Abstract:Lately, a great deal of attention is being paid to capsule coating, since the coat protects active pharmaceutical ingredients (APIs) from damage, as is in the case of tablet and pellet. However, moisture and heat sensitivity of gelatin shells make it challenging to coat capsules using the conventional aqueous coating techniques. In an effort to overcome this challenge, the present study aims to coat capsules using two different coating techniques: electrostatic dry powder coating (EDPC) and dip coating (DC). Both capsule coatings and free films were prepared by these two coating techniques, and the effects of coating formulations and processing conditions on the film quality were investigated. The corresponding drug in vitro release and mechanisms were characterized and compared. The results of dissolution tests demonstrated that the drug release behavior of both EDPC and DC coated capsules could be optimized to a sustained release of 24 h, following the Fick's diffusion law. The results of this study suggest that EDPC method is better than DC method for coating capsules, with respect to the higher production efficiency and better stability, indicating that this dry coating technology has promised in gelatin capsule coating applications.

Abstract:BackgroundProbiotics and omega-3 rich oils are two health-beneficial bioactives added to a variety of foods to enhance their functional properties. However, these bioactives are highly sensitive to degradation during food handling and transit through the gastrointestinal tract (GIT).Scope and approachThe goal of this review was to describe the benefits and drawbacks of extrusion technology in the food industry, as well as the viability of probiotics and oxidative stability of omega-3 rich oils encapsulated by extrusion technology after exposure to in vitro and in vivo systems, storage, and in the food system. This review also highlights the co-encapsulation concept of multiple bioactives along with the synergistic effects of co-encapsulated probiotics and omega-3 rich oils.Key findings and conclusionsThe survival of probiotics encapsulated using the extrusion process was found to be satisfactory in both the actual food system and the in vivo model. However, in the case of omega-3 rich oils, there is a lack of evidence to support the stabilization effect of the encapsulation technique using in vivo and real food models. A few studies found higher viability of probiotics and oxidative stability of omega-3 rich oils in co-encapsulation of these bioactives in one matrix rather than their single encapsulation by the extrusion technique. For the commercial application of co-encapsulated probiotics and omega-3 rich oils, future research should focus on researching the interactions of co-encapsulated probiotics and omega-3 rich oils with real food systems, as well as in vivo and clinical studies to determine product functionality and efficacy.

Hydrocolloids, including proteins and carbohydrates, natural or modified, are a group of ingredients used as the wall material in extrusion. Hydrocolloids, due to their specific properties, can significantly improve the probiotic survivability of the final powder during the microencapsulation process and storage. The present article will discuss the different kinds of hydrocolloids used for microencapsulation of probiotics by extrusion and co-extrusion, along with new sources of novel gums and their potential as wall material.

Abstract:This review described and explains the encapsulation of bioactive compounds by extrusion technologies. Bioactive compounds have many health benefits, but several drawbacks such as a high organoleptic impact due to the bitterness and acrid taste of some compounds, and limited shelf life reduce the utilization of bioactive compounds in food. Encapsulation of bioactive compounds to prevent their several drawbacks and to increase their utilization in food has been achieved by 'extrusion' technology. The extrusion technologies discussed in the review are hot-melt extrusion, melt injection extrusion process, co-extrusion, and electrostatic extrusion. Extrusion technology as a mode of encapsulation of bioactive compounds as increased the number of bioactive compounds that can be encapsulated. Also, extrusion technology helps to reduce the particle size of encapsulated bioactive compounds which increase their application in the food industry. The reduction in the particle size of the extrudate helps to increase the shelf life of encapsulated bioactive compounds and aid-controlled release in the targeted site in the body.

Abstract:Encapsulation of bioactive compound has some encountered issues associated particularly to their mode of fabrication, design, and biodegradability. For these reasons, selecting appropriate coating materials like hydroxyl cellulose has several advantages such as modification of moisture, crystal growth, and viscosity. In addition, starches, maltodextrins, whey proteins, chitosan, zein/pectin, and alginates are considered an important coating materials used during microencapsulation processing steps. Thus, the variability in the hydrophilicity, lipophilicity and chargeability levels of those coating materials along with the encapsulation is considered among the molecular aspects in forming genius emulsion vesicles, microgels, solid lipid nanoparticles, liposomes, or core shell nanoparticles. These encapsulation designs are capable in promoting variable physicochemical and therapeutic features such as intra-atomic dimensions, dispersed volumes, release kinetics, and stability levels. Under these desired structural and molecular parameters, in addition to proper optimization of processing conditions, perfect microcarriers and microcapsules could be achieved. Moreover, appropriate control of copolymerization and crystallization of the coating materials with some antagonistic pharmaceutical drugs needs special emphasize. In general, this manuscript reviews various important aspects of structured materials used along with their production and potential uses for enhancing targeted delivery and release of therapeutic foods.

