Abstract:Commercially available cellulase cocktails frequently demonstrate high efficiency in hydrolyzing easily digestible pretreated biomass, which often lacks hemicellulose and/or lignin fractions. However, the challenge arises with enzymatic hydrolysis of mildly pretreated lignocellulosic biomasses, which contain cellulose, hemicellulose and lignin in high proportions. This study aimed to address this question by evaluating the supplementation of a commercial cellulolytic cocktail with accessory hemicellulases and two additives (H 2 O 2 and Tween® 80). Statistical optimization methods were employed to enhance the release of glucose and xylose from mildly pretreated sugarcane bagasse. The optimized supplement composition resulted in the production of 304 and 124 mg g −1 DM of glucose and xylose, respectively, significantly increasing glucose release by 84% and xylose release by 94% compared to using only the cellulolytic cocktail. This enhancement might be attributed to a coordinated hemicellulases action degrading hemicellulose, creating more space for cellulase activity, potentially boosted by the presence of H 2 O 2 and Tween® 80. However, the addition of different concentrations of H 2 O 2 in combination with hemicellulase and Tween® 80 did not result a significant difference on sugar release, which could be attributed to the limited range of concentrations studied (5 to 65 μM). The results obtained in this study using the mix of three supplements were also compared to the addition of only hemicellulase and only Tween® 80 to the cellulolytic cocktail. A significant increase in glucose release of 39 and 41%, respectively, was observed when using the optimized combination. For xylose, the increase was 38 and 41%, respectively. This study underscores the substantial potential in optimizing enzyme cocktails for the hydrolysis of mildly pretreated lignocellulosic biomass by using enzymes and additive combinations tailored to the specific biomass composition.

Abstract:This study evaluated innovative approaches for the enzymatic hydrolysis of lignocellulosic biomass. More specifically, assays were performed to evaluate the supplementation of the commercial cellulolytic cocktail Cellic® CTec2 (CC2) with LPMO (GcLPMO9B), H2O2, or cello-oligosaccharide dehydrogenase (CelDH) FgCelDH7C in order to boost the LPMO action and improve the saccharification efficiency of biomass into monosaccharides. The enzymatic hydrolysis was carried out using sugarcane bagasse pretreated by hydrodynamic cavitation-assisted oxidative process, 10% (w/w) solid loading, and 30 FPU CC2/g dry biomass. The results were compared in terms of sugars release and revealed an important influence of the supplementations at the initial 6 h of hydrolysis. While the addition of CelDH led to a steady increase in glucose production to reach 101.1 mg of glucose/g DM, accounting for the highest value achieved after 72 h of hydrolysis, boosting the LPMOs activity by the supplementation of pure LPMOs or the LPMO co-substrate H2O2 were also effective to improve the cellulose conversion, increasing the initial reaction rate of the hydrolysis. These results revealed that LPMOs play an important role on enzymatic hydrolysis of cellulose and boosting their action can help to improve the reaction rate and increase the hydrolysis yield. LPMOs-CelDH oxidative pairs represent a novel potent combination for use in the enzymatic hydrolysis of lignocellulose biomass. Finally, the strategies presented in this study are promising approaches for application in lignocellulosic biorefineries, especially using sugarcane bagasse as a feedstock.

Abstract:<p>The acquisition during biomass saccharification of elevated levels of fermentable sugars with lower cellulase concentration is central to ensuring an economically viable and industrially relevant hydrolytic process. Thus, using a new cellulase preparation (LT4) at low cellulase loading (2 mg protein/g dried substrate), this study assessed the possible boosting effect of integrating accessory enzymes and additives on high-solids hydrolysis of sugarcane bagasse via fed-batch feeding. Hydrolysis which commenced with initial 8% solids loading and subsequent substrate feeding of 4% solids at 6 h, 18 h, and 24 h respectively, proved optimal for the 20% high-solids saccharification producing 158 g/L total sugars and 83% glucose yield after 72 h with the combined optimized additives and accessory enzymes. The results obtained indicate that the integration of accessory enzymes and additives offers a benignant approach to minimizing the enzyme load and cost of high solids saccharification of lignocellulosic heteropolymers while also boosting enzyme hydrolytic performance.</p>

