Abstract:Mosquito behavioral assays are an important component in vector research and control tool development. Here, we present a protocol for rearing Anopheles mosquitoes, performing host-seeking behavioral bioassays, and collecting 3D flight tracks in a large wind tunnel. We describe steps for setting up host-seeking landing assays, both as a non-choice and as a dual-choice assay, and analyzing flight tracks. This protocol can be applied in the research of several behavioral traits, including nectar seeking, resting, mating, and oviposition behavior. Development of novel compact wind tunnel for testing efficacy of insecticide formulated products in mosquitoes

Abstract:Background: Ultra-low volume (ULV) space sprays aerosolize insecticide formulated products (FP) to contact flying mosquitoes, while barrier sprays expose mosquitoes to FP residue on vegetation and other surfaces. Centers for Disease Control and Prevention bottle bioassays used to assess insecticide resistance are based on residual active ingredient (AI) exposure and do not directly relate to FP efficacy. The current pilot study developed a novel compact wind tunnel for mosquito exposure to FP. Caged Aedes albopictus and Culex pipiens/quinquefasciatus were exposed to undiluted Biomist®3 + 15 FP (permethrin AI) or air (control) within the wind tunnel, transferred to new cages, and held in a 28 °C incubator. Separate mosquitoes were exposed to residual permethrin AI (8 μg mL-1) in bottle bioassays. Mortality was monitored 15, 30, 60, and 120 min post-exposure. Results: Chi-square tests (P < 0.05) showed significantly higher mortality in Aedes compared to Culex populations for most time points in both bioassay and wind tunnel exposure groups. As expected, mosquitoes exposed to Biomist®3 + 15 showed higher mortality rates than bottle bioassay exposure to permethrin. Two Culex colonies resistant to permethrin in bottle bioassays were susceptible to Biomist®3 + 15 in the wind tunnel. Conclusion: The novel compact wind tunnel developed here may be an alternative to field trials for testing FP efficacy, avoiding factors such as weather, logistical planning, and extended personnel hours. The wind tunnel could allow programs to conveniently test efficacy of multiple FP. Comparisons of different insecticide exposure methods provide practical information to inform operational decisions. © 2024 The Authors. Video Recording Can Conveniently Assay Mosquito Locomotor Activity

Abstract:Anopheles gambiae and Aedes aegypti are perhaps the best studied mosquito species and important carriers of human malaria and arbovirus, respectively. Mosquitoes have daily rhythms in behaviors and show a wide range of activity patterns. Although Anopheles is known to be principally nocturnal and Aedes principally diurnal, details of mosquito activity are not easily assayed in the laboratory. We recently described FlyBox, a simple tracking system for assaying Drosophila locomotor activity rhythms and thought that it might also be applicable to monitoring mosquito activity. Indeed, we show here that FlyBox can easily, conveniently, affordably and accurately measure the activity of Anopheles as well as Aedes over several days. The resulting profiles under light-dark as well as constant darkness conditions are compatible with results in the literature, indicating that this or similar systems will be useful in the future for more detailed studies on a range of insect species and under more diverse laboratory conditions.

Abstract:High speed video observations of free flying male Aedes aegypti mosquitoes, the dengue and yellow fever vector, along with custom measurement methods, enable measurement of wingbeat frequency, body position and body orientation of mosquitoes during flight. We find these mosquitoes flap their wings at approximately 850 Hz. We also generate body yaw, body pitch and wing deviation measurements with standard deviations of less than 1 degree and find that sideways velocity and acceleration are important components of mosquito motion. Rapid turns involving changes in flight direction often involve large sideways accelerations. These do not correspond to commensurate changes in body heading, and the insect's flight direction and body heading are decoupled during flight. These findings call in to question the role of yaw control in mosquito flight. In addition, using orientation data, we find that sideways accelerations are well explained by roll-based rotation of the lift vector. In contrast, the insect's body pitch angle does not correlate with its forward acceleration. This implies that controlling body roll is important to mosquito dynamics. The dynamic importance of stabilizing body pitch and body yaw is less clear.

Abstract:Mosquito flight dynamics was considered using an advanced mathematical model with experimentally determined and biologically relevant parameters. The model was developed using a self-consistent algorithm. It described mosquito's body and wing oscillations using mechanics equations and aerodynamic flows, simultaneously. Six equations were used for the mechanics of the mosquito (relative to the center of mass), and Navier–Stokes equations simulated aerodynamics. Numerical methods employed deformable computational mesh with specific mesh construction near the wing boundaries. Using size, shape and weight characteristics of the Culex quinquefasciatus (male) mosquito the flight dynamics was computed and analyzed. It was determined that the mosquito lift force was proportional to the square of the wing frequency and wing size. Specifically, we found a critical frequency of approximately 820 Hz that corresponded to mosquito freezing when the lift force compensated gravity. This number was consistent to experimentally determined mosquito flight characteristics. Mechanism of mosquito's lift force generation was discussed.

