Abstract:The distal histidine mutations of dehaloperoxidase-hemoglobin A (DHP A) to aspartate (H55D) and asparagine (H55N) have been prepared to study the role played by the distal histidine in both activation and protection against oxidation by radicals in heme proteins. The H55D and H55N mutants of DHP A have similar to 6-fold and similar to 11-fold lower peroxidase activities than wild type enzyme toward the oxidation of 2,4,6-trichlorophenol (TCP) to yield 2,6-dichloroquinone (DCQ) in the presence of H2O2. The origin of the lower rate constants may be the solvent-exposed conformations of distal D55 and N55, which would have the dual effect of destabilizing the binding of H2O2 to the heme iron, and of removing the acid-base catalyst necessary for the heterolytic O-O bond cleavage of heme-bound H2O2 (i.e., compound 0). The partial peroxidase activity of H55D can be explained if one considers that there are two conformations of the distal aspartate (open and closed) by analogy with the distal histidine. We hypothesize that the distal aspartate has an active conformation in the distal pocket (closed). Although the open form is observed in the low-temperature X-ray crystal structure of ferric H55D, the closed form is observed in the FTIR spectrum of the carbonmonoxy form of the H55D mutant. Consistent with this model, the H55D mutant also shows inhibition of TCP oxidation by 4-bromophenol (4-BP). Consistent with the protection hypothesis, compound ES, the tyrosyl radical-containing ferryl intermediate observed in WT DHP A, was not observed in H55D.
Comprehensive identification of novel post-translational modifications in cellular peroxiredoxin 6

Abstract:Peroxiredoxin 6 (PRDX6), a 1-Cys peroxiredoxin, is a bifunctional enzyme acting both as a glutathione peroxidase and a phospholipase A2. However, the underlying mechanisms and their regulation mechanisms are not well understood. Because post-translational modifications (PTMs) have been shown to play important roles in the function of many proteins, we undertook, in this study, to identify the PTMs in PRDX6 utilizing proteomic tools including nanoUPLC-ESI-q-TOF MS/MS employing selectively excluded mass screening analysis (SEMSA) in conjunction with MOD(i) and MODmap algorithm. We chose PRDX6 obtained from liver tissues from two inbred mouse strains, C57BL/6J and C3H/HeJ, which vary in their susceptibility to high-fat diet-induced obesity and atherosclerosis, and a B16F10 melanoma cell line for this study. When PRDX6 protein samples were separated on 2D-PAGE based on pI, several PRDX6 spots appeared. They were purified and the low abundant PTMs in each PRDX6 spot were analyzed. Unexpected mass shifts (Δm = -34, +25, +64, +87, +103, +134, +150, +284 Da) observed at active site cysteine residue (Cys47) were quantified using precursor ion intensities. Mass differences of -34, +25, and +64 Da are presumed to reflect the conversion of cysteine to dehydroalanine, cyano, and Cys-SO(2) -SH, respectively. We also detected acrylamide adducts of sulfenic and sulfinic acids (+87 and +103 Da) as well as unknown modifications (+134, +150, +284 Da). Comprehensive analysis of these PTMs revealed that the PRDX6 exists as a heterogeneous mixture of molecules containing a multitude of PTMs. Several of these modifications occur at cysteine residue in the enzyme active site. Other modifications observed, in PRDX6 from mouse liver tissues included, among others, mono- and dioxidation at Trp and Met, acetylation at Lys, and deamidation at Asn and Gln. Comprehensive identification of the diverse PTMs occurring in this bifunctional PRDX6 enzyme should help understand how PRDX6 plays key roles in oxidative stresses.

