Abstract:Carvedilol is a non-selective beta/alpha1 blocker that is broadly used in treatment of arrhythmia, congestive heart failure, hypertension, and myocardial infarction. Enhancing carvedilol low solubility and dissolution rate would help in improving the efficiency of the tablet dosage form. In this study, different types of silica nanoparticle (NPs) (SBA-16, MCM-41 and ZSM-5) were synthesized. Coupling of SBA-16 with 3-Aminopropyl-triethoxysilane (APTES) was performed to improve surface characteristics of SBA-16. Different methods to load carvedilol on these carriers were used with high-performance liquid chromatography (HPLC) as a method of analysis for carvedilol. For the characterization of the loaded NPs Fourier -transform infrared spectroscopy (FT-IR), X ray crystallography (XRD) and Therapeutic goods administration (TGA) were used. MCM-41 loaded with Carvedilol was decided to be formulated as tablet dosage and compared with tablets contain carvedilol without carrier. The results showed that SBA-16, SBA-16 coupled, ZSM-5 and MCM-41 carriers were used successfully to load carvedilol in a good drug load percent of 64.5, 69.3, 82 and 90%, respectively. MCM-41 gave highest loading efficiency of carvedilol using solvent evaporation method and the dissolution of carvedilol from powder was superior to the pure carvedilol where it gave 100% release of the drug at 15 minutes compared to 30% of pure carvedilol. MCM-41 loaded with carvedilol was successfully formulated as tablet dosage form according to the specifications of USP and the release of carvedilol from the prepared tablet was 99% at 15 min compared to the carvedilol released from tablets prepared using carvedilol powder which gave 38% which indicates the efficiency of the carrier in increasing the solubility and dissolution of carvedilol, a class II BCS drugs.

Abstract:Carvedilol (CARV) -a nonselective β-receptor antagonist indicated in treatment of hypertension-suffers from poor oral bioavailability due to its poor solubility and hepatic first-pass metabolism. This study aimed to optimize CARV-loaded cubosomes to enhance oral CARV bioavailability and prolong its action in the treatment of hypertension. D-optimal design was used to optimize CARV CUBs based on either Polyvinyl alcohol (PVA) or Cremophor RH 40 (RH 40) as secondary stabilizers. Furthermore, the effect of Poloxamer P407 percentage and secondary stabilizer concentration was studied. Comparing PVA-based to the RH 40-based CUBs, CARV-CUBs-RH40 showed lower particle size (PS), higher polydispersity index (PDI), lower zeta potential (ZP) and slightly lower CARV content. Optimized PVA-based and RH 40-based CUBs showed PS of 141 ± 1 nm and 105 ± 0 nm, PDI of 0.166 ± 0.00 and 0.247 ± 0.01, ZP of 4.9 ± 0.3 mV and 6.3 ± 0.3 and CARV content of 105.0 ± 1.4 % and 93.6 ± 0.9 %, respectively. TEM imaging confirmed the characteristic cubic morphology of the prepared CUBs. Moreover, the prepared CARV-CUBs exhibited an in vitro sustained release behavior and comparable in vivo antihypertensive performance. Abstract:Cardiovascular diseases remain a leading cause of morbidity and mortality worldwide, requiring innovative therapeutic strategies. This project explores a nano-pharmaceutical approach to enhance the efficacy of cardiovascular drugs, focusing on carvedilol and curcumin. These agents, known for their potential cardiovascular benefits, are encapsulated within Soluplus® micelles to form a novel drug delivery system. The novelty of this formulation lies in its ability to significantly improve the solubility of both carvedilol and curcumin, which have traditionally been limited by their hydrophobic nature. By utilizing Soluplus® micelles, we have developed a unique delivery system that optimizes the therapeutic potential of both drugs. The nanomicelles were meticulously characterized for drug loading, size distribution, and morphological features. The carvedilol and curcumin release patterns were investigated, revealing sustained and controlled release profiles. Additionally, the antioxidant capacity of the micellar formulation was evaluated, demonstrating the preservation of curcumin's antioxidative properties. In vivo studies using spontaneously hypertensive male rats explored the pharmacokinetics and hemodynamic effects of the nanomicellar system. These results indicated successful encapsulation of both drugs without altering their plasma profiles. Furthermore, the administration of carvedilol and curcumin micelles exhibited a more significant reduction in mean arterial pressure compared to individual drug administration, suggesting a potential synergistic effect. In conclusion, this nano-pharmaceutical approach offers a promising avenue for cardiovascular therapy, providing a platform for combined drug delivery and potential synergistic effects. The optimized formulation could lead to improved patient outcomes and enhanced cardiovascular health.

