Abstract:The Hami melon (Cucumis melo var. reticulatus Naud.) is a tropical fruit with a short storage period. To prolong freshness, the fruit is stored at low temperatures (3 degrees C), although this can lead to chilling injury (CI) involving skin lesions, numerous tiny brown spots on the rind of the fruit, susceptibility to decay, especially upon re warming, the formation of small pits, and browning of the surface associated with a loss of sensory quality. Our preliminary research revealed that pretreatment with 15 mmol L-1 oxalic acid (OA) for 10 min at 25 degrees C effectively reduced CI in Hami melons stored at 3 degrees C. The current work was conducted to confirm the above results and determine whether enzymes involved in antioxidative pathways play a role in the decreased CI. The activities of enzymes related to antioxidative pathways and expression levels of the corresponding genes were measured. OA pretreatment prior to storage at 3 degrees C for 42 d increased the activities of glutathione reductase (GR), ascorbate peroxidase (APX), and peroxidase (POD), and led to higher expression levels of the Cm-GR, Cm-APX, and Cm-POD genes relative to the control group. The OA-treated group also maintained higher contents of ascorbic acid (MA) and glutathione(GSH) compared with the control group. OA treatment inhibited H2O2 and O-2(-) accumulation, delayed membrane lipid oxidation, and led to a lower CI index compared with the control fruit. These results indicate that enzymes involved in antioxidative pathways play a role in the alleviation of CI upon OA treatment.

Abstract:Harvested cucumber (Cucumis sativus L. cv. Jinyou-1) fruit were placed at 37°C (heat treatment) or at 20°C (control) for 24h prior to storage at 2°C. In order to determine the responses of phospholipase D (PLD) and lipoxygenase (LOX) to chilling stress, their activities and the expression pattern of the PLD gene in cucumber fruit were measured during storage. Chilling injury assessed as electrolyte leakage and malondialdehyde (MDA) content was alleviated by heat treatment. Heated fruit showed reduced activities of LOX and PLD with suppressed expression of PLD mRNA compared with control fruit over the storage. The data indicate that PLD and LOX may be the major lipid-degradative enzymes involved in the induction of chilling injury in cucumber fruit. Our results suggest that PLD and LOX might be associated with the initiation of chilling injury by involvement in membrane deterioration.
Research of the Effects of Oxalic Acid-treatment on the Chilling Injury and Quality of Harvested Hami Melon

Abstract:Hami melon"Xizhoumi 25" was used in this experiment. Melons were dipped in different concentra-tions of oxalic acid (OA ) with 10, 15 mmol/ L and 20 mmol/ L (add 0.5 mL/L tuwen-20)and melons were dipped in normal temperature distilled water was control at 25 ℃ for 10 min, after the surface were dried com-pletely and stored at 3 ℃-5 ℃ for 42 days, chilling injury index and quality in fruits were measured during the storage period. Compared with that in control, OA treatment inhibited the decrease of firmness and ascorbic acid (ASA)contents and increased the contents of total soluble solids from the initial storage to 21 day (P < 0.05), in this respect, 15 mmol/L OA was the most effective treatment. In addition treatment with 15 mmol/L OA inhib-ited the increase of respiration rate and electrolyte leakage, reducing the chilling injury index of Hami melon. But 20 mmol/L OA promoted the chilling injury index and electrolyte leakage, meanwhile which restrained the decline of chlorophyll content compared to the controls. The results indicated 15 mmol/L OA was the most effec-tive treatment to reduce the CI(chilling injury index)and extend the storage quality.

