Abstract:The botanical origin of honey is subjected to severe controls by Food Control Institutions, both for health protection and for frauds prevention. The complexity of honey makes it very difficult to verify the botanical origin. Among the available validated methods, sensory analysis and melissopalynology are the most widely employed. These methods require a long time and deep consolidated expertise. To shorten analysis time while simplifying the analytical procedure, head-space flash gas chromatography was applied in the present study. Chromatographic peak areas were processed by chemometrics (in particular principal components analysis and linear discriminant analysis). Three hundred and thirty-nine honey samples from twelve categories of unifloral honey were analyzed: acacia, citrus, chestnut, thistle, tree heath, eucalyptus, sunflower, rhododendron, lime, French honeysuckle, fir honeydew, and wood honeydew. Each sample was a priori classified by sensory analysis. The multivariate models were validated by cross validation and test-set validation, with predictive abilities always higher than 80%: good results were obtained both in calibration and in prediction mode, showing a good agreement between this new approach and the traditional one for the determination of the botanical origin of honey.

Abstract:The mandate assigned by the European Union to the Community Reference Laboratory for residues at the Istituto Superiore di Sanità covers, among other things, the assessment of trace elements in living animals as well as their products. To better protect the health of the consumer, this task aims in particular at harmonizing the procedures in use at the National Reference Laboratories (NRLs) for residues of the Member States for such determinations as well as at developing new approaches wherever the need arises. In this context an investigation was undertaken to appraise the average levels of a number of key elements in several types of honey with special regard to the influence of the various processing steps. Instrumental methods of election for the analysis of this matrix turned out to be inductively coupled plasma atomic emission spectrometry (ICP-AES) and inductively coupled plasma mass spectrometry (ICP-MS) depending on the actual concentrations of the elements of interest. Dissolution of the samples for suitable presentation to the analytical systems could be easily achieved by gentle heating at ca. 50 degrees C, sonication and addition of high-purity water. The ranges ascertained are as follows (in ng g(-1)): As, <0.50-0.70; Cd, <0.50-0.74; Cr, 1.03-3.93; Cu, 144-216; Fe, 191-651; Mn, 223-580; Ni, 17-49; Pb, 3.20-186; Pt, <0.50; Sn, <4-27; V, 1.22-1.94; and Zn, 565-1144. As a rule, concentrations of elements in honey from different beehives were similar. A few exceptions were noted for As, Cu, Fe, Ni and Zn. On the other hand, although data obtained with different analytical approaches for the same types of honey and beehive were generally in good agreement, yet some inconsistencies occurred, as in the case of Cu in freshly collected, extracted and ripened honey, Ni in extracted honey and Fe in ripened honey. These could not be traced back to specific procedural facts; rather, they should be ascribed to the inherent heterogeneity of the raw material. The experience gained with this exploratory study will be exploited to set up wider surveys and to plan the preparation of a new certified reference material in a matrix of honey to the benefit of NRLs for residues.

Abstract:High-performance anion exchange chromatography with pulsed amperometric detection (HPAEC–PAD) combined with chemometric analysis was developed to describe, for the first time, the sugar profile of sixty-one honeys of different botanical origin produced in southern Italy (Calabria Region). The principal component and linear discriminant analysis used to describe the variability of sugar data were able to discriminate the honeys according to their botanical origin with a correlation index higher than 90%. For the purpose of the robustness of the conclusions of this study, the analytical advantages of the HPAEC–PAD method have been statistically demonstrated compared to the official Italian HPLC–RI method (Refractive Index detection). Finally, as the characterization of the floral and geographical origin of honey became an important issue due to high consumer demand, 13 acacia honeys originating from Europe and China were studied by using the same method. By chemometric method it was possible to discriminate the different geographical origin with an index of 100%. All results proved the possibility to identify the sugar profile obtained by HPAEC–PAD combined with a robust statistical analysis, as a tool of authentication.

