Abstract:The separation of charge variants is a challenge for the production of monoclonal antibodies (mAbs). Cation exchange chromatography (CEX) is normally used to accomplish this task. In this study, with IgG1-type mAb as the model protein, charge variants separation was investigated under various conditions including resins, elution mode, gradient slope and flow rate. The trade-off between main peak purity and recovery was focused to evaluate the separation performance. The results indicated that the diameter of resin particles was the critical factor. For SP Sepharose Fast Flow (90 mu m diameter resin), the overlap between main peak and charge variants was too serious to meet the separation requirements. For Monomix MC30 SP (30 mu m diameter resin), the purity of main peak was improved to 93.4% under 0.05 pH/CV and 100 cm/h. For Nano SP-15L (15 mu m diameter resin), near 100% purity of main peak could be obtained with a recovery of 56.5% under 0.05 pH/CV and 100 cm/h. Two types of elution gradient, salt gradient and pH gradient, were compared and it was found that there was no obvious difference after the optimization and both showed some improvements with a shallower gradient. In addition, slower flow rate could improve the separation of main peak. In general, smaller resin particle size, shallower gradient slope and slower flow rate could improve the separation performance of main peak and charge variants under both salt gradient and pH gradient elution, which should be selected according to the actual requirements.
A Novel Twin‐column Continuous Chromatography Approach for Separation and Enrichment of Monoclonal Antibody Charge Variants

Abstract:Abstract Downstream processing of mAb charge variants is difficult owing to their similar molecular structures and surface charge properties. This study aimed to apply a novel twin‐column continuous chromatography (called N‐rich mode) to separate and enrich acidic variants of an IgG1 mAb. Besides, a comparison study with traditional scaled‐up batch‐mode cation exchange (CEX) chromatography was conducted. For the N‐rich process, two 3.93 mL columns were used, and the buffer system, flow rate and elution gradient slope were optimized. The results showed that 1.33 mg acidic variants with nearly 100% purity could be attained after a 22‐cycle accumulation. The yield was 86.21% with the productivity of 7.82 mg/L/h. On the other hand, for the batch CEX process, 4.15 mL column was first used to optimize the separation conditions, and then a scaled‐up column of 88.20 mL was used to separate 1.19 mg acidic variants with the purity of nearly 100%. The yield was 59.18% with the productivity of 7.78 mg/L/h. By comparing between the N‐rich and scaled‐up CEX processes, the results indicated that the N‐rich method displays a remarkable advantage on the product yield, i.e. 1.46‐fold increment without the loss of productivity and purity. Generally, twin‐column N‐rich continuous chromatography displays a high potential to enrich minor compounds with a higher yield, more flexibility and lower resin cost.
Development of a Hybrid Biomimetic Ligand with High Selectivity and Mild Elution for Antibody Purification

Abstract:A hybrid biomimetic ligand composed of three amino acid residues and a hydrophobic charge-induction group was developed and its performance of immunoglobulin G (IgG) purification from human serum was evaluated. The sequence of peptides of the hybrid ligand (Phenylalanine-Tyrosine-Glutamate-"5-amino-benzimidazole", FYE-ABI) was designed and evaluated via molecular simulation, and the ligand was synthesized and coupled onto agarose beads. Adsorption isotherm results showed that the optimum IgG adsorption was found at pH 7.0 with saturated adsorption capacity of 91 mg/mL resin and dissociation constant of 1.6 mu M. The hybrid biomimetic resin showed high separation purity of 99.0% and recovery of 91.5% when used in IgG separation from human serum albumin (HSA) containing solutions. Moreover, study on IgG separation from human serum showed that the hybrid biomimetic ligand could provide comparable IgG purity (93.9%) and recovery (88.9%), but much milder elution condition (pH 4.5) as those of Protein A affinity chromatography. The combination of biomimetic and hydrophobic charge-induction chromatography presented high selectivity and mild elution for antibody purification.
Process design and application of twin-column continuous chromatography for antibody affinity separation