Abstract:Stable angina or angina pectoris is referred to as uneasiness/pain in the chest, resulting from coronary heart disease (CHD). Ivabradine Hydrochloride (IBH) is a recently approved drug to manage stable angina and heart failure symptoms. The approved IBH tablets exhibit some technical shortcomings i.e. short half-life (2 h), variable systemic absorption, and high first-pass metabolism (>50%). Therefore, utilizing a nanoformulation technique, we have designed a differentiated and innovative formulation of IBH. The IBH loaded polymeric nanoparticles (IBH-PNPs) are being developed for per-oral delivery by double emulsion method, using Poly lactic-co-glycolic acid (PLGA) as polymer, and d-α-tocopherol polyethylene glycol 1000 succinate (TPGS) as a stabilizer. The pre-formulation studies performed are UV spectroscopy, FT-IR, and X-ray diffraction. The Box-Behnken design was exploited for formulation optimization. The optimized formulation was characterized for its particle size, zeta potential, morphology, entrapment efficiency, in-vitro release, stability studies, ex-vivo permeability, and in-vivo pharmacodynamics study. The optimized IBH-PNPs were found to be spherical (<200nm) and exhibited normal size distribution under transmission electron microscopy and atomic force microscopy respectively. The zeta potential and entrapment efficiency were found to be −43.75 mv and 60 ± 4.8% respectively. Developed IBH-PNPs analyzed for in-vitro drug release where they exhibited biphasic release. The ex-vivo drug permeation study showed 1.85 folds increment in intestinal permeability as compared to IBH tablets. The in-vivo anti-anginal efficacy studies were performed using vasopressin-induced angina model in Wistar rats. Developed formulation was found to have a therapeutic effect for three days.

Intranasal Delivery of Carvedilol- and Quercetin-Encapsulated Cationic Nanoliposomes for Cardiovascular Targeting: Formulation and In Vitro and Ex Vivo Studies

Abstract:Carvedilol (CVD), an adrenoreceptor blocker, is a hydrophobic Biopharmaceutics Classification System class II drug with poor oral bioavailability due to which frequent dosing is essential to attain pharmacological effects. Quercetin (QC), a polyphenolic compound, is a potent natural antioxidant, but its oral dosing is restricted due to poor aqueous solubility and low oral bioavailability. To overcome the common limitations of both drugs and to attain synergistic cardioprotective effects, we formulated CVD- and QC-encapsulated cationic nanoliposomes (NLPs) in situ gel (CVD/QC-L.O.F.) for intranasal administration. We designed CVD- and QC-loaded cationic nanoliposomal (NLPs) in situ gel (CVD/QC-L.O.F.) for intranasal administration. In vitro drug release studies of CVD/QC-L.O.F. (16.25%) exhibited 18.78 ± 0.57% of QC release and 91.38 ± 0.93% of CVD release for 120 h. Ex vivo nasal permeation studies of CVD/QC-L.O.F. demonstrated better permeation of QC (within 96 h), i.e., 75.09% compared to in vitro drug release, whereas CVD permeates within 48 h, indicating the better interaction between cationic NLPs and the negatively charged biological membrane. The developed nasal gel showed a sufficient mucoadhesive property, good spreadability, higher firmness, consistency, and cohesiveness, indicating suitability for membrane application and intranasal administration. CVD-NLPs, QC-NLPs, and CVD/QC-NLPs were evaluated for in vitro cytotoxicity, in vitro ROS-induced cell viability assessment, and a cellular uptake study using H9c2 rat cardiomyocytes. The highest in vitro cellular uptake of CVD/QC-cationic NLPs by H9c2 cells implies the benefit of QC loading within the CVD nanoliposomal carrier system and gives evidence for better interaction of NLPs carrying positive charges with the negatively charged biological cells. The in vitro H2O2-induced oxidative stress cell viability assessment of H9c2 cells established the intracellular antioxidant activity and cardioprotective effect of CVD/QC-cationic NLPs with low cytotoxicity. These findings suggest the potential of cationic NLPs as a suitable drug delivery carrier for CVD and QC combination for the intranasal route in the treatment of various cardiovascular diseases like hypertension, angina pectoris, etc. and for treating neurodegenerative disorders.
Development of Carvedilol Nanoformulation-Loaded Poloxamer-Based In Situ Gel for the Management of Glaucoma

