proteinaceous adhesives for strong wet adhesion. The mechanisms underlying the strong adhesion of these natural adhesive proteins provide rich information for biomimetic efforts. Here, combining atomic force microscopy (AFM) imaging and force spectroscopy, we examine the effects of pH on the self-assembly and adhesive properties of cp19k, a key barnacle underwater adhesive protein. For the first time, we confirm that the bacterial recombinant Balanus albicostatus cp19k (rBalcp19k), which contains no 3,4-dihydroxyphenylalanine (DOPA) or any other amino acids with post-translational modifications, can self-assemble into aggregated nanofibers at acidic pHs. Under moderately acidic conditions, the adhesion strength of unassembled monomeric rBalcp19k on mica is only slightly lower than that of a commercially available mussel adhesive protein mixture, yet the adhesion ability of rBalcp19k monomers decreases significantly at increased pH. In contrast, upon preassembly at acidic and low-salinity conditions, rBalcp19k nanofibers keep stable in basic and high-salinity seawater and display much stronger adhesion and thus show resistance to its adverse impacts. Besides, we find that the adhesion ability of Balcp19k is not impaired when it is combined with an N-terminal Thioredoxin (Trx) tag, yet whether the self-assembly property will be disrupted is not determined. Collectively, the self-assembly-enhanced adhesion presents a previously unexplored mechanism for the strong wet adhesion of barnacle cement proteins and may lead to the design of barnacle-inspired adhesive materials.

Abstract:Water molecules pose a significant obstacle to conventional adhesive materials. Nevertheless, some marine organisms can secrete bioadhesives with remarkable adhesion properties. For instance, mussels resist sea waves using byssal threads, sandcastle worms secrete sandcastle glue to construct shelters, and barnacles adhere to various surfaces using their barnacle cement. This work initially elucidates the process of underwater adhesion and the microstructure of bioadhesives in these three exemplary marine organisms. The formation of bioadhesive microstructures is intimately related to the aquatic environment. Subsequently, the adhesion mechanisms employed by mussel byssal threads, sandcastle glue, and barnacle cement are demonstrated at the molecular level. The comprehension of adhesion mechanisms has promoted various biomimetic adhesive systems: DOPA-based biomimetic adhesives inspired by the chemical composition of mussel byssal proteins; polyelectrolyte hydrogels enlightened by sandcastle glue and phase transitions; and novel biomimetic adhesives derived from the multiple interactions and nanofiber-like structures within barnacle cement. Underwater biomimetic adhesion continues to encounter multifaceted challenges despite notable advancements. Hence, this work examines the current challenges confronting underwater biomimetic adhesion in the last part, which provides novel perspectives and directions for future research.

Quantifying cation-π interactions in marine adhesive proteins using single-molecule force spectroscopy

Abstract:In marine adhesives, cation-π interactions play an important role in their liquid-liquid phase separation process and underlying their strong interfacial bonding. However, it remains challenging to study the strength of cation-π interactions at the single-molecule level. Here, we engineer a recombinant chimeric polyprotein containing the mussel foot proteins-5 (MFP5) and a finger print domain GB1 to unambiguiously quantify the strength of cation-π interactions using atomic force microscopy (AFM)-based single-molecule force spectroscopy. Our results show that the formation of intermolecular cation-π interactions can be triggered at elevated salt concentrations, consistent with previous ensemble studies. Individual cation-π interaction ruptures at about 70 pN at a pulling speed of 1.6 μm s−1, comparable to the strength of other non-covalent interactions. The strength of cation-π interactions is weakly dependent on pH, which is in stark contrast with the hydrogen bonding and charge-charge interactions. Moreover, we find that the position of the cation-π bonds are formed randomly along the polyprotein chains. The propensity of forming long range cation-π interactions increases considerably when increasing the pH from 4 to 8, presumably due to the neutralization of the positive changes of MFP5. Our study directly quantifies the mechanical strength of cation-π bonds in the biological relavent settings and reveals key design parameters that may inspire the design of biomimetic strong underwater adhesives.

