Abstract:In this work, a high-performance conjugated microporous polymer (CMP) decorated with BiOBr (Tr(PhXOD) 3 -CMP/BiOBr) is synthesized to application in construction of ultrasensitive photoelectrochemical (PEC) biosensor for sensing miRNA-122, by firstly coupling with efficient clip toehold-mediated allosteric bicycle strand displacement (ABSD). Notably, the Tr(PhXOD) 3 -CMP/BiOBr not only owns self-enhanced D-A-D structure that extremely shortens migration distance of photo-generated electron, but also forms Z-type heterostructure for accelerating electron-hole separation, thereby significantly enhancing the photocurrent with 10-fold higher than commonly used methods. Meanwhile, the clip toehold-mediated ABSD based on ternary linkage structure transformation avoids the attrition of invading strand, endowing the conservation of high concentration for undergoing rapid reaction with high-efficiency DNA amplification, which dramatically improves reaction time and superior target conversion. The experimental results indicate that proposed PEC biosensor had a high sensitivity to miRNA-122 with a detection limit of 0.49 fM, which provides a newly organic/inorganic photosensitive nanomaterials and efficient DNA strand displacement in bioanalytical and early clinical disease diagnosis.

Laboratory Experiments for Wave-Driven Sand Transport Prediction

Abstract:Since the first NCK-Days 20 years ago, a significant body of large-scale laboratory experiments on wave-driven sand transport has been conducted in oscillatory flow tunnels and large wave flumes. The experiments have yielded measures of net sand transport rates for a wide range of flow and sand conditions and have provided insights that have informed the development of practical predictors for sand transport in oscillatory flows and under waves. An overview of these experiments and a commentary on some of the important insights and quantitative results is presented. Particular attention is given to unsteady aspects of the sand flux for ripple regime and fine sand sheet-flow conditions, the role of flow acceleration on bed shear stress and net transport, and the differences in ne transport and transport processes occurring in tunnel oscillatory flows and occurring under progressive surface waves.
Long-lived Charge Carriers in Mn-doped CdS Quantum Dots for Photoelectrochemical Cytosensing.

Abstract:Photoelectrochemical (PEC) biosensing with semiconductor quantum dots (QDs) has received great attention because it integrates the advantages of both photo-excitation and electrochemical detection. During the photon-to-electricity conversion in PEC processes, electron-hole (charge) separation competes with electron-hole recombination, and the net effect essentially determines the performance of PEC biosensors. Herein, we propose a new approach for slowing down electron-hole recombination to increase charge separation efficiency for PEC biosensor development. Through doping with Mn(2+) , a pair of d bands ((4) T1 and (6) A1 ) is inserted between the conduction and valence bands of CdS QDs, which alters the electron-hole separation and recombination dynamics, allowing the generation of long-lived charge carriers with ms-scale lifetime that decay about 10(4) -10(5) -fold more slowly than in the case of undoped QDs. Photocurrent tests indicated that Mn(2+) doping resulted in an approximately 80 % increase in photocurrent generation compared with undoped CdS QDs. For application, the Mn-doped CdS QDs were coated on the surface of a glassy carbon electrode and functionalized with a cell surface carbohydrate-specific ligand (3-aminophenylboronic acid). In this way, a sensitive cytosensor for K562 leukemia cells was constructed. Moreover, the sugar-specific binding property of 3-aminophenylboronic acid allowed the electrode to serve as a switch for the capture and release of cells. This has been further explored with a view to developing a reusable PEC cytosensing platform.

Abstract:Development of novel photoactive materials is of great significance for improving the analytical performance of photoelectrochemical (PEC) biosensors. Herein, sensitive PEC biosensor was developed based on organic PM6:Y6 p-n heterojunction as photoactive matrix for circulating tumor cells (CTCs) detection. Y6, as organic acceptor that combines with the organic donor PM6 forms p-n heterojunction promoted separation of photo-generated charges with high charge transfer efficiency, resulting in the enhancement of photocurrent intensity. The detection of CTCs was achieved through the traditional "sandwich" protocol. Magnetic nanobeads (MNs) decorated with anti-epithelial cell attachment molecule antibody (anti-EpCAM) were used for capturing and separating CTCs, while the Au-aptamer probe and silver staining reaction were designed for dual signal amplification. The localized surface plasmon resonance (LSPR) of gold nanoparticles (Au NPs), and silver nanoparticles (Ag NPs) that generated by silver staining reaction improved photocurrent response of PEC sensor. This PEC biosensor was applied to detect MCF-7 and displayed an ultra-low detection limit of 9 cell mL-1 and linear range from 10 to 10000 cell mL-1. This work explored a path for the application of organic semiconductors as photoactive materials in PEC sensing, which may find broad applications for detecting varied analytes.

