Abstract:Visfatin play an essential role in the central regulation of appetite in birds. This study aimed to determine role of intracerebroventricular (ICV) injection of the visfatin on food intake and its possible interaction with neuropeptide Y (NPY) and nitric oxide system in neonatal broiler chicken. In experiment 1, neonatal chicken received ICV injection visfatin (1, 2 and 4 μg). In experiment 2, chicken received ICV injection of B5063 (NPY 1 receptor antagonist 1.25 μg), visfatin (4 μg) and co-injection of the B5063 + Visfatin. In experiments 3–6, SF22 (NPY 2 receptor antagonist 1.25 μg), SML0891 (NPY 5 receptor antagonist 1.25 μg), L–NAME (nitric oxide synthase inhibitor, 100 nmol) and L-arginine (Precursor of nitric oxide, 200 nmol) were injected instead of B5063. Then the amount of cumulative food was measured at 30, 60 and 120 min after injection. Obtained data showed, injection visfatin (2 and 4 μg) increased food intake compared to control group (P < 0.05). Co-injection of the B5063 + Visfatin decreased visfatin-induced hyperphagia compared to control group (P < 0.05). Co-injection of the L–NAME + Visfatin amplified visfatin-induced hyperphagia compared to control group (P < 0.05). The result showed that visfatin has hyperphagic role and this effect mediates via NPY 1 and nitric oxide system in neonatal chicken.

Abstract:The development of potential therapeutic and prophylactic vaccines for human papillomavirus (HPV) infection is a very exciting area of HPV research. There are a number of features of HPV biology that makes the development of a vaccine particularly difficult, although there are several examples of vaccines that have had spectacular success in the prevention of other viral diseases. Our poor understanding of the immune response to HPV infection is the first problem. We do not understand the mechanism by which spontaneous clearing of warts is generated and therefore cannot particularly target this pathway in the development of a vaccine. Furthermore, there is no in vitro culture system nor an animal model for HPV. Another problem is that there is no ready source of live virus that might be exploited for a live attenuated viral vaccine, such as was used with poliovirus. Although most other viruses spend a portion of their life cycle in the systemic circulation where they are vulnerable to neutralizing antibodies, HPV remain exclusively in the epithelium and thus antibodies must transverse the basement membrane and reach the other layers of the skin or mucosa to be effective in preventing infection. Significant progress is being made in the development of potential vaccine candidates despite these and other confounding factors.
Characterization of the feeding inhibition and neural activation produced by dorsomedial hypothalamic cholecystokinin administration.

Abstract:Within the dorsomedial hypothalamus (DMH), cholecystokinin (CCK) has been proposed to modulate neuropeptide Y (NPY) signaling to affect food intake. However, the neural circuitry underlying the actions of this CCK-NPY signaling system in the controls of food intake has yet to be determined. We sought to characterize the feeding inhibition and brain neural activation produced by CCK administration into the DMH of rats. We determined the time course of feeding inhibitory effects of exogenous DMH CCK, assessed NPY gene expression in the DMH in response to DMH CCK administration, and characterized c-Fos activation in the entire brain induced by CCK injection into the DMH using c-Fos like immunohistochemistry. We found that parenchymal injection of CCK into the DMH decreased food intake during the entire 22 h observation period, with a primary effect in the first 4 h, and down-regulated NPY gene expression in the DMH. c-Fos immunohistochemistry revealed that DMH CCK increased the number of c-Fos positive cells in the paraventricular nucleus (PVN), arcuate nucleus, suprachiasmatic nucleus and retrochiasmatic area as well as in the contralateral DMH. This pattern of activity is different from that produced by peripherally administered CCK which is short acting and primarily activates neurons in the nucleus of the solitary tract and area postrema, as well as the PVN and DMH. Together, these data suggest that DMH CCK plays an important role in the control of food intake, and does so by activating different pathways from those activated by peripheral CCK.
The Effects of Intrauterine Undernutrition on Pancreas Ghrelin and Insulin Expression in Neonate Rats.

