Abstract:BACKGROUND: Over the course of evolution, plants have developed various sophisticated defense mechanisms to resist pests and diseases. The phytohormone abscisic acid (ABA) has an important role in the growth and development of plants and confers tolerance to selected abiotic stressors, such as drought. Previous studies have shown that ABA promotes the deposit of callose in response to piercing/sucking insect pests. The English grain aphid, Sitobion avenae Fabricius, causes huge losses in rice and is especially harmful to rice seedlings. RESULTS: Exogenous ABA promoted growth and reduced the feeding behavior of S. avenae nymphs in rice. Our results suggested that enhanced trichome density and increased expression of related genes may be associated with rice resistance to aphids. An analysis of volatiles revealed the production of seven compounds associated with pest resistance. CONCLUSION: These results indicate that ABA reduces aphid feeding in rice. Our findings provide a basis for understanding ABA-mediated defense responses in rice and provide insights on more environmentally-friendly approaches to control. Comprehensive Insights into Sitobion avenae Preferences and Performance on Pakistan's Wheat Cultivars Leading to Identification of Potential RNAi Targets

Abstract:Sitobion avenae, a notable hemipteran pest, poses a significant economic threat to Triticum aestivum due to its short generation times and high reproductive rates. Challenges like the development of insecticide resistance, the limited impact of insecticidal genes, and associated risks led to seeking a more precise approach like RNA interference. This study evaluated S. avenae response on seven different local cultivars (Anaj-2021, Subhani-2022, Fakhar-e-Bhakkar-2021, Akbar-2019, Mexi-Pak-2022, Barani-2022, & Dilkash-2022) through aphid preference test, aphid choice assay, and aphid performance test. Further, differential proteomics of S. avenae (pre- and post-feeding on susceptible and resistant wheat cultivars) was performed using Sodium Dodecyl Sulphate-Polyacrylamide Gel Electrophoresis. Among the local wheat cultivars, Anaj-2021 was regarded as the most susceptible cultivar while Barani-2022 was declared the most resistant. The differential proteome analysis of Anaj-2021 (S), and Barani-2022 (R) show 11 upregulated proteins including Glutathione S- transferase, Cathepsin, Carbonic anhydrases, Ecdysone induced protein, Odorant binding protein 3, Heat shock protein, Salivary effector protein, SID1-like protein, Sodium channel protein, chemosensory protein, and trypsin were upregulated in S. avenaeon wheat feeding as compared to non-feeding. Trypsin, cathepsin-B and carbonic anhydrases are connected to detoxification and digestion. While odorant binding proteins, salivary effector proteins, sodium channel proteins and ecdysone- induced proteins facilitate feeding process in S. avenae. The enhanced expression of proteins having detoxification, digestion or defense activity implicates their essential role in the survival of S. avenae. Therefore, these proteins have the potential to serve as RNA interference targets, against which double-stranded RNA could be designed and expressed in wheat cultivars to make them resistant to local S. avenae infestation and avert yield loss.

Threonyl-tRNA Synthetase Gene, a Potential Target for RNAi-based Control of Three Rice Planthoppers

