Abstract:Uncontrolled hemorrhage is a main cause of pre-hospital death. Given the importance of hemostatic wound dressings in pre-hospital emergency treatment, novel composite materials are required for fast hemostasis, synergistic bacterial ablation with negligible resistance and wound healing acceleration. Herein, multifunctional SCTF cryogels were fabricated by the simultaneous cross-linking of sodium alginate (SA) and tannic acid (TA) with Fe3+ ions. As a result, the prepared SCTF cryogels consisted of Fe3+/TA-based metal phenolic networks (MPNs) and Fe3+/SA-based 3D skeleton for collagen (CA). MPNs endowed the cryogels with photothermal effect, photothermal-enhanced Fenton activity and pH/photothermal dual-responsive release property of TA and Fe2+, which were beneficial for the antibacterial capacity. Due to the intrinsic high porosity, in vitro and in vivo assays demonstrated that SCTF cryogels possessed good hemostatic capacity. Moreover, the synergistic photothermal therapy (PTT), chemodynamic therapy (CDT) and pH/photothermal responsive chemo-therapy dramatically enhanced the bactericidal efficacy of SCTF cryogels both in vitro and in vivo. Eventually, their outstanding healing-accelerating effects were confirmed via animal experiments, which were attributed to the presence of CA and TA. Therefore, the developed composite materials could offer new strategy on exploiting multifunctional wound dressing for clinical applications in the future.

Abstract:The development of novel wound dressings, such as aerogels, with rapid hemostasis and bactericidal capacities for pre-hospital care is necessary. To prevent the occurrence of bacterial resistance, antibacterial photodynamic therapy (aPDT) with broad-spectrum antibacterial ability and negligible bacterial resistance has been intensively studied. However, photosensitizers often suffer from poor water solubility, short singlet oxygen (1O2) half-life and restricted 1O2 diffusion distance. Herein, sodium alginate was covalently modified by photosensitizers and phenylboronic acid, and cross-linked by Ca(II) ions to generate SA@TPAPP@PBA aerogel after lyophilization as an antibacterial photodynamic wound dressing. Afterwards, its photodynamic and bacterial capture activities were intensively evaluated. Furthermore, its hemostasis and bactericidal efficiency against Staphylococcus aureus were assessed via in vitro and in vivo assays. First, chemical immobilization of photosensitizers led to an enhancement of its solubility. Moreover, it showed an excellent hemostasis capacity. Due to the formation of reversible covalent bonds between phenylboronic acid and diol groups on bacterial cell surface, the aerogel could capture S. aureus tightly and dramatically enhance aPDT. To sum up, the prepared aerogel illustrated excellent hemostasis capacity and antibacterial ability against S. aureus. Therefore, they have great potential to be utilized as wound dressing in clinical trials.

Alginate-derived hydrogels synergetic with metal-phenolic nanospheres for chronic wound therapy

Abstract:Management of diabetic wounds presents a global health challenge due to elevated levels of ROS in the wound microenvironment, persistent dysregulation of inflammation modulation, and limitations in commercially available dressings. Addressing this issue, we have developed a pH-responsive and glucose-sensitive multifunctional hydrogel dressing that dynamically responds to the wound microenvironment and enables on-demand drug release. The dressing incorporates a matrix material based on aminophenylboronic acid-functionalized alginate and polyhydroxy polymer, alongside an enhancer phase consisting of a self-assembled metal-phenol coordination nanospheres formed by tannic acid and iron ions. Through the dynamic borate ester bonds and catechol-metal ion coordination bonds, the dressing exhibits remarkable shape adaptability, self-healing capability, tissue adhesiveness, antioxidant activity, and photothermal responsiveness, without additional curatives or crosslinking agents. As a wound dressing, it elicits macrophage polarization towards an anti-inflammatory phenotype while maintaining long-lasting antimicrobial effects. In a diabetic mouse model of full-thickness wound infections, it effectively mitigated inflammation and vascular damage, significantly expediting the wound healing process with a commendable 97.7% wound closure rate. This work provides a new direction for developing multifunctional smart hydrogel dressings that can accelerate diabetic wound healing for human health.

