Abstract:Cell therapies have great potential for the treatment of many different diseases, while the direct application of cells to the targeted location leads to limited therapeutic outcomes due to the low cell engraftment and cell survival rate. Injectable hydrogels have been developed to facilitate cell delivery; however, those currently developed hydrogel systems still face the limited cell survival rate. Here, an injectable and self-healable hydrogel is reported through the combination of hyperbranched PEG-based multi-hydrazide macro-crosslinker (HB-PEG-HDZ) and aldehyde-functionalized hyaluronic acid (HA-CHO), with gelatin added to increase the crosslinking density and cell activity. The hydrogels can be formed only in 7 s due to the relatively high content of the functional end groups. The reversible crosslinking mechanism between the hydrazide and aldehyde groups endows the hydrogel with shear-thinning and self-healing properties. The hydrogels with gelatin exhibit relatively better mechanical properties and cell activity. The hydrogels can improve the survival, attachment, and engraftment of injected cells due to the rapid sol-gel transition, which can promote an enhanced regenerative response.

Abstract:Hydrogel microfibers have many fascinating applications as microcarriers for drugs, factors, and cells, such as 3D cell culture, building micro-organoids, and transplantation therapy due to their simple structures. It is unknown whether cell-laden fiber can become a standard-use product like woundplast. Here, from the technical and practical view, the elements required for user-oriented microfibers are first discussed: i) the materials used should promote cell functionalization and be easily processed; ii) follow a manufacturing method for mass fabrication; iii) have the ability to be stored long-term and be available for immediate use. Here, it is demonstrated that bioactive microfibers can be simply fabricated with coaxial bioprinting using gelatin methacrylate due to its tunable biological and mechanical properties. Additionally, programmed microfibers and 3D constructs with controllable composition can also be fabricated. These microfibers can be used to directly build organoids and complex co-culture tissue models. In the present study, vascular organoid, angiogenic sprouts, and tumor angiogenesis are demonstrated. It is also demonstrated, for the first time, that the cell-laden microfibers can be stored long-term via cryopreservation. These results show that cell-laden structures can be developed as a novel type of organoid product, which will open more avenues for tissue engineering and clinical organ repair.

Abstract:Over the recent years, various materials have been introduced as potential 3D cell culture scaffolds. These include protein extracts, peptide amphiphiles, and synthetic polymers. Hydrogel scaffolds without human or animal borne components or added bioactive components are preferred from the immunological point of view. Here we demonstrate that native nanofibrillar cellulose (NFC) hydrogels derived from the abundant plant sources provide the desired functionalities. We show 1) rheological properties that allow formation of a 3D scaffold in-situ after facile injection, 2) cellular biocompatibility without added growth factors, 3) cellular polarization, and 4) differentiation of human hepatic cell lines HepaRG and HepG2. At high shear stress, the aqueous NFC has small viscosity that supports injectability, whereas at low shear stress conditions the material is converted to an elastic gel. Due to the inherent biocompatibility without any additives, we conclude that NFC generates a feasible and sustained microenvironment for 3D cell culture for potential applications, such as drug and chemical testing, tissue engineering, and cell therapy.

Fabrication of Liver Microtissue with Liver Decellularized Extracellular Matrix (decm) Bioink by Digital Light Processing (DLP) Bioprinting

Abstract:As one of the most effective treatments of end-stage liver disease, liver transplantation still suffers from a shortage of donor organs or a low degree of engraftment. Thus, alternatives to liver transplantation, such as liver support systems, have to be extensively explored. In this study, a novel liver microtissue with an inner gear-like structure, which achieved a larger body surface area, was designed and manufactured to improve hepatic functional restoration. The liver-specific bioinks were developed by combining photocurable methacrylated gelatin (GelMA) with liver decellularized extracellular matrix (dECM), and human-induced hepatocytes (hiHep cells) were encapsulated to form cell-laden bioinks. The mechanical properties, swelling, and cytocompatibility of GelMA/dECM bioinks were carefully characterized before 3D printing. Then, the digital light process (DLP)-based bioprinting was used to fabricate the liver microtissue, and liver dECM was found to improve both the printability and cell viability of GelMA bioinks. hiHep cells were also found to spread farther and have better hepatocyte-specific functions (albumin secretion and urea) in the liver microtissue when liver dECM was added to the GelMA bioinks. Our results provide a promising liver dECM-based cell-laden bioink for liver microtissue fabrication, which would be a potential liver tissue engineering product to help restore hepatic functions.