Abstract:Background RNA interference (RNAi) has potential as a new strategy for pest control. However, the current overemphasis on the control of a single pest increased control costs. The aim of this study was to find a green method of controlling several pests without affecting the natural enemies with a single target gene. One possible RNAi target is the threonyl-tRNA synthetase (ThrRS), which is conserved and plays a significant role in protein biosynthesis. Results In this study, one threonyl-tRNA synthetase gene (NlthrS) was identified from the brown planthopper (Nilaparvata lugens). Spatio-temporal expression pattern analysis showed that NlthrS was highly expressed in the ovary, late embryogenesis, nymphs and female adults. In addition, RNAi-mediated knockdown of NlthrS caused 85.6% nymph mortality, 100% female infertility, molting disorder, extended nymph duration and shortened adult longevity. Target-specific results were obtained when dsNlthrS was used to interfere with the whiteback planthopper (Sogatella furcifera), small brown planthopper (Laodelphax striatellus), zig-zag winged leafhopper (Inazuma dorsalis) and their natural enemy (green mirid bug, Cyrtorhinus lividipennis). In addition, dsNlthrS could cause high mortalities of three species of planthoppers (85.6-100%), while only dsNlthrS-1 led to the death (97.3%) of I. dorsalis that was not affected by dsNlthrS-2. Furthermore, neither dsNlthrS-1 nor dsNlthrS-2 could influence the survival of C. lividipennis. Conclusion The results reveal the biological functions of ThrRS in N. lugens in addtion to its protein synthesis, deepening our understanding of tRNA synthase in insects and providing a new method for the control of several rice pests via one dsRNA design. Abstract:The current methods for pest managements in cereal crops rely mainly on the applications of conventional chemicals, which are known to have low pest control efficiencies and are less environmentally friendly. The primary aim of this research endeavor was to devise a specific, efficient and eco-friendly biological agent capable of mitigating maize lethal necrosis (MLN), a viral disease that affected maize productivity and mainly caused by coinfection of maize chlorotic mottle virus (MCMV) and sugarcane mosaic virus (SCMV). Through this study, we have found that silencing Ferredoxin 3 (ZmFd3) expression in maize can significantly reduce the damages caused by MCMV and/or SCMV infections. Based on this finding, we have developed a liposome nanoparticles (LNPs)mediated nanotechnology to suppress MCMV and/or SCMV infections. Using this nanotechnology, we have confirmed that the cationic/anionic liposomes (CLPs and ALPs) can be used to package and promote ZmFd3dsRNA spread in maize plants. Through leaf spray, we have determined that the anti-MCMV and anti-SCMV activities of ZmFd3@CLPs and ZmFd3@ALPs nanoparticles can reach 52.46 similar to 79.83 % at 6 days post virus inoculation (dpi). Because ZmFd3 is involved in hypersensitive response (HR), and can interact with MCMV p7b and SCMV HC-Pro, we designed an FHP-dsRNA to target ZmFd3 , MCMV p7b , and SCMV HC-Pro transcripts for silencing in plants simultaneously. The result showed that both FHP@CLPs and FHP@ALPs nanoparticles can provide excellent, although short, protection to maize and sorghum plants to MCMV and SCMV infections. Overall, this study has developed a novel nanotechnology that can be used to mitigate virus infection in cereal crops through RNA interference (RNAi).
Application of Double-Strand RNAs Targeting Chitin Synthase, Glucan Synthase, and Protein Kinase Reduces Fusarium graminearum Spreading in Wheat

Abstract:Controlling the devastating fungal pathogen Fusarium graminearum (Fg) is a challenge due to inadequate resistance in nature. Here, we report on the identification of RNAi molecules and their applications for controlling Fg in wheat through silencing chitin synthase 7 (Chs7), glucan synthase (Gls) and protein kinase C (Pkc). From transgenic Fg strains four RNAi constructs from Chs7 (Chs7RNAi−1, −2, −3, and −4), three RNAi constructs from Gls (GlsRNAi−2, −3, and −6), and one RNAi construct from Pkc (PkcRNAi−5) were identified that displayed effective silencing effects on mycelium growth in medium and pathogenicity in wheat spikes. Transcript levels of Chs7, Gls and Pkc were markedly reduced in those strains. Double-strand RNAs (dsRNAs) of three selected RNAi constructs (Chs7RNAi-4, GlsRNAi-6 and PkcRNA-5) strongly inhibited mycelium growth in vitro . Spray of those dsRNAs on detached wheat leaves significantly reduced lesion sizes; the independent dsRNAs showed comparable effects on lesions with combination of two or three dsRNAs. Expression of three targets Chs7, Gls, and Pkc was substantially down-regulated in Fg-infected wheat leaves. Further application of dsRNAs on wheat spikes in greenhouse significantly reduced infected spikelets. The identified RNAi constructs may be directly used for spray-induced gene silencing and stable expression in plants to control Fusarium pathogens in agriculture.

Abstract:Abstract Diseases adversely affect grain yield of crop plants. Leaf rust is a major disease of wheat. As race-specific resistance breaks down, introduction of newer sources of resistance often from older accessions is necessary. Linkage drag from the donor accessions adversely impacts grain yield. As a result, CIMMYT breeding effort has shifted to using durable resistance, also referred to as race non-specific host resistance, which allows slow rusting but maintains grain yield. One of the key genes for durable resistance is Lr67 and the resistant form of this gene is absent in CIMMYT elite lines. Hence, we have initiated efforts to convert the susceptible copy of Lr67 into its resistant form directly in these elite lines using gene editing. This would eliminate backcrossing and thus save time as well as eliminate linkage drag that would accompany the resistant copy of the gene if it were to be introgressed from an older accession. As first steps, we have isolated and sequenced the genomic copies from each of the A, B, and D genome of the Lr67 gene from three elite lines and an experimental line. Identification of more than 50 single nucleotide polymorphisms (SNPs) in the open reading frames (ORF) among these lines would be useful in designing the guide RNA molecules with precision. Further, we have streamlined genetic transformation of these elite lines, a prerequisite step for gene editing.
Double-stranded RNA in the biological control of grain aphid (Sitobion avenae F.)