Abstract:Conjugated microporous polymers (CMPs), in which rigid building blocks form robust networks, are usually synthesized as insoluble and unprocessable powders. We developed a methodology using electropolymerization for the synthesis of thin CMP films. The thickness of these films is synthetically controllable, ranging from nanometers to micrometers, and they are obtained on substrates or as freestanding films. The CMP films combine a number of striking physical properties, including high porosity, extended π conjugation, facilitated exciton delocalization, and high-rate electron transfer. We explored the CMP films as versatile platforms for highly sensitive and label-free chemo- and biosensing of electron-rich and electron-poor arenes, metal ions, dopamine, and hypochloroic acid, featuring rapid response, excellent selectivity, and robust reusability.

Abstract:A sensitive photoelectrochemical (PEC) approach for Pb2+ sensing was achieved firstly with the coupling of G-quadruplex-based DNAzyme and biocatalytic precipitation (BCP) on a CdS quantum dots electrode. Pb2+ can induce the conformational change of K+-stabilized G-quadruplex-based DNAzyme, leading to the decrease of the catalytic activity to H2O2-mediated 4-chloro-1-naphthol oxidation (BCP reaction). From the respective BCP reaction that monitors the event of DNAzyme deactivation, a novel PEC Pb2+ sensor could be realized with a detection limit of 1.0×10−8M.

Abstract:Sensitive and accurate analysis of DNA is crucial to better understanding of DNA functions and early diagnosis of fatal disease. Herein, an enhanced photoelectrochemical (PEC) DNA biosensor was proposed based on dual-signal amplification via coupling inorganic-organic nanocomposites sensitization with λ-exonuclease (λ-Exo)-assisted target recycling. The short DNA sequence about chronic myelogenous leukemia (CML, type b3a2) was selected as target DNA (tDNA). ZnO nanoplates were deposited with CdS nanocrystals to form ZnO/CdS hetero-nanostructure, and it was used as PEC substrate for immobilizing hairpin DNA (hDNA). CdTe quantum dots (QDs) covalently linked with meso-tetra(4-carboxyphenyl)porphine (TCPP) to form CdTe/TCPP inorganic-organic nanocomposites, which were utilized as sensitization agents labeling at the terminal of probe DNA (pDNA). When the hDNA-modified sensing electrode was incubated with tDNA and λ-Exo, hDNA hybridized with tDNA, and meanwhile it could be recognized and cleaved by λ-Exo, resulting in the release of tDNA. The rest of nonhybridized hDNA would continuously hybridize with the released tDNA, cleave by λ-Exo, and set free the tDNA again. After λ-Exo-assisted tDNA recycling, more amounts of short DNA (sDNA) fragments coming from digestion of hDNA produced on the electrode and hybridized with CdTe/TCPP-labeled pDNA (pDNA-CdTe/TCPP conjugates). In this case, the sensitization of CdTe/TCPP inorganic-organic nanocomposites occurred, which evidently extend the absorption range and strengthened the absorption intensity of light energy, and accordingly the photocurrent signal significantly promoted. Through introducing the dual-signal amplification tactics, the developed PEC assay allowed a low calculated detection limit of 25.6 aM with a wide detection scope from 0.1 fM to 5 pM for sensitive and selective determination of tDNA.

