Abstract:BACKGROUND: Salinity is the main abiotic stress that affects seed germination, plant growth and crop production. Plant growth begins with seed germination, which is closely linked to crop development and final yields. Morus alba L. is a well-known saline-alkaline tree with economic value in China, and the most prominent method of expanding mulberry tree populations is seed propagation. Understanding the molecular mechanism of Morus alba L. salt tolerance is crucial for identifying salt-tolerant proteins in seed germination. Here, we explored the response mechanism of mulberry seed germination to salt stress at physiological and protein omics levels.METHODS: Tandem mass tag (TMT)-based proteomic profiling of Morus alba L. seeds germinated under 50 mM and 100 mM NaCl treatment for 14 days was performed, and the proteomic findings were validated through parallel reaction monitoring (PRM).RESULTS: Physiological data showed that salt stress inhibited the germination rate and radicle length of mulberry seeds, decreased the malondialdehyde (MDA) content and significantly increased superoxide dismutase (SOD), peroxidase (POD), and catalase (CAT) activities. Then, a TMT marker technique was used to analyze the protein groups in mulberry seeds with two salt treatment stages, and 76,544 unique peptides were detected. After removing duplicate proteins, 7717 proteins were identified according to TMT data, and 143 (50 mM NaCl) and 540 (100 mM NaCl) differentially abundant proteins (DAPs) were screened out. Compared with the control, in the 50 mM NaCl solution, 61 and 82 DAPs were upregulated and downregulated, respectively, and in the 100 mM NaCl solution, 222 and 318 DAPs were upregulated and downregulated, respectively. Furthermore, 113 DAPs were copresent in the 50 mM and 100 mM NaCl treatments, of which 43 were upregulated and 70 were downregulated. Gene Ontology (GO) annotation and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis revealed that the DAPs induced by salt stress during mulberry seed germination were mainly involved in photosynthesis, carotenoid biosynthesis and phytohormone signaling. Finally, PRM verified five differentially expressed proteins, which demonstrated the reliability of TMT in analyzing protein groups.CONCLUSIONS: Our research provides valuable insights to further study the overall mechanism of salt stress responses and salt tolerance of mulberry and other plants.

Abstract:Twenty-five promising salt-tolerant mulberry germplasms from different Morus species were evaluated for growth under varying salinity levels (10.20-25.60 dS m-1) typical of the coastal regions in South 24 Parganas, West Bengal. Evaluations were conducted using in vitro axillary bud culture and field experiments under natural conditions to identify superior salt-tolerant germplasms. Soil sample analysis revealed significant variation in salinity levels (34.37-17.09 dS m-1) across different areas, with the highest in Kultali and the lowest in Canning I and II. Among the 25 germplasms, 6 were identified as highly salt-tolerant, 6 as moderately high salt-tolerant, 11 as salt-tolerant, and 2 as salt-sensitive. Survivability rate and root length were found to have the highest correlation with salt tolerance during early development. The six highly salt-tolerant germplasms, including English Black, Kolitha-3, C776, Rotundiloba, BC259, and S1 were further tested in field trials. English Black showed the highest survivability rate of 69.2 % in soil salinity of 18-20 dS m-1. Results from in vitro and field trials were consistent, with a strong positive correlation between survivability rate and root length. This study establishes an effective method for evaluating salt tolerance in mulberry, providing a foundation for more efficient assessments.
Morphological, Physiological, and Biochemical Composition of Mulberry (Morus spp.) under Drought Stress

Abstract:Drought is the most important environmental factor inhibiting plant photosynthesis. In this study, the morphological characteristics, biomass allocation, and physiological and biochemical characteristics of four potted mulberry plants under drought stress were analyzed. The study revealed the drought tolerance differences of four mulberry potted seedlings in semi-arid sandy areas of China. Combined with the results of two−way ANOVA, under normal growth conditions, Shensang No. 1 and Ji'an grew well and produced higher benefits, which was attributed to their larger leaf areas, biomass, and total Chl contents, and there were significant differences between their other traits (p < 0.05). Drought stress led to a decrease in the photosynthetic capacity of the mulberry leaves, and the drought resistance capabilities of the four mulberry trees were different. Among the trees, Aerxiang and Fujia were less affected by drought, and their cultivation in a naturally arid environment was able to achieve certain drought resistance effects. The branch length, total leaf area, and specific leaf area were significantly differently correlated with the biomass components' dry leaf weight, dry branch weight, dry root weight, total biomass, and root−to−crown ratio (p < 0.05), and there was also a significant positive correlation with the photosynthetic fluorescence parameters GS, PIABS, ABS/RC, and TRo/RC and the biochemical parameters NSC (p < 0.05). Studies have shown that plant biomass and physiological and biochemical characteristics jointly affect plant growth. Our research results will help in the screening of mulberry trees, providing data support for the strategic planning of subsequent breeding, and maximizing the quality and resource benefits of mulberry trees.

