Abstract:We reported important marker‐trait associations for sustainable grain production under drought and heat stress at anthesis. Likewise, the mechanisms of drought and heat resilience including gene expression and physiological traits (activities of carbohydrate metabolic and antioxidant enzymes, and endogenous hormonal responses) were explored. Moreover, identified pre‐breeding traits and genotypes can be utilized in breeding new wheat cultivars resilient to future adverse environments. Extreme weather including heat waves and drought episodes are expected to increase in intensity and duration due to climate change. Wheat, being a major crop is under extreme threat to these stresses especially at the reproductive stage. This review addresses the potential of diverse wheat germplasm (originated from landraces and synthetic derivatives) to cope with drought and heat stress at the flowering stage. Here, important marker‐trait associations were reported for sustainable grain production under drought and heat stress at anthesis. Likewise, the mechanisms of drought and heat resilience including gene expression and physiological traits (activities of carbohydrate metabolic and antioxidant enzymes, and endogenous hormonal responses) were explored. These studies helped to understand the genetic and physiological basis of drought and heat tolerance and certain pre‐breeding traits related to osmotic adjustment, phytohormonal regulation, antioxidant metabolism, and the expression of novel genes were identified. Moreover, identified pre‐breeding traits and genotypes can be utilized in breeding wheat cultivars resilient to future adverse environments.

Abstract:Background: As a result of the world population and climate change impact increases (especially in arid environments), there is a critical need for high-yield, drought-tolerant wheat. Synthetic hexaploid wheat derived lines (SHW-DL), were created artificially by crossing different durum wheat cultivars (AABB) with accessions of Aegilops tauschii (DD), a beneficial source of new genes for common bread wheat (Triticum aestivum L). Here, we studied the response of a panel of 91 SHW-DL for drought tolerance based on physiological, antioxidant enzyme activities, and drought tolerance indices. Results: A wide range of variation and high values of heritability observed for grain yield, physiological and antioxidant traits indicating that the SHW-DL panel constitutes a valuable gene source for drought tolerance improvement of wheat. Despite decreases in grain yield (YLD), leaf area index (LAI), and relative water content (RWC) an increase in the content of malondialdehyde (MDA) was observed. Moreover, drought streass increased the antioxidant enzyme activities of ascorbate peroxidase (APX), catalase (CAT) and peroxidase (POD), and also photosynthetic pigments, proline (Pro), and MDA content. With higher values of grain yield, physiological and biochemical traits such as photosynthetic pigments, and RWC, and lower content of MDA, and peroxidase (H2O2) activity, SHW-DL performed better compared to common wheat lines under water stress conditions. Conclusions: Different responses to water stress within the germplasm and between synthetic and common wheat suggest that selection for adaptive and suitable genotypes is possible for drought tolerance in synthetic wheat germplasm. Genotypes 54, 98, 102, 105, 122, 124, 143, 159, 196, and 198 were identified to be directly used in breeding programs or indirectly by crossing them with other wheat germplasm collections.

Abstract:The present study was conducted to evaluate the effect of a stay-green trait to stabilize the yield responses under combined heat and drought stress on wheat. Ten Recombinant Inbred Lines (RILs) of wheat along with their parents (HI1500/DBW 43) were evaluated under timely sown with irrigation (control) and without irrigation (drought stress), and late sown with irrigation (heat stress) and without irrigation (combined heat and drought stress). We observed that combined stress of drought and heat stress was more detrimental than the individual stresses for plants. GCP 6 and GCP 33 RILs showed 50.08% and 49.52% while GCP 23 and GCP 30 showed 61.05% and 62.17% decrease in yield under combined stress as compared to the control condition. We found that GCP 6 and GCP 33 RILs stabilized their yield by maintaining stay-green traits such as retaining a higher amount of chlorophyll, photosynthesis rate and leaf area duration at the anthesis stage under combined stress as compared to other RILs. Further, Real-Time gene expression data inferred that GCP 6 and GCP 33 RILs maintained stay-green traits by up-regulating TaCHLD as well as down-regulating TaPaO, TaPPH and TaSGR1 genes at anthesis stage under combined stress, that are known chlorophyll biosynthesis (TaCHLD) and degradation genes respectively. It was also revealed that GCP 6 and GCP 33 RILs maintained higher photosynthesis rate during the anthesis stage under combined stress due to increased carbon fixation resulting from higher TaRbcs, PsbP and TaPsbA genes expression. This study showed that stay-green traits help in stabilizing the yield attributes under combined heat and drought stress in wheat.

