Abstract:The effect of fungal infection on the reproductive potential of two-spotted spider mite, Tetranychus urticae , was evaluated as part of the full biocontrol potential of three entomopathogenic fungi by modeling of fecundity probability. Female mites (≤2-day-old) on leaves were exposed to the sprays of Beauveria bassiana , Paecilomyces fumosoroseus and Metarhizium anisopliae at the concentrations of 1.13 × 10 3 , 1.55 × 10 3 and 0.95 × 10 3 deposited conidia mm −2 and then individually reared at 25°C and 12:12 L:D for oviposition. Mite mortalities 10 days after spraying were 73.1, 75.4 and 67.9% in the fungal treatments versus 15.5% in control. On average, females infected by the three fungal species survived 5.8, 6.2 and 6.3 days, and laid 3.1, 4.0 and 4.0 eggs per capita, respectively. These were 3–4 fold lower than the control fecundity at 12.3. The cumulative probabilities [ P ( m ≤ N )] for the counts of infected and non-infected (control) females laying m eggs per capita ( m ≤ N ) during 10 days fit very well the equation P ( m ≤ N ) = 1/[1 + exp( a + bm )] ( r 2 ≥ 0.98), yielding a solution to the probability for the female mites to achieve a specific fecundity { P ( m ≤ N )− P [ m ≤ ( N − 1)]}. Consequently, the infected mites had 71–78% chance to lay ≤5 eggs per capita but only 5–8% to deposit >10 eggs despite some variation among the tested fungi. In contrast, the chances for the non-infected mites to achieve the low and high fecundities were 23 and 55%. The fitted probabilities provide a full coverage of the fecundity potential of infected versus non-infected mites and are more informative than the mean fecundities.

Abstract:Cyetpyrafen belonging to mitochondrial electron transport inhibitors of complex II (METI II) has been widely applied to manage pest mites in China. To investigate the adaption of Tetranychus urticae in the evolution of cyetpyrafen resistance, a study of cross resistance, mode of inheritance and fitness comparison of resistance using indoor cyetpyrafen resistant strain (resistance ratio, RR > 2, 000-fold) was executed. Cyet-R showed serious cross resistance to cyenopyrafen (>2500-fold) and cyflumetofen (~190-fold). The number of resistant genes was evaluated via chi-square (χ2) test and the concentration-response curve regarding goodness-of-fit between observed and the expected mortality. The LC50s of F1RS (Cyet-R♀ × Tu-YN♂) and F1SR (Tu-YN♀ × Cyet-R♂) were 3126.30 mg/L and 2743.97 mg/L, respectively, without significance, suggesting autosomal inheritance. The degree of dominance (D) values of F1RS and F1SR ranged between 0 and 1, revealing an incompletely dominant inheritance in the tested population of Tetranychus urticae. Plots of concentration-response data for the orthogonal backcross and reverse backcross progenies showed a significant deviation from the expected lines, pointing out a polygenic inheritance. Besides, lifetable analysis showed a fitness advantage of Cyet-R with a significantly decreased adult preadult period and significantly increased total fecundity. This study suggested that cyetpyrafen resistance against T. urticae was inherited as autosomal, incompletely dominant and multigenetic and characterized with serious cross resistance and fitness advantage. Therefore, rational application and preventive strategy should be considered to sustain the efficacy of cyetpyrafen against T. urticae.