Abstract:Objective:To investigate the feasibility of using nectar flower as a contrast to find chemical markers for honey.Methods:Acacia honey and aqueous hydrochloric acid extract of acacia flower were extracted and purified by D-101 macroporous resin and then analyzed by UPLC-MS/MS.Separation was performed on an Inertsil ODS-3 column (2.1 mm × 75 rmm,2 μm),with methano1-0.02％ formic acid solution as mobile phase in gradient elution at a flow rate of 0.4 mL· min-1.The column temperature was controlled at 40 ℃ and the injection volume was 2 μL.Electrospray ionization (ESI)source was applied in multiple reaction monitoring(MRM)mode,taking 16 kinds of flavonoids as the reference,such as catechin,epicatechin,rutin,narirutin,morin,myricetin,kaempferol,pinocembrin,etc.Results:Naringenin,quercetin,genistein,luteolin,kaempferol,apigenin and wogonin were found in both acacia honey and acacia flower,and the proportion of each component in the total content of common components was 5.75:23.21:5.00:30.37:32.47:2.94:0.25 and 2.45:23.05:0.88:5.41:66.04:2.13:0.03,respectively.There was a certain degree of correlation between content of flavonoids in acacia honey and locust flower.Compounds with high content in acacia flower were also relatively high in acacia honey,vice versa.Finally,a potential marker of genistein was found in acacia honey.Conclusion:It was feasible to find markers for honey by using nectar flower as a "contrast".

Abstract:Honey is a natural sweetener constituted by numerous macro- and micronutrients. Carbohydrates are the most representative, with glucose and fructose being the most abundant. Minor honey components like volatile organic compounds (VOCs), minerals, vitamins, amino acids are able to confer honey-specific properties and are useful to characterize and differentiate between honey varieties according to the botanical origin. The present work describes the chemical characterization of honeys of different botanical origin (multifloral, acacia, apple–dandelion, rhododendron, honeydew, and chestnut) produced and collected by beekeepers in the Trentino Alto-Adige region (Italy). Melissopalynological analysis was conducted to verify the botanical origin of samples and determine the frequency of different pollen families. The carbohydrate composition (fourteen sugars) and the profile of VOCs were evaluated permitting to investigate the relationship between pollen composition and the chemical profile of honey. Statistical analysis, particularly partial least squares discriminant analysis (PLS-DA), demonstrates the importance of classifying honey botanical origin on the basis of effective pollen composition, which directly influences honey's biochemistry, in order to correctly define properties and value of honeys.

Abstract:Honey is a highly consumed natural product produced by bees which is susceptible to fraudulent practices, some of them regarding its botanical origin. Two HPLC-UV non-targeted fingerprinting approaches were evaluated in this work to address honey characterization, classification, and authentication based on honey botanical variety. The first method used no sample treatment and a universal reversed-phase chromatographic separation. On the contrary, the second method was based on an off-line SPE preconcentration method, optimized for the isolation and extraction of polyphenolic compounds, and a reversed-phase chromatographic separation optimized for polyphenols as well. For the off-line SPE method, the use of HLB (3 mL, 60 mg) cartridges, and 6 mL of methanol as eluent, allowed to achieve acceptable recoveries for the selected polyphenols. The obtained HPLC-UV fingerprints were subjected to an exploratory principal component analysis (PCA) and a classificatory partial least squares-discriminant analysis (PLS-DA) to evaluate their viability as sample chemical descriptors for authentication purposes. Both HPLC-UV fingerprints resulted to be appropriate to discriminate between blossom honeys and honeydew honeys. However, a superior performance was accomplished with off-line SPE HPLC-UV polyphenolic fingerprints, being able to differentiate among the different blossom honey samples under the study (orange/lemon blossom, rosemary, thyme, eucalyptus, and heather). In general, this work demonstrated the feasibility of HPLC-UV fingerprints, especially those obtained after off-line SPE polyphenolic isolation and extraction, to be employed as honey chemical descriptors to address the characterization and classification of honey samples according to their botanical origin.