Abstract:A novel enhanced photoelectrochemical (PEC) DNA biosensor, based on a compact heterojunction g-C3N4/MoS2 and co-sensitization effect with CdSe quantum dots (QDs), was first proposed for simple and accurate analysis of a short ssDNA. In this work, the g-C3N4/MoS2 was successfully synthesized and used as the electrode matrix material to construct PEC biosensor. 2D/2D heterojunction was formed between g-C3N4 and MoS2, which could promote the separation of photogenerated electron-hole pairs resulting in an enhanced photocurrent. In the presence of target DNA, CdSe QDs labeled reporter DNA was complementary pairing with target DNA which was specific recognized by capture DNA loading on self-assembled CdS QDs film, leading to close contact between CdSe QDs and g-C3N4/MoS2 modified electrode surface, thereby resulting in the enhanced photocurrent intensity due to the co-sensitization effect. Under the optimal operating conditions, the photoelectrochemical biosensor demonstrated favorable accuracy and could respond to 0.32 pM (S/N = 3) with a linear concentration range from 1.0 pM to 2.0 mu M. Moreover, the proposed PEC DNA biosensor exhibits high sensitivity, excellent specificity, acceptable reproducibility and accuracy, showing a promising potential in DNA bioanalysis and other relative fields. Perylene-Based Photoactive Material as a Double-Stranded DNA Intercalating Probe for Ultrasensitive Photoelectrochemical Biosensing

Abstract:Photoelectrochemical (PEC) sensing techniques have attracted considerable concerns because of the intrinsic merit of complete separation between the excitation light and responsive current but still remain a great challenge for further potential application. It is assigned to the scarcity of photoactive materials with narrow band gap, good biosafety, and high photon-to-electron conversion efficiency and unfavorable processing methods for photoactive materials on indium tin oxide. Herein, we employed a perylene-based polymer (PTC-NH2) with exceptional photoelectrical properties to develop a red-light-driven PEC sensor for ultrasensitive biosensing based on its superior electrostatic intercalation efficiency in double-stranded DNA to that in single-stranded DNA, with DNA adenine methyltransferase (Dam MTase) as the model target. The prepared PTC-NH2 was characterized by Fourier transform infrared spectroscopy, ultraviolet-visible spectroscopy, and PEC techniques, and the results demonstrated that PTC-NH2 rather than metal oxides/metal sulfides/C3N4/metal complexes enjoyed the prominent capacity of converting light to current. Benefiting from the unique PEC properties of PTC-NH2 and target-initiated hybridization chain reaction (HCR) signal amplification, ultrasensitive detection of Dam MTase was accessibly realized with the detection limit of 0.015 U/mL, which is lower than that of PEC, electrochemical, or fluorescent biosensors previously reported. Furthermore, the proposed PEC sensor has been also applied in screening Dam MTase activity inhibitors. Therefore, the perylene-based PEC sensor exhibits great potential in early accurate diagnosis of DNA methylation-related diseases.

Abstract:Photocathodic biosensor possesses excellent anti-interference capability in bioanalysis, which however suffers from high electron-hole recombination rate with low photocurrent. Herein, a high-performance inorganic organic P 3 60 @ZnO nanosphere with cascade energy band arrangement was synthesized as photoactive signal probe, which inherited the advantages of inorganic strong optical absorptivity and organic high mobility for photo-generated holes. Specifically, the well-matched band gap endowed not only the improved life for light generated carrier and promoted separation of electron-hole pairs, but also the expansion of charge-depletion layer, significantly improving the photoelectric conversion efficiency for acquiring an extremely high photocathodic signal that increased by 30 times compared with individual materials. Accordingly, by integrating with the efficient amplification of DNA nanonet derived from clamped hybrid chain reaction (C-HCR), a sensitive P 3 60 @ZnO nanosphere based photocathodic biosensor was proposed for accurate detection of p53. The experimental results showed that the biosensor had a wide detection range from 0.1 fM to 10 nM and a low detection limit of 0.37 fM toward p53, offering a new avenue to construct sensitive PEC platform with superior anti-interference ability and hold a prospective application in early disease diagnosis and biological analysis.

