Abstract:Constructed wetlands (CWs) amended with biochar have attracted much attention for nitrate removal treating secondary effluent. However, little is acknowledged about the linkage among the nitrate removal performance, microbial metabolic pathway of nitrate, and biochar properties. Herein, biochars pyrolyzed under 300°C, 500°C, and 700°C (BC300, BC500, and BC700, respectively) were used in CWs to reveal the relationship. Results showed that CWs amended with BC300 (59.73%), BC500 (53.27%), and BC700 (49.07%) achieved higher nitrogen removal efficiency, compared with the control (39.51%). Metagenomic analysis showed that biochars could enrich the genes, which encoded key enzymes (adenosine triphosphate production, and electrons generation, transportation, and consumption) involved in carbon and nitrate metabolism. Further, biochar pyrolyzed under lower temperature, with higher oxygen content, molar O/C ratio, and the electron donating capacity, in CWs could obtain higher nitrate removal efficiency. Overall, this research offers new understandings for the promotion of denitrification in CWs amended with biochar.

Abstract:The denitrification process in constructed wetlands (CWs) is responsible for most of the nitrous oxide (N2O) emissions, which is an undesired impact on the ecology of sewage treatment systems. This study compared three types of CWs filled with gravel (CW-B), gravel mixed with natural pyrite (CW-BF), or biochar (CW-BC) to investigate their impact on microbiota and genetic potential for N2O generation during denitrification under varying chemical oxygen demand (COD) to nitrate (NO3--N) ratios. The results showed that natural pyrite and biochar were superior in enhancing COD (90.6-91.2 %) and NO3--N removal (90.0-93.5 %) in CWs with a COD/NO3--N ratio of 9. The accumulation of NO2--N during the denitrification process was the primary cause of N2O emission, with the fluxes ranging from 95.6-472.0 μg/(m2·h) in CW-B, 92.9-400 μg/(m2·h) in CW-BF, and 54.0-293.3 μg/(m2·h) in CW-BC. The addition of biochar significantly reduced N2O emissions during denitrification, while natural pyrite had a lesser inhibitory effect on N2O emissions. The three types of substrates also influenced the structure of microbiota in the biofilm, with natural pyrite enriched nitrogen transformation microorganisms, especially for denitrifiers. Notably, biochar significantly enhanced the abundance of nosZ and the ratio of nosZ/(norB + norC), which are critical factors in reducing N2O emissions from CWs. Overall, the results suggest that the biochar-induced changes in microbiota and genetic potential during denitrification play a significant role in preventing N2O production in CWs, especially when treating sewage with a relatively high COD/NO3--N ratio.

Abstract:The interactive effects of both biochar (BC) and electrochemistry (EC) can affect nitrogen (N) removal process. However, little is known about how this function in constructed wetland (CW) systems. In this study, an electrochemically (EC) coupled BC-amended saturated subsurface vertical flow constructed wetland (BECW) systems were established to enhance nitrogen (N) removal. Other three CW systems: without BC and EC (CW); with EC only (ECW); and with BC only (BCW) were performed as controls. Results indicated that the total nitrogen (59.88%–93.03%) and nitrate‑nitrogen (83.14%–100%) of the BECW system were significantly enhanced (p < 0.05) compared with the control systems. Treated WWTP tail-water could meet Class-IV of the Surface Water Quality Standard (GB3838–2002) in China by the BECW system. The enhanced N removal in the BECW system could be attributed to (1) the autotrophic denitrification process in which H2 and Fe2+ provided by the cathode and anode acted as electron donors; (2) BC addition acting as substrate could improve the activity, diversity and richness of microorganisms. Microbial community analysis further indicated that high N removal in the BECW system was significantly dependent on the synergy between the heterotrophic and autotrophic denitrifiers, facilitated by BC and EC interaction. Results illustrate that the BECW system is a feasible and eco-sustainable technology for treating low C/N tail-water from WWTPs. This work provides a novel and fundamental understanding of the electrochemically coupled biochar-amended CW system. These results could serve as a theoretical basis for the engineered applications in the deep purification of WWTPs' tail-water.

