Abstract:Dominance, semidominance, and recessiveness are important modes of Mendelian inheritance. The phytohormone gibberellin (GA) regulates many plant growth and developmental processes. The previously cloned semidominant GA-insensitive (GAI) genes Reduced height1 (Rht1) and Rht2 in wheat (Triticum aestivum) were the basis of the Green Revolution. However, no completely dominant GAI gene has been cloned. Here, we report the molecular characterization of Rht-B1c, a dominant GAI allele in wheat that confers more extreme characteristics than its incompletely dominant alleles. Rht-B1c is caused by a terminal repeat retrotransposons in miniature insertion in the DELLA domain. Yeast two-hybrid assays showed that Rht-B1c protein fails to interact with GA-INSENSITIVE DWARF1 (GID1), thereby blocking GA responses and resulting in extreme dwarfism and pleiotropic effects. By contrast, Rht-B1b protein only reduces interaction with GID1. Furthermore, we analyzed its functions using near-isogenic lines and examined its molecular mechanisms in transgenic rice. These results indicated that the affinity between GID1 and DELLA proteins is key to regulation of the stability of DELLA proteins, and differential interactions determine dominant and semidominant gene responses to GA.

Abstract:Pyrrolidine-amide oligonucleotide mimics (POMs) can cross-pair strongly with complementary parallel and antiparallel DNA and RNA targets in a sequence-specific fashion. As a result POMs have significant potential for applications including in vivo gene silencing, diagnostics and bioanalysis. To further modulate the DNA- and RNA-recognition properties and fine-tune the physiochemical properties of POMs for nucleic acid targeting, backbone-extended pyrrolidine-amide oligonucleotide mimics (bePOM I and II) were introduced. The bePOMs differ from the original POMs through the insertion of an additional methylene group into the backbone units, which increases the flexibility of the oligomers. bePOM I and II oligomers were synthesised using solid-phase peptide chemistry. Interestingly, UV thermal denaturation and circular dichroism studies reveals bePOM I and II can hybridise with complementary RNA, but not DNA.
Phenotypic and transcriptomic characterization of a wheat tall mutant carrying an induced mutation in the C-terminal PFYRE motif of RHT-B1b

Abstract:Background: As central regulators of the gibberellic acid (GA) signaling pathway in plants, DELLA proteins function as growth repressors and affect diverse biological processes. The wheat RHT-B1b and RHT-D1b semi-dwarfing alleles, which encode GA-insensitive DELLA proteins, have been widely adopted in modern wheat varieties to improve lodging tolerance and harvest index. However, the molecular mechanisms by which DELLA modulates these responses in wheat remain largely unknown. Results: We identified a tall tetraploid wheat mutant line carrying an induced missense mutation (E529K) in the PFYRE motif of RHT-B1b that partially suppressed the semi-dwarf phenotype. The height-increasing effect of RHT-B1bE529K relative to RHT-B1b (19 cm or 21% increase) was significantly smaller than the effect of RHT-B1a (33 cm or 34% increase) relative to RHT-B1b in the same field experiment. The RHT-B1bE529K mutation was also associated with length increases in coleoptiles, seedling shoots, and stem internodes relative to the RHT-B1b allele. We detected no significant differences in germination rate, seedling root length, tiller number, flag leaf size, spike length, or yield components. Using RNA-seq, we compared gene expression profiles of plants encoding RHT-B1b and RHT-B1bE529K in coleoptile, first leaf, and elongating peduncles. We detected limited overlap among tissues of the genes differentially regulated by the two genotypes, and more genes upregulated (77%) than downregulated (23%) in RHT-B1bE529K relative to RHT-B1b. These results suggest that the wheat DELLA protein affects the transcriptome in a tissue-specific manner and that the mutation mainly eliminates or reduces repression functions of the RHT-B1 protein. Our study identified distinct sets of potential DELLA direct or indirect target genes involved in cell wall and carbohydrate metabolisms, cell cycle/division, and hormone pathways. Conclusions: We identified the hypomorphic RHT-B1bE529K allele that confers an intermediate plant height and coleoptile elongation. This allele can be useful in rain-fed wheat breeding programs where the strong reduction in height and biomass associated with RHT-B1b has detrimental effects. Transcriptomic characterization of different tissues from the plants encoding RHT-B1bE529K and RHT-B1b provided valuable information for identifying DELLA downstream GA response genes in wheat.
Identification of a Novel Locus Qgw12/ospub23 Regulating Grain Shape and Weight in Rice (oryza Sativa L.)