Abstract:Ghrelin has a correlation with insulin secretion, beta-cell development, and diabetes in crucial development period. The aim of this study was to compare the changes in plasma ghrelin, insulin, and glucose concentrations, and variation of ghrelin expression in the pancreas in response to intrauterine malnutrition in newborn rats. Pregnant rats at day 2 were randomly divided into two groups: nourished (fed ad libitum; NR) and undernourished rats (UR). The offspring of NR were defined as normal-birth-weight group (NBW, n = 79) and those of UR were defined as low-birth-weight group (LBW, n = 74). Plasma glucose, ghrelin, and serum insulin of both dams and their pups were analyzed at the first day after birth. The entire pancreas was collected for determination of ghrelin and insulin mRNAs, and quantification of pancreas ghrelin and insulin. Immunohistochemical double staining and confocal microscopy were performed on rat pancreas. Birth weight was 5.81 +/- 0.64 and 4.76 +/- 0.23 g in NBW group and LBW group respectively. Fasting plasma ghrelin concentrations in UR group (1382 (1287-1513) pg/ml) were higher than that of NR group (1072 (974-1205) pg/ml). Plasma ghrelin concentrations in the LBW group (2176 (2031-2384) pg/ml) were significantly lower than that of the NBW group (2493 (2311-2675) pg/ml). Undernutrition caused a decrease in plasma insulin concentrations in both UR dams and LBW pups (P < 0.001). Ghrelin mRNA and total ghrelin of pancreas were significantly affected by intrauterine nutrition state. Pancreas insulin concentrations were significantly affected by intrauterine nutrition (P = 0.007). The majority of ghrelin-producing cells were present at the periphery of islets in the NBW group. Ghrelin was colocalized with insulin in ss-cells in LBW group. The percentage of ghrelin-positive cells in the islets of LBW group was significantly higher than that of the NBW group (P < 0.01). Intrauterine undernutrition may affect the birth weight, plasma insulin and ghrelin levels, islet ghrelin expression, and ghrelin cell distribution. It will be interesting to investigate intrauterine nutrition which is involved in islet ghrelin expression and ghrelin cell distribution.
Zinc and CGRP on Feed Intake of Piglets: Effects and Mechanism

Abstract:This experiment was conducted to study the effects of zinc and calcitonin gene-related peptide(CGRP) on feed intake and its regulation mechanism of piglets.Thirty crossed-bred(Duroc×Landrace×Large White) pigs with an average body weight of 9.57 kg were randomly assigned to five groups(control group Ⅰ,trial group Ⅰ,trial groupⅡ,control group Ⅱ and trial group Ⅲ) with 6 piglets per group.Piglets in all the groups were fed with the same basal diet.The feeding trial lasted for 14 days.After 24-hour fasting(free access to water),piglets in control group Ⅰ,trial group Ⅰ and trial group Ⅱ were injected 0,2 and 4 mg/kg BW zinc,and then slaughtered at 24-hour after injection.Piglets in control group Ⅱ and trial group Ⅲ were injected 0 and 0.05 mg/kg BW CGRP,and then slaughtered at 2-hour after injection.The results showed as follows: 1) compared with the control group Ⅰ,feed intake of trial group Ⅰ and trial group Ⅱ were significantly increased(P0.05) and decreased(P0.05),respectively.Compared with the control group Ⅱ,feed intake in trial group Ⅲ were significantly decreased(P0.05).2) There were no significant differences on serum glucose and leptin levels by injection of zinc(P0.05),but the triglyceride(TG),insulin and glucagon levels were significantly decreased(P0.05).Injection of CGRP significantly decreased serum glucose,TG and insulin levels(P0.05) and increased glucagon levels(P0.05),but had no effect on leptin levels(P0.05).3) Compared with the control group Ⅰ,the expression levels of NPY mRNA in trial group Ⅰ and trial group Ⅱ were significantly increased(P0.05),but the CGRP and CCK mRNA expression levels were significantly decreased(P0.05).Compared with the control group Ⅱ,the CGRP and CCK mRNA expression level were significantly increased(P0.05),while NPY mRNA expression levels was decreased significantly(P0.05) in trial group Ⅲ.These results indicate that zinc can stimulate intake of piglets through inducing NPY mRNA expression and inhibiting CGRP and CCK mRNA expression.However,CGRP can compromise feed intake of piglets by inhibiting NPY mRNA expression and inducing CGRP as well as CCK mRNA expression.