Abstract:The liver, as one of the vital organs in the body, plays a crucial role in various bodily functions. Numerous factors can cause liver damage, that the sole remedy for severe liver conditions is the transplantation of healthy liver tissue. In response to the transplantation challenges, innovative approaches involving hydrogel-based technologies have emerged, leading to the creation of highly functionalized tissues. The development of three-dimensional printing and patterning of cell-laden biomaterial matrices offers promising advances for creating tissue-specific structures in tissue engineering and bioprinting. However, the matrix materials currently employed in bioprinting liver microtissue often fail to capture the complexity of the natural extracellular matrix (ECM), hindering their ability to restore innate cellular shapes and functions. Liver ECM-based hydrogels are increasingly recognized for their potential as biomimetic 3D cell culture systems that facilitate the exploration of liver disease, metabolism, and toxicity mechanisms. Yet, the conventional production of these hydrogels relies on slow thermal gelation processes, which restrict the manipulation of their mechanical characteristics. In this research, we introduce a novel approach with a functionalized photocrosslinkable liver decellularized extracellular matrix (dECM). By combining liver dECM methacrylate (LdECMMA) with gelatin methacrylate (GelMA), we achieved accelerated crosslinking under visible light irradiation and the ability to tune the mechanical, rheological, and physiological properties of the material. We encapsulated human hepatocellular carcinoma cells within an optimal concentration of the GelMA-LdECMMA hybrid hydrogel and examined cell proliferation and function over an extended period. The findings revealed that the GelMA-LdECMMA hybrid hydrogel enhances liver cell proliferation and function, holding significant promise for applications in drug screening and liver cancer metastasis research.

Abstract:The rapid development of additive manufacturing techniques in the field of tissue regeneration offers unprecedented success for artificial tissue and organ fabrication. However, some limitations still remain for current bioinks, such as the compromised cell viability after printing, the low cross-linking efficiency leading to poor printing resolution and speed due to the relatively slow gelation rate, and the requirement of external stimuli for gelation. To address these problems, herein, a biocompatible and printable instant gelation hydrogel system has been developed based on a designed hyperbranched poly(ethylene glycol) (PEG)-based multihydrazide macro-cross-linker (HB-PEG-HDZ) and an aldehyde-functionalized hyaluronic acid (HA-CHO). HB-PEG-HDZ is prepared by the postfunctionalization of hyperbranched PEG-based multivinyl macromer via thiol-ene chemistry. Owing to the high functional group density of HB-PEG-HDZ, the hydrogel can be formed instantly upon mixing the solutions of two components. The reversible cross-linking mechanism between the hydrazide and aldehyde groups endows the hydrogel with shear-thinning and self-healing properties. The minimally toxic components and cross-linking chemistry allow the resulting hydrogel to be a biocompatible niche. Moreover, the fast sol-to-gel transition of the hydrogel, combining all of the advanced characteristics of this platform, protects the cells during the printing procedure, avoids their damage during extrusion, and improves the transplanted cell survival.