Abstract:Volatile profiles of unifloral honeys and meads prepared in different ways (boiled-saturated, not boiled-unsaturated) were investigated by headspace solid-phase micro extraction (HS-SPME) and dehydration homogeneous liquid-liquid extraction (DHLLE) followed by GC-FID/MS analyses. The obtained data were analyzed by principal component analysis (PCA) to evaluate the differences between the investigated products. The volatile profiles of honey as well as the boiled and the not boiled meads prepared from it showed significant discrepancies. The meads contained more aliphatic acids and esters but fewer monoterpenes and aliphatic hydrocarbons than the honey. Significant/substantial differences were found between the boiled (more aliphatic alcohols and acids) and the not boiled meads (more aliphatic hydrocarbons and esters). Some compounds related to yeast metabolism, such as tryptophol, may be considered markers of honey fermentation. This research allowed us to identify chemical markers of botanical origin, retained and detectable in the meads: 4-isopropenylcyclohexa-1,3-diene-1-carboxylic acid and 4-(1-hydroxy-2-propanyl)cyclohexa-1,3-diene-1-carboxylic acid for linden; valeric acid, γ-valerolactone, p-hydroxybenzoic acid for buckwheat; 4-hydroxybenzeneacetic acid, homovanillic acid and trans-coniferyl alcohol for honeydew; and methyl syringate for canola.
The Use of Right Angle Fluorescence Spectroscopy to Distinguish the Botanical Origin of Greek Common Honey Varieties

Abstract:The standardization of the botanical origin of honey reflects the commercial value and quality of honey. Nowadays, most consumers are looking for a unifloral honey. The aim of the present study was to develop a novel method for honey classification using chemometric models based on phenolic compounds analyzed with right angle fluorescence spectroscopy, coupled with stepwise linear discriminant analysis (LDA). The deconstructed spectrum from three-dimensional-emission excitation matrix (3D-EEM) spectra provided a correct classification score of 94.9% calibration and cross-validation at an excitation wavelength (λex) of 330 nm. Subsequently, a score of 81.4% and 79.7%, respectively, at an excitation wavelength (λex) of 360 nm was achieved. Each chemometric model confirmed its power through the external validation with a score of 82.1% for both. Differentiation could be correlated with hydroxycinnamic and hydroxybenzoic acids, which absorb in this region of the spectrum. Fluorescence spectroscopy constitutes a rapid and sensitive technique, which, when combined with the stepwise algorithm and LDA method, can be used as a reliable and predictive authentication tool for honey. This study indicates that the developed methodology is a promising technique for determination of the botanical origin of common Greek honey varieties. Our long-term ambition is to support producers and suppliers to remain in a competitive national and international market.