Abstract:In this study, an efficient coupling surfactant to ethylene glycol pretreatment (EG) and enzymatic hydrolysis of sugarcane bagasse (SCB) was proposed to enhance fermentable sugar production with lower pretreatment energy consumption and reduced enzyme loading. Under optimized conditions, 5 % Tween 80-assisted EG pretreatment of SCB achieved 80.5 % delignification while retaining cellulose (91.6 %) and hemicellulose (81.6 %) content. This led to an enhanced glucose yield of 81.3 %, compared to 65.1 % without Tween 80. The addition of Tween 80 to pretreatment modified residual lignin through etherification, reduced phenolic hydroxyl groups and enhanced hydrophilicity by 23.7 % and 9.4 %, respectively, compared to the lignin sample without Tween 80. Consequently, this modification alleviated non-productive adsorption between lignin and enzymes, improving substrate hydrolyzability. Moreover, when 4.5 % Triton-X 100 was introduced during the hydrolysis of Tween 80-assisted EG-pretreated substrates, a maximum glucose yield of 91.8 % and xylose yield of 92.6 % were achieved. Energy and enzyme cost analyses revealed a reduction of 35 % in pretreatment energy consumption and a 58.8 % decrease in enzyme costs, thanks to the synergistic action of surfactants in EG pretreatment and enzymatic hydrolysis. Overall, integrating surfactants into pretreatment and enzymatic hydrolysis holds promise for highly efficient conversion of SCB into fermentable sugars in enzyme-mediated lignocellulosic biorefineries.

Abstract:Bioproducts production using monomeric sugars derived from lignocellulosic biomass presents several challenges, such as to require a physicochemical pretreatment to improve its conversion yields. Hydrothermal lignocellulose pretreatment has several advantages and results in solid and liquid streams. The former is called hemicellulosic hydrolysate (HH), which contains inhibitory phenolic compounds and sugar degradation products that hinder microbial fermentation products from pentose sugars. Here, we developed and applied a novel enzyme process to detoxify HH. Initially, the design of experiments with different redox activities enzymes was carried out. The enzyme mixture containing the peroxidase (from Armoracia rusticana) together with superoxide dismutase (from Coptotermes gestroi) are the most effective to detoxify HH derived from sugarcane bagasse. Butanol fermentation by the bacteria Clostridium saccharoperbutylacetonicum and ethanol production by the yeast Scheffersomyces stipitis increased by 24.0× and 2.4×, respectively, relative to the untreated hemicellulosic hydrolysates. Detoxified HH was analyzed by chromatographic and spectrometric methods elucidating the mechanisms of phenolic compound modifications by enzymatic treatment. The enzyme mixture degraded and reduced the hydroxyphenyl- and feruloyl-derived units and polymerized the lignin fragments. This strategy uses biocatalysts under environmentally friendly conditions and could be applied in the fuel, food, and chemical industries.
Enhanced Production of Clean Fermentable Sugars by Acid Pretreatment and Enzymatic Saccharification of Sugarcane Bagasse

Abstract:Sugarcane bagasse (SCB), an agro-industrial byproduct generated by a sugar mill, holds a substantial carbohydrate content of around 70 wt.%, comprising cellulose and hemicellulose. Saccharification plays a pivotal role in the conversion of SCB into second-generation (2G)-ethanol and valuable compounds, which is significantly aided by thermochemical pretreatments. In this study, SCB underwent diluted sulfuric acid pretreatment (2% H2SO4, 80 rpm, 200 °C, 20 min), resulting in the removal of 77.3% of the xylan. The hemicellulosic hydrolysate was analyzed to identify the sugars and degraded products acting as microbial inhibitors. The acid hydrolysate showed a xylose yield of 68.0% (16.4 g/L) and a yield of 3.8 g/L of acetic acid. Afterward, the hemicellulosic hydrolysate was concentrated 2.37 times to obtain a xylose-rich stream (39.87 g/L). The sequential detoxification, employing calcium oxide and activated carbon, removed the inhibitory compounds, including acetic acid, while preserving the xylose at 38.10 g/L. The enzymatic saccharification of cellulignin at 5% and 10% of the total solids (TSs) yielded comparable reducing sugar (RS) yields of 47.3% (15.2 g/L) and 47.4% (30.4 g/L), respectively, after 96 h, employing a 10 FPU/g enzyme loading of Cellic® CTec3 (Novozymes Inc. Parana, Brazil). In summary, these findings outline an integrated green chemistry approach aimed at addressing the key challenges associated with pretreatment, concentration, detoxification, and enzymatic hydrolysis to produce fermentable sugars.