Value of excretory cystography in the diagnosis of bladder tumours.

Abstract:The value of excretory cystography in diagnosing bladder tumours was assessed in a randomised study. The 10-min, 20-min, and post-voiding films of intravenous urograms of 75 patients with biopsy-proved bladder tumours and of 25 patients with proved normal bladders were randomly reviewed. Overall, 60 of 75 bladder tumours (80%) were correctly diagnosed by review of the roentgenograms. Specificity and sensitivity were comparable (94 and 96% respectively) for patients with papillary tumours greater than or equal to 0.5 cm in diameter, but specificity was low (23%) for those with tumours smaller than 0.5 cm. It is concluded that excretory cystography makes a considerable contribution to the establishment of the diagnosis of papillary bladder tumours greater than or equal to 0.5 cm in diameter.

Abstract:Wastewater polluted by heavy metals with low C/N ratio posed a big challenge to constructed wetlands (CWs) for sustainable advanced wastewater purification. In this study, biochar coupled with pyrite was successfully applied in CWs to simultaneously remove organics, total nitrogen (TN), total phosphorus (TP) and heavy metals, with the highest removal efficiency of 95.78%, 98.78%, 96.22% and 99.91%, respectively. Batch tests proved that the complexation of ferric hydroxide from pyrite oxidation played an important role in TP and heavy metals removal. Meanwhile, fluorescence excitation-emission matrix combined with parallel factor analysis (EEM-PARAFAC) further manifested that protein-like substances contributed a lot to heavy metals stabilization. Moreover, the multiplication of Proteobacteria , Chloroflexi at phylum level and Dechloromonas at genus level, which were capable of heterotrophic or autotrophic nitrate reducing process indicated that the denitrification capacity of CWs was significantly enhanced by biochar and pyrite. This study provided a feasible insight into the application of CWs with biochar and pyrite addition to enhance the treatment of low C/N wastewater.

Abstract:Biochar is considered as black gold in environmental remediation field due to its high surface area, light density, great adsorption capacity. Biochar has high potential to improve the wastewater purification in constructed wetlands (CWs), which has been increasingly investigated in this decade. This paper reviews the characteristics of biochar, methods of preparation, pollutant removal effects, and the greenhouse gas (GHG) emission reduction in CWs systematically, which could guide further studies of biochar application in CWs. It was found that the biochar applied in CWs were mainly produced by pyrolysis of terrestrial plants, while the biochar produced from marine biology or by other procedures may have different performance due to the various properties, which needed further investigation. Biochar affected CW performance through multiple pathways, including influencing the physicochemical & hydraulic conditions and biological behavior in CWs. The quantified enhancement of pollutant removal by biochar addition in CWs was firstly summarized. Research data proved that medians of COD, ammonia and TN removal in biochar-added CWs were 109%, 145% and 178% as much as those in non-biochar CWs, respectively. Moreover, the most significant improvement showed under oxygen-shortage condition in CWs, rather than aeration condition. However, it remains unclear which pathway dominates the enhanced pollutant removal. Besides, biochar commonly decreased N2O emission from CWs, while the opposite results were also reported and a comprehensive explanation has not been reached. Therefore, future mechanism studies are needed to advance our understanding on biochar's behavior in CWs for wastewater treatment.

Fe-modified biochar enhances microbial nitrogen removal capability of constructed wetland

