Abstract:The growing concern about pollution and toxicity in aquatic as well as terrestrial organisms is predominantly caused due to waterborne exposure and poses a risk to environmental systems and human health. This study addresses the co-toxic effects of cadmium (Cd) and ketoprofen (KPF), representing heavy metal and pharmaceutical discharge pollutants, respectively, in aquatic ecosystems. A 96-h acute toxicity assessment was conducted using zebrafish embryos. The results indicated that high dosages of KPF (10, 15, and 100 μg/mL) and Cd (10 and 15 μg/mL) reduced survivability and caused concentration-dependent deformities such as scoliosis and yolk sac edema. These findings highlight the potential defects in development and metabolism, as evidenced by hemolysis tests demonstrating dose-dependent effects on blood cell integrity. Furthermore, this study employs adult zebrafish for a 42-day chronic exposure to Cd and KPF (10 and 100 μg/L) alone or combined (10 + 10 and 100 + 100 μg/L) to assess organ-specific Cd and KPF accumulation in tissue samples. Organ-specific accumulation patterns underscore complex interactions impacting respiratory, metabolic, and detoxification functions. Prolonged exposure induces reactive oxygen species formation, compromising antioxidant defense systems. Histological examinations reveal structural changes in gills, gastrointestinal, kidney, and liver tissues, suggesting impairments in respiratory, osmoregulatory, nutritional, and immune functions. This study emphasizes the importance of conducting extensive research on co-toxic effects to assist with environmental risk assessments and safeguard human health and aquatic ecosystems.

Vitamin C Mitigates Oxidative Stress and Behavioral Impairments Induced by Deltamethrin and Lead Toxicity in Zebrafish

Abstract:Environmental contamination from toxic metals and pesticides is an issue of great concern due to their harmful effects to human health and the ecosystems. In this framework, we assessed the adverse effects when aquatic organisms are exposed to toxicants such as deltamethrin (DM) and lead (Pb), alone or in combination, using zebrafish as a model. Moreover, we likewise evaluated the possible protective effect of vitamin C (VC) supplementation against the combined acute toxic effects of the two toxicants. Juvenile zebrafish were exposed to DM (2 μg L-1) and Pb (60 μg L-1) alone and in combination with VC (100 μg L-1) and responses were assessed by quantifying acetylcholinesterase (AChE) activity, lipid peroxidation (MDA), some antioxidant enzyme activities (SOD and GPx), three-dimension locomotion responses and changes of elements concentrations in the zebrafish body. Our results show that VC has mitigative effects against behavioral and biochemical alterations induced by a mixture of contaminants, demonstrating that it can be used as an effective antioxidant. Moreover, the observations in the study demonstrate zebrafish as a promising in vivo model for assessing the neuroprotective actions of bioactive compounds.
Acute exposure to waterborne cadmium induced oxidative stress and immunotoxicity in the brain, ovary and liver of zebrafish (Danio rerio)