Abstract:The objective of the current study was to fabricate a thermosensitive in situ gelling system for the ocular delivery of carvedilol-loaded spanlastics (CRV-SPLs). In situ gel formulations were prepared using poloxamer analogs by a cold method and was further laden with carvedilol-loaded spanlastics to boost the precorneal retention of the drug. The gelation capacity, rheological characteristics, muco-adhesion force and in vitro release of various in situ gel formulations (CS-ISGs) were studied. The optimized formula (F2) obtained at 22% w/v poloxamer 407 and 5% w/v poloxamer 188 was found to have good gelation capacity at body temperature with acceptable muco-adhesion properties, appropriate viscosity at 25 °C that would ease its ocular application, and relatively higher viscosity at 37 °C that promoted prolonged ocular residence of the formulation post eye instillation and displayed a sustained in vitro drug release pattern. Ex vivo transcorneal penetration studies through excised rabbit cornea revealed that F2 elicited a remarkable (p ˂ 0.05) improvement in CRV apparent permeation coefficient (Papp = 6.39 × 10−6 cm/s) compared to plain carvedilol-loaded in situ gel (CRV-ISG; Papp = 2.67 × 10−6 cm/s). Most importantly, in normal rabbits, the optimized formula (F2) resulted in a sustained intraocular pressure reduction and a significant enhancement in the ocular bioavailability of carvedilol, as manifested by a 2-fold increase in the AUC0–6h of CRV in the aqueous humor, compared to plain CRV-ISG formulation. To sum up, the developed thermosensitive in situ gelling system might represent a plausible carrier for ophthalmic drug delivery for better management of glaucoma.

Abstract:The setting stress in composite resins was studied as a function of restoration shape. The shape is described by the configuration factor, C, the ratio of the restoration's bonded to unbonded (free) surfaces. In an experimental set-up, the shape of the restoration was simulated by cylindrical forms of various dimensions. The shrinkage stress was measured continuously. It was shown that in most of the clinically relevant cavity configurations, the stress-relieving flow is not sufficient to preserve adhesion to dentin by dentin-bonding agents.

Abstract:Stable angina (angina pectoris) is a type of chest pain that happens when the heart muscle needs more oxygen than usual but it's not getting it at that moment because of heart disease. The drawback of the marketed formulation is required repeated administration of the drug due to low bioavailability. A recently licenced medication called ivabradine hydrochloride (IVB) is used to treat stable angina and signs of heart failure. Technical problems in the approved IBH tablets include a two-hour half-life, erratic systemic absorption, and a high rate of first-pass metabolism (> 50%). We therefore created a distinctive and cutting-edge formulation of IVB using a nano formulation approach like nanostructured lipid carriers (NLCs). The response surface method with a three-level Box–Behnken design was used for the creation and improvement of IVB. The optimized formulation was proceeded for physicochemical characterizations like particle size, zeta potential, morphology (TEM and SEM), entrapment efficiency, in-vitro release, stability studies, compatibility study (DSC, FTIR and XRD), hemocompatibility study, ex-vivo permeability, and in-vivo angina study. As results, optimized formulation was found to be 114.45±5.14 nm particle size with 83.45±3.23% entrapment efficiency and biphasic release. The formulation showed spherical and compatible with excipients, no interaction was observed and hemocompatible. The IBH-NLCss showed more permeability (1.85 folds) as compared to the marketed dosage form. The in-vivo pharmacological activity was established in terms of decrease in severity and duration of ST-segment depression in vasopressin-induced angina model in experimental rats. Carvedilol Precipitation Inhibition by the Incorporation of Polymeric Precipitation Inhibitors Using a Stable Amorphous Solid Dispersion Approach: Formulation, Characterization, and In Vitro In Vivo Evaluation

