Abstract:In temperate regions of the United States, female Anopheles mosquitoes respond to low temperatures and short photoperiods by entering an overwintering dormancy or diapause. Diapause in Anopheles results in reduced frequency of blood-feeding and reproductive arrest, indicating a period when pathogen transmission by these mosquitoes is unlikely. However, it is unclear precisely how late into the fall and how early in the spring these mosquitoes are biting, reproducing, and potentially transmitting pathogens. This is further complicated by the lack of clear markers of diapause in Anopheles (e.g., changes in egg follicle length). Our goal was to characterize the seasonal reproductive activity of female Anopheles in central Ohio, United States and evaluate egg follicle length as an indicator of Anopheles diapause. We used traditional mosquito traps and aspirators to collect Anopheles from urban woodlots and culverts, respectively, from late September 2021 through mid-May 2022 in central Ohio. By measuring their egg follicle length, reproductive status, and blood-feeding status, we found that egg follicle length is not a reliable indicator of Anopheles diapause. We also found that a small proportion of An. punctipennis (Say), An. perplexens (Ludlow), and An. quadrimaculatus (Say) continued to bite and reproduce into early November 2021 and that females of these species terminated reproductive dormancy and began biting by mid-March 2022. This period of reproductive activity extends beyond current mosquito surveillance and control in Ohio. Our findings suggest that within temperate regions of North America, Anopheles have the capacity to transmit pathogens throughout the spring, summer, and fall.

Abstract:In temperate regions, female Culex pipiens biotype pipiens mosquitoes undergo diapause in winter. Diapausing biotype pipiens mosquitoes are potentially important winter reservoirs for mosquito-borne viruses, such as West Nile virus (WNV), Usutu virus (USUV), and Sindbis virus (SINV). Mosquitoes in diapause have not taken a bloodmeal prior to winter. Therefore, they do not become infected by taking an infectious bloodmeal and as a consequence, vertical transmission is considered the primary mechanism of mosquito-borne virus overwintering. Prior to winter, biotype pipiens mosquitoes build up fat reserves, which they utilize throughout winter. Furthermore, earlier studies have indicated that larger body size is correlated with increased survival during winter. However, not much is known about lipid utilization and body size of wild biotype pipiens mosquitoes in diapause. Therefore, we performed monthly collections of diapausing biotype pipiens mosquitoes in two consecutive winters (2020/2021 and 2021/2022) in bunkers of the New Hollandic Waterline in the Netherlands. Every month, we checked the proportion of inseminated and parous females via microscopy. In addition, we measured wing length as proxy for body size, and assessed total lipid content. Furthermore, we monitored indoor temperature in the overwintering locations. We show that the overwintering sites in our study provide relatively stable environments, in which temperatures rarely drop below 0 °C. The vast majority of biotype pipiens females were inseminated (84.1 %) and nulliparous (97.5 %). We detected differences in body size between but not within the two years of sampling. Additionally, we detected a difference in lipid content between the sampling years. We confirm that the vast majority of diapausing biotype pipiens females are inseminated and nulliparous. This indicates that they did not blood feed prior to winter, which underscores the likeliness of vertical transmission being the primary mechanism behind virus overwintering. The detected difference in body size between years can most likely be attributed to differences in summer conditions the mosquitoes were exposed to as larvae, although this needs confirmation. The difference in lipid depletion could not be explained by differences in climatic conditions. To shed more light on the links between climatic conditions, body size, lipid depletion and the consequences for mosquito population dynamics and arbovirus transmission, future experimental work, for example by arbovirus exposure followed by artificially induced diapause, is desired.

Abstract:Usutu virus (USUV), which is closely related to West Nile virus (WNV), sharing a similar ecology and transmission cycle, was first reported in the UK in the southeast of England in 2020. Both USUV and WNV are emerging zoonotic viruses hosted by wild birds. The 2020 finding of USUV in England raised awareness of this virus and highlighted the importance of understanding the seasonality of Culex pipiens sensu lato ( Cx. pipiens s.l.), the main enzootic vector of these viruses. Zoos are prime locations for trapping mosquitoes because of their infrastructure, security, and range of vertebrate hosts and aquatic habitats.