Effects of indoor air movement and ambient temperature on mosquito (Anopheles gambiae) behaviour around bed nets: implications for malaria prevention initiatives

Abstract:Background: Until recently, relatively little research has been done on how mosquitoes behave around the occupied bed net in the indoor environment. This has been partly remedied in the last few years through laboratory and field studies, most of these using video methods and mosquito flight tracking. Despite these recent advances, understanding of the mosquito-bed net environment system, and the principles that underlie mosquito behaviour within it, is limited. This project aimed to further understand this system by studying the effects of gently moving air (such as might be introduced through room design to make the indoor environment more comfortable and conducive to ITN use) and warmer vs. cooler ambient conditions on mosquito activity around ITNs and other bed nets. Methods: The activity of colonized female Anopheles gambiae around an occupied untreated bed net set up in a mosquito-proof tent in a large laboratory space was recorded under different ambient conditions using a laser detection-video recording system. Conditions tested were 'cool' (23-25 °C) and 'warm' (27-30 °C) air temperatures and the presence or absence of a cross-flow produced by a small central processing unit (CPU) fan pointed at the side of the net so that it produced a 'low-' or 'high-' speed cross-draught (approx. 0.1 and 0.4 m/s, respectively). Near-net activity in recordings was measured using video image analysis. Results: In cool, still air conditions, more than 80% of near-net activity by An. gambiae occurred on the net roof. Introduction of the low-speed or high-speed cross-draught resulted in an almost total drop off in roof activity within 1 to 2 min and, in the case of the high-speed cross-draught, a complementary increase in activity on the net side. In warm, still conditions, near-net activity appeared to be lower overall than in cool, still air conditions and to be relatively less focussed on the roof. Introduction of the high-speed cross-draught in warm conditions resulted in a decrease in roof activity and increase in side activity though neither effect was statistically significant. Conclusions: Results are interpreted in terms of the flow of the stimulatory odour plume produced by the net occupant which, consistent with established principles of fluid dynamics, appears to rise quickly and remain more intact above the net occupant in cool, still air than in warm, still air. Cross-draught effects are ascribed to the changes they cause in the flow of the host odour plume as opposed to mosquito flight directly. The implications of these results for house designs that promote indoor air movement, on bed net design, and on other vector control measures are discussed. How mosquitoes approach a net is influenced both by indoor temperature and ventilation and their interaction. This system is in need of further study.

Abstract:Mosquito vectors of pathogens (e.g., Aedes, Anopheles, and Culex spp. which transmit dengue, Zika, chikungunya, West Nile, malaria, and others) are of increasing concern for global public health. These vectors are geographically shifting under climate and other anthropogenic changes. As small-bodied ectotherms, mosquitoes are strongly affected by temperature, which causes unimodal responses in mosquito life history traits (e.g., biting rate, adult mortality rate, mosquito development rate, and probability of egg-to-adult survival) that exhibit upper and lower thermal limits and intermediate thermal optima in laboratory studies. However, it remains unknown how mosquito thermal responses measured in laboratory experiments relate to the realized thermal responses of mosquitoes in the field. To address this gap, we leverage thousands of global mosquito occurrences and geospatial satellite data at high spatial resolution to construct machine-learning based species distribution models, from which vector thermal responses are estimated. We apply methods to restrict models to the relevant mosquito activity season and to conduct ecologically plausible spatial background sampling centered around ecoregions for comparison to mosquito occurrence records. We found that thermal minima estimated from laboratory studies were highly correlated with those from the species distributions (r = 0.87). The thermal optima were less strongly correlated (r = 0.69). For most species, we did not detect thermal maxima from their observed distributions so were unable to compare to laboratory-based estimates. The results suggest that laboratory studies have the potential to be highly transportable to predicting lower thermal limits and thermal optima of mosquitoes in the field. At the same time, lab-based models likely capture physiological limits on mosquito persistence at high temperatures that are not apparent from field-based observational studies but may critically determine mosquito responses to climate warming. Our results indicate that lab-based and field-based studies are highly complementary; performing the analyses in concert can help to more comprehensively understand vector response to climate change.
On the Role of Vortical Structures in Aerodynamic Performance of a Hovering Mosquito