Abstract:BACKGROUND:Because of an increasing discrepancy between the number of potential liver graft recipients and the number of organs available, scientists are trying to create artificial liver to mimic normal liver function and therefore, to support the patient's liver when in dysfunction. 3D printing technique meets this purpose. The present study was to test the feasibility of 3D hydrogel scaffolds for liver engineering.METHODS:We fabricated 3D hydrogel scaffolds with a bioprinter. The biocompatibility of 3D hydrogel scaffolds was tested. Sixty nude mice were randomly divided into four groups, with 15 mice in each group: control, hydrogel, hydrogel with L02 (cell line HL-7702), and hydrogel with hepatocyte growth factor (HGF). Cells were cultured and deposited in scaffolds which were subsequently engrafted into livers after partial hepatectomy and radiation-induced liver damage (RILD). The engrafted tissues were examined after two weeks. The levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), albumin, total bilirubin, CYP1A2, CYP2C9, glutathione S-transferase (a-GST), and UDP-glucuronosyl transferase (UGT-2) were compared among the groups. Hematoxylin-eosin (HE) staining and immunohistochemistry of cKit and cytokeratin 18 (CK18) of engrafted tissues were evaluated. The survival time of the mice was also compared among the four groups.RESULTS:3D hydrogel scaffolds did not impact the viability of cells. The levels of ALT, AST, albumin, total bilirubin, CYP1A2, CYP2C9, a-GST and UGT-2 were significantly improved in mice engrafted with 3D scaffold loaded with L02 compared with those in control and scaffold only (P<0.05). HE staining showed clear liver tissue and immunohistochemistry of cKit and CK18 were positive in the engrafted tissue. Mice treated with 3D scaffold+L02 cells had longer survival time compared with those in control and scaffold only (P<0.05).CONCLUSION:3D scaffold has the potential of recreating liver tissue and partial liver functions and can be used in the reconstruction of liver tissues.
Optimization of collagen type I-hyaluronan hybrid bioink for 3D bioprinted liver microenvironments

Abstract:Current 3D printing of tissue is restricted by the use of biomaterials that do not recapitulate the native properties of the extracellular matrix (ECM). These restrictions have thus far prevented optimization of composition and structure of the in vivo tissue microenvironment. The artificial nature of currently used biomaterials affects cellular phenotype and function of the bioprinted tissues, and results in inaccurate modeling of disease and drug metabolism significantly. Collagen type I is the major structural component in the ECM, and is widely used as a 3D hydrogel, but is less applicable for 3D bioprinting due to low viscosity and slow polymerization. We have hypothesized that a combination of hyaluronic acid with collagen I yields a bioink with the properties required for extrusion bioprinting, while supporting native cell-matrix interactions and preservation of the native microenvironment properties. To test this hypothesis, we tested the viscoelastic properties of three bioink formulations -2:1, 3:1, and 4:1 collagen type I to hyaluronic acid, and examined cellular behavior in order to determine an optimal formulation that allows for bioprinting while supporting biological activity. We then employed this formulation to bioprint 3D liver tissue constructs containing primary human hepatocytes and liver stellate cells and tested the effects of acetaminophen, a common liver toxicant. Our results have shown that the combination of methacrylated collagen type I and thiolated hyaluronic acid yield a simple, printable bioink that allows for modulation that was directly related to stromal cell elongation. Further, the bioink adequately allowed for implementation as a support hydrogel for hepatocytes which were able to remain viable over two weeks and responded to drug treatment appropriately.

3D Printing of Vascularized Hepatic Tissues with a High Cell Density and Heterogeneous Microenvironment.

Abstract:Three-dimensional bioprinting has emerged as an appealing approach for creating functional tissues; however, a lack of suitable bioinks with high cell density and printability has greatly limited our ability to print functional tissues. We address this limitation by developing a granular cell aggregate-based biphasic (GCAB) bioink based on densely packed cell aggregates. The GCAB bioink exhibited the desired shear-thinning and shear-recovery properties for extrusion bioprinting and hyperelastic behaviors postprinting for modeling the mechanical characteristics of soft biological tissues. The GCAB bioink displayed a high cell density (∼1.7 × 108cells cm-3) without compromising viability (∼83%). We printed dense hepatic tissue constructs with enhanced vascularization and metabolic functions by preorganization of GCAB bioink with a defined heterogeneous microenvironment. By simultaneously printing the GCAB bioink and an endothelial cell-laden gelatin bioink, we successfully produced functional hepatic tissues with a high cell density and a perfusable vascular network. The design of the generalizable GCAB bioink opens new avenues to create functional tissues for therapeutic applications.
Preparation of Chitosan-based Injectable Hydrogels and Its Application in 3D Cell Culture

