Abstract:The quest for sustainable strategies aimed at increasing the bioactive properties of plant-based foods has grown quickly. In this work, we investigated the impact of exogenously applied phenolics, i.e., chlorogenic acid (CGA), hesperidin (HES), and their combinations (HES + CGA), on Lactuca sativa L. grown under normal- and mild-salinity conditions. To this aim, the phenolic profile, antioxidant properties, and enzyme inhibitory activity were determined. The untargeted metabolomics profiling revealed that lettuce treated with CGA under non-stressed conditions exhibited the highest total phenolic content (35.98 mg Eq./g). Lettuce samples grown under salt stress showed lower phenolic contents, except for lettuce treated with HES or HES + CGA, when comparing the same treatment between the two conditions. Furthermore, the antioxidant capacity was investigated through DPPH (2,2-diphenyl-1-picrylhydrazyl), ABTS (2,20-azinobis-(3-ethylbenzothiazoline-6-sulfonate)), and FRAP (ferric reducing antioxidant power) assays, coupled with metal-chelating activity and phosphomolybdenum capacity. An exciting increase in radical scavenging capacity was observed in lettuce treated with exogenous phenolics, in both stress and non-stress conditions. The inhibitory activity of the samples was evaluated against target health-related enzymes, namely cholinesterases (acetylcholinesterase; AChE; butyryl cholinesterase; BChE), tyrosinase, α-amylase, and α-glucosidase. Lettuce treated with HES + CGA under non-stress conditions exhibited the strongest inhibition against AChE and BChE, while the same treatment under salinity conditions resulted in the highest inhibition capacity against α-amylase. Additionally, CGA under non-stress conditions exhibited the best inhibitory effect against tyrosinase. All the functional traits investigated were significantly modulated by exogenous phenolics, salinity, and their combination. In more detail, flavonoids, lignans, and stilbenes were the most affected phenolics, whereas glycosidase enzymes and tyrosinase activity were the most affected among enzyme assays. In conclusion, the exogenous application of phenolics to lettuce represents an effective and green strategy to effectively modulate the phenolic profile, antioxidant activity, and enzyme inhibitory effects in lettuce, deserving future application to produce functional plant-based foods in a sustainable way.

Abstract:The roles of nitrogen availability in determining the phenolic accumulation of vegetables have been widely studied, but the underlying mechanism involved remains unknown. Thus, primary and secondary metabolites profiling of lettuce leaves were performed using non-targeted metabolomics analysis. The results showed that carbon metabolism, amino acid metabolism, and phenolic biosynthesis metabolism in lettuce were significantly affected by low nitrogen supply (LN). The phenolic content was significantly increased in LN-treated lettuce, indicating that the activated phenolic biosynthesis was triggered by the LN treatment. The reduced citrate cycle and enhanced glucose and sucrose content suggested there is a relative excess of carbon resources in LN-treated lettuce. In addition, the decreased nitrogen-rich amino acids (glutamine and aspartate acid) and the maintained phenylalanine content indicated the redirection of nitrogen resources to phenylalanine biosynthesis. Meanwhile, no significant changes of chlorophyll content were observed in LN-treated lettuce leaves. The LN-treated lettuce showed lower glutamine synthetase activity but higher glutamate synthase activity compared to control. These findings together suggest that LN treatment may increase the phenolic accumulation in lettuce by effectively redirecting more carbon and nitrogen resources to the phenolic biosynthesis pathway.
Enhancement of Polyphenolic Metabolism As an Adaptive Response of Lettuce (lactuca Sativa) Roots to Aluminum Stress.