Abstract:Grain aphid (Sitobion avenae F.) is the most dominant and destructive pest of wheat, which causes significant yield loss of cereal plants each year by inflicting damage both through the direct effects of feeding and by vectoring debilitating plant viruses. In this study, we performed de novo transcriptome sequencing of grain aphid via Roche 454 GS-FLX pyrosequencing. A total of 1,106,696 reads were obtained and assembled into 32,277 unigenes, of which 25,389, 21,635, and 16,211 unigenes matched the Nt, Nr, and Swiss-Prot databases, respectively. Functional annotation of these unigenes revealed not only the presence of genes that encode the key components of RNAi machinery such as Dicer and Argonaute but also the genes encoding the TAR RNA binding protein (TRBP) and the SID-1 protein, which function in assisting the RNA-induced silencing complex (RISC) formation in microRNA (miRNA) pathway and mediating a systemic RNA interference (RNAi) effect though a cellular uptake mechanism. Furthermore, among a set of 66 unigenes selected for a double-stranded RNA (dsRNA) artificial diet assay, four novel effective RNAi targets, which led to high mortality of aphids due to the down-regulation of the expression of the respective target gene, were identified. Moreover, the expansion of systemic RNAi effect in grain aphid was observed by adding the fluorescently labeled dsRNA in an artificial diet assay.

Spray method application of transdermal dsRNA delivery system for efficient gene silencing and pest control on soybean aphid Aphis glycines

Abstract:RNA interference (RNAi) has been recognized as a novel and safe strategy in pest management due to its high sequence-dependent specificity. However, the existing dsRNA delivery methods largely restrict the application of the RNAi-based pest management strategy; thus, we previously constructed a nanocarrier-based transdermal dsRNA delivery system on the soybean aphid Aphis glycines with the help of nanocarrier and detergent. In the current study, we improved our transdermal dsRNA delivery system with a smaller and cheaper nanocarrier to investigate the efficacy of spraying aphid-infested soybean seedlings to apply our RNA pesticide. A dsRNA/nanocarrier/detergent formulation was performed, and the dsRNA could penetrate the aphid body wall within 4 h with the help of nanocarrier through the topical application. Four potential RNAi target genes (TREH, ATPD, ATPE and CHS1) were selected and cloned, and their dsRNA fragments were synthesized and tested through the transdermal dsRNA delivery system. The delivered dsRNA efficiently silenced the target gene expression with the knockdown effects ranging from 86.86 to 58.87% and resulted in a high mortality up to 81.67% (dsATPD + dsATPE) through the topical application, when through the spray method, with the highest percent mortality of 78.50% (dsATPD + dsCHS1). Our novel transdermal dsRNA delivery system not only provides a powerful tool for gene functional analysis in laboratory, but also shows a great potential for the pest management in the field, which will promote the practice and development of RNAi-based pest management.

Abstract:BACKGROUND:The double-spotted leaf beetle (DLB), Monolepta hieroglyphica, is becoming a significant corn pest in China. It mainly attacks corn silk and developing kernels during the adult stage and is causing significant corn yield loss in north-eastern China. The damage caused by DLB is expected to worsen as pesticide usage is likely to decrease along with the upcoming commercial planting of transgenic lepidopteran-resistant maize in China. Therefore, it is highly desirable to develop transgenic corn for DLB resistance. RESULTS:Three target genes, MhSsj1, MhSnf7 and MhSec23A were cloned from DLB by their sequence similarity to their corresponding homologous genes known for their effectiveness as RNA interference (RNAi) targets in western corn rootworm (WCR, Diabrotica virgifera virgifera). Injection of the double-stranded RNAs (dsRNAs) of MhSsj1, MhSnf7 and MhSec23A to DLB adults was highly effective to suppress the messenger RNAs (mRNAs) of these genes and resulted in high mortality. Furthermore, a synergistic effect was observed among the dsRNAs of these three target genes. Transgenic maize plants simultaneously transcribing dsRNAs of any two of the three target genes were found to be highly resistant to DLB adults, showcasing the potential of utilizing RNAi-based strategy for transgenic DLB control. CONCLUSION:MhSsj1, MhSnf7 and MhSec23A are effective RNAi target genes and transgenic corn based on suppression of these genes by RNAi are effective for controlling adult DLB. © 2024 Society of Chemical Industry.