Abstract:Durum wheat performance in the Mediterranean climate is limited when water scarcity occurs before and during anthesis. The present research was performed to determine the effect of drought stress on several physiological and agro-morphological traits in 17 durum wheat genotypes under two conditions (control and drought) over two years. The results of analysis of variance indicated that the various durum wheat genotypes responded differently to drought stress. Drought stress significantly reduced the grain filling period, plant height, peduncle length, number of spikes per plot, number of grains per spike, thousand grains weight, grain yield, biomass, and harvest index in all genotypes compared to the control condition. The heatmap-based correlation analysis indicated that grain yield was positively and significantly associated with phenological characters (days to heading, days to physiological maturity, and grain filling period), as well as number of spikes per plant, biomass, and harvest index under drought conditions. The yield-based drought and susceptible indices revealed that stress tolerance index (STI), geometric mean productivity (GMP), mean productivity (MP), and harmonic mean (HM) were positively and significantly correlated with grain yields in both conditions. Based on the average of the sum of ranks across all indices and a three-dimensional plot, two genotypes (G9 and G12) along with the control variety (G1) were identified as the most tolerant genotypes. Among the investigated genotypes, the new breeding genotype G12 showed a high drought tolerance and yield performance under both conditions. Hence, this genotype can be a candidate for further multi-years and locations test as recommended for cultivation under rainfed conditions in arid and semi-arid regions.

Abstract:Stomatal density (SD) and stomatal index (SI) on the abaxial flag leaf epidermis, gas exchange of flag leaves and yield of six wheat ( Triticum aestivum L.) cultivars differing in drought resistance were studied in plant growth chamber. There were significant differences among cultivars in SD, SI, epidermal cell density (ED), photosynthetic rate (P n ), transpiration rate (E), instantaneous transpiration efficiency (ITE), shoot dry weight yield (Ys), grain yield (Yd) and whole growth season water use efficiency (WUE), but not in stomatal conductance (g s ). Wheat cultivars with moderate drought resistance had higher P n and Yd than cultivars with strong or weak drought resistance. SD was negatively correlated with ITE (r=−0.85) and WUE (r=−0.84), but not with P n , E and Yd. There were significant relationships between ED and Yd (r=−0.92), between SI and P n (r=0.84). These results revealed that breeding wheat cultivar only to raise drought resistance might cause yield decrease when water stress was not too severe. Wheat cultivars with lower SD might improve ITE and WUE, but P n and yield (Ys and Yd) were not influenced significantly. Greater ED was not beneficial to Yd. SI might be a criterion to use in selecting parents for producing high-yielding wheat cultivars.