The Deconstruction of the Lignocellulolytic Structure of Sugarcane Bagasse by Laccases Improves the Production of H2 and Organic Acids

Abstract:The production of biofuels using sugarcane bagasse (SCB) as substrate can be considered an environmentally friendly approach, due to the possibility of combining energy production with the reuse of agroindustrial wastes. This study was undertaken to explore the applicability of a new extract with the enzymes (Lac mix ) isolated from Chaetomium cupreum for SCB pretreatment. Lac mix was more active at pH of 2.2 to 4 and 50 to 60 °C. Further, the individual and mutual effects of SCB concentration (6.6 to 23.4 g L − 1 ), enzyme concentration (0.066 to 0.234 U L − 1 ), and incubation time of the SCB with Lac mix (19 to 221 min) on SCB pretreatment were evaluated using a response surface methodology and central composite design. The optimized conditions were 23.4 g L − 1 SCB, 0.234 U mL − 1 laccases, and 2.44 h resulting in 547 ± 108 mg L − 1 of total sugars. This value agrees with the predicted value (455 ± 41 mg L − 1 ) by the statistical model. Through the SCB pretreated with Lac mix fermentation, 96.1% more H 2 and 22.5% more organic acids were observed compared to SCB without pretreatment. Therefore, laccases improve delignification, maximizing biomass fermentation for biofuel production.

Abstract:Hepatic glucuronidation of the asymmetrical natural bilirubin molecule results in formation of two different positional isomers, bilirubin C-8 monoglucuronide and bilirubin C-12 monoglucuronide. In view of the existence of multiple isoforms of UDPglucuronyltransferase, which is the microsomal enzyme system responsible for bilirubin esterification, we performed kinetic analysis of microsomal glucuronidation of bilirubin and a number of its structural congeners to determine whether synthesis of the two monoglucuronide isomers involved two distinct substrate-binding sites or reflected two different modes of binding to a single catalytic site. Both isomers were found in all tested species (man, rat, guinea pig, sheep), but there were marked species differences in the C-8/C-12 ratio of monoglucuronide found in bile or formed by liver microsomes. Correspondence between in vivo and in vitro results for such regioselectivity of glucuronidation was excellent in each species. On the basis of our results of kinetic analysis of bilirubin esterification at variable pigment substrate concentrations and inhibition studies with alternative substrates, we postulate that both natural monoglucuronide isomers are synthesized at a single binding site. Possible mechanisms responsible for the markedly regioselective esterification of bilirubin by rat and sheep liver were investigated by study of glucuronidation of selected structural analogues of the pigment. Our results do not support explanations of regioselectivity of bilirubin glucuronidation in terms of (i) preferential binding of either the C-8- or C-12-containing dipyrrolic half of the asymmetrical bilirubin molecule or (ii) enantioselective complexation of bilirubin UDPglucuronyltransferase to one of the two chirality enantiomers of intramolecularly hydrogen-bonded bilirubin.(ABSTRACT TRUNCATED AT 250 WORDS)
Semi-Simultaneous Saccharification and Fermentation Improved by Lignin and Extractives Removal from Sugarcane Bagasse