Abstract:Cadmium (Cd) is an environmental contaminant that poses serious risks to aquatic organisms and their associated ecosystem. The mechanisms underlying Cd-induced oxidative stress and immunotoxicity in fish remain largely unknown. In this study, adult female zebrafish were exposed to 0 (control), 1mgL-1 Cd for 24h and 96h, and the oxidative stress and inflammatory responses induced by Cd were evaluated in the brain, liver and ovary. Reactive oxygen species (ROS), nitric oxide (NO), and malondialdehyde (MDA) increased in a time-dependent manner after treatment with Cd in the brain and liver. The increase may result from the disturbance of genes including copper and zinc superoxide dismutase (Cu/Zn-SOD), catalase (CAT), inducible nitric oxide synthase (iNOS), and ciclooxigenase-2 (COX-2) at mRNA, protein and activity levels. Although ROS, NO and MDA were not significantly affected by Cd in the ovary, the up-regulation of Cu/Zn-SOD, CAT, iNOS, and COX-2 was observed. Exposure to Cd induced a sharp increase in the protein levels of tumor necrosis factor alpha (TNF-α) in the brain, liver and ovary, possibly contributing to activate inflammatory responses. Furthermore, we also found a dramatic increase in mRNA levels of NF-E2-related factor 2 (Nrf2) and nuclear transcription factor κB (NF-κB) at 24h in the liver and ovary. The corresponding changes in the mRNA levels of Kelch-like-ECH-associated protein 1 (Keap1a and Keap1b) and the inhibitor of κBα (IκBαa and IκBαb) may contribute to regulate the transcriptional activity of Nrf2 and NF-κB, respectively. Contrarily, mRNA levels of Nrf2, NF-κB, Keap1, Keap1b, IκBαa and IκBαb remained stable at 24 and 96h in the brain. Taken together, we demonstrated Cd-induced oxidative stress and immunotoxicity in fish, possibly through transcriptional regulation of Nrf2 and NF-κB and gene modifications at transcriptional, translational, post-translational levels, which would greatly extend our understanding on the Cd toxicity.
Oxidative Stress, Neurotoxicity, and Intestinal Microbial Regulation after a Chronic Zinc Exposure: an Experimental Study on Adult Zebrafish (danio Rerio)

Abstract:Zinc is one of the heavy metals present in textile wastewater with high concentrations. However, the chronic toxic effects of zinc on aquatic vertebrates are still ambiguous. Zinc accumulation in zebrafish after chronic zinc exposure and toxic effects on the intestines, muscles, and gills were investigated in this study. The results showed that a significant accumulation of zinc in the intestine, muscle, and gill was observed after 25 d of zinc exposure. The toxic effects of zinc were mainly in the form of zinc-induced oxidative stress in zebrafish, potential neuro-toxicity, and changes in intestinal microbes. Significant changes in the levels of superoxide dismutase, catalase, metallothionein, glutathione, and malondialdehyde indicated that zinc damaged the antioxidant system of adult zebrafish. Zinc exposure resulted in a significant decrease in acetylcholinesterase activity and abnormal neural signaling. Furthermore, zinc exposure resulted in increased intestinal microbial richness and decreased the Simpson index in adult zebrafish. At the phylum and genus levels, the predominant microbes in the intestine are altered by zinc. In summary, this study provides an analysis of the toxic effects of chronic zinc exposure on adult zebrafish and the potential mech-anisms, which are important for assessing the dual effects of zinc on aquatic organisms.
Effects of Single and Combined Ciprofloxacin and Lead Treatments on Zebrafish Behavior, Oxidative Stress, and Elements Content

Abstract:Even though the toxic effects of antibiotics and heavy metals have been extensively studied in the last decades, their combined adverse impact on aquatic organisms is poorly understood. Therefore, the objective of this study was to assess the acute effects of a ciprofloxacin (Cipro) and lead (Pb) mixture on the 3D swimming behavior, acetylcholinesterase (AChE) activity, lipid peroxidation level (MDA-malondialdehyde), activity of some oxidative stress markers (SOD-superoxide dismutase and GPx-glutathione peroxidase), and the essential elements content (Cu-copper, Zn-zinc, Fe-iron, Ca-calcium, Mg-magnesium, Na-sodium and K-potassium) in the body of zebrafish (Danio rerio). For this purpose, zebrafish were exposed to environmentally relevant concentrations of Cipro, Pb, and a mixture for 96 h. The results revealed that acute exposure to Pb alone and in mixture with Cipro impaired zebrafish exploratory behavior by decreasing swimming activity and elevating freezing duration. Moreover, significant deficiencies of Ca, K, Mg, and Na contents, as well as an excess of Zn level, were observed in fish tissues after exposure to the binary mixture. Likewise, the combined treatment with Pb and Cipro inhibited the activity of AChE and increased the GPx activity and MDA level. The mixture produced more damage in all studied endpoints, while Cipro had no significant effect. The findings highlight that the simultaneous presence of antibiotics and heavy metals in the environment can pose a threat to the health of living organisms.