Abstract:Drought and heat stress are potential problems that can reduce wheat yield, particularly during the terminal growth stages in arid and semiarid regions of the world. The current study intended to examine the impact of individual and combined drought and heat stress on the biochemical contents (antioxidant enzymes, proline, soluble proteins, and soluble sugars), physiological parameters (chlorophyll content, cell membrane stability, photosynthesis, stomatal conductance, and transpiration), plant–water relations (relative water content, water potential, osmotic potential, and pressure potential), agronomic traits (flag leaf area, plant height, number of tillers per plant, spike length, grains per spike, and thousand-grain weight), and gene expression ( TaHSF1a, TaWRKY-33, TaNAC2L , and TaGASR1 ) in four different thermostable and drought-tolerant wheat genotypes (i.e., Gold-16, HS-240, Suntop, and Hemai-13) collected from different countries. The tri-replicate experiment was conducted using two factorial arrangements in a randomized complete block design (RCBD). All measured traits, except total soluble sugars, proline, and cell membrane stability index, showed significant reduction under both combined and individual treatments. Furthermore, correlation analysis revealed a significant association between biochemical and physiological characteristics and crop agronomic productivity. Furthermore, principal component analysis (PCA) and heatmap analysis demonstrated significant levels of variation in traits according to the type of stress and nature of wheat genotype. The spectrographs and micrographs generated by scanning electron microscopy for the selected high- and low- tolerance samples revealed clear differences in mineral distribution and starch granulation. All studied genes showed comparatively high levels of relative expression under combined treatments of drought and heat stress in all wheat genotypes, but this expression was the highest in 'Gold-16' followed by 'HS-240', 'Suntop', and 'Hemai-13'. Overall, this study concluded that plants are proactive entities and they respond to stresses at all levels; however, the tolerant plants tend to retain the integrity of their biochemical, physiological, and molecular equilibrium.

Abstract:In wheat, water deficit during meiosis of pollen mother cells greatly reduces seed set and grain number. A promising option to avoid grain losses and maintain wheat productivity under water stress is to exploit conservative water-use strategies during reproduction. In this work, two cultivars known to be adapted to different environments were studied. Water stress, with or without a polymer spray known to reduce stomatal conductance, was applied to both cultivars just prior to meiosis. Two experiments were carried out in a phenotyping platform to (1) assess and validate daily non-destructive estimation of projected leaf area and to (2) evaluate different water-use (WU) strategies across the meiotic period and their effect on physiology and yield components. Gladius displays an elevated breakpoint (BP) in the regression of WU against fraction of transpirable soil water (FTSW) for both daily and night-time WU suggesting higher conservative whole-plant response when compared to Paragon. At the same time, Gladius maintained flag leaf gas-exchange with a significant reduction at ~ 0.2 FTSW only, suggesting an uncoupled mechanism of WU reduction that optimized the water resource available for flag leaf gas-exchange maintenance. Abstract:The decrease in water resources due to the excessive use of water for irrigation purpose and climatic changes represents a serious world-wide threat to food security. In this regards, 50 wheat accessions were analyzed, using completely random factorial design at the seedlings stage under normal and drought stress conditions. Significant variation was detected among all accessions under both conditions. All characters studied showed variations in the mean values in water deficit environments in studied gemplasm at seedling stage. As seedling fresh weight, dry weight, relative water content, cell membrane thermo-stability, chlorophyll a & b were positively associated among themselves under drought conditions which showed the significance of these attribute for water deficit areas in future wheat breeding programs. Based on their performance, five accessions namely G-1, G-6, G-11, G-16 and G-21 were selected as drought tolerant and threes accessions namely G-3, G15 and G-25 were selected as drought susceptible. The choice of wheat accessions based on the characteristics of the seedlings is informal, low-priced and less hassle. Likewise, the seedlings attributes exhibit moderate to high variation with an additive genetics effects on the environments. Best performance accessions under water deficit environment will be beneficial in future wheat breeding schemes and early screening for the attributes suggested in current experiment will be useful for producing best-yielded and drought-tolerance wheat genotypes to sustainable food security.

