Abstract:Fungi Penicillium oxalicum SL2 was applied for Pb2+ bioremediation in aqueous solution in this study. After 7 days of incubation at different initial concentrations of Pb2+ (0, 100, 500 and 2500 mg L-1), most of Pb2+ were removed (90, 98.3, and 86.2%), the maximum Pb content in mycelium reached about 155.6 mg g(-1) dw. Meanwhile, the formation of extracellular secondary minerals and intracellular Pb-complex were observed and identified, the speciation of Pb in mycelium was also detected by X-ray absorption near-edge structure (XANES) spectroscopy, i.e., Pb-oxalate, Pb-citrate, Pb-hydrogen phosphate and Pb-glutathione analogues. In addition, content of glutathione and oxidized glutathione was increased under the exposure of Pb2+, which implied that glutathione might play a key role in Pb immobilization and detoxification in P. oxalicum SL2. This study elucidated partial mechanisms of Pb immobilization and speciation transformation of this strain, providing an alternative biomaterial in the bioremediation of Pb-contaminated wastewater.
Dual Regulatory Role of Penicillium Oxalicum SL2 in Soil: Phosphorus Solubilization and Pb Stabilization.

Abstract:The mechanisms of the P. oxalicum SL2-mediated microbial community on phosphorus solubilization and Pb stabilization were investigated through a 90-day soil experiment. In the treatments inoculated with P. oxalicum SL2, the amount of P. oxalicum SL2-GFP remained at 77.8%-138.6% of the initial inoculation amount after 90 days, and the available phosphorus (AP) content increased 21.7%-40.8% while EDTA-Pb decreased 29.9%-43.2% compared with CK treatment. SEM-EDS results showed that P. oxalicum SL2 changed the agglomeration degree of microaggregates and promoted the combination of Pb with C and O elements. These phenomena were enhanced when applied with Ca3(PO4)2. Microbial community analysis showed that P. oxalicum SL2 improved soil microbial activity, in which the fungi absolute abundance increased about 15 times within 90 days. Correlation analyses and a partial least-squares path model showed that the activation of Penicillium, Ascobolus, Humicola, and Spizellomyces in a fungal community increased the content of oxalate and AP, which directly decreased EDTA-Pb content, while the change of Bacillus, Ramlibacter, Gemmatimonas, and Candidatus Solibacter in the bacterial community regulated Fe/Mn/S/N cycle-related functions, thus promoting the conversion of Pb to oxidizable state. Our findings highlight that P. oxalicum SL2 enhanced the microbial-induced phosphate precipitation process by activating soil microbial communities and regulating their ecological functions.
Colonization of Penicillium Oxalicum SL2 in Pb-contaminated Paddy Soil and Its Immobilization Effect on Soil Pb.

Abstract:Penicillium oxalicum SL2 (SL2) is a previously screened Pb-tolerant fungus that can promote crops growth. The relationship between SL2 colonization and Pb immobilization was studied to provide a theoretical basis for microbial remediation of Pb-contaminated paddy soil. In this study, green fluorescent protein (GFP) labeled SL2 was inoculated into different Pb-contaminated paddy soils (S1-S6). The Pb extracted from the soil by HNO3, EDTA and CaCl2 were used to characterize the available Pb. The results showed that the colonization of SL2 was divided into lag phase (0-7 days), growth phase (7-30 days), and mortality phase (30-90 days). SL2 colonized well in sandy soils rich in clay and total phosphorus with initial pH of 4.5-7.0. In addition, SL2 increased soil pH and decreased soil Eh, which was beneficial to immobilize Pb. In different soils, the highest percentages of CaCl2-Pb, EDTA-Pb, and HNO3-Pb immobilized by SL2 were 34.34%-40.53%, 17.05%-20.11%, and 7.39%-15.62%, respectively. Pearson correlation analysis showed that the percentages of CaCl2-Pb and EDTA-Pb immobilized by SL2 were significantly positively correlated with the number of SL2 during the growth phase. SL2 mainly immobilized Pb in the growth phase and a higher peak number of SL2 was beneficial to the immobilization of Pb.
Biochemical Mechanism Underlying the Synthesis of Pbs Nanoparticle and Its In-Situ Photo Effect on Shinellazoogloeoides Pq7