Abstract:Mulberry is an economically important crop for sericulture in China. Mulberry plantations are shifting inland, where they face high salinity. Arbuscular mycorrhizal fungi (AMF) reportedly enhance mulberry's tolerance to salinity. Here, we assessed if additional adaptive advantages against salinity are provided by sex differences beyond those provided by mycorrhizal symbiosis. In a pot experiment, male and monoecious plants were exposed to three salinity regimes (0, 50, and 200 mM NaCl) and two mycorrhiza-suppressed conditions (with or without benomyl application) for more than 16 months. We noticed that salinity alone significantly decreased the mycorrhizal colonization rate, salinity tolerance, K + concentrations, and the ionic ratios of all plants. Mycorrhizal association mildly ameliorated the salt-induced detrimental effects, especially for monoecious plants, and sex-specific responses were observed. Meanwhile, both sexes had adopted different strategies to enhance their salinity resistance. Briefly, mycorrhizal monoecious plants exhibited a higher net photosynthetic rate and lower translocation of Na + from root to shoot compared with mycorrhizal males under saline conditions. Their salt tolerance was probably due to the Ca 2+ /Na + in roots. In comparison, male plants exhibited lower Na + acquisition, more Na + translocated from root to shoot, higher root biomass allocation, and higher N concentrations under harsh saline conditions, and their salt tolerance was mainly related to the K + /Na + in their shoots. In conclusion, our results highlight that AMF could be a promising candidate for improving plant performance under highest salinity, especially for monoecious plants. Cultivators must be mindful of applying fungicides, such as benomyl, in saline areas.

New Genes Identified as Modulating Salt Tolerance in Maize Seedlings Using the Combination of Transcriptome Analysis and BSA

Abstract:(1) Background: Salt stress is an abiotic factor that limits maize yield and quality. A highly salt-tolerance inbred AS5 and a salt-sensitive inbred NX420 collected from Ningxia Province, China, were used to identify new genes for modulating salt resistance in maize. (2) Methods: To understand the different molecular bases of salt tolerance in AS5 and NX420, we performed BSA-seq using an F2 population for two extreme bulks derived from the cross between AS5 and NX420. Transcriptomic analysis was also conducted for AS5 and NX420 at the seedling stage after treatment with 150 mM of NaCl for 14 days. (3) Results: AS5 had a higher biomass and lower Na+ content than NX420 in the seedling stage after treatment with 150 mM NaCl for 14 days. One hundred and six candidate regions for salt tolerance were mapped on all of the chromosomes through BSA-seq using F2 in an extreme population. Based on the polymorphisms identified between both parents, we detected 77 genes. A large number of differentially expressed genes (DEGs) at the seedling stage under salt stress between these two inbred lines were detected using transcriptome sequencing. GO analysis indicated that 925 and 686 genes were significantly enriched in the integral component of the membrane of AS5 and NX420, respectively. Among these results, two and four DEGs were identified as overlapping in these two inbred lines using BSA-seq and transcriptomic analysis, respectively. Two genes (Zm00001d053925 and Zm00001d037181) were detected in both AS5 and NX420; the transcription level of Zm00001d053925 was induced to be significantly higher in AS5 than in NX420 (41.99 times versus 6.06 times after 150 mM of NaCl treatment for 48 h), while the expression of Zm00001d037181 showed no significant difference upon salt treatment in both lines. The functional annotation of the new candidate genes showed that it was an unknown function protein. (4) Conclusions: Zm00001d053925 is a new functional gene responding to salt stress in the seedling stage, which provides an important genetic resource for salt-tolerant maize breeding.