Abstract:The rice pest Nilaparvata lugens (the brown planthopper, BPH) has developed different levels of resistance to at least 11 chemical pesticides. RNAi technology has contributed to the development of environmentally friendly RNA biopesticides designed to reduce chemical use. Consequently, more precise targets need to be identified and characterized, and efficient dsRNA delivery methods are necessary for effective field pest control. In this study, a low off-target risk ds NlUAP fragment (166 bp) was designed in silico to minimize the potential adverse effects on non-target organisms. Knockdown of NlUAP via microinjection significantly decreased the content of UDP- N -acetylglucosamine and chitin, causing chitinous structural disorder and abnormal phenotypes in wing and body wall, reduced fertility, and resulted in pest mortality up to 100 %. Furthermore, ds NlUAP was loaded with ROPE@C, a chitosan-modified nanomaterial for spray application, which significantly downregulated the expression of NlUAP , led to 48.9 % pest mortality, and was confirmed to have no adverse effects on Cyrtorhinus lividipennis , an important natural enemy of BPH. These findings will contribute to the development of safer biopesticides for the control of N. lugens .

Spraying of dsRNA molecules derived from Phytophthora infestans, along with nanoclay carriers as a proof of concept for developing novel protection strategy for potato late blight

Abstract:Background: Phytophthora infestans is a late blight-causing oomycetes pathogen. It rapidly evolves and adapts to the host background and new fungicide molecules within a few years of their release, most likely because of the predominance of transposable elements in its genome. Frequent applications of fungicides cause environmental concerns. Here, we developed target-specific RNA interference (RNAi)-based molecules, along with nanoclay carriers, that when sprayed on plants are capable of effectively reducing late blight infection. Results: Targeted the genes unique to sporulation, early satge infection and the metabolism pathway stages based on in an our own microarray data. We used nanoclay as a carrier for sorbitol dehydrogenase, heat shock protein 90, translation elongation factor 1-α, phospholipase-D like 3 and glycosylphosphatidylinositol-anchored acidic serine-threonine-rich HAM34-like protein double-stranded (ds)RNAs, which were assessed by culture bioassay, detached leaf assay and spray methods, and revealed a reduction in growth, sporulation and symptom expression. Plants sprayed with multigene targeted dsRNA-nanoclay showed enhanced disease resistance (4% disease severity) and less sporulation (<1 × 103 ) compared with plants sprayed with dsRNA alone. Conclusion: The use of nanoclay with multigene targeted dsRNA was assumed to be involved in effective delivery, protection and boosting the action of RNAi as a spray-induced gene silencing approach (SIGS). A significant reduction in growth, sporulation, disease severity and decreased gene expression authenticates the effects of SIGS on late blight progression. This study demonstrated as a proof of concept the dsRNA-nanoclay SIGS approach, which could be used as an alternative to chemical fungicides and transgenic approaches to develop an environmentally friendly novel plant protection strategy for late blight. © 2022 Society of Chemical Industry.
Bacillus subtilis 26D Triggers Induced Systemic Resistance against Rhopalosiphum padi L. by Regulating the Expression of Genes AGO, DCL and microRNA in Bread Spring Wheat

Abstract:Bacillus subtilis 26D is a plant growth-promoting endophytic bacteria capable of inducing systemic resistance through the priming mechanism, which includes plant genome reprogramming and the phenomenon of RNA interference (RNAi) and microRNA (miRNAs). The phloem-feeding insect bird cherry-oat aphid Rhopalosiphum padi L. is a serious pest that causes significant damage to crops throughout the world. However, the function of plant miRNAs in the response to aphid infestation remains unclear. The results of this work showed that B. subtilis 26D stimulated aphid resistance in wheat plants, inducing the expression of genes of hormonal signaling pathways ICS, WRKY13, PR1, ACS, EIN3, PR3, and ABI5. In addition, B. subtilis 26D activated the RNAi mechanism and regulated the expression of nine conserved miRNAs through activation of the ethylene, salicylic acid (SA), and abscisic acid (ABA) signaling pathways, which was demonstrated by using treatments with phytohormones. Treatment of plants with SA, ethylene, and ABA acted in a similar manner to B. subtilis 26D on induction of the expression of the AGO4, AGO5 and DCL2, DCL4 genes, as well as the expression of nine conserved miRNAs. Different patterns of miRNA expression were found in aphid-infested plants and in plants treated with B. subtilis 26D or SA, ethylene, and ABA and infested by aphids, suggesting that miRNAs play multiple roles in the plant response to phloem-feeding insects, associated with effects on hormonal signaling pathways, redox metabolism, and the synthesis of secondary metabolites. Our study provides new data to further elucidate the fine mechanisms of bacterial-induced priming. However, further extensive work is needed to fully unravel these mechanisms.