Abstract:Green lacewing, Chrysoperla carnea (Stephens) is a generalist predator used as a biological control agent in agro ecosystems. In order to use chemical and biological control in an integrated way, it is advantageous to know about natural enemy resistance response to a selected chemical. To determine C . carnea spirotetramat resistance potential, a population collected from the field was selected in the laboratory. Then we determined how spirotetramat resistance was inherited and how much it impacts the fitness of C . carnea . After eighteen selections with spirotetramat, the selected population (Spiro-Sel) of C . carnea had a 47-fold of resistance when compared to an UNSEL population. Inheritance results showed that spirotetramat resistance was inherited as an autosomal, incompletely dominant and polygenic trait. The values of effective dominance decreased from 0.87 (incomplete dominant) to 0.00 (complete recessive) as the concentration of spirotetramat increased from 625 mg/L to 10000 mg/L. The Spiro-Sel strain had no cross resistance to chlorfenapyr (1.10-fold), deltamethrin (1.26-fold) and chlorpyrifos (1.27-fold). After 7 generations without selection pressure resistance to all experimental insecticides in the Spiro-Sel strain was stable. Fitness data of the Spiro-Sel, Cross A, Cross B, UNSEL and susceptible strains of C . carnea showed that spirotetramat resistance increased the fitness of the selected green lacewing population. Life history parameters like fecundity, net reproductive rate, and relative fitness of the Spiro-Sel strain significantly increased when compared to the susceptible or unselected strains of C . carnea . These findings show that C . carnea is a perfect candidate for integrated pest management (IPM) programmes that combine biological control methods with selective pesticide applications to manage a variety of insect pests. Additionally, it would reduce the possibility of pests developing pesticide resistance despite repeated applications. It would be an excellent choice for widespread releases and be effective in most spray programs.

[Cystic lymphangioma of the liver].

Abstract:The authors presented a rare condition of cystic lymphangion of the liver in a female child of 14 months, comprising the whole left lobe leading to compression of the surrounding intestinal organs and partly of some chest organs. By a transversal supraumbilical laparotomy, the cystic tumor of the left liver lobe was identified, and a left lobectomy of the liver was made by removing all cystic formations. It was hystologically acknowledged that it was about a cystic lymphangiom of the liver. Postoperatively the course ran regularly and with no complications. The control examinations showed that the function of the liver was not damaged. The cystic changes of the liver are most frequently in the framework of the polycystic diseases when even other organs are invaded. All cysts of the liver can be divided into congenital cysts or acquired. The presented case was most probably about a congenital anomaly, which was getting augmented gradually and resulted to a compression of the surrounding organs.
[Effect of color mutation on the function of the adrenal cortex in chronic feed stress in American mink].

Abstract:Effect of color mutations, headlund and aleutian, on the adrenocortical function was studied in American mink under stress conditions. The adrenocortical function was analysed in the course of gland's incubation in vitro. Natural environmental factors (sharp fall of temperature) and artificially limited feed served as stress factors. It was shown that minks heterozygous for color mutant alleles studied were more resistant to the stressors. This is associated with genetically mediated peculiarities of the action of hypophysial adrenocortical system which is characterized by more adequate reactivity towards stimuli acting. Thus, the effect of heterosis in minks heterozygous for color alleles may well be caused by their increased resistance to stress.

Abstract:Ketoenol acaricides have been widely used to control Tetranychus urticae populations across the world. However, control failure due to resistance development is an increasing concern. To sustain resistance management, it is therefore important to understand the molecular mechanisms underlying resistance, as well as understand the level of cross-resistance they convey between different ketoenol acaricides, such as spirodiclofen or spiromesifen. A T. urticae population with moderate levels of resistance to ketoenols was collected from a carnation greenhouse and further selected in the laboratory with spirodiclofen and spiromesifen, separately, until high levels of resistance were achieved. Synergism assays indicated the involvement of P450 monooxygenases and, to a lesser extent, carboxyl/cholinesterases in resistance. Genome-wide gene expression analysis of ketoenol-selected populations compared to the initial field-collected population and a susceptible reference laboratory population further supported the hypothesis of P450-mediated resistance to ketoenols. In addition to metabolic resistance, target-site resistance was also investigated, but no amino acid substitutions in the carboxyl-transferase (CT) domain of the acetyl-CoA carboxylase (ACCase), the target-site of ketoenols, were found in the studied populations. However, increased expression of ACCase was found in the spiromesifen-selected, but not in the spirodiclofen-selected population. Finally, changes in resistance levels of some commonly used acaricides were identified after selection with spiromesifen or spirodiclofen.