Abstract:Seed propagation is the main method of mulberry expansion in China, an important economic forest species. However, seed germination is the most sensitive stage to various abiotic stresses, especially salinity stress. To reveal the molecular regulatory mechanism of mulberry seed germination under salt stress, flavonoid metabolomics and transcriptomics analyses were performed on mulberry seeds germinated under 50 and 100 mmol/L NaCl stress. Analysis of the flavonoid metabolome revealed that a total of 145 differential flavonoid metabolites (DFMs) were classified into 9 groups, 40 flavonols, 32 flavones, 16 chalcones and 14 flavanones. Among them, 61.4% (89) of the DFMs accumulated continuously with increasing salt concentration, reaching the highest level at a 100 mmol/L salt concentration; these DFMs included quercetin-3-O-glucoside (isoquercitrin), kaempferol (3,5,7,4'-tetrahydroxyflavone), quercetin-7-O-glucoside, taxifolin (dihydroquercetin) and apigenin (4',5,7-trihydroxyflavone), indicating that these flavonoids may be key metabolites involved in the response to salt stress. Transcriptional analysis identified a total of 3055 differentially expressed genes (DEGs), most of which were enriched in flavonoid biosynthesis (ko00941), phenylpropanoid biosynthesis (ko00940) and biosynthesis of secondary metabolites (ko01110). Combined analysis of flavonoid metabolomic and transcriptomic data indicated that phenylalanine ammonia-lyase ( PAL ), 4-coumarate-CoA ligase ( 4CL ), chalcone synthase ( CHS ), flavonol synthase ( FLS ), bifunctional dihydroflavonol 4-reductase/flavanone 4-reductase ( DFR ) and anthocyanidin reductase ( ANR ) were the key genes involved in flavonoid accumulation during mulberry seed germination under 50 and 100 mmol/L NaCl stress. In addition, three transcription factors, MYB, bHLH and NAC, were involved in the regulation of flavonoid accumulation under salt stress. The results of quantitative real-time PCR (qRT‒PCR) validation showed that the expression levels of 11 DEGs, including 7 genes involved in flavonoid biosynthesis, under different salt concentrations were consistent with the transcriptomic data, and parallel reaction monitoring (PRM) results showed that the expression levels of 6 key enzymes (proteins) involved in flavonoid synthesis were consistent with the accumulation of flavonoids. This study provides a new perspective for investigating the regulatory role of flavonoid biosynthesis in the regulation of mulberry seed germination under salt stress at different concentrations.

Abstract:Salt damage is one of severe abiotic stresses which restricts yield and quality of crops all over the world. In China, saline alkali soil area is increasing mainly for environmental pollution and unreasonable fertilization in recent decades. Although cotton (Gossypium hirsutum L.) is classified as a strong salt-resistant plant, it is sensitive to salinity at seedling and flowering stages. Thus, in order to improve salt tolerance of cottons, systematical study on salt tolerance mechanism becomes more and more necessary. In other words, research on the growth and physiological characteristics can not only explore the mechanism of salt tolerance of plant, but also lay the foundation for breeding new salt-tolerant varieties.In this experiment, solution cultures with 0 (CK), 150 and 300 mmol/L NaCl were conducted to study the effect of salt stress on characteristics of seedling growth, antioxidant enzyme activity, malonaldehyde (MDA) content and inorganic ion concentration of two upland cotton cultivars (cv. Zaoshuchangrong7, salt-tolerant and cv. Nandanbadidahua, salt-sensitive). All the samples were collected after one week treatment. Concentrations of Na+, K+, Ca2+, and Mg2+ of different organs (root, stem and leaf) were detected by inductively coupled plasma-atomic emission spectroscopy (ICP-AES). The result showed that the salt-tolerant cotton had a larger growth quantity of root under moderate salt stress than the salt-sensitive cotton. For instance, under the 150 mmol/L NaCl treatment, the salt-tolerant cv. Zaoshuchangrong7 had 14.3%, 10.9%and 21.7% more in primary lateral root number, taproot length, and root mass than the control, respectively. Also, the higher net photosynthetic rate and antioxidant enzyme activity could be maintained in the leaf of salt-tolerant cotton under the salt stress. For example, the activities of superoxide dismutase (SOD) and peroxidase (POD) were increased gradually in the salt-tolerant cv. Zaoshuchangrong7 along with the increase of salt stress, while the activities of these enzymes increased firstly and then reduced in salt-sensitive cv. Nandanbadidahua. Furthermore, there was a regular distribution of inorganic ion concentrations of the salt-tolerant cv. Zaoshuchangrong7 under the same NaCl stress: greatly higher concentration of Na + in the root than in the stem and leaf; higher concentrations of K+, Ca2+, and Mg2+, but lower ratio of Na+/K+ in the shoot. It was indicated that the salt-tolerant cv. Zaoshuchangrong7 could store up part of Na + in the root thereby to relieve Na + toxicity in the shoot. However, MDA content of salt-sensitive cv. Nandanbadidahua increased more dramatically than the salt-tolerant cv. Zaoshuchangrong7 when the NaCl concentration climbed from 150 to 300 mmol/L. Under the 150 and 300 mmol/L NaCl treatments, the MDA contents of salt-sensitive cv. Nandanbadidahua increased 30.3% and 83.5%, respectively, when compared with the control, and remained higher level than the salt-tolerant cv. Zaoshuchangrong7. To sum up, this study reveals that protection from ion toxicity and oxidative damage by regulating the root growth, enhancing the activity of antioxidant enzyme and coordinating the ion homeostasis in different organs may be major physiological mechanisms of salt-tolerant cotton in response to salt damage.
Transcriptome and Metabolome Analysis Reveals Salt-Tolerance Pathways in the Leaves and Roots of ZM-4 (Malus zumi) in the Early Stages of Salt Stress