Abstract:INTRODUCTION Health technology assessment (HTA) is a tool based on scientific evidence useful for making decisions about the introduction of technological advances in healthcare systems. However, there are multiple elements that influence the actual use of the results of HTA. These elements are related both to the characteristics of HTA and to the stakeholders involved -health professionals, politicians, and society. The aim of this article was to describe the main elements that influence the applicability of HTA in practice, as well as the use of HTA in clinical practice and health policy. METHODS A literature search was performed in PubMed. The references retrieved, as well as additional bibliography and gray literature dealing with different aspects of HTA practice identified from the references retrieved, were reviewed. The results are presented descriptively, with analysis of the studies grouped according to the various HTA-related topics. RESULTS Key elements that could be improved are standardization of the evaluation process, description of the organization's mission, prioritization of the technologies to be assessed, the clarity of the presentation of results, inclusion of values other than scientific evidence, such as social and cost-effectiveness values, and increased international coordination. CONCLUSIONS For many people, the rationale for HTA is its explicit role in decision making, despite its problems. The immediate future of HTA would seem to include making the assessment process explicit, and the inclusion of HTA in decision making. This entails prioritization of technologies, setting limits on resource allocation and interrelation with political processes.
Porous hollow carbon nanobubbles@ZnCdS multi-shelled dodecahedral cages with enhanced visible-light harvesting for ultrasensitive photoelectrochemical biosensors

Abstract:Herein, novel photoactive materials, MOF-derived porous hollow carbon nanobubbles@ZnCdS multi-shelled dodecahedral cages (C@ZnCdS MSDCs)， were synthesized via continuous chemical etching, sulfurization, cation-exchange and calcination strategies. Due to the synergistic effect between the porous shells and the carbon-layer coating， C@ZnCdS MSDCs displayed superior photoelectrochemical (PEC) performance. The synthesized C@ZnCdS MSDCs were assembled onto TiO2 modified ITO electrodes to form a type-II heterostructures. Then, Au nanoparticles (NPs) were deposited on the surface of ITO/TiO2/C@ZnCdS MSDCs. Benefiting from the unique structure and performance merits of photoactive materials, a label-free PEC sensing platform based on ITO/TiO2/C@ZnCdS MSDCs/Au was successfully constructed for CEA detection. Under optimal conditions, the PEC biosensor exhibited a wide linear range (0.00005-500 ng mL-1) and low detection limit (2.28 fg mL-1). The proposed PEC biosensor also showed good stability, specificity, reproducibility and acceptability in human serum. The prepared C@ZnCdS MSDCs would be a promising photoactive material for PEC biosensors. Most importantly, this work opens up new horizons for the application of MOFs-derived hollow carbon materials in sensing.
A versatile dendritical amplification photoelectric biosensing platform based on Bi2S3 nanorods and a perylene-based polymer for signal "on" and "off" double detection of DNA

Abstract:A novel versatile dendritical amplification photoelectric (PEC) biosensing platform using Bi2S3 nanorods and perylene-based polymer (PTC-NH2) as double signal probes is proposed for the detection of trace target DNA. Bi2S3 nanorods as efficient photoactive materials were firstly immobilized on the Au nanoparticle (NP) modified electrode and generated a high PEC signal. Exonuclease III (Exo III)-assisted target recycling generated a large number of DNA product chains (PC), PC hybridized with hairpin DNA (H1) on AuNPs/Bi2S3/ITO, and then triggered rolling circle amplification (RCA) and the hybridization chain reaction (HCR) to form the dendritic structure with abundant DNA duplexes. After large amounts of Mn(iii) meso-tetra(N-methyi-4-pyridyl)porphine pentachloride (MnPP) were embedded in the dendritic structure, they efficiently quenched the PEC signal, realizing the "signal off" detection of the target. In addition, the dendritic structure was also formed on AuNPs/ITO, and large amounts of PTC-NH2 molecules as the PEC probe were inserted into the dendritic structure, achieving a highly enhanced PEC current for the "signal on" detection of the target. This biosensing platform with double signals exhibits good analytical performance with wide linear ranges. This study develops a new DNA nanostructure-amplified versatile PEC sensing platform for bioanalysis, with promising potential for application in gene therapy and clinical analysis.
Semiconducting Organic-Inorganic Nanodots Heterojunctions: Platforms for General Photoelectrochemical Bioanalysis Application