Abstract:A potentiometric electronic tongue (PE-tongue) and a voltammetric electronic tongue (VE-tongue) were used as rapid techniques to classify and predict the honey samples from different floral and geographical origins. The PE-tongue, which was named alpha-ASTREE, was developed by Alpha M.O.S. (Toulouse, France), and it comprises seven potentiometric chemical sensors. The VE-tongue was self-developed at Zhejiang University and comprises six metallic working sensors. Four types of honey of different floral origins (acacia, buckwheat, data, and motherwort) and four types of acacia honey of different geographical origins were classified by both multisensor systems. Multivariate statistical data analysis techniques such as principal component analysis (PCA) and discriminant function analysis (DFA) were used to classify the honey samples. Both types of electronic tongue have good potential to classify the honey samples, and the positions of the data point for the samples in the PCA score plots based on the VE-tongue were much more closely grouped. Three regression modes, principal component regression (PCR), partial least squares regression (PLSR), and least squared-support vector machines (LS-SVM), were applied for category forecasting. These regression models exhibited a clear indication of the prediction ability of the two types of electronic tongue, and a positive trend in the prediction of the floral and geographical origin of honey was found. Moreover, the performance of these regression models for predicting the four types of honey of different geographical origins by the VE-tongue is very stable. (C) 2014 Elsevier B.V. Abstract:The botanical origin of honey determines its composition and hence properties and product quality. As a highly valued food product worldwide, assurance of the authenticity of honey is required to prevent potential fraud. In this work, the characterisation of Spanish honeys from 11 different botanical origins was carried out by headspace gas chromatography coupled with mass spectrometry (HS-GC-MS). A total of 27 volatile compounds were monitored, including aldehydes, alcohols, ketones, carboxylic acids, esters and monoterpenes. Samples were grouped into five categories of botanical origins: rosemary, orange blossom, albaida, thousand flower and "others" (the remaining origins studied, due to the limitation of samples available). Method validation was performed based on linearity and limits of detection and quantification, allowing the quantification of 21 compounds in the different honeys studied. Furthermore, an orthogonal partial least squares-discriminant analysis (OPLS-DA) chemometric model allowed the classification of honey into the five established categories, achieving a 100% and 91.67% classification and validation success rate, respectively. The application of the proposed methodology was tested by analysing 16 honey samples of unknown floral origin, classifying 4 as orange blossom, 4 as thousand flower and 8 as belonging to other botanical origins.

Abstract:The objective of this study is to test the feasibility of multi-isotopic and elemental analyses combined with chemometric techniques for differentiating the botanical origins of major honey products in China. The stable isotope and elemental compositions of 57 honey samples from four major floral origins in China (i.e., rape honey, acacia honey, vitex honey, and jujube honey) were analyzed using stable isotope ratio mass spectrometry (IRMS) and inductively coupled plasma mass spectrometry (ICP-MS), respectively. The results showed that hydrogen and oxygen isotopes could be more suitable than the carbon isotope for discriminating the floral origins of major honeys in China. There were significant differences in the contents of most elements between or among different floral origins. The combination of IRMS and ICP-MS methods provides the most effective and accurate approach (in most cases close to 100% accuracy) for classifying Chinese honeys according to their floral origins.
Fast Classification of Geographical Origins of Honey Based on Laser-Induced Breakdown Spectroscopy and Multivariate Analysis

Abstract:Traceability of honey is highly required by consumers and food administration with the consideration of food safety and quality. In this study, a technique named laser-induced breakdown spectroscopy (LIBS) was used to fast trace geographical origins of acacia honey and multi-floral honey. LIBS emissions from elements of Mg, Ca, Na, and K had significant differences among different geographical origins. The clusters of honey from different geographical origins were visualized with principal component analysis. In addition, support vector machine (SVM) and linear discrimination analysis (LDA) were used to quantitively classify the origins. The results indicated that SVM performed better than LDA, and the discriminant results of multi-floral honey were better than acacia honey. The accuracy and mean average precision for multi-floral honey were 99.7% and 99.7%, respectively. This study provided a fast approach for geographical origin classification, and might be helpful for food traceability.

Application of the Dehydration Homogeneous Liquid–Liquid Extraction (DHLLE) Sample Preparation Method for Fingerprinting of Honey Volatiles

Abstract:Recently, we proposed a new sample preparation method involving reduced solvent and sample usage, based on dehydration homogeneous liquid–liquid extraction (DHLLE) for the screening of volatiles and semi-volatiles from honey. In the present research, the method was applied to a wide range of honeys (21 different representative unifloral samples) to determine its suitability for detecting characteristic honey compounds from different chemical classes. GC-FID/MS disclosed 130 compounds from different structural and chemical groups. The DHLLE method allowed the extraction and identification of a wide range of previously reported specific and nonspecific marker compounds belonging to different chemical groups (including monoterpenes, norisoprenoids, benzene derivatives, or nitrogen compounds). For example, DHLLE allowed the detection of cornflower honey chemical markers: 3-oxo-retro-α-ionols, 3,4-dihydro-3-oxoedulan, phenyllactic acid; coffee honey markers: theobromine and caffeine; linden honey markers: 4-isopropenylcyclohexa-1,3-diene-1-carboxylic acid and 4-(2-hydroxy-2-propanyl)cyclohexa-1,3-diene-1-carboxylic acid, as well as furan derivatives from buckwheat honey. The obtained results were comparable with the previously reported data on markers of various honey varieties. Considering the application of much lower volumes of very common reagents, DHLLE may provide economical and ecological advantages as an alternative sample preparation method for routine purposes.