Abstract:The breeding of salt-tolerant rootstock relies heavily on the availability of salt-tolerant Malus germplasm resources. The first step in developing salt-tolerant resources is to learn their molecular and metabolic underpinnings. Hydroponic seedlings of both ZM-4 (salt-tolerant resource) and M9T337 (salt-sensitive rootstock) were treated with a solution of 75 mM salinity. ZM-4's fresh weight increased, then decreased, and then increased again after being treated with NaCl, whereas M9T337′s fresh weight continued to decrease. The results of transcriptome and metabolome after 0 h (CK) and 24 h of NaCl treatment showed that the leaves of ZM-4 had a higher content of flavonoids (phloretinm, naringenin-7-O-glucoside, kaempferol-3-O-galactoside, epiafzelechin, etc.) and the genes (CHI, CYP, FLS, LAR, and ANR) related to the flavonoid synthesis pathway showed up-regulation, suggesting a high antioxidant capacity. In addition to the high polyphenol content (L-phenylalanine, 5-O-p-coumaroyl quinic acid) and the high related gene expression (4CLL9 and SAT), the roots of ZM-4 exhibited a high osmotic adjustment ability. Under normal growing conditions, the roots of ZM-4 contained a higher content of some amino acids (L-proline, tran-4-hydroxy-L-prolin, L-glutamine, etc.) and sugars (D−fructose 6−phosphate, D−glucose 6−phosphate, etc.), and the genes (GLT1, BAM7, INV1, etc.) related to these two pathways were highly expressed. Furthermore, some amino acids (S-(methyl) glutathione, N-methyl-trans-4-hydroxy-L-proline, etc.) and sugars (D-sucrose, maltotriose, etc.) increased and genes (ALD1, BCAT1, AMY1.1, etc.) related to the pathways showed up-regulation under salt stress. This research provided theoretical support for the application of breeding salt-tolerant rootstocks by elucidating the molecular and metabolic mechanisms of salt tolerance during the early stages of salt treatment for ZM-4.

Research on Physiological Characteristics and Differential Gene Expression of Rice Hybrids and Their Parents under Salt Stress at Seedling Stage

Abstract:Soil salinization is one of the most important abiotic stresses which can seriously affect the growth and development of rice, leading to the decrease in or even loss of a rice harvest. Increasing the rice yield of saline soil is a key issue for agricultural production. The utilization of heterosis could significantly increase crop biomass and yield, which might be an effective way to meet the demand for rice cultivation in saline soil. In this study, to elucidate the regulatory mechanisms of rice hybrids and their parents that respond to salt stress, we investigated the phenotypic characteristics, physiological and biochemical indexes, and expression level of salt-related genes at the seedling stage. In this study, two sets of materials, encapsulating the most significant differences between the rice hybrids and their parents, were screened using the salt damage index and a hybrid superiority analysis. Compared with their parents, the rice hybrids Guang-Ba-You-Hua-Zhan (BB1) and Y-Liang-You-900 (GD1) exhibited much better salt tolerance, including an increased fresh weight and higher survival rate, a better scavenging ability towards reactive oxygen species (ROS), better ionic homeostasis with lower content of Na+ in their Na+/K+ ratio, and a higher expression of salt-stress-responsive genes. These results indicated that rice hybrids developed complex regulatory mechanisms involving multiple pathways and genes to adapt to salt stress and provided a physiological basis for the utilization of heterosis for improving the yield of rice under salt stress.

Abstract:Salinity is a major abiotic stress affecting agricultural production. To understand the genetic behavior of salinity tolerance traits, a half‐diallel cross was made among six barley cultivars (Hordeum vulgare L.), with contrasting levels of known tolerance, to study the combining ability of salinity tolerance on the basis of K+ loss from plant roots under saline conditions. The glasshouse pot experiments showed that the six parents were significantly different in salinity tolerance and those tolerances were highly correlated with the K+ flux measurements. The combining ability analysis showed that the variances of both general combining ability (GCA) and specific combining ability (SCA) were highly significant. Two tolerant cultivars, CM72 and Numar, showed significantly higher GCA for salinity tolerance (less K+ loss under salinity stress). Cultivars with medium GCA were YU6472 and Yan90260. Salinity tolerance was mainly controlled by additive effects with the tolerance allele showing partial dominance. High positive SCA was also found between two tolerant cultivars and between tolerant and medium‐tolerant cultivars, indicating possible different tolerant genes or some minor genes in these cultivars. The combination of these genes from different sources of tolerant cultivars should produce cultivars with even greater tolerance.
Identification and functional verification of salt tolerance hub genes in Salix matsudana based on QTL and transcriptome analysis

