Abstract:A simple and label-free electrochemical aptasensor for bisphenol A (BPA) determination was developed based on gold nanoparticles dotted graphene (GNPs/GR) nanocomposite film modified glassy carbon electrode (GCE). The electrochemical probe of ferricyanide was used to investigate the interactions between aptamer and BPA. The resulting GNPs/GR layer exhibited good current response for BPA detection. The highly conductive and biocompatible nanostructure of GNPs/GR nanocomposite was characterised by atomic force microscope (AFM), scanning electron microscopy (SEM) and cyclic voltammetry (CV). The peak current change (ΔI) of ferricyanide was linear with the concentration of BPA in the range from 0.01 μM to 10 μM with the detection limit of 5 nM. The proposed aptasensor is rapid, convenient and low-cost for effective sensing of BPA. Particularly, the aptasensor was applied successfully to determine BPA in milk products, and the average recovery was 105%.

Abstract:Bisphenol A (BPA) is an endocrine disrupting chemical having estrogenic properties that can enter the body through leaking from food containers or the food chain. An upconversion nanoparticle-based aptasensor integrated with microfluidic chip was simply designed to achieve the determination of BPA in foodstuff. Core-shell structured upconversion nanoparticles with 6.41-fold luminescence enhancement were used as fluorescence donors and gold nanoparticles as fluorescence acceptors to construct fluorescence resonance energy transfer (FRET) system. The BPA aptamer serves as a link between the building of the FRET system and the BPA-specific recognition component. The fishbone-structured microfluidic chip only needs 0.5 μL sample solution and 4 min to determine the presence of BPA. The proposed method has good linearity for BPA in the range of 0.02–100 ng/mL with calibration curve of y = 0.0918 x + 0.6477 and limit of detection of 0.014 ng/mL. It is tremendously competitive to perform on-chip analysis that enables microsampling and rapid determination.

Abstract:Bisphenol A (BPA) is a known endocrine disruptor and one of the most serious environmental contaminants, often present at low levels in various water sources. Therefore, it is very important and necessary to develop a fast, cost-effective, sensitive, and selective method for on-site detection of BPA. Herein, we developed a portable, evanescent, wave fiber-optic aptasensor for rapid, on-site detection of BPA with high sensitivity and selectivity. In this system, the probe DNA molecule, which is the complementary sequence of a small part of the BPA aptamer, was covalently immobilized onto the optical fiber sensor surface. Using an indirect competitive detection mode, samples containing different concentrations of bisphenol A were premixed with a given concentration of fluorescence-labeled BPA aptamer, which binds to bisphenol A with high specificity. Then, the sample mixture was pumped to the sensor surface, and a higher concentration of BPA led to less fluorescence-labeled BPA aptamer hybridized with surface immobilized probe DNA and thus to a lower fluorescence signal. The developed sensing system exhibits a sensitive response to BPA in the range of 2 nM to 100 nM with a low detection limit of 1.86 nM (0.45 ng ml(-1)) under the optimal conditions. The biosensors showed good reproducibility, stability, and selectivity for BPA detection. Finally, this proposed sensor was successfully employed to determine the presence of BPA in wastewater samples.

Abstract:Rapid and sensitive analysis of bisphenol A (BPA) is essential for preventing health risks to humans and animals. Hence, a signal-amplified electrochemical aptasensor without repetitive polishing and modification of working electrode was developed for BPA using Au-decorated magnetic reduced graphene oxide (Au/MrGO)-based recognition probe (RP) and DNA nanospheres (DNS)-based signal probe (SP) cooperative signal amplification. The DNS served as a signal molecule carrier and signal amplifier, while Au/MrGO acted as a signal amplifier and excellent medium for magnetic adsorption and separation. Moreover, utilizing the excellent magnetic properties of Au/MrGO eliminates the need for repetitive polishing and multi-step direct modification of the working electrode while ensuring that all detection processes take place in solution and that used Au/MrGO can be easily recycled. The proposed aptasensor exhibited not only good stability and selectivity, but also excellent sensitivity with a limit of detection (LOD) of 8.13 fg/mL (S/N = 3). The aptasensor's practicality was proven by spiking recovery tests on actual water samples and comparing the results with those detected by HPLC. The excellent sensitivity and selectivity make this aptasensor an alternative and promising avenue for rapid detection of BPA in environmental monitoring.