Abstract:Wound infection resulting in delayed wound healing and wound deterioration remains a clinical challenge. Recently, multifunctional hydrogel dressing was a promising strategy which has attracted wide attention in preventing wound infection and promoting wound healing. In this study, a hybrid hydrogel made of gelatin (GL), tannic acid (TA), oxidized sodium alginate (OSA), and zinc oxide nanoparticles (ZnO NPs) was prepared mainly by double network cross-linking approach, named tannic acid-gelatin/oxidized sodium alginate/zinc oxide (TA-GL/OSA/ZnO). The composite hydrogels exhibited improved mechanical properties, which provided by TA modified the structure of GL network, Schiff base reaction between GL and OSA, and the strengthening effect of ZnO NPs. Meanwhile, the composite hydrogel showed high antibacterial activity against Staphylococcus aureus (S. aureus) (97.8 % ± 0.9 %) and Escherichia coli (E. coli) (96.6 % ± 1.2 %), attributed to the synergistic effect of TA and ZnO NPs. Furthermore, benefiting from the good antioxidative properties of TA, the sustain-released Zn2+ with the good capability to kill bacteria, and promoting the regeneration of skin epithelial tissues in BALB/c mice constantly, the multifunctional hydrogel had a significant therapeutic effect on wound healing and broad application prospects.

Abstract:Bacterial infections in wounds continue to be a major challenge in clinical settings worldwide and represent a significant threat to human health. This work proposes novel expandable and versatile methods for solidifying sodium alginate (SA) with metal ions (such as Fe3+, Co2+, Ni2+, Cu2+, and Zn2+) to create Metal-Alginate (M-Alg) hydrogel with adjustable morphology, composition, and microstructure. It conforms to the wound site, protects against second infection, reduces inflammation, and promotes the healing of infected wounds. Among these hydrogels, Cu-Alginate (Cu-Alg) shows excellent sterilization effect and good efficacy against both gram-positive and gram-negative bacteria, including multidrug-resistant (MDR) strains such as Methicillin-resistant Staphylococcus aureus (MRSA) and Carbapenem-resistant Klebsiella pneumoniae (CRKP) due to its dual antibacterial mechanisms: contact-killing and reactive oxygen species (ROS) burst. Importantly, it exhibits low cytotoxicity and biodegradability. This simple and cost-effective gel-based system has the potential to introduce an innovative approach to the management of wound infection and offers promising new perspectives for the advancement of wound care practice.

Abstract:Microorganisms induced wound infection and the accompanying excessive inflammatory response is the daunting problems in wound treatment. Due to the lack of corresponding biological functions, traditional wound dressings cannot effectively protect the wound and are prone to induce local infection, excessive inflammation, and vascular damage, resulting in prolonged unhealing. Here, a mussel-inspired strategy was adopted to prepare a multifunctional hydrogel created by H2O2/CuSO4-induced rapid polydopamine (PDA) deposition on carboxymethyl chitosan (CMC)/sodium alginate (Alg) based hydrogel, termed as CAC/PDA/Cu(H2O2). The prepared CAC/PDA/Cu(H2O2) hydrogel features excellent biocompatibility, adequate mechanical properties, and good degradability. Moreover, the CAC/PDA/Cu(H2O2) hydrogel can not only realize antibacterial, and anti-inflammatory effects, but also promote angiogenesis to accelerate wound healing in vitro thanks to the composite PDA/Cu(H2O2) coatings. Significantly, CAC/PDA/Cu(H2O2) hydrogel illustrates excellent therapeutic effects in Methicillin-resistant Staphylococcus aureus (MRSA) induced-rat infection models, which can efficiently eliminate MRSA, dramatically reduce inflammatory expression, promote angiogenesis, and ultimately shorten the wound healing time. CAC/PDA/Cu(H2O2) hydrogel exhibited the best wound healing rate on days 7 (80.63 ± 2.44 %), 11 (92.45 ± 2.26 %), and 14 (97.86 ± 0.66 %). Thus, the multifunctional hydrogel provides a facile and efficient approach to wound management and represents promising potential in the therapy for wound healing.