Abstract:An increased demand for natural products nowadays most specifically probiotics (PROs) is evident since it comes in conjunction with beneficial health effects for consumers. In this regard, it is well known that encapsulation could positively affect the PROs' viability throughout food manufacturing and long-term storage. This paper aims to analyze and review various double/multilayer strategies for encapsulation of PROs. Double-layer encapsulation of PROs by electrohydrodynamic atomization or electrospraying technology has been reported along with layer-by-layer assembly and water-in-oil-in-water (W1/O/W2) double emulsions to produce multilayer PROs-loaded carriers. Finally, their applications in food products are presented. The resistance and viability of loaded PROs to mechanical damage, during gastrointestinal transit and shelf life of these trapping systems, are also described. The PROs encapsulation in double- and multiple-layer coatings combined with other technologies can be examined to increase the opportunities for new functional products with amended functionalities opening a novel horizon in food technology.

Influence of internal composition on physicochemical properties of alginate aqueous-core capsules

Abstract:Hypothesis: To enhance physicochemical properties of alginate aqueous-core capsules, conventional strategies were focused in literature on designing composite and coated capsules. In the present study, own effect of liquid-core composition on mechanical and release properties was investigated. Experiments: Capsules were prepared by dripping a CaCl2 solution into an alginate gelling solution. Viscosity of CaCl2 solution was adjusted by adding cationic, anionic and non-ionic naturally derived polymers, respectively chitosan, xanthan gum and guar gum. In parallel, uniform alginate hydrogels were prepared by different methods (pouring, in situ forming and mixing). Mechanical stability of capsules and plane hydrogels were respectively evaluated by compression experiments and small amplitude oscillatory shear rheology and then correlated. Capsules permeability was evaluated by monitoring diffusion of encapsulated cochineal dye, riboflavin and BSA. The core-shell interactions were investigated by ATR-FTIR. Findings: Results showed that inner polymer had an impact on membrane stability and could act as an internal coating or provide mechanical reinforcement. Mechanical properties of alginate capsules were in a good agreement with rheological behavior of plane hydrogels. Release behavior of the entrapped molecules changed considerably. This study demonstrated the importance of aqueous-core composition, and gave new insights for possible adjusting of microcapsules physicochemical properties by modulating core-shell interactions.

Abstract:Encapsulation has emerged as a vitally important tool for protecting the integrity of critical chemicals and improving the delivery mechanisms of natural/synthetic drugs, enabling the controlled release of ingredients and the maintenance of their chemical, physical, and biological properties. It is well known that essential oils (EOs) provide a valuable alternative for food preservation, as they help reducing the deterioration of foodstuffs as well as the proliferation of pathogens. Nevertheless, EOs are highly volatile and lipophilic, rendering them insoluble in aqueous systems. In addition, their secondary metabolites are extremely susceptible to environmental factors such as humidity, temperature, light, and oxygen. Therefore, encapsulation of EOs is an innovative option not only for preserving these substances but also for promoting their stability, controlling their release, and optimizing their efficiency and bioavailability. In this sense, this review aimed to describe current techniques and approaches used to incorporate natural hydrophobic compounds, covering EOs. It also examines whether encapsulation technology can be used efficiently in drug discovery and development. Studies have shown that microencapsulation, the use of nanoparticle, and liposomal are the most effective techniques for encapsulating EOs. Other encapsulation systems included spray drying, coacervation, and emulsification.