Abstract:Polyphenols, pivotal secondary metabolites, are involved in plant adaption to abiotic stresses. Here, we investigated the role and metabolism profile of polyphenols under aluminum (Al) stress in different lettuce genotypes grown in 0.5 mM CaCl2 solution with AlCl3 (pH = 4.5). The complementary use of high-resolution mass spectrometry and quantitative biochemical approaches allowed the characterization of total and unique phenols, as well as their roles in Al tolerance. By comparing the most tolerant and sensitive genotype, 8 polyphenols, including 4 phenolic acids, 2 flavonoids, 1 xanthone and 1 unknown compound, were identified in the roots of the tolerant genotype. The total phenolic and flavonoid contents significantly increased in the tolerant genotype under Al stress. Seedlings with more phenolic accumulation usually performed greater Al tolerance. Meanwhile, principal enzymes related to phenolic biosynthesis significantly increased in roots of the tolerance genotype after Al treatment, with phenylalanine ammonia lyase (PAL), cinnamate 4-hydroxylase, and 4-coumarate coenzyme A ligase increased by 16,18 and 30%, respectively. The elevated total phenolics were significantly suppressed by AIP, a highly specific PAL inhibitor. Consequently, the antioxidant capacity was inhibited, leading to lettuce sensitivity to Al stress. These results clearly suggested the enhancement of unique polyphenolic biosynthesis as an adaptive strategy of lettuce to Al stress by protecting plants from oxidative stress. (C) 2020 Elsevier Ltd. Role of Sucrose in Modulating the Low-Nitrogen-induced Accumulation of Phenolic Compounds in Lettuce (lactuca Sativa L.).

Abstract:BACKGROUND Phenolic compounds are phytochemicals present in vegetables which contribute to human health. Although nitrogen deficiency and sucrose (Suc) are linked to phenolic production in vegetables, the relationship between them in the regulation of phenolic biosynthesis remains unknown. This study investigated the potential role of Suc in regulating phenolic biosynthesis of lettuce under low-nitrogen (LN) conditions. RESULTS Our results showed that LN treatment significantly increased Suc content in lettuce by inducing rapid increases in activities of sucrose synthesis-related enzymes. Exogenous Suc further stimulated LN-induced phenolic accumulation in lettuce by upregulating the expression of genes (PAL,CHS,F3H,DFR,F35HandUFGT) involved in phenolic biosynthesis. The opposite effects were true for exogenous 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU) application. No changes were observed in chlorophyll content in LN-treated lettuce, in either the presence or absence of Suc application. Notably, exogenous DCMU resulted in decreases of maximum quantum efficiency of photosystem II (PSII) photochemistry, actual efficiency of PSII and electron transport rate in PSII and increase of quantum yield of non-regulated energy dissipation in PSII in lettuce under LN conditions, whereas these effects were reversed on Suc application. Exogenous Suc also increased glutamine synthetase and glutamate synthase activities in LN-treated lettuce. CONCLUSIONS These results suggest that Suc is involved in LN-induced phenolic production in lettuce by enhancing photosynthetic and nitrogen assimilation efficiency to increase the supply of carbon resources and precursors for phenolic biosynthesis. Abstract:Intensive and indiscriminate use of insecticides in agroecosystems causes phytotoxic disturbances in non-target crops. However, the mechanisms by which plants reprogram cellular metabolites to resist and tolerate such agrochemicals remain unclear. Here, the interaction between lettuce plants with imidacloprid and fenvalerate was investigated by the complementary use of physiological and metabolomic analyses. Neither imidacloprid nor fenvalerate induced overt phytotoxicity in lettuce seedlings. The plant biomass, chlorophyll fluorescence, lipid peroxidation, and membrane integrity were not significantly affected by the selected insecticides. Flavonoid content decreased by 25% in lettuce leaves under fenvalerate exposure, whereas polyphenol and flavonoid contents were not significantly altered by imidacloprid. Although the content of most of the nutrient element in the leaves remained the same following pesticide treatment, iron content decreased by 28.1% under imidacloprid exposure but increased by 22.8% under fenvalerate exposure. Metabolomic analysis revealed that the selected insecticides induced extensive metabolic reprogramming in lettuce roots and shoots. Imidacloprid dramatically increased the metabolism of several amino acids (arginine, cysteine, homoserine, and 4-hydroxyisoleucine), whereas markedly decreased the metabolism of various carbohydrates (glucose, raffinose, maltotetraose, maltopentaose, and stachyose). Fenvalerate did not significantly alter amino acid metabolism but decreased carbohydrate metabolism. Additionally, the relative abundance of most organic acids and polyphenolic compounds decreased significantly after pesticide exposure. These results suggest that plants might program their primary and secondary metabolism to resist and tolerate insecticides. The findings of this study provide important information on how neonicotinoid and pyrethroid insecticides affect the health and physiological state of plants, which are ultimately associated with crop yield and quality.
The Strength of the Nutrient Solution Modulates the Functional Profile of Hydroponically Grown Lettuce in a Genotype-Dependent Manner