Abstract:The elemental analysis of honey is a useful tool for regional and botanical authentication of honey, particularly when combined with palynology and carbohydrate analysis. A comprehensive database that includes trace element levels in honey with known regional and botanical sources is needed to evaluate its usefulness for honey source provenance. In this study, honey samples collected from discrete sources in Montana, North Dakota, and South Dakota were analyzed by ICP-MS and INAA. Sucrose was measured by NMR to confirm that samples were not adulterated with sugar. Palynology was used to assess the pollen source. The elements B, Na, Mg, Al, P, K, Ca, Mn, Fe, Cu, Zn, Rb, Sr, Co, Mo, Cs, Ba, and Ce, among others, were measured by ICP-MS, and the elements Na, Cl, Sc, Cr, Fe, Zn, Se, Br, and Hg were measured by INAA. The duplicate elements from the two methods were compared. A cluster analysis and a principal component analysis were conducted to determine if the regional or botanical sources could be distinguished by their trace element content. The results have demonstrated that elemental analysis may be useful in discriminating honey sourced from Montana against honey sourced from North and South Dakota.

Identification of geographical and botanical origin of Mediterranean honeys using UV-vis spectroscopy and multivariate statistical analysis

Abstract:The present study aimed to differentiate Mediterranean honey samples with regards to their botanical and geographical origin, using UV-vis spectroscopy, combined with multivariate statistical analysis and chemometrics. A total of 151 commercial honey samples were collected from different countries (Greece, Malta, Spain, Tunisia, and Turkey), which were characterized as thyme, pine, or polyfloral. Discrimination of geographical and botanical origin was achieved successfully with Principal Component Analysis (PCA) together with Random Forest (RF), Partial Least Squares – Discriminant Analysis (PLS – DA), and Data Driven – Soft Independent Modelling of Class Analogies (DD-SIMCA) using the spectral range of 220–550 nm. DD-SIMCA achieved very high accuracy, sensitivity, and specificity on the discrimination of both the geographical and botanical origin of honey samples. The results support the good predictive capability of UV-vis spectroscopy combined with chemometrics for the determination of honey origin. This methodology could be utilized for the rapid and non-destructive assessment of Mediterranean honey origin.

Abstract:A novel Fe3O4@MDN magnetic nanomaterial was successfully developed as a magnetic dispersive solid-phase extraction adsorbent for the determination of triazole pesticide residuals in honey samples by high performance liquid chromatography coupled with triple quadrupole mass spectrometry (HPLC-MS/MS). The macromolecular structure named MDN was coated on the surface of a copolymer synthesized with silyl reagents methacrylic acid-3-(trimethoxysilyl) propyl ester (MPS) and tetraethyl orthosilicate (TEOS) based on ferriferrous oxide (Fe3O4), whose characteristics were confirmed by a scanning electron microscope (SEM). The proposed magnetic dispersive solid-phase extraction (MDSPE) method provided a rapid, environmentally friendly and magnetic stuff recyclable approach for the triazole pesticide analysis. By means of optimal conditions, the recoveries of 15 triazoles (spiked at 20 mu g kg(-1)) ranged from 90.5 to 105.9%. The method showed good linearity, sensitivity in the range of mu g per kilogram and repeatability (relative standard deviation below 6.8%, n = 5). The proposed method was confirmed to be a simple, rapid and efficient method for the analysis of triazole pesticide residuals in honey samples.
