Abstract:Adenosine has received great attentions acting as a potential biomarker for monitoring lung cancer. Most of the reported studies for adenosine detection require large instruments and complicated procedures. Herein, a sensitive, rapid and in-situ colorimetric aptasensor was developed for adenosine detection. Moreover, a homemade biomimetic electronic-eye (E-eye) was established and utilized as a portable in-time detection equipment. The entire measurement can be completed within 20 min, including the combination of aptamer with adenosine or AuNPs and the detection of adenosine. Four different kinds of aptamer were compared and the results showed that the AuNPs-aptamer-biotin system was the most stable and with the widest detection range of 5.0 mu M-60.0 mu M and the lowest LOD of 0.17 mu M. Moreover, the artificial urine samples were also tested with a linear range from 5.0 to 50.0 mu M and a LOD of 0.48 mu M. The results validated that the aptasensor together with the E-eye can be a promising platform for adenosine detection. (C) 2020 Elsevier B.V. All rights reserved.

Abstract:Cellular adenosine triphosphate (ATP) release detection is of great importance owing to its signaling function. For increasing the electrode surface area and simultaneously maintaining the enzyme activity, a 3D Pt nanoflowers decorated micro-needle tip electrode has been fabricated through a simple method in this work. A dual enzyme ATP biosensor was developed by the co-immobilization of glucose oxidase and hexokinase on the surface of the 3D Pt nanoflowers decorated micro-needle tip. The simply fabricated ATP biosensor can be used for in situ determination of cellular ATP release with a sensitivity of 0.84 +/- 0.10 nA mu M-1 mm(-2) to ATP and a low limit of detection (5 mu M, S/N = 3). The proposed 3D Pt nanoflowers decorated microelectrodes are promising to be used for the determination of other neurotransmitters and cell secretions.

Abstract:Adenosine triphosphate (ATP) plays a crucial role in energy metabolism and extracellular purinergic signaling. A 3D bimetallic Au/Pt nanoflowers decorated ATP microelectrode biosensor prepared by facile and effective template-free electrodeposition was firstly reported, realizing local detection of cellular ATP secretion. The ATP biosensor was developed by co-immobilization of glucose oxidase and hexokinase, exhibiting long-term stability (79.39 ± 9.15% of its initial value remained after 14 days at 4 °C) and high selectivity with a limit of detection down to 2.5 μM (S/N = 3). The resulting ATP biosensor was then used for direct in situ monitoring of ATP secreted from living cells (PC12) with the stimulation of high K+ solutions. The obtained current was about 21.6 ± 3.4 nA (N = 6), corresponding to 12.2 ± 2.8 μM ATP released from cells, right in the micromolar range and consistent with the suggested levels. The 3D bimetallic Au/Pt nanoflowers possess excellent catalytic activity and large electroactive surface area, contributing to enzymatic activity preservation and long-term stability. This work provides a promising platform for long-time monitoring of other neurotransmitters and secretions in cellular glycolysis and apoptosis processes in the future.
In vivo Monitoring of Serotonin by Nanomaterial Functionalized Acupuncture Needle

Abstract:Acupuncture treatment is amazing but controversial. Up to now, the mechanism of treating diseases by acupuncture and moxibustion is still unclear, especially the occurrence of the molecular events in local acupoints. Herein, we report an extremely stable microsensor by modifying carbon nanotube (CNT) to the tip surface of acupuncture needle and applying this CNT-modified acupuncture needle for real time monitoring of serotonin (5-HT) in vivo. To stabilize CNT modification on the needle tip surface, poly(3,4-ethylenedioxythiophene)(PEDOT) was employed as glue water to stick CNT on the needle. The detection limit of the CNT-modified needle was found to be approximately 50 nM and 78 nM in the PBS and the cell medium, respectively. In addition, the needle showed good selectivity to some inflammatory mediators and some electroactive molecules. For the first time, the CNT-modified needle could be directly probed into rat body for real time monitoring of 5-HT in vivo, showing a great potential for better understanding the mechanism of acupuncture treatment.