Abstract:Salt tolerance hub genes play a vital role in the response to salt stress. Exploring the salt tolerance hub genes of Salix matsudana (Koidz.), an allotetraploid willow with certain salt tolerance, will be conducive to further research on the molecular mechanism of salt tolerance in trees. Here, 114 F 1 populations of crossing salt-tolerant willow "9901" and salt-sensitive willow "Yanjiang" were used to perform the QTL of salt-tolerant traits and transcriptome analysis. 46 QTLs related to salt tolerance traits were obtained, of which 9 QTLs can regulate multiple traits. A total of 23795 differentially expressed genes (DEGs) were obtained by RNA-seq in different salt treatment periods (0 h/4 h/8 h/12 h). Weighted gene co-expression network analysis (WGCNA) and time-ordered gene co-expression network (TO-GCN) analysis on these DEGs revealed candidate hub genes in regulating salt tolerance. Combining the QTL and transcriptome results, the number of salt tolerance hub genes was cut down and several hub genes were selected for further functional verification. qRT-PCR, VIGS and salt-sensitive yeast complementation experiment showed that the silencing of one of salt tolerance hub gene, SmERF B1-2 significantly reduced salt tolerance in S. matsudana . And the expression of SmERF B1-2 in salt-sensitive yeast strain AXT3K rescued its salt sensitive phenotype. In this study, the combination of QTL and transcriptome was shown to be effective in screening the salt-tolerant hub gene of S. matsudana . This salt-tolerant hub gene identified in this way provides new insight into the molecular mechanisms of salt tolerance in trees and a theoretical basis for molecular breeding of salt-tolerant trees.

2 A Framework for Telestroke Network Design

Abstract:Stroke is the third leading cause of death and leading cause of adult long-term disability in western industrialized countries. In stroke units providing special expertise, stroke patients receive highly efficacious care (Pollack et al., 2007). However, stroke units cannot be implemented in particular in sparsely populated rural areas because of a shortage of experienced neurologists (Audebert & Schwamm, 2009). As compensatory measures, telemedical solutions are increasingly applied within “telestroke” networks to provide neurological expertise from “hubs” to small primary care hospitals (“spokes”) (Audebert 2006; Müller et al., 2006). Due to various environmental factors and personal preferences, different types of telestroke networks have emerged especially in Europe and the United States within the last ten years (Schwamm et al., 2009a; Günzel et al., 2010). In these networks, various pilot studies have demonstrated that valid decisions on thrombolytic therapy, the most important and time-critical therapy for the majority of acute stroke patients, and on a variety of further special interventions can be made accurately with the aid of telemedical expert support (Audebert et al., 2006; Audebert et al., 2009). Since telemedicine has proven to be a valid supplemental procedure in the treatment of acute stroke patients, primary care hospitals, especially in less populated regions, increasingly show interest in complementing their range of care by teleconsultations (Ickenstein et al., 2010). Those primary care hospitals may have totally different characteristics, some are e.g. small and located far away from the next certified stroke unit, while others are mediumsized and have neurology experts available on weekdays, but need support for night shifts and weekends. From a health economic point of view, these and many other different types of hospitals could benefit from telestroke care. However, current telemedicine networks focus in their approaches mostly on one specific group of primary care hospitals, thereby excluding others and thus withholding specialized stroke care from a large number of stroke patients. The situation may even worsen when national reimbursement regulations for telestroke care within the DRG system will be introduced. Those regulations will most likely be tied to current practice of the dominant telestroke network design in the respective country, thereby raising an entry barrier for all other primary care hospitals. To address these important issues for the first time, comparative health economic research into optimizing health care provision by both local stroke units and combinations of established telestroke network concepts is needed – and it must specifically take into account different regional environments. Abstract:Gamma-aminobutyric acid (GABA) has been reported to accumulate in plants when subjected to salt stress, and GABA-transaminase (GABA-T) is the main GABA-degrading enzyme in the GABA shunt pathway. So far, the salt tolerance mechanism of the GABA-T gene behind the GABA metabolism remains unclear. In this study, the cDNA (designated MuGABA-T) of GABA-T gene was cloned from mulberry, and our data showed that MuGABA-T protein shares some conserved characteristics with its homologs from several plant species. MuGABA-T gene was constitutively expressed at different levels in mulberry tissues, and was induced substantially by NaCl, ABA and SA. In addition, our results demonstrated that exogenous application of GABA significantly reduced the salt damage index and increased plant resistance to NaCl stress. We further performed a functional analysis of MuGABA-T gene and demonstrated that the content of GABA was reduced in the transgenic MuGABA-T Arabidopsis plants, which accumulated more ROS and exhibited more sensitivity to salt stress than wild-type plants. However, exogenous application of GABA significantly increased the activities of antioxidant enzymes and alleviated the active oxygen-related injury of the transgenic plants under NaCl stress. Moreover, the MuGABA-T gene was overexpressed in the mulberry hairy roots, and similar results were obtained for sensitivity to salt stress in the transgenic mulberry plants. Our results suggest that the MuGABA-T gene plays a pivotal role in GABA catabolism and is responsible for a decrease in salt tolerance, and it may be involved in the ROS pathway in the response to salt stress. Taken together, the information provided here is helpful for further analysis of the function of GABA-T genes, and may promote mulberry resistance breeding in the future.