Abstract:Winter survival consists a major component of insect vectors life history in temperate environments that is directly related with early and later population growth next season with major consequences in the epidemiology of vectored diseases. The common European mosquito Culex pipiens is a major vector of the West Nile Virus (WNV) in Europe, including Greece. West Nile Virus outbreaks are frequently reported in Greece over the last 2 decades and Thessaly, Central Greece, is included in the affected areas. Here we report on overwintering trials conducted in three regions of Thessaly to investigate the overwintering dynamics of the subspecies of the Cx. pipiens complex, Cx. pipiens f. pipiens. Two overwintering experiments regarding adults of Cx. pipiens f. pipiens carried out in two coastal areas of Thessaly (Nea Anchialos and Volos) and an inland area (Kalamaki). Results demonstrated the successful overwintering of Cx. pipiens f. pipiens females, as well as the failure of males to survive in all three regions considered. Successful overwintering females were capable of initiating egg laying following a blood meal in spring onsetting the first summer generation. Nonetheless, mortality patterns differ between the coastal and the inland area as well as among different cohorts of adults.

Light Pollution Disrupts Seasonal Differences in the Daily Activity and Metabolic Profiles of the Northern House Mosquito, Culex pipiens

Abstract:The Northern House mosquito, Culex pipiens, is an important disease vector, and females are capable of surviving the winter in a state of overwintering diapause. This species' diapause response has been extensively studied, and recent evidence suggests that the circadian clock is involved in measuring seasonal changes in daylength to initiate the diapause response. However, differences in the circadian activity of diapausing and non-diapausing Cx. pipiens have not been thoroughly investigated. Additionally, recent findings indicate that artificial light at night (ALAN) can disrupt mosquito diapause, potentially prolonging the mosquito biting season. We compared the circadian locomotor activity of mosquitoes reared in diapause-averting, long-day conditions and diapause-inducing, short-day conditions with and without ALAN to elucidate the interplay between circadian activity, diapause, and light pollution. We also uncovered metabolic differences between mosquitoes reared under diapausing and non-diapausing photoperiods with and without ALAN by measuring the concentration of protein, fructose, glycogen, water-soluble carbohydrates, and lipids. We found that ALAN exposure altered several diapause-associated phenotypes including slightly, but not significantly, increasing activity levels in short day-reared mosquitoes; and preventing some short day-reared mosquitoes from accumulating lipids. ALAN also significantly reduced glycogen and water-soluble carbohydrate levels in long day-reared mosquitoes. Based on our findings, light pollution may decrease insect fitness by perturbing metabolism, and may also impact several phenotypes associated with insect diapause, potentially extending the mosquito biting season and preventing insects in urban environments from overwintering successfully.

Abstract:Insects often exhibit irruptive population dynamics determined by environmental conditions. We examine if populations of the Culex tarsalis mosquito, a West Nile virus (WNV) vector, fluctuate synchronously over broad spatial extents and multiple timescales and whether climate drives synchrony in Cx. tarsalis , especially at annual timescales, due to the synchronous influence of temperature, precipitation, and/or humidity. We leveraged mosquito collections across 9 National Ecological Observatory Network (NEON) sites distributed in the interior West and Great Plains region USA over a 45-month period, and associated gridMET climate data. We utilized wavelet phasor mean fields and wavelet linear models to quantify spatial synchrony for mosquitoes and climate and to calculate the importance of climate in explaining Cx. tarsalis synchrony. We also tested whether the strength of spatial synchrony may vary directionally across years. We found significant annual synchrony in Cx. tarsalis , and short-term synchrony during a single period in 2018. Mean minimum temperature was a significant predictor of annual Cx. tarsalis spatial synchrony, and we found a marginally significant decrease in annual Cx. tarsalis synchrony. Significant Cx. tarsalis synchrony during 2018 coincided with an anomalous increase in precipitation. This work provides a valuable step toward understanding broadscale synchrony in a WNV vector.

The effects of forced-egg retention on the blood-feeding behavior and reproductive potential of Culex pipiens (Diptera: Culicidae)

Abstract:High rates of West Nile virus (WNV) transmission to humans are associated with exceptionally hot and dry summers. This is paradoxical since the eggs of Culex vectors of WNV depend on the persistence of containers with water, which decline during droughts. We examined the effects of forced-egg retention on the reproductive success of female Culex pipiens as well as behavioral responses, such as likelihood of secondary blood meals. As controls we examined the effects of female age and delayed mating. We found that early mating is essential to achieve reproductive success and, consistent with an "all-or-none" ovipositing strategy, C. pipiens females are able to retain considerable reproductive potential while searching for oviposition sites. Specifically, although forced-egg retention resulted in significant decreases in fitness, the decline was moderate for 5 weeks and most can be accounted for by increases in female age. Consequently, no females took blood more than once per gonotrophic cycle, which eliminates the possibility that heightened vectorial capacity due to multiple blood-feedings increases WNV transmission during periods of drought. Instead, our findings suggest that during droughts populations of C. pipiens have time to locate the remaining water holes, which are associated with human populations and WNV-competent bird species.
Transcription profiling reveals tissue-specific metabolic pathways in the fat body and ovary of the diapausing mosquito Culex pipiens