Abstract:Peroxiredoxins (Prdxs) with a single peroxidative cysteine (Cp) in a conserved motif PXXX(T/S)XXCp within its thioredoxin fold, have been classified as the Prdx6 family. The single CP of Peroxiredoxin 6 (Prdx6) uses various external electron donors including glutathione, thioredoxin, and ascorbic acid for its peroxidase activity. Prdx6 proteins also exhibit Ca2+-independent phospholipase A2 (PLA2), lysophosphosphatidylcholine acyltransferase (LPCAT), and chaperone activities depending on cellular localization and the oxidation and oligomerisation states of the protein. Although there are wide variations in both the structure and function of the Prdx6 family members in various organisms and under varying physiological conditions, all Prdx6 proteins show the unique structural trait of a long C-terminal extension that is also seen in Prdx1, but not in other members of the Prdx family. Physiologically, Prdx6s play important roles in: protection against oxidative stress, cell membrane repair, mammalian lung surfactant turnover, activation of some nicotinamide adenine dinucleotide phosphate oxidases (NADPH oxidases types 1 and 2), and in regulation of seed germination in plants. Of crucial importance will be further evaluation of the role of Prdx6 enzymes in cancer spread and metastasis.This review describes the unique structural features of Prdx6 enzymes that are associated with its multifunctional physiological activities.

The Effect of Loudness Variation on Velopharyngeal Function in Children with 22q11.2 Deletion Syndrome: A Pilot Study

Abstract:Objective: Children with 22q11.2 deletion syndrome (22qDS) often require surgical intervention to treat velopharyngeal dysfunction (VPD). Although some studies have documented improved velopharyngeal (VP) closure under increased speaking effort, currently no studies have examined the effect of similar behavioral speech modifications on VP closure in children with 22qDS. The purpose of this pilot study was to explore the effect of loudness on VP closure during speech in children with 22qDS and persisting VPD. Patients and Methods: Four children with 22qDS, posterior pharyngeal flap, and persisting mild VPD underwent pressure-flow testing while repeating words at habitual and increased loudness levels. Using a single-subject A-B design, descriptive statistics and graphical measures were used to examine differences in VP orifice area (VPA) and timing of closure in the habitual versus loud condition. Results: Results were mixed. Median VPA decreased during some stimuli for 3 participants, but increased for 1 subject when speaking louder. Median duration of nasal airflow decreased for 3 participants in the loud condition. Conclusion: This study presents preliminary aerodynamic data regarding the plasticity of VP physiology in the 22qDS group. Further research is needed to determine how loudness impacts VP function in children with 22qDS.

Abstract:Peroxiredoxin 6 (Prdx6) is a multifunctional eukaryotic antioxidant enzyme. Mammalian Prdx6 possesses peroxidase activity against a wide range of organic and inorganic hydroperoxides, as well as exhibits phospholipase A2 (aiPLA2) activity, which plays an important role in the reduction of oxidized phospholipids and cell membrane remodeling. Exogenous Prdx6 has recently been shown to be able to penetrate inside the cell. We hypothesized that this entry may be due to the phospholipase activity of Prdx6. Experiments using exogenous Prdx6 in three cell lines (3T3, A549, RAW 264.7) demonstrated that it is the phospholipase activity that promotes its penetration into the cell. Overoxidation of Prdx6 led to a suppression of the peroxidase activity and a 3-to-4-fold growth of aiPLA2, which enhanced the efficiency of its transmembrane transport into the cells by up to 15 times. A mutant form of Prdx6-S32A with an inactivated phospholipase center turned out to be unable to enter the cells in both the reduced and oxidized state of the peroxidase active center. Previously, we have shown that exogenous Prdx6 has a significant radioprotective action. However, the role of phospholipase activity in the radioprotective effects of Prdx6 remained unstudied. Trials with the mutant Prdx6-S32A form, with the use of a total irradiation model in mice, showed a nearly 50% reduction of the radioprotective effect upon aiPLA2 loss. Such a significant decrease in the radioprotective action may be due to the inability of Prdx6-S32A to penetrate animal cells, which prevents its reduction by the natural intracellular reducing agent glutathione S-transferase (πGST) and lowers the efficiency of elimination of peroxides formed from the effect of ionizing radiation. Thus, phospholipase activity may play an important role in the reduction of oxidized Prdx6 and manifestation of its antioxidant properties.