Abstract:Key message A major quantitative trait locus (qGW12) for grain shape and weight has been isolated in rice, corresponding to LOC_Os12g17900/OsPUB23, and its encoded protein interacts with OsMADS1. Grain shape in rice is an important trait that influences both yield and quality. The primary determinants of grain shape are quantitative trait loci (QTLs) inherited from natural variation in crops. In recent years, much attention has been paid to the molecular role of QTLs in regulating grain shape and weight. In this study, we report the cloning and characterization of qGW12, a major QTL regulating grain shape and weight in rice, using a series of chromosome fragment substitution lines (CSSLs) derived from Oryza sativa indica cultivar 9311 (acceptor) and Oryza rufipogon Griff (donor). One CSSL line, Q187, harboring the introgression of qGW12, exhibited a significant decrease in grain-shape-related traits (including grain length and width) and thousand-grain weight compared to the cultivar 9311. Subsequent backcrossing of Q187 with 9311 resulted in the generation of secondary segregating populations, which were used to fine-map qGW12 to a 24-kb region between markers Seq-44 and Seq-48. Our data indicated that qGW12 encodes a previously unreported U-box type E3 ubiquitin ligase, designated OsPUB23, which exhibited E3 ubiquitin ligase activity. Overexpression of OsPUB23 in rice resulted in higher plant yield than the wild type due to an increase in grain size and weight. Conversely, loss of OsPUB23 function resulted in the opposite tendency. Yeast two-hybrid screening and split luciferase complementation assays revealed that OsPUB23 interacts with OsMADS1. The functional characterization of OsPUB23 provides new genetic resources for improving of grain yield and quality in crops.

Abstract:Dwarfing and semi-dwarfing are important agronomic traits that have great potential for the improvement of wheat yields. Rht12, a dominant gibberellic acid (GA)-responsive dwarfing gene from the gamma-ray-induced wheat mutant Karcagi 522M7K, is located in the long arm of chromosome 5A, which is closely linked with the locus Xwmc410. Rht12 is likely an ideal gene for GA biosynthesis and deactivation research in common wheat. However, information on the Rht12 locus and sequence is lacking. In this study, Rht12 significantly shortened stem cell length and decreased GA biosynthetic components. Using bulked segregant RNA-Seq, wheat 660k single nucleotide polymorphism chip detection, and newly developed simple sequence repeat markers, Rht12 was mapped to a 11.21-Mb region at the terminal end of chromosome 5AL, and was found to be closely linked with the Xw5ac207SSR marker with a 10.73-Mb fragment deletion in all of the homologous dwarfing plants. Transcriptome analyses of the remaining 483-kb region showed significantly higher expression of the TraesCS5A01G543100 gene encoding the GA metabolic enzyme GA 2-β-dioxygenase in dwarfing plants than in high stalk plants, suggesting that Rht12 reduces plant height by activating TaGA2ox-A14. Taken together, our findings will promote cloning and functional studies of Rht12 in common wheat.

Abstract:The utilization of reduced plant height genes Rht-B1b and Rht-D1b encoding homeologous DELLA proteins led to the wheat Green Revolution (GR). However, the specific functions of the GR genes in yield determination and the underlying regulatory mechanisms remained unknown. Here we validated that Rht-B1b as a representative of the GR genes affected plant architecture and yield component traits. The up-regulation of Rht-B1b reduced plant height, leaf size and grain weight, but increased tiller number, tiller angle, spike number per unit area (SN), and grain number per spike (GNS). Dynamic investigations showed that Rht-B1b increased SN by improving tillering initiation not outgrowth and enhanced GNS by promoting floret fertility. Rht-B1b reduced plant height by reducing cell size in the internodes, and reduced grain size or weight by decreasing cell number in the pericarp. Transcriptome analyses uncovered that Rht-B1b regulated many homologs of previously reported key genes for given traits and several putative integrators for different traits. These findings specify the pleiotropic functions of Rht-B1b in improving yield and broaden new insights into the regulatory mechanism underlying plant morphogenesis and yield formation.