Abstract:Cell-matrix adhesion depends on the collective behaviours of clusters of receptor-ligand bonds called focal contacts between cell and extracellular matrix. While the behaviour of a single molecular bond is governed by statistical mechanics at the molecular scale, continuum mechanics should be valid at a larger scale. This paper presents an overview of a series of recent theoretical studies aimed at probing the basic mechanical principles of focal contacts in cell-matrix adhesion via stochastic-elastic models in which stochastic descriptions of molecular bonds and elastic descriptions of interfacial traction-separation are unified in a single modelling framework. The intention here is to illustrate these principles using simple analytical and numerical models. The aim of the discussions is to provide possible clues to the following questions: why does the size of focal adhesions (FAs) fall into a narrow range around the micrometre scale? How can cells sense and respond to substrates of varied stiffness via FAs? How do the magnitude and orientation of mechanical forces affect the binding dynamics of FAs? The effects of cluster size, cell-matrix elastic modulus, loading direction and cytoskeletal pretension on the lifetime of FA clusters have been investigated by theoretical arguments as well as Monte Carlo numerical simulations, with results showing that intermediate adhesion size, stiff substrate, cytoskeleton stiffening, low-angle pulling and moderate cytoskeletal pretension are factors that contribute to stable FAs. From a mechanistic point of view, these results provide possible explanations for a wide range of experimental observations and suggest multiple mechanisms by which cells can actively control adhesion and de-adhesion via cytoskeletal contractile machinery in response to mechanical properties of their surroundings.
Why Are Superhydrophobic Surfaces So Sticky? the Crucial Roles of Van Der Waals Force at Nanoscale in Wet Adhesion

Abstract:Recent experiments found that the superhydrophobic surfaces with surface nanostructures can be highly adhesive. In order to understand the underlying mechanisms of this novel property of the nanostructured surfaces, a theoretical model which takes into account the van der Weals interaction between the solid surface and the liquid was proposed. We showed that the role of the van der Waals interaction between the non-contacting surfaces of the solid and liquid becomes more dominant at small scale. The adhesive force caused by this non-contacting interaction via van der Weals force is inversely proportional to the size of the surface microstructures, which might explain why the nanostructured surfaces can be both superhydrophobic and highly adhesive. Our predictions on the adhesion force of these nanostructured surfaces are consistent with the experimental results. This work might provide useful guidelines for the design of novel devices for manipulating objects at small scale.
Influence of Surface Roughness on Wet Adhesion of Biomimetic Adhesive Pads with Planar Microstructures

Abstract:Biological adhesive pads of some reptiles and insects, such as tree frogs and grasshoppers, are covered with planar microstructures and have strong and stable adhesive ability on both wet and dry substrates. These adhesion forces do not mainly come from van-der-Waals force but wet adhesion. In this study, the influence of substrates' surface roughness on the wet adhesion of man-made adhesive pads inspired by tree frog toe pads is investigated experimentally. Biomimetic polydimethylsiloxane adhesive pads with planar hexagon microstructures with a microchannels width of 10 mm are fabricated by combining electroforming with soft lithography. Experiments of wet adhesive force between the pads and sandpaper slices with different average surface roughness are carried out at various preloads. Results show that the rougher surface leads to the decrement of wet adhesion force. It is also observed that if the microcosmic profile height of the substrates is near or less than the width of microchannels in the biomimetic adhesive pads, the microstructures and preloads can increase significantly the wet adhesive force, otherwise the microstructures and preloads do not contribute indistinctively. The experimental results can be explained by analysing the relation between the solid contact area and the area with a liquid bridge.

Abstract:Significance The need for bio-inspired wet adhesives has significantly increased in the past few decades (e.g., for dental and medical transplants, coronary artery coatings, cell encapsulants, etc.). However, the molecular basis behind catechol-facilitated adhesion to organic surfaces remains unclear, thus hindering synthesis and optimization of novel underwater adhesives. The present combined experimental and theoretical study reconciles bioadhesion measurements of novel catechol-containing peptides to self-assembled monolayers (SAMs) with all-atom molecular dynamics simulations, yielding a comprehensive framework that explicitly identifies the basis for underwater adhesion. Simulations and surface forces apparatus measurements agree with one another, and both approaches show strong peptide adhesion to hydrophobic SAMs, and weak peptide adhesion to hydrophilic SAMs, providing a starting point for the development of next-generation underwater glues.
Adaptive Adhesions of Barnacle-Inspired Adhesive Peptides