Abstract:Mosquitoes have slimmer wings, higher flapping frequencies, and much lower amplitudes than most other insects. These unique features signify special aerodynamic mechanisms. Besides the leading-edge vortex, which is one of the most common mechanisms of flapping-wing flight, mosquitoes have two distinctive mechanisms: trailing-edge vortex and rotational drag. In this study, the three-dimensional flow field around a hovering mosquito is simulated by using the immersed boundary method. The numerical results agree well with previous experimental data. Mechanisms unique to mosquitoes are identified from the instantaneous pressure and vorticity fields. The flow domains, containing several vortical structures produced by the flapping wings, are divided into different regions for quantitatively analyzing the contribution of vortical structures to the lift. Advection of the trailing-edge vortex and production of the leading-edge vortex each contribute peaks in lift. Passive deformation of the wings is also important, as it stabilizes delayed stall and decreases by 26% the maximum aerodynamic power required for hovering flight. In addition, the lift coefficient and power economy are improved as the Reynolds number increases, which explains the better ability of larger mosquitoes to seek and feed on hosts from the aerodynamic point of view. Acoustotactic response of mosquitoes in untethered flight to incidental sound

Abstract:Mosquitoes are vectors for some of the most devastating diseases on the planet. Given the centrality of acoustic sensing in the precopulatory behavior of these vectors, the use of an exogenous acoustic stimulus offers the potential of interfering with the courtship behavior of these insects. Previous research on the acoustotactic response of mosquitoes has been conducted on tethered preparations using low-intensity sound stimuli. To quantify differences in acoustotactic responses between mosquitos of distinct sex and species, we examined the effects of incidental sound stimuli on the flight behavior of free-flying male vs. female Aedes aegypti and Anopheles gambiae mosquitoes. The key variables were sound frequency (100-1000 Hz) and intensity (67-103 dB, measured at 12.5 cm from the source), and the acoustotactic response was measured in terms of the relative increase in flight speed in response to the stimulus. The data show, for the first time, significant sex- and species-specific differences in acoustotactic responses. A. aegypti exhibited a greater response to sound stimulus compared to An. gambiae, and the response also extended over a larger range of frequencies. Furthermore, the males of both species displayed a greater acoustotactic response than females, with An. gambiae females exhibiting minimal response to sound.
A Method for Repeated, Longitudinal Sampling of Individual Aedes aegypti for Transmission Potential of Arboviruses

Abstract:Mosquito-borne viruses are the cause of significant morbidity and mortality worldwide, especially in low- and middle-income countries. Assessing risk for viral transmission often involves characterization of the vector competence of vector–virus pairings. The most common determination of vector competence uses discreet, terminal time points, which cannot be used to investigate variation in transmission aspects, such as biting behavior, over time. Here, we present a novel method to longitudinally measure individual biting behavior and Zika virus (ZIKV) transmission. Individual mosquitoes were exposed to ZIKV, and from 9 to 24 days post-exposure, individuals were each offered a 180 μL bloodmeal every other day. Biting behavior was observed and characterized as either active probing, feeding, or no bite. The bloodmeal was then collected, spun down, serum collected, and tested for ZIKV RNA via qRT-PCR to determine individuals’ vector competence over time. This included whether transmission to the bloodmeal was successful and the titer of expectorated virus. Additionally, serum was inoculated onto Vero cells in order to determine infectiousness of positive recovered sera. Results demonstrate heterogeneity in not only biting patterns but expectorated viral titers among individual mosquitoes over time. These findings demonstrate that the act of transmission is a complex process governed by mosquito behavior and mosquito–virus interaction, and herein we offer a method to investigate this phenomenon.

Abstract:Blood feeding represents a critical event in the life cycle of female mosquitoes. In addition to providing nutrients to the mosquito, blood feeding facilitates the transmission of parasites and viruses to hosts, potentially having devastating health consequences. Despite this, our understanding of these short, yet important bouts of behaviour is incomplete. How and where a mosquito decides to feed and the success of feeding can influence the transmission of pathogens, while a more thorough understanding may allow interventions to reduce or prevent infections. Recent advances in machine vision and automated tracking presents the opportunity to observe and understand blood feeding behaviour of mosquitoes at unprecedented spatial and temporal resolution. Here, we combine these technologies with novel designs for behavioural arenas and controllable artificial host cues to enable detailed observations of biting behaviour using relatively inexpensive and readily available materials. Combined with a workflow for quantitative image analysis, we are able to describe nuanced, high resolution biting behaviour under tightly controlled conditions.
A new Y‐tube olfactometer for mosquitoes to measure the attractiveness of host odours