Abstract:Watermelon (Citrullus lanatus Thunb. Matsum and Nakai) fruits were held in 50μll−1 ethylene at 20°C following an 18-h exposure to 5μll−1 1-methylcyclopropene (1-MCP) to investigate the involvement of ethylene perception in the regulation of placental tissue water-soaking through phospholipase-mediated phospholipid (PL) degradation. Water-soaking was accompanied by increase in phospholipases C (PLC, EC 3.1.4.3) and D (PLD, EC 3.1.4.4), and lipoxygenase (LOX, EC 1.13.11.12) activities, decreases in phosphatidylcholine (PC) and phosphatidylinositol (PI), and increases in phosphatidic acid (PA). These changes were evident within 2 days of ethylene exposure, coincident with accelerated softening, increased electrolyte leakage and extractable juice, and appearance of water-soaking. Air-stored fruits did not exhibit water-soaking, and the activities of lipid-degrading enzymes and PL levels remained constant throughout the 8-day storage period. Treatment of fruit with 5μll−1 1-MCP prior to ethylene exposure reduced ethylene-induced increases in the activities of lipid-degrading enzymes, and PL degradation, and completely prevented water-soaking and the attendant increases in electrolyte leakage and extractable juice. The results demonstrated that water-soaking in watermelon fruit requires competent ethylene responsiveness and is associated with ethylene-inducible PL degradation. This represents the first report wherein 1-MCP imparts to a fully ripe fruit complete protection against deleterious effects of exogenous ethylene. Watermelon fruit maintained under simulated commercial storage conditions (up to 3 weeks at 13°C ) without exposure to exogenously provided ethylene also benefited significantly from prior exposure to 1-MCP.

Abstract:Having been held in 10 μL/L 1-methylcyclopropene (1-MCP) or air for 18 h, seedless watermelon (Citrullus lanatus Thunb. Mansfeld, cv. Millionaire) fruit was cut to obtain pericarp cylinders (7 mm in diameter, 40 mm thick), which were rinsed with 2% CaCl2 or deionized water and then stored at 10 °C. Tissue firmness, electrolyte leakage and activities of phospholipase C (PLC), phospholipase D (PLD) and lipoxygenase (LOX) were determined. Results suggested that 2% CaCl2 stimulated activities of phospholipase C (PLC), phospholipase C (PLD) and lipoxygenase (LOX), but maintained tissue firmness throughout storage. CaCl2 alone may not be sufficient to maintain quality of fresh-cut watermelon and even would exert negative effects for stimulating lipolytic enzymes. 1-MCP counteracted CaCl2 in regulation of PLC, PLD and LOX. Combination of 1-MCP and CaCl2 retarded the ripening process, as illustrated by higher firmness and lower activities of lipolytic enzymes in relative to the control.
Changes of Chilling Tolerance in Different Sections of Cucumber Fruit.

Abstract:To determine the chilling tolerance of cucumber fruit after harvest,cucumber(Cucumis sativus L.'Jinyou 1')fruit were stored at 2 ℃ for 9 days before ripening at 20 ℃ for 2 days.Chilling injury index,electrolyte leakage,activities of phospholipase D(PLD)and lipoxygenase(LOX),membrane-associated Ca2+ content in the calyx,middle and stalk sections of cucumber fruit were determined,respectively.Chilling injury occurred initially in the calyx area,and developed toward the middle and stalk end.After storage,chilling injury index,electrolyte leakage,activities of PLD and LOX in calyx section were highest,and lowest in stalk section of the fruit.Content of membrane-associated Ca2+ increased from calyx to stalk.Date indicated that increasing tolerance to chilling temperature from calyx to stalk of cucumber fruit was significantly related with the stimulation of PLD and LOX activities and decrease of membrane-associated Ca2+ content.
Responses of Phospholipase D and Lipoxygenase to Mechanical Wounding in Postharvest Cucumber Fruits.

Abstract:This study was to investigate the responses of phospholipase D (PLD) and lipoxygenase (LOX) to mechanical wounding in postharvest cucumber (Cucumis sativus L. cv. Biyu-2) fruits. Membrane-associated Ca2+ content, activities and gene expression of PLD and LOX, and contents of phosphatidylcholine (PC), phosphatidylinositol (PI), and phosphatidic acid (PA) were determined in cucumber fruits following mechanical wounding. Results show that PLD and LOX activities increased with the PLD and LOX mRNAs which are upregulated upon wounding, while membrane-associated Ca2+ content decreased. Accompanying with the increase of PLD and LOX activities, accumulation of PA and losses of PC and PI were observed in all fruits, but there were differences of degrees between wounded and control fruits. Results suggest that PLD and LOX might be the main hydrolytic enzymes of phospholipids in postharvest cucumber fruits participating in the mechanical wounding injury. The activation of PLD and LOX might be the result of gene expression, which could be stimulated by the Ca2+ flowing from the membrane to the cytoplasm upon receiving the wounding signals.
Relationship Between Heat Shock Alleviating Chilling Injury and Lipoxygenase of Persimmon Fruit