Abstract:Bisphenol A migrated from plastic products can function as an endocrine-disrupting compound, leading to serious diseases. The construction of rapid, cost-effective, and user-friendly sensors to accurately monitor BPA at ultra-low levels has great significance for protecting human health. Here, we proposed an aptasensor combined with long-period fiber gratings (LPFGs) for the label-free detection of BPA molecules. A long DNA aptamer against BPA molecules was selected and immobilized on the surface of the LPFG through chemical modification. When exposed to BPA solutions, the long aptamer provides a large conformation transition to amplify the perturbation of the refractive index near the fiber surface, which is conducive to producing a measurable shift of the transmission spectra and improving the sensitivity of the BPA detection. As a result, an ultralow detection limit of 0.232 ng/mL was achieved, which is lower than the standard limit (1.5 ng/mL) in drinking water set by European Union (EU). The proposed sensor offers an effective strategy for BPA detection, featuring the advantages of a simple structure, low cost, excellent reusability, high sensitivity, and reliability.

Abstract:Bisphenol A (BPA) is widely used in the manufacture of polycarbonate and epoxy-resin-based products, and BPA contamination often happens in a variety of types of environment and food stuffs. BPA can cause many harmful effects to health due to its high toxicity. The rapid detection of BPA is of great significance in environmental monitoring and food safety. Nucleic acid aptamers show advantages in biosensors due to good chemical stability, the ease of labeling functional groups, and target binding that induces conformation change. Here, we performed a thorough characterization of the binding performance of one 60-nt anti-BPA DNA aptamer with isothermal titration calorimetry (ITC). We found the crucial region of the aptamer sequence for affinity binding with BPA, and the aptamer was able to be truncated to 29-nt DNA without losing affinity. We then developed a simple reagent-less electrochemical aptamer-based sensor for rapid BPA detection with this engineered aptamer. The truncated aptamer with a redox tag methylene blue (MB) was immobilized on a gold electrode. BPA-binding induced the conformation change of the MB-labeled aptamer, moving the MB close to the electrode surface and causing a significant current increase in MB in square wave voltammetry (SWV). Under optimized conditions, we achieved the quantitative detection of BPA with a detection limit of BPA at 0.1 μM. This sensor showed quick response to BPA and could be regenerated by washing with deionized water. This sensor was selective, and it allowed detecting BPA in complex samples, showing its potential in practice. This study will help in further applications of the aptamers of BPA.

Abstract:We prepared a series of diverse metal ions-doped titanium-based metal-organic frameworks [Ti-MOFs(M2+), M2+ = Co2+, Fe2+, and Ni2+] and used them as novel scaffolds to construct aptasensors that can sensitively determine trace bisphenol A (BPA). Ti-MOF was prepared using 6-(5-carboxypyridin-2-yl) pyridine-3-carboxylic acid as building block, for which different metal ions (Co2 +, Fe2+, and Ni2+) were coordinated with N atoms bearing on pyridine of Ti-MOF. The series of Ti-MOFs(M2+) were composed of irregular nanoparticles, Ti clusters and mixed metal valence states of Co2+/Co3+, Fe2+/Fe3+ or Ni2+/Ni3+. A large amounts of BPA-targeted aptamer strands were anchored over the series of Ti-MOFs(M2+) via the combined interaction, such as sr-sr stacking, hydrogen bond, and metal-N coordination, leading to the excellent sensing ability toward BPA. Among different Ti-MOFs(M2+), the Ti-MOF(Co2+) exhibited a high electrochemical activity and strong stabilization ability for the aptamer-BPA complex. The corresponding aptasensor exhibited an ultra-low detection limit of 0.09 fg mL(-1) within the range of 1 fg mL(-1) to 1 ng mL(-1), along with high selectivity, good stability, and outstanding reproducibility. Further, it demonstrated the extensive serviceability in different environments, including milk, river water, and human serum. Abstract:A facile label-free aptamer-based electrochemical biosensor has been developed for sensitive detection of bisphenol A (BPA) based on a dual-signaling amplification strategy. The aptasensor was constructed by electrodeposition of gold nanoparticles (AuNPs) onto a glassy carbon electrode (GCE), where a thiolated-modified BPA aptamer was immobilized through self-assembly and then hybridized with the biotin-modified complementary DNA probe (cDNA) to form a double-stranded DNA. Upon the highly specific interaction between the target BPA and its aptamer, cDNA was released from the electrode surface and BPA was immobilized on the sensing interface. Streptavidin-modified horseradish peroxidase-functionalized gold nanoparticle (avidin-HRP-AuNP) was chosen as the nanoprobe, due to its catalytic activity to the oxidation of hydroquinone (HQ) in the presence of H2O2. As a result, the captured amounts of avidin-HRP-AuNP decrease with the increase of the BPA concentration and produce a series of decreasing catalytic peak currents. In addition, BPA has a redox activity and could provide an additional signal transformation. By superimposing the two signal changes, BPA was detected sensitively in a linear range from 0.001 to 1nM with a detection limit of 0.41pM. The aptasensor exhibited good selectivity toward BPA even in the presence of the interferents at 100-fold concentrations. This method would be readily applicable for sensitive detection of other redox analytes, merely by changing the anti-BPA aptamer/cDNA pair with a correspondent anti-target molecule aptamer and cDNA.