Abstract:Metal sulfide nanoparticles (NPs) with semiconductor potentials are valuable bioremediation end-products that attract great research interests. However, biochemical mechanisms underlying their biosynthesis and photo-effects remain elusive. In this study, we found that biofilm lifestyle remarkably improved lead resistance and PbS-NP biosynthesis in Shinella zoogloeoides PQ7. Surprisingly, biosynthesis of PbS-NP required more than cysteine and H2S production. Transcriptomic and metabolomic analysis indicated that PQ7 responded to lead stress by changing metabolic activities in ABC transporters, oxidative phosphorylation, EPS production, quorum sensing, protein de novo synthesis, flagella assembly and antioxidative reactions, etc. The elevated EPS production and quorum sensing gene expression echoed the favorable roles of biofilm formation in lead resistance. Biosynthesis of PbS-NP required proper oxygen supply, and was impeded by adding kanamycin or using yeast extract as the sole nutrient supply. Investigations on NAD/NADH, ATP, ROS and GSH productions indicated that biosynthesis of PbS-NP was corelated with cellular respiration, energy metabolism, and redox status. Finally, we proved that PbS-NP had the dose-dependent in-situ photo effect on PQ7’s growth and ROS production. This is the first report that pinpoints the role of cellular respiration in PbS-NP biosynthesis, which is essential for further mechanism study and the development of bioremediation techniques.
Proteomics Study on Immobilization of Pb(II) by Penicillium Polonicum.

Abstract:Lead (Pb) is widely distributed in nature and has important industrial applications, while being highly toxic. In this study, the Pb(II) biosorption and immobilization behavior of Penicillium polonicum was investigated through surface morphology observation and multiple experimental analysis. In addition, the molecular mechanism of Pb(II) immobilization was further explored through proteomics. The analysis of the removal ability of P. polonicum to Pb(II) has found that P. polonicum could remove Pb(II) up to 95% (initial 4 mM Pb(II)) in 12 d. Scanning Electron Microscope (SEM) revealed a large amount of Pb(II) adsorbed on the cell wall. Raman and Energy Disperse Spectroscopy (EDS) revealed the formation of large amounts of PbC2O4 minerals extracellularly. Field Emission Higheresolution Transmission Electron Microscopy (FEeTEM) found that [Pb-5(PO4)(3)Cl] formed on the cell surface and inside the cells. The iTRAQ technique was used to analyze the characteristics of the changes of proteins during the action between Pb(II) and P. polonicum, which further revealed the mechanism of P. polonicum against Pb(II) and biomineralization. It was found that differential proteins in terms of redox, ion binding, metabolic process and ribosome synthesis were predominant in the GO analysis. Together with some of the characterization experiments above, the mechanisms mentioned above was well explained. The up-regulated expression of related proteins involved in respiratory metabolic pathways, antioxidant stress, and degradation of intracellular hazardous substances in the P. polonicum intracellularly such as succinate dehydrogenase, ATPase and cytochrome c oxidase, could explain the high tolerance of P. polonicum to Pb(II). The up regulation of OAH was responsible for extracellular PbC2O4 production. The up regulation of proteins such as TXN and GFA promoted Pb-glutathione (PbeGSH) complex formation. This study explores the mechanism of Pb removal by fungi from the proteomic level, and provides new ideas and ways for Pb biogeochemical research. (c) 2022 British Mycological Society. The Synthesis of PbS NPs and Biosorption of Pb(II) by Shinella Zoogloeoides PQ7 in Aqueous Conditions