Abstract:In order to study the mechanism of chilling injury and to determine the efficacy of hot shock in delaying fruit chilling injury, the persimmon fruit were treated in hot air at 48℃ for 3 hours after harvest, then stored at 1℃. Chilling injury index, lipoxygenase activity and fatty acid relative content of persimmon fruit were investigated. Results showed that heat shock retarded the activity of lipoxygenase. The chilling injury index and the relative content of palmitic acid and stearic acid of persimmon fruit by heat shock was significantly lower than that of control. While the relative content of linoleic acid and linolenic acid in heat shock persimmon fruit were significantly higher than that of control fruit. But there was no significantly difference in oleic acid relative content. These results indicated that heat shock in persimmon could delay chilling injury through inhibition of lipoxygenase activity and elevation of linoleic acid and linolenic acid relative content.

Abstract:Chilling injury (CI) is a physiological disorder induced by cold, which heavily limit crop production and postharvest preservation worldwide. Methyl jasmonate (MeJA) can alleviate CI in various fruit species, including peach; however, the underlying molecular mechanism is poorly understood. Here, changes in contents of phenolics, lipids, and jasmonic acid (JA) and gene expressions are compared between MeJA and control fruit. Exogenous MeJA inhibited expressions of PpPAL1, PpPPO1, and PpPOD1/2 but did not affect the phenolic content. Furthermore, MeJA fruit showed lower relative electrolyte leakage, indicating less membrane damage. Meanwhile, the enrichment of linoleic acid in the potential lipid biomarkers, especially phosphatidylcholine, phosphatidylethanolamine, and phosphatidylglycerol, coincided with lower expressions of PpFAD8.1 but higher PpLOX3.1 and JA content. In the JA signaling pathway, MeJA significantly upregulated expressions of PpMYC2.2 and PpCBF3 but downregulated PpMYC2.1. In conclusion, adjustments of fatty acids in phospholipids contribute to MeJA-induced alleviation of CI in peach fruit via induction of the JA-mediated C-repeat-binding factor pathway.

Abstract:Persimmon fruit quickly softens after harvest, which limits its industrial value. The hormone melatonin has been reported to delay fruit ripening and softening. However, the effect of melatonin on postharvest persimmon fruit remains inconclusive. In this study, we found 10 μM melatonin treatment delayed the softening of persimmon fruit, accompanying with lower levels of endogenous ethylene and abscisic acid (ABA). Further, transcriptomic and metabolomic analysis were applied to investigate the regulatory mechanism. In total, 6831 differentially expressed genes (DEGs) and 1224 differentially accumulated metabolites (DAMs) were identified. We also investigated the metabolism pathways and regulatory networks incorporating crucial structural genes and TF during persimmon fruit softening. They were involving functions of saccharides, organic acids, ethylene biosynthesis, and ABA biosynthesis. In conclusion, our results indicate that melatonin can delay the biosynthesis of ethylene and ABA during persimmon softening and provide new insights into the regulatory pathways of melatonin-maintained persimmon fruit quality.
Identification of Down-regulated Genes Modulated by an Alternative Oxidase Pathway under Cold Stress Conditions in Watermelon Plants

Abstract:This study investigated the cold responses of watermelon plants modulated by the alternative oxidase (AOX) pathway. Salicylhydroxamic acid was used in watermelon (Citrullus lanatus) plants in order to chemically inhibit the AOX pathway. After exposure to cold temperature (10°C) for 24 h, the SHAM-treated plants showed more serious cold-induced damage compared to non-treated plants. To investigate how the AOX pathway modulated the watermelon plants’ responses to cold, a polymerase chain reaction (PCR)-based suppression subtractive hybridization technique was used to isolate genes differently expressed between cold-stressed leaves of SHAM-treated and non-treated watermelon germplasm “IVSM No. 9”. After sequencing 400 randomly chosen cDNA clones, 314 uniquely expressed sequence tags (ESTs) were obtained. A total of 114 ESTs encoded unknown proteins or had no similarity in the database. Identified genes with significant protein homology mainly participated in stress and defense responses, metabolism and energetic processes, regulation of gene expression, signal transduction, protein fate and synthesis, cellular transport and others. These results indicated that watermelon plants could not respond appropriately to cold stress and suffer greater damage with an inhibited AOX pathway, thus, the AOX pathway had an important role in defining the plants cold responses.
Comparative Dynamics of Ethylene Production and Expression of the ACS and ACO Genes in Normal-Ripening and Non-Ripening Watermelon Fruits