The salivary effector protein Sg2204 in the greenbug Schizaphis graminum suppresses wheat defence and is essential for enabling aphid feeding on host plants

Abstract:Aphids secrete diverse repertoires of salivary effectors into host plant cells to promote infestation by modulating plant defence. The greenbug Schizaphis graminum is an important cereal aphid worldwide. However, the secreted effectors of S. graminum are still uncharacterized. Here, 76 salivary proteins were identified from the watery saliva of S. graminum using transcriptome and proteome analyses. Among them, a putative salivary effector Sg2204 was significantly up-regulated during aphid feeding stages, and transient overexpression of Sg2204 in Nicotiana benthamiana inhibited cell death induced by BAX or INF1. Delivering Sg2204 into wheat via the type III secretion system of Pseudomonas fluorescens EtAnH suppressed pattern-triggered immunity (PTI)-associated callose deposition. The transcript levels of jasmonic acid (JA)- and salicylic acid (SA)-associated defence genes of wheat were significantly down-regulated, and the contents of both JA and SA were also significantly decreased after delivery of Sg2204 into wheat leaves. Additionally, feeding on wheat expressing Sg2204 significantly increased the weight and fecundity of S. graminum and promoted aphid phloem feeding. Sg2204 was efficiently silenced via spray-based application of the nanocarrier-mediated transdermal dsRNA delivery system. Moreover, Sg2204-silenced aphids induced a stronger wheat defence response and resulted in negative impacts on aphid feeding behaviour, survival and fecundity. Silencing of Sg2204 homologues from four aphid species using nanocarrier-delivered dsRNA also significantly reduced aphid performance on host plants. Thus, our study characterized the salivary effector Sg2204 of S. graminum involved in promoting host susceptibility by suppressing wheat defence, which can also be regarded as a promising RNAi target for aphid control.

Abstract:Background: Cotton is a cash crop majorly affected by many hemipteran pests, among them the cotton mealybug, Phenacoccus solenopsis. Cotton mealybug attack has a devastating effect on cotton production and causes huge yield losses. Results: In this study, 25 potential RNA interference (RNAi) target genes were selected from the iBeetle database and a transcriptome data set for P. solenopsis. To assess the effectiveness of the selected target genes, three methods were utilized to deliver double-stranded (ds)RNA (ingestion, artificial diet bioassay and transient gene silencing). dsRNA molecules at different concentrations were fed to insects and insect mortality was recorded for each target gene. Based on the mortality data, three genes, Krüppel homologue-1, ADP-ATP/Translocase and IDGF-1, were selected for further gene expression studies using a reduced concentration of dsRNA (5 μg/ml). Of the three genes, Krüppel homologue-1 showed significantly downregulated expression (by 70.81% and 84.33%) at two different time points (8 and 14 days). An RNAi silencing construct was designed for Krüppel homologue-1 under control of the double enhancer CamV35S promoter in the plant binary vector. Significant downregulation of gene expression, by 66.69% and 81.80%, was found for Krüppel homologue-1 using transient gene silencing at the same time intervals. Conclusion: This work provides the first evidence for targeting the Krüppel homologue-1 gene in a hemipteran pest, P. solenopsis, using RNAi technology through oral delivery and in planta-based transient gene silencing methods. © 2020 Society of Chemical Industry.

Abstract:The problem of team control is of recent interest in intelligent systems and robotics due to the broad range of applications of multi-agent systems in space missions, operations in hazardous environments, and military missions. Such systems can exhibit flexibility, reliability through redundancy, and simple hardware. This paper considers control problems in unicycle-like mobile robots with restricted inputs. The concept of flatness will be applied for generating reference trajectories, and tracking controllers will be established for the considered system with bounds on the input signals. The rigid formation control problem will then be studied and solved based on the established tracking controllers. The proposed controllers which account for the system physical constraints ensure asymptotic convergence of a group of N agents towards a desired rigid formation.