Abstract:In order to improve process throughput and reduce operation cost of Protein A affinity chromatography, a twin-column continuous chromatography method was studied for monoclonal antibody (mAb) separation. Effects of residence time in the column and loading protein concentration were investigated, and the separation conditions were optimized for mAb capture from cell culture broth. Two typical Protein A affinity resin (MabSelect SuRe and UniMab) were used and the breakthrough curves were measured under different residence times and IgG concentrations, and their effects on the productivity, resin capacity utilization and buffer consumption were evaluated. The results indicate that the twin-column continuous chromatography method has higher productivity and capacity utilization, and lower buffer consumption when comparing to traditional batch chromatography at low residence times. The separation performance of continuous chromatography for mAb capture from cell culture broth was similar to batch chromatography with purity > 97%, yield > 87%, aggregates < 2% and host cell proteins < 300 ppm. However, the capacity utilization increased from 20% ~30% to 70% ~80%, and the buffer consumption reduced to 1/5~1/4 of traditional batch processes. The results demonstrate that the twin-column continuous chromatography method shows great potential for the mAb production.

Abstract:Antibody, especially monoclonal antibody has become an important therapeutic drug which has a broad market demand and prospect.Nowadays the bottleneck of antibody production has shifted from antibody expression to downstream processing.The typical antibody purification process starts with protein A affinity chromatographic to capture the antibody and includes two subsequent polishing steps for impurity removal, such as ionic exchange, hydrophobic interaction or hydroxyapatite chromatography.There are some limitations restricting the development in antibody industry.Therefore, the researchers are committed to develop novel purification methods, including mixed-mode chromatography (MMC) and short peptide biomimetic chromatography which remedy the weakness of traditional methods.According to the recent advances in the purification of monoclonal antibody drugs, protein A affinity chromatography and the novel chromatographic methods and their applications in monoclonal antibody purification are reviewed in the present article.
Integration of Expanded Bed Adsorption and Hydrophobic Charge-Induction Chromatography for Monoclonal Antibody Separation

Abstract:The integration of expanded bed adsorption (EBA) with hydrophobic charge-induction chromatography (HCIC) affords a promising new technology to capture antibody from the complex feedstock. New EBA resin T-ABI was prepared with hydrophobic charge-induction ligand 5-aminobenzimidazole (ABI) and used to separate monoclonal antibody (mAb) from CHO cell culture broth. The static and dynamic adsorption behaviors of hIgG and mAb were investigated, and the typical properties of pH dependence and salt tolerance were found. High dynamic binding capacities at 10% breakthrough (18.1-21.4 mg/mL resin) were obtained even for high operation velocities (711-1203 cm/h) in an expanded bed. By optimization of loading pH, elution pH, and expansion factor, hIgG could be separated from the protein mixture (2 mg/mL hIgG and. 10 mg/mL bovine serum albumin) with high efficiency. Finally, mAb was separated directly from the CHO cell culture broth with T-ABI EBA under optimized conditions (loading at pH 7.0, elution at pH 4.0 or 4.5, expansion factor of 2.0), and the purity reached 93.7-97.7% with the recovery of 72.6-79.4%. The results indicated that T-ABI resin was suitable to capture hIgG from the complicated feedstock. New separation process with T-ABI EBA showed a promising potential for antibody purification with high productivity and process efficiency, which combined the advantages of HCIC and EBA.