Abstract:Bacterial infections and excessive oxidative stress seriously hinder the healing of skin wounds. Traditional wound dressings can only serve as physical barriers and lack active molecules essential for actively promoting wound healing. Herein, an antibacterial and antioxidant liquid metal inorganic active material is developed for wound repair through in situ polymerization of chitosan/acrylic acid precursor solution initiated by tannic acid-coated liquid metal nanoparticles, without extra initiators and ultraviolet (UV) light. The tannic acid component enables the inorganic active material to exhibit antioxidant property, which can remove 90% of free radicals and relieve cellular oxidative stress. The chitosan component endows the inorganic active material with antibacterial property, effectively inhibiting the growth of Staphylococcus aureus and Escherichia coli (killing ratio: 90%). In vivo experiment demonstrates that this inorganic active material can promote the healing of Staphylococcus aureus -infected wound, achieving a closure rate of 98.16% on the 9th day. Meanwhile, this inorganic active material exhibits good electrical conductivity, enabling timely and stable monitoring of human joint movements. This work offers a simple strategy for developing multifunctional inorganic active material, which holds great potential for wound repair and motion monitoring.

A low-swelling alginate hydrogel with antibacterial hemostatic and radical scavenging properties for open wound healing

Abstract:Development of a low-cost and biocompatible hydrogel dressing with antimicrobial, antioxidant, and low swelling properties is important for accelerating wound healing. Here, a multifunctional alginate hydrogel dressing was fabricated using the D-(+)-gluconic acidδ-lactone/CaCO3system. The addition of hyaluronic acid and tannic acid (TA) provides the alginate hydrogel with anti-reactive oxygen species (ROS), hemostatic, and pro-wound healing properties. Notably, soaking the alginate hydrogel in a poly-ϵ-lysine (EPL) aqueous solution enables the alginate hydrogel to be di-crosslinked with EPL through electrostatic interactions, forming a dense network resembling 'armor' on the surface. This simple one-step soaking strategy provides the alginate hydrogel with antibacterial and anti-swelling properties. Swelling tests demonstrated that the cross-sectional area of the fully swollen multifunctional alginate hydrogel was only 1.3 times its initial size, thus preventing excessive wound expansion caused by excessive swelling. After 5 h ofin vitrorelease, only 7% of TA was cumulatively released, indicating a distinctly slow-release behavior. Furthermore, as evidenced by the removal of 2,2-diphenyl-1-picrylhydrazyl free radicals, this integrated alginate hydrogel systems demonstrate a notable capacity to eliminate ROS. Full-thickness skin wound repair experiment and histological analysis of the healing site in mice demonstrate that the developed multifunctional alginate hydrogels have a prominent effect on extracellular matrix formation and promotion of wound closure. Overall, this study introduces a cost-effective and convenient multifunctional hydrogel dressing with high potential for clinical application in treating open wounds.
Facile Synthesis of Thermo-Sensitive Composite Hydrogel with Well Dispersed Ag Nanoparticles for Application in Superior Antibacterial Infections