Abstract:The northern house mosquito, Culex pipiens, employs diapause as an essential survival strategy during winter, inducing important phenotypic changes such as enhanced stress tolerance, lipid accumulation, and extended longevity. During diapause, the cessation of reproductive development represents another distinctive phenotypic change, underlining the need for adjusted modulation of gene expressions within the ovary. Although considerable advancements in screening gene expression profiles in diapausing and non-diapausing mosquitoes, there remains a gap in tissue-specific transcriptomic profiling that could elucidate the complicated formation of diverse diapause features in Cx. pipiens. Here, we filled this gap by utilizing RNA sequencing, providing a detailed examination of gene expression patterns in the fat body and ovary during diapause compared to non-diapause conditions. Functional annotation of upregulated genes identified associations with carbohydrate metabolism, stress tolerance, immunity, and epigenetic regulation. The validation of candidate genes using quantitative real-time PCR verified the differentially expressed genes identified in diapausing mosquitoes. Our findings contribute novel insights into potential regulators during diapause in Cx. pipiens, thereby opening possible avenues for developing innovative vector control strategies.
Effects of seasonality and developed land cover on Culex mosquito abundance and microbiome diversity

Abstract:The vectorial capacity of mosquitoes, which influences the dynamics of vector-borne disease transmission, is intricately linked to mosquito abundance and the composition and diversity of their associated microbiomes. However, the influence of environmental factors on mosquito populations and microbiome diversity remains underexplored. Here we examined the effects of seasonality and developed land cover on Culex mosquito abundance and bacterial diversity. Biweekly field sampling of female Culex mosquitoes was conducted using gravid and CDC light traps, spanning summer to autumn across varying developed land cover levels in two urban areas in Central Illinois. Mosquito abundance was assessed by the number of mosquitoes captured per trap night and compared across seasons and developed levels. The mean mosquito abundance for gravid and light traps was 12.96 ± 2.15 and 7.67 ± 1.44, respectively. Notably, higher levels of developed land cover exhibited higher Culex abundance than the low level for light traps, but no significant difference was found between summer and early autumn. In gravid traps, no significant differences were detected across seasons or developed levels. Microbial analysis of the mosquito microbiome revealed that Proteobacteria and Wolbachia , with a mean relative abundance of 80.77 and 52.66% respectively, were identified as the most dominant bacterial phylum and genus. Their relative abundance remained consistent across seasons and developed land cover levels, with negligible variations. Alpha diversity, as measured by observed species, Chao1, Shannon, and Simpson, showed slightly higher values in early-autumn compared to late-summer. A notable pattern of bacterial diversity, as indicated by all four diversity indexes, is evident across varying levels of land development. Significantly, high or intermediate developed levels consistently showed reduced alpha diversity when compared to the lower level. This underscores the pronounced impact of anthropogenic ecological disturbances in shaping mosquito microbiomes. Beta diversity analysis revealed no significant dissimilarities in bacterial community composition across seasons and developed levels, although some separation was noted among different levels of developed land cover. These findings highlight the significant role of environmental factors in shaping mosquito abundance and their associated microbiomes, with potential implications for the vectorial capacity in the transmission of vector-borne diseases.

Abstract:Mosquito-borne diseases represent a significant threat to human and animal health in the United States. Several viruses, including West Nile, Saint Louis encephalitis, and Eastern equine encephalitis are endemic. In humans, the disease is typically detected during the summer months, but not during the winter months. The ability of these viruses to reemerge year after year is still not fully understood, but typically involves persistence in a reservoir host or vector during periods of low transmission. Mosquito species are known to overwinter at different life stages (adults, larvae, or eggs) in manufactured or natural sites. Gopher tortoise burrows are known to serve as refuge for many vertebrate and invertebrate species in pine savannas. In this study, we surveyed the interior of gopher tortoise burrows for overwintering mosquitoes. We identified 4 species (Anopheles crucians s.l., Culex erraticus, Mansonia dyari, and Uranotaenia sapphirina). Cx. erraticus was the most abundant, and its presence and abundance increased in winter months, implying that this species utilized gopher tortoise burrows for overwintering. Bloodfed Cx. erraticus and An. crucians s.l. females were detected. While An. crucians s.l. fed exclusively on the white-tailed deer, Cx. erraticus had a more diverse host range but fed primarily on the gopher tortoise. Tortoises and other long-lived reptiles like the American alligator have been shown to sustain high viremia following West Nile virus (WNV) and Eastern equine encephalitis virus (EEEV) infection and therefore could play a role in the maintenance of these viruses. In addition, Cx. erraticus is naturally infected with WNV and is a known bridge vector for EEEV. As such, these overwintering sites may play a role in perpetuating over-winter arboviral activity in Florida.
Temperature and photoperiod differentially impact maternal phenotypes in diapause egg-laying Aedes albopictus mosquitoes