Abstract:An amorphous solid dispersion (ASD) of carvedilol (CVL) was prepared via the solvent evaporation method, using cellulose derivatives as polymeric precipitation inhibitors (PPIs). The prepared ASDs existed in the amorphous phase, as revealed by X-ray powder diffraction (XRPD) and scanning electron microscopy (SEM). The Fourier-transform infrared spectroscopy (FT-IR) and differential scanning calorimetry (DSC) results confirmed the compatibility between CVL and the polymers used. The ASDs characteristics were evaluated, with no change in viscosity, a pH of 6.8, a polydispersity index of 0.169, a particle size of 423-450 nm, and a zeta potential of 3.80 mV. Crystal growth inhibition was assessed for 180 min via an infusion precipitation study in simulated intestinal fluid (SIF). The interactions between the drug and polymers were established in great detail, using nuclear magnetic resonance (NMR) spectroscopy, nuclear Overhauser effect spectroscopy (NOESY), and Raman spectroscopy studies. Dielectric analysis was employed to determine the drug-polymer interactions between ion pairs and to understand ion transport behavior. In vivo oral kinetics and irritation studies performed on Wistar rats have demonstrated promising biocompatibility, stability, and the enhanced bioavailability of CVL. Collectively, the stable ASDs of CVL were developed using cellulose polymers as PPIs that would inhibit drug precipitation in the gastrointestinal tract and would aid in achieving higher in vivo drug stability and bioavailability.

Abstract:Background: In the current work, co-rotating twin-screw processor (TSP) was utilized to formulate solid crystal suspension (SCS) of carvedilol (CAR) for enhancing its solubility, dissolution rate, permeation and bioavailability using mannitol as a hydrophilic carrier. Methods: In-silico molecular dynamics (MD) studies were done to simulate the interaction of CAR with mannitol at different kneading zone temperatures (KZT). Based on these studies, the optimal CAR: mannitol ratios and the kneading zone temperatures for CAR solubility enhancement were assessed. The CAR-SCS was optimized utilizing Design-of-Experiments (DoE) methodology using the Box-Behnken design. Saturation solubility studies and in vitro dissolution studies were performed for all the formulations. Physicochemical characterization was performed using differential scanning calorimetry , Fourier transform infrared spectroscopy, X-ray diffraction studies, and Raman spectroscopy analysis. Ex vivo permeation studies and in vivo pharmacokinetic studies for the CAR-SCS were performed. Stability studies were performed for the DoE-optimized CAR-SCS at accelerated stability conditions at 40 ºC/ 75% RH for three months. Results: Experimentally, the formulation with CAR: mannitol ratio of 20:80, prepared using a KZT of 120 ºC at 100 rpm screw speed showed the highest solubility enhancement accounting for 50-fold compared to the plain CAR. Physicochemical characterization confirmed the crystalline state of DoE-optimized CAR-SCS. In-vitro dissolution studies indicated a 6.03-fold and 3.40-fold enhancement in the dissolution rate of optimized CAR-SCS in pH 1.2 HCl solution and phosphate buffer pH 6.8, respectively, as compared to the pure CAR. The enhanced efficacy of the optimized CAR-SCS was indicated in the ex vivo and in vivo pharmacokinetic studies wherein the apparent permeability was enhanced 1.84-fold and bioavailability enhanced 1.50-folds compared to the plain CAR. The stability studies showed good stability concerning the drug content. Conclusions: TSP technology could be utilized to enhance the solubility, bioavailability and permeation of poor soluble CAR by preparing the SCS.
Potential of βC-Loaded Silica Nanoparticles in the Management of L-NAME –Induced Hypertension in Experimental Rats

Abstract:Natural bioactive compounds, like β-carotene (βC), are widely recommended as pharmaceutical supplements that are meant to aid in the treatment of diseases such as cardiovascular diseases. However, extreme hydrophobic nature, chemical instability, and low bioavailability of βC limit its clinical application. The purpose of this study was to synthesize and characterize silica nanoparticles (SiNPs) and βC-loaded silica nanoparticles (βC-SiNPs), and then study its activity as an antihypertensive agent in an experimental animal model. The sol–gel method was utilized in the preparation of SiNPs using different solvent systems. The loading of βC was accomplished through an in situ-loading method. The physicochemical characterization of obtained SiNPs was carried out to obtain information about morphology, average particle size, chemical integrity, thermal properties, and colloidal stability. In addition, the encapsulation efficiency (EE%) and release profile in either simulated gastric fluid or in physiological conditions were also investigated. The antihypertensive activity was evaluated in NG-nitro-L-arginine methyl ester hydrochloride (L-NAME)-induced hypertensive rat model. The results showed that the sol–gel synthesis SiNPs exhibited a spherical shape nanoparticle with an average size of 122 ± 38 nm for methanol/water mixture and 598 ± 123 nm for ethanol/water mixture. However, the sample with 0.05 g βC content revealed less average particle size (103 ± 22 nm) with a high EE% of 86%. The encapsulation efficiency (EE %) ranged from 36.4 to 86.8% according to the loading method and βC content. In addition, the drug release studies showed two distinct release steps due to initial burst effect followed by a controlled release. Indeed, the lowest dose of (βC-SiNPs) exhibited ameliorative effects against L-NAME-induced hypertension in adult male Wistar rats. Heart tissues and blood were obtained after rats were administered L-NAME and/or βC-SiNPs orally for 2 weeks for additional histological and biochemical studies. The results revealed that L-NAME therapy resulted in significant cardiac pathological damage as well as a rise in heart function analyses as serum LDH and creatine kinase (CK-MB). In conclusion, nanostructured vehicles (i.e., silica NPs) could be a promising strategy for controlling release of hydrophobic bioactive compounds in pharmaceutical industry and act as antihypertensive agent by restoring biochemical parameters and the histopathological changes in cardiac muscles.