Abstract:Animals at the neonatal stage have to eat more to support better growth and health. However, it is difficult to understand the mechanism of feeding during an early stage of life in the brain of the rodent model. Chickens are precocial and they can look for their food by themselves right after hatching. Neonatal chicks have a relatively large-sized brain; therefore, the drugs are easy to administer centrally and changes in food intake can be clearly monitored. Sleeping status, which affects food intake, can be estimated from the posture. The closest vertebrate outgroup to mammals is birds, but it was reported that the organization of the human genome is closer to that of the chicken than the mouse. Thus, it is important to understand the central mechanism of feeding regulation in the neonatal chicks. In neuropeptides, the number of candidates as the orexigenic factor was less than those as the anorexigenic factor, even at an early growth stage. Some of the neuropeptides have reverse effects, e.g., ghrelin and prolactin releasing peptides, or no effects compared to the effects confirmed in mammals. Some of the genetic differences between meat-type (broiler) and layer-type chickens would explain the difference in food intake. On the other hand, it was difficult to explain the feeding mechanism by neuropeptides alone, as neonatal chicks have a repeated feeding, sleeping, and resting behavior within a short period. Some of the amino acids and their metabolites act centrally to regulate feeding with sedative and hypnotic effects. In conclusion, endogenous neuropeptides and endogenous and/or exogenous nutrients like amino acids collaborate to regulate feeding behavior in neonatal chicks.

In Ovo Leptin Administration Affects Hepatic Lipid Metabolism and Microrna Expression in Newly Hatched Broiler Chickens.

Abstract:BACKGROUND:A leptin-like immunoreactive substance has been found in chicken eggs and has been implicated in serving as a maternal signal to program offspring growth and metabolism. In the present study, we investigated the effects of in ovo leptin administration on hatch weight, serum and hepatic concentrations of metabolites and hormones, as well as on the expression of genes involved in hepatic lipid metabolism and the predicted microRNAs (miRNAs) targeting the affected genes. To this end we injected fertile eggs with either 0.5 μg of recombinant murine leptin or vehicle (PBS) before incubation.RESULTS:Prenatally leptin-exposed chicks showed lower hatch weight, but higher liver weight relative to the body weight, compared to the control group. In ovo leptin treatment increased the hepatic content and serum concentration of leptin in newly hatched chickens. The hepatic contents of triglycerides (TG) and total cholesterol (Tch) were decreased, whereas the serum levels of TG, Tch and apolipoprotein B (ApoB) were increased. The hepatic mRNA expression of sterol regulator element binding protein 1 (SREBP-1c), SREBP-2, hydroxy-3-methylglutaryl coenzyme A reductase (HMGCR) and cholesterol 7α-hydroxylase 1 (CYP7A1) was significantly up-regulated, as was the protein content of both SREBP-1c and SREBP-2 in hepatic nuclear extracts of leptin-treated chickens. Moreover, out of 12 miRNAs targeting SREBP-1c and/or HMGCR, five were significantly up-regulated in liver of leptin-treated chicks, including gga-miR-200b and gga-miR-429, which target both SREBP-1c and HMGCR.CONCLUSIONS:These results suggest that leptin in ovo decreases hatch weight, and modifies hepatic leptin secretion and lipid metabolism in newly hatched broiler chickens, possibly via microRNA-mediated gene regulation.
Central serotoninergic system mediates Neuromedin S (NMS) induced hypophagia in layer-type chicken

Abstract:According to previous studies, the serotonergic system plays a remarkable role in regulating meal intake, however, its role in the mediation of feeding caused by neuromodulin S (NMS) has not been investigated in birds. In the present research, 5 trials were considered in order to determine the links between these systems. In treatment 1, chickens received intracerebroventricular (ICV) infusion of NMS (0.25, 0.5, and 1 nmol). NMS (1 nmol), SB242084 (1.5 μg), a 5-HT2C receptor antagonist, and NMS + SB242084 were injected in the treatment 2. In subsequent treatments, instead of SB242084, 8-OH-DPAT (agonist of 5-HT1A receptor, 15.25 nmol), PCPA (serotonin synthesis inhibitor, 1.25 μg), and Fluoxetine (serotonin reuptake inhibitor, 10 μg) were applied. Then, total food consumption was recorded for 120 minutes. Based on observations, NMS dose-dependently attenuated meal intake (P<0.05). Hypophagia was diminished with NMS + SB242084 administration (P<0.05). ICV infusion of NMU + 8-OH-DPAT had no significant effect on the hypophagia (P≥0.05). The NMS-induced decreasing feed intake was attenuated with co-infusion of NMS+ PCPA (P< 0.05). Also, hypophagia was strengthened by NMS+ Fluoxetine co-infusion (P< 0.05). According to the results, the NMS-induced hypophagia is possibly mediated via 5-HT2c receptors in layer-type chickens.
Effect of early feed restriction on metabolic programming and compensatory growth in broiler chickens.