Abstract:The increasing demand for efficient and robust processes in the purification of monoclonal antibodies (mAbs) has recently brought frontal chromatography to the forefront. Applied during the polishing step, it enables the removal of high molecular weight aggregates from the target product, achieving high purities. Typically, this process is operated in batch using a single column, which makes it intrinsically subjected to a purity-yield tradeoff. This means that high purities can only be achieved at the cost of lowering the product yield and vice versa. Recently, a two-column continuous implementation of frontal chromatography, referred to as Flow2, was developed. Despite being able of alleviating the purity-yield tradeoff typical of batch operations, the increase in the number of process parameters complicates its optimal design, with the risk of not exploiting its full potential. In this study, we developed an ad hoc design procedure (DP) suitable for the optimization of both batch frontal chromatography and Flow2 in terms of purity, yield, and productivity. This procedure provided similar results as a multiobjective optimization based on genetic algorithm but with lower computational effort. Then, batch and Flow2 operated at their optimal conditions were compared. Besides showing a more favorable Pareto front of yield and productivity at a specified purity, the Flow2 process demonstrated improved robustness compared to the batch process with respect to modifications in the loading linear velocity, washing buffer ionic strength and loading time, thus providing an appealing operation for integrated processes.
Evaluating Antibody Monomer Separation from Associated Aggregates Using Mixed-Mode Chromatography

Abstract:Aggregate level is a key factor when assessing quality attributes of antibody product, since the aggregation might impact the biological activity of biopharmaceutical. Effective separation methods are usually essential in manufacturing processes. In this study, three mixed-mode resins, i.e. Capto adhere and two home-made resins with benzylamine and butylamine as the functional ligands (named BA and AB), were used to investigate the separation performances of antibody aggregate removal processes. In addition, two traditional resins (Q Sepharose FF and Phenyl Sepharose 6 FF) were also investigated to explore the role of molecular interaction in the aggregate removal. The results indicated that the removal efficiency was highly dependent on the mass loading. With the sample load of 50mg/ml resin both Capto adhere and BA resins can significantly reduce the aggregate level from 20.5% to 2.6% and 2.4%, respectively, while a relatively low degree of aggregate reduction was found with the traditional resins. The research indicates that both hydrophobic interaction and electrostatic interaction are critical for the aggregate removal and the cooperativity of different molecular interactions is important for the effective aggregates removal with mixed-mode resins.
Coupling cation and anion exchange chromatography for fast separation of monoclonal antibody charge variants

Abstract:A design procedure for the separation of charge variants of a monoclonal antibody (mAb) was developed, which was based on the coupling of cation-exchange chromatography (CEX) and anion-exchange chromatography (AEX) under high loading conditions. The design of the coupled process was supported by a dynamic model. The model was calibrated on the basis of band profiles of variants determined experimentally for the mAb materials of different variant compositions. The numerical simulations were used to select the coupling configuration and the loading conditions that allowed for efficient separation of the mAb materials into three products enriched with each individual variant: the acidic (av), main (mv) and basic (bv) one. In the CEX section, a two-step pH gradient was used to split the loaded mass of mAb into a weakly bound fraction enriched with av and mv, and a strongly bound fraction containing the bv-rich product. The weakly bound fraction was further processed in the AEX section, where the mv-rich product was eluted in flowthrough, while the av-rich product was collected by a step change in pH. The choice of flow distribution and the number of columns in the CEX and AEX sections depended on the variant composition of the mAb material. For the selected configurations, the optimized mAb loading density in the CEX columns ranged from 10 to 26 mg mL-1, while in the AEX columns it was as high as 300 or 600 mg mL-1, depending on the variant composition of the mAb material. By proper selection of the loading condition, a trade-off between yield and purity of the products could be reached.