Abstract:Selective laser sintering (SLS) has drawn attention for the fabrication of three-dimensional oral dosage forms due to the plurality of drug formulations that can be processed. The aim of this work was to employ SLS with a CO2 laser for the manufacturing of carvedilol personalised dosage forms of various strengths. Carvedilol (CVD) and vinylpyrrolidone-vinyl acetate copolymer (Kollidon VA64) blends of various ratios were sintered to produce CVD tablets of 3.125, 6.25, and 12.5 mg. The tuning of the SLS processing laser intensity parameter improved printability and impacted the tablet hardness, friability, CVD dissolution rate, and the total amount of drug released. Physicochemical characterization showed the presence of CVD in the amorphous state. X-ray micro-CT analysis demonstrated that the applied CO2 intensity affected the total tablet porosity, which was reduced with increased laser intensity. The study demonstrated that SLS is a suitable technology for the development of personalised medicines that meet the required specifications and patient needs.

Abstract:Carvedilol, a β-blocker prescribed for chronic heart failure, suffers from poor bioavailability and rapid first pass metabolism when administered orally. Herein, we present the development of tip microarray patches (MAPs) composed of ternary cyclodextrin (CD) complexes of carvedilol for transdermal delivery. The ternary complex with hydroxypropyl γ-cyclodextrin (HPγCD) and poly(vinyl pyrrolidone) (PVP) reduced the crystallinity of carvedilol, as evidenced by DSC, XRD, NMR, and SEM analysis. MAPs were fabricated using a two-step process with the ternary complex as the needle layer. The resulting MAPs were capable of breaching ex vivo neonatal porcine skin to a depth ≈600 μm with minimal impact to needle height. Upon insertion, the needle dissolved within 2 h, leading to the transdermal delivery of carvedilol. The MAPs displayed minimal toxicity and acceptable biocompatibility in cell assays. In rats, MAPs achieved significantly higher AUC levels of carvedilol than oral administration, with a delayed T<sub>max</sub> and sustained plasma levels over several days. These findings suggest that the carvedilol-loaded dissolving MAPs have the potential to revolutionise the treatment of chronic heart failure.

Preparation and Characterization of Bovine Serum Albumin Nanoparticles of Curcumin using a Chemometric Approach

Abstract:Bovine Serum Albumin (BSA) has been presupposed to be a versatile protein polymer for targeted drug delivery. BSA nanoparticles can lead to passive targeting of drugs to the inflamed joint via the Enhanced Permeability and Retention (EPR) effect and due to their specific affinity towards the inflamed joint because of the inadequacy of protein in the affected region. Hence, the aim of the study was to develop BSA nanoparticles loading curcumin (BSA_CUR_NPs) by nanoparticle albuminbound technology and its optimization was conducted by 33 Box Behnken Design (BBD) in order to achieve the desired particle size and entrapment efficiency. Further, the optimised nanoparticles were also assessed for polydispersity index, zeta potential, total drug content, and in-vitro drug release study. The response surface plots and equations generated by BBD predicted the relationship between variables under study. The optimised formulation C12 was found to have a particle size of 207.1 ± 1.36nm, PDI of 0.138 ± 0.03, entrapment efficiency of 75.04 ± 0.06 %, total drug content of 91.40 ± 0.08% and zeta potential of -32.9mV. The optimised nanoparticles exhibited good sustained release for up to 8 days. The use of a chemometric approach led to the development of BSA_CUR_NPs with the desired characteristics with a less experimental procedure. Therefore, it presents an important model for producing the nanoparticles of the desired characteristics using albumin as a polymer for the enhanced and sustained delivery of loaded drugs to the inflamed joint.
Solubility and Stability of Carvedilol in Deep Eutectic Solvents: A Step Towards a Sustainable Pharmaceutical Formulation in the Liquid State