Abstract:Increasing heavy metal pollution in water continues to endanger human health. The genus Shinella has potential for heavy metal bioremediation but has rarely been studied. In this study, we report that Shinella zoogloeoides PQ7 turns black in the presence of lead ions. Transmission electron microscopy (TEM), Scanning electron microscopy–energy dispersive X-ray spectroscopy (SEM–EDS), X-ray photoelectron spectroscopy (XPS) and X-ray diffraction (XRD) indicated that PbS nanoparticles (NPs) were synthesized by PQ7. Moreover, PQ7 was used as a biosorbent to remove Pb(II) from aqueous solutions. Biosorption performance was evaluated in terms of contact time, pH, biomass dosage and initial Pb(II) concentration. The equilibrium and kinetic data were consistent with the Freundlich isotherm model (R2 = 0.986) and pseudo-second-order model (R2 = 0.977), respectively. The maximum (qmax) Pb(II) adsorption reached 222.22 mg/g, which was higher than that of other bacteria reported in previous literature. SEM–EDS, XRD and Fourier transform infrared (FTIR) analyses also confirmed the adsorption of Pb(II) by the PQ7 cells. In conclusion, PQ7 is a promising strain in removing and recovering Pb(II) from wastewater.
The colonization of Penicillium oxalicum SL2 on rice root surface increased Pb interception capacity of iron plaque and decreased Pb uptake by roots

Abstract:The exploration of microbial resources to reduce Pb accumulation in rice attracted great attention. In this study, we found Penicillium oxalicum SL2, a Pb-tolerant strain with good capability of dissolving phosphorus and stabilizing Pb in soil, was able to colonize on the root surface of rice seedlings without additional carbon sources, and promoted the secretion of metabolites related to amino acid metabolism, organic acid metabolism, signal transduction and other pathways in rhizosphere exudates, in which the secretion of oxalate increased by 47.7 %. However, P. oxalicum SL2 increased Fe(II) proportion and Fe availability on the root surface, resulting in iron plaque content decrease. Moreover, by converting root surface Pb from Pb-Fe state to PbC 2 O 4 and Pb-P compounds, P. oxalicum SL2 increased Pb intercept capacity of iron plaque by 118.0 %. Furthermore, P. oxalicum SL2 regulated element distribution on the root surface, and reduced the relative content of Pb on the maturation zone of root tip, which was conducive to reducing Pb uptake by apoplastic pathway and the risk of Pb accumulation in root system. Our findings further revealed the interaction between P. oxalicum SL2 and rice root, providing a theoretical basis for the development and application of microbial agents in Pb-contaminated farmland.

Abstract:Heavy metal contamination in agricultural soil and its associated risk of food safety are of great concern globally. It is therefore an urgent need to develop sustainable option to mitigate the accumulation of metals in crop plants. Here we investigated the potential of phosphorus-solubilizing fungus, Penicillium oxalicum SL2, on regulating the bioavailability of Pb in a lead (Pb) polluted soil-rice system. Our results showed that the content of Pb in rice grain was significantly decreased by ~80% with the application of P. oxalicum SL2. The competition between oxalate and phosphate for the complexation of Pb showed to be effective in mediating the bioavailability of Pb, and such impact varied with water fluctuation in paddy soil. The solubilization of phosphorus also played an important role in alleviating the dissolution of iron plaque caused by oxalic acid, which helped maintaining the biomass of iron plaque as a barrier to the uptake of Pb by root. The predominant indigenous microbial community was not affected by the inoculation with P. oxalicum SL2, suggesting it as an eco-friendly strain. Therefore, we suggest P. oxalicum SL2 as a promising fungus in enhancing the safe use of moderately Pb polluted paddy soil for safe rice.
Oxalic Acid Enhances Bioremediation of Cr(VI) Contaminated Soil Using Penicillium Oxalicum SL2.

Abstract:Oxalic acid is the most abundant low molecular weight organic acid (LMWOA) in many environments and offers enormous prospects for treating Cr(VI) contamination. In this study, laboratory batch experiments were conducted to estimate the roles of oxalic acid in Cr(VI) removal by Penicillium oxalicum SL2. Oxalic acid changed the initial pH and provided a suitable condition for the growth of strain SL2 when the penicillium was applied to bioremediation of Cr(VI) contamination in alkaline soil. Gompertz model analysis indicated that initial pH affected the lag time of the growth curve of strain SL2. Scanning electron microscopy and scanning transmission X-ray microscopy analysis showed strain SL2 sufficiently contacted with contaminated soil and reduced Cr(VI) to Cr(III) in the hyphae. The results suggested that oxalic acid could enhance the bioremediation efficiency of strain SL2 though improving chromium bioleaching from the contaminated soil and strengthening Cr(VI) removal in the leaching solution. This study provided oxalic acid as a green reagent for stimulating Cr(VI) removal by strain SL2 and would expand knowledge on the roles of LMWOA in Cr(VI) bioremediation.

