Abstract:For over forty years, a sugar of rare configuration known as trehalose (two molecules of glucose linked at their 1-carbons), has been recognised for more than just its roles as a storage compound. The ability of trehalose to protect an extensive range of biological materials, for instance cell lines, tissues, proteins and DNA, has sparked considerable interest in the biotechnology and pharmaceutical industries. Currently, trehalose is now being investigated as a promising therapeutic candidate for human use, as it has shown potential to reduce disease severity in various experimental models. Despite its diverse biological effects, the precise mechanism underlying this observation remain unclear. Therefore, this review delves into the significance of trehalose biosynthesis pathway in the development of novel drug, investigates the inhibitors of trehalose synthesis and evaluates the binding efficiency of T6P with TPS1. Additionally, it also emphasizes the knowledge about the protective effect of trehalose on modulation of autophagy, combating viral infections, addressing the conditions like cancer and neurodegenerative diseases based on the recent advancement. Furthermore, review also highlight the trehalose's emerging role as a surfactant in delivering monoclonal antibodies that will further broadening its potential application in biomedicines.

Abstract:Trehalose is a naturally occurring, non-reducing saccharide widely distributed in nature. Over the years, research on trehalose has revealed that this initially thought simple storage molecule is a multifunctional and multitasking compound protecting cells against various stress factors. This review presents data on the role of trehalose in maintaining cellular homeostasis under stress conditions and in the virulence of bacteria and fungi. Numerous studies have demonstrated that trehalose acts in the cell as an osmoprotectant, chemical chaperone, free radical scavenger, carbon source, virulence factor, and metabolic regulator. The increasingly researched medical and therapeutic applications of trehalose are also discussed.

Abstract:Trehalose is a natural, non-reducing disaccharide synthesized in some bacteria, fungi, plants, and insects. Due to its advantageous physical and chemical properties, trehalose can stabilize proteins and membranes, and protect cells from desiccation, heating, and freezing. Vertebrates do not synthesize trehalose, but the beneficial effects of trehalose have been demonstrated in numerous diseases as it eliminates aggregates, misfolded proteins, and damaged organelles, and reduces hyperinflammation and oxidative stress. Trehalose induces autophagy through nuclear translocation and activation of transcription factor EB (TFEB) in an mTOR-independent manner, but increases the expression of SQSTM1/p62 and has antioxidant properties in an autophagy-independent manner. Furthermore, trehalose induces apoptosis in tumor cells by increasing membrane fluidity through the activation of caspase 3, 6 and the JNK (c-Jun N-terminal kinase) pathway. Overall, in this review, previous knowledge on the therapeutic potential of trehalose in various diseases such as dry eye syndrome, neurodegenerative diseases, and tumors, was summarized, focusing on the underlying molecular mechanisms.

Abstract:Cryopreservation is crucial to fields including immune and stem cell therapies, reproductive technology, blood banking, regenerative medicine and across all biotechnology. During cryopreservation, cryoprotectants are essential to protect cells from the damage caused by exposure to freezing temperatures. The most common penetrating cryoprotectants, such as DMSO and glycerol do not give full recovery and have a cytotoxicity limit on the concentration which can be applied. The non-reducing disaccharide trehalose has been widely explored and used to supplement these, inspired by its use in nature to aid survival at extreme temperatures and/or desiccation. However, trehalose has challenges to its use, particular its low membrane permeability, and how its protective role compares to other sugars. Here we review the application of trehalose and its reported benefit and seek to show where chemical tools can improve its function. In particular, we highlight emerging chemical methods to deliver (as cargo, or via selective permeation) into the intracellular space. This includes encapsulation, cell penetrating peptides or (selective) modification of hydroxyls on trehalose.

Abstract:Trehalose is a naturally occurring, non-reducing disaccharide comprising two covalently-linked glucose molecules. It possesses unique physiochemical properties, which account for multiple biological roles in a variety of prokaryotic and eukaryotic organisms. In the past few decades, intensive research on trehalose has uncovered its functions, and extended its uses as a sweetener and stabilizer in the food, medical, pharmaceutical, and cosmetic industries. Further, increased dietary trehalose consumption has sparked research on how trehalose affects the gut microbiome. In addition to its role as a dietary sugar, trehalose has gained attention for its ability to modulate glucose homeostasis, and potentially as a therapeutic agent for diabetes. Abstract:Some spoilage yeasts are able to develop resistance to commonly used weak-acid preservatives. We studied the trehalose metabolism and its regulation in Saccharomyces cerevisiae in response to propionic acid stress. We show interruption of trehalose synthetic pathway caused the mutant hypersensitive to the acid stress, while its overexpression conferred acid-tolerance to yeast. Interestingly, this acid-tolerance phenotype was largely independent of trehalose but relied on the trehalose synthetic pathway. We demonstrate trehalose metabolism played a vital role in regulation of glycolysis flux and Pi/ATP homeostasis in yeast during acid-adaptation, and the PKA and TOR signaling pathways were involved in regulating trehalose synthesis at transcriptional level. This work confirmed the regulatory function of trehalose metabolism and improved our understanding of molecular mechanism of acid-adaptation of yeast. By exemplifying trehalose metabolism interruption limited the growth of S. cerevisiae exposed to weak acids, and trehalose pathway overexpression conferring acid-resistance to Yarrowia lipolytica enhanced citric acid production, this work provides new insights into the development of efficient preservation strategies and robust organic acid producers.