Abstract:AbstractDrought poses a severe threat to global wheat production and world food security. This study was conducted to evaluate bread wheat genotypes for drought tolerance based on different morphological, biochemical, and physiological traits. The responses of 25 bread wheat genotypes of Pakistani origin were evaluated based on morphological, biochemical, and physiological traits under optimal (70% water holding capacity (WHC)) and drought stress (35% WHC) conditions. Sufficient genotypic variability was noted in the tested genotypes on the morphological (shoot length, relative leaf water contents, and seedling dry weight), biochemical (membrane injury index, leaf free proline, and total soluble phenolic contents), and physiological traits (chlorophyll meter value, intercellular CO2 concentration (Ci), stomatal conductance (gs), photosynthetic rate (A), ratio of photosynthetic rate and intercellular CO2 concentration (A/Ci), transpiration rate (T), and instantaneous water use efficiency (WUEins)) under both control and drought stress conditions. The principal component analysis (PCA) under drought stress showed 78.3% variability of morpho-biochemical traits and 67.0% of physiological traits. Bi-plot of PCA showed the genotypes SQU-2, SQU-4, SQU-10, and SQU-11 are tolerant under drought stress and grouped based on high A, and A/Ci. Similarly, the genotypes SQU-8, SQU-9, SQU-14, SQU-15, SQU-16, and SQU-24 were also tolerant and grouped based on high WUEins and Ci under drought stress. In conclusion, significant genotypic variability was found among the tested wheat genotypes based on morphological, biochemical, and physiological traits, which can be used in future breeding programs to develop drought-tolerant bread wheat genotypes. The genotypes SQU-9, SQU-10, SQU-7, and SQU-13 had better growth under drought stress than other genotypes due to the accumulation of free proline and soluble phenolics which helped maintain tissue water status and sustained the photosynthesis.

Abstract:Drought is one of the main environmental factors affecting crop growth, and breeding drought-tolerant cultivars is one of the most economic and effective ways of increasing yields and ensuring sustainable agricultural production under drought stress. To facilitate the breeding of drought-tolerant wheat, this study was conducted to evaluate genotypic differences in the drought tolerance of 334 wheat genotypes collected from China and Australia with the aim of screening for drought-tolerant and -sensitive genotypes and to elucidate the corresponding physiological mechanisms. A hydroponic-air experiment (roots exposed to air for 7 h/d and continued for 6 d) showed significant genotypic differences in shoot and root dry weights among the genotypes. The relative shoot and root dry weights, expressed as the percentage of the control, showed a normal distribution, with variation ranges of 20.2–79.7% and 32.8–135.2%, respectively. The coefficients of variation were in the range of 18.2–22.7%, and the diversity index was between 5.71 and 5.73, indicating a rich genetic diversity among the wheat genotypes for drought tolerance. Using phenotypic differences in relative dry weights in responses to drought stress, 20 of each of the most drought-tolerant and drought-sensitive genotypes were selected; these were further evaluated in pot experiments (watering withheld until the soil moisture content reached four percent). The results showed that the trends in drought tolerance were consistent with the hydroponic-air experiment, with genotypes W147 and W235 being the most drought-tolerant and W201 and W282 the most sensitive. Significant genotypic differences in water use efficiency in response to drought were observed in the pot experiment, with the drought-tolerant genotypes being markedly higher and the two sensitive genotypes being no different from the control. A marked increase in bound water content in the drought stress plants was observed in the two drought-tolerant genotypes, while a decrease occurred in the free water. The reductions in photochemical efficiencies of PSII, transpiration rates, net photosynthesis rates, chlorophyll contents and stomatal conduction in the drought-sensitive genotypes W201 and W282 under drought stress were higher than the two tolerant genotypes. This study provides a theoretical guide and germplasm for the further genetic improvement of drought tolerance in wheat.