Abstract:Arbuscular mycorrhizal fungi (AMF) are often considered bioameliorators. AMF can promote plant growth under various stressful conditions; however, differences between male and female clones in mycorrhizal strategies that protect plants from the detrimental effects of salinity are not well studied. In this study, we aimed to examine the interactive effects of salinity and AMF on the growth, photosynthetic traits, nutrient uptake, and biochemical responses of Morus alba males and females. In a factorial setup, male and female M. alba clones were subjected to three salinity regimes (0, 50, and 200 mM NaCl) and planted in soil with or without Funneliformis mosseae inoculation. The results showed that NaCl alone conferred negative effects on the growth, salinity tolerance, photosynthetic performance, and shoot and root ionic ratios (K + /Na + , Ca 2+ /Na + , and Mg 2+ /Na + ) in both sexes; in contrast, mycorrhizal inoculation mitigated the detrimental effects of salinity. Furthermore, the mycorrhizal effects were closely correlated with Mn 2+ , proline, and N concentrations. Females benefited more from AMF inoculation as shown by the enhancements in their biomass accumulation, and N, proline, K + , Mg 2+ , Fe 2+ , Zn 2+ , and Mn 2+ concentrations than males with mycorrhizal inoculation under saline conditions. In comparison, male plants inoculated with AMF showed improvements in biomass allocated to the roots, P, and peroxidase concentrations under saline conditions. These sex-specific differences suggest that male and female mulberry clones adopted different mycorrhizal strategies when growing under saline conditions. Overall, our results provide insight into the sex-specific difference in the performance of AMF-associated mulberry clones, suggesting that female mulberry could be more suitable for vegetation remediation than the male one, due to its higher salinity tolerance.

Abstract:Marginal lands, such as those with saline soils, have potential as alternative resources for cultivating dedicated biomass crops used in the production of renewable energy and chemicals. Optimum utilization of marginal lands can not only alleviate the competition for arable land use with primary food crops, but also contribute to bioenergy products and soil improvement. Miscanthus sacchariflorus and M. lutarioriparius are prominent perennial plants suitable for sustainable bioenergy production in saline soils. However, their responses to salt stress remain largely unexplored. In this study, we utilized 318 genotypes of M. sacchariflorus and M. lutarioriparius to assess their salt tolerance levels under 150 mM NaCl using 14 traits, and subsequently established a mini-core elite collection for salt tolerance. Our results revealed substantial variation in salt tolerance among the evaluated genotypes. Salt-tolerant genotypes exhibited significantly lower Na + content, and K + content was positively correlated with Na + content. Interestingly, a few genotypes with higher Na + levels in shoots showed improved shoot growth characteristics. This observation suggests that M. sacchariflorus and M. lutarioriparius adapt to salt stress by regulating ion homeostasis, primarily through enhanced K + uptake, shoot Na + exclusion, and Na + sequestration in shoot vacuoles. To evaluate salt tolerance comprehensively, we developed an assessment value (D value) based on the membership function values of the 14 traits. We identified three highly salt-tolerant, 50 salt-tolerant, 127 moderately salt-tolerant, 117 salt-sensitive, and 21 highly salt-sensitive genotypes at the seedling stage by employing the D value. A mathematical evaluation model for salt tolerance was established for M. sacchariflorus and M. lutarioriparius at the seedling stage. Notably, the mini-core collection containing 64 genotypes developed using the Core Hunter algorithm effectively represented the overall variability of the entire collection. This mini-core collection serves as a valuable gene pool for future in-depth investigations of salt tolerance mechanisms in Miscanthus .