Abstract:This study investigates the use of aqueous eutectic mixtures to enhance the solubility of carvedilol, a drug with low solubility and bioavailability. It was found that mixtures of citric acid and choline chloride increased the maximum solubility. Additionally, the stability of carvedilol in these mixtures was demonstrated. These findings contribute to the sustainable development of liquid medications. Carvedilol is used to treat chronic hypertension, chronic stable angina pectoris and concomitant treatment heart failure and class II of the Biopharmaceutics Classification System (low solubility and poor bioavailability). This drug is only formulated as tablets, and liquid oral formulations are not available on the market. In this work, several aqueous eutectic mixtures formed by xylitol, citric acid, sorbitol and glucose (HBD) and choline chloride as HBA have been used to improve the solubility. Additionally, SwissADME was used to obtain physicochemical and pharmacokinetic parameters to further knowledge on the ADME (absorption, distribution, metabolism and excretion) parameters for carvedilol. The results reveal that the solubility of carvedilol increases as the content of water decreases, and the mixture formed by citric acid and choline chloride (CACh10) was the most soluble. The stability study shows that carvedilol in DES is stable during the test time, as the maximum and minimum concentration losses were 19.0 % and 0.2 %, respectively. These results help advance the green and sustainable development of new medicines in the liquid state.

Abstract:BACKGROUND: Niosomes are non-ionic surfactant vesicles used as drug carriers for encapsulating both hydrophobic and hydrophilic drugs. The aim of this study is to evaluate the effect of different surfactants on the physical properties and stability of carvedilol niosomes designed to improve oral bioavailability.MATERIALS AND METHODS: Different niosomal formulations were prepared using a film hydration method, with various mixtures of different non-ionic surfactants including Span 20, 40, and 60, and also Tween 20, 40, and 60, along with cholesterol. The physicochemical characteristics of the formulations were evaluated in vitro.RESULTS: The drug encapsulation efficiency was reduced by using lauryl (C12) chain containing surfactants, that is, Span/Tween. Cholesterol content and drug entrapment were the main factors affecting the mean particle size of the niosomes. The drug release profiles from most of the formulations were fitted well with the Baker-Lonsdale model, indicating a diffusion-based drug release mechanism. Niosomes prepared from 50 and 40% of the cholesterol with 25 or 30% of Span/Tween 60 showed the highest stability due to their high transition temperature and solid state feature of these surfactants.CONCLUSIONS: From the results obtained, it may be concluded that nanoniosomes are promising stable carriers for the oral delivery of carvedilol.

Abstract:Nanotechnology has emerged as an inspiring tool for the effective delivery of drugs to help treat Coronary heart disease (CHD) which represents the most prevalent reason for mortality and morbidity globally. The current study focuses on the assessment of the cardioprotective prospective ofanovel combination nanoformulation of sericin and carvedilol. Sericin is a silk protein obtained from Bombyx mori cocoon and carvedilol is a synthetic nonselective β-blocker. In this present study, preparation of chitosan nanoparticles was performed via ionic gelation method and were evaluated for cardioprotective activity in doxorubicin (Dox)-induced cardiotoxicity. Serum biochemical markers of myocardial damage play a substantial role in the analysis of cardiovascular ailments and their increased levels have been observed to be significantly decreased in treatment groups. Treatment groups showed a decline in the positivity frequency of the Troponin T test as well. The NTG (Nanoparticle Treated Group), CSG (Carvedilol Standard Group), and SSG (Sericin Standard Group) were revealed to have reduced lipid peroxide levels (Plasma and heart tissue) highly significantly at a level of p < 0.01 in comparison with the TCG (Toxic Control Group). Levels of antioxidants in the plasma and the cardiac tissue were also established to be within range of the treated groups in comparison to TCG. Mitochondrial enzymes in cardiac tissue were found to be elevated in treated groups. Lysosomal hydrolases accomplish a significant role in counteracting the inflammatory pathogenesis followed by disease infliction, as perceived in the TCG group. These enzyme levels in the cardiac tissue were significantly improved after treatment with the nanoformulation. Total collagen content in the cardiac tissue of the NTG, SSG, and CSG groups was established to be highly statistically significant at p < 0.001 as well as statistically significant at p < 0.01, respectively. Hence, the outcomes of this study suggest that the developed nanoparticle formulation is effective against doxorubicin-induced cardiotoxicity.