Abstract:The physiological changes in Tetranychus urticae selected with spiromesifen+abamectin (S+A) mixture formulation were demonstrated by biochemical and molecular methods. The susceptible (GSS) population of T. urticae, was selected with S+A and made resistant with increasing doses of the mixture formulation. The resistant population was divided into two, and selection pressure was terminated in one population which is named IR (Interrupt resistant). In the other population, the selection with S+A was continued and the population named IR2. Activities of some detoxification enzymes (esterase, glutathione-S-transferase (GST) and cytochrome P450) were investigated in the GSS and IR populations. The activity of esterase, GST and P450 enzymes were increased by 1.77, 2.57 and 2.58-fold, respectively, in the IR population compared to the GSS population. Also, investigation of synergistic effects with esterase inhibitor (TTP), GST inhibitor (DEM) and P450 inhibitor (PBO) in this population revealed that all three synergists showed significant synergistic effects in IR population. In molecular studies, when the population was screened for the presence of two previously identified glutamate channel mutations (G314D and G326E) by qPCR with TaqMan probes no mutations could be detected in glutamate channels. and the CTD domain of acetyl coenzyme-A carboxylase (ACCase). Furthermore, possible genetic mutations in the biotin carboxylase domain (BCD) and carboxyl transferase domain (CTD) of the ACCase target sites were determined by sequencing. Although a single amino acid mutation G37D in the BCD of the ACCase gene of the IR and IR2 populations was detected, its association with spiromesifen resistance was not confirmed. The results indicated that increased detoxification and possible target site mutation may be responsible for the S+A resistance in the IR and IR2 populations.

Abstract:Musca domestica L., a pest of animals and humans, has developed resistance to alpha-cypermethrin, a pyrethroid insecticide commonly used to control medically important pests in many countries, including Saudi Arabia. We investigated the mechanism underlying the development of alpha-cypermethrin resistance and life history characteristics of alpha-cypermethrin-susceptible (Alpha-SS) and alpha-cypermethrin-resistant (Alpha-RS) M. domestica using the age-stage, two-sex life table theory, which is crucial for developing a future rational management strategy and minimizing the negative effects of alpha-cypermethrin on the environment. Our results showed that Alpha-RS M. domestica had a 405.93-fold increase in resistance to alpha-cypermethrin relative to Alpha-SS M. domestica. This increase in the resistance toward insecticide was attributed to metabolic enzymes, such as glutathione S-transferases, specific esterases, and cytochrome P450 monooxygenases. Furthermore, Alpha-RS M. domestica exhibited lower relative fitness (0.50), longevity, survival rate, life expectancy, reproductive values, intrinsic rate of increase, net reproductive rate, fecundity, maternity, and finite rate of increase, along with shorter larval, female preadult, and adult durations than Alpha-SS M. domestica, indicating fitness costs associated with most parameters. However, no significant differences were found between the strains in the following parameters: egg, pupa, and male preadult durations; adult preoviposition, total preoviposition, and oviposition periods; female ratio; and total generation time. Additionally, Alpha-RS M. domestica had a markedly lower intrinsic rate of increase, net reproductive rate, and finite rate of increase than Alpha-SS M. domestica. The results of this study suggest that alpha-cypermethrin resistance may lead to dominant fitness costs in M. domestica. Overall, these findings will aid in the development of rational control strategies for M. domestica as well as help to reduce pesticide pollution.

Abstract:Tetranychus urticae (Koch) (Acari: Tetrancyhidae) is one of the most economically important pests among spider mites, due to the number of hosts that it harms in agricultural fields and greenhouses, and to its remarkable ability to develop resistance to chemicals applied in a short time. Pyrethroids are among the most widely used insecticides to control this pest. In this study, the biochemical mechanisms and inheritance mode of lambda cyhalothrin and bifenthrin resistance in T. urticae were investigated. A greenhouse population (BEYDOM) of T. urticae was selected for eight cycles in the laboratory with lambda cyhalothrin and bifenthrin by a spray tower-Petri dish method using the LC90 of a standard susceptible population (GSS). After eight selection cycles, the BEYDOM population was found 18.24-fold resistant to lambda cyhalothrin and 74.23-fold resistant to bifenthrin compared with the GSS based on LC50 values. Reciprocal and backcross studies revealed that the inheritance of lambda cyhalothrin resistance was sex-linked, incompletely dominant, and was probably under the control of more than one gene at lower doses and a single gene at higher doses, while inheritance to bifenthrin resistance was autosomal and incompletely recessive and may be controlled by one gene. Biochemical and synergism experiments suggested that monooxygenases and carboxylesterase enzymes play an essential role in the resistance to these pyrethroids, whereas glutathione S-transferases were not found to have a significant effect. These results provide valuable information in terms of designing an effective resistance management strategy to retard or avoid the development of resistant T. urticae populations.