Abstract:To improve the nitrogen removal capability of constructed wetlands, the biochar, produced from bamboo, activated with HCl and coated with Fe (FeCl3·6H2O), and then was added as a substrate into the systems. Three horizontal subsurface flow constructed wetlands (HSCWs) was established to treat the low C/N tailwater from the wastewater treatment plant: C-HSCW (quartz sand + soil), B-HSCW (quartz sand + soil + unmodified biochar), and FeB-HSCW (quartz sand + soil + Fe-modified biochar). Under different combinations of hydraulic retention time and nitrogen loading, the FeB-HSCW revealed extremely effective nitrogen removal, compared to the C-HSCW and B-HSCW. The highest removal efficiencies of NO3--N (95.30%), TN (86.68%), NH4+-N (86.33%), NO2--N (79.35%) and COD (63.36%) were obtained in FeB-HSCW with the hydraulic retention time of 96 h. and low influent nitrogen loading (C/N of 2.5). Nitrogen mass balance analysis showed that microbial processes played the most important role of nitrogen removal in HSCWs and the Fe-modified biochar significantly enhanced the microbial nitrogen removal. A total of 128.40 g nitrogen was removed by microorganisms in FeB-HSCW (average removal rate of 2.52 g N/(m3·d1)), much higher than that in other two HSCWs. The contributions of microorganisms, substrate storage and plant uptake on the total amount of nitrogen removal in the FeB-HSCW was 92.69%, 2.97% and 4.34%, respectively. Moreover, FeB significantly increased the abundances of genes involved in nitrogen removal. The copy numbers of bacterial 16S rRNA and amx, as well as of genes nirS, nirK, nosZ-I, nosZ-II, and hzsA were 1.3- to 27.8-fold higher in the FeB-HSCW than that in the other two HSCWs. Thus, Fe-modified biochar provides a feasible and effective amendment for constructed wetlands to improve the nitrogen removal, particularly nitrate-N, for low C/N wastewaters by enhancing the microbial nitrogen removal capacity (mainly of the denitrification).

Abstract:Constructed wetlands (CWs) are well recognized as a cost-effective and environmentally friendly eco-technology for contaminated water remediation, especially in decentralized communities and rural areas. As a critical component in CWs, the substrate determines the wastewater treatment performance and ecological effects of CW systems. Biochar, a versatile and green carbonaceous biomaterial produced by carbonization of various biomasses, has recently been amended into CWs or biofilters (BFs) as a novel alternative substrate for wastewater treatment. In this paper, the performance of biochar amendment in CWs/biofilters with respect to the removal of nitrogen, phosphorus, organic contaminants, heavy metals and pathogens and its concurrent ecological benefits in improving macrophyte growth and mitigating greenhouse gas (GHG) emissions are summarized and evaluated. Furthermore, the mechanisms involved in these benefits, which are considerably different from those in batch tests, are analysed and elucidated, especially highlighting the roles of microorganisms. The high potential for recycling spent biochar media in CWs/BFs as a soil ameliorant/remediator is proposed. Collectively, it is highly promising to apply biochar as a substrate in CW/BF treatment systems, and more relevant research and practical applications of biochar substrates in CWs/BFs, especially the durability, recycling and possible negative effects of biochar, are required in the future.

Abstract:Biochar was utilized to intensify constructed wetland (CW) for further organic and nitrogen removal from secondary wastewater. Four sets of non-aerated biochar amended vertical flow CW (VFCW) were developed to investigate the synergistic effects of biochar and microbes on pollutant removal. Results showed that the average COD and nitrogen removal efficiencies of VFCW1 (with 1% w/w biochar with microbe and plants) achieved 89.1±5.6% and 90.2±3.1% respectively, and their corresponding removal rates of 10.2±0.8 mg-COD/(m3.d) and 3.57±0.3 mg-TN/(m3.d) which were 35 and 52.3% higher than control. The biochar's dissolved organic carbon release in VFCWs indicated that water and acidic media portray the optimum conditions for nitrogen removal. The 16S RNA gene sequencing analysis indicated that in the biochar-amended VFCWs, bacterial phylum Proteobacteria (24.13-51.95%) followed by Chloroflexi (5.64-25.01%), Planctomycetes (8.48-14.43%), Acidobacteria (2.29-11.65%) were abundantly enhanced. Conclusively, incorporating biochar in non-aerated VFCWs is an efficient technique for enhancing nitrogen removal from secondary effluent.