Abstract:In this study, semiconducting organic polymer dots (Pdots) and inorganic quantum dots (Qdots) were first utilized to construct the organic-inorganic nanodots heterojunction for the photoelectrochemical (PEC) bioanalysis application. Specifically, n-type CdS Qdots, p-type CdTe Qdots, and tetraphenylporphyrin (TPP)-doped poly[(9,9-dioctylfluoreny1-2,7-diyl)-co(1,4-benzo-{2,1',3}-thiadazole)] (PFBT) Pdots were fabricated, and their energy levels, that is, their valence band (VB)/conduction band (CB) or lowest unoccupied molecular orbital (LUMO)/highest occupied molecular orbital (HOMO) values, were also determined. Then, these nanodots were integrated to construct four types of p-n and p-p organic-inorganic nanodots heterojunctions, that is, CdS Qdots/TPP-doped PFBT Pdots, TPP-doped PFBT Pdots/CdS Qdots, CdTe Qdots/TPP-doped PFBT Pdots, and TPP-doped PFBT Pdots/CdTe Qdots, on the transparent glass electrode. Upon light irradiation, four heterojunctions exhibited different PEC behaviors with some having prominent photocurrent enhancement. With the model molecule L-cysteine (L-cys) as target, the proposed PEC sensor exhibited good performances. In brief, this work presents the first semiconducting organic-inorganic nanodots heterojunction for PEC bioanalysis application, which could be easily used as a general platform for future PEC bioanalysis building. Besides, it is expected to inspire more interest in the design, development, and implementation of various organic-inorganic heterojunctions for advanced PEC bioanalysis in the future.

Abstract:Integrating the advantages of excellent anti-interference and high stability superior to photoanode, cathodic photoelectrochemical (PEC) bioanalysis is promising and competitive in precise monitoring targets in complex matrices. However, serious consideration of the photocathode is far behind the anodic one. Herein, we report a high quality MoS2 QDs-BiOI p-n heterojunctioned material and exemplify the feasibility of constructing cathodic PEC aptasensing platform. Intense visible light-harvesting, high photoelectric conversion efficiency and magnified photocurrent is observed, which origin is detailed explored by various spectroscopic and electrochemical study. Taking the tumor necrosis factor-alpha (TNF-α) as a model analyte and by coupling the simplest target-induced conformational change mechanism, high selectivity and ultrasensitive self-power cathodic PEC detection is exemplified. Stable and reproducible PEC responses are achieved in a wide linear range of 10 fg/mL to 0.1 ug/mL with an ultralow detection limit (5.2 fg/mL) surpassing most sensors reported so far. Monitoring TNF-α in biological system is realized with desired accuracy and satisfactory recovery.

Rationality and cost-effectiveness of diagnosis and treatment of group A streptococci in primary care patients with pharyngitis.

Abstract:Rationality and cost-effectiveness of several strategies for diagnosing and treating sore throat patients were evaluated in primary care. In an empirical material of 2016 patients, 3 different bacteriological methods (traditional culture, slide culture, and latex agglutination test) for the detection of group A beta-hemolytic streptococci (GAS) were compared. Sensitivity analyses were done for several variables, including GAS test performance, testing frequencies, medication rates, and accuracy of clinical evaluation. The amounts of unnecessary medication varied from 19 to 33%, and there were 2.2-8.9% of untreated GAS infections. Strategies based on bacteriology achieved more rational and economic results than treating no one, treating all patients, or using clinical evaluation without bacteriology. The cost-effectiveness of the latex agglutination test depended on the sensitivity of the test. The results were further sensitive to sick leave lengths, cost of antibacterials, and clinical accuracy, but not to GAS test cost. Rapid tests for GAS detection can be recommended, if the sensitivity of the test in the actual working conditions is acceptable.
A duple nanozyme stimulating tandem catalysis assisted multiple signal inhibition strategy for photoelectrochemical bioanalysis

Abstract:A sandwich-type photoelectrochemical (PEC) immunosensor was developed for cytokeratin 19 fragment 21-1 (CYFRA21-1) ultrasensitive detection relying on a multiple signal inhibition strategy. In this work, Bi10O6S9 nanosheet arrays (BiOS) sensitized strontium titanate nanocubes (SrTiO3 NCs) was used as photoactive matrix. Bi10O6S9 with the large specific surface area and narrow band gap could improve the utilization of light to provide strong initial PEC signal. Besides, bovine serum albumin stabilized gold nanoparticles (BSA@Au NPs) anchored on Cu7S4 polyhedron composite (BSA-Au@Cu7S4 polyhedron) was utilized as an effective PEC signal regulator. As a duple nanozyme with the glucose oxidase (GOx)-like activity and the peroxidase-like activity, BSA@Au NPs induced intriguing tandem biocatalytic precipitation (BCP). On the one hand, the BCP could produce precipitations which obstructed the electron transfer between the photoelectrode and electron donor, leading a decrease of PEC signal; On the other hand, the competitive light absorption between BSA-Au@Cu7S4 polyhedron and photoactive matrix caused a further reduction of PEC signal. Generally, the PEC signal was quenched sharply taking advantage of multiple signal inhibition strategy including tandem BCP strategy, competitive absorption of light and steric hindrance, which improved the detection sensitivity of PEC immunosensor. The suggested PEC immunosensor displayed a good linearity in the range of 0.1 pg/mL -50 ng/mL for CYFRA21-1 detection with a low detection limit of 1.12 fg/mL (S/N = 3). The skillful strategy has infinite potential in biological samples analysis.