Abstract:Selection and utilization of salt-tolerant crops are essential strategies for mitigating salinity damage to crop productivity with increasing soil salinization worldwide. This study was conducted to identify salt-tolerant white clover ( Trifolium repens ) genotypes among 37 materials based on a comprehensive evaluation of five physiological parameters, namely, chlorophyll (Chl) content, photochemical efficiency of PS II (Fv/Fm), performance index on an absorption basis (PIABS), and leaf relative water content (RWC), and to further analyze the potential mechanism of salt tolerance associated with changes in growth, photosynthetic performance, endogenous polyamine metabolism, and Na + /K + uptake and transport. The results showed that significant variations in salt tolerance were identified among 37 genotypes, as PI237292 and Tr005 were the top two genotypes with the highest salt tolerance, and PI251432 and Korla were the most salt-sensitive genotypes compared to other materials. The salt-tolerant PI237292 and Tr005 not only maintained significantly lower EL but also showed significantly better photosynthetic performance, higher leaf RWC, underground dry weight, and the root to shoot ratio than the salt-sensitive PI251432 and Korla under salt stress. Increases in endogenous PAs, putrescine (Put), and spermidine (Spd) contents could be key adaptive responses to salt stress in the PI237292 and the Tr005 through upregulating genes encoding Put and Spd biosynthesis ( NCA , ADC , SAMDC , and SPDS2 ). For Na + and K + accumulation and transport, higher salt tolerance of the PI237292 could be associated with the maintenance of Na + and Ca + homeostasis associated with upregulations of NCLX and BTB/POZ . The K + homeostasis-related genes ( KEA2 , HAK25 , SKOR , POT2 / 8 / 11 , TPK3 / 5 , and AKT1 / 5 ) are differentially expressed among four genotypes under salt stress. However, the K + level and K + /Na + ratio were not completely consistent with the salt tolerance of the four genotypes. The regulatory function of these differentially expressed genes (DEGs) on salt tolerance in the white clover and other leguminous plants needs to be investigated further. The current findings also provide basic genotypes for molecular-based breeding for salt tolerance in white clover species.

Abstract:Soil salinity is a major factor threatening the production of crops around the world. Smooth bromegrass ( Bromus inermis L.) is a high-quality grass in northern and northwestern China. Currently, selecting and utilizing salt-tolerant genotypes is an important way to mitigate the detrimental effects of salinity on crop productivity. In our research, salt-tolerant and salt-sensitive varieties were selected from 57 accessions based on a comprehensive evaluation of 22 relevant indexes, and their salt-tolerance physiological and molecular mechanisms were further analyzed. Results showed significant differences in salt tolerance between 57 genotypes, with Q25 and Q46 considered to be the most salt-tolerant and salt-sensitive accessions, respectively, compared to other varieties. Under saline conditions, the salt-tolerant genotype Q25 not only maintained significantly higher photosynthetic performance, leaf relative water content (RWC), and proline content but also exhibited obviously lower relative conductivity and malondialdehyde (MDA) content than the salt-sensitive Q46 ( p < 0.05). The transcriptome sequencing indicated 15,128 differentially expressed genes (DEGs) in Q46, of which 7,885 were upregulated and 7,243 downregulated, and 12,658 DEGs in Q25, of which 6,059 were upregulated and 6,599 downregulated. The Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis showed that the salt response differences between Q25 and Q46 were attributed to the variable expression of genes associated with plant hormone signal transduction and MAPK signaling pathways. Furthermore, a large number of candidate genes, related to salt tolerance, were detected, which involved transcription factors (zinc finger proteins) and accumulation of compatible osmolytes (glutathione S -transferases and pyrroline-5-carboxylate reductases), etc. This study offers an important view of the physiological and molecular regulatory mechanisms of salt tolerance in two smooth bromegrass genotypes and lays the foundation for further identification of key genes linked to salt tolerance.

Abstract:Abstract Background Saline, alkaline, and saline-alkaline stress severely affect plant growth and development. The tolerance of plants to these stressors has long been important breeding objectives, especially for woody perennials like apple. The aims of this study were to identify quantitative trait loci (QTLs) and to develop genomics-assisted prediction models for salt, alkali, and salt-alkali tolerance in apple rootstock. Results A total of 3258 hybrids derived from the apple rootstock cultivars ‘Baleng Crab’ ( Malus robusta Rehd., tolerant) × ‘M9’ ( M. pumila Mill., sensitive) were used to identify 17, 13, and two QTLs for injury indices of salt, alkali, and salt–alkali stress via bulked segregant analysis. The genotype effects of single nucleotide polymorphism (SNP) markers designed on candidate genes in each QTL interval were estimated. The genomic predicted value of an individual hybrid was calculated by adding the sum of all marker genotype effects to the mean phenotype value of the population. The prediction accuracy was 0.6569, 0.6695, and 0.5834 for injury indices of salt, alkali, and salt–alkali stress, respectively. SNP182G on MdRGLG3 , which changes a leucine to an arginine at the vWFA-domain, conferred tolerance to salt, alkali, and salt-alkali stress. SNP761A on MdKCAB , affecting the Kv_beta domain that cooperated with the linked allelic variation SNP11, contributed to salt, alkali, and salt–alkali tolerance in apple rootstock. Conclusions The genomics-assisted prediction models can potentially be used in breeding saline, alkaline, and saline-alkaline tolerant apple rootstocks. The QTLs and the functional markers may provide insight for future studies into the genetic variation of plant abiotic stress tolerance.