Abstract:In this study, a novel spectroelectrochemical method was proposed for neurotransmitters detection. The central sensing device was a hybrid structure of nanohole array and gold nanoparticles, which demonstrated good conductivity and high localized surface plasmon resonance (LSPR) sensitivity. By utilizing such specially-designed nanoplasmonic sensor as working electrode, both electrical and spectral responses on the surface of the sensor could be simultaneously detected during the electrochemical process. Cyclic voltammetry was implemented to activate the oxidation and recovery of dopamine and serotonin, while transmission spectrum measurement was carried out to synchronously record to LSPR responses of the nanoplasmonic sensor. Coupling with electrochemistry, LSPR results indicated good integrity and linearity, along with promising accuracy in qualitative and quantitative detection even for mixed solution and in brain tissue homogenates. Also, the detection results of other negatively-charged neurotransmitters like acetylcholine demonstrated the selectivity of our detection method for transmitters with positive charge. When compared with traditional electrochemical signals, LSPR signals provided better signal-to-noise ratio and lower detection limits, along with immunity against interference factors like ascorbic acid. Taking the advantages of such robustness, the coupled detection method was proved to be a promising platform for point-of-care testing for neurotransmitters.

Abstract:Cellular adenosine triphosphate (ATP) release detection is of great significance due to its signaling function. A triple-enzyme microelectrode array ATP biosensor was developed by the co-immobilization of glucose oxidase, hexokinase and horseradish peroxidase. To increase the surface area, improve the sensitivity and lower the limit of detection, Lithops-like Pt nanorods based on Au spikes were fabricated for the detection of cellular ATP release. The proposed MEA integrated with the Lithops-like Pt nanorods can be used for real-time measurement of local ATP release from single cells. The results will contribute to the determination of other neurotransmitters and cell secretions at the single cell level.

Abstract:Adenosine can be a possible biomarker for monitoring the progress of lung cancer. Here, a sensitive and easily operated colorimetric methods based on aptasensor for the detection of adenosine has been developed. Gold nanoparticles (AuNPs) will aggregate in high-salt solutions, which causes color change, and single-stranded DNA will prevent this from happening. Based on changes in solution color, adenosine can be quantitatively determined. Besides a home-made bionic electronic-eye (E-eye) system was developed as a portable in-situ detection platform. Two different aptamers were used for comparison for adenosine detection. The linear correlation existed between 5.0-60.0μM with a detection limit of 0.17μM in water with modified aptamer. Moreover, the performance of the method was validated by testing the artificial urine samples with the linear range from 5.0 to 50.0μM and a detection limit of 0.48μM in artificial urine. This simple and cost-effective approach indicating its great potential for point-of-care applications.

Abstract:A rigorous preselection of identified compounds by in vitro cellular screening is necessary prior to using the drug candidates for the further time consuming and parallel monitoring stage. In this paper we introduced an integrated cell based chip which can detect changes of electrophysiological parameters by using microelectronic chips such as light-addressable potentiometric sensors(LAPS), micro-electrodes array sensors(MEAS) and interdigitated array sensor(IDA) etc. to record the extracellular metabolism chemical and biological substances, action potential and living characteristics of cells representing the different response. In this research we presented novel ideas including an integrating multifunctional cell sensors chip, obtaining the living cells as the source of cell sensors with cardiac myocytes. Further developments will be done by culturing living olfactory and taste cells on surface of the devices. Now, we have completed the fabrication of the sensor and obtained some promising experiments results in different sensor units. This research will be interesting and useful for parallel extracellular monitoring.

Abstract:Reduced nicotinamide adenine dinucleotide (NADH) plays crucial roles in maintaining cell growth and differentiation, cytoprotection and energy metabolism. And it is imperative to precisely monitor the variable quantity of NADH. Herein, the connotative feature of acupuncture needle (AN) was explored as electrochemical matrix. Poly-3, 4-ethylenedioxythiophene-liganded gold nanoparticle modified AN electrode (ANE/AuNPs/PEDOT) was prepared via a simple electrosynthesis technology. Working together by charge transfer and reaction has a strong response to the electron-transfer of “N” in NADH, which is coordinately presented as a current peak. Interestingly, the behavior of electron-transfer was affected by ambient environment. After the conditional optimization, the fabricated sensor exhibited good linear range of 20 ∼ 500 μmol/L with a detection limit of 18.5 μmol/L. In addition, the sensor can be successfully applied to analysis NADH in serum. Furthermore, the microelectrode exhibits the real-time monitor ability for NADH that released from MDA-MB231 cells. This sensor also possesses the significant performance of high selectivity, stability and reproducibility. Based on the acupuncture history, the developed ANE should be a fascinating microsensor besides the low price, fast response and user-friendly operation.