Abstract:The effect of early feed restriction on metabolic programming and compensatory growth was studied in broiler chickens. A total of 480 female 1-d-old broiler birds (Aconred) were randomly allocated to ad libitum and feed-restricted groups, each of which was replicated 6 times with 40 birds per replicate. Broilers were provided commercial diets. Feed-restricted broilers were deprived of feed for 4 h per day from 1 to 21 d of age. Effects of treatments were determined at 21 and 63 d of age. In feed-restricted birds at 21 d of age, BW, average daily gain and average daily feed intake, breast muscle (P < 0.01), carcass yield (P < 0.05), and abdominal fat (P < 0.05) were decreased. Ether extract content in breast muscle was increased (P < 0.01), whereas CP content was slightly decreased. Triiodothyronine (P < 0.01) and thyroxine (P < 0.05) were decreased in serum. Free fatty acid and very low density lipoprotein were slightly increased in serum, whereas triglyceride and glucose were decreased (P < 0.01). Activities of NADPH-generating enzymes in liver including malic dehydrogenase, isocitrate dehydrogenase, and glucose-6-phosphate remained unchanged in ad libitum birds, whereas hormone-sensitive lipase activity was increased (P < 0.01). In feed-restricted birds at 63 d of age, BW, average daily gain, average daily feed intake, carcass yield, breast muscle yield, and serum triiodothyronine and thyroxine remained as ad libitum birds, whereas abdominal fat yield was increased (P < 0.05). Ether extract content in breast muscle was decreased (P < 0.01), whereas CP content was increased (P < 0.05). Activities of NADPH-generating enzymes were significantly increased, except abdominal malic dehydrogenase and hormone-sensitive lipase activity was decreased (P < 0.01) in liver and abdominal fat. Lipoprotein lipase activity was increased (P < 0.05) in abdominal fat. In summary, feed restriction severely affected growth performance and lipid metabolism in broilers in the early period. Because there was no statistical difference among the final BW, near full compensatory growth was achieved. In addition, early feed restriction might have induced prolonged metabolic programming in chicks and led to adult obesity.
Promotion of adipogenesis by neuropeptide Y during the later stages of chicken preadipocyte differentiation

Abstract:Neuropeptide Y (NPY) promotes adipogenesis in both birds and mammals, although mechanisms in avians remain unclear. The objective of this study was thus to evaluate effects of NPY on chick preadipocyte proliferation and differentiation. Preadipocytes were treated with 0, 1, 10, or 100 nmol/L NPY and gene expression and cellular proliferation were evaluated at 12, 24, and 48 h. At 12 h posttreatment, mRNA abundance of topoisomerase II alpha (TOP2A), and thioredoxin-dependent peroxidase 2 was upregulated and NPY was downregulated in response to NPY (0 vs. 100 nmol/L) in preadipocytes. Cells were also treated with NPY during differentiation and harvested at 8, 10, and 12 days postinduction of differentiation. At day 8 postinduction of differentiation, there was increased lipid accumulation (0 vs. 10 and 100 nmol/L), expression of CCAAT/enhancer binding protein β and fatty acid binding protein 4 (FABP4) (0 vs. 100 nmol/L), and sterol regulatory element-binding protein (0 vs. 10 and 100 nmol/L) mRNA in NPY-treated cells. The number of proliferating cells decreased on day 8 in response to NPY (0 vs. 10 nmol/L). At day 10, FABP4 and Kruppel-like factor 7 mRNAs were downregulated (0 vs. 10 and 100 nmol/L, and 100 nmol/L, respectively), and at day 12, TOP2A mRNA was down-regulated (0 vs. 100 nmol/L) in response to NPY treatment. Activity of glycerol-3-phosphate dehydrogenase (G3PDH) was increased on days 10 and 12 in NPY-treated cells (0 vs. 100 nmol/L). Increased gene expression of proliferation markers in preadipocytes, and during differentiation increased expression of transcription factors and a fatty acid transporter, increased lipid accumulation, and increased activity of G3PDH suggest that NPY may enhance preadipocyte activity, adipogenesis, and promotes lipid accumulation throughout chicken adipocyte differentiation.