Abstract:Abstract Background: To improve our understanding about the physiological mechanism of grain yield reduction at anthesis, three spring wheat genotypes [L 1 (advanced line), L 2 (Vorobey) and L 3 (Punjab-11)] having contrasting yield potential under drought in field were investigated under controlled greenhouse conditions., drought stress was imposed at anthesis stage by withholding irrigation until all plant available water was depleted, while well-watered control plants were kept at 95% pot water holding capacity. Results: Compared to genotype L 1 and L 2 , pronounced decrease in grain number (NGS), grain yield (GY) and harvest index (HI) were found in genotype L 3 , mainly due to its greater kernel abortion (KA) under drought. A significant positive correlation of leaf monodehydroascorbate reductase (MDHAR) with both NGS and HI was observed. In contrast, significant negative correlations of glutathione S-transferase (GST) and vacuolar invertase (vacInv) both within source and sink with NGS and HI were found. Likewise, a significant negative correlation of leaf abscisic acid (ABA) with NGS was noticed. Moreover, leaf aldolase and cell wall peroxidase (cwPOX) activities were significantly and positively associated with thousand kernel weight (TKW). Conclusion: Distinct physiological markers correlating with yield traits and higher activity of leaf aldolase and cwPOX may be chosen as predictive biomarkers for higher TKW. Also, higher activity of MDHAR within the leaf can be selected as a predictive biomarker for higher NGS in wheat under drought. Whereas, lower activity of vacInv and GST both within leaf and spike can be selected as biomarkers for higher NGS and HI. The results highlighted the role of antioxidant and carbohydrate-metabolic enzymes in the modulation of source-sink balance in wheat crops, which could be used as bio-signatures for breeding and selection of drought-resilient wheat genotypes for a future drier climate.
Favorable Alleles for Stem Water-Soluble Carbohydrates Identified by Association Analysis Contribute to Grain Weight under Drought Stress Conditions in Wheat

Abstract:Drought is a major environmental constraint to crop distribution and productivity. Stem water-soluble carbohydrates (WSC) buffer wheat grain yield against conditions unfavorable for photosynthesis during the grain filling stage. In this study, 262 winter wheat accessions and 209 genome-wide SSR markers were collected and used to undertake association analysis based on a mixed linear model (MLM). The WSC in different internodes at three growth stages and 1000-grain weight (TGW) were investigated under four environmental regimes (well-watered, drought stress during the whole growth period, and two levels of terminal drought stress imposed by chemical desiccation under the well-watered and drought stress during the whole growth period conditions). Under diverse drought stress conditions, WSC in lower internodes showed significant positive correlations with TGW, especially at the flowering stage under well-watered conditions and at grain filling under drought stress. Sixteen novel WSC-favorable alleles were identified, and five of them contributed to significantly higher TGW. In addition, pyramiding WSC favorable alleles was not only effective for obtaining accessions with higher WSC, but also for enhancing TGW under different water regimes. During the past fifty years of wheat breeding, WSC was selected incidentally. The average number of favorable WSC alleles increased from 1.13 in the pre-1960 period to 4.41 in the post-2000 period. The results indicate a high potential for using marker-assisted selection to pyramid WSC favorable alleles in improving WSC and TGW in wheat.

Abstract:Abstract This study aims to evaluate the role of genetic diversity originated from three-way crosses of landraces and synthetic bread wheat derivatives attempted with elite cultivars to identify the genotypes having better grain yield as well as better starch contents under flowering stage drought stress. In total, 50 genotypes comprised of 47 derivatives of landraces and synthetics, and three standard varieties were grown in the fields of CIMMYT, Obregon following alpha lattice design with two replications. Drought stress was imposed by stopping the irrigation before anthesis and post-anthesis stages. Different traits related to physiological adaptations, earliness, grain yield and starch contents were decreased under drought stress except amylose which increased significantly. Canopy temperature depression was not significantly affected by drought stress. Further, we identified strong correlation of grain yield with plant height, normalized difference in vegetation index, plant maturity, spike length, grain number and thousand grain weight but negative with canopy temperature depression. Non-significant correlation of amylose and amylopectin was recorded with grain yield. Results revealed that genetic diversity derived from landraces and synthetic bread wheat can improve grain yield and starch contents but simultaneous improvement of both is elusive.