Abstract:Watermelon fruit is traditionally classified as non-climacteric, but it has been reported to be hypersensitive to exogenous ethylene: resulting in accelerated softening and water-soaking. The previous studies have reported a modest, but significant increase in ethylene production in strawberry, grape, etc., which has stimulated interest in exploring the possible role of this hormone in ripening of non-climacteric fruits. In this study, both ethylene emission and content of internal ethylene (CIE) in watermelon fruit were determined, with tomato fruit used as the reference. Throughout the development and ripening, the average CIE levels in flesh was higher than the nearly undetectable ethylene emission from fruits, and revealed a distinct rise of ethylene production in flesh in normal-ripening line 97103, but not in non-ripening line PI296341-FR. In parallel with the patterns of CIE, the activity of 1-aminocyclopropane-1-carboxylate synthase (ACS) and 1-aminocyclopropane-1-carboxylate oxidase (ACO) both increased in 97103 flesh as ripening progressed, but stayed at a constant and low level in PI296341-FR flesh. Eight ACS and eight ACO isoforms were identified in watermelon genome by full-length alignment and domain analysis of protein sequences, and among which, four ACS and four ACO genes were selected based on transcriptome profiling of the fruit development for further assay using qPCR. The expression of two ACS isoforms, Cla022653 and Cla011522, as well as two ACO isoforms, Cla014827 and Cla016287, were significantly up-regulated in 97103 flesh during ripening compared with PI296341-FR, indicating that these four genes could potentially play a more important role in ethylene biosynthesis in normal-ripening watermelon flesh.
Chitosan treatment reduces softening and chilling injury in cold-stored Hami melon by regulating starch and sucrose metabolism

Abstract:Cold-stored Hami melon is susceptible to chilling injury, resulting in quality deterioration and reduced sales. Pre-storage treatment with chitosan reduces fruit softening and chilling injury in melon; however, the underlying mechanism remains unclear. In this study, Gold Queen Hami melons were treated with 1.5% chitosan solution for 10 min before cold storage at 3°C and then the effect of chitosan was examined on fruit firmness, weight loss, chilling injury, soluble solid content (SSC), pectin, and soluble sugar contents of melon fruit. Also, the enzyme activities and gene expressions related to fruit softening and starch and sucrose metabolism were investigated. Chitosan treatment reduced the fruit softening and chilling injury, maintained the high levels of starch and sucrose contents, and regulated the enzyme activities and gene expressions related to starch and sucrose metabolism. Fruit firmness was significantly positively correlated with sucrose and starch contents. Altogether, we uncovered the potential mechanism of chitosan coating mitigating melon softening and chilling injury through the regulation of starch and sucrose metabolism.

Abstract:To investigate the physiological and molecular properties relating to cell wall carbohydrate metabolism in fruit, the ultrastructure and polysaccharides compositions of the cell wall, as well as the fruit quality and activities of enzymes relating to fruit softening, were studied for three Hami melon varieties (‘Xizhoumi 17’, ‘Jinhuami 25’, and ‘Chougua’) representing three different storability levels. The results showed that ‘Chougua’ maintained a higher firmness on day 18, with the lowest decay incidence (0%). ‘Chougua’ showed a better storage quality and intact cell wall structure. The molecular weight and monosaccharide composition of cell wall polysaccharides for Hami melons underwent great changes during storage, and the degradation of pectin polysaccharides was obvious, involving the depolymerization of macromolecular polymers accompanied by the production of new macromolecular polymers and composition changes in pectin monosaccharides (glucose, galactose, and arabinose) during the softening process of the Hami melons. Polygalacturonase, pectin methylesterase, xyloglucan endo-transglycosylase/hydrolase, α-arabinofuranosidase, β-galactosidase, and cellulase were associated with fruit softening at different stages of storage. There were similar softening mechanisms in the three Hami melons. This study will provide reference for further study on the fruit softening mechanisms of Hami melons.