Abstract:In a new bioassay a small Y‐shaped wind tunnel is used to quantitatively investigate the responses of mosquitoes to host odours. Female yellow fever mosquitoes, Aedes aegypti (L.) (Diptera: Culicidae) were tested to (1) a human hand, (2) an extract with human skin residues, (3) L‐(+)‐lactic acid, and (4) carbon dioxide. The responses to the skin extract followed a sigmoid dose response curve. The most effective dose attracted 80–90% of the mosquitoes within 30 s and was as effective as the human hand. L‐(+)‐lactic acid was identified in this extract and found attractive for mosquitoes also when presented alone. Carbon dioxide stimulated taking flight and was attractive, an effect which was synergistically enhanced in combination with L‐(+)‐lactic acid. The presented bioassay is especially suited to test the behavioural effects of synthetic odours as well as of natural odour sources. Due to the fast response of the mosquitoes, the sensitivity, and the simplicity of the testing procedure it is a potent tool in the search for new attractive components.

Mating versus alternative blood sources as determinants to mosquito abundance and population resilience

Abstract:A deterministic nonlinear ordinary differential equation model for mosquito dynamics in which the mosquitoes can quest for blood either within a human population or within non-human/vertebrate populations is derived and studied. The model captures both the mosquito's aquatic and terrestrial forms and includes a mechanism to investigate the impact of mating on mosquito dynamics. The model uses a restricted form of homogeneous mixing based on the idea that the mosquito has a blood-feeding habit by accounting for the mosquitoes' blood-feeding preferences as well as its gonotrophic cycle. This characterization allows us to compartmentalise the total mosquito population into distinct compartments according to the spatial location of the mosquito (breeding site, resting places and questing places) as well as blood-fed status. Issues of overcrowding and intraspecific competition both within the aquatic and the terrestrial stages of the mosquito's life forms are addressed and considered in the model. Results show that the inclusion of mating induces bi-stability; a phenomenon whereby locally stable trivial and non-trivial equilibria co-exist with an unstable non-zero equilibrium. The local nature of the stable equilibria is demonstrated by numerically showing that the long-term state of the system is sensitive to initial conditions. The bi-stability state is analogous to the phenomenon of the Allee effect that has been reported in population biology. The model's results, including the derivation of the threshold parameter of the system, are comprehensively tested via numerical simulations. The output of our model has direct application to mosquito control strategies, for it clearly shows key points in the mosquito's developmental pathway that can be targeted for control purposes.

Mosquitoes escape looming threats by actively flying with the bow wave induced by the attacker

Abstract:To detect and escape looming threats, night-flying insects must rely on other senses than vision alone. Nocturnal mosquitoes can evade looming objects in the dark, but how they achieve this is still unknown. Here, we show how night-active female malaria mosquitoes escape from rapidly looming objects that simulate defensive actions of blood-hosts. First, we quantified the escape performance of flying mosquitoes from an event-triggered mechanical swatter, showing that mosquitoes use swatter-induced airflow to increase their escape success. Secondly, we used high-speed videography and deep-learning-based tracking to analyze escape flights in detail, showing that mosquitoes use banked turns to evade the threat. By combining escape kinematics data with numerical simulations of attacker-induced airflow and a mechanistic movement model, we unraveled how mosquitoes control these banked evasive maneuvers: they actively steer away from the danger, and then passively travel with the bow wave produced by the attacker. Our results demonstrate that night-flying mosquitoes can detect looming objects when visual cues are minimal, suggesting that they use attacker-induced airflow both to detect the danger and as a fluid medium to move with away from the threat. This shows that escape strategies of flying insects are more complex than previous visually induced escape flight studies suggest. As most insects are of similar or smaller sizes than mosquitoes, comparable escape strategies are expected among millions of flying insect species. The here-observed escape maneuvers are distinct from those of mosquitoes escaping from odor-baited traps, thus providing new insights for the development of novel trapping techniques for integrative vector management.
The Antenna Base Plays a Crucial Role in Mosquito Courtship Behavior

Abstract:Mosquitoes are vectors of diseases like malaria, dengue fever, yellow fever, chikungunya and Zika. For mosquito control it is crucial to understand their hearing system, as mosquitoes' courting behavior is mostly auditory. Many nonlinear characteristics of the mosquito hearing organ have been observed through behavioral studies and neural measurements. These enable mosquitoes to detect and synchronize to other mosquitoes. Many hypotheses concerning the role of the flagellum and the fibrillae of the antenna in mosquito hearing have been made, and neural processes have been considered as the origin of the nonlinearities. In this study we introduce a geometric model based on the morphology of the mosquito antenna base. The model produces many of the observed nonlinear characteristics, providing evidence that the base of the antenna plays a crucial role in mosquito hearing. Even without neural processing, the antenna response to sound produces behaviorally relevant cues that can inform about the presence, location and sex of other mosquitoes.