Abstract:The process of countercurrent multistage precipitation (CnMP) was developed for reduction of the acidic variant ( av ) content in monoclonal antibody (mAb) pools. The process was performed in polyethylene glycol (PEG) solutions of low ionic strength, at which av was the most prone to precipitate. To design the process, a mathematical model was formulated, which consisted of steady-state mass balance equations and underlying thermodynamic dependencies. The model was solved for different combinations of the process variables, including the protein concentration in the feed material and the PEG concentration in subsequent precipitation stages. The model solution was projected onto two-dimensional planes, where the performance indicators, such as the separation yield and the av reduction level, were correlated with the process variables. CnMP overperformed crosscurrent multistage precipitation (CsMP) and ion exchange chromatography (IEX) used in previous studies; in the comparison with CsMP, the 3-stage CnMP process allowed up to 28% increase in yield and up to 25% increase in the av reduction level, and in the comparison with IEX, up to 37% increase in yield and up 12.5% increase in the av reduction level. The yield benefit of CnMP depended on the composition of the feed material and demand for the av reduction. The concept was experimentally verified; the 2- and 3-stage CnMP processes were performed using the input process variables provided by the model solution. The performance indicators calculated by the model and experimentally measured were in a good agreement, which indicated both feasibility and predictability of CnMP. Abstract:Molecular interactions between ligands and target biomolecules are crucial in the development of chromatographic techniques for the separation and purification of biotherapeutics. In this study, the role of functional moieties on a mixed-mode ligand (phenylalanine-tyrosine-glutamate-5-aminobenzimidazole) for human immunoglobulin G purification was investigated and a detailed mechanism was discussed. A similar ligand with glutamic acid substituted by glutamine (phenylalanine-tyrosine-glutamine-5-aminobenzimidazole) together with other resins including a commercial resin (CM Bestarose Fast Flow), phenylalanine-tyrosine-glutamate, glutamate-5-aminobenzimidazole, and 5-aminobenzimidazole resins were prepared for comparison. Molecular dynamics simulation and experimental studies were used to analyze the difference between these ligands. The results showed that the carboxyl group of phenylalanine-tyrosine-glutamate-5-aminobenzimidazole contributed 70% of the electrostatic interaction during human immunoglobulin G binding, and 5-aminobenzimidazole provided electrostatic repulsion for desorption, which showed low selectivity and binding capacities at pH 4.0 (dynamic binding capacities at 10% breakthrough of human immunoglobulin G = 1.0 mg/ml resin, dynamic binding capacities at 10% breakthrough of human serum albumin = 1.2 mg/ml resin) when used as an individual resin ligand. The results showed in this study demonstrated that it is possible to achieve optimal antibody separation and purification through reasonable ligand design by understanding the performance of key functional moieties in binding and elution processes.
Predicting Colloidal Stability of High-Concentration Monoclonal Antibody Formulations in Common Pharmaceutical Buffers Using Improved Polyethylene Glycol Induced Protein Precipitation Assay.

Abstract:Colloidal stability is an important consideration when developing high concentration mAb formulations. PEG-induced protein precipitation is a commonly used assay to assess the colloidal stability of protein solutions. However, the practical usefulness and the current theoretical model for this assay have yet to be verified over a large formulation space across multiple mAbs and mAb-based modalities. In the present study, we used PEG-induced protein precipitation assays to evaluate colloidal stability of 3 mAbs in 24 common formulation buffers at 20 and 5 °C. These prediction assays were conducted at low protein concentration (1 mg/mL). We also directly characterized high concentration (100 mg/mL) formulations for cold-induced phase separation, turbidity, and concentratibility by ultrafiltration. This systematic study allowed analysis of the correlation between the results of low concentration assays and the high concentration attributes. The key findings of this study include the following: (1) verification of the usefulness of three different parameters (Cmid, μB, and Tcloud) from PEG-induced protein precipitation assays for ranking colloidal stability of high concentration mAb formulations; (2) a new method to implement PEG-induced protein precipitation assay suitable for high throughput screening with low sample consumption; (3) improvement in the theoretical model for calculating robust thermodynamic parameters of colloidal stability (μB and εB) that are independent of specific experimental settings; (4) systematic evaluation of the effects of pH and buffer salts on colloidal stability of mAbs in common formulation buffers. These findings provide improved theoretical and practical tools for assessing the colloidal stability of mAbs and mAb-based modalities during formulation development.