Abstract:Peroxiredoxin 6 (Prdx6), a protein with both peroxidase and PLA2 activities, plays a critical role in antioxidant defense of lung and other organs. We investigated the relative importance of these 2 activities of Prdx6 in the “repair” of peroxidized cell membranes in pulmonary microvascular endothelial cells (PMVECs) and isolated mouse lungs. Lipid peroxidation was evaluated by measurement of thiobarbituric acid reactive substances (TBARS) or oxidation of diphenyl‐1‐pyrenylphosphine (DPPP). PMVECs were treated with tert‐butyl hydroperoxide (tBOOH, 0.2 mM) or Cu2+ /ascorbate for 4 h and then evaluated at 0,2,4 and 6 h after removal of the oxidant. Values for lipid peroxidation returned to control levels in 2 h with wild type (WT) PMVECs but were unchanged at 6 h after oxidant removal in Pxdx6 null PMVECs. An intermediate degree of repair was observed with lentivirus transfected C47S or D140A mutant Prdx6 PMVECs; the former express only PLA2 activity and the latter only peroxidase activity. Similar results were obtained with the isolated mouse lungs from WT, Prdx6 null and Prdx6‐C47S or ‐D140A “knock‐in” mice that were observed for 3 h following 60 min of perfusion with 15 mM tBOOH. These results indicate that the peroxidase and phospholipase A2 activities of Prdx6 both play a critical role in the repair of peroxidized phospholipids in cell membranes and the recovery of lung cells from peroxidative stress.

Abstract:Peroxiredoxins (Prxs) are members of the antioxidant enzymes necessary for every living object in the three domains of life and play critical roles in controlling peroxide levels in cells. This comprehensive literature review aims to elucidate the peroxidase activity of Prxs, examining their roles and significance for organisms across various taxa. Ironically, the primary role of the Prxs is the peroxidase activity, which comprises the reduction of hydrogen peroxide and other organic hydroperoxides and decreases the risk of oxidative damage in the cells. The above enzymatic activity occurs through the reversible oxidation-reduction catalyzed by cysteine residues in the active site by forming sulfenic acid and reduction by intracellular reductants. Structurally and functionally, Prxs function as dimers or decamers and show different catalytic patterns according to their subfamilies or cellular compartments. Compared to the mechanisms of the other two subgroups of Prxs, including 2-Cys Prxs and atypical Prxs, the 1-Cys Prxs have monomer-dimer switch folding coupled with catalytic activity. In addition to their peroxidase activity, which is widely known, Prxs are becoming acknowledged to be involved in other signaling processes, including redox signaling and apoptosis. This aversion to oxidative stress and regulation by the cellular redox state places them at the heart of adaptive cellular responses to changes in the environment or manifestations of diseases. In conclusion, based on the data obtained and on furthering the knowledge of Prxs' structure and function, these enzymes may be classified as a diverse yet essential family of proteins that can effectively protect cells from the adverse effects of oxidative stress due to peroxidase activity. This indicates secondary interactions, summarized as peroxide detoxification or regulatory signaling, and identifies their applicability in multiple biological pathways. Such knowledge is valuable for enhancing the general comprehension of essential cellular functions and disclosing further therapeutic approaches to the diseases caused by the increased production of reactive oxygen species.

Abstract:The expression of the phospholipase A2 activity (aiPLA2) of peroxiredoxin 6 (Prdx6) in the cell cytoplasm is physiologically relevant for the repair of peroxidized cell membranes, but aiPLA2 assay in vitro indicates that, unlike assay at pH 4, activity at cytosolic pH is essentially absent with non-oxidized substrate. However, the addition of glutathione (GSH) to the assay medium significantly increased aiPLA2 activity at cytosolic pH, while oxidized GSH (GSSG) and several other thiols had no effect. By mass spectroscopy (ESI MS), the addition of GSH to Prdx6 paradoxically led to oxidation of its conserved Cys47 residue to a sulfinic acid. The effect of GSH on PLA2 activity was abolished by incubation under anaerobic conditions, confirming that auto-oxidation of the protein was the mechanism for the GSH effect. Analysis by circular dichroism (CD) and tryptophan fluorescence showed alterations of the protein structure in the presence of GSH. Independently of GSH, the oxidation of Prdx6 by exposure to H2O2 or the presence of oxidized phospholipid as substrate also significantly increased aiPLA2 activity at pH 7. We conclude that the oxidation of the peroxidatically active Cys47 of Prdx6 results in an increase of aiPLA2 activity at pH 7 without effect on the activity of the enzyme at pH 4.