Abstract:Phosphate-solubilizing bacteria (PSB) promotes the formation of mineralized precipitation through phosphorous dissolution and mineralization, forming stable lead (Pb(II)) minerals and reducing the migration of Pb(II) in the environment. In this study, a Pb-tolerant strain Ochrobactrum sp. J023 from a contaminated soil around a battery factory in Jiangsu Province, China, was screened for experiments to investigate the phosphate solubilization and mineralization mechanism of this strain. The organic acids and the acid phosphatase produced by the bacteria have a synergistic effect on phosphate dissolution. When the pH of the culture medium decreased to the lowest 4.55, the amount of soluble phosphate and the activity of acid phosphatase reached the maximum 161.29 mg L−1 and 61.98 U mL−1, and there was a significant correlation between the concentration of soluble phosphate and the activity of acid phosphatase (R = 0.832**, P < 0.05). It was found that acetic acid played the most important role in the secreted organic acids. During the mineralization reaction, the extracellular polymeric substances (EPS) chelates part of the Pb(II) on the surface of the cell wall, preventing the metal Pb from penetrating into the cell, thus providing protection to the strain. Meanwhile, due to the nucleation sites provided by cell surface groups (carboxyl and phosphate groups), a large number of metal ions are absorbed to promote the formation of crystallization. The final mineralized product of Pb(II) by strain J023 was pyroxite (Pb5(PO4)3X, where X = Cl, OH). The mechanism of phosphate dissolution and mineralization proposed by us is that the organic acids and acid phosphatases secreted by phosphate-solubilizing bacteria promote the increase of PO43− concentration in the solution, the complexation of metal cations and cell surface groups will induce the formation of mineralized precipitation under the catalysis of enzyme. Therefore, it is a promising strategy for bioremediation of lead pollution by screening functional strains with strong abilities of phosphate solubility and mineralization.

Penicillium oxalicum augments soil lead immobilization by affecting indigenous microbial community structure and inorganic phosphate solubilization potential during microbial-induced phosphate precipitation

Abstract:Phosphate-solubilizing microorganisms (PSMs) are critically important for increasing soil phosphate (P) and decreasing lead (Pb) bioavailability during microbial-induced phosphate precipitation (MIPP). However, their relative contributions to the indigenous soil microbial communities and P-cycling genes during the MIPP process remain unclear. In this study, inoculation of the PSM P. oxalicum in hydroxyapatite-cultured and Pbcontaminated soil increased soil phosphatase activities, available P (AP) concentrations and reduced available Pb levels. Metagenomics revealed a 3.9-44.0% increase in the abundance of P-cycling genes by P. oxalicum inoculation. No P-cycling genes were assigned to Penicillium. While P. oxalicum increased the complexity of microbial community co-occurrence networks, and improved the directly interrelationships between Penicillium and genera containing P-cycling gene. These results suggesting that P. oxalicum obviously positively affected the regulation of indigenous P-cycling functional communities during the MIPP process. Inorganic P solubilization genes (gcd, ppa, and ppx) have been shown to affect soil AP, suggesting that inorganic P solubilization is the major driver of Pb immobilization improvement following P. oxalicum inoculation. These results enhance our understanding of the significant ecological role of PSMs in governing soil P-cycling and alleviating Pb2+ biotoxicity during the MIPP process.
Enhanced Lead (Pb) immobilization in red soil by phosphate solubilizing fungi associated with tricalcium phosphate influencing microbial community composition and Pb translocation in Lactuca sativa L.