Abstract:In recent years, trehalose, a natural disaccharide, has attracted growing attention because of the discovery of its potential to induce autophagy. Trehalose has also been demonstrated to preserve the protein's structural integrity and to limit the aggregation of pathologically misfolded proteins. Both of these properties have made trehalose a promising therapeutic candidate to target autophagy-related disorders and protein aggregation diseases. Unfortunately, trehalose has poor bioavailability due to its hydrophilic nature and susceptibility to enzymatic degradation. Recently, trehalose-bearing carriers, in which trehalose is incorporated either by chemical conjugation or physical entrapment, have emerged as an alternative option to free trehalose to improve its efficacy, particularly for the treatment of neurodegenerative diseases, atherosclerosis, nonalcoholic fatty liver disease (NAFLD), and cancers. In the current Perspective, we discuss all existing literature in this emerging field and try to identify key challenges for researchers intending to develop trehalose-bearing carriers to stimulate autophagy or inhibit protein aggregation.

Abstract:Trehalose is a natural non-reducing sugar that is found in the vast majority of organisms such as bacteria, yeasts, invertebrates and even in plants. Regarding its features, it is considered as a unique compound. It plays a key role as a carbon source in lower organisms and as an osmoprotectant or a stabilizing molecule in higher animals and plants. Although in plants it is present in a minor quantity, its levels rise upon exposure to abiotic stresses. Trehalose is believed to play a protective role against different abiotic stressful cues such as temperature extremes, salinity, desiccation. Moreover, it regulates water use efficiency and stomatal movement in most plants. Detectable endogenous trehalose levels are vital for sustaining growth under stressful cues. Exogenously applied trehalose in low amounts mitigates physiological and biochemical disorders induced by various abiotic stresses, delays leaf abscission and stimulates flowering in crops. External application of trehalose also up-regulates the stress responsive genes in plants exposed to environmental cues. The genetically modified plants with trehalose biosynthesis genes exhibit improved tolerance against stressful conditions. An increased level of trehalose has been observed in transgenic plants over-expressing genes of microbial trehalose biosynthesis. However, these transgenic plants display enhanced tolerance to heat, cold, salinity, and drought tolerance. Due to multiple bio-functions of this sugar, it has gained considerable ground in various fields. However, exogenous use of this bio-safe sugar would only be possible under field conditions upon adopting strategies of low-cost production of trehalose. In short, trehalose is a unique chemical that preserves vitality of plant life under harsh ecological conditions. Certainly, the new findings of this disaccharide will revolutionize a wide array of new avenues.

Abstract:The disaccharide trehalose plays a crucial role in multiple facets of the stress biology of yeasts and fungi. Here, we evaluate the properties, cellular and ecophysiological roles, metabolism, and stress-protection mechanisms of trehalose. We integrate disparate sources of knowledge across these topics, and bring new information about the mechanisms by which trehalose stabilises biomacromolecules and how trehalose metabolism is regulated thus giving rise to its diverse roles in fungi including stress protector, carbohydrate reserve, and regulatory/signaling molecule. We also present new findings about the effect of trehalose on microbial adaptation, complexity and spatio-temporal heterogeneity of microbial populations, and implications for industrial processes that apply fungi. Based on the elucidation of the structures of enzymes involved in trehalose synthesis, their catalytic mechanisms, and the regulation of trehalose synthesis, we discuss prospects for the development of more-efficient fungicides. Current humanitarian crises, such as overpopulation, global warming, malnutrition, immunocompromised conditions, and usage of immunosuppressant drugs, are making the incidence of human pathogens increases. Furthermore, fungal infections can be difficult to treat due to the conserved biochemistry between human and fungi cells. Serendipitously, however, trehalose is not synthesised by mammals, which makes trehalose synthesis an interesting target for the development of new therapies.