Abstract:Fabricating a self-powered photoelectrochemical (PEC) sensor with superior performances and cost-effective design is highly desirable but still remains challenges. Herein, we develop a self-powered PEC molecular imprinted (MIP) sensor integrating WO3/TiO2 photoanode and gold nanoparticles modified carbon intercalated MoS2 photocathode for highly sensitive and specific detection of chloroquine phosphate (CQP). By constructing a reasonable stepped energy band structure, the electrons are efficiently transferred driven by the Fermi energy level gap between photoanode and photocathode, which amplifies the cathodic photocurrent. Furthermore, the mechanism of electron transfer driven by the Fermi energy level gap in the self-powered sensor was demonstrated for the first time by density functional theory (DFT). The Molecular imprinted polymer was modified on the photocathode as recognition element, which avoids the interference of reducing substances in the samples and complex biometric reactions. Under optimal conditions, the proposed MIP sensor for CQP presents the detection limit of 0.11 μM with a broad linear range from 10−6 M to 10−3 M. This work presented an innovative design strategy for constructing self-powered sensor.

Abstract:In this work, we report a new concept of upconversion-powered photoelectrochemical (PEC) bioanalysis. The proof-of-concept involves a PEC bionanosystem comprising a NaYF4:Yb,Tm@NaYF4 upconversion nanoparticles (UCNPs) reporter, which is confined by DNA hybridization on a CdS quantum dots (QDs)/indium tin oxide (ITO) photoelectrode. The CdS QD-modified ITO electrode was powered by upconversion absorption together with energy transfer effect through UCNPs for a stable photocurrent generation. By measuring the photocurrent change, the target DNA could be detected in a specific and sensitive way with a wide linear range from 10 pM to 1 μM and a low detection limit of 0.1 pM. This work exploited the use of UCNPs as signal reporters and realized upconversion-powered PEC bioanalysis. Given the diversity of UCNPs, we believe it will offer a new perspective for the development of advanced upconversion-powered PEC bioanalysis.

A novel electrochemiluminescence biosensor based on the self-ECL emission of conjugated polymer dots for lead ion detection

Abstract:The poly[(9,9-dioctylfuorenyl-2,7-diyl)-alt-co-(1,4-benzo-{2,1′,3}-thiadiazole)] (PFBT) was carboxyl-functionalized to prepare polymer dots (C-PFBT Pdots), which served as a self-ECL emitter for producing an extraordinary ECL signal without any exogenous coreactants. The C-PFBT Pdots–modified electrode captured the substrate DNA and further hybridized with a ferrocene (Fc)-labeled DNA. The ECL emission of C-PFBT Pdots was quenched by Fc (a signal off state). After the DNAzyme was added, the DNAzyme-substrate hybrids were formed through hybridizing between DNAzyme and substrate and the Fc-labeled DNA was released. In the presence of target Pb2+, the DNAzyme-substrate hybrids could be specifically recognized and cleaved to release the DNAzyme and Pb2+. Ultimately, the released DNAzyme would further hybridize with the substrate for producing the DNAzyme-substrate hybrids and then were cleaved by the released Pb2+. As a result, the DNA walking machine was generated and the substantial Fc was away from C-PFBT Pdots to obtain a signal on state. Such a strategy achieved a sensitive detection of Pb2+ and the detection limit was as low as 0.17 pM. Moreover, making this ECL biosensor for an intracellular Pb2+ detecting, a convincing performance was achieved. The self-ECL emitter C-PFBT Pdots combining with the quencher Fc provided a new strategy and platform for constructing a coreactant-free ECL assay.