Abstract:Peroxiredoxin 6 (Prdx6), a bifunctional 25‐kDa protein with both peroxidase and phospholipase A2 (PLA2) activities, is expressed in all major organs with particularly high levels in the lung. Prdx6 can reduce phosphatidylcholine hydroperoxides with a high rate constant similar to H2O2 (~106M−1s−1). In vitro, recombinant Prdx6 binds only to oxidized phospholipid substrate at pH 7.0 but not to reduced substrate. Consistent with its antioxidant functions and preferential binding to oxidized phospholipids we hypothesized that Prdx6 can repair oxidized membrane without hydrolyzing reduced phospholipids. Plasma membranes (PM) preparations from A549 cells treated with either H2O2 (400μM) or t‐butyl hydroperoxide (800μM) for 6 hr showed translocation of Prdx6 as compared to PMs from control untreated cells. H2O2 treated A549 cells showed a 3.8 and 3.6 fold increase in DPPP fluorescence and TBARS, respectively, indicating lipid peroxidation. H2O2 induced enrichment of Prdx6 in PM, DPPP fluorescence, and TBARS values returned to basal levels when cells were either allowed to recover for 6 hr post H2O2 treatment or pretreated with catalase (100U/ml). In conclusion, using intact lung epithelial cells, we establish a mechanism by which Prdx6 mediates reduction of oxidized phospholipids such as those present in the plasma membrane of cells exposed to oxidative stress.

Abstract:Peroxiredoxin 6 (Prdx6) is a member of an evolutionary ancient family of peroxidase enzymes with diverse functions in the cell. Prdx6 is an important enzymatic antioxidant. It reduces a wide range of peroxide substrates in the cell, thus playing a leading role in the maintenance of the redox homeostasis in mammalian cells. Beside peroxidase activity, Prdx6 has been shown to possess an activity of phospholipase A2, an enzyme playing an important role in membrane phospholipid metabolism. Moreover, Prdx6 takes part in intercellular and intracellular signal transduction due to its peroxidase and phospholipase activity, thus facilitating the initiation of regenerative processes in the cell, suppression of apoptosis, and activation of cell proliferation. Being an effective and important antioxidant enzyme, Prdx6 plays an essential role in neutralizing oxidative stress caused by various factors, including action of ionizing radiation. Endogenous Prdx6 has been shown to possess a significant radioprotective potential in cellular and animal models. Moreover, intravenous infusion of recombinant Prdx6 to animals before irradiation at lethal or sublethal doses has shown its high radioprotective effect. Exogenous Prdx6 effectively alleviates the severeness of radiation lesions, providing normalization of the functional state of radiosensitive organs and tissues, and leads to a significant elevation of the survival rate of animals. Prdx6 can be considered as a potent and promising radioprotective agent for reducing the pathological effect of ionizing radiation on mammalian organisms. The radioprotective properties and mechanisms of radioprotective action of Prdx6 are discussed in the current review.
Structural Snapshots of Yeast Alkyl Hydroperoxide Reductase Ahp1 Peroxiredoxin Reveal a Novel Two-cysteine Mechanism of Electron Transfer to Eliminate Reactive Oxygen Species

Abstract:Peroxiredoxins (Prxs) are thiol-specific antioxidant proteins that protect cells against reactive oxygen species and are involved in cellular signaling pathways. Alkyl hydroperoxide reductase Ahp1 belongs to the Prx5 subfamily and is a two-cysteine (2-Cys) Prx that forms an intermolecular disulfide bond. Enzymatic assays and bioinformatics enabled us to re-assign the peroxidatic cysteine (C(P)) to Cys-62 and the resolving cysteine (C(R)) to Cys-31 but not the previously reported Cys-120. Thus Ahp1 represents the first 2-Cys Prx with a peroxidatic cysteine after the resolving cysteine in the primary sequence. We also found the positive cooperativity of the substrate t-butyl hydroperoxide binding to Ahp1 homodimer at a Hill coefficient of ∼2, which enabled Ahp1 to eliminate hydroperoxide at much higher efficiency. To gain the structural insights into the catalytic cycle of Ahp1, we determined the crystal structures of Ahp1 in the oxidized, reduced, and Trx2-complexed forms at 2.40, 2.91, and 2.10 Å resolution, respectively. Structural superposition of the oxidized to the reduced form revealed significant conformational changes at the segments containing C(P) and C(R). An intermolecular C(P)-C(R) disulfide bond crossing the A-type dimer interface distinguishes Ahp1 from other typical 2-Cys Prxs. The structure of the Ahp1-Trx2 complex showed for the first time how the electron transfers from thioredoxin to a peroxidase with a thioredoxin-like fold. In addition, site-directed mutagenesis in combination with enzymatic assays suggested that the peroxidase activity of Ahp1 would be altered upon the urmylation (covalently conjugated to ubiquitin-related modifier Urm1) of Lys-32.