Abstract:The strategy of robust adhesion employed by barnacles renders them fascinating biomimetic candidates for developing novel wet adhesives. Particularly, barnacle cement protein 19k (cp19k) has been speculated to be the key adhesive protein establishing the priming layer in the initial barnacle cement construction. In this work, we systematically studied the sequence design rationale of cp19k by designing adhesive peptides inspired by the low-complexity STGA-rich and the charged segments of cp19k. Combining structure analysis and the adhesion performance test, we found that cp19k-inspired adhesive peptides possess excellent disparate adhesion strategies for both hydrophilic mica and hydrophobic self-assembled monolayer surfaces. Specifically, the low-complexity STGA-rich segment offers great structure flexibility for surface adhesion, while the hydrophobic and charged residues can contribute to the adhesion of the peptides on hydrophobic and charged surfaces. The adaptive adhesion strategy identified in this work broadens our understanding of barnacle adhesion mechanisms and offers valuable insights for designing advanced wet adhesives with exceptional performance on various types of surfaces.
Interferon alpha treatment and retreatment of hepatitis B e antigen-negative chronic hepatitis B.

Abstract:BACKGROUND AND AIMS In hepatitis B e antigen (HBeAg)-negative chronic hepatitis B (CHBe-), interferon (IFN) achieves very low biochemical sustained response rates. No information exists on retreatment. METHODS Two hundred sixteen CHBe- patients treated for 5 or 12 months with 3 MU IFN-alpha2b thrice weekly and retreated (51 patients, 60 courses) because of no response or relapse were retrospectively analyzed. RESULTS After 7.0 years of median follow-up, 39 naive patients (18.1%) were still in biochemical and virologic remission after a single IFN course. Longer treatment and a biochemical response within 4 months were significant predictors, inversely related to relapse by multivariate analysis (relative hazard [RH], 0.611; 95% confidence interval [CI], 0.448-0.834 and RH, 0.290; 95% CI, 0.192-0.438, respectively). Retreatment resulted in 18.4% sustained response by intention-to-treat (18 of 98 patients). Patients with sustained response had persistently normal alanine aminotransferase levels, undetectable hepatitis B virus (HBV) DNA by molecular hybridization, and significant improvement of histologic grade, and 32% of them lost hepatitis B surface antigen. In sustained responders, serum HBV DNA was undetectable or very low at the end of treatment and at the end of follow-up (median 3934 and 903 copies/mL, respectively) by quantitative polymerase chain reaction. CONCLUSIONS IFN induced long-term biochemical and virologic remission in approximately 18% of naive or retreated patients with CHBe-. Sustained responders exhibited significant histologic improvement and a high rate of HBsAg loss.
Toward Bioinspired Wet Adhesives: Lessons from Assessing Surface Structures of the Suction Disc of Intertidal Clingfish

Abstract:The clingfish attaches to rough surfaces with considerable strength using an intricate suction disc, which displays complex surface geometries from structures called papillae. However, the exact role of these structures in adhesion is poorly understood. To investigate the relationship between papillae geometry and adhesive performance, we developed an image processing tool that analyzed the surface and structural complexity of papillae, which we then used to model hydrodynamic adhesion. Our tool allowed for the automated analysis of thousands of papillae in specimens across a range of body sizes. The results led us to identify spatial trends in papillae across the complex geometry of the suction disc and to establish fundamental structure-function relationships used in hydrodynamic adhesion. We found that the surface area of papillae changed within a suction disc and with fish size, but that the aspect ratios and channel width between papillae did not. Using a mathematical model, we found that the surface structures can adhere considerably when subjected to disturbances of moderate to high velocities. We concluded that a predominant role of the papillae is to leverage hydrodynamic adhesion and wet friction to reinforce the seal of the suction disc. Overall, the trends in papillae characteristics provided insights into bioinspired designs of surface microstructures for future applications in which adhesion is necessary to attach to diverse surfaces (in terrestrial or aquatic environments), even when subjected to disturbance forces of randomized directionality.
The physico-chemistry of adhesions of protein resistant and weak polyelectrolyte brushes to cells and tissues