Abstract:Elevated salinity significantly threatens cotton growth, particularly during the germination and seedling stages. The utilization of primitive species of Gossypium hirsutum, specifically Gossypium purpurascens, has the potential to facilitate the restoration of genetic diversity that has been depleted due to selective breeding in modern cultivars. This investigation evaluated 45 G. purpurascens varieties and a salt-tolerant cotton variety based on 34 morphological, physiological, and biochemical indicators and comprehensive salt tolerance index values. This study effectively identified a total of 19 salt-tolerant and two salt-resistant varieties. Furthermore, transcriptome sequencing of a salt-tolerant genotype (Nayanmian-2; NY2) and a salt-sensitive genotype (Sanshagaopao-2; GP2) revealed 2776, 6680, 4660, and 4174 differentially expressed genes (DEGs) under 0.5, 3, 12, and 24 h of salt stress. Gene ontology enrichment analysis indicated that the DEGs exhibited significant enrichment in biological processes like metabolic (GO:0008152) and cellular (GO:0009987) processes. MAPK signaling, plant-pathogen interaction, starch and sucrose metabolism, plant hormone signaling, photosynthesis, and fatty acid metabolism were identified as key KEGG pathways involved in salinity stress. Among the DEGs, including NAC, MYB, WRKY, ERF, bHLH, and bZIP, transcription factors, receptor-like kinases, and carbohydrate-active enzymes were crucial in salinity tolerance. Weighted gene co-expression network analysis (WGCNA) unveiled associations of salt-tolerant genotypes with flavonoid metabolism, carbon metabolism, and MAPK signaling pathways. Identifying nine hub genes (MYB4, MYB105, MYB36, bZIP19, bZIP43, FRS2 SMARCAL1, BBX21, F-box) across various intervals offered insights into the transcriptional regulation mechanism of salt tolerance in G. purpurascens. This study lays the groundwork for understanding the important pathways and gene networks in response to salt stress, thereby providing a foundation for enhancing salt tolerance in upland cotton.

Abstract:Identification of tolerant mulberry genotypes and elucidation of stress-tolerant mechanisms are two priority areas need to be addressed for sustainable mulberry leaf production. In the present study, 16 mulberry genotypes were evaluated under at low (75% FC), medium (50% FC), and high moisture stress (25% FC) conditions along with well-irrigated plants (100% FC) and measured the photosynthetic efficiency and antioxidant potential. In well-irrigated plants, photosynthetic rate was high in S13 (16.45 μmol/m 2 /s), Mysore Local, RC2, and AGB8 (13.62–16 μmol/m 2 /s). All genotypes showed a gradual decline in photosynthetic rate during the progression of moisture stress (low to high stress). MSG2, AGB8, RC1, RC2, and Nagalur Estate showed high photosynthetic rate (7.45–10.15 μmol/m 2 /s) under high moisture stress, whereas low photosynthetic rate was recorded in Mysore Local (3.46 μmol/m 2 /s) and K2 (3.50 μmol/m 2 /s). All tolerant genotypes showed higher activities of SOD (104.25–122.44 Units/g) and POX (25.61–33.73 Units/g) activities under high moisture stress. Bundi-3, S34, RC2, and AGB8 showed higher SOD activity and low enzyme activity was observed in K2. Non-enzymatic antioxidants and osmolytes were high in moisture stress-tolerant genotypes. This study identified that high photosynthetic rate is one of the important physiological indicator for moisture stress tolerance in mulberry. Also higher activities of antioxidant enzymes (SOD and POX), presence of high quantity of non-enzymatic antioxidants (ascorbic acids, reduced glutathione, and phenols), and accumulation of osmolytes (proline and glycine betaine) are identified as stress tolerance biochemical markers in mulberry genotypes. Evaluation Index (EI) value was calculated and genotypes suitable for well-irrigated condition as well as for low, medium, and high moisture stress conditions were identified based on EI values.