Abstract:An immunotoxin was synthesized with pokeweed antiviral protein and an IgG1 monoclonal antibody directed against human B and pre-B cells. The B43 murine monoclonal antibody does not react with normal human bone marrow precursor cells. The immunotoxin bound to all Burkitt's lymphoma cell lines that were tested but not to human peripheral blood T cells. The ability of antibody-toxin conjugate to inhibit human lymphoblast cell lines was checked in a clonogenic assay system. The immunotoxin in the presence of chloroquine elicited 5.8 logs of killing of Burkitt's lymphoma cells (B-ALL). The efficient inhibition of clonogenic growth of B-ALL cells was not affected by the presence of normal bone marrow cells. The immunotoxin was not very toxic to pluripotent stem cells; less than 50% of the stem cells were lost under conditions where 5.6 logs of clonogenic lymphoma cells were eliminated from a 100-fold excess of normal marrow cells. Further, when assayed by long-term human bone marrow cultures, immunotoxin treatment did not result in a significant loss of pluripotent precursor cells.

Abstract:The introduction of dwarfing genes triggered a wave of "green revolution". A number of wheats dwarfing genes have been reported in previous studies, and only a small fraction of these have been applied to production practices. Therefore, the development of novel dwarfing genes for wheat is of great value. In this study, a novel dwarfing site, Rht-yz , identified in the Yanzhan mutation, is located on chromosome 4B (30-33MB) and its mechanism of action is different from that of Rht-B1b (C-T mutation), but whether it affects the Rht-B1a ( TraesCS4B02G043100 ) or other genes is unclear. Exogenously applied GA 3 experiments showed that Rht-yz is one of the gibberellin-insensitive dwarf genes. The effects of the dwarf gene Rht-yz on agronomic traits in wheat were evaluated in the field using Yanzhan, Yanzhan mutations, F 2:3 and F 3:4 lines. The results showed that Rht-yz improved lodging resistance by reducing plant height, increasing diameter, wall thickness and mechanical strength of the basal stem. In terms of yield traits, Rht-yz had negative effects on tiller number plant -1 , biomass plant -1 and yield plant -1 , but had no significant effect on harvest index, 1000-kernel weight and spike traits. In addition, Rht-yz significantly increased crude protein, wet gluten and starch content. Therefore, the rational use of the new dwarfing site Rht-yz has potential and value in dwarf wheat breeding.

Abstract:The application of dwarfing genes in wheat breeding effectively reduced plant height, improved lodging resistance and increased the harvest index. Although the gibberellin-responsive (GAR) dwarf gene Rht15 did not negatively affect coleoptile length or root traits while reducing the plant height, it reduced the yield plant −1 . The gibberellin-insensitive (GAI) dwarf gene Rht1 can increase wheat yield under favorable conditions, but it reduces coleoptile length and affects root traits. This study was conducted to investigate whether the interaction of Rht15 and Rht1 can alleviate their negative effects. To analyze the interaction of Rht15 and Rht1 , field experiments were conducted with the F 5 , F 6 and F 7 lines of the four genotypes, Rht15 + Rht1, Rht15, Rht1 and tall lines detected using Rht15 and Rht1 molecular markers, derived from Durox ( Rht15 ) and Kronos ( Rht1 ), by comparing their effects on seedling traits, plant height and internode length, lodging resistance, flag leaf, yield and quality traits in durum wheat. The results showed that although the interaction of Rht15 and Rht1 did not overcome the inhibitory effect of Rht1 on coleoptile growth, it alleviated the negative effect of Rht1 on maximum root length (+14.8%). The grain yield plant −1 (+7.2%) of the Rht15 lines was improved under the action of Rht1 but was still lower than that of the Rht1 lines (−12.7%). The Rht15 + Rht1 lines had the strongest effect on reducing plant height and center of gravity height (−42.2% and −41.5%), which significantly improved lodging resistance (+175%). The flag leaf area (+97.4%) and SPAD value (+11.6%, +9.6% and +10.6%) were also significantly increased. In addition, the coleoptile length of the Rht15 lines was not significantly different from that of the tall lines, the plant height and center of gravity height were significantly reduced (−30.5% and −34.0%), and the lodging resistance was significantly increased (+176.8%). The Rht1 lines had increased grain yield (+12.7%) due to an increased 1000-kernel weight (+12.9%) compared to the tall lines. In conclusion, the combination of Rht15 and Rht1 alleles enhanced the advantages and partially alleviated the defects in some traits when compared to the individual alleles, but the grain yield plant −1 was still lower than the Rht1 lines. This reflected the limitations of Rht15 , and further confirmed the yield advantages of Rht1 .