Abstract:Neuropeptide Y (NPY) receptors and its ligands, NPY, peptide YY (PYY) and pancreatic polypeptide (PP), are suggested to regulate many physiological processes including food intake in birds. However, our knowledge regarding this avian NPY system remains rather limited. Here, we examined the tissue expression of NPY, PYY and PP and the gene structure, expression and signaling of three NPY receptors (cY1, cY4 and cY6) in chickens. The results showed that 1) NPY is widely expressed in chicken tissues with abundance noted in the hypothalamus via quantitative real-time PCR, whereas PYY is highly expressed in the pancreas, gastrointestinal tract and various brain regions, and PP is expressed almost exclusively in the pancreas; 2) cY1, cY4 and cY6 contain novel non-coding exon(s) at their 5'-UTR; 3) The wide tissue distribution of cY1 and cY4 and cY6 were detected in chickens by quantitative real-time PCR and their expression is controlled by the promoter near exon 1, which displays strong promoter activity in DF-1 cells as demonstrated by Dual-luciferase reporter assay; 4) Monitored by luciferase reporter assays, activation of cY1 and cY4 expressed in HEK293 cells by chicken NPY1-36, PYY1-37, and PP1-36 treatment inhibits cAMP/PKA and activates MAPK/ERK signaling pathways, while cY6-expressing cells show little response to peptide treatment, indicating that cY1 and cY4, and not cY6, can transmit signals in vitro. Taken together, our study offers novel information about the expression and functionality of cY1, cY4, cY6 and their ligands in birds, and helps to decipher their conserved roles in vertebrates.
The effect of brain-derived neurotrophic factor (BDNF) on food consumption of broilers: Role of serotonergic receptors

Abstract:Previously, the effects of brain-derived neurotrophic factor (BDNF) on feed intake of mammals were studied, however, its role in avian feeding regulation remains unclear. Therefore, the present study aimed to investigate the possible effects of BDNF on the food consumption of broilers and its interaction with the serotonergic system. In the first treatment, broilers received intracerebroventricular (ICV) infusion of BDNF (7.5, 15, and 30 μg). BDNF (30 μg), Fluoxetine (10 μg), a serotonin reuptake inhibitor, and BDNF + Fluoxetine were injected in the second treatment. In subsequent treatments, instead of Fluoxetine, PCPA (serotonin synthesis inhibitor, 1.25 μg), SB242084 (antagonist of 5-HT2C receptor, 1.5 μg), and 8-OH-DPAT (agonist of 5-HT1A receptor, 15.25 nmol) were applied. Then, total food intake was recorded for 2 hours. According to the results, BDNF injection (15 and 30 μg) caused a significant decrease in the food intake of broilers compared to the control group (P<0.05). Simultaneous administration of PCPA and SB242084 with BDNF suppressed BDNF-induced hypophagia (P<0.05), while the injection of fluoxetine + BDNF strengthened this hypophagic effect (P<0.05). The decrease in feed intake caused by BDNF was not significantly changed by simultaneous injection of BDNF + 8-OH-DPAT (P≥0.05). According to the findings, it seems that BDNF reduces food consumption in broilers and this effect is probably mediated via 5-HT2c receptors.
Chicken adaptive response to low energy diet: main role of the hypothalamic lipid metabolism revealed by a phenotypic and multi-tissue transcriptomic approach