Plasticity of the Peroxidase AhpC Links Multiple Substrates to Diverse Disulfide-Reducing Pathways in Shewanella Oneidensis

Abstract:AhpC is a bacterial representative of 2-Cys peroxiredoxins (Prxs) with broad substrate specificity and functional plasticity. However, details underpinning these two important attributes of AhpC remain unclear. Here, we studied the functions and mechanisms of regulation of AhpC in the facultative Gram-negative anaerobic bacterium Shewanella oneidensis, in which AhpC's physiological roles can be conveniently assessed through its suppression of a plating defect due to the genetic loss of a major catalase. We show that successful suppression can be achieved only when AhpC is produced in a dose- and time-dependent manner through a complex mechanism involving activation of the transcriptional regulator OxyR, transcription attenuation, and translation reduction. By analyzing AhpC truncation variants, we demonstrate that reactivity with organic peroxides (OPs) rather than H2O2 is resilient to mutagenesis, implying that OP reduction is the core catalytic function of AhpC. Intact AhpC could be recycled only by its cognate reductase AhpF, and AhpC variants lacking the Prx domain or the extreme C-terminal five residues became promiscuous electron acceptors from the thioredoxin reductase TrxR and the GSH reductase Gor in addition to AhpF, implicating an additional dimension to functional plasticity of AhpC. Finally, we show that the activity of S. oneidensis AhpC is less affected by mutations than that of its Escherichia coli counterpart. These findings suggest that the physiological roles of bacterial AhpCs are adapted to different oxidative challenges, depending on the organism, and that its functional plasticity is even more extensive than previously reported.

Abstract:Chromosome segregation in mitosis is orchestrated by dynamic interactions between spindle microtubules and centromeres, which in turn are governed by protein kinase- and phosphatase-signaling cascades. Previous studies showed that overexpression of human phosphatase HsCdc14A, an antagonist of cyclin-dependent kinase 1, affects several aspects of cell division. However, the molecular mechanism underlying HsCdc14A regulation in mitosis has remained elusive. Here we show that HsCdc14A activity is regulated by an auto-inhibitory mechanism via its intra-molecular association. Our biochemical study demonstrated that Polo-like kinase 1 (PLK1) interacts with and phosphorylates HsCdc14A. This phosphorylation partially releases the auto-inhibition of HsCdc14A judged by its phosphatase activity in vitro. To examine the functional relevance of such phospho-regulation of HsCdc14A in vivo, a phospho-mimicking mutant of HsCdc14A was expressed in HeLa cells. Importantly, overexpression of the phospho-mimicking mutants caused aberrant chromosome alignment with a prometaphase delay, suggesting the temporal regulation of HsCdc14A activity is critical for orchestrating mitotic events. Given the fact that HsCdc14A forms an intra-molecular association and PLK1-mediated phospho-regulation promotes HsCdc14A phosphatase activity, we propose that PLK1-HsCdc14A interaction provides a temporal regulation of HsCdc14A in chromosome segregation during mitosis.
Anti-oxidant and Pro-Oxidant Effects of Peroxiredoxin 6: A Potential Target in Respiratory Diseases

Abstract:Peroxiredoxin 6 (PRDX6) is widely distributed in several organs, especially the lungs. The role of PRDX6 in oxidative stress is controversial and even contradictory, as indicated by research conducted over the past 20 years. PRDX6 has anti-oxidant or pro-oxidant effects on oxidative stress in different diseases. It can even exhibit both anti-oxidant and pro-oxidant effects in the same disease. These findings are attributed to the fact that PRDX6 is a multifunctional enzyme. The peroxidase and phospholipase A2 activity of PRDX6 is closely related to its anti-oxidant and pro-oxidant effects, which leads to the conflicting regulatory effects of PRDX6 on oxidative stress in respiratory diseases. Moreover, PRDX6 interacts with multiple redox signaling pathways to interfere with cell proliferation and apoptosis. PRDX6 has become a new target in respiratory disease research due to its important regulatory role in oxidative stress. In this paper, the role of PRDX6 in oxidative stress in respiratory diseases and the research progress in targeting PRDX6 are reviewed.