Abstract:Considering that functional components of plant foods are mainly secondary-metabolism products, we investigated the shaping of health-promoting compounds in hydroponically grown butterhead lettuce (Lactuca sativa L. var. capitata) as a function of the strength of the nutrient solution utilized. To this aim, untargeted metabolomics profiling, in vitro antioxidant capacity (total phenolics, 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), cupric reducing antioxidant capacity (CUPRAC), and ferric reducing antioxidant power (FRAP) assays), and the inhibition of selected enzyme activities were investigated in two butterhead lettuce cultivars with different pigmentation, i.e., green and red Salanova. Full-strength nutrition, together with half- and quarter-strength solutions of macronutrients, was tested. Our results indicate that by reducing the nutrients strength, we could elicit a distinctive shaping of the phenolic profile of lettuce. It is noteworthy that only specific classes of phenolics (namely, lignans and phenolic acids, followed by flavones and anthocyanins) were modulated by the induction of nutritional eustress (fold-change values in the range between −5 and +11). This indicates that specific responses, rather than a generalized induction of phenolic compounds, could be observed. Nonetheless, a genotype-dependent response could be observed, with the red cultivar being much more responsive to nutritional deprivation than the green Salanova lettuce. Indeed, analysis of variance (ANOVA) confirmed a genotype x nutrition interaction in red Salanova (p < 0.001). As a consequence of the changes in phenolic composition, also the antioxidant capacity (p < 0.001) and amylase inhibition (p < 0.001) properties were affected by the growing conditions. However, the effect on cholinesterase and tyrosinase inhibition was poorly affected by the nutritional strength. Provided that yields are not compromised, the application of a controlled nutritional eustress in hydroponically cultivated lettuce may represent a valuable strategy to produce food with tailored functional features in a sustainable manner.

Effect of glycine nitrogen on lettuce growth under soilless culture: a metabolomics approach to identify the main changes occurred in plant primary and secondary metabolism

Abstract:Background: Lettuce is a significant source of antioxidants and bioactive compounds. Nitrate is a cardinal fertilizer in horticulture and influences vegetable yield and quality; however, the negative effects of nitrate on the biosynthesis of flavonoids require further study. It is expected that using fertilizers containing organic nitrogen (N) could promote the synthesis of health-promoting compounds. Results: Lettuces were hydroponically cultured in media containing 9 mmol L-1 nitrate or 9 mmol L-1 glycine for 4 weeks. Primary and secondary metabolites were analyzed using gas chromatography/mass spectrometry (GC/MS) and ultra-performance liquid chromatography/ion mobility spectrometry/quadrupole time-of-flight mass spectrometry (UPLC/IMS/QTOF-MS). Data analysis revealed that 29 metabolites were significantly altered between nitrate and glycine treatments. Metabolites were tentatively identified by comparison with online databases, literature and standards and using collision cross-section values. Significant differences in flavonoid biosynthesis, phenolic biosynthesis and the tricarboxylic acid (TCA) cycle response were observed between N sources. Compared with nitrate, glycine promoted accumulation of glycosylated flavonoids (quercetin 3-glucoside, quercetin 3-(6″-malonyl-glucoside), luteolin 7-glucuronide, luteolin 7-glucoside), ascorbic acid and amino acids (l-valine, l-leucine, l-glutamine, asparagine, l-serine, l-ornithine, 4-aminobutanoic acid, l-phenylalanine) but reduced phenolic acids (dihydroxybenzoic acid hexose isomers 1 and 2, chicoric acid, chicoric acid isomer 1) and TCA intermediates (fumaric, malic, citric and succinic acids). Conclusion: The novel methodology applied in this study can be used to characterize metabolites in lettuce. Accumulation of glycosylated flavonoids, amino acids and ascorbic acid in response to glycine supply provides strong evidence supporting the idea that using amino acids as an N source alters the nutritional value of vegetable crops. © 2017 Society of Chemical Industry.