Abstract:Spray-induced gene silencing represents an eco-friendly approach for crop protection through the use of double-stranded RNA (dsRNA) to activate the RNA interference (RNAi) pathway, thereby silencing crucial genes in pathogens. The major challenges associated with dsRNA are its limited stability and poor cellular uptake, necessitating repeated applications for effective crop protection. In this study, RNA nanoparticles (NPs) were proposed as effectors in plants and pathogens by inducing the RNAi pathway and silencing gene expression. RNA structural motifs, such as hairpin-loop, kissing-loop, and tetra-U motifs, were used to link multiple siRNAs into a long, single-stranded RNA (lssRNA). The lssRNA, synthesized in Escherichia coli, self-assembled into stable RNA nanostructures via local base pairing. Comparative analyses between dsRNA and RNA NPs revealed that the latter displayed superior efficacy in inhibiting spore germination and mycelial growth of Botrytis cinerea. Moreover, RNA NPs had a more robust protective effect on plants against B. cinerea than did dsRNA. In addition, RNA squares are processed into expected siRNA in plants, thereby inhibiting the expression of the target gene. These findings suggest the potential of RNA NPs for use in plant disease control by providing a more efficient and specific alternative to dsRNA without requiring nanocarriers.

Abstract:Over the last decade, several studies have revealed the enormous potential of RNA-silencing strategies as a potential alternative to conventional pesticides for plant protection. We have previously shown that targeted gene silencing mediated by an in planta expression of non-coding inhibitory double-stranded RNAs (dsRNAs) can protect host plants against various diseases with unprecedented efficiency. In addition to the generation of RNA-silencing (RNAi) signals in planta, plants can be protected from pathogens, and pests by spray-applied RNA-based biopesticides. Despite the striking efficiency of RNA-silencing-based technologies holds for agriculture, the molecular mechanisms underlying spray-induced gene silencing (SIGS) strategies are virtually unresolved, a requirement for successful future application in the field. Based on our previous work, we predict that the molecular mechanism of SIGS is controlled by the fungal-silencing machinery. In this study, we used SIGS to compare the silencing efficiencies of computationally-designed vs. manually-designed dsRNA constructs targeting ARGONAUTE and DICER genes of Fusarium graminearum (Fg). We found that targeting key components of the fungal RNAi machinery via SIGS could protect barley leaves from Fg infection and that the manual design of dsRNAs resulted in higher gene-silencing efficiencies than the tool-based design. Moreover, our results indicate the possibility of cross-kingdom RNA silencing in the Fg-barley interaction, a phenomenon in which sRNAs operate as effector molecules to induce gene silencing between species from different kingdoms, such as a plant host and their interacting pathogens.

Abstract:With the growing global demands on sustainable food production, one of the biggest challenges to agriculture is associated with crop losses due to parasitic nematodes. While chemical pesticides have been quite successful in crop protection and mitigation of damage from parasites, their potential harm to humans and environment, as well as the emergence of nematode resistance, have necessitated the development of viable alternatives to chemical pesticides. One of the most promising and targeted approaches to biocontrol of parasitic nematodes in crops is that of RNA interference (RNAi). In this study we explore the possibility of using biostimulants obtained from metabolites of soil streptomycetes to protect wheat (Triticum aestivum L.) against the cereal cyst nematode Heterodera avenae by means of inducing RNAi in wheat plants. Theoretical models of uptake of organic compounds by plants, and within-plant RNAi dynamics, have provided us with useful insights regarding the choice of routes for delivery of RNAi-inducing biostimulants into plants. We then conducted in planta experiments with several streptomycete-derived biostimulants, which have demonstrated the efficiency of these biostimulants at improving plant growth and development, as well as in providing resistance against the cereal cyst nematode. Using dot blot hybridization we demonstrate that biostimulants trigger a significant increase of the production in plant cells of si/miRNA complementary with plant and nematode mRNA. Wheat germ cell-free experiments show that these si/miRNAs are indeed very effective at silencing the translation of nematode mRNA having complementary sequences, thus reducing the level of nematode infestation and improving plant resistance to nematodes. Thus, we conclude that natural biostimulants produced from metabolites of soil streptomycetes provide an effective tool for biocontrol of wheat nematode.