Abstract:Traditional constructed wetlands (CWs) still face tremendous challenges, including low nitrogen removal, unstable operation, undesired refractory substances degradation and poor reoxygenation capacity, in their practical applications. Recently, biochar has been regarded as a promising alternative substrate in CWs to enhance pollutants removal via inner environment adjustment, plants growth facilitating and biotoxins adsorption. Based on VOSviewer analysis, this study comprehensively summarizes the mechanisms, application, and performances of biochar addition in CWs. First, physicochemical properties of biochar, including pore sizes/pore volumes, surface aeras, functional groups and chemical compositions, with different preparation methods were comparatively analyzed. Then, the mechanisms and applications of (modified) biochar in CWs and its improving effects on typical pollutants (such as nitrogen, phosphorus, complex organics and heavy metals) removal were summarized. Moreover, the mechanisms and strengthened effect of novel Fe-C micro-electrolysis substrates on pollutants reduction in CWs was also elucidated. Finally, the prospectives and challenges were proposed to offer a deep insight in biochar for its engineering application in CWs.

Abstract:Biochar (BC) has been widely utilized in constructed wetland (CW) for intensifying the removal of pollutants from low carbon/nitrogen (C/N) wastewater. However, there are knowledge gaps about the optimal proportion of BC as an external carbon substrate. This study developed four sets of aerated vertical flow (VF) CWs with different BC amendment proportions (0%, 25%, 50%, 75%) to explore the influence of BC dosage on the treatment performance, enzyme activity and microbial community. The results showed that all CWs achieved better COD (above 90%) and NH4+-N (above 99%) removal due to adequate oxygen supplement, while the best TN removal (67%) was obtained in CW with 50% BC addition. TP removal ascended gradually with the increasing BC amendment proportion. The activity of enzymes and the proportion of Proteobacteria and Patescibacteria at the phylum level were appreciably enhanced with the addition of BC.

Abstract:Constructed wetlands (CWs) are frequently used for effective biological treatment of nitrogen-rich wastewater with external carbon source addition; however, these approaches often neglect the interaction between plant litter and biochar in biochar-amended CW environments. To address this, we conducted a comprehensive study to assess the impacts of single or combined addition of common reed litter and reed biochar (pyrolyzed at 300 and 500 °C) on nitrogen removal, greenhouse gas emission, dissolved organic matter (DOM) dynamics, and microbial activity. The results showed that combined addition of reed litter and biochar to CWs significantly improved nitrate and total nitrogen removal compared with biochar addition alone. Compared to those without reed litter addition, CWs with reed litter addition had more low-molecular-weight and less aromatic DOM and more protein-like fluorescent DOM, which favored the enrichment of bacteria associated with denitrification. The improved nitrogen removal could be attributed to increases in denitrifying microbes and the relative abundance of functional denitrification genes with litter addition. Moreover, the combined addition of reed litter and 300 °C-heated biochar significantly decreased nitrous oxide (30.7 %) and methane (43.9 %) compared to reed litter addition alone, while the combined addition of reed litter and 500 °C-heated biochar did not. This study demonstrated that the presences of reed litter and biochar in CWs could achieve both high microbial nitrogen removal and relatively low greenhouse gas emissions.

Abstract:Microcystins (MCs) pollution caused by eutrophication and climate change has posed a serious threat to ecosystems and human health. Constructed wetlands (CWs) with biochar addition volume ratios of 0% (BC0-CWs), 10% (BC10-CWs), 20% (BC20-CWs) and 50% (BC50-CWs) were set up to evaluate the efficiency of biochar-amended CWs for eutrophication and MCs pollution control. The results illustrated that removal efficiencies of both NH4+-N and NO3--N were enhanced by biochar addition to varying degrees. The average TP and MC-LR removal efficiencies increased with increasing biochar addition ratios, and the average TP and MC-LR removal efficiencies in biochar-amended CWs were significantly (p < 0.05) improved by 5.64-9.58% and 10.74-14.52%, respectively, compared to that of BC0-CWs. Biochar addition changed the microbial community diversity and structure of CWs. The relative abundance of functional microorganisms such as Burkholderiaceae, Nitrospiraceae, Micrococcaceae, Sphingomonadaceae and Xanthomonadaceae was promoted by biochar addition regardless of addition ratios. The higher relative abundance of the above microorganisms in BC20-CWs and BC50-CWs may contribute to their better removal performance compared to other CWs. The concentrations of extracellular polymeric substance (EPS) in biochar-amended CWs were significantly (p < 0.05) lower than that in BC0-CWs, which can reduce the risk of system clogging. This study demonstrated that biochar addition may be a potential intensification strategy for eutrophication and MCs pollution control by CWs. Considering both the removal performance and economic cost, a biochar addition ratio of 20% was recommended as an optimal addition ratio in practical application.