Abstract:Lignocellulosic biomass and agro-industrial residues are a source of fermentable sugars; however, pretreatments are needed to overcome biomass recalcitrance. This study evaluated the effect of sugarcane bagasse hydrolysis and fermentation in response to dilute acid pretreatment. In natura bagasse, extractive-free bagasse, partially delignified bagasse, and bagasse with added butylated hydroxytoluene antioxidant were pretreated with diluted acid and investigated in semi-simultaneous saccharification and fermentation (S-SSF). The effect of butylated hydroxytoluene (BHT) resulted in lower yields of inhibitors in the liquid fraction of the acid pretreatment (0.01 g L−1 of furfural, 0.01 g L−1 of 5-hydroxymethylfurfural, and 0.68 g L−1 of acetic acid). Partially delignified material and material with BHT resulted in biomass with low hemicellulose and lignin contents, indicating that BHT influenced lignin removal. Extractives removal showed benefits for the acid pretreatment, decreasing the dioxane-soluble material, and a higher yield of glucose and ethanol via S-SSF for the partially delignified material. Enzymatic saccharification of partially delignified material showed 87% of cellulose conversion (24 h with 15 FPU/g), and after 48 h of S-SSF (25 FPU/g), residual 7.06 g L−1 of glucose and production of 15.17 g L−1 of ethanol were observed. The low content of extractives, lignin, and dioxane soluble material resulted in better cellulose accessibility and ethanol yield. Chemical compounds can help remove lignin from biomass favoring ethanol production by S-SSF.

Abstract:To boost the enzymatic hydrolysis of CuCl 2 -catalyzed organosolv pretreated sugarcane bagasse, the single and composite additives were introduced to decrease the negative influence of residual lignin. By adding the optimal loadings of composite additives (e.g., 30 mg Tween 80, 40 mg tea saponin, 40 mg bovine serum albumin (BSA), and 0.8 mg xylanase based on per g substrate), the highest glucose yield reached 96.5% after hydrolysis for 48 h. Furthermore, the composite additives were introduced to high-solids enzymatic hydrolysis, and the partial supplement of the pretreated substrate and enzyme increased the highest glucose concentration to 194.3 g/L (including 10.8 g/L oligomer glucose), accounting for 88.8% of total fermentable sugars (218.9 g/L). This fed-batch mode along with the composite additives provided a beneficial approach for high-solids enzymatic hydrolysis, the high proportion of glucose was also convenient for commercial bioethanol production using industrial bacterial strains.

Abstract:In the present study, a sustainable pretreatment methodology combining liquid hot water and deep eutectic solvent is proposed for the efficient fractionation of hemicellulose, cellulose, and lignin from sugarcane bagasse, thereby facilitating the comprehensive utilization of both C5 and C6 sugars. The application of this combined pretreatment strategy to sugarcane bagasse led to notable enhancements in enzymatic saccharification and subsequent fermentation. Experiment results demonstrate that liquid hot water-deep eutectic solvent pretreatment yielded 85.05 ± 0.66 g·L −1 of total fermentable sugar (glucose: 60.96 ± 0.21 g·L −1 , xylose: 24.09 ± 0.87 g·L −1 ) through enzymatic saccharification of sugarcane bagasse. Furthermore, fermentation of the pretreated sugarcane bagasse hydrolysate yielded 34.33 ± 3.15 g·L −1 of bioethanol. These findings confirm the effectiveness of liquid hot water-deep eutectic solvent pretreatment in separating lignocellulosic components, thus presenting a sustainable and promising pretreatment method for maximizing the valuable utilization of biomass resources.

[The new euthanasia-discussion from the psychiatric viewpoint].

Abstract:The practice to kill terminally ill patients on their own demand has resulted in the Netherlands in a decriminalisation of active euthanasia which thus has fundamentally changed the way to deal with dying patients. Sooner or later this development will extend to other European countries as well as to the USA. Involuntary euthanasia of severely handicapped newborn children or of demented persons is propagated by the practical ethics of P. Singer and other representatives of utilitarianistic philosophy. According to the standpoint of utilitarianism a human being should only have the right to live as long as he or she is a person, i. e. has rationality and self-consciousness. The next step toward the elimination of elderly people can easily be predicted. For economical reasons these persons may be withheld from life-saving medical treatment or may be supposed to commit suicide. A moral pressure is created to make a decision for suicide as soon as severe invalidity occurs. The consideration of such ideas shows that in today's debate on euthanasia the issue is no longer the right of a few severely and terminally ill human beings to their "own death". Instead, the right to live of a large group of handicapped and "socially useless" or "unproductive" persons is at stake. This is the danger of today's discussion of euthanasia.
Enzymatic Hydrolysis Strategies for Cellulosic Sugars Production to Obtain Bioethanol from Eucalyptus globulus Bark