Abstract:Lettuce is a vegetable that contributes vitamins, minerals, fibre, phenolic compounds and antioxidants to the human diet. In the search for improving production conditions and crop health, the use of microorganisms with plant growth-promoting capabilities, such as soil yeasts (PGPY), in conjunction with nanotechnology could offer sustainable development of agroecosystems. This study evaluated the synthesis of health-promoting bioactive compounds in lettuce under the application of soil yeast and an iron nanoparticle (NP-Fe2O3) encapsulated in alginate beads. Two yeast strains, Candida guillermondii and Rhodotorula mucilaginosa, and a consortium of both yeasts were used in the presence and absence of Fe2O3-NPs. Phenolic compounds were identified and quantified via HPLC-ESI-Q-ToF and antioxidant activity. Ten phenolic compounds were identified, highlighting the chicoric acid isomer and two quercetin glycosides with high concentrations of up to 100 μg g−1 in treatments with C. guillermondii. Treatments with R. mucilaginosa and NPs-Fe2O3 presented an increase in antioxidant activity, mainly in TEAC, CUPRAC and DPPH activities in leaves, with significant differences between treatments. Therefore, the use of encapsulated soil yeasts is a viable alternative for application in vegetables to improve the biosynthesis and accumulation of phenolic compounds in lettuce and other crops.

Abstract:Beneficial plant–microbe interaction for enhancing crop yield and quality is a sustainable way to achieve eco-friendly, desirable agricultural productions. The main objective of this experiment was to evaluate the individual and combined effects of an arbuscular mycorrhizal fungus (AMF) strain (Funneliformis mosseae) and a seaweed extract (SWE) derived from Ascophyllum nodosum, on the growth and physiological responses of lettuce (Lactuca sativa L.). Lettuce plants were inoculated with commercial AMF inoculum (5 g kg−1 soil), and SWE foliar application was done at three levels (0.5, 1.5, and 3 g L−1). The findings revealed that AMF along with SWE generated the greatest impact. In fact, co-application of AMF inoculation and 3 g L−1 SWE considerably enhanced root colonization, chlorophyll a, chlorophyll b, total chlorophyll, carotenoids, and mineral content in the shoots and roots (N, P, K, Ca, Fe, Zn, and Mn content) of lettuce plants. This combination improved initial fluorescence (F0), photochemical efficiency of PSII (FV/Fm) and Y(NO) and total antioxidant activity (TAA), whereas the maximum fluorescence, (Fm) and Y(II), showed the highest increase in lettuce plants treated with AMF and 1.5 g L−1 SWE. Furthermore, AMF inoculation along with SWE, at concentrations 1.5 and 3 g L−1, considerably enhanced variable fluorescence (FV) and the activity of water decomposition in electron donor photosystem II (FV/F0). As a result of these findings, it can be stated that the co-application of AMF and SWE positively improves the growth and development of lettuce plants.

Placenta praevia: incidence, risk factors and outcome

Abstract:Abstract Objective: Aim of this study was to evaluate the incidence, potential risk factors and the respective outcomes of pregnancies with placenta praevia. Methods: Data were prospectively collected from women diagnosed with placenta praevia in 10 Austrian hospitals in in the province of Styria between 1993 and 2012. We analyzed the incidence, potential risk factors and the respective outcomes of pregnancies with placenta praevia. Differences between women with major placenta praevia (complete or partial placenta praevia) and minor placenta praevia (marginal placenta praevia or low-lying placenta) were evaluated. Results: 328 patients with placenta praevia were identified. The province wide incidence of placenta praevia was 0.15%. Maternal morbidity was high (ante-partum bleeding [42.3%], post-partum hemorrhage [7.1%], maternal anemia [30%], comorbid adherent placentation [4%], and hysterectomy [5.2%]) and neonatal complications were frequent (preterm birth [54.9%], low birth weight <2500 g [35.6%], Apgar-score after five minutes <7 [5.8%], and fetal mortality [1.5%]. Women with major placenta praevia had a significant higher incidence of preterm delivery, birthweight <2500 g and Apgar-score after five minutes <7. Conclusions: Placenta praevia was associated with adverse maternal (34.15%) and neonatal (60.06%) outcome. The extent of placenta praevia was not related with differences regarding risk factors and maternal outcome.
Elucidating the genetic basis of antioxidant status in lettuce (Lactuca sativa)