Abstract:One of the physiological functions of cellular prion protein (PrP C ) is believed to work as a cellular resistance to oxidative stress, in which the octarepeats region within PrP plays an important role. However, the detailed mechanism is less clear. In this study, the expressing plasmids of wild-type PrP (PrP-PG5) and various PrP mutants containing 0 (PrP-PG0), 9 (PrP-PG9) and 12 (PrP-PG12) octarepeats were generated and PrP proteins were expressed both in E. coli and in mammalian cells. Protein aggregation and formation of carbonyl groups were clearly seen in the recombinant PrPs expressed from E. coli after treatment of H 2 O 2 . MTT and trypan blue staining assays revealed that the cells expressing the mutated PrPs within octarepeats are less viable than the cells expressing wild-type PrP. Statistically significant high levels of intracellular free radicals and low levels of glutathione peroxidase were observed in the cells transfected with plasmids containing deleted or inserted octarepeats. Remarkably more productions of carbonyl groups were detected in the cells expressing PrPs with deleted and inserted octarepeats after exposing to H 2 O 2 . Furthermore, cells expressing wild-type PrP showed stronger resistant activity to the challenge of H 2 O 2 at certain extent than the mutated PrPs and mock. These data provided the evidences that the octarepeats number within PrP is critical for maintaining its activity of antioxidation. Loss of its protective function against oxidative stress may be one of the possible pathways for the mutated PrPs to involve in the pathogenesis of familial Creutzfeldt-Jacob diseases.

Abstract:Peroxiredoxin 6 (Prdx6) is a Ca2+-independent intracellular phospholipase A2 (called aiPLA2) that is localized to cytosol, lysosomes, and lysosomal-related organelles. Activity is minimal at cytosolic pH but is increased significantly with enzyme phosphorylation, at acidic pH, and in the presence of oxidized phospholipid substrate; maximal activity with phosphorylated aiPLA2 is ∼2 µmol/min/mg protein. Prdx6 is a “moonlighting” protein that also expresses glutathione peroxidase and lysophosphatidylcholine acyl transferase activities. The catalytic site for aiPLA2 activity is an S32-H26-D140 triad; S32-H26 is also the phospholipid binding site. Activity is inhibited by a serine “protease” inhibitor (diethyl p-nitrophenyl phosphate), an analog of the PLA2 transition state [1-hexadecyl-3-(trifluoroethyl)-sn-glycero-2-phosphomethanol (MJ33)], and by two naturally occurring proteins (surfactant protein A and p67phox), but not by bromoenol lactone. aiPLA2 activity has important physiological roles in the turnover (synthesis and degradation) of lung surfactant phospholipids, in the repair of peroxidized cell membranes, and in the activation of NADPH oxidase type 2 (NOX2). The enzyme has been implicated in acute lung injury, carcinogenesis, neurodegenerative diseases, diabetes, male infertility, and sundry other conditions, although its specific roles have not been well defined. Protein mutations and animal models are now available to further investigate the roles of Prdx6-aiPLA2 activity in normal and pathological physiology.