Abstract:Cellulosic sugars production for the valorization of lignocellulosic biomass residues in an industrial site has economic benefits and is promising if integrated into a biorefinery. Enzymatic hydrolysis (EH) of pretreated Eucalyptus globulus bark, an industrial residue of low-economic value widely available in Portuguese pulp and paper mills, could be an excellent approach to attain resource circularity and pulp mill profitability. This work evaluated the potential for improving cellulosic sugars concentrations by operating with high solids loading and introducing the additives Triton X-100, PEG 4000 and Tween 80 using a commercial enzymatic consortium with a dosage of 25 FPU gcarbohydrates−1. Additives did not improve enzymatic hydrolysis performance, but the effect of increasing solids loading to 14% (w/v) in batch operation was accomplished. The fed-batch operation strategy was investigated and, when starting with 11% (w/v) solids loading, allowed the feeding of 3% (w/v) fresh feedstock sequentially at 2, 4 and 6 h, attaining 20% (w/v) total solids loading. After 24 h of operation, the concentration of cellulosic sugars reached 161 g L−1, corresponding to an EH conversion efficiency of 76%. Finally, the fermentability of the fed-batch hydrolysate using the Ethanol Red® strain was evaluated in a 5 L bioreactor scale. The present results demonstrate that Eucalyptus globulus bark, previously pretreated by kraft pulping, is a promising feedstock for cellulosic sugars production, allowing it to become the raw material for feeding a wide range of bioprocesses.

Abstract:By screening out total nine sugarcane cultivars that contain directly-fermentable sugars, this study examined largely varied hexoses yields from direct enzymatic hydrolysis of bagasse residues. Using two representative sugarcane bagasse substrates, this study sorted out optimal K3PO4 pretreatments by performing classic response surface methodology, which respectively extracted 58% and 63% lignin and significantly reduced cellulose CrI and DP values in two pretreated bagasse residues. These consequently led to remarkably raised cellulose accessibility by 1.4- and 3.6 folds for near-complete biomass enzymatic saccharification with hexoses yields close to 100% (% cellulose). Notably, unlike physical (hot water, steam explosion) and chemical (acid, alkali) pretreatments as previously examined, this optimal pretreatment (6.4% K3PO4, 144.0 °C, 1 h) could not significantly extract arabinose-rich hemicellulose, suggesting that almost all xylose of hemicelluloses was applicable for engineered yeast co-fermentation to achieve maximum bioethanol at 29% (% dry matter) without the necessity of solid-liquid separation, water washing, and detoxification process. Furthermore, this study collected all K3PO4 liquid waste as chemical fertilizer into rice culture for much enhanced plant growth and biomass production up to 1.5 folds. Therefore, this work has proposed a mechanism model to highlight a cost-effective and green-like strategy for high yield of bioethanol with potential value-added bioproducts by using sugarcane bagasse and other lignocellulose residues.