Abstract:In this study, we have described a facile process for fabrication of multifunctional composite hydrogel, in which sodium alginate was subjected to cross-linking using Ca2+ derived from ZnO/CaCO₃/Ag composite nanospheres. The ZnO/CaCO₃/Ag composite nanospheres were prepared based on our previously reported AA-[Zn(OH)₄]2- composite nanosphere reaction conducted with silver and calcium salt following hydrothermal method, that led to the disintegration and release of Ca2+ under acidic conditions for application as a cross-linking agentto catalyze reaction with sodium alginate. Ag nanoparticles were well-dispersed in the multifunctional composite hydrogel, exhibiting excellent antibacterial activity. Additionally, polydopamine (PDA) with photothermal effect was also added to obtain a multifunctional composite hydrogel, and this hydrogel showed photothermal conversion performance and facilitated the release of Ag+ to achieve the rapid antibacterial effect. Simultaneously, PDA NPs could scavenge free radicals and improve cell adhesion. All such features would promote wound healing. The potent antimicrobial activity of the prepared composite hydrogel was demonstrated in the mouse model of S. aureus infection, and biosafety of the hydrogel was confirmed by conducting histopathological examination in the mouse model. This type of multifunctional hydrogel wound dressing with photosensitive and antibacterial properties presents with broad applications and prospects in antibacterial treatment.
Tannic acid based multifunctional hydrogels with mechanical stability for wound healing

Abstract:Conventional wound dressings have poor tissue adhesion and mechanical stability, restricting their applications in dynamic motion environments. Tannic acid (TA) was ideal candidates for current dressing materials due to their well-known antioxidant and anti-inflammatory properties. However, the inevitable polymerization problem of TA limited the one-step synthesis of dressings. Herein, we reported a simple one-pot method to prepare double-network hydrogels containing N-acryloyl glycinamide (NAGA), N-hydroxyethyl acrylamide (HEAA) and TA. The resulting NHT hydrogel exhibited excellent tensile properties, fatigue resistance, and notch insensitivity to ensure mechanical stability under large deformation and stress in vitro. The NHT hydrogel also demonstrated room-temperature self-healing, broad adhesion to various substrates, synergistic swelling ability. In addition, catechol and benzene rings from TA helped shield against UV radiation and acted as free radical scavengers to relieve oxidative stress in wound damage. As a result, full-layer wounds in mice treated with NHT patches showed a higher healing rate, in which epithelialization was completed within 14 days. The integrated function enables hydrogel to maintain mechanical stability in dynamic motion environments with high strain and defects, with great potential for future clinical translation.

Abstract:Rapid hemostasis and wound healing are crucial in emergency trauma situations for saving patients' lives. Traditional hemostatic materials often have drawbacks such as slow hemostasis and susceptibility to post-hemostasis bacterial infections. Therefore, there is an urgent need for advanced wound dressing materials that can provide both rapid hemostasis and antimicrobial properties. In this study, we designed and fabricated a biocompatible hemostatic silk fibroin (SF) aerogel loaded with aggregation-induced emission (AIE)-featured berberine (BBr) and Ti3C2Tx MXene. The resulting SFMB aerogel demonstrates robust mechanical properties and a porous structure that enables quick hemostasis. This aerogel exhibits photodynamic and photothermal responses for antimicrobial activity, releases BBr upon light exposure to enhance bacterial-killing efficiency (99.33 % in vitro and 99.09 % in vivo), and effectively promotes the healing of infected wounds in vivo through combined photothermal/photodynamic antibacterial and anti-inflammatory mechanisms. Furthermore, the aerogel shows high hemocompatibility and cytocompatibility, supporting its potential biomedical applications. Overall, the synthesized SFMB aerogel holds promise for treating bacteria-infected wounds and for use in first aid applications in clinical settings.
Bimetal-organic framework/GOx-based hydrogel dressings with antibacterial and inflammatory modulation for wound healing