Abstract:OBJECTIVE:To investigate the roles of peroxiredoxin (Prdx) 6 in the regulation of oxidative stress to lipopolysaccharide (LPS)-induced acute lung injury (ALI) in mice.METHODS:Prdx6 knockout mice were tested for their genotype by PCR and Prdx6 protein expression was measured in lungs by immunohistochemistry. Eighteen male Prdx6 knockout mice were randomised to the Prdx6 knockout control group and the Prdx6 knockout LPS 24 h group, with 9 mice in each group. Eighteen male wild-type C57BL/6J mice were randomised to the wild-type control group and the wild-type LPS 24 h group, with 9 mice in each group. ALI was induced by intratracheal administration of 5mg/kg LPS. Twenty-four hours after stimulation, lung tissue slides were stained with hematoxylin and eosin for histological evaluation. The concentration of protein in the bronchial alveolar lavage fluid (BALF) was measured by using the Micro BCA Protein Assay Kit. Levels of reactive oxygen species (ROS) in the lungs were quantified by measurement of hydrogen peroxide (H(2)O(2)). Lipid and protein peroxidation were measured as levels of malondialdehyde (MDA) and protein carbonylation. H(2)O(2), MDA, protein carbonyl, total superoxide dismutase (SOD) activity, and total antioxidative capacity (TAOC) in lungs were measured by using assay kits from the manufacture.RESULTS:Prdx6 knockout mice presented their genotype and there was no Prdx6 protein expression in the lung. Increased polymorphonuclear cells in alveoli and bronchial wall thickening were observed in the lungs of LPS groups, which were more severe in Prdx6 knockout LPS 24 h group compared with wild-type LPS 24 h group. LPS instillation induced a significant elevated protein concentration in BALF in wild-type LPS 24 h group (441 ± 54) mg/L compared with wild-type control group (168 ± 20) mg/L (t = -4.71, P < 0.01). Significantly increased protein level in BALF was observed in Prdx6 knockout LPS 24 h group (770 ± 66) mg/L compared with wild-type LPS 24 h group (t = -3.69, P < 0.01). LPS instillation induced a significantly decreased SOD activity in wild-type LPS 24 h group (16.0 ± 1.2) U/mg protein compared with wild-type control group (26.5 ± 3.9) U/mg protein (t = 6.22, P < 0.01). SOD activity in Prdx6 knockout LPS 24 h group (14.5 ± 5.3) U/mg protein was not statistical different from wild-type LPS 24 h group (t = 0.56, P = 0.60). LPS instillation induced a significantly increased H(2)O(2) and MDA in wild-type LPS 24 h group [H(2)O(2)(52.3 ± 7.8) nmol/g protein; MDA (3.3 ± 0.5) nmol/mg protein] compared with wild-type control group [H(2)O(2)(29.5 ± 3.2) nmol/g protein, (t = -4.25, P < 0.01); MDA (1.6 ± 0.8) nmol/mg protein, (t = -5.94, P < 0.01)]. Significantly increased H(2)O(2) and MDA [H(2)O(2)(73.5 ± 12.4) nmol/g protein, (t = -3.01, P = 0.02); MDA (5.9 ± 0.9) nmol/mg protein, (t = -6.01, P < 0.01)] were observed in Prdx6 knockout LPS 24 h group compared with wild-type LPS 24 h group. No significant difference of protein carbonylation was observed in wild-type LPS 24 h group (6.9 ± 1.2) nmol/mg protein compared with wild-type control group (6.1 ± 0.9) nmol/mg protein (t = -1.62, P = 0.15). Significantly increased protein carbonylation (8.9 ± 0.9) nmol/g protein was observed in Prdx6 knockout LPS 24 h group compared with wild-type LPS 24 h group (t = -2.76, P = 0.03). LPS instillation induced a significantly decreased TAOC in wild-type LPS 24 h group (4.7 ± 0.6) U/mg protein compared with wild-type control group (6.5 ± 0.4) U/mg protein (t = 3.35, P < 0.01). Significantly decreased TAOC was observed in Prdx6 knockout LPS 24 h group (3.9 ± 0.4)U/mg protein compared with wild-type LPS 24 h group (t = 2.44, P = 0.04). The above parameters were not statistically different between Prdx6 knockout control group and wild-type control group.CONCLUSION:Deletion of peroxiredoxin 6 exaggerated LPS-induced acute lung injury with increased oxidative stress.

Abstract:Peroxiredoxin 6 (Prdx6), a unique 1-Cys member of the peroxiredoxin family, exhibits peroxidase activity, phospholipase activity, and lysophosphatidylcholine acyltransferase (LPCAT) activity. Prdx6 has been known to be an important enzyme for the maintenance of lipid peroxidation repair, cellular metabolism, inflammatory signaling, and antioxidant damage. Growing research has demonstrated that the altered activity of this enzyme is linked with various pathological processes including central nervous system (CNS) disorders. This review discusses the distinctive structure, enzyme activity, and function of Prdx6 in different CNS disorders, as well as emphasizing the significance of Prdx6 in neurological disorders.