Abstract:Abstract Background The wheat dwarfing gene increases lodging resistance, the grain number per spike and harvest index. Dwarf Polish wheat ( Triticum polonicum L., 2n = 4x = 28, AABB, DPW), initially collected from Tulufan, Xinjiang, China, carries a semi-dwarfing gene Rht-dp on chromosome 4BS. However, Rht-dp and its dwarfing mechanism are unknown. Results Homologous cloning and mapping revealed that Rht-dp is the ‘Green Revolution’ gene Rht-B1b . A haplotype analysis in 59 tetraploid wheat accessions showed that Rht-B1b was only present in T. polonicum . Transcriptomic analysis of two pairs of near-isogenic lines (NILs) of DPW × Tall Polish wheat ( Triticum polonicum L., 2n = 4x = 28, AABB, TPW) revealed 41 differentially expressed genes (DEGs) as potential dwarfism-related genes. Among them, 28 functionally annotated DEGs were classed into five sub-groups: hormone-related signalling transduction genes, transcription factor genes, cell wall structure-related genes, reactive oxygen-related genes, and nitrogen regulation-related genes. Conclusions These results indicated that Rht-dp is Rht-B1b , which regulates pathways related to hormones, reactive oxygen species, and nitrogen assimilation to modify the cell wall structure, and then limits cell wall loosening and inhibits cell elongation, thereby causing dwarfism in DPW.

Abstract:Reducing plant height (PH) is one of the core contents of the "Green Revolution", which began in the 1960s in wheat. A number of 27 reduced-height ( Rht ) genes have been identified and a great number of quantitative trait loci (QTLs) for PH have been mapped on all 21 chromosomes. Nonetheless, only several genes regulated PH have been cloned. In this study, we found the interval of QTL QPh-1B included an EST-SSR marker swes1079 . According to the sequence of swes1079 , we cloned the TaOSCA1.4 gene. We developed a CAPS marker to analyze the variation across a natural population. The result showed that the PH was significantly different between the two haplotypes of TaOSCA1.4–1B under most of the 12 environments and the average values of irrigation and rainfed conditions. This result further demonstrated that TaOSCA1.4 was associated with PH. Then, we validated the TaOSCA1.4 via RNAi technology. The average PHs of the wild-type (WT), RNAi lines 1 (Ri-1) and 2 (Ri-2) were 94.6, 83.6 and 79.2 cm, respectively, with significant differences between the WT and Ri-1 and Ri-2. This result indicated that the TaOSCA1.4 gene controls PH. TaOSCA1.4 is a constitutively expressed gene and its protein localizes to the cell membrane. TaOSCA1.4 gene is a member of the OSCA gene family, which regulates intracellular Ca 2+ concentration. We hypothesized that knock down mutants of TaOSCA1.4 gene reduced regulatory ability of Ca 2+ , thus reducing the PH. Furthermore, the cell lengths of the knock down mutants are not significantly different than that of WT. We speculate that TaOSCA1.4 gene is not directly associated with gibberellin (GA), which should be a novel mechanism for a wheat Rht gene.

