Abstract:Cleidocranial dysplasia (CCD) is an autosomal-dominant disorder caused by a lack of function of one or more alleles of the RUNX2 gene. Mutations of the RUNX2 gene were analyzed in a family with CCD, and a novel nonsense mutation was identified, c. 1096G >T, p.E366X, which was predicted to cause a number of potential dysfunctions. Western blot analysis showed that the novel mutation created a shortened protein product, which lost 155 aa in the C-terminal domain. The mutant protein was detected to be localized mostly in the cytoplasm, not in the nucleus, which demonstrated that transport of the RUNX2 protein into the nucleus was disturbed by the p.E366X mutation. For the first time, RUNX2(+/m) dental pulp cells (DPCs) were isolated from two permanent incisors of the CCD patient. Compared to RUNX2(+/+) controls, RUNX2(+/m) DPCs presented an impeded progression from the G1 to the S phase in the cell cycle, a lower rate of proliferation, weaker ability of calcification, and distinct ultrastructure. More interestingly, the ultrastructural analysis and energy dispersive X-ray spectrometry (EDS) analysis showed that the CCD tooth exhibited insufficient mineralization of enamel and dentin. This study suggests that the truncated RUNX2 mutant protein may be responsible for the alterations of RUNX2(+/m) DPCs, and RUNX2 gene may be involved in dental development by affecting the cell growth and differentiation, which provides new insights into understanding of dental abnormalities in CCD patients. Abstract:To investigate whether crosstalk between RUNX2 and miRNAs is involved in tooth eruption regulated by dental follicle cells (DFCs) and the possible molecular mechanism. Blood samples and embedded dental follicles were collected from patients with cleidocranial dysplasia, and RUNX2 gene mutations were analyzed, then RUNX2(+/m) DFCs were isolated and identified. The characteristics of RUNX2(+/m) DFCs were analyzed. The differential expression of miRNAs was detected between the RUNX2(+/m) DFCs and RUNX2(+/+) DFCs by microarray, and target genes were predicted by miRGen. miR-146a was chosen for further investigation, and its effects in DFCs were analyzed by transfecting its mimics and inhibitors, and expression of genes involved in tooth eruption were detected. A novel insertion mutation (c.309_310insTG) of RUNX2 gene was identified which had an effect on the characteristics of DFCs. Compared with the RUNX2(+/+) DFCs, there were 69 microRNAs more than twofold up-regulated and 54 microRNAs more than twofold down-regulated in the RUNX2(+/m) DFCs. Among these, miR-146a decreased significantly in RUNX2(+/m) DFCs, and expression of RUNX2, CSF-1, EGFR, and OPG was significantly altered when miR-146a was overexpressed or inhibited. RUNX2 gene mutation contributes to the characteristic change of DFCs, and the crosstalk between RUNX2 gene and miRNAs may be one of the key regulatory mechanisms of differentiation of DFCs. J. Cell. Abstract:Objective:To obtain human dental follicle cells with RUNX2 gene mutation.Methods:Patients with cleidocranial dysplasia were collected,and RUNX2 gene mutation was identified.The dental follicle cells were cultured from unerupted dental follicle in vitro,and the same mutation site was identified.Results:The novel insertion mutation was identified as c.309_310insTG in exon 2 of RUNX2 gene by genome detection with whole blood,and the dental follicle cells with the same mutation were successfully cultured and identified.The expression of RUNX2 was decreased in RUNX2+/m dental follicle cells.Conclusions:Human dental follicle cells with RUNX2 mutation were successfully cultured and identified,and it contributes to the further function study of RUNX2 gene during tooth eruption.
A Novel RUNX2 Mutation in Cleidocranial Dysplasia Patients.

Abstract:Cleidocranial dysplasia (CCD) is an autosomal-dominant heritable skeletal disease caused by heterozygous mutations in the RUNX2 gene. Here, the RUNX2 gene was analyzed within a CCD family from China, and a novel missense mutation (c. 475G --> C [p.G159R]) was identified. Normal and mutant RUNX2 expression vectors were then constructed and expressed transiently in NIH3T3 cells. Immunofluorescent staining and Western blotting showed that wild-type RUNX2 protein was localized exclusively in the nucleus; however, the mutant protein was found in both the nucleus and the cytoplasm, which demonstrated that transport of the RUNX2 mutant into the nucleus was disturbed by the G159R mutation. Therefore, we suggest that G159 is very important to promote RUNX2 nuclear localization. According to clinical analysis, the patient displays severe dysplasia of bones and relatively low-grade craniofacial abnormality, and we infer that G159 may be vital for normal skeletal development, other than control of tooth number. These findings confirm that mutations in the RUNX2 gene are associated with the pathogenesis of CCD across different ethnic backgrounds.
RUNX2 mutation reduces osteogenic differentiation of dental follicle cells in cleidocranial dysplasia