Abstract:A diet rich in phytonutrients from fruit and vegetables has been acknowledged to afford protection against a range of human diseases, but many of the most popular vegetables are low in phytonutrients. Wild relatives of crops may contain allelic variation for genes determining the concentrations of these beneficial phytonutrients, and therefore understanding the genetic basis of this variation is important for breeding efforts to enhance nutritional quality. In this study, lettuce recombinant inbred lines, generated from a cross between wild and cultivated lettuce (Lactuca serriola and Lactuca sativa, respectively), were analysed for antioxidant (AO) potential and important phytonutrients including carotenoids, chlorophyll and phenolic compounds. When grown in two environments, 96 quantitative trait loci (QTL) were identified for these nutritional traits: 4 for AO potential, 2 for carotenoid content, 3 for total chlorophyll content and 87 for individual phenolic compounds (two per compound on average). Most often, the L. serriola alleles conferred an increase in total AOs and metabolites. Candidate genes underlying these QTL were identified by BLASTn searches; in several cases, these had functions suggesting involvement in phytonutrient biosynthetic pathways. Analysis of a QTL on linkage group 3, which accounted for >30% of the variation in AO potential, revealed several candidate genes encoding multiple MYB transcription factors which regulate flavonoid biosynthesis and flavanone 3-hydroxylase, an enzyme involved in the biosynthesis of the flavonoids quercetin and kaempferol, which are known to have powerful AO activity. Follow-up quantitative RT-PCR of these candidates revealed that 5 out of 10 genes investigated were significantly differentially expressed between the wild and cultivated parents, providing further evidence of their potential involvement in determining the contrasting phenotypes. These results offer exciting opportunities to improve the nutritional content and health benefits of lettuce through marker-assisted breeding.
Reduced nitrogen supply enhances the cellular antioxidant potential of phenolic extracts through the alteration of phenolic composition in lettuce (Lactuca sativa L.).

Abstract:BACKGROUND Nitrogen availability is an important environmental factor that determines the production of phenolic compounds in vegetables, but the relationship between low nitrogen-induced alterations of phenolic compounds in vegetable crops and the cellular antioxidant activities of these compounds remains unclear. This study investigated the effect of reduced nitrogen supply (0.05 mmol L-1 nitrate) on phenolic metabolism in lettuce and the protective role of phenolic extracts against H2O2-induced oxidative stress in Caco-2 cells by determining cell damage, reactive oxygen species (ROS) content and antioxidant enzyme activities. RESULTS Reduced nitrogen supply significantly improved the accumulation of phenolic compounds in lettuce, which was partially correlated with the upregulation of genes related to the phenolic synthesis pathway. Phenolic extracts from lettuce cultivated in low-nitrogen medium exhibited a better protective effect against H2O2-induced oxidative damage in Caco-2 cells than those from lettuce cultivated with adequate nitrogen. These extracts act by increasing the activities of antioxidant enzymes and, subsequently, by inhibiting ROS overproduction, which leads to a decrease in mitochondrial membrane and DNA damage. The results of HPLC and correlation analyses implied that the improvement in the protective capacity of lettuce extracts after low-nitrogen treatment may be related, not only to the increased content of phenolic compounds, but also to the increased percentage contribution of chlorogenic acid and quercetin derivatives to the total phenolic content. CONCLUSION Reduction in nitrogen supply can be a powerful strategy to modify phenolic metabolism and composition in lettuce and, consequently, to improve their antioxidant capacity. Growth and Antioxidant Responses of Lettuce (Lactuca sativa L.) to Arbuscular Mycorrhiza Inoculation and Seaweed Extract Foliar Application