Abstract:Background: Production conditions of layer chicken can vary in terms of temperature or diet energy content compared to the controlled environment where pure-bred selection is undertaken. The aim of this study was to better understand the long-term effects of a 15%-energy depleted diet on egg-production, energy homeostasis and metabolism via a multi-tissue transcriptomic analysis. Study was designed to compare effects of the nutritional intervention in two layer chicken lines divergently selected for residual feed intake. Results: Chicken adapted to the diet in terms of production by significantly increasing their feed intake and decreasing their body weight and body fat composition, while their egg production was unchanged. No significant interaction was observed between diet and line for the production traits. The low energy diet had no effect on adipose tissue and liver transcriptomes. By contrast, the nutritional challenge affected the blood transcriptome and, more severely, the hypothalamus transcriptome which displayed 2700 differentially expressed genes. In this tissue, the low-energy diet lead to an over-expression of genes related to endocannabinoid signaling (CN1R, NAPE-PLD) and to the complement system, a part of the immune system, both known to regulate feed intake. Both mechanisms are associated to genes related polyunsaturated fatty acids synthesis (FADS1, ELOVL5 and FADS2), like the arachidonic acid, a precursor of anandamide, a key endocannabinoid, and of prostaglandins, that mediate the regulatory effects of the complement system. A possible regulatory role of NR1H3 (alias LXRα) has been associated to these transcriptional changes. The low-energy diet further affected brain plasticity-related genes involved in the cholesterol synthesis and in the synaptic activity, revealing a link between nutrition and brain plasticity. It upregulated genes related to protein synthesis, mitochondrial oxidative phosphorylation and fatty acid oxidation in the hypothalamus, suggesting reorganization in nutrient utilization and biological synthesis in this brain area. Conclusions: We observed a complex transcriptome modulation in the hypothalamus of chicken in response to low-energy diet suggesting numerous changes in synaptic plasticity, endocannabinoid regulation, neurotransmission, lipid metabolism, mitochondrial activity and protein synthesis. This global transcriptomic reprogramming could explain the adaptive behavioral response (i.e. increase of feed intake) of the animals to the low-energy content of the diet.
Effect of Dietary N-carbamylglutamate on Development of Ovarian Follicles Via Enhanced Angiogenesis in the Chicken.

Abstract:N-carbamylglutamate (NCG), an analogue of N-acetyl-L-glutamate (NAG), can increase arginine synthesis in mammals and improve the reproductive performance. However, the effect of NCG on poultry laying performance is still unclear. This study investigated the effect of dietary NCG on development of chicken ovarian follicles. The dosage and timing for NCG administration were evaluated for its effect on follicular development. Results showed that supplementation with 1% NCG in the diet for 14 D led to accelerated development of growing follicles (over 60 μm in oocyte diameter) and significantly increased feed intake and feed efficiency. Plasma amino acids (AA) analysis showed that feeding with 1% NCG significantly increased of plasma AA levels. RNA-seq analysis revealed that NCG supplementation upregulated expression of genes related to angiogenesis and cell proliferation, but downregulated expression of apoptosis-related genes. Meanwhile, RT-qPCR and Western blot analysis validated the RNA-seq results. Moreover, NCG enhanced plasma NO level; upregulated expression of PKG-I, Raf1, and p-p38; and increased angiogenesis of the ovaries. In conclusion, dietary NCG (1% for 14 D) can promote development of ovarian follicles by increasing angiogenesis in ovaries of the chicken.
Peripheral 5-HT Mediates Gonadotropin-Inhibitory Hormone-Induced Feeding Behavior and Energy Metabolism Disorder in Chickens via the 5-HT2C Receptor

Abstract:Introduction: Since the discovery of gonadotropin-inhibitory hormone (GnIH), it has been found to play a critical role in reproduction in vertebrates. Recently, a regulatory role of GnIH in appetite and energy metabolism has emerged, although its precise physiological mechanisms remain unknown. Methods: Thus, the present study evaluated the effects of a single or long-term intraperitoneal GnIH treatment on the food intake, weight, and glucolipid metabolism of chickens, as well as investigating the possible neuroendocrinology factors and mechanisms involved in GnIH-induced obesity and glucolipid metabolism disorder. Results: Our results show that the intraperitoneal administration of GnIH to chickens resulted in a marked body mass increase, hyperlipidemia, hyperglycemia, and glucose intolerance. Subsequently, the results of metabolomics studies and the pharmacological inhibition of the 5-HT2C receptor revealed that blocking the 5-HT2C receptor reinforced the effects of GnIH on food intake, body weight, and blood glucose and lipid levels, resulting in even worse cases of GnIH-induced hyperglycemia, hyperlipidemia, and hepatic lipid deposition. This suggests that, via the 5-HT2C receptor, peripheral 5-HT may act as a negative feedback regulator to interplay with GnIH and jointly control energy balance homeostasis in chickens. Discussion: Our present study provides evidence of cross-talk between GnIH and 5-HT in food intake and energy metabolism at the in vivo pharmacological level, and it proposes a molecular basis for these interactions, suggesting that functional interactions between GnIH and 5-HT may open new avenues for understanding the mechanism of the neuroendocrine network involved in appetite and energy metabolism, as well as providing a new therapeutic strategy to prevent obesity, diabetes, and metabolic disorders.