Abstract:Heavy metal pollution has a harmful impact on the health of all living beings. The goal of this study was to compare the impacts of zinc (Zn), lead (Pb), iron (Fe), and manganese (Mn) on two Egyptian streams in Aswan Governorate. In addition, aquatic Chironomidae larvae were exposed to a variety of heavy metal concentrations for two days. The mortality rate was determined, and the median lethal concentrations (LC50) were estimated. The most harmful metal for Chironomidae larvae was Pb, followed by Zn, Fe, and Mn. Metal bioconcentration in Chironomidae larvae increases with increasing concentrations of metals. The enzymatic response to heavy metals uptake in Chironomidae larvae; namely, lipid peroxidation, superoxide dismutase, catalase, glutathione-S-transferase, and glutathione peroxidase were determined. Although the Pb-treated group recorded the highest lipid peroxidation, it showed the lowest glutathione peroxidase activity. The glutathione-S-transferase enzyme was less active in the Zn-treated group, as was the superoxide dismutase enzyme, and the catalase enzyme was the least active in the Zn-treated group. Changes in randomly amplified polymorphic DNA (RAPD) profiles were detected following heavy metal treatments at various doses when compared to controls. This study gives insight into how cruise ship activities and industrial pollution in rivers endanger aquatic life and hence, human life.
Ecotoxicological Assessment of Heavy Metal and Its Biochemical Effect in Fishes

Abstract:Level of toxic heavy metal concentration like lead (Pb), chromium (Cr), cadmium (Cd), iron (Fe), copper (Cu), zinc (Zn), and nickel (Ni) in thirty-six soft and hard organs and their impact on lipid profile of Hypophthalmichthys molitrix and Catla catla fish species inhibiting in Tanda Dam reservoir were investigated. The heavy metal concentrations in water, sediment, and fish of the different regions in the reservoir were determined with atomic absorption spectrophotometer. Lipid profile was carried out by AOAC official methods. The results showed that Pb was dominant among all the heavy metals in six organs, and its maximum concentration of Pb (22.5 mg kg-1 and 32.9 mg kg-1) was observed in scales in Hypophthalmichthys molitrix and tail of Catla catla, respectively. The maximum concentrations of Cd were observed in the head, scales, fins, and gills of Catla catla. The bioaccumulation of heavy metals was significantly different at (p ≤ 0.01) within the organs and between the fish species. The lipid concentration was minimum in those organs where the concentrations of heavy metals were maximum. It is clear from the findings that heavy metal accumulation reduces the lipid content of fish. It is inevitable to monitor the Tanda Dam reservoir to safeguard human health.
Examining the subchronic (28-day) effects of aqueous Cd-BaP co-exposure on detoxification capacity and cardiac function in adult zebrafish (Danio rerio)

Abstract:The present study aimed to examine the effects of environmentally relevant concentrations of cadmium (Cd) and Benzo[a]Pyrene (BaP) in the adult zebrafish (Danio rerio). To this end, fish were exposed to either 1 or 10 μg/L Cd or 0.1 or 1 μg/L BaP in isolation, or a co-exposure containing a mixture of the two toxicants. Our results showed extensive modulation of the expression of key antioxidant genes (GPx, SOD1, catalase), detoxifying genes (MT1, MT2, CYP1A1) and a stress biomarker (HSP70) differing between control, single toxicant groups and co-exposure groups. We additionally carried out histopathological analysis of the gills, liver, and hearts of exposed animals, noting no differences in tissue necrosis or apoptosis. Finally, we carried out ultrasonographic analysis of cardiac function, noting a significant decrease of E-wave peak velocity and end diastolic volume in exposed fish. This in turn was accompanied by a decrease in stroke volume and ejection fraction, but not cardiac output in co-exposed fish. The present study is the first to demonstrate that a subchronic aqueous exposure to a Cd-BaP mixture can extensively modulate detoxification capacity and cardiac function in adult zebrafish in a tissue-specific manner.