Abstract:Biofertilizers, such as arbuscular mycorrhiza fungi (AMF) and seaweed extract (SWE), have been effective in environmental and agricultural ecosystems. In this study, the effects of AMF, SWE, and their co-application were assayed on the growth and antioxidant potential of lettuce plants. The experiment was conducted as a factorial based on a completely randomized design with two factors and four replications under greenhouse conditions. The first factor was AMF (Glomus mosseae) at two levels consisting of AMF application (20 g pot−1), and without using AMF; and the second factor was SWE foliar spraying (Ascophyllum nodosum) at 0.5, 1.5 and 3 g L−1 concentration. The results revealed that the highest root colonization (85%) belonged to AMF and SWE (3 g L−1) × AMF; the lowest colonization rate (65%) was observed for AMF × SWE (0.5 g L−1) treatment. The highest growth parameters (leaf number, shoot and root fresh weight, head diameter), biochemical traits (total soluble proteins, carbohydrates content) and TAA, total antioxidant activity by FRAP method and ascorbic acid, total phenolics, and flavonoids content were obtained with the co-applications. Therefore, the best results of the evaluated traits were achieved with AMF × SWE (3 g L−1). The TAA value was increased three-fold compared to the control. Total phenolics and flavonoids content were 2.24 and 6.59 times higher than the control, respectively. On the other hand, leaf dry weight was decreased with the further growth of the plants. Overall, the co-application of AMF with SWE can be recommended to producers as an alternative and environment-friendly strategy to improve the qualitative and quantitative traits of the lettuce crop.
Regulated Water Deficits Improve Phytochemical Concentration in Lettuce

Abstract:In a growth chamber study, lettuce ( Lactuca sativa ) plants were used to evaluate the effects of water deficits on health-promoting phytochemicals with antioxidant properties. Lettuce plants were treated with water stress by withholding water once at 6 weeks after sowing for 2 days or multiple times at 4 weeks for 4 days, at 5 weeks for 3 days, and at 6 weeks for 2 days. Water stress increased the total phenolic concentration and antioxidant capacity in lettuce. Young seedlings, 7 days after germination, had the highest total phenolic concentration and antioxidant capacity, and also, younger plants were typically more responsive to water stress treatments in accumulating the antioxidants than older plants. Phenylalanine ammonia lyase and γ-tocopherol methyltransferase genes, involved in the biosynthesis of phenolic compounds and vitamin E, respectively, were activated in response to water stress, although no activation of L-galactose dehydrogenase was detected. Lettuce plants subjected to multiple water stress treatments accumulated significant amounts of chicoric acid compared with the control plants. Although the increase in antioxidant activity in water stress-treated plants at harvest was not as great as in young seedlings, it was significantly higher than the control. One-time water stress treatment of lettuce at the time of harvest did not result in any adverse effect on plant growth. Thus, these results show that mild water stress in lettuce applied just before harvest can enhance its crop quality with regard to its phytochemical concentration without any significant adverse effect on its growth or yield.

Spatio-Temporal Dynamics of Lettuce Metabolome: A Framework for Targeted Nutritional Quality Improvement

Abstract:Lettuce (Lactuca sativa L.) is a popular leafy vegetable valued for its dietary fiber, antioxidants, and beneficial vitamins. This study presents a comprehensive spatio-temporal analysis of the lettuce metabolome, revealing complex dynamics in metabolite accumulation influenced by plant age, leaf position, proximodistal distribution within a leaf, and head closure. Samples were collected from plants at five maturity stages (ranging from baby leaf to full commercial maturity and eventually to bolting) and from five leaf positions (from the apex to the base of each plant). A widely targeted metabolomics approach identified 1905 compounds, with flavonoids, phenolic acids, and lipids as the largest classes. Younger plants exhibited higher levels of flavonoids, while older plants accumulated more saccharides and amino acids. Metabolites showed distinct proximodistal distributions, with flavonoids and vitamins concentrated at leaf tips and terpenoids declining from base to tip. Head closure significantly reduced levels of flavonoids, retinol (vitamin A1), and riboflavin (vitamin B2), while it was associated with increased content of other beneficial vitamins, such as thiamine (B1), pantothenate (B5), and pyridoxine (B6). Broad-sense heritability (H2) estimates for metabolites yielded mean H2 values of 0.648 and 0.743 for plants at baby-leaf and commercial maturity stages, respectively. The overall highest heritability was observed in tannins (H2 = 0.909) in younger plants and chalcones (H2 = 0.894) in older plants, suggesting strong genetic control over specific metabolite classes and subclasses. These findings offer a robust framework for optimizing lettuce's nutritional profile by linking metabolite distributions to developmental processes, plant architecture, and genetic regulation. By leveraging these insights, breeders and producers can develop targeted strategies to enhance metabolite content through optimized breeding and harvesting strategies.