Abstract:Abstract Background Hami melon ( Cucumis melo var. saccharinus ) is a popular fruit in China because of its excellent taste, which is largely determined by its physicochemical characteristics, including flesh texture, sugar content, aroma, and nutrient composition. However, the mechanisms by which these characteristics are regulated have not yet been determined. In this study, we monitored changes in the fruits of two germplasms that differed in physicochemical characteristics throughout the fruit development period. Results Ripe fruit of the bred variety ‘Guimi’ had significantly higher soluble sugar contents than the fruit of the common variety ‘Yaolong.’ Additionally, differences in fruit shape and color between these two germplasms were observed during development. Comparative transcriptome analysis, conducted to identify regulators and pathways underlying the observed differences at corresponding stages of development, revealed a higher number of differentially expressed genes (DEGs) in Guimi than in Yaolong. Moreover, most DEGs detected during early fruit development in Guimi were associated with cell wall biogenesis. Temporal analysis of the identified DEGs revealed similar trends in the enrichment of downregulated genes in both germplasms, although there were differences in the enrichment trends of upregulated genes. Further analyses revealed trends in differential changes in multiple genes involved in cell wall biogenesis and sugar metabolism during fruit ripening. Conclusions We identified several genes associated with the ripening of Hami melons, which will provide novel insights into the molecular mechanisms underlying the development of fruit characteristics in these melons.

Abstract:Cuticle wax chemicals are cultivar-dependent and contribute to storage quality. Few research reported on wax analysis between melting flesh-type (MF; ‘Jinhuami 25’) and nonmelting flesh-type (NMF; ‘Xizhoumi 17’ and ‘Chougua’) Hami melons. Chemicals and crystal structures of Hami melon cuticular wax, cell wall metabolism related to fruit melting, and fruit physiology were analyzed to observe wax functions. Results showed that Hami melon cuticle wax predominantly consists of esters, alkanes, alcohols, aldehydes, and terpenoids. MF-type has a lower alkane/terpenoid ratio, concomitant to its higher weight loss and cuticle permeability. Micromorphology of wax crystals appears as numerous platelets with irregular crystals, and the transformation of wax structure in NMF Hami melon is delayed. Waxy components affect cell wall metabolism and physiological quality, which results in the pulp texture difference between MF-type and NMF-type during storage. Results provide a reference for the regulation of wax synthesis in both types of melons.

CETH effectively compromised internal browning incidence, accompanied by the inhibited activity of peroxidase but enhanced phenolic content, and less membrane leakage with reduced H2O2 and malondialdehyde contents. Intriguingly, CETH elevated levels of phospholipids and unsaturation of their acyl chains, coincident with the lower transcript levels of phospholipase D alpha 1 but higher fatty acid desaturase2/8.1, and the enhanced sphingolipid contents and biosynthesis concomitant with higher transcript levels of glucosylceramide synthase but lower inositol phosphorylceramide synthase3. Therefore, CETH can ameliorate fruit CI through adjusting phenolic and lipid metabolism, especially comprehensive remodeling of phospholipids and sphingolipids to contribute to the membrane stability.
Elucidating the Molecular Responses to Waterlogging Stress in Cucumis melo by Comparative Transcriptome Profiling