Abstract:World wheat grain yields increased substantially in the 1960s and 1970s because farmers rapidly adopted the new varieties and cultivation methods of the so-called ‘green revolution’ 1 , 2 , 3 , 4 . The new varieties are shorter, increase grain yield at the expense of straw biomass, and are more resistant to damage by wind and rain 3 , 4 . These wheats are short because they respond abnormally to the plant growth hormone gibberellin. This reduced response to gibberellin is conferred by mutant dwarfing alleles at one of two Reduced height-1 ( Rht-B1 and Rht-D1 ) loci 4 , 5 . Here we show that Rht-B1/Rht-D1 and maize dwarf-8 ( d8 ) 6 , 7 are orthologues of the Arabidopsis Gibberellin Insensitive ( GAI ) gene 8 , 9 . These genes encode proteins that resemble nuclear transcription factors and contain an SH2-like 10 domain, indicating that phosphotyrosine may participate in gibberellin signalling. Six different orthologous dwarfing mutant alleles encode proteins that are altered in a conserved amino-terminal gibberellin signalling domain. Transgenic rice plants containing a mutant GAI allele give reduced responses to gibberellin and are dwarfed, indicating that mutant GAI orthologues could be used to increase yield in a wide range of crop species.

Abstract:Plant height (PH) plays a pivotal role in plant morphological architecture and is associated with yield potential in wheat. For the quantitative trait locus (QTL) analysis, a recombinant inbred line population was developed between varieties differing significantly in PH. Two major QTL were identified on chromosomes 4B (QPh.sicau-4B) and 6D (QPh.sicau-6D) in multiple environments, which were then validated in two different backgrounds by using closely linked markers. QPh.sicau-4B explained 10.1–21.3% of the phenotypic variance, and the location corresponded to the dwarfing gene Rht-B1. QPh.sicau-6D might be a novel QTL for PH, explaining 6.6–13.6% of the phenotypic variance and affecting spike length, thousand-kernel weight, and spikelet compactness. Three candidate genes associated with plant growth and development were identified in the physical interval of QPh.sicau-6D. Collectively, we identified a novel stable and major PH QTL, QPh.sicau-6D, which could aid in the development of closely linked markers for marker-assisted breeding and cloning genes underlying this QTL.

Abstract:Spike compactness (SC) is strongly associated with wheat (Triticum aestivum L.) grain yield. In this study, we conducted a quantitative trait locus (QTL) analysis using a doubled haploid (DH) population derived from a cross between two common wheat varieties with contrasting spike morphology, revealing 16 stable QTLs associated with SC. The effect of a major QTL, QSc.cau-6B.1, was validated in 231 F7 recombinant inbred lines (RILs) derived from the same cross as the DH population. Using two residual heterozygous lines (RHLs), we delimited QSc.cau-6B.1 to an approximately 0.5-Mbp physical interval containing four high-confidence genes. The tetratricopeptide repeat-TraesCS6B03G1214400 (TaTPR-B1) was the priority candidate gene according to sequence and expression variations between near-isogenic lines. Accordingly, TaTPR-B1 knockout in the common wheat variety 'CB037' significantly increased SC compared to the wild type (WT). Conversely, TaTPR-B1 overexpression in the common wheat variety 'Fielder' significantly decreased SC compared to the WT. Moreover, we developed a PCR-based marker targeting the 32-bp insertion/deletion (InDel) between the two TaTPR-B1 alleles, which could be practical and valuable in modern wheat breeding programs for diagnostic purposes. Collectively, these findings provide insight into the genetic basis of SC in common wheat and present a valuable target with a breeder-friendly diagnostic marker for gene pyramid breeding.

Abstract:Severe visual disturbance associated with lesions of the optic nerve developed in a Wilson's disease patient who had been receiving DL-penicillamine therapy for more than 2 years. Prompt relief was achieved with pyridoxine hydrochloride. These results suggest that the optic nerve lesions resulted from a penicillamine-induced pyridoxine deficiency. Furthermore, results of urinary xanthurenic acid excretion studies made on the patient before and after tryptophan loading tests, which were conducted during periods of no therapy, penicillamine therapy, and penicillamine plus pyridoxine therapy, indicate an alteration in pyridoxine metabolism in the presence of penicillamine. Although no other similar cases have been described, the present results indicate the possibility that patients on prolonged high dosage levels of penicillamine may require pyridoxine supplementation of their diet.