Abstract:The protocol presents a facile, efficient, and versatile method to prepare chitosan-based hydrogels using dynamic imine chemistry. The hydrogel is prepared by mixing solutions of glycol chitosan with a synthesized benzaldehyde terminated polymer gelator, and hydrogels are efficiently obtained in several minutes at room temperature. By varying ratios between glycol chitosan, polymer gelator, and water contents, versatile hydrogels with different gelation times and stiffness are obtained. When damaged, the hydrogel can recover its appearances and modulus, due to the reversibility of the dynamic imine bonds as crosslinkages. This self-healable property enables the hydrogel to be injectable since it can be self-healed from squeezed pieces to an integral bulk hydrogel after the injection process. The hydrogel is also multi-responsive to many bio-active stimuli due to different equilibration statuses of the dynamic imine bonds. This hydrogel was confirmed as bio-compatible, and L929 mouse fibroblast cells were embedded following standard procedures and the cell proliferation was easily assessed by a 3D cell cultivation process. The hydrogel can offer an adjustable platform for different research where a physiological mimic of a 3D environment for cells is profited. Along with its multi-responsive, self-healable, and injectable properties, the hydrogels can potentially be applied as multiple carriers for drugs and cells in future bio-medical applications.
A highly biocompatible bio-ink for 3D hydrogel scaffolds fabrication in the presence of living cells by two-photon polymerization

Abstract:Bio-inks with living cells encapsulation are core materials for 3D printing of biomimetic cell culture models in vitro. In this study, a highly biocompatible bio-ink containing a hydrophilic benzylidene cyclopentanone photoinitiator (Y4) and a glycidyl methacrylate modified gelatin (Gel-GM) was prepared, and 3D hydrogel scaffolds with normal mouse fibroblast cells (L929) encapsulation were accurately fabricated by two-photon polymerization on a multiphoton confocal scanning laser microscopy. No apparent damage to L929 cells was observed within the laser-exposed region during the fabrication process. After four Days’ incubation, the embedded L929 cells showed a vigorous proliferation and occupied most of the available space within the 3D hydrogel scaffolds, which indicated that the bio-ink would have great potential for 3D bioprinting.

Abstract:<p>Direct injection of cell‐laden hydrogels shows high potential for tissue regeneration in translational therapy. The traditional cell‐laden hydrogels are often used as bulk space fillers to tissue defects after injection, likely limiting their structural controllability. On the other hand, patterned cell‐laden hydrogel constructs often necessitate invasive surgical procedures. To overcome these problems, herein, a unique strategy is reported for encapsulating living human cells in a pore‐forming gelatin methacryloyl (GelMA)‐based bioink to ultimately produce injectable hierarchically macro‐micro‐nanoporous cell‐laden GelMA hydrogel constructs through 3D extrusion bioprinting. The hydrogel constructs can be fabricated into various shapes and sizes that are defect‐specific. Due to the hierarchically macro‐micro‐nanoporous structures, the cell‐laden hydrogel constructs can readily recover to their original shapes, and sustain high cell viability, proliferation, spreading, and differentiation after compression and injection. In addition, in vivo studies further reveal that the hydrogel constructs can integrate well with the surrounding host tissues. These findings suggest that the unique 3D‐bioprinted pore‐forming GelMA hydrogel constructs are promising candidates for applications in minimally invasive tissue regeneration and cell therapy.</p>

Preparation, Mechanical and Biological Properties of Inkjet Printed Alginate/Gelatin Hydrogel