Abstract:Pollution is a world problem with immeasurable consequences. Heavy metal compounds are frequently found as components of anthropogenic pollution. Here we evaluated the effects of the treatment with cadmium acetate, lead acetate, mercury chloride, and zinc chloride in acetylcholinesterase activity and gene expression pattern, as well as the effects of these treatments in antioxidant competence in the brain of an aquatic and well-established organism for toxicological analysis, zebrafish (Danio rerio, Cyprinidae). Mercury chloride and lead acetate promoted a significant decrease in acetylcholinesterase activity whereas they did not alter the gene expression pattern. In addition, the antioxidant competence was decreased after exposure to mercury chloride. The data presented here allowed us to hypothesize a signal transmission impairment, through alterations in cholinergic transmission, and also in the antioxidant competence of zebrafish brain tissue as some of the several effects elicited by these pollutants.

Abstract:Abstract Increasing industrial activity causes the release of chemical compounds into aquatic habitats, including toxic heavy metals like cadmium and medications like ketoprofen, posing considerable ecological concerns. Although previous studies have shown that cadmium and ketoprofen individually cause cognitive impairment, there is a lack of information on the combined neurological effects of the two substances. We investigated the neurological consequences of persistent cadmium exposure in the presence of ketoprofen on adult zebrafish, providing an essential model for understanding cumulative impacts on vertebrate organisms. Behavioral assessments, bioaccumulation rates, biochemical studies, and histopathological exams were conducted over 42 days in authentic environmental settings. The results of our study show that cadmium (10 μg/L) and ketoprofen (10 and 100 μg/L) at environmentally relevant concentrations had a significant impact on locomotor activity, social interactions, and cognitive responses, indicating cumulative neurotoxicity in co‐exposure groups compared to single pollutant groups. Biochemical tests show disturbances in antioxidant defense systems, while histological examinations reveal structural changes in zebrafish brain regions. Ketoprofen influences cadmium accumulation in the brain, underscoring the importance of conducting complete evaluations to understand the intricate interactions between environmental pollutants. This study improves our understanding of the complex interactions between heavy metals and medications, stressing the need to consider combined exposure when assessing the neurological effects on vertebrate models.

Abstract:Heavy metals are polluting the freshwater ecosystems with hazardous impacts on aquatic animals especially fish. Considerable amounts of Cadmium (Cd) in aquatic ecosystems are posing a serious threat to sustainable growth of aquaculture industry. Objective: To investigate acute toxicity of Cd (96-hour LC50 and lethal concentrations) was determined by utilizing probit analysis method for the freshwater fish Labeo rohita. Methods: Different organs of fish (liver, gills and muscles) were also analyzed for accumulation of Cd by inductively coupled plasma mass spectrometry (ICP-MS). Results: Acute toxicity of Cd (96-hour LC50) for Labeo rohita was calculated as 159.59 mg/L. Bioaccumulation of Cd in different tissues of fish was examined in the following order liver>gills> muscles. Comet assay was performed for the evaluation of DNA damage by different sub-lethal doses (1/2, 1/3, 1/4 of LC50) of Cd. Conclusions: This research will aid in control of heavy metals contamination and conservation of fish species Labeo rohita in natural aquatic habitats of Pakistan.

Abstract:The concentrations of 14 metals, namely, chromium (Cr), manganese (Mn), cobalt (Co), nickel (Ni), copper (Cu), zinc (Zn), selenium (Se), arsenic (As), strontium (Sr), cadmium (Cd), tin (Sn), antimony (Sb), mercury, (Hg), and lead (Pb), have been studied in freshwater mollusk Lamellidens marginalis collected from Dhimbe reservoir. In addition, also studied the cellular and antioxidative status to assess metal contamination as pollution biomarkers. The levels of all the 14 metals were elevated in different sampling sites of Dhimbe reservoir. The level of metals in the L. marginalis was followed as Mn > Zn > Ni > Sr > Cr > Cu > Hg > Se > Sn > Pb > As > Co > Cd > Sb. The oxidative biomarkers viz. catalase (CAT), superoxide dismutase (SOD), and glutathione-S-transferase (GST); glycolytic enzymes viz. lactate dehydrogenase (LDH) and malate dehydrogenase (MDH); and protein metabolic enzymes viz. aspartate amino transferase (AST), alanine amino transferase (ALT), and lipid peroxidation (LPO) were significantly (p < 0.01) higher, while neurotransmitter enzymes such as acetylcholine esterase (AChE) were significantly inhibited in muscle of L. marginalis (p < 0.01) collected from different sampling sites. Therefore, based on our results, we could recommend that the Dhimbe reservoir is moderately metal contaminated and investigation based on biochemical parameters such as antioxidative enzymes, cellular biomarkers, and AChE are strong indicators for metal contamination; hence, these parameters are successfully applied as reliable biomarkers for monitoring contaminated freshwater ecosystems.