Abstract:Aedes albopictus (Skuse 1894) mosquitoes can transmit deadly arboviruses and are globally invasive due to their ability to survive in both tropical and temperate climates. Although adults cannot survive harsh winters, females are capable of anticipating seasonal change and producing overwintering diapause (DP) eggs that remain in a state of arrested development over the winter and hatch when favorable conditions return in the spring. While low temperatures can facilitate DP entry under short photoperiods, temperature signals alone are not sufficient to induce DP. To identify maternal phenotypes predictive of DP egg production in laboratory conditions, we characterized aspects of maternal physiology and behavior to identify those that correlate with DP egg production and changes in photoperiod, versus changes in temperature. Neither changes in temperature nor photoperiod impacted protein preference, blood meal consumption, or total number of eggs produced per female. Egg retention and oviposition timing were influenced by temperature, independent of DP egg production. However, females housed under short photoperiod conditions showed increased starvation resistance, despite showing similar levels of locomotor activity and internal stores of triacylglycerols, glucose, glycogen, and trehalose compared to females housed in long photoperiods. These results suggest that temperature and photoperiod differentially affect maternal phenotypes and identify starvation resistance as a maternal phenotype that is influenced by photoperiod and correlates with DP egg status. Aedes albopictus mosquitoes can survive in temperate climates because females are able to detect changes in temperature and day length in the autumn to produce "diapause" eggs that are provisioned with extra nutrients to survive harsh winters by remaining in a state of arrest and hatching in the spring. Although temperature and day length normally change in concert with each other throughout the year, studies have shown that day length is the most important cue used by female mosquitoes to initiate diapause egg production. In this study, the authors examined aspects of feeding and reproduction to determine which of these correlated with day length. Many aspects of reproduction were influenced by temperature; however, housing female mosquitoes under autumn-like short-day conditions led to increased starvation resistance. Surprisingly these females survive longer even though they do not appear to expend less energy compared to females housed in spring-like long-day conditions at the same temperature. This suggests that the mosquito mothers undergo changes in their metabolism that correlate with the production of DP eggs that can survive the winter. This new knowledge may lead to novel targets to disrupt seasonal reproduction in mosquitoes and limit the parts of the world where they can survive.

Abstract:Seasonal patterns of mosquito population density and their vectorial capacity constitute major elements to understand the epidemiology of mosquito-borne diseases. Using adult mosquito traps, we compared the population dynamics of major mosquito species (Culex pipiens, Aedes albopictus, Anopheles spp.) in an urban and a wetland rural area of Attica Greece. Pools of the captured Cx. pipiens were analyzed to determine infection rates of the West Nile virus (WNV) and the Usutu virus (USUV). The data provided were collected under the frame of the surveillance program carried out in two regional units (RUs) of the Attica region (East Attica and South Sector of Attica), during the period 2017–2018. The entomological surveillance of adult mosquitoes was performed on a weekly basis using a network of BG-sentinel traps (BGs), baited with CO2 and BG-Lure, in selected, fixed sampling sites. A total of 46,726 adult mosquitoes were collected, with larger variety and number of species in East Attica (n = 37,810), followed by the South Sector of Attica (n = 8916). The collected mosquitoes were morphologically identified to species level and evaluated for their public health importance. Collected Cx. pipiens adults were pooled and tested for West Nile virus (WNV) and Usutu virus (USUV) presence by implementation of a targeted molecular methodology (real-time PCR). A total of 366 mosquito pools were analyzed for WNV and USUV, respectively, and 38 (10.4%) positive samples were recorded for WNV, while no positive pool was detected for USUV. The majority of positive samples for WNV were detected in the East Attica region, followed by the South Sector of Attica, respectively. The findings of the current study highlight the WNV circulation in the region of Attica and the concomitant risk for the country, rendering mosquito surveillance actions and integrated mosquito management programs as imperative public health interventions.