Abstract:Two spotted spider mite Tetranychus urticae Koch is an economically serious pest posing threat to major vegetable crops. Roving survey in and around Coimbatore region revealed that farmers do not target mites with acaricides instead they use higher dose of insecticides at frequent intervals which results in development of resistance. The bioassay results revealed that fenpropathrin (2.07 to 6.86-folds) and fenazaquin (2.74 to 7.13-folds) exhibit higher susceptibility, whereas diafenthiuron (5.35 to 12.25-folds) revealed a low to moderate level of resistance. The propargite (43.80 to 60.63-folds) and chlorfenapyr (61.01 to 75.10-folds) exhibited high resistance, followed by spiromesifen (222.28 to 300.26-folds) and buprofezin (382.60 to 417.87-folds), with extremely high level of resistance. The higher specific activity of GST (4.54-folds), MFO (10.06-folds) and CarE (15.06-folds) in Puthupalayam population suggested the role of biochemical resistance. A significant positive correlation was observed between diafenthiuron and CarE activity (r = 0.981*), fenpropathrin and MFO activity (r = 0.964*).

Abstract:The Eastern Cooperative Oncology Group evaluated cyclophosphamide 600 mg/m2 intravenously daily × 4 (total dose each cycle 2400 mg/m2) as an aggressive approach to the treatment of patients with advanced multiple myeloma. The overall objective response rate is 43%. This includes a 38% response rate for all previously treated patients and a 29% response rate for patients refractory to prior therapy with cyclophosphamide. The objective response duration was 3 months and the survival of responding patients 9 months. A subjective response rate of 63% was observed, characterized by effective pain relief and improved performance. Sixty‐nine percent of patients experienced leukocyte cell nadirs < 500/mm2 with a mean time to marrow recovery of 17 days. Thrombocytopenia was less severe but required platelet transfusion in 43% of patients. Bone marrow toxicity was encountered in all patients, and death in aplasia is a significant risk. Strict adherence to entry criteria, and a systematic plan for hospitalization for antibiotic and blood component support is required for treatment with this regimen.
Effect of Chlorantraniliprole on Life History Traits and Relative Fitness of Laboratory-Selected Resistant and Susceptible Populations of Spodoptera litura (Lepidoptera: Noctuidae)

Abstract:Insecticides are widely used to control pests and improve agricultural yield. The use of indiscriminate amounts and persistent pesticides has not only resulted in insect pest resistance challenges but has also harmed non-target organisms and contaminated the environment. We assessed the cost-effectiveness of Spodoptera litura (Fabricius) resistance to chlorantraniliprole (CTPR) as part of insect resistance control programs. S. litura larvae were collected from the fields and treated with chlorantraniliprole for 15 generations. Compared to the unselected population (Unsel-Lab), the chlorantraniliprole-selected population (CTPR-Sel) of S. litura exhibited a resistance level of 98.23 times. The findings demonstrated that the chlorantraniliprole-resistant genotype had lower egg-to-adult survivability, longer egg-to-adult developing times, and lower fecundity than the chlorantraniliprole-susceptible genotype. Compared to the Unsel-Lab population, the CTPR-Sel population of S. litura had a poorer relative fitness (0.44) at LC30 and shorter male/female longevity. Demographic parameters, including net reproductive rate (R0) and intrinsic (rm) and finite rate of increase (λ), were lower in the CTPR-Sel strain than in Unsel-Lab S. litura. These findings demonstrate that chlorantraniliprole resistance in S. litura has fitness costs at the individual and population levels, implying that removing the selecting agent from the environment might result in less resistance and opportunities for susceptibility restoration. As a result, the current work could help to determine effective management strategies to prevent chlorantraniliprole resistance in S. litura.