Abstract:Arsenic (As) and chromium (Cr), two well-known cytotoxic and carcinogenic metals are reported to coexist in industrial effluents and groundwater. Their individual toxicities have been thoroughly studied but the combined effects, especially the mechanism of toxicity and cellular stress response remain unclear. Considering co-exposure as a more realistic scenario, current study compared the individual and mixture effects of As and Cr in the liver of zebrafish (Danio rerio). Fish were exposed to environmentally relevant concentrations of As and Cr for 15, 30 and 60 days. ROS generation, biochemical stress parameters like lipid peroxidation, reduced glutathione content, catalase activity and histological alterations were studied. Results showed increase in ROS production, MDA content and GSH level; and vicissitude in catalase activity as well as altered histoarchitecture, indicating oxidative stress conditions after individual and combined exposure of As and Cr which were additive in nature. This study also included the expression of Nrf2, the key regulator of antioxidant stress responses and its nuclear translocation. Related antioxidant and xenobiotic metabolizing enzyme genes like keap1, nqo1, ho1, mnsod and cyp1a were also studied. Overall results indicated increased nrf2, nqo1, ho1, mnsod expression at all time points and increased cyp1a expression after 60 days exposure. Emphasizing on the Nrf2-Keap1 pathway, this study exhibited additive or sometimes synergistic effects of As and Cr in zebrafish liver.

Abstract:In this study, laboratory experiments were conducted to investigate the combined effect of zinc and pH on metal accumulation and oxidative stress biomarkers in Carassius auratus. Fish were exposed to 0.1 and 1.0mg Zn/L at three pH values (5.0, 7.25, 9.0) for 3, 12, and 30 d. After each exposure, the contents of three trace elements (Zn, Fe and Cu) were determined in liver. Generally, longer exposure to zinc (12d and 30 d) increased hepatic Zn and Cu deposition, but decreased Fe content. Increasing accumulation of Zn in the tissue was also observed with increasing zinc concentration in the exposure medium. Moreover, hepatic antioxidant enzyme activities including superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx), together with the level of glutathione (GSH) were measured to evaluate the oxidative stress status. The decreases in the four measured biochemical parameters after 3d exposure might reflect the failure of the antioxidant defense system in neutralizing the ROS generated during the metabolic process, while the recovery of the antioxidants at days 12 and 30 suggested a possible shift toward a detoxification mechanism. With regard to the influence of pH on zinc toxicity, the general observation was that the living environment became more stressful when the water conditions changed from an acidic state toward a near-neutral or alkaline state.

Abstract:Zebrafish ( Danio rerio ) is ideal for studying the effects of toxins like lead or plumbum (Pb) which persist in the environment and harm body systems when absorbed. Increasing Pb concentration could result in a higher mortality rate and alteration of behavior and metabolism. The present study evaluates the acute toxicity effect of Pb on metabolome and behavior in adult zebrafish. The zebrafish were exposed to various Pb concentrations ranging from 0 to 30 mg/L for different periods (24, 48, and 72 h) before the fish samples were subjected to Nuclear Magnetic Resonance (NMR)-multivariate data analysis (MVDA) with additional support from behavioral assessment. The behavior of zebrafish was significantly altered after Pb inducement and the differential metabolites increased in low (5 mg/L) while decreased in high (10 mg/L) Pb concentrations. An ideal Pb induction could be achieved by 5 mg/L concentration in 24 h, which induced significant metabolite changes without irreversible damage. Continuing research on the effects of lead toxicity is crucial to develop effective prevention and treatment strategies.