Abstract:Antibiotic resistance of bacteria and persistent inflammation are critical challenges in treating bacteria infected wounds. Thus, it is urgent to develop versatile wound dressings that possess high-efficiency antibacterial performance and inflammation regulation. Herein, we have successfully constructed a hydrogel wound dressing consisting of the bimetallic metal-organic framework (MOF) loaded with glucose oxidase (GOx), termed as MOF(Fe-Cu)/GOx-polyacrylamide (PAM) gel. Hydrogel dressings can provide an efficient cascade-catalyzed system to accelerate wound healing via synergistic antibacterial and inflammatory modulation. Importantly, the catalytic property of the bimetallic MOF(Fe-Cu) is about five times that of the monometallic MOF(Fe). Based on such a cascade-catalyzed system, the abundant gluconic acid and H2O2 can be continuously produced by decomposing glucose via GOx. Such gluconic acid can notably improve the peroxidase performance of MOF(Fe-Cu), which can further efficiently decompose H2O2 to achieve the antibacterial. Meanwhile, MOF (Fe Cu)/GOx PAM gel can induce macrophages to change into an M2 phenotype, which can accelerate the transformation of the wound microenvironment to a remodeling state and then accelerate angiogenesis and neurogenesis. This work provides multifunctional bioactive materials for accelerating wound healing and will have great potential in clinical applications. STATEMENT OF SIGNIFICANCE: Antibiotic resistance and persistent inflammation are still the critical reasons for the slow healing of bacteria infected wounds. Herein, we prepared a hydrogel wound dressing composed of bimetallic metal organic framework (MOF) loaded with glucose oxidase (GOx). The catalytic activity of the bimetallic MOF(Fe-Cu) is significantly enhanced due to doping of copper, which makes it possess outstanding antibacterial ability based on cascade catalysis. Such dressing can promote the remodeling of inflammatory immunity by regulating macrophage polarization to suppress over-reactive inflammation, further accelerating the healing of bacteria-infected wounds. This study provides an innovative and effective way to accelerate the healing of bacteria infected wound by combining bacteria killing and inflammation modulation.

Abstract:The skin, the primary barrier of the body, is inevitably broken. However, the development of materials that facilitate wound healing with sustained antimicrobial, hemostatic, and biocompatible properties remains a formidable challenge. In this article, we prepared a photopolymerizable composite hydrogel consisting of a hydrogel matrix, a hemostatic/antibacterial agent, and a photothermal therapy agent. The photopolymerizable hydrogel matrix was prepared by grafting the photoinitiator and polymerizable active monomer onto the chitosan chain segment, which exhibits excellent biocompatibility. Furthermore, linalool is adsorbed on the surface of halloysite nanotubes (HNTs) to form a hemostatic and antibacterial. Meanwhile, dopamine is employed as a coating material for hollow glass microsphere (HGM), which enables them to function as photothermal therapy agents. Upon exposure to near-infrared radiation, the PHA hydrogel releases linalool molecules from the surface of the HNTs, which diffuse into the hydrogel matrix, resulting in a sustained antimicrobial effect. At the same time, rapid curing of the photopolymerizable hydrogel under UV light forms a physical barrier that synergistically enhances the hemostatic properties of the HNTs. From the above, the results pave the way to develop a potential hemostatic antimicrobial dressing for clinical use in wound healing.

Abstract:Hydrogel dressings with similar structural characteristics to the extracellular matrix and tunable physicochemical properties have become promising candidates for wound healing. However, the fabrication of an ideal hydrogel dressing with low-cost, good biocompatibility, excellent hemostatic capacity, potent and broad-spectrum antibacterial activity remains a huge challenge. Herein, a lignocellulose-based nanocomposite hydrogel (ATC/SA/PVA) is fabricated by simply mixing Ag nanoparticles loaded, tannic acid-decorated lignocellulose nanofibrils with sodium alginate and polyvinyl alcohol. Based on the dynamic borate ester bonds and multiple weak hydrogen bonds, the fabricated hydrogel exhibits excellent flexibility and self-healing performance. Its highly porous structure endows the gel excellent blood and tissue exudates absorption ability. Interestingly, the release behavior of Ag nanoparticles from hydrogel displays pH dependence, which can facilitate the accumulation of Ag nanoparticles at the wound site, thereby accelerating the process of wound healing. In vitro antibacterial assay demonstrates the potent antibacterial ability of hydrogel against both Gram-positive (S. aureus) and negative bacteria (E. coli). More importantly, in vivo investigations reveal that such hydrogel can effectively accelerate tissue regeneration and wound healing with no obvious adverse effects. All these results suggest that this nanocomposite hydrogel would be a promising candidate to accelerate wound healing.