Abstract:Waterlogging is a serious abiotic stressor that drastically hinders the growth and productivity of melon (Cucumis melo) around the world, due to the reduction in available oxygen levels in the waterlogged tissues. However, the mechanism underlying the responses to waterlogging stress in melon is largely unknown. In this study, physiological and transcriptome data of the waterlogging-sensitive accession ‘L39’ and the waterlogging-tolerant accession ‘L45’ were investigated under conditions of normal water supply and waterlogging stress. The results showed that ‘L45’ exhibited higher chlorophyll contents and lower REL (relative electrolyte leakage) and MDA (malondialdehyde) contents compared with ‘L39’ under waterlogging stress. Additionally, waterlogging stress only led to the stomatal closure and chloroplast damage of ‘L39’. In total, 1748 genes were differentially expressed in the leaves of waterlogging-stressed ‘L45’ compared with control, whereas 3178 genes were differentially expressed in ‘L39’. Further analysis indicated that genes related to chlorophyll synthesis and photosynthesis were more depressed in ‘L39’, while sugar cleavage, glycolysis, and fermentation genes were highly induced in ‘L39’ compared with ‘L45’. The expression of genes involved in ROS (reactive oxygen species) scavenging and hormone signaling significantly differed between ‘L39’ and ‘L45’ in their response to waterlogging stress. Moreover, a total of 311 differentially expressed transcription factors were waterlogging-responsive, among which members of the ERF (ethylene response factor), bHLH (basic helix-loop-helix), and WRKY families might play crucial roles in waterlogging tolerance in melon. This study unraveled the molecular responses to waterlogging stress in melon and could provide helpful candidate genes for further molecular breeding of waterlogging-tolerant melon varieties.

Abstract:Abstract Fruit ripening is influenced by multiple plant hormones and the regulation of genes. However, studies on posttranscriptional regulators (e.g., miRNAs) of fruit growth and ripening are limited. We used miRNA sequencing and degradome methods to identify miRNAs and their target genes in melon ( Cucumis melo cv. Hetao melon). A total of 61 conserved miRNAs and 36 novel miRNAs were identified from fruit growth, ripening, climacteric, and postclimacteric developmental stage samples, of which 32 conserved miRNAs were differentially expressed between developmental stage samples. Sixty-two target genes of 43 conserved miRNAs and 1 novel miRNA were identified from degradome sequencing. To further investigate miRNA influencing fruit ripening, transgenic melon plants overexpressing pre-cme-miR393 (cme-miR393 - OE) were generated and characterized. The results showed that fruit ripening was delayed in cme-miR393-OE transgenic lines compared to nontransgenic fruits. The target of cme-miR393 was also identified, and the expression of CmAFB2 was repressed in transgenic plants. These results provide evidence that miRNA regulates melon fruit ripening and provide potential targets to improve the horticultural traits of melon fruit.

Abstract:Low temperature storage prolongs the postharvest life of various fresh fruit and vegetables. However, tropical fruit may suffer from cold stress during storage. Here, Jiashi melons was stored at 3 °C, and 0.5 °C for a period of 36 days, with storage at 21 °C serving as a control. Rotting was delayed at the lower temperatures compared to the control. Storage at 21 °C resulted in more severe weight loss and a high rotting rate. By day 36, the weight loss rate as a result of moisture reduction reached 11% in the control, but only 8% at the lower temperatures. Chilling injury was observed at both low temperatures over time, but its onset was later for storage at 3 °C. The fruit stored at 3 °C also scored better for rotting rate, weight loss rate, and a chilling injury index than fruit stored at 0.5 °C. Storage at 3 °C was associated with higher activity of peroxidase (POD), catalase (CAT), fatty acid desaturase (FAD), and adenosine triphosphatase (ATPase). The transcriptomes of the fruit were sequenced for the time points 0 d, 12 d and 24 d at all three storage temperatures. This identified a total of 415 co-expressed differential genes that are mainly involved in regulation of transcription, signal transduction, response to stress, lipid metabolism and other processes. The findings were corroborated by targeted RT-qPCR analysis. The expression levels of differentially expressed genes (DEGs) indicated that storage at 3 °C induced overexpression of the early cold-responsive genes ICE1 and calcium-dependent protein kinase (CDPK). Extended storage at 3 °C reduced the damage of low-temperature induced oxidative stress and altered lipid metabolism that otherwise would negatively affect cell membrane functions, so that cell homeostasis was maintained for longer. This was enabled by increased expression of MYB, auxin response factor, heat shock protein (HSPs), FAD synthetase and ATPase. This study provides insights in the molecular responses of Jiashi melon towards cold stress, and provides a theoretical basis for the development of new storage and preservation technologies.