Abstract:3D printing has made remarkable progress in soft tissue reconstruction enabling the custom design of complex material implants with patient specific geometry. The aim of this study was to inkjet print mechanically reinforced biocompatible hydrogels. Here, we developed a double crosslinked ink by optimizing the rheological properties of solutions of sodium alginate (NaAlg), NaAlg/transglutaminase (TG), CaCl2 and gelatin/CaCl2. The results showed that a two-component ink system comprising NaAlg (4% w/v)/TG (0.8% w/v) and gelatin (4% w/v)/CaCl2 (3% w/v) gave optimum printability. The mechanical and biological properties of printed alginate/gelatin hydrogels prepared from inks with different gelatin contents, and incorporated fibroblasts, were characterized by Scanning Electron Microscope (SEM), mechanical testing and laser confocal microscopy. The compressive moduli of alginate/gelatin hydrogels could be adjusted from 19.2 kPa ± 1.2 kPa to 65.9 kPa ± 3.3 kPa by increasing the content of gelatin. After incubation for 7 d, fibroblasts had permeated all printed hydrogels and the rate of proliferation increased with increasing gelatin content. The highest cell proliferation rate (497%) was obtained in a hydrogel containing 4.5% (w/v) gelatin. This study offers a new strategy for the fabrication of 3D structures used to mimic the function of native tissues.

Fibrin Hydrogels for Endothelialized Liver Tissue Engineering with a Predesigned Vascular Network

Abstract:The design and manufacture of a branched vascular network is essential for bioartificial organ implantation, which provides nutrients and removes metabolites for multi-cellular tissues. In the present study, we present a technology to manufacture endothelialized liver tissues using a fibrin hydrogel and a rotational combined mold. Both hepatocytes and adipose-derived stem cells (ADSCs) encapsulated in a fibrin hydrogel were assembled into a spindle construct with a predesigned multi-branched vascular network. An external overcoat of poly(dl-lactic-co-glycolic acid) was used to increase the mechanical properties of the construct as well as to act as an impervious and isolating membrane around the construct. Cell survivability reached 100% in the construct after 6 days of in vitro culture. ADSCs in the spindle construct were engaged into endothelial cells/tissues using a cocktail growth factor engagement approach. Mechanical property comparison and permeability evaluation tests all indicated that this was a viable complex organ containing more than two heterogeneous tissue types and a functional vascular network. It is, therefore, the first time an implantable bioartificial liver, i.e., endothelialized liver tissue, along with a hierarchical vascular network, has been created.
A facile, versatile hydrogel bioink for 3D bioprinting benefits long-term subaqueous fidelity, cell viability and proliferation

Abstract:Both of the long-term fidelity and cell viability of three-dimensional (3D)-bioprinted constructs are essential to precise soft tissue repair. However, the shrinking/swelling behavior of hydrogels brings about inadequate long-term fidelity of constructs, and bioinks containing excessive polymer are detrimental to cell viability. Here, we obtained a facile hydrogel by introducing 1% aldehyde hyaluronic acid (AHA) and 0.375% N-carboxymethyl chitosan (CMC), two polysaccharides with strong water absorption and water retention capacity, into classic gelatin (GEL, 5%)-alginate (ALG, 1%) ink. This GEL-ALG/CMC/AHA bioink possesses weak temperature dependence due to the Schiff base linkage of CMC/AHA and electrostatic interaction of CMC/ALG. We fabricated integrated constructs through traditional printing at room temperature and in vivo simulation printing at 37°C. The printed cell-laden constructs can maintain subaqueous fidelity for 30 days after being reinforced by 3% calcium chloride for only 20 s. Flow cytometry results showed that the cell viability was 91.38 ± 1.55% on day 29, and the cells in the proliferation plateau at this time still maintained their dynamic renewal with a DNA replication rate of 6.06 ± 1.24%. This work provides a convenient and practical bioink option for 3D bioprinting in precise soft tissue repair.