Abstract:Abstract Bacteria possess diverse metabolic and genetic processes, resulting in the inability of certain bacteria to degrade trehalose. However, some bacteria do have the capability to degrade trehalose, utilizing it as a carbon source, and for defense against environmental stress. Trehalose, a disaccharide, serves as a carbon source for many bacteria, including some that are vital for pathogens. The degradation of trehalose is carried out by enzymes like trehalase (EC 3.2.1.28) and trehalose phosphorylase (EC 2.4.1.64/2.4.1.231), which are classified under the glycoside hydrolase families GH37, GH15, and GH65. Numerous studies and reports have explored the physiological functions, recombinant expression, enzymatic characteristics, and potential applications of these enzymes. However, further research is still being conducted to understand their roles in bacteria. This review aims to provide a comprehensive summary of the current understanding of trehalose degradation pathways in various bacteria, focusing on three key areas: (i) identifying different trehalose-degrading enzymes in Gram-positive and Gram-negative bacteria, (ii) elucidating the mechanisms employed by trehalose-degrading enzymes belonging to the glycoside hydrolases GH37, GH15, and GH65, and (iii) discussing the potential applications of these enzymes in different sectors. Notably, this review emphasizes the bacterial trehalose-degrading enzymes, specifically trehalases (GH37, GH15, and GH65) and trehalose phosphorylases (GH65), in both Gram-positive and Gram-negative bacteria, an aspect that has not been highlighted before.

Abstract:The search for effective treatment for neurodegeneration implies attacking the multiple mechanisms of this pathology. Such properties were found in disaccharide trehalose, which shows therapeutic effects in models of many diseases and has been approved by the FDA for use in humans. Trehalose consists of two glucose residues bonded together by a flexible α-1-1'-glycosidic bond, giving it chaperone-like activity. Due to this, it prevents abnormal folding of aberrant proteins and has the properties of a cryo- and bioprotector. However, the main therapeutic effect is determined by the induction of mTOR-independent autophagy mediated by AMPK kinase as the main target. The result is a weakening of the accumulation of cytotoxic proteins and factors and an increase in cell viability. Autophagy activation depends on trehalose-induced lysosome and autophagosome biogenesis through activation of transcription factors TFEB and FOXO1. Trehalose has an anti-inflammatory effect closely related to the inhibition of oxidative stress. Trehalose-induced enhancement of endogenous antioxidant defense involves the regulator Nrf2. The review considers the neuroprotective effects of trehalose in models of major neurodegenerative diseases such as Parkinson's, Alzheimer's, Huntington's and others. Overall, trehalose shows high therapeutic potential in the treatment of experimental neurodegeneration and thus stimulating the study of its clinical application.

Abstract:<p>Trehalose is a natural disaccharide with a remarkable ability to stabilize biomolecules. In recent years, trehalose has received growing attention as a neuroprotective molecule and has been tested in experimental models for different neurodegenerative diseases. Although the underlying neuroprotective mechanism of trehalose's action is unclear, one of the most important hypotheses is autophagy induction. The chaperone‐like activity of trehalose and the ability to modulate inflammatory responses has also been reported. There is compelling evidence that the dysfunction of autophagy and aggregation of misfolded proteins contribute to the pathogenesis of Alzheimer's disease (AD) and other neurodegenerative disorders. Therefore, given the linking between trehalose and autophagy induction, it appears to be a promising therapy for AD. Herein, the published studies concerning the use of trehalose as a potential therapy for AD are summarized, providing a rationale for applying trehalose to reduce Alzheimer's pathology.</p>

Abstract:The increasing concern over sugar-related health issues has sparked research interest in seeking alternatives to sucrose. This review aims to provide a comprehensive overview of the properties of trehalulose, emphasizing its health benefits. The industrial prospects of trehalulose as sweetener and reducing agent, particularly in food and beverages pharmaceutical, and cosmeceutical sectors, are explored. Additionally, the review delves into the sources of trehalulose and the diverse organisms capable of producing trehalulose. The biosynthesis of this sugar primarily involves an enzyme-mediated process. Thus, these enzymes' properties, mechanisms, and the heterologous expression of genes associated with trehalulose production are explored. The strategies discussed in this review can be improved and applied to establish trehalulose bio-factories for efficient synthesis of trehalulose in the future. With further research and development, trehalulose holds promise as a valuable component across various industries.