Abstract:Bisphenol A (BPA) is one of the environmental endocrine disruptors (EDCs), and BPA contamination in environment can cause high risks to human health. Rapid determination of BPA on sites is in high demand in environmental analysis. Taking advantage of aptamers as affinity ligands and fluorescence anisotropy (FA) analysis, we developed a simple and rapid FA assay for BPA by employing a single tetramethylrhodamine (TMR) labeled short 35-mer DNA aptamer against BPA. The assay is based on the BPA-binding induced conformation change of TMR-labeled aptamer and alteration of interaction between TMR and guanine bases, resulting in change of FA signals. We screened the FA change of aptamer probes having TMR label on a specific site of the aptamer upon BPA addition. The aptamer with a TMR label on the 22nd T base showed large FA-decreasing response to BPA and maintained good binding affinity to BPA. By using this TMR-labeled aptamer, we achieved FA detection of BPA with a detection limit of 0.5 μmol/L under the optimized conditions. This assay was selective towards BPA and enabled the detection of BPA spiked in tap water sample, showing the potential applications on water samples.

Abstract:In this study, a novel electrochemical sensor was constructed based on gold nanoparticles-MoS2 nanoflowers/ionic liquid functionalized graphene (AuNPs/MoS2-NFs/IL-graphene) nanocomposites for low level detection of bisphenol (BPA). The prepared nanocomposites were characterized by scanning electron microscopy, transmission electron microscopy, X-ray diffraction, X-ray photoelectron spectroscopy, and Fourier transform infrared spectroscopy. The prepared nanocomposites were also explored the effect of pH and scan rate to the results. The electrochemical performance of the sensor was then evaluated by cyclic voltammetry and differential pulse voltammetry, and it was showed that the sensor had prominent electrochemistry activity towards the BPA. Under the optimized experimental conditions, a large linear relationship in the range of 0.05–4.0 μM was detected between the peak current and the concentration of BPA, and the detection limit was 0.028 μM. The results showed that the sensor had high sensitivity, great selectivity, and excellent stability. Finally, the developed electrochemical sensor was successfully applied toward the determination of BPA in lake water samples and the satisfactory recoveries ranged from 95.7% to 105.4%. Thus, this sensor provided a promising candidate for BPA detection in the environmental water.

Abstract:We report a method for trace detection of bisphenol A (BPA) based on fluorescence resonance energy transfer (FRET) between lanthanide-doped KGdF4 nanoparticles (KGdF4: Tb3+ NPs) as a donor, and gold nanoparticles (AuNPs) that act as an acceptor. The KGdF4: Tb3+ NPs were modified with an aptamer recognizing BPA, and AuNPs were modified with the complementary DNA (cDNA) of the aptamer. The donor-acceptor pair was fabricated by hybridizing the aptamers and their cDNA. Thus, fluorescence of the KGdF4: Tb3+ NPs could be quenched due to the good overlap between the KGdF4: Tb3+ NP fluorescence emission and the AuNP absorption spectrum. However, on addition of BPA, the aptamers preferentially bound to BPA, forming KGdF4: Tb3+ NP-aptamer-BPA complexes. As a result, the FRET was disrupted, and the fluorescence was restored. The recovered fluorescence of the KGdF4: Tb3+ NPs was linearly proportional to the concentration of BPA in the range of 0.5 to 100 ng mL(-1), with detection limits as low as 0.16 ng mL(-1). The assay was applied to real samples, and the results were consistent with the results obtained using high performance liquid chromatography (HPLC) methods. We presume that this strategy can be extended to the detection of other contaminants by simply substituting the aptamer.