Abstract:Charge heterogeneity among therapeutic monoclonal antibodies (mAbs) is considered an important critical quality attribute and requires careful characterization to ensure safe and efficacious drug products. The charge heterogeneity among mAbs is the result of chemical and enzymatic post-translational modifications and leads to the formation of acidic and basic variants that can be characterized using cation exchange chromatography (CEX). Recently, the use of mass spectrometry-compatible salt-mediated pH gradients has gained increased attention to elute the proteins from the charged stationary phase material. However, with the increasing antibody product complexity, more and more selectivity is required. Therefore, in this study, we set out to improve the selectivity by using a solvent-enriched mobile phase composition for the analysis of a variety of mAbs and bispecific antibody products. It was found that the addition of the solvents to the mobile phase appeared to modify the hydrate shell surrounding the protein and alter the retention behavior of the studied proteins. Therefore, this work demonstrates that the use of solvent-enriched mobile phase composition could be an attractive additional method parameter during method development in CEX.
Modeling the impact of amino acid substitution in a monoclonal antibody on cation exchange chromatography

Abstract:A vital part of biopharmaceutical research is decision making around which lead candidate should be progressed in early‐phase development. When multiple antibody candidates show similar biological activity, developability aspects are taken into account to ease the challenges of manufacturing the potential drug candidate. While current strategies for developability assessment mainly focus on drug product stability, only limited information is available on how antibody candidates with minimal differences in their primary structure behave during downstream processing. With increasing time‐to‐market pressure and an abundance of mAbs in development pipelines, developability assessments should also consider the ability of mAbs to integrate into the downstream platform.This study investigates the influence of amino acid substitutions in the complementarity‐determining region (CDR) of a full‐length IgG1 mAb on the elution behavior in preparative cation exchange (CEX) chromatography. Single amino acid substitutions within the investigated mAb resulted in an additional positive charge in the light chain (L) and heavy chain (H) CDR, respectively. The mAb variants showed an increased retention volume in linear gradient elution compared to the wild type antibody. Further, the substitution of tryptophan with lysine in the H‐CDR3 increased charge heterogeneity of the product. A multi‐scale in silico analysis, consisting of homology modeling, protein surface analysis, and mechanistic chromatography modeling increased understanding of the adsorption mechanism. Abstract:Immunoglobulin G (IgG) is the most common monoclonal antibody (mAb) grown for therapeutic applications. While IgG is often selectively isolated from cell lines using protein A (ProA) chromatography, this is only a stepping stone for complete characterization. Further classification can be obtained from weak cation exchange chromatography (WCX) to determine IgG charge variant distributions. The charge variants of monoclonal antibodies can influence the stability and efficacy in vivo, and deviations in charge heterogeneity are often cell-specific and sensitive to upstream process variability. Current methods to characterize IgG charge variants are often performed off-line, meaning that the IgG eluate from the ProA separation is collected, diluted to adjust the pH, and then transferred to the WCX separation, adding time, complexity, and potential contamination to the sample analysis process. More recently, reports have appeared to streamline this separation using in-line two-dimensional liquid chromatography (2D-LC). Presented here is a novel, 2D-LC coupling of ProA in the first dimension ( 1 D) and WCX in the second dimension ( 2 D) chromatography. As anticipated, the initial direct column coupling proved to be challenging due to the pH incompatibility between the mobile phases for the two stages. To solve the solvent compatibility issue, a size exclusion column was placed in the switching valve loop of the 2D-LC instrument to act as a means for the on-line solvent exchange. The efficacy of the methodology presented was confirmed through a charge variant determination using the NIST monoclonal antibody standard (NIST mAb), yielding correct acidic, main, and basic variant compositions. The methodology was employed to determine the charge variant profile of IgG from an in-house cultured Chinese hamster ovary (CHO) cell supernatant. It is believed that this methodology can be easily implemented to provide higher-throughput assessment of IgG charge variants for process monitoring and cell line development.