Abstract:During the summers of 2017-2019, 60 human cases of Jamestown Canyon virus-associated disease were reported in the State of Wisconsin, U.S.A; by comparison, there were 28 cases in the 5 years prior. Jamestown Canyon virus (JCV, Peribunyaviridae: Orthobunyavirus) is a zoonotic, mosquito-borne virus that is endemic throughout North America. The proposed transmission cycle for JCV involves horizontal transmission by a variety of mammal-feeding mosquito species and deer hosts, and transseasonal maintenance by vertical transmission in Aedes mosquito species. Although some of the earliest work on JCV transmission and disease was done in Wisconsin (WI), little is known about the spectrum of mosquitoes that are currently involved in transmission and maintenance of JCV, which is key to inform the approach to control and prevent JCV transmission, and to understand why case numbers have increased dramatically in recent years. Therefore, we undertook an intensive surveillance effort in Sawyer and Washburn counties, WI between April and August of 2018 and 2019, in an area with a concentration of JCV human cases. Larval and adult stages of mosquitoes were surveyed using larval dippers and emergence traps, light traps, resting boxes, a Shannon-style trap, and backpack aspirator. In total, 14,949 mosquitoes were collected in 2018, and 28,056 in 2019; these specimens represent 26 species in 7 genera. Suspect vector species were tested for JCV by polymerase chain reaction (PCR); of 23 species that were tested, only Aedes provocans yielded JCV positive results. In 2018, a single pool of Ae. provocans tested positive. In 2019, with more focused early season surveillance, we detected JCV in 4 pools of adult mosquitoes, and one pool that consisted of lab-raised adults that were collected as larvae. Material from all of these PCR-positive samples also yielded infectious virus in cell culture. Overall, these data provide new insight into the seasonality and habitat preferences for 26 mosquito species in Northern WI, which will be useful to inform future surveillance efforts for JCV. The results underscore the importance of Ae. provocans as a vector species involved in transseasonal maintenance of JCV in this region.

Yearly seasonal outbreaks continue to persist, and the city and suburbs of Chicago, Illinois, is considered a "hot spot" for WNV activity. To interrupt WNV transmission, ground ultra-low volume (ULV) adulticide applications are regularly used to reduce Culex pipiens L. and Culex restuans Theobold (Diptera: Culicidae) abundance and infection. The real-world effectiveness of adulticide applications has not been comprehensively assessed, and prior studies, including our own investigation, have yielded inconclusive or conflicting results. Therefore, we expanded our prior work and evaluated the effects of 5 sequential weekly truck-mounted ULV adulticide applications in large residential areas in the northern suburbs of Chicago, Illinois, in 2019 and 2020. Each day, Cx. pipiens and Cx. restuans host-seeking and gravid mosquitoes were collected to assess abundance, age structure, and WNV infection rates. Adulticide applications resulted in significant reductions of both host-seeking and gravid abundance on the night of treatment. The reduction in host-seeking mosquitoes was followed by a reduction in gravid mosquitoes trapped 3 and 4 days after adulticide application and an increase in the proportion of nulliparous mosquitoes. WNV infection rates were significantly reduced in treatment sites as compared to untreated sites when infection rates were higher in 2020. This large-scale study provides comprehensive evidence that ground ULV adulticide applications are an effective tool in an integrated mosquito management program for combating WNV vectors and infection risk.
Climate, landscape, and life history jointly predict multidecadal community mosquito phenology

Abstract:Phenology of adult host-seeking female mosquitoes is a critical component for understanding potential for vector-borne pathogen maintenance and amplification in the natural environment. Despite this importance, long-term multi-species investigations of mosquito phenologies across environments and differing species' life history traits are rare. Here we leverage long-term mosquito control district monitoring data to characterize annual phenologies of 7 host-seeking female mosquito species over a 20-year time period in suburban Illinois, USA. We also assembled data on landscape context, categorized into low and medium development, climate variables including precipitation, temperature and humidity, and key life history traits, i.e. overwintering stage and Spring–Summer versus Summer–mid-Fallseason fliers. We then fit linear mixed models separately for adult onset, peak abundances, and flight termination with landscape, climate and trait variables as predictors with species as a random effect. Model results supported some expectations, including warmer spring temperatures leading to earlier onset, warmer temperatures and lower humidity leading to earlier peak abundances, and warmer and wetter fall conditions leading to later termination. However, we also found sometimes complex interactions and responses contrary to our predictions. For example, temperature had generally weak support on its own, impacting onset and peak abundance timing; rather temperature has interacting effects with humidity or precipitation. We also found higher spring precipitation, especially in low development contexts, generally delayed adult onset, counter to expectations. These results emphasize the need to consider how traits, landscape and climatic factors all interact to determine mosquito phenology, when planning management strategies for vector control and public health protection.