Adenoviruses in faeces of children with acute gastroenteritis in Rio De Janeiro, Brazil

Abstract:Faeces from 746 children less than 5 years old with acute gastroenteritis were screened for the presence of adenovirus particles or antigens by immunoelectron microscopy (IEM) and enzyme immunoassay (EIA). Thirty‐five samples were positive by both IEM and EIA, two only by IEM, and two only by EIA, giving a total of 39 (5.2%) samples with positive results. Of these, 25 could be propagated in HEp2 cells and were neutralized by one of the antisera to adenovirus types 1 to 18. The remaining 14 samples could be propagated only in the 293 permanent line of human cells transformed by adenovirus type 5 DNA [Graham et al, 1977] and were not neutralized by antisera to adenovirus types 1 to 31. An EIA carried out by the antibody‐capture technique, using antiserum specific for “enteric” adenoviruses [Johansson et al, 1979], gave positive results with all isolates that could be propagated only in 293 cells and with none of those capable of growing in HEp2 cells.
Ameliorating effects of natural herbal supplements against water-borne induced toxicity of heavy metals on Nile tilapia, (Oreochromis niloticus)

Abstract:The efficacy of herbal supplements in mitigating heavy metals (HMs) toxicity was investigated using a widely grown fish, the Nile tilapia (Oreochromis niloticus). The experiment was conducted over two phases: during the stress phase, the experimental fishes were exposed to sub-lethal concentrations of HMs, including lead, cadmium, zinc, and copper for 15 days; following which during the feeding phase, herbal supplements were given for 70 days to ameliorate their effects. Seven groups were established: the control negative group (CON-ve), control positive group (CON+ve, without any treatment), and five groups with supplementation of 1% turmeric (TUR), cinnamon (CIN), ginger (GIN), garlic (GAR), and their mixture (MIX), respectively. A total of 315 fishes were distributed evenly in experimental tanks (15 fishes per tank, in triplicates). The results revealed that exposure to HMs led to significant (p < 0.05) alterations in all the tested parameters, i.e., liver damage and growth reduction. The herbal supplements, especially the MIX groups, ameliorated the harmful effects of HMs and restored fish growth, digestibility, carcass composition, and liver health. In conclusion, the study demonstrated that the herbal supplements were effective in reducing the HMs-linked toxicity in Nile tilapia. Future studies pertaining to the mechanisms facilitated by the various herbal bioactive substances-linked tolerance to HMs in fishes are warranted.

Abstract:Prolonged exposure to environmentally relevant amounts of cadmium (Cd) in aquatic environments, even at small doses (0.1 and 1 μg/L), might endanger the health of underwater creatures. This research delved into the impacts of a four-month cadmium exposure on Mozambique tilapia ( Oreochromis mossambicus ), aiming to uncover the mechanisms behind it. Through close examination, we found that the 4-momth cadmium exposure led to harmful effects on the fish's gills, muscles, brain, and intestines. This exposure also triggered changes in gene expressions in the brain and liver, affected the respiratory system and weakened liver's ability to detoxify and defend against potential infections. Looking deeper into the fish's gut, we noticed alterations in energy-related genes and disruptions in immune pathways, making it more susceptible to illnesses. The exposure to cadmium also had an impact on the fish's gut and water-dwelling microorganisms, reducing diversity and encouraging harmful microbial communities. Interestingly, some gut microbes seemed to assist in breaking down and detoxifying cadmium, which could potentially protect the fish. Taken together, prolonged low-level cadmium exposure impaired gill, muscle, and brain function, suppressed immunity, disrupted intestines, and altered microbial balance, leading to hindered growth. These insights illuminate cadmium's impact on fish, addressing vital environmental concerns.