Abstract:Chemical communication via neurotransmitters plays a central role in the brain. Nevertheless, techniques for recording neurochemical signals in vivo are still limited. Here, the fabrication of a novel needle field‐effect transistor biosensor based on an acupuncture needle which is capable of real‐time monitoring neurotransmitters released in vivo is shown. Chemical communication via neurotransmitters is central to brain functions. Nevertheless, in vivo real‐time monitoring of neurotransmitters released in the brain, especially the electrochemically inactive molecules, remains a great challenge. In this work, a novel needle field‐effect transistor (FET) microsensor based on an acupuncture needle is proposed, which is demonstrated to be capable of real‐time monitoring dopamine molecules as well as neuropeptide Y in vivo. The FET microstructure is fabricated by successively wrapping an insulating layer and a gold layer on the top of the needle, where the needle and the Au served as the source and drain, respectively. After assembling reduced graphene oxide (RGO) between the source and drain electrodes, the specific aptamer is immobilized on the RGO, making this needle‐FET biosensor highly selective and sensitive to real‐time monitor neurotransmitters released from rat brain, even in a Parkinson's diseases model. Furthermore, the needle‐FET biosensor is applied to detect a variety of targets including hormones, proteins, and nucleic acid. By constructing a FET sensing interface on an acupuncture needle and implanting the sensor in a rat's brain for in vivo detection, this work provides a new sight in the FET domain and further expands the species of real‐time in vivo detection.

Abstract:Serotonin (5-hydroxytryptamine, 5-HT) is a pivotal monoamine neurotransmitter, which is widely distributed in human brain for biological, physical and psychopathological processes. The content of 5-HT can support diagnose of various diseases. To selectively detect 5-HT is very important in clinical medicine. Here, a novel microbiosensor for 5-HT is studied on acupuncture needle. Molecularly imprinted film enwrapped 5-HT was electropolymerized onto bimetallic gold/platinum (Au/Pt) nanoparticles on acupuncture needle microelectrode (ANME). Au/Pt nanostructure exhibited active sites to catalyze the oxidation of 5-HT and bind the generated polymer. 5-HT can be enwrapped by the functional monomer of pyrrole (Py) in the process of electropolymerization with suitably electroactive conformation. Comparing with interfaces of single metal or molecularly imprinted layer, synergistic microbiosensor exhibit better performance for 5-HT. 5-HT can be adsorbed and catalytically oxidized by the imprinted cavities. Under optimized conditions, the peak current linearly increases with the concentration of 5-HT from 0.03 to 500 μM, and a detection limit of 0.0106 μM is obtained. The performance of this microbiosensor is competitive with previous studies. Furthermore, the prepared microbiosensor showed effective application to analyze 5-HT in human serum and urine. Interestingly, the microbiosensor expressed the real-time monitoring ability to 5-HT from stimulated PC12 cells by K+. The microbiosensor also exhibited high selectivity, stability and reproducibility, which is promising in view of the low price, fast response and simple operation.

Abstract:This paper introduces a new technique combining microdialysis with patch clamp to detect the changes in ATP (adenosine triphosphate) concentration in acupoints during acupuncture. The microdialysis probe was implanted into the Zusanli acupoint (ST 36) of adult SD (Sprague Dawley) rats to sample acupoint fluid containing ATP released during acupuncture. Then, the fluid with ATP was delivered in real time to 293 T cells overexpressing P2X3 receptors, with which we could carry out patch clamp experiments. The results showed that changes in membrane currents could reflect changes in the concentration of ATP. Thus, we can successfully detect ATP released in acupoints during acupuncture in real time. This technique provides us with a new way to study the mechanism of acupuncture signal initiation.
Direct Acupuncture of Nitric Oxide by an Electrochemical Microsensor with High Time-Space Resolution

Abstract:Measurement of signal molecule is critically important for understanding living systems. Nitric oxide (NO) is a key redox signal molecule that shows diverse roles in virtually all life forms. However, probing into NO's activities is challenging as NO has restricted lifetime (<10 s) and limited diffusion distance (usually <200 mu m). So, for the direct acupuncture of NO within the time-space resolution, an electrochemical microsensor has been designed and fabricated in this work. Fabrication of the microsensor is achieved by (1) selective assembly of an electrocatalytic transducer, (2) attaching the transducer on carbon fiber electrode, and (3) covered it with a screen layer to reduce signal interference. The fabricated microsensor exhibits high sensitivity (LOD, 13.5 pM), wide detection range (100 pM-5 mu M), and good selectivity. Moreover, studies have revealed that the availability of the sensor for efficient detection of NO is due to the formation of a specific DNA/porphyrin hybrid structure that has synergetic effects on NO electrocatalysis. Therefore, NO release by cells and tissues can be directly and precisely traced, in which we have obtained the release pattern of NO by different cancer cell lines, and have known its dynamics in tumor microenvironment. The fabricated electrocatalytic microsensor may provide a unique and useful tool for the direct assay of NO with high time-space resolution, which promisingly gives a technical solution for the bioassay of NO in living systems.