Abstract:To resolve the occurrence of unfulfillable detection in high-salts foods, we used fluorescence resonant energy transfer (FRET) sensors based on nanoparticle upconversion. In this study, we developed a novel FRET sensor for the detection of bisphenol A (BPA) in high-salt foods. We based this approach on the assembly of aptamer modified upconversion nanoparticles (DNA1-UCNPs) and complementary DNA modified metal organic frames (DNA2-MOFs), which possessed corresponding wavelength absorption. Targeting BPA signal transduction was performed using the BPA aptamer, via competitive recognition between the BPA analyte and complementary DNA sequences in a high-salt solution. Sensor adaption in high-salt samples was attributed to functional hydrophilic groups, modified in the MOFs, and the enhanced colloidal stability of these MOFs. The MOF-UCNP assembly displayed considerable analytical performance in terms of BPA detection, with a linear range of 0.1–100 nM, and a limit of detection (LOD) of 0.02 nM, in a 340 mM NaCl food sample (the energy drink, Gatorade). Thus, this method provides a solid basis for small molecules detection in high-salt foods.

A semi-covalent molecularly imprinted fluorescent sensor for highly specific recognition and optosensing of bisphenol A

Abstract:A novel mesoporous fluorescent molecularly imprinted sensor for selective detection of bisphenol A (BPA) in food materials was fabricated via a semi-covalent imprinting method. The imprinting precursor that served as an alternative template molecule for BPA was prepared via thermally reversible isocyanate bonding, which effectively improved the imprinting efficiency for the molecularly imprinted sensor. Carbon dots (CDs) were embedded in mesoporous silica as signal recognition elements that exhibited quenching upon BPA binding. Subsequently, through the sol-gel process, the molecularly imprinted layer was coated on the CDs silica layer and provided specific recognition sites for BPA. The composite of CDs embedded in the mesoporous molecularly imprinted polymer (CDs@MIP) was characterized with scanning electron microscopy, transmission electron microscopy, Fourier transform infrared spectroscopy, Brunauer-Emmett-Teller measurements and thermogravimetric analysis. The mechanism of carbon dots quenching and the high selectivity of CDs@MIP towards BPA were explored. The linear response range of the sensor was from 0.025 mg L-1 to 2 mg L-1 with a limit of detection of 0.016 mg L-1. The method was successfully applied for the determination of food samples and recoveries ranged from 92.5% to 101.1%. The BPA contents in actual samples were determined using high performance liquid chromatography and the proposed sensor, showing no significant difference between the two methods.

Abstract:Okadaic acid (OA) is one of the most common marine biological toxins, which can cause human poisoning or even death. It is desirable and essential to develop novel approaches for OA detection in a convenient, practical and low-cost manner. In this study, a novel piezoelectric aptasensor was designed for the detection of OA. The aptamers specifically binding with OA were used as the sensing elements. Quartz crystal micmbalance (QCM) device was used as the transducer for responsive signal detection. Gold nanoparticles (AuNPs) were used to amplify the detection signals. The results showed that the linear detection range of OA was 0.5 similar to 200 nM with a detection limit of 0.32 nM. This biosensor provides a novel approach for inexpensive, ultrasensitive, well-specific and label-free detection of OA as well as other marine biological toxins with minor modifications.