Abstract:The main objective of the present study was to assess the effects of sulfur (S) nutrition on plant growth, overall quality, secondary metabolites, and antibacterial and radical scavenging activities of hydroponically grown lettuce cultivars. Three lettuce cultivars, namely, Pazmanea RZ (green butterhead, V1), Hawking RZ (green multi-leaf lettuce, V2), and Barlach RZ (red multi-leaf, V3) were subjected to two S-treatments in the form of magnesium sulfate (+S) or magnesium chloride (−S). Significant differences were observed under −S treatments, especially among V1 and V2 lettuce cultivars. These responses were reflected in the yield, levels of macro- and micro-nutrients, water-soluble sugars, and free inorganic anions. In comparison with the green cultivars (V1 and V2), the red-V3 cultivar revealed a greater acclimation to S starvation, as evidenced by relative higher plant growth. In contrast, the green cultivars showed higher capabilities in production and superior quality attributes under +S condition. As for secondary metabolites, sixteen compounds (e.g., sesquiterpene lactones, caffeoyl derivatives, caffeic acid hexose, 5-caffeoylquinic acid (5-OCQA), quercetin and luteolin glucoside derivatives) were annotated in all three cultivars with the aid of HPLC-DAD-MS-based untargeted metabolomics. Sesquiterpene lactone lactucin and anthocyanin cyanidin 3-O-galactoside were only detected in V1 and V3 cultivars, respectively. Based on the analyses, the V3 cultivar was the most potent radical scavenger, while V1 and V2 cultivars exhibited antibacterial activity against Staphylococcus aureus in response to S provision. Our study emphasizes the critical role of S nutrition in plant growth, acclimation, and nutritional quality. The judicious-S application can be adopted as a promising antimicrobial prototype for medical applications.

Responses of salicylic acid encapsulation on growth, photosynthetic attributes and ROS scavenging system in Lactuca sativa exposed to polycyclic aromatic hydrocarbon pollution

Abstract:Salicylic acid (SA) is a phytohormone that plays a key role in the regulation of the defense response against environmental variables in plants, and it provides increased yield and stress tolerance when exogenously applied to plants as a growth regulator. The role of SA-mediated signals in abiotic stress tolerance varies according to the species, stressor, application method, and dose. This study investigated the effects of salicylic acid (SA, 0.1 mg ml-1) or β-cyclodextrin encapsulated salicylic acid (e-SA, 0.1 mg ml-1) treatments on growth parameters, gas exchange, photosynthesis efficiency, and antioxidant capacity in lettuce seedlings exposed to polycyclic aromatic hydrocarbon pollution. Fluorene (FLN, 100 mg L-1) contamination resulted in a 27% growth rate and a 14% water content reduction in lettuce leaves. Significant suppressions of stomatal conductance, carbon assimilation, and PSII photochemistry were detected in plants under stress. FLN + SA and FLN + e-SA treatments regulated plant-water relations by stimulating proline accumulation and relieving stomatal limitations. As indicated by the high Fv/Fm ratio, photosynthesis efficiency was recovered in FLN + SA and FLN + e-SA group plants. FLN stress caused high oxidative stress in lettuce leaves and increased lipid peroxidation level by 40%. However, especially e-SA application to plants under stress, increased SOD activity by 3-fold and CAT activity by 80% and was successful in preventing H2O2 accumulation and lipid peroxidation. Both SA and e-SA treatments partially activated the AsA-GSH cycle. As a result, direct SA application was effective in mitigating stress-induced physiological limitations with high SA accumulation in the tissues, while encapsulated SA treatment was more effective in regulating photosynthetic and biochemical reactions, alleviating oxidative damage by activating the antioxidant defense, and promoting growth under stress with moderate SA accumulation.