Abstract:Denitrification is an important biological process in wastewater treatment plants (WWTPs). However, a low carbon-to-nitrogen (C/N) ratio limits the availability of organic carbon, potentially reducing denitrification efficiency. This study investigates the impact of sludge-derived biochar on the nitrogen removal of activated sludge for low C/N ratio municipal wastewater. Sludge-based biochar was characterized by its physicochemical properties, revealing that biochar prepared at 400 °C exhibited the highest specific surface area and the most favorable surface functional groups for electron transfer. The results from batch tests showed that adding 4 g/L of biochar dosage enhanced denitrification rates and total nitrogen (TN) removal efficiency the most. Sequencing batch reactors (SBRs) experiments further confirmed that biochar dosgae improved the removal efficiencies of COD, NH 4 + -N, and TN, achieving stable values of 97.2 ± 1.2 %, 99.2 ± 0.6 %, and 83.8 ± 2.4 %, respectively. Metabolic and electrochemical analyses revealed that biochar addition enhanced the activity of denitrification enzymes, increasing the ammonia oxidation rate by 12.9 ± 0.7 %, nitrite oxidation rate by 14.7 ± 1.2 %, nitrate reduction rate by 36.9 ± 1.5 %, and nitrite reduction rate by 16.4 ± 0.8 %. The relative abundance of denitrification functional genes ( amoA, nirS, nirK, narG, nosZ ) increased, and the activities of the corresponding enzymes (AMO, NXR, NAP, NIR) rose by 23±6 %, 53±5 %, 260±15 %, and 55±7 %, respectively. This increase in enzyme activity suggested enhanced denitrification processes, which was further supported by the 60.1 ± 3.7 % increase in electron transfer system activity (ETSA), indicating that biochar acted as an electron shuttle. This study proposes a potential sustainable approach for sludge recycling and enhanced wastewater nitrogen removal under low C/N conditions.

Abstract:Considering the unsatisfied denitrification performance of carbon-limited wastewater in iron-based constructed wetlands (ICWs) caused by low electron transfer efficiency of iron substrates, utilization of plant-based conductive materials in-situ for improving the long-term reactivity of iron substrates was proposed to boost the Fe (III)/Fe (II) redox cycle thus enhance the nitrogen elimination. Here, we investigated the effects of withered Iris Pseudacorus biomass and its derived biochar on nitrogen removal for 165 days in ICWs. Results revealed that accumulate TN removal capacity in biochar-added ICW (BC-ICW) increased by 14.7% compared to biomass-added ICW (BM-ICW), which was mainly attributed to the synergistic strengthening of iron scraps and biochar. The denitrification efficiency of BM-ICW improved by 11.6% compared to ICWs, while its removal capacity declined with biomass consumption. Autotrophic and heterotrophic denitrifiers were enriched in BM-ICW and BC-ICW, especially biochar increased the abundance of electroactive species ( Geobacter and Shewanella , etc.). An active iron cycle exhibited in BC-ICW, which can be confirmed by the presence of more liable iron minerals on iron scraps surface, the lowest Fe (III)/Fe (II) ratio (0.51), and the improved proportions of iron cycling genes ( feoABC, korB, fhuF, TC.FEV.OM , etc.). The nitrate removal efficiency was positively correlated with the nitrogen, iron metabolism functional genes and the electron transfer capacity (ETC) of carbon materials ( P < 0.05), indicating that redox-active carbon materials addition improved the iron scraps bioavailability by promoting electron transfer, thus enhancing the autotrophic nitrogen removal. Our findings provided a green perspective to better understand the redox properties of plant-based carbon materials in ICWs for deep bioremediation in-situ .