Abstract:The non-specific adhesion of polymers and soft tissues is of great interest to the field of biomedical engineering, as it will shed light on some of the processes that regulate interactions between scaffolds, implants and nanoparticles with surrounding tissues after implantation or delivery. In order to promote adhesion to soft tissues, a greater understanding of the relationship between polymer chemistry and nanoscale adhesion mechanisms is required. In this work, we grew poly(dimethylaminoethyl methacrylate) (PDMAEMA), poly(acrylic acid) (PAA) and poly(oligoethylene glycol methacrylate) (POEGMA) brushes from the surface of silica beads, and investigated their adhesion to a variety of substrates via colloidal probe-based atomic force microscopy (AFM). We first characterised adhesion to a range of substrates with defined surface chemistry (self-assembled monolayers (SAMs) with a range of hydrophilicities, charge and hydrogen bonding), before studying the adhesion of brushes to epithelial cell monolayers (primary keratinocytes and HaCaT cells) and soft tissues (porcine epicardium and keratinized gingiva). Adhesion assays to SAMs reveal the complex balance of interactions (electrostatic, van der Waals interactions and hydrogen bonding) regulating the adhesion of weak polyelectrolyte brushes. This resulted in particularly strong adhesion of PAA brushes to a wide range of surface chemistries. In turn, colloidal probe microscopy on cell monolayers highlighted the importance of the glycocalyx in regulating non-specific adhesions. This was also reflected by the adhesive properties of soft tissues, in combination with their mechanical properties. Overall, this work clearly demonstrates the complex nature of interactions between polymeric biomaterials and biological samples and highlights the need for relatively elaborate models to predict these interactions.
Insight into the adhesion propensities of extracellular polymeric substances (EPS) on the abiotic surface using XDLVO theory

Abstract:The adhesion of microorganisms is ubiquitous and plays a central role in a wide spectrum of problems, including biomedical study, industrial application and environmental microbiology. To know more about mechanisms underlying microalgal early-stage adhesion, the soluble EPS (sEPS) and the bound EPS (bEPS) of Chlorella vulgaris were extracted and characterized for their adhesion behaviors on the hydrophilic polymethyl methacrylate (PMMA) and the hydrophobic polytetrafluoroethylene (PTFE) surfaces in this work. It was found that the sEPS and the bEPS were dominated by hydrophilic fractions (57.6%) and hydrophobic fractions (49.6%) respectively. As a result, the bEPS always showed a higher adhesion propensity than the sEPS, and its adhesion on PTFE was more significant than that on PMMA. According to the extended Derjaguin-Landau-Verwey-Overbeek (XDLVO) theory, the adhesion behavior was dominated by the Lewis acid-base interfacial free energy (∆GmwfAB) of algal EPS. The bEPS could promote adhesive propensity of both two materials, whereas the sEPS would repulse the hydrophilic surfaces. Additionally, the dissolved organic carbon (DOC) was shown to be a more suitable index for the description of EPS adhesion. This work presents evidence on the mechanisms of the EPS-mediated adhesion process in algal biofilm formation.

Abstract:Antifouling properties of materials play crucial roles in many important applications such as biomedical implants, marine antifouling coatings, biosensing, and membranes for separation. Poly(ethylene glycol) (or PEG) containing polymers and zwitterionic polymers have been shown to be excellent antifouling materials. It is believed that their outstanding antifouling activity comes from their strong surface hydration. On the other hand, it is difficult to develop underwater glues, although adhesives with strong adhesion in a dry environment are widely available. This is related to dehydration, which is important for adhesion for many cases while water is the enemy of adhesion. In this research, we applied sum frequency generation (SFG) vibrational spectroscopy to investigate buried interfaces between mussel adhesive plaques and a variety of materials including antifouling polymers and control samples, supplemented by studies on marine animal (mussel) behavior and adhesion measurements. It was found that PEG containing polymers and zwitterionic polymers have very strong surface hydration in an aqueous environment, which is the key for their excellent antifouling performance. Because of the strong surface hydration, mussels do not settle on these surfaces even after binding to the surfaces with rubber bands. For control samples, SFG results indicate that their surface hydration is much weaker, and therefore mussels can generate adhesives to displace water to cause dehydration at the interface. Because of the dehydration, mussels can foul on the surfaces of these control materials. Our experiments also showed that if mussels were forced to deposit adhesives onto the PEG containing polymers and zwitterionic polymers, interfacial dehydration did not occur. However, even with the strong interfacial hydration, strong adhesion between mussel adhesives and antifouling polymer surfaces was detected, showing that under certain circumstances, interfacial water could enhance the interfacial bio-adhesion.