Abstract:Sugarcane bagasse—a lignocellulosic material derived from sugarcane after crushing and extracting of its juice—is a promising feedstock for bioethanol production. The current study presented a two-step alkaline and acid pretreatment of sugarcane bagasse using sodium hydroxide and maleic acid as a strategy to enhance enzymatic digestibility of sugarcane bagasse. The two-step alkaline and acid pretreatment resulted to a significant increase in glucan content (69.72%) with accompanying decreases in xylan (12.99%) and lignin (4.89%) contents. Preliminary study on enzymatic hydrolysis of the two-step alkaline and acid-pretreated sugarcane bagasse with low solid loading and low cellulase concentrations resulted to high glucose yields. Optimization of high-solid enzymatic hydrolysis of two-step alkaline and acid-pretreated sugarcane bagasse was executed based on Box-Behnken design (BBD). BBD—a response surface methodology—is a powerful method that can be used to test several parameters by applying a minimum number of experimental trials. The optimal level of process variables as indicated by the statistical model namely solid loading (20%, w/w), incubation time (72 h), enzyme loading (2 FPU/g glucan), and substrate feeding (mode 2) were validated. Optimization of the process parameters resulted to 61.85% of glucose yield which was close to its predicted value (65.71%). The current study highlights the potential of high-solid enzymatic hydrolysis using two-step alkaline and acid-pretreated sugarcane bagasse for efficient fermentable sugar production.

Abstract:<p>The effect of NaOH catalytic ethanol pretreatment under various temperatures (130–180 °C) and time (15–90 min) on the chemical composition and enzymatic saccharification of sugarcane bagasse was investigated in this study. The results showed that NaOH catalytic ethanol pretreatment assisted delignification and the reservation of cellulose and hemicellulose. When sugarcane bagasse was pretreated at 180 °C for 30 min, a substantial glucose yield of 91.6% was obtained after hydrolysis for 72 h, representing 94.6% of glucose in pretreated residue. This yield was promoted with respect to the compositional change and surface alteration of pretreated substrate. With the supplement of Tween 80, the enzyme usage would be saved by 50% and the enzymolysis time could be shortened to 24 h while obtaining comparable glucose yield. This study provided an economical feasible and gradual process for the generation of glucose, which was followed by fermentation and conversion to platform chemicals.</p>

Abstract:An essential step in the conversion of lignocellulosic biomass to ethanol and other biorefinery products is conversion of cell wall polysaccharides into fermentable sugars by enzymatic hydrolysis. The objective of the present study was to understand the mode of action of hemicellulolytic enzyme mixtures for pretreated sugarcane bagasse (PSB) deconstruction and wheat arabinoxylan (WA) hydrolysis on target biotechnological applications. In this study, five hemicellulolytic enzymes—two endo-1,4-xylanases (GH10 and GH11), two α-L-arabinofuranosidases (GH51 and GH54), and one β-xylosidase (GH43)—were submitted to combinatorial assays using the experimental design strategy, in order to analyze synergistic and antagonistic effects of enzyme interactions on biomass degradation. The xylooligosaccharides (XOSs) released from hydrolysis were analyzed by capillary electrophoresis and quantified by high-performance anion exchange chromatography with pulsed amperometric detection (HPAEC–PAD). Based on this analysis, it was possible to define which enzymatic combinations favor xylose (X1) or XOS production and thus enable the development of target biotechnological applications. Our results demonstrate that if the objective is X1 production from WA, the best enzymatic combination is GH11 + GH54 + GH43, and for xylobiose (X2) production from WA, it is best to combine GH11 + GH51. However, if the goal is to produce XOS, the five enzymes used in WA hydrolysis are important, but for PSB hydrolysis, only GH11 is sufficient. If the final objective is bioethanol production, GH11 is responsible for hydrolyzing 64.3 % of hemicellulose from PSB. This work provides a basis for further studies on enzymatic mechanisms for XOS production, and the development of more efficient and less expensive enzymatic mixtures, targeting commercially viable lignocellulosic ethanol production and other biorefinery products.

Enhancement of Lignin Removal and Enzymolysis of Sugarcane Bagasse by Ultrasound-Assisted Ethanol Synergized Deep Eutectic Solvent Pretreatment