Abstract:Fasting-induced hypothalamic metabolic reprogramming is involved in regulating energy homeostasis and appetite in mammals, but this phenomenon remains unclear in poultry. In this study, the expression patterns of a panel of genes related to neuropeptides, glucose, and lipid metabolism enzymes in the hypothalamus of chickens during fasting and refeeding were characterized by microarray analysis and quantitative PCR. Results showed that 48 h of fasting upregulated (P < 0.05) the mRNA expressions of orexigenic neuropeptide Y and agouti-related protein but downregulated (P < 0.05) that of anorexigenic neuropeptide pro-opiomelanocortin; growth hormone-releasing hormone; islet amyloid polypeptide; thyroid-stimulating hormone, β; and glycoprotein hormones, α polypeptide. After 48 h of fasting, the mRNA expression of fatty acid β-oxidation [peroxisome proliferator-activated receptor α (PPARα), carnitine palmitoyltransferase 1A, and forkhead box O1], energy sensor protein [sirtuin 1 (SIRT1) and forkhead box O1], and glycolysis inhibitor (pyruvate dehydrogenase kinase, isozyme 4) were enhanced, but that of fatty acid synthesis and transport associated genes (acetyl-CoA carboxylase α, fatty acid synthase, apolipoprotein A-I, endothelial lipase, and fatty acid binding protein 7) were suppressed. Liver and muscle also demonstrated similar expression patterns of genes related to glucose and lipid metabolism with hypothalamus, except for that of acetyl-CoA carboxylase α, acyl-CoA synthetase long-chain family member 4, and apolipoprotein A-I. The results of intracerebroventricular (ICV) injection experiments confirmed that α-lipoic acid (ALA, pyruvate dehydrogenase kinase, isozyme 4 inhibitor, 0.10 μmol) and NADH (SIRT1 inhibitor, 0.80 μmol) significantly suppressed the appetite of chickens, whereas 2-deoxy-d-glucose (glycolytic inhibitor, 0.12 to 1.20 μmol) and NAD+ (SIRT1 activator, 0.08 to 0.80 μmol) increased feed intake in chickens. The orexigenic effect of NAD+ was also blocked by cotreatment with NADH. However, ICV injection of either GW7647 (PPARα agonist) or GW6471 (PPARα antagonist) showed no effects on feed intake. Results suggested that hypothalamic glycolysis (inhibited by ALA and promoted by 2-deoxy-d-glucose) and SIRT1 (inhibited by NADH and promoted by NAD+), not PPARα, were probably involved in feed intake regulation in chickens.

Abstract:The goal of this study was to investigate the influence of l-arginine (l-Arg) supplementation on diet-induced changes in hypothalamus and liver proteome of laying hens. Layers were fed either an isonitrogenous control diet or a l-Arg diet. The test included a 2-week acclimation period and a 12-week experimental period. Eight layers per group were sacrificed at terminal of the experiment underwent 12 fasting. Blood and tissue samples of hypothalamus and liver were collected for further analysis. The levels of serum nitric oxide and hypothalamus neuropeptide Y of layers in l-Arg group were increased in comparison with those in control group. Quantitative proteomic analyses showed that a total of 3,715 hypothalamus proteins (235 differentially expressed) and 3797 liver proteins (373 differentially expressed) were detected between control and l-Arg-fed groups. A further enriched Gene Ontology term analysis of proteins found that 17 hypothalamus proteins (11 upregulated and six downregulated) and 29 liver proteins (14 upregulated and 15 downregulated) were altered differentially between the two groups. Our findings revealed the changes in metabolic and hormonal signals in central nervous system and peripheral tissues by responding to l-Arg feeding, which provides a possible way to gain a better understanding of l-Arg function in laying hens.