Abstract:<span>The microencapsulation of essential oils by complex coacervation continues to attract considerable attention due to high payload, increased thermal stability and sustained release of core materials. This review recapitulates the thermal properties of coacervates and essential oil microcapsules prepared by complex coacervation method using protein/polysaccharide and polysaccharide/polysaccharide. The authors discussed the factors affecting coacervation and the thermal properties of coacervates. Besides, this review describes the microencapsulation processes physicochemical properties and release characteristics of essential oils microcapsules based on complex coacervation method. Finally, the review concentrates on the antimicrobial properties and the applications of essential oils microcapsules in food and nutrition. Despite extensive research conducted on the preparation of essential oils microcapsules prepared by complex coacervation, the application of this technique in encapsulating essential oils exposed to high temperatures during processing and storage remains a current area of research. Therefore, the research consolidated in this review describes a high degree of thermal stability of essential oils microcapsules prepared with complex coacervation that is yet understood, which can be readily utilized in the food and pharmaceutical industries.</span>

Microencapsulation of Bifidobacterium lactis and Lactobacillus plantarum within a Novel Polysaccharide-Based Core-Shell Formulation: Improving Probiotic Viability and Mucoadhesion

Abstract:Probiotics health benefits are hampered by long-term storage, gastrointestinal transit, and lack of adequate colonization within the colon. To this end, we have designed a core-shell structure that features an acid resistant core formulation with low water activity composed of alginate, hydroxypropyl methyl cellulose, and gellan gum (AHG) and a mucoadhesive shell made from chemically modified carboxymethyl chitosan with polyethylenimine (PEI-CMC). The structure of the core-shell microparticles was examined using scanning electron microscopy, and rheological measurements confirmed the improved ionic interactions between the core and the shell using the PEI-modified CMC. Simulated release from core-shell microparticles using polystyrene beads showed preferential release under intestinal conditions. PEI-CMC coating yielded improvements in mucoadhesion that was consistent with a positive shift in surface charge of the particles. Ex vivo studies using Bifidobacterium lactis probiotic bacteria demonstrated a 1.1 × 105-fold improvement in bacterial viability with encapsulation under storage conditions of high humidity and temperature (30 °C). When exposed to simulated gastric fluid, encapsulation increased the probiotic viability by 3.0 × 102-fold. In vivo studies utilizing bioluminescent Lactobacillus plantarum in mice revealed that encapsulation extended the duration of the signal within the gut and resulted in higher plate counts in suspensions isolated from the cecum. Conversely, we observed an abrupt loss of signal in the gut of the free probiotic. In conclusion, this core-shell system is suitable for improving probiotic shelf life and maximizing delivery to and retention by the colon.

Abstract:As a translucent functional gel with biodegradability, non-toxicity and acid resistance, gellan gum has been widely used in probiotic packaging, drug delivery, wound dressing, metal ion adsorption and other fields in recent years. Because of its remarkable gelation characteristics, gellan gum is suitable as the shell material of microcapsules to encapsulate functional substances, by which the functional components can improve stability and achieve delayed release. In recent years, many academically or commercially reliable products have rapidly emerged, but there is still a lack of relevant reports on in-depth research and systematic summaries regarding the process of microcapsule formation and its corresponding mechanisms. To address this challenge, this review focuses on the formation process and applications of gellan gum-based microcapsules, and details the commonly used preparation methods in microcapsule production. Additionally, it explores the impact of factors such as ion types, ion strength, temperature, pH, and others present in the solution on the performance of the microcapsules. On this basis, it summarizes and analyzes the prospects of gellan gum-based microcapsule products. Fabrication of thermo-responsive multicore microcapsules using a facile extrusion process

Abstract:A process employing extrusion was used to produce multicore microcapsules composed of multiple beads. The inner beads were made from κ-carrageenan (κ-c), a thermo-responsive linear sulphated polymer whose gelling temperature ranges at 40–60 °C, depending on the concentration of κ-c polymer and the amount of potassium chloride used for gelation. The resulting beads were then enveloped by chitosan through gelation with sodium triphosphate. The pesticide ammonium glufosinate was encapsulated in the κ-c/chitosan multicore microcapsules for demonstration of controlled release of the encapsulant. It was found that in response to an external stimulus, such as elevated temperature or solar simulation, the microcapsules exhibit the gradual release of encapsulated pesticide molecules from multicore microcapsules, compared with beads only. This process of making multicore microcapsules can be extended to other polymer pairs based on applications. This work is relevant to agriculture, where the controlled-release of the pesticides or fertilizers could be triggered by the sun and/or temperature changes, thus extending the residual period of the chemicals as well as decreasing the extent of pollution by leaching of abundant chemicals.