Abstract:Phosphate (P) minerals and phosphate solubilizing fungi (PSF) play essential roles in lead (Pb) immobilization, but their roles in driving Pb bioavailability and ecological risks in red soil remains poorly understood. In this study, the inoculation of P. oxalicum and TCP successfully enhanced available P (AP) and urease concentrations in artificially Pb contaminated red soil. Combined P. oxalicum and TCP inoculation significantly reduced Pb bioavailability, bioaccessibility, leachability and mobility by increasing soil AP concentration and forming stable Pb-P compounds during the 21-day experiment. Soil AP and Pb bioavailability play an important role in shifting soil microbial communities induced by co-occurrence of P. oxalicum and TCP. Combined P. oxalicum and TCP could notably promote the relative abundances of predominant soil genus to enhance microbial resistance to soil Pb. Likewise, coexistence of P. oxalicum and TCP showed the highest biomass and better branch root development of Pb-stressed in lettuces (Lactuca sativa L.) in pot experiment, and significantly reduced up to 88.1% of Pb translocation from soil to root over control. The reductions of Pb translocation and accumulation in root in P. oxalicum + TCP treatment could enhance the oxidase activities and alleviate the oxidative damages of H2O2 and O2.- in shoot tissues. Our study provided strong evidence to use PSF associated with P materials for the stable and eco-friendly soil Pb remediation.

Abstract:Microbial-induced phosphate (P) precipitation (MIPP) based on P-solubilizing microorganisms (PSM) is regarded as a promising approach to bioimmobilize environmental lead (Pb). Nevertheless, the underlying changes of Pb 2+ biotoxicity in PSM during MIPP process were rarely discussed. The current study explored the Pb 2+ immobilization and metabolic changes in PSM Penicillium oxalicum postexposure to Pb 2+ and/or tricalcium phosphate (TCP). TCP addition significantly increased soluble P concentrations, accelerated extracellular Pb mineralization, and improved antioxidative enzyme activities in P. oxalicum during MIPP process. Secondary Pb 2+ biomineralization products were measured as hydroxypyromorphite [Pb 10 (PO 4 ) 6 (OH) 2 ]. Using untargeted metabolomic and transcriptomics, we found that Pb 2+ exposure stimulated the membrane integrity deterioration and nucleotide metabolism obstruction of P. oxalicum . Correspondingly, P. oxalicum could produce higher levels of gamma-aminobutyric acid (GABA) to enhance the adaptive cellular machineries under Pb 2+ stress. While the MIPP process improved extracellular Pb 2+ mineralization, consequently alleviating the nucleotide metabolism inhibition and membrane deterioration. Multi-omics results suggested that GABA degradation pathway was stimulated for arginine biosynthesis and TCA cycle after Pb 2+ mineralization. These results provided new biomolecular information underlying the Pb 2+ exposure biotoxicities to microorganisms in MIPP before the application of this approach in environmental Pb 2+ remediation.

Abstract:Phosphorus (P) containing minerals are identified as effective Pb stabilizers in soil, while their low solubility limit the Pb immobilization efficiency. In this work, the combination of phosphate solubilizing fungi (PSF) Penicillium oxalicum and tricalcium phosphate (TCP) was constructed and applied to improve Pb immobilization stabilities in medium and soils. P. oxalicum + TCP could significantly improve Pb2+ removal to above 99% under different TCP/Pb2+ and pH values. TCP and P. oxalicum could remarkably immobilize Pb by ion exchange, and PbC2O4 precipitation or surface adsorption, respectively. While the enhanced Pb immobilization in P. oxalicum + TCP was explained by stronger Pb2+ interaction with tryptophan protein-like substances in extracellular polymeric substance, and the formation of the most stable Pb-phosphate compound hydroxypyromorphite (Pb5(PO4)3OH). Toxicity characteristic leaching procedure test showed that only 0.91% of Pb2+ was leachable in P. oxalicum + TCP treatment, significantly lower than that in P. oxalicum (2.90%) and TCP (7.52%) treatments. In addition, the lowest soil exchangeable Pb fraction (37.1%) and the highest available soil P (88.0 mg/kg) were both found in P. oxalicum + TCP treatment. By synergistically forming stable Pb-containing products, thus the combination of PSF and P minerals could significantly improve Pb2+ immobilization and stability in soils.