Abstract:The effect of methyl jasmonate (MeJA) foliar spray on the activity of antioxidant enzymes-Superoxide dismutase (SOD), Catalase (CAT), Ascorbate peroxidase (APX), and Guaiacol peroxidase (GPX)-along with assessments of total phenolic and flavonoid contents and antioxidant activity (IC50), was examined in Prickly lettuce (Lactuca serriola L.). The study involved treating plants with three MeJA solutions (0, 200, and 400 µM) and harvesting samples at four distinct time intervals. Varied MeJA concentrations and time intervals resulted in a substantial increase in the activity of all the antioxidant enzymes investigated in this study. Both concentration levels and time courses exhibited progressive outcomes. Moreover, MeJA treatment led to elevated levels of total phenolic and flavonoid contents, reaching peaks of 17.02 (mg GAL/g DW) and 8.3 (mg QUE/g DW), respectively, particularly in response to the 400 µM concentration. However, the total flavonoid content did not show any significant variation between the two concentrations. Based on the half-maximal inhibitory concentration (IC50) values, the antioxidant activity in MeJA-treated plants was found to be lower compared to the controls. However, our findings suggest that, under specific conditions discussed in this study, MeJA has the potential to enhance the nutritional value of L. serriola.

Abstract:Increasing demands by consumers for fresh, nutritional, and convenient food has led to the increase of fresh-cut produce market. Nowadays, there is a turn towards the investigation of natural products (i.e., essential oils, organic acids, and edible coatings) in an effort to lower the usage of chemical synthetic compounds (i.e., chlorine) as postharvest sanitizers. The aim of the present study was to assess the effectiveness of Origanum majorana essential oil (EO), ascorbic acid (AA), chitosan, and their combinations on quality attributes of fresh-cut lettuce stored for six days at 7 °C. When applied, Chitosan+AA resulted to a less acceptable product (visual quality and aroma), while the application of marjoram EO was able to preserve the visual quality of fresh-cut lettuce and at the same time resulted in a pleasant aroma. The application of EO+AA and Chitosan+AA increased total phenolics and antioxidant levels of fresh-cut lettuce on the fourth and sixth day of storage. The EO and EO+AA increased damage index (hydrogen peroxide and lipid peroxidation) of fresh-cut lettuce, while at the same time these treatments decreased the activity of enzymes related with plant tissue browning (i.e., peroxidase activity and polyphenol oxidase). Chitosan decreased total valuable counts and yeasts and molds counts on the sixth day of storage, while EO, AA, EO+Chitosan, and Chitosan+AA decreased yeasts and molds after four days of application. The findings of the present work indicating that the combination of marjoram EO, AA, and chitosan could be considered further as alternative means for fresh-cut produce preservation.

Abstract:The use of biostimulants in modern agriculture has rapidly expanded in recent years, owing to their beneficial effects on crop yield and product quality, which have come under the scope of intensive research. Accordingly, in the present study we appraised the efficacy of two plant-derived biostimulants, the legume-derived protein hydrolysates Trainer® (PH), and the tropical plant extract Auxym® (TPE) on two lettuce cultivars (green and red salanova®) in terms of morpho-physiological and biochemical traits (primary and secondary metabolites). The two cultivars differed in their acquisition capacity for nitrate and other beneficial ions, their photosynthetic and transpiration rates, and their ability to synthetize and accumulate organic acids and protective metabolites. The biostimulant effect was significant for almost all the parameters examined but it was subjected to significant cultivar × biostimulant interactions, denoting a cultivar-dependent response to biostimulant type. Notwithstanding this interaction, biostimulant application could potentially improve the yield and quality of lettuce by stimulating plant physiological processes, as indicated by the SPAD index (leaf chlorophyll index), ACO2 (assimilation rate), E (transpiration), and WUEi (intrinsic water use efficiency), and by increasing concurrently the plant mineral content (total N, K, Ca, Mg) and the biosynthesis of organic acids (malate, citrate), phenols (caffeic acid, coumaroyl quinic acid isomer 1, dicaffeoylquinic acid isomer 1), and flavonoids (quercetin-3-O-glucuronide, quercetin-3-O-glucoside). Biostimulant action may facilitate the bio-enhancement of certain lettuce cultivars that are otherwise limited by their genetic potential, for the accumulation of specific compounds beneficial to human health.