Abstract:Co-precipitation is generally refers to the co-precipitation of two solids and is widely used to prepare active-loaded nanoparticles. Here, it is demonstrated that liquid and solid can precipitate simultaneously to produce hierarchical core-shell nanocapsules that encapsulate an oil core in a polymer shell. During the co-precipitation process, the polymer preferentially deposits at the oil/water interface, wetting both the oil and water phases; the behavior is determined by the spreading coefficients and driven by the energy minimization. The technique is applicable to directly encapsulate various oil actives and avoid the use of toxic solvent or surfactant during the preparation process. The obtained core-shell nanocapsules harness the advantage of biocompatibility, precise control over the shell thickness, high loading capacity, high encapsulation efficiency, good dispersity in water, and improved stability against oxidation. The applications of the nanocapsules as delivery vehicles are demonstrated by the excellent performances of natural colorant and anti-cancer drug-loaded nanocapsules. The core-shell nanocapsules with a controlled hierarchical structure are, therefore, ideal carriers for practical applications in food, cosmetics, and drug delivery.

Abstract:Bioactive substances can enhance host health by modulating biological reactions, but their absorption and utilization by the body are crucial for positive effects. Encapsulation of probiotics is rapidly advancing in food science, with new approaches such as 3D printing, spray-drying, microfluidics, and cryomilling. Co-encapsulation with bioactives presents a cost-effective and successful approach to delivering probiotic components to specific colon areas, improving viability and bioactivity. However, the exact method by which bioactive chemicals enhance probiotic survivability remains uncertain. Co-crystallization as an emerging encapsulation method improves the physical characteristics of active components. It transforms the structure of sucrose into uneven agglomerated crystals, creating a porous network to protect active ingredients. Likewise, electrohydrodynamic techniques are used to generate fibers with diverse properties, protecting bioactive compounds from harsh circumstances at ambient temperature. Electrohydrodynamic procedures are highly adaptable, uncomplicated, and easily expandable, resulting in enhanced product quality and functionality across various food domains. Furthermore, food byproducts offer nutritional benefits and technical potential, aligning with circular economy principles to minimize environmental impact and promote economic growth. Hence, industrialized nations can capitalize on the growing demand for functional foods by incorporating these developments into their traditional cuisine and partnering with businesses to enhance manufacturing and production processes.

Abstract:The coating process for solid dosage forms is widely used in the pharmaceutical industry but presents challenges for small-scale production, needed in personalized medicine and clinical or galenic settings. This study aimed to evaluate immersion coating, a cost-effective small-scale method, for enteric-coated gelatin capsules using standard equipment. Two enteric coating polymers and different polymer concentrations were tested, along with API solubility. Results were compared with commercially available enteric capsule shells. Successful preparation of enteric coating capsules via immersion necessitates a comprehensive grasp of API and enteric polymer behavior. However, utilizing commercially available enteric capsule shells does not guarantee ease or robustness, as their efficacy hinges on the attributes of the active ingredient and excipients. Notably, coating with Eudragit S100 stands out for its superior process robustness, requiring minimal or no development time, thus representing the best option for small-scale enteric capsule production.

Abstract:Microencapsulation is an advanced methodology for the protection, preservation, and/or delivery of active materials in a wide range of industrial sectors, such as pharmaceuticals, cosmetics, fragrances, paints, coatings, detergents, food products, and agrochemicals. Polymeric materials have been extensively used as microcapsule shells to provide appropriate barrier properties to achieve controlled release of the encapsulated active ingredient. However, significant limitations are associated with such capsules, including undesired leaching and the nonbiodegradable nature of the typically used polymers. In addition, the energy cost of manufacturing microcapsules is an important factor to be considered when designing microcapsule systems and the corresponding production processes. Recent factors linked to UN sustainability goals are modifying how such microencapsulation systems should be designed in pursuit of "ideal" microcapsules that are efficient, safe, cost-effective and environmentally friendly. This review provides an overview of advances in microencapsulation, with emphasis on sustainable microcapsule designs. The key evaluation techniques to assess the biodegradability of microcapsules, in compliance with recently evolving European Union requirements, are also described. Moreover, the most common methodologies for the fabrication of microcapsules are presented within the framework of their energy demand.