Abstract:Most current researches focus solely on reducing soil chromium availability. It is difficult to reduce soil Cr(VI) concentration below 5.0 mg kg -1 using single remediation technology. This study introduced a sustainable soil Cr (VI) reduction and stabilization system, Penicillium oxalicum SL2 -nanoscale zero-valent iron (nZVI), and investigated its effect on Cr(VI) reduction efficiency and microbial ecology. Results showed that P. oxalicum SL2 -nZVI effectively reduced soil total Cr(VI) concentration from 187.1 to 3.4 mg kg -1 within 180 d, and remained relatively stable at 360 d. The growth curve of P. oxalicum SL2 and microbial community results indicated that gamma-ray irradiation shortened the adaptation time of P. oxalicum SL2 and facilitated its colonization in soil. P. oxalicum SL2 colonization activated nZVI and its derivatives, and increased soil iron bioavailability. After restoration, the negative effect of Cr(VI) on soil microorganisms was markedly alleviated. Cr(VI), Fe(II), bioavailable Cr/Fe, Eh, EC and urease (SUE) were the key environmental factors of soil microbiota. Notably, Penicillium significantly stimulated the growth of urease-positive bacteria, Arthrobacter , Pseudarthrobacter , and Microvirga , synergistically reducing soil chromium availability. The combination of P. oxalicum SL2 and nZVI is expected to form a green, economical and long-lasting Cr(VI) reduction stabilization strategy.

Abstract:The ubiquity and persistence of organophosphate esters (OPEs) and heavy metal (HMs) pose global environmental risks. This study explored tris(2-chloroisopropyl)phosphate (TCPP) biomineralization coupled to lead (Pb 2+ ) biostabilization driven by denitrifying bacteria (DNB). The domesticated DNB achieved synergistic bioremoval of TCPP and Pb 2+ in the batch bioreactor (efficiency: 98 %).TCPP mineralized into PO 4 3− and Cl − , and Pb 2+ precipitated with PO 4 3− . The TCPP-degrading/Pb 2+ -resistant DNB: Achromobacter , Pseudomonas , Citrobacter , and Stenotrophomonas , dominated the bacterial community, and synergized TCPP biomineralization and Pb 2+ biostabilization. Metagenomics and metaproteomics revealed TCPP underwent dechlorination, hydrolysis, the TCA cycle-based dissimilation, and assimilation; Pb 2+ was detoxified via bioprecipitation, bacterial membrane biosorption, EPS biocomplexation, and efflux out of cells. TCPP, as an initial donor, along with NO 3 − , as the terminal acceptor, formed a respiratory redox as the primary energy metabolism. Both TCPP and Pb 2+ can stimulate phosphatase expression, which established the mutual enhancements between their bioconversions by catalyzing TCPP dephosphorylation and facilitating Pb 2+ bioprecipitation. TCPP may alleviate the Pb 2+ -induced oxidative stress by aiding protein phosphorylation. 80 % of Pb 2+ converted into crystalized pyromorphite. These results provide the mechanistic foundations and help develop greener strategies for synergistic bioremediation of OPEs and HMs.

Abstract:Currently, lead (Pb) has become a severe environmental pollutant and fungi hold a promising potential for the remediation of Pb-containing wastewater. The present study showed that Penicillium polonicum was able to tolerate 4 mmol/L Pb(II), and remove 90.3% of them in 12 days through three mechanisms: extracellular immobilization, cell wall adsorption, and intracellular bioaccumulation. In this paper. the three mechanisms were studied by Raman, X-ray diffraction analysis (XRD), scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR) and transmission electron microscopy (TEM). The results indicated that Pb(II) was immobilized as lead oxalate outside the fungal cell, bound with phosphate, nitro, halide, hydroxyl, amino, and carboxyl groups on the cell wall, precipitated as pyromorphite [Pb-5(PO4)(3)Cl] on the cell wall, and reduced to Pb(0) inside the cell. These combined results provide a basis for additionally understanding the mechanisms of Pb(II) removal by P. polonicum and developing remediation strategies using this fungus for lead-polluted water.
Pb(II) bioremediation using fresh algal-bacterial aerobic granular sludge and its underlying mechanisms highlighting the role of extracellular polymeric substances