Abstract:A low-cost and green biorefinery will increase the economy and revenue from lignocellulosic biomass. This study demonstrated the effect of two-pot sequential pretreatment, comprising of ultrasound ethanol (USEL) synergized ternary deep eutectic solvent (TDES, choline chloride: glycerol: FeCl3 center dot 6H(2)O), with the aim to investigate the effects of USEL, TDES, and USEL thorn TDES pretreatments on enhancing delignification of sugarcane bagasse (SCB). The results showed that under the optimum TDES pretreatment conditions (FeCl3 center dot 6H(2)O, 120 degrees C, 3 h), the cellulose content and lignin removal rate (LRR) of SCB reached 47.67 and 82.71%, respectively. Under the optimum synergy conditions (USEL conditions: 100% ethanol, 20 + 28 + 40 kHz, 240 W, 60 min; TDES conditions: FeCl3 center dot 6H(2)O, 120 degrees C, 3 h), the cellulose content and LRR of SCB reached 66.17 and 86.39%, respectively. After enzymatic hydrolysis, the SCB pretreated with USEL + TDES shows a saccharification rate of 90.31%, which was substantially higher than that of the SCB pretreated with TDES alone (85.68%). X-ray diffraction, particle size, brunaueremmet-teller, scanning electron microscopy, and optical microscopy analyses were conducted to confirm the efficiency of USEL + TDES on pretreating SCB. Meanwhile, 2D-HSQC NMR analysis revealed that regenerated lignin exhibited well-preserved structures (beta-O-4, beta-beta linkages), making it suitable for depolymerization into monoaromatic compounds. Overall, this work demonstrated that biomass pretreatment with the USEL + TDES was promising for a low-cost biorefinery to efficiently fractionate lignocellulosic biomass into glucose and high-quality lignin with tailored chemical structures. A Comparison of Different Oxidative Pretreatments on Polysaccharide Hydrolyzability and Cell Wall Structure for Interpreting the Greatly Improved Enzymatic Digestibility of Sugarcane Bagasse by Delignification

Abstract:In order to confirm the contribution of delignification to the increase in lignocellulosic cellulose digestibility, several laboratory oxidative pretreatments under mild conditions, including alkaline-hydrogen peroxide (AP), two-step alkaline/peracetic acid (APAA) and sodium chlorite (SC) pretreatments were employed to achieve selective delignification of sugarcane bagasse and retained most of the hemicelluloses (xylan) in the pretreated solids. Four commercial cellulase cocktails were used to test the enzymatic hydrolyzability of pretreated substrates. Results revealed that delignification indeed could greatly improve the final (120 h) cellulose hydrolysis with relatively high final (120 h) glucan conversion (> 90%) by different cellulase cocktails even if the substrates still had a high hemicelluloses content. However, the xylan conversion seemed to be more greatly dependent on the pretreatments and cellulase cocktails used. AP and APAA pretreatments resulted in the disappearance of middle lamella and liberation of cellulose fibers with significant etching, deformation and fracture of cell wall structure. SC pretreatment greatly modified the sugar bagasse surface morphology to make the surface much coarser. The cell wall also underwent serious fracture and deformation with some middle lamella disappearing. However, no significant alteration on the structure of pure cellulose was observed by SC oxidative pretreatment of filter paper. Oxidative pretreatment might also modify lignin structure and surface properties thus greatly reducing the non-specific adsorption of enzymes. The obtained results strongly support the conclusion that delignification under mild pretreatment condition can be very helpful to improve the enzymatic hydrolysis of lignocellulosic cellulose by commercial cellulase cocktails even if the substrates has a high hemicelluloses content.

Abstract:During the bioconversion of lignocellulose to ethanol, the biomass always undergoes pretreatment in order to increase the enzymatic digestibility of cellulose. In present work, we conducted the pretreatment of sugarcane bagasse with aqueous acetic acid for delignification and alkali for deacetylation respectively (Acetoline process) to increase cellulose accessibility for enzymatic hydrolysis. The effects of several factors on the pretreatment effectiveness were investigated. The enzymatic digestibility of pretreated bagasse was further studied. The enzymatic glycan conversion of pretreated solid reached about 80% when it was digested under 7.5% solid consistency with cellulase of 15 FPU/g solid and beta-glucosidase of 10 CBU/g solid for 48 h. Compared with dilute acid pretreatment, Acetoline pretreatment could obtain higher enzymatic glycan conversion. The experimental results indicate that Acetoline is an effective pretreatment method to increase the enzymatic digestibility of sugarcane bagasse.