Abstract:Male mosquitoes form aerial aggregations, known as swarms, to attract females and maximize their chances of finding a mate. Within these swarms, individuals must be able to recognize potential mates and navigate the dynamic social environment to successfully intercept a mating partner. Prior research has almost exclusively focused on the role of acoustic cues in mediating the ability of male mosquitoes to recognize and pursue flying females. However, the role of other sensory modalities in this behavior has not been explored. Moreover, how males avoid collisions with one another in the dense swarm while pursuing females remains poorly understood. In this study, we combined free-flight and tethered flight simulator experiments to demonstrate that swarming Anopheles coluzzii mosquitoes integrate visual and acoustic information to track conspecifics and avoid collisions. Our tethered experiments revealed that acoustic stimuli gated mosquito steering responses to visual objects simulating nearby mosquitoes, especially in males that exhibited attraction to visual objects in the presence of female flight tones. Additionally, we observed that visual cues alone could trigger changes in wingbeat amplitude and frequency of mosquitoes. These findings were corroborated by our free-flight experiments, which revealed that mosquitoes modulate their flight responses to nearby conspecifics in a similar manner to tethered animals, allowing for collision avoidance within swarms. Together, these results demonstrate that both males and females integrate multiple sensory inputs to mediate swarming behavior, and for males, the change in flight kinematics in response to multimodal cues allows them to simultaneously track females while avoiding collisions.

Abstract:Automated misting systems are a convenient way for homeowners or small businesses to control adult mosquitoes. One such system was presented to the Anastasia Mosquito Control District (AMCD) for evaluation to control caged Aedes aegypti. The system consisted of 3 spray tanks, 2 pumps, water level sensors, and flow meters, and was controlled through an Android tablet loaded with dedicated control software. The evaluation of the system included calibration tests, droplet characterization, spray dispersion in the open field, and effectiveness testing using bio-assay cages for mortality assessment. For these tests, a loop of 14 nozzles 4 m apart was connected and held at 1 m height utilizing a total of 120 m tube. All nozzles were arranged in a 16 × 12 m rectangle laid in the East-West direction. Water was sprayed for calibration and droplet size measurements at pressures of 13.0, 15.5, and 18 bar; water and 10% red dye solution for spray dispersion at 18 bar pressure, and 0.17% solution of equalizer 20-20 was sprayed at 18 bar pressure for mortality tests. All 3 replicated tests were conducted in the morning between 9:00 and 11:30 am. During this time, temperature ranged from 21-26°C, relative humidity from 54-95 %, and wind speed from 0 - 2 km/hr. The combined flow rate from all 14 nozzles was significantly affected by pressure and was in agreement with the machine-calculated flow rate. There was a similar flow rate from all nozzles, indicated by a standard error of 0.82 mL/min. The droplet characteristics represented by DV0.1, DV0.5, and DV0.9 were not affected by nozzles but decreased with an increase in pressure as expected. The percentage of coverage on the cards, an indicator of spray dispersion, ranged from 20 -100 %, and it was found to increase in the direction of the wind. Mosquito mortality showed a similar trend of increasing in the wind direction and ranged from 30 to 100%. There was no effect of the location of cages on mosquito mortality. These results indicate that the effectiveness of this spray depends upon wind direction. The results, however, may be different when there is no wind, which may be the case during the times these applications are made.

Abstract:A 35-year-old female who suffered advanced gastric cancer (classified Borrmann type 3) with para-aortic lymph nodes swelling received chemotherapy preoperatively. These lymph nodes, which could be detected by abdominal CT scan, were highly suspected of metastasis from gastric cancer. The regimens of neoadjuvant chemotherapy were Mitomycin C (MMC) 8 mg/m2 and Cis-platinum (CDDP) 75 mg/m2, administered intravenously as one course. After she was given these two courses, we performed total gastrectomy, pancreatico-splenectomy, and lymph-adenectomy curatively. The patient has been maintained in an outpatient clinic for one and half years without any evidence of recurrence.