Abstract:Monoclonal antibodies (mAbs) have become fundamental across various sectors, with growing demands in both therapeutic and non-therapeutic applications. For the antibodies capture step, precipitation stands out as an alternative or precursor method to chromatographic techniques. However, research on unconventional precipitants, such as polyethylene glycol (PEG) and ZnCl 2 , for diluted non-therapeutic mAbs is still scarce. This study evaluates the potential and challenges of combining PEG 6000 kDA and ZnCl 2 as precipitants to capture anti-human IgG 1 mAbs from highly diluted cell culture supernatants. We established mAbs solubility curves, identified optimal conditions for batch precipitation, fine-tuned the resolubilization step and conducted a viscosity analysis for the precipitates. We achieved 100% yield and 59% purity for the redissolution step, after using a 12% PEG 6000/3 mmol L −1 ZnCl 2 precipitation condition with a 1:1 dilution ratio at pH 6.0 for resolubilization. Nevertheless, results also indicate that the resolubilization step potentially leads to mAb fragmentation and that protein aggregates might impact mAb viscosity and flow behavior. In conclusion, the combination of these agents is promising for mAbs capture, however, pre-concentration techniques are advised for severely diluted systems and caution toward ZnCl 2 is recommended, especially concerning the pH of the system and ZnCl 2 concentration.

Abstract:Cation exchange chromatography (CEX) of therapeutic monoclonal antibodies is generally performed with either salt gradient (MES buffer + NaCl) or using commercial pH gradient buffer. The goal of this study was to find out some alternative buffer systems for CEX separation of mAbs, which may offer alternative selectivity, while maintaining similar peak shapes. Among the new buffers that were tested, (N-morpholino)ethanesulfonic acid (MES) / 1,3-diamino-2-propanol (DAP), and citric acid / 2-(cyclohexylamino)ethanesulfonic acid (CHES) systems were particularly promising, especially when combining them with a moderate salt gradient of NaCl. This two buffer system provides an equivalent or slightly better separation than the standard, mobile phases for therapeutic mAbs. It was also demonstrated that working with salt-mediated pH gradients, allows to extend the possibilities in method development, since the concentration of salt in the mobile phase has a significant impact on selectivity. Using HPLC modeling software (Drylab), it was possible to successfully develop CEX methods for authentic mAb samples within only 6 h, by optimizing the gradient steepness and salt concentration in the B eluent.

Abstract:Peptide mapping with mass spectrometry (MS) is an important tool for protein characterization in the biopharmaceutical industry. Historically, peptide mapping monitors post-translational modifications (PTMs) of protein products and process intermediates during development. Multi-attribute monitoring (MAM) methods have been used previously in commercial release and stability testing panels to ensure control of selected critical quality attributes (CQAs). Our goal is to use MAM methods as part of an overall analytical testing strategy specifically focused on CQAs, while removing or replacing historical separation methods that do not effectively distinguish CQAs from non-CQAs due to co-elution. For example, in this study, we developed a strategy to replace a profile-based ion-exchange chromatography (IEC) method using a MAM method in combination with traditional purity methods to ensure control of charge variant CQAs for a commercial antibody (mAb) drug product (DP). To support this change in commercial testing strategy, the charge variant CQAs were identified and characterized during development by high-resolution LC-MS and LC-MS/MS. The charge variant CQAs included PTMs, high molecular weight species, and low molecular weight species. Thus, removal of the IEC method from the DP specification was achieved using a validated LC-MS MAM method on a QDa system to directly measure the charge variant PTM CQAs in combination with size exclusion chromatography (SE-HPLC) and capillary electrophoresis (CE-SDS) to measure the non-PTM charge variant CQAs. Bridging data between the MAM, IEC, and SE-HPLC methods were included in the commercial marketing application to justify removing IEC from the DP specification. We have also used this MAM method as a test for identity to reduce the number of QC assays. This strategy has received approvals from several health authorities.