Abstract:Disturbed permanent tooth eruption is common in cleidocranial dysplasia (CCD), a skeletal disorder caused by heterozygous mutation of RUNX2, but the mechanism underlying is still unclear. As it is well known that dental follicle cells (DFCs) play a critical role in tooth eruption, the changed biological characteristics of DFCs might give rise to disturbance of permanent tooth eruption in CCD patients. Thus, primary DFCs from one CCD patient and normal controls were collected to investigate the effect of RUNX2 mutation on the bone remodeling activity of DFCs and explore the mechanism of impaired permanent tooth eruption in this disease. Conservation and secondary structure analysis revealed that the RUNX2 mutation (c.514delT, p.172fs) found in the present CCD patient was located in the highly conserved RUNT domain and converted the structure of RUNX2. After osteogenic induction, we found that the mineralised capacity of DFCs and the expression of osteoblast-related genes, including RUNX2, ALP, OSX, OCN and Col Iα1, in DFCs was severely interfered by the RUNX2 mutation found in CCD patients. To investigate whether the osteogenic deficiency of DFCs from the CCD patient can be rescued by RUNX2 restoration, we performed 'rescue' experiments. Surprisingly, the osteogenic deficiency and the abnormal expression of osteoblast-associated genes in DFCs from the CCD patient were almost rescued by overexpression of wild-type RUNX2 using lentivirus. All these findings indicate that RUNX2 mutation can reduce the osteogenic capacity of DFCs through inhibiting osteoblast-associated genes, thereby disturbing alveolar bone formation, which serves as a motive force for tooth eruption. This effect may provide valuable explanations and implications for the mechanism of delayed permanent tooth eruption in CCD patients.

Abstract:Cleidocranial dysplasia (CCD) is an autosomal dominant inheritable skeletal disease caused by heterozygous mutations in an osteoblast-specific transcription factor, RUNX2. Mutational analyses of RUNX2 were done on 4 unrelated Chinese patients with CCD. One nonsense and 3 missense mutations were detected, including one novel mutation, a heterozygous G to C transition mutation at nucleotide 475 in exon 2, which converts glycine to arginine at codon 159 (G159R). Two mutations, R225W and R391X, were reported in Chinese patients with CCD for the first time. Our findings show that R225 mutations interfere with nuclear accumulation of RUNX2 protein, and that a lack of nuclear RUNX2 protein accumulation is at least one of the causes of haploinsufficiency in these cases. Body stature was significantly reduced in the 3 male and 1 female cases. The cases all had malformations of the tarsometatarsal joints. In 1 case, the humeroulnar joints and humeroradial joints were abnormal, and the elbow looked like a triangle. The data suggest that an impaired runt domain contributes to the short stature of CCD patients. We postulate that RUNX2 influences joint formation by affecting the differentiation pathways of chondrocytes and osteoblasts.

Abstract:Objective:To study the RUNX2 gene mutation in a patient with cleidocranial dysplasia(CCD).Method: Examination was carried out over the oral cavity and the entire body,and the patient was diagnosed as CCD.Genomic DNA was extracted from whole blood,and the RUNX2 gene was amplified by PCR from genomic DNA.Mutations were analyzed using BLAST nucleotide program.Besides,dental follicle cells from both CCD patient and control group were cultured.cDNA sequence was analyzed to confirm the mutation.Result:We found a novel insertion mutation of TG in the exon 2.And the same mutation was found in dental follicle cells cDNA too(c.309_310insTG).Conclusion:We successfully found a novel mutation of RUNX2,which supplemented the gene mutation database of CCD.

Abstract:OBJECTIVES Kleptomania, a disabling impulse control disorder, is characterized by the repetitive and uncontrollable theft of items that are of little use to the afflicted person. Despite its relatively long history, kleptomania remains poorly understood to the general public, clinicians, and sufferers. METHOD This article reviews the literature for what is known about the clinical characteristics, family history, neurobiology, and treatment options for individuals with kleptomania. RESULTS Kleptomania generally has its onset in late adolescence or early adulthood and appears to be more common among women. Lifetime psychiatric comorbidity is frequent, mainly with other impulse control (20-46%), substance use (23-50%) and mood disorders (45-100%). Individuals with kleptomania suffer significant impairment in their ability to function socially and occupationally. Kleptomania may respond to cognitive behavioral therapy and various pharmacotherapies (lithium, anti-epileptics, and opioid antagonists). CONCLUSIONS Kleptomania is a disabling disorder that results in intense shame, as well as legal, social, family, and occupational problems. Large scale treatment studies are needed.