Abstract:Nitrous oxide (N2O) emissions from constructed wetlands (CWs) are accompanying problems and have attracted much attention in recent years. CWs filled with different substrates (gravel, biochar, zeolite, and pyrite) were constructed to investigate the nitrogen removal performance and N2O emissions, which named C-CWs, B-CWs, Z-CWs, and P-CWs, respectively. C-CWs showed the poorest nitrogen removal performance in all CWs. Although B-CWs exhibited the highest fluxes of N2O emissions, the percentage of N2O emissions in nitrogen removal (0.15%) was smaller than that of C-CWs (0.18%). In addition, microbiological analysis showed that compared with C-CWs, CWs filled with biochar, zeolite, and pyrite had higher abundance of nitrifying and denitrifying microorganisms and lower abundance of N2O producing bacteria. In conclusion, biochar, zeolite, and pyrite were more favorable kinds of substrate than the conventional substrates of gravel for the nitrogen removal and reduction of N2O emissions from CWs.

Abstract:Constructed wetland (CW) is an eco-friendly and economically treatment for the deep nitrogen removal . Microbial denitrification, a key process in CW, is temperature-sensitive, with decreased nitrogen removal efficiency at low temperature. To enhance the nitrogen removal efficiency at low temperature, this study explored the use of functional substrates, focusing on iron sulfide (FeS) nanoparticles and biochar (BC). The results showed that immobilized BC/FeS significantly improved nitrate (NO 3 -N) and total nitrogen (TN) removal in CW at low temperature (8–13 °C), which reached 93.59 ± 2.07 and 80.06 ± 3.00 %, respectively, with 30 % immobilized BC/FeS. Increasing the dosage of immobilized BC/FeS accelerated denitrification rate and reduced nitrite (NO 2 -N) accumulation. Autotrophic denitrification was the primary pathway, with the contribution of autotrophic denitrification increasing from 64.06 % (10 % immobilized BC/FeS) to 83.25 % (30 % immobilized BC/FeS). Sulfurimonas was the dominant denitrifying bacteria identified as a cold-tolerant denitrifying bacteria. The study underlines the potential of composite electron donor substrates to enhance nitrogen removal in CW, providing a sustainable solution to nitrogen pollution in cold climates.

Abstract:We have determined the nucleotide sequence of three positive single-stranded RNA coliphages and have used this information, together with the known sequences of the related phages Q beta and SP, to construct a secondary structure model for the two distal domains of Q beta RNA. The 3' terminal domain, which is about 100 nucleotides long, contains most of the 3' untranslated region and folds into four short, regular hairpins. The adjacent 3' replicase domain contains about 1100 nucleotides. Hairpins in this protein-coding domain are much longer and more irregular than in the 3' untranslated region. Both domains are defined by long-distance interactions. The secondary structure is not a collection of hairpin structures connected by single-stranded regions. Rather, the RNA stretches between the stem-loop structures are all involved in an extensive array of long-distance interactions that contract the molecule to a rigid structure in which all hairpins are predicted to have a fixed position with respect to each other. A general feature of the model is that helices tend to be organized in four-way junctions with little or no unpaired nucleotides between them. As a result, there is a potential for coaxial stacking of adjacent stems. The essential features of the model are supported by the S1 nuclease cleavage pattern. Viral RNA sequences are strongly constrained by their coding function. As a result, structural evolution by simple base-pair substitution is not always possible, as this usually requires the juxtaposition of the codon wobble positions in stems. Rather, we often observe co-ordinate base substitutions that maintain the stem, but tend to change the position at which bulges or internal loops are found. Structures that differ in this way are apparently equally fit. Also, the relative position of hairpin loops can shift several nucleotides through an alignment based on maximal sequence identity i.e. amino acid homology. The fact that these structural irregularities do not occur at the 3' untranslated region suggests indeed that the coding function of the RNA constrains the secondary structure. Hairpins with the stable tetraloop motif GNRA and UNCG or their complement are over-represented. This suggests their involvement in segregation of plus and minus strand. The genome of the coliphages contains a well-defined high affinity binding site for the coat protein, which serves to suppress replicase translation and also acts as a nucleation point in capsid formation. Close to the 3' end we find an additional conserved helix that meets the described consensus criteria for coat-protein binding.