However, one of the key problems hindering the application and development of 3D printing in TE area is the poor mechanical property of bio-inks. In this work, we aimed to design a high-strength hydrogel system based on dialdehyde cellulose nanocrystals (DAC) and gelatin (GEL) as a new bio-ink for 3D printing of scaffolds. The DAC were prepared and used as a natural crosslinker to interact with the GEL through a Schiff base reaction. The mechanical test results indicated that the breaking strength of the optimal 4:8-DAC/GEL sample was almost 41.3-fold greater than that of the GEL hydrogel. According to the rheological test results, the 4:8-DAC/GEL sample incubated for 3 h was proposed as a bio-ink for 3D printing. Then, the printing conditions, including the printing pressure and nozzle speed, as well as additional crosslinking conditions for a freshly printed scaffold, i.e., the crosslinking time and temperature, were optimized. Crosslinked scaffolds with adjustable porosity and good fidelity were successfully obtained. The biocompatibility of 4:8-DAC/GEL was also investigated by CCK-8 and Hoechst 33342/PI double-staining assays. Collectively, these results confirm the good potential of the 4:8-DAC/GEL hydrogel as a 3D bio-ink for application in tissue repair.

Abstract:Liver diseases represent a considerable burden to patients and healthcare systems. Hydrogels play an important role in the engineering of soft tissues and may be useful for embedding hepatocytes for different therapeutic interventions or the development of in vitro models to study the pathogenesis of liver diseases or testing of drugs. Here, we developed two types of hydrogels by crosslinking hydrazide-functionalized gelatin with either oxidized dialdehyde hyaluronan or alginate through the formation of hydrazone bonds. Gel formulations were studied through texture analysis and rheometry, showing mechanical properties comparable to those of liver tissue while also demonstrating long-term stability. The biocompatibility of hydrogels and their ability to host hepatocytes was studied in vitro in comparison to pure gelatin hydrogels crosslinked by transglutaminase using the hepatocellular line HepG2. It was found that HepG2 cells could be successfully embedded in the hydrogels, showing no signs of gel toxicity and proliferating in a 3D environment comparable to pure transglutaminase cross-linked gelatin hydrogels used as control. Altogether, hydrazide gelatin in combination with oxidized polysaccharides makes stable in situ gelling systems for the incorporation of hepatocytes, which may pave the way for use in liver tissue engineering and drug testing.

Abstract:Cell adhesion is the physical basis of tissue formation. Developing a kind of bioink with tunable mechanical properties and the promotion of cell adhesion is a major challenge for tissue engineering. In the present paper, a kind of hydrogel with good printable performance was developed, and cell adhesion was promoted in cell-laden DLP (Digital Light Processing) 3D bioprinting as a flexible solution for customized 3D structures. The hydrogel was prepared by modifying gelatin methacryloyl (GelMA)/hyaluronic acid methacryloyl (HAMA) with silk fibroin methacryloyl (SilMA). The three kinds of materials contain significant components of the extracellular matrix (ECM), such as glycosaminoglycan and proteins. Rheological results indicate that the multicomponent hydrogel precursors exhibit good photocuring behavior and are very suitable for DLP 3D bioprinting. The mechanical and biological properties of hydrogel can be tuned flexibly by SilMA content. Besides this, the SilMA/GelMA/HAMA hydrogel shows higher adhesion strength than the GelMA/HAMA hydrogel. The addition of SilMA into GelMA/HAMA significantly increased cell actin expression and promoted cell adhesion. Furthermore, the cell-laden 3D printed SilMA/GelMA/HAMA porous scaffold structure can remain stable after 7 days of in vitro culture. The survival rate of L929 fibroblasts can be up to 87%, exhibiting a good cell morphology and proliferation. All these results indicate that the SilMA/GelMA/HAMA hydrogel promoting cell adhesion will have broad implications for customized 3D structures in tissue engineering.