Abstract:Electrochemical aptamer-based (EAB) sensors are among the only generalizable sensing platforms that resist fouling enough to function directly in bodily fluids and in situ in the living body. Already, these sensors have enabled the real-time measurement of a range of small molecule targets in vivo, including pharmaceuticals and drugs of abuse. EAB sensors consist of an electrode-attached, redox reporter-labeled aptamer that binds to its target species. Target binding causes a shift in electron-transfer rate between the redox reporter and electrode surface, easily detected by a range of electrochemical techniques. Here, we explore the application of EAB sensors to minimally-invasive molecular monitoring. Specifically, we adapt the platform to a microneedle device format. Microneedles can provide direct access to the skin’s interstitial space – a bodily compartment that holds valuable information ranging from drug distribution and immune status. And because microneedles can penetrate into the top layers of the skin with minimal nerve endings, they can avoid eliciting notable pain responses. In this work, we demonstrate the substantial miniaturization of the sensing platform into electrodes thinner than a human hair, and then integrate them with ultra-fine, hollow microneedles. To ensure needle sterilizability and sharpness while avoiding complex fabrication, we utilize commercial off the shelf stainless steel microneedles. Using an EAB sensor responding to the high-toxicity small molecule drug, vancomycin, as a model system, we achieve quantitative measurements directly in body temperature, undiluted whole blood. Building on this milestone, we explore the application of the sensors when inserted into porcine skin. In porcine skin (a common proxy for human skin), we observe facile microneedle penetration and stable electrochemical signaling. Because of the individual addressability of the sensing electrodes and the generalizability of the aptamer platform, this technology is capable of multiplexed, simultaneous detection of many different molecular targets. Thus, microneedle-enabled aptamer-based sensors offer a promising route forward for non-invasive physiological monitoring – applicable to therapeutic drug monitoring, pain-free biomarker detection, and disease management.
Platinum Black/Gold Nanoparticles/Polyaniline Modified Electrochemical Microneedle Sensors for Continuous In Vivo Monitoring of pH Value

Abstract:Continuous in vivo monitoring (CIVM) of pH value is essential for personalized medicine, as many diseases are closely related to acid-base imbalances. However, conventional pH meters are limited in their ability to perform CIVM due to excessive blood consumption, large device volume, frequent calibration, and inadequate real-time monitoring. There is thus an urgent need for a portable method for CIVM of pH value. To address this need, we propose a minimally invasive, continuous monitoring solution in the form of an implantable pH microneedle sensor (MNS) in this study. The MNS is based on the integration of an acupuncture needle (AN) and a Ag/AgCl reference electrode. We fabricate the sensor by electrochemically depositing platinum black and gold nanoparticles onto the AN and further modifying it with polyaniline to increase its sensitivity to hydrogen ions. The pH value is obtained by calculating the open circuit voltage between the modified AN and the reference electrode. The resulting MNS demonstrates excellent selectivity and a high nernstian response to pH (-57.4 mV per pH) over a broad range (pH = 4.0 to pH = 9.0). Both in vitro and in vivo experiments have verified the performance of the sensor, showcasing its potential for biomedical research and clinical practice. The MNS provides an alternative to conventional pH meters, offering a less invasive and more convenient way to perform CIVM of pH value. Moreover, this electrochemical implantable sensor based on AN and silver wires provides a simple and sensitive method for continuous in vivo detection of other biomarkers.
Studies of Selective Ion-Sensitive Microelectrode Technique for the Detection of Neurotransmitter in Vivo

Abstract:The neurotransmitters such as acetylcholine/choline (Ach/ch), serotonin and dopamine play very important roles in the course of neurological diseases. Traditional methods used in the detection of these neurotransmitters have enzymatic and radiochemical methods, fluorescence spectroscopy, gas chromatography, high performance liquid chromatography, thin layer chromatography etc. But these methods cannot be used in the direct and continuous detection of neurotransmitter activity in a single cell. Here, a new ion-selective microelectrode technique has been developed. The advantage of the ion-sensitive microelectrode technique is in its very high spatial resolution, by which some interesting ion activity inside and outside the living cells can be monitored continuously for a relatively long time without great damage to the living cells. All these virtues of this technique force the authors to develop some new ion-sensitive microelectrode especially for acetylcholine/choline, serotonin, histamine and dopamine. The results obtained showed that the technique could be used successfully in the detection of neurotransmitters at the cellular level, and the technique has promising prospects in the field of neuroscience
In vivo monitoring of calcium ions in rat cerebrospinal fluid using an all-solid-state acupuncture needle based potentiometric microelectrode