Abstract:OBJECTIVE:To analyze the mutations of the RUNX2 gene in a family with cleidocranial dysplasia (CCD).METHODS:The general health status of family members with CCD was investigated through propositus verification method. Oral specialized examination and radiological examination were performed. The peripheral venous blood of the proband and her parents and sisters was collected. Genomic DNA was extracted, and the RUNX2 gene from this DNA was amplified by polymerase chain reaction (PCR). DNA sequences were analyzed with the Blastn program.RESULTS:After Blastn analysis, heterozygous C to T transition mutation at nucleotide 568 occurred in exon 2, which converted arginine to tryptophane at codon 190 (c.568C>T, CGG-->TGG).CONCLUSION:RUNX2 gene is responsible for the CCD in the Chinese family under study. The c.568C>T mutation is the molecular basis of the CCD in the family.

Abstract:As a nonlinear system, a recurrent neural network generally has an incremental gain different from its induced norm. While most of the previous research efforts were focused on the latter, this paper presents a method to compute an effective upper bound of the former for a class of discrete-time recurrent neural networks, which is not only applied to systems with arbitrary inputs but also extended to systems with small-norm inputs. The upper bound is computed by simple optimizations subject to linear matrix inequalities.

Abstract:Cleidocranial dysplasia (CCD), a skeletal disorder characterized by delayed permanent tooth eruption and other dental abnormalities, is caused by heterozygous RUNX2 mutations. As an osteoblast-specific transcription factor, RUNX2 plays a role in bone remodeling, tooth formation and tooth eruption. To investigate the crosstalk between RUNX2 and 1α,25-dihydroxyvitamin D3 (1α,25-(OH) 2 D 3 ) in human dental follicle cells (hDFCs) during osteoclast formation, we established a co-culture system of hDFCs from CCD patient and healthy donors with peripheral blood mononuclear cells (PBMCs). Expression of the osteoclast-associated genes and the number of TRAP + cells were reduced in CCD hDFCs, indicating its suppressed osteoclast-inductive ability, which was reflected by the downregulated RANKL/OPG ratio. In addition, 1α,25-(OH) 2 D 3 -stimulation elevated the expression of osteoclast-related genes, as well as RANKL mRNA levels and RANKL/OPG ratios in control hDFCs. Conversely, RUNX2 mutation abolished this 1α,25-(OH) 2 D 3 -induced RANKL gene activation and osteoclast formation in CCD hDFCs. Therefore, RUNX2 haploinsufficiency impairs dental follicle-induced osteoclast formation capacity through RANKL/OPG signaling, which may be partially responsible for delayed permanent tooth eruption in CCD patients. Furthermore, this abnormality was not rescued by 1α,25-(OH) 2 D 3 application because 1α,25-(OH) 2 D 3 -induced RANKL activation in hDFCs is mediated principally via the RUNX2 -dependent pathway.

Effect of the RUNX2 Gene Mutations on Subnuclear Localization of the RUNX2 Protein Mutants

Abstract:Objective To observe the effect of the RUNX2 gene mutations on subnuclear localization of the RUNX2 mutant protein by transfection.Methods Total RNA was extracted and reverse-transcribed from blood of a patient with cleidocranial dysplasia,and the eukaryotic expression vector pEGFP-C1-RUNX2 was constructed by subcloning the objective fragments.The subnuclear localization of RUNX2 wild and mutant protein was observed by transfected into NIH3T3 fibroblast cells.Results The vector pEGFP-C1-RUNX2 was constructed and identified successfully.The results of transfection showed that both c.674 G>A and c.673 C>T mutation sites can contribute to partial loss of nuclear localization regardless of presence of intact nuclear localization signal,causing dual localization to both the cytoplasm and nucleus,and the wild RUNX2 protein was observed in nuclear exclusively.Conclusion Different mutation sites of RUNX2 gene can contribute to change of subnuclear localization of RUNX2 protein,and the deficiency in nuclear might be responsible for the dysfunction of RUNX2 protein in pathology of cleidocranial dysplasia.
A Study on the Ultrastructure of Hypocalcification in Patient with Cleidocranial Dysostosis