Abstract:Frequent occurrence of wound infection caused by multiple-resistant bacteria (MRB) has posed a serious challenge to the current healthcare system relying on antibiotics. The development of novel antimicrobial materials with high safety and efficacy to heal wound infection is of great importance in combating this crisis. Herein, we prepared a promising antibacterial hydrogel by cross-linking ferrous ions (Fe2+) with the deprotonated carboxyl anion in sodium alginate (Na-ALG) to cure wound infections caused by methicillin-resistant Staphylococcus aureus (MRSA). Interestingly, ferrous-modified Na-ALG (Fe-ALG) hydrogel demonstrated better properties compared to the traditional Na-ALG-based hydrogels, including injectability, self-healing, appropriate fluidity, high-water retention, potent MRSA-killing efficacy, and excellent biocompatibility. Importantly, the addition of Fe2+ enhances the antibacterial efficacy of the Na-ALG hydrogel, enabling it to effectively eliminate MRSA and accelerate the healing of antibiotic-resistant bacterial-infected wounds in a remarkably short period (10 days). This modification not only facilitates wound closure and fur generation, but also mitigates systemic inflammation, thereby effectively impeding the spread of MRSA to the lungs. Taken together, Fe-ALG hydrogel is a promising therapeutic material for treating wound infections by Staphylococcus aureus, especially by antibiotic-resistant strains like MRSA.

Abstract:Uncontrollable bleeding and infection are two of the most common causes of trauma-related death. Yet, developing safe materials with high hemostatic and antibacterial effectiveness remains a challenge. Keratin-based biomaterials have been reported to exhibit the functions of enhancing platelet binding and activating and facilitating fibrinogen polymerization. In this study, we designed hemostatic material with good biodegradability, biocompatibility, hemostatic ability, and antibacterial function to solve the shortcomings of common hemostatic materials. The methylene blue-loaded keratin/alginate composite scaffolds were prepared by the freeze-gelation method. The composite scaffolds exhibited over 1600% liquid absorption, well-interconnected pores, good biocompatibility, and biodegradability. We find that the keratin/alginate composite scaffolds' synergistic action may significantly reduce hemostasis time. To prevent infection, the drug-loaded scaffolds generated a high burst release by absorbing wound exudate in the early stages of wound healing. The results obtained by the antimicrobial photoinactivation assay in vitro suggest that an antimicrobial photodynamic effect might be triggered, thereby preventing the fast growth of colonies.

Abstract:Chronic wounds, especially diabetic, foot and pressure ulcers are a major health problem affecting >10 % of the world's populace. Calcium phosphate materials, particularly, bioactive glasses (BG), used as a potential material for hard and soft tissue repair. This study combines nanostructured 45S5 BG with titania (TiO 2 ) and alumina (Al 2 O 3 ) into a composite via simple sol-gel method. Prepared composites with alginate (Alg) formed a bioactive nanocomposite hydrogel membrane via freezing method. X-ray diffraction revealed formation of two phases such as Na 1.8 Ca 1.1 Si 6 O 14 and β-Na 2 Ca 4 (PO 4 ) 2 SiO 4 in the silica network. Fourier transformed InfraRed spectroscopy confirmed the network formation and cross-linking between composite and alginate. <2 % hemolysis, optimal in vitro degradation and porosity was systematically evaluated up to 7 days, resulting in increasing membrane bioactivity. Significant cytocompatibility, cell migration and proliferation and a 3–4-fold increase in Collagen (Col) and Vascular Endothelial Growth Factor (VEGF) expression were obtained. Sustained delivery of 80 % Dox in 24 h and effective growth reduction of S. aureus and destruction of biofilm development against E. coli and S. aureus within 24 h. Anatomical fin regeneration, rapid re-epithelialization and wound closure were achieved within 14 days in both zebrafish and in streptozotocin (STZ) induced rat in vivo animal models with optimal blood glucose levels. Hence, the fabricated bioactive membrane can act as effective wound dressing material, for diabetic chronic infectious wounds.