Abstract:Acupuncture needles are regarded as ideal materiel for the development of microelectrodes for in vivo sensing. In this work, an all-solid-state ion-selective microelectrode (ISμE) has been developed by coating a calcium ion-selective membrane on an acupuncture needle tip with a diameter of less than 80 μm, which is modified with poly(3,4-ethylenedioxythiophene)-poly(sodium 4-styrenesulfonate) as solid contact. The proposed Ca2+-ISμE shows a Nernstian response toward Ca2+ in the range from 1.0 × 10-6 to 3.1 × 10-3 M with a slope of 30.8 ± 0.9 mV/decade (R2 = 0.999), and the detection limit is 1.2 × 10-7 M. The Ca2+-ISμE has been used for in vivo monitoring of the calcium changes in rat cerebrospinal fluid (CSF) under the injury of spinal cord transection. The results demonstrate that the calcium concentration in CSF increases sharply from the normal level of 20.6 ± 1.72 μM (n = 3) to 133.2 ± 7.63 μM (n = 3) with a severe fluctuation after spinal cord damage. Thus, the proposed Ca2+-ISμE is available for in vivo monitoring of calcium ions with high temporal resolution and flexibility. The detection system can be extended to measure other ions in CSF by changing different ion-selective membranes.
An aptamer-integrated conductive microneedle biosensor for real-time transdermal cortisol monitoring

Abstract:Cortisol, known as the 'stress hormone', plays a crucial role in the regulation of metabolism, the promotion of secondary sex characteristics, and the maintenance of normal biological functions. A conductive microneedle (MN)-based electrochemical biosensor modified with dendritic Au nanoparticles (AuNPs) was developed for real-time monitoring of cortisol levels in interstitial fluid (ISF) to address the current challenges of time-consuming and laborious cortisol detection. The dendritic AuNPs provide signal amplification due to their high surface area and abundance of active sites, and the amine group modification of the cortisol aptamer facilitates its connection to an Au MN electrode with surface-modified dendritic AuNPs. Through parameters optimization and utilization of differential pulse voltammetry (DPV), the MN biosensor exhibits a wide detection range (1–1000 nM) covering normal human cortisol levels. Notably, it demonstrates low computational detection limits of 0.17 nM and 0.22 nM in phosphate buffered saline (PBS) and simulated ISF respectively, along with exceptional stability, selectivity, and repeatability. Furthermore, the successful detection of cortisol in the healthy volunteers' ISF reveals a significant circadian rhythm and a correlation between cortisol levels in the ISF and blood. Together these findings indicate the promise of aptamer-integrated microneedle biosensors for facilitating simple, sensitive, and disposable cortisol assays.

The Real-Time Detection of Acupuncture-Induced Extracellular ATP Mobilization in Acupoints and Exploration of Its Role in Acupuncture Analgesia.

Abstract:Our and in vitro studies had confirmed that mechanosensitive ATP release and accumulation in acupoints was elicited by acupuncture (AP), which might be a pivotal step for triggering AP analgesia. But to date, the dynamics of extracellular ATP (eATP) in the interstitial space during AP process was poorly known, mainly due to the low temporal resolution of the current detection approach. This study attempted to capture rapid eATP signals in vivo in the process of needling, and further explored the role of this eATP mobilization in initiating AP analgesic effect. Ipsilateral 20-min needling was applied on Zusanli acupoint (ST36) of complete Freund's adjuvant (CFA)-induced ankle arthritis rats. Pain thresholds were assessed in injured-side hindpaws. eATP in the interstitial space was microdialyzed and real-time quantified by luciferin-luciferase assay at 1-min interval with the aid of the microfluid chip. We revealed in behavioral tests that modulation of eATP levels in ST36 influenced AP analgesic effect on ankle arthritis. A transient eATP accumulation was induced by needling that started to mobilize at 4 min, climbed to the peak of 11.21 nM within 3.25 min and gradually recovered. Such AP-induced eATP mobilization was significantly impacted by ankle inflammation, needling depth, needle manipulation, and the presence of local ecto-nucleotidases. This work reveals that needling elicits a transient eATP mobilization in acupoints, which contributes to initiating AP analgesia. This study will help us better understand the peripheral mechanism of AP analgesia and guide clinicians to optimize the needle manipulations to improve AP efficacy.