Abstract:Objective:To observe the ultrastructure of the CCD teeth caused by mutation of runx2 gene.Methods:Deciduous teeth and mandibular permanent teeth from CCD patients were collected.Ground sections were made.The ultrastructure changes of CCD teeth were observed by scanning electron microscope and the component content of Ca and P in enamel and dentin were analyzed by energy-dispersive spectrometer(EDS).Results:Compared with normal teeth,CCD teeth had more enamel lamellae and tufts,straighter EDJ,irregular distribution of dentinal tubule and thinner cementum.The contents of Ca and P declined.Conclusion:The mutation of runx2 gene has influence on the structure formation of CCD teeth and their element composition,which demonstrates the importance of runx2 gene in the normal development of the teeth.
Effect of RUNX2 mutation on tranforming grouth factor-β relevant signal passway in dental pulp cells

Abstract:Objective To investigate different expression profile of TGF-β/BMP signaling pathway in dental pulp cells with mutation of RUNX2 gene by PCR array.Methods The dental pulp cells with mutation of RUNX2 gene had been successfully separated and identified in the patients with cleidocranial dysplasia,which was compared with the corresponding normal dental pulp cells,and the different gene profile was detected by PCR array about TGF-β / BMP signaling pathway,then data was analyzed.Results The data analysis of PCR array showed that there were 18 genes which were up-regulated in dental pulp cells with mutation of RUNX2 gene compared with normal ones,and 14 genes were down-regulated.Conclusion The characteristics of dental pulp cells was affected by the related genes about TGF-β / BMP signaling pathway regulated by RUNX2 gene.

Abstract:Journal of Dental Research, Ahead of Print. RUNX2 is a master osteogenic transcription factor, and mutations in RUNX2 cause the inherited skeletal disorder cleidocranial dysplasia (CCD). Studies have revealed that RUNX2 is not only a downstream target of the bone morphogenetic protein (BMP) pathway but can also regulate the expression of BMPs. However, the underlying mechanism of the regulation of BMPs by RUNX2 remains unknown. In this project, we diagnosed a CCD patient with a 7.86-Mb heterozygous deletion on chromosome 6 containing all exons of RUNX2 by multiplex ligation-dependent probe amplification (MLPA) and whole-genome sequencing (WGS). Bone marrow mesenchymal stem cells (BMSCs) were further extracted from patient alveolar bone fragments (CCD-BMSCs), an excellent natural model to explore the possible mechanism. The osteogenic differentiation ability of CCD-BMSCs was severely affected by RUNX2 heterozygous deletion. Also, BMP4 decreased most in BMP ligands, and CHRDL1, a BMP antagonist, was abnormally elevated in CCD-BMSCs. Furthermore, BMP4 treatment essentially rescued the osteogenic capacity of CCD-BMSCs, and RUNX2 overexpression reversed the abnormal expression of BMP4 and CHRDL1. Notably, we constructed CRISPR/Cas9 Runx2+/m MC3T3-E1 cells, which simulated a variant in CCD-BMSCs, to exclude the interference of other gene deletions and the heterogeneity of the genetic background of primary cells, and verified all findings from the CCD-BMSCs. Moreover, the luciferase reporter experiment showed that RUNX2 could inhibit the transcription of CHRDL1. Through immunofluorescence, the inhibitory effect of CHRDL1 on BMP4/Smad signaling was confirmed in MC3T3-E1 cells. These results revealed that RUNX2 regulated the BMP4 pathway by inhibiting CHRDL1 transcription. Abstract:OBJECTIVE To use pedigree analysis to evaluate the feasibility of a major locus model for deafness in Dalmatians. ANIMALS 605 purebred Dalmatians from 42 families. PROCEDURE Hearing loss was evaluated through the brainstem auditory-evoked response. Dogs were classified into mutually exclusive categories: normal hearing, unilaterally deaf, or bilaterally deaf. Information was collected on sex, coat color, presence or absence of a color patch at birth, and eye color. Statistical analyses were performed by use of regressive logistic models designed for complex segregation analysis. Genetic correlations among eye color, deafness, and color patch were estimated. RESULTS Prevalence of hearing loss was 11% for dogs classified as unilaterally deaf and 5% for dogs that were bilaterally deaf. Complex segregation analysis detected statistical evidence of a single allele with an expected frequency of 0.21 that had an effect on the prevalence of deafness. Results of analyses suggested that this locus cannot completely explain the inheritance and incidence of deafness in Dalmatians. Genetic correlation estimates among deafness, eye color, and color patch revealed strong interrelationships among these characteristics. CONCLUSIONS AND CLINICAL RELEVANCE To reduce the incidence of hearing loss in Dalmatians, unilaterally deaf, blue-eyed dogs should not be considered as potential parents.