Abstract:Fitness costs associated with resistance genes expressed in the absence of insecticides affect the evolution of insecticide resistance and the outcome of resistance management programmes. However, measurements of fitness costs may not be straightforward as they vary with environmental conditions. The diamondback moth, Plutella xylostella L. (Lepidoptera: Plutellidae), has developed resistance to spinosad, the first insecticide of the Naturalyte class, after only a few years of field application of this product. In this study, we compared the performance of two homogenous strains of P. xylostella , one susceptible (SS) and the other resistant (RR) to spinosad at an unfavourable, low natural temperature regime, a favourable median-fluctuating temperature regime and an unfavourable high-fluctuating temperature regime. The RR strain showed only marginal fitness cost at the median temperature regime. At the low temperature regime, the RR strain failed to produce any viable offspring, while the SS strain achieved positive population growth. At the high temperature regime, the RR strain showed a 33% decrease in intrinsic rate of increase compared to the SS strain. The results demonstrate that fitness costs of resistance to spinosad are temperature-dependent, increasing in scale at unfavourably low and high temperatures; costs were particularly high at low temperatures. Suggestions for designing effective management programmes are made to delay or avoid development of resistance to spinosad by P. xylostella under different temperature conditions.

Abstract:Event-related potentials were recorded to examine the electrophysiologic correlates of the evaluation of possible answers provided during a semantic, a numeric, and a figure inference task. We examined two conditions: the probe stimulus given was what participants initially generated (Correct answer) or it was unexpected and incorrect (Unexpected Incorrect answer). Results showed that Unexpected Incorrect answers elicited a more negative ERP deflection (N320) than did Correct answers between 300–400 ms under all inference tasks. The generator of N320 effect was localized near the anterior cingulate cortex. The N320 effect, therefore, reflects an expectation violation while participants performed semantic, numeric, or figure inference task.

Abstract:Two-spotted spider mite (TSSM) Tetranychus urticae (Koch) is an important agricultural pest that causes considerable yield losses to over 150 field and greenhouse crops. Mitochondrial electron transport inhibitors (METI) acaricides are commonly used to control mite species in commercial Canadian greenhouses. Development of resistance to METIs in TSSM populations have been reported worldwide, but not until recently in Canada. The objectives of this study were to: 1) monitor the acaricide-susceptibility in greenhouse TSSM populations, and 2) investigate the resistance to pyridaben, a METI acaricide, in greenhouse resistant and pyridaben-selected (SRS) mite strains. The increased mortality to the pyridaben sub-lethal concentration (LC30) when SRS mites were exposed to piperonyl butoxide (PBO), a general cytochrome P450 monooxygenase inhibitor, and higher P450 activity compared to the greenhouse strain (RS) mites, indicated that P450s may be at least partially responsible for the resistance. The molecular mechanisms of target site insensitivity-mediated resistance in the pyridaben resistant strain of TSSM were investigated by comparing the DNA sequence of NADH dehydrogenase subunits TYKY and PSST, NADH-ubiquinone oxidoreductase chain 1 and 5 (ND1, ND5) and the NADH-ubiquinone oxidoreductase subunit 49 kDa from SRS to the reference strain (SS) and RS. Despite a number of nucleotide substitutions, none correlated with the pyridaben resistance. Understanding the underlying mechanisms of TSSM adaptation to acaricides is an essential part of resistance management strategy in any IPM program. The findings of this study will encourage growers to apply acaricides with different modes of action to reduce the rate at which acaricide resistance will occur in greenhouse TSSM populations.

Abstract:Abstract Resistance to permethrin has been reported in Pakistani strains of Musca domestica . The present study explored the performance of biological traits and analyzed life tables to determine whether there is any detrimental effect of permethrin resistance on the fitness of permethrin-resistant strains [an isogenic resistant strain (Perm-R) and a field strain (Perm-F)] compared to a susceptible strain (Perm-S). Perm-R and Perm-F exhibited 233.93- and 6.87-fold resistance to permethrin, respectively. Life table analyses revealed that the Perm-R strain had a significantly shorter preadult duration, longer longevity, shorter preoviposition period, higher fecundity, finite rate of increase, intrinsic rate of increase, net reproductive rate and a shorter mean generation time, followed by the Perm-F strain when compared to the Perm-S strain. Data of the performance of biological traits reveled that permethrin resistance strains had a better fit than that of the Perm-S strain. The enhanced fitness of resistant strains of M. domestica may accelerate resistance development to permethrin and other pyrethroids in Pakistan. Some possible measures to manage M. domestica and permethrin resistance in situations of fitness advantage are discussed.