Abstract:We need to understand how zoonotic diseases make their way in urbanized areas, and one way is via gathering well-structured temporal and spatial accounts of zoonotic disease vectors. Due to its large biotic and abiotic heterogeneity, Mexico City provides favorable environmental conditions for the proliferation of mosquitoes including vector species of medical importance. Hereby we present the results of one year sampling (May 2023-April 2024) in eight contrasting locations in Mexico City. In total, 4,861 mosquitoes belonging to 11 species were captured, with Culex stigmatosoma and Culex restuans emerging as the most dominant, while Anopheles aztecus, Culex erythrothorax and Aedes trivittatus were the least abundant. Monthly fluctuations in the composition and abundance of species were observed at each site, highlighting cemeteries as the places with the highest mosquito density, making them key areas for monitoring. This research should serve local health authorities to guide mosquito control strategies, and two key candidates are Culex quinquefasciatus and Cx. restuans , particularly from June to November which is when their abundance reaches a peak.

Abstract:Background: Mosquitoes are major vectors of arboviruses and other vector-borne diseases, making them a significant public health concern worldwide. Mitigation of arboviral outbreaks relies largely on the use of insecticides, but the effectiveness of such responses is threatened by the evolution of insecticide resistance. Monitoring mosquito susceptibility to different insecticides is therefore vital for informed decisions regarding outbreak responses. In this study, we elucidate the patterns of resistance to two insecticide classes within the primary vectors of West Nile virus in the northeast and midwestern regions of the continental United States, Culex pipiens and Culex restuans. Methodology/Principal Findings: Egg collections were performed throughout Illinois from 2018-2020, and adults were tested for insecticide resistance to permethrin and malathion. Individuals from each sampling location were sequenced to determine the presence of kdr target-site mutations, and biochemical assays were performed to determine increases in detoxification enzymes and insensitive acetylcholinesterase. Results from the bottle assays indicate variable resistance rates in Illinois, however lowered mortality was found in most of the regions that were tested. The kdr mutation (L1014F) was present in 50% of Culex pipiens sequenced, and more prevalent in southern Illinois compared with northern and central (p < 0.001). Different mechanisms were predictive of resistance by species and insecticide, with permethrin resistance being affected by kdr-allele frequency and oxidase levels and malathion resistance by α- and β-esterases in Culex pipiens. For Culex restuans α-esterase and oxidase levels were predictive of permethrin resistance while β-esterase and insensitive acetylcholinesterase levels were predictive of malathion resistance. Conclusions/Significance: We documented variation in insecticide resistance levels that appear to be driven by population differences in kdr mutation rates and metabolic resistance mechanisms. The presence of different mechanisms in species and regions has implications for approaches to resistance management and highlights the need to implement and maintain insecticide resistance monitoring practices.

Abstract:Abstract Mosquito-borne disease remains a significant burden on global health. In the United States, the major threat posed by mosquitoes is transmission of arboviruses, including West Nile virus by mosquitoes of the Culex genus. Virus metagenomic analysis of mosquito small RNA using deep sequencing and advanced bioinformatic tools enables the rapid detection of viruses and other infecting organisms, both pathogenic and non-pathogenic to humans, without any precedent knowledge. In this study, we sequenced small RNA samples from over 60 pools of Culex mosquitoes from two major areas of Southern California from 2017 to 2019 to elucidate the virome and immune responses of Culex . Our results demonstrated that small RNAs not only allowed the detection of viruses but also revealed distinct patterns of viral infection based on location, Culex species, and time. We also identified miRNAs that are most likely involved in Culex immune responses to viruses and Wolbachia bacteria, and show the utility of using small RNA to detect antiviral immune pathways including piRNAs against some pathogens. Collectively, these findings show that deep sequencing of small RNA can be used for virus discovery and surveillance. One could also conceive that such work could be accomplished in various locations across the world and over time to better understand patterns of mosquito infection and immune response to many vector-borne diseases in field samples.