Abstract:Constructing advanced substrates with excellent features is promising for sensitive surface-enhanced Raman spectroscopy (SERS) detection. Here a novel capillary monolithic 3D structural-substrate SERS platform with Au@cDNA@Ag@Cyanine 3-aptamer nanoparticles (Au@cDNA@Ag@Cy3-Apt NPs) was fabricated for rapid, highly specific profiling of ultra-trace Bisphenol A (BPA). The proposed SERS platform combined both in-capillary SERS and aptamer-affinity recognition strategies, in which the superior SERS properties of Au–Ag NPs, aptamer selectivity, and the advantages of capillary monolith were integrated. A 3D hierarchically porous network was constructed in the monolithic column, which was endowed with rich hotspots for SERS, rapid sample permeation, and better analysis efficiency than most plane-shaped SERS modes. By varying the amount of Ag + precursor, the Ag-shell thickness on SERS was finely tuned to guarantee Cy3 label in proximity to the plasmonic surface. Based on the biorecognition of aptamer, the selective identification of BPA occurred and exhibited a significant change in SERS intensity without obvious interference. As a result, the monolithic SERS platform featured facile operation, excellent specificity, and rapid analysis (10 min, much less than the solution-based or planar substrate SERS modes). Ultra-high sensitivity and robust reproducibility for BPA analysis was achieved with a low limit of detection (LOD) at 9.12 × 10 - 4 ng/L. The feasibility of this SERS platform for monitoring BPA in water and milk samples was also validated. This work lights a new access to capillary monolithic SERS-sensing platform for ultrasensitive and specific analysis of BPA.

Abstract:A novel "signal-on" electrochemical aptasensor was developed for ultrasensitive and specific detection of BPA, using single-walled carbon nanotubes (SWCNT) as the electro-catalytic probe for further signal amplification. The BPA aptamer was then loaded onto the sensing platform by hybridization with complementary DNA (CDNA). In the presence of BPA it combines with the aptamer and the BPA-aptamer conjugate was released from the electrode;subsequently the added SWCNT and CDNA assembled quickly. Thus, the dual-amplification of the "signal-on" electrochemical aptasensor takes effect. The [Fe (CN)6]3−/4− redox probe signal (∆I) detected by DPV (differential pulse voltammetry) is proportional to the negative logarithm of BPA concentration between 10−19 M and 10−14 M. The detection limit is 0.08 aM. Importantly, the proposed biosensor represents a successful application for determination of BPA in human serum and lake water.

Abstract:In this paper, a novel bimetallic Fe–Cu metal–organic framework combined with 1,3,5-benzenetricarboxylic acid (Fe–Cu-BTC) are synthesized using hydrothermal reaction. The bimetallic Fe–Cu-BTC with high BET (1504 cm 3 g −1 ) and high Langmuir surface area (1831 cm 3 g −1 ) is composited by gold nanoparticles to improve the conductivity and to develop their synergistic effect. A novel bisphenol A (BPA) sensor was prepared by dropcasting Fe–Cu-BTC on glassy carbon electrodes (GCE) followed by AuNPs electrodeposition. The Fe–Cu-BTC framework were characterized by X-ray photoelectron spectroscopy (XPS), X-ray diffraction (XRD), scanning electron microscopy (SEM), transmission electron microscopy studies (TEM), FT-IR, BET measurements and EDX spectra. Cyclic voltammetry (CV) and differential pulse voltammetry (DPV) were carried out for surveying the electrochemical properties of the sensors and for the quantification of BPA. Two linear ranges of BPA concentrations 0.1–1.0 μM and 1.0–18 μM with 18 nM limit of detection were obtained. The developed sensor was used to measure the concentration of BPA in samples extracted from rain coat with the recovery ranging from 85.70 to 103.23%.

Abstract:Residue of sulfamethazine (SMZ), a typical short-acting drug to prevent bacterial infections, in food is a threat to human health. A ternary heterogeneous metal-organic framework hybrid (Zn/Fe-MOF@PDANSs) of Zn-TCPP-MOF, MIL-101 (Fe) and polydopamine nanoparticles (PDANSs) was proposed to establish an aptasensor for the sensitive and selective detection of SMZ. In this sensor, Zn-TCPP-MOF and FAM emitted fluorescence at 609 nm and 523 nm, respectively, and the fluorescence of FAM-ssDNA could be quenched when it was adsorbed on the surface of MOF hybrid. In the presence of SMZ, the fluorescence of FAM-ssDNA recovered due to the dropping from MOF hybrid, while the fluorescence of MOF hybrid remained. With this strategy, a wide concentration range and high sensitivity of SMZ were detection. And the ternary Zn/Fe-MOF@PDANSs sensor exhibited more excellent performance than binary Zn/Fe-MOF aptasensor. In addition, the sensor showed pleasurable selectivity, and was utilized for SMZ determination in authentic chicken and pork samples, implying the fascinating potential in practical application.