Abstract:Plant abiotic stresses endanger crop production and food security to a growing degree under the present climate change scenario. This calls for effective measures to be deployed to increase the level of agricultural production to meet the needs of soaring world population. Application of osmo-protectants and soluble sugars were reported to counter abiotic stresses in many crop species. Trehalose (Tre) is one such non-reducing sugar found in bacteria and yeasts, where it serves as source of carbon, and in higher plants and animals, where it acts as osmo-protectant. Tre is involved in various physiological, biochemical and molecular mechanisms associated with plant growth, development and defense against drought, salinity, cold, heat, UV rays, nutrient deficiency and heavy metal stresses. It helps to maintain cellular integrity under stress by upgrading the antioxidant defense system. However, Tre amounts are lower than those needed to assure adequate plant stress tolerance. Interestingly, Tre supplementation up-regulates stress response genes and induces the accumulation of various osmolytes, including proline, glycine betaine and soluble sugars, which confer different kinds of stress tolerance. Alternatively, the development of transgenic plants with genes for Tre biosynthesis leads to appreciable tolerance against different stresses. However, some transgenic plants over-expressing Tre biosynthesis genes are adversely affected. This work aims to systematically review Tre's role as stress tolerance molecule and its crosstalk with other osmolytes under stress conditions, explaining mechanism of stress tolerance and pointing out areas for future research. It is evidenced that this compound owns a promising future as osmo-protectant in the coming years. The present review is intended as means to enrich the awareness on Tre potential benefits, in order to help the scientists as well as the practitioners to improve crop behavior and ultimate production under stress conditions.

Abstract:Cardiovascular diseases (CVDs) as environmental-influenced disorders, are a major concern and the leading cause of death worldwide. A range of therapeutic approaches has been proposed, including conventional and novel methods. Natural compounds offer a promising alternative for CVD treatment due to their ability to regulate molecular pathways with minimal adverse effects. Trehalose is natural compound and disaccharide with unique biological functions and cardio-protective properties. The cardio-protective effects of trehalose are generated through its ability to induce autophagy, which is mediated by the transcription factors TFEB and FOXO1. The stimulation of TFEB plays a significant role in regulating autophagy genes and autophagosome formation. Activation of FOXO1 through dephosphorylation of Foxo1 and blocking of p38 mitogen-activated protein kinase (p38 MAPK) also triggers autophagy dramatically. Trehalose has been shown to reduce CVD risk factors, including atherosclerosis, cardiac remodeling after a heart attack, cardiac dysfunction, high blood pressure, and stroke. It also reduces structural abnormalities of mitochondria, cytokine production, vascular inflammation, cardiomyocyte apoptosis, and pyroptosis. This review provides a molecular overview of trehalose's cardioprotective functions, including its mechanisms of autophagy and its potential to improve CVD symptoms based on clinical evidence.

Abstract:Trehalose is a stable disaccharide that consists of two glucose units linked primarily by an α,α-(1 → 1)-linkage, and it has been found in a wide variety of organisms. In these organisms, trehalose functions not only as a source of carbon energy but also as a protector against various stress conditions. In addition, this disaccharide is attractive for use in a wide range of applications due to its bioactivities. In trehalose metabolism, direct trehalose-hydrolyzing enzymes are known as trehalases, which have been reported for bacteria, archaea, and eukaryotes, and are classified into glycoside hydrolase 37 (GH37), GH65, and GH15 families according to the Carbohydrate-Active enZyme (CAZy) database. The catalytic domains (CDs) of these enzymes commonly share (α/α)<sub>6</sub>-barrel structures and have two amino acid residues, Asp and/or Glu, that function as catalytic residues in an inverting mechanism. In this review, I focus on diverse and common features of trehalases within different GH families and their contributions to microbial trehalose metabolism.

Abstract:Vinblastine and catharanthine, mainly derived from Catharanthus roseus, are highly effective anticancer agents. With the increasing demand for Catharanthus roseus as their raw material, the cultivation gradually spread from south to north, in which planting and breeding are seriously affected by the cold climate in high-latitude regions. This study explores the effect of trehalose on C. roseus under low-temperature stress. Based on low-temperature stress experiments on C. roseus seedlings as the experimental material, it was discovered that exogenous application of trehalose effectively increased plant chlorophyll content and antioxidant enzyme activity. l -phenylalanine, quercetin, apigenin, chrysin and genistein accumulated significantly under low-temperature stress, by 115.3%, 91.6%, 125.7% and 123.6%, respectively, compared to CK, whereas the accumulation decreased after exogenous trehalose application. In addition, alkaloids production in C. roseus was inhibited. To further understand the effect of exogenous trehalose on alkaloids content, dynamic expression changes in the 4 enzyme genes involved in alkaloids metabolism were analyzed. In summary, CrSTR and CrSGD expression was significantly upregulated under low-temperature, while the expression of CrTDC and CrLAMT was downregulated. Exogenous trehalose significantly increased the expression level of all 4 genes. These findings revealed the positive regulation of trehalose on alkaloid metabolism in C. roseus . Consequently, this study provides a theoretical basis for low-temperature stress resistance improvement in C. roseus , suggesting exogenous trehalose applications in industrial production to promote the planting and breeding process.