Abstract:Lead (Pb) discharged from rural industries poses a significant threat to the environment and human health. Algal-bacterial aerobic granular sludge (A-B AGS) is a promising alternative for sewage treatment with high efficiency and good settleability. In this study, Pb(II) biosorption using fresh A-B AGS was investigated for the first time. The important role of extracellular polymeric substances (EPS) was revealed with the involved mechanisms being clarified. The desorbents for Pb recovery from Pb-loaded A-B AGS were also screened. Results showed that A-B AGS has an excellent maximum Pb adsorption capacity of 72.4 mg·g-1 at pH 6.0. EPS plays an important role in keeping microbial activity, Pb bonding, and providing metal ions (Ca, Na and Mg) for Pb ion exchanges. Electrostatic interaction, ion exchange, and bonding to functional groups may occur orderly in the Pb biosorption process and the formation of pyromorphite (Pb5(PO4)3Cl) contributes to Pb biosorption. About 66 % of the adsorbed Pb was accumulated in the A-B AGS microbial cells. Na2EDTA (0.05 M) can recover 60.3 % of the loaded Pb with the highest microbial activity of granules being remained. All the findings will provide the theoretical basis for the large-scale application of A-B AGS to bioremediate Pb(II)-containing wastewater.
Adaptation strategies and functional transitions of microbial community in pyrene-contaminated soils promoted by lead with Pseudomonas veronii and its extracellular polymeric substances

Abstract:Pyrene was designated as a remediation target in this study, and low contamination of lead (Pb) was set to induce heavy metal stress. Pseudomonas veronii and its extracellular polymeric substances (EPSs) were chosen for biofortification, with the aim of elucidating the structural, metabolic, and functional responses of soil microbial communities. Community analysis of soil microorganisms using high-throughput sequencing showed that the co-addition of P. veronii and EPSs resulted in an increase in relative abundance of phyla associated with pyrene degradation, and formed a symbiotic system dominated by Firmicutes and Proteobacteria, which involved in pyrene metabolism. Co-occurrence network analysis revealed that the module containing P. veronii was the only one exhibiting a positive correlation between bacterial abundance and pyrene removal, indicating the potential of bioaugmentation in enriching functional taxa. Biofortification also enhanced the abundance of functional gene linked to EPS production (biofilm formation- Pseudomonas aeruginosa ) and pyrene degradation. Furthermore, 17 potential functional bacteria were screened out using random forest algorithm. Lead contamination further promoted the growth of Proteobacteria, intensified cooperative associations among bacteria, and increased the abundance of bacteria with positive correlation with pyrene degradation. The results offer novel perspectives on alterations in microbial communities resulting from the synergistic impact of heavy metal stress and biofortification.

Abstract:Background and aims Salix integra has a high tolerance to lead (Pb) stress, and has been applied in the phytostabilization of Pb-Zn mine tailings in East China. Mechanisms for Pb sequestration and detoxification were poorly understood in Salix plants. The present study aimed to elucidate where Pb was localized and how Pb was combined in roots of S. integra for a better understanding of Pb tolerance strategies. Methods Histochemical methods combined with synchrotron-based X-ray fluorescence microscopy, transmission electron microscopy and fourier transform infrared spectroscopy were used to explore the distribution and chemical forms of Pb at cellular and subcellular levels in S. integra . Results The accumulation of Pb displayed a fast linear uptake in roots and stems in the short-term period, whereas a slower Freundlich-like uptake in leaves. Micro-XRF and histochemical staining revealed that the absorbed Pb in roots was mostly restricted in the cortex, and partly translocated in the stele. At the subcellular level, most Pb in roots was localized in the cell walls and trophoplast fraction, and the free -OH and pectin C—O/C—O—S groups may be involved in the cell wall sequestration. It was also found that the majority of Pb in roots existed as phosphate and oxalate. Conclusions S. integra has a high uptake potential of accumulating Pb in their roots. The absorbed Pb was primarily localized in the cell walls and combined with -OH or pectin groups, and precipitation as phosphate or oxalate may be responsible for the detoxification in root cells.