Abstract:Abstract Background To improve our understanding about the physiological mechanism of grain yield reduction at anthesis, three spring wheat genotypes [L 1 (advanced line), L 2 (Vorobey) and L 3 (Punjab-11)] having contrasting yield potential under drought in field were investigated under controlled greenhouse conditions, drought stress was imposed at anthesis stage by withholding irrigation until all plant available water was depleted, while well-watered control plants were kept at 95% pot water holding capacity. Results Compared to genotype L 1 and L 2 , pronounced decrease in grain number (NGS), grain yield (GY) and harvest index (HI) were found in genotype L 3 , mainly due to its greater kernel abortion (KA) under drought. A significant positive correlation of leaf monodehydroascorbate reductase (MDHAR) with both NGS and HI was observed. In contrast, significant negative correlations of glutathione S-transferase (GST) and vacuolar invertase (vacInv) both within source and sink were found with NGS and HI. Likewise, a significant negative correlation of leaf abscisic acid (ABA) with NGS was noticed. Moreover, leaf aldolase and cell wall peroxidase (cwPOX) activities were significantly and positively associated with thousand kernel weight (TKW). Conclusion Distinct physiological markers correlating with yield traits and higher activity of leaf aldolase and cwPOX may be chosen as predictive biomarkers for higher TKW. Also, higher activity of MDHAR within the leaf can be selected as a predictive biomarker for higher NGS in wheat under drought. Whereas, lower activity of vacInv and GST both within leaf and spike can be selected as biomarkers for higher NGS and HI. The results highlighted the role of antioxidant and carbohydrate-metabolic enzymes in the modulation of source-sink balance in wheat crops, which could be used as bio-signatures for breeding and selection of drought-resilient wheat genotypes for a future drier climate.

Abstract:Heat and drought are among the major obstacles confronting crop production under climate change. The present study was conducted to evaluate 50 diverse wheat genotypes for cell membrane stability (CMS) and chlorophyll content at seedling and anthesis stages under heat and drought stress conditions, to understand the effect of the two abiotic factors and to find promising genotypes for future breeding. Experiments were conducted in the glasshouse (seedling stage) and the field (anthesis stage). Analysis of variance showed significant variation (P ≤ 0.05) for all of the traits at seedling and anthesis stages. High levels of broad-sense heritability and genetic advance at 5% selection intensity indicated the presence of a high genetic component of variation and potential for genetic improvement through selection among the existing genetic variation. CMS showed a significant positive correlation with 1000-grain weight (TGW) under heat and drought conditions at both seedling and anthesis stages. Chlorophyll a/b ratio at seedling stage exhibited a significant negative correlation (r = –0.39, P < 0.05) with TGW under heat stress. Total chlorophyll content was significantly (r = 0.42, P < 0.05) correlated with TGW under heat stress at anthesis. Genotypes ETAD248 and ETAD7 showed the highest CMS and TGW values, whereas their chlorophyll a/b values were lowest, at both seedling and anthesis stages under heat and drought stress conditions. Higher CMS and total chlorophyll content, and lower chlorophyll a/b, were found to be useful indicators to identify genotypes with high TGW under heat and drought stress conditions. This study indicated the possibility of using seedling resistance as an indicator for later stage response in breeding for heat and drought resistance. The resistant genotypes identified can be used as potential germplasm in breeding programs.
Genetic and morpho-physiological analyses of the tolerance and recovery mechanisms in seedling stage spring wheat under drought stress

Abstract:Drought is one of the complex abiotic stresses that affect the growth and production of wheat in arid and semiarid countries. In this study, a set of 172 diverse spring wheat genotypes from 20 different countries were assessed under drought stress at the seedling stage. Besides seedling length, two types of traits were recorded, namely: tolerance traits (days to wilting, leaf wilting, and the sum of leaf wilting), and recovery traits (days to regrowth, regrowth biomass, and drought survival rate). In addition, tolerance index, recovery index, and drought tolerance index (DTI) were estimated to select the most drought tolerant genotypes. Moreover, leaf protein content (P), amino acid (AM), proline content (PRO), glucose (G), fructose (F), and total soluble carbohydrates (TSC) were measured under control and drought conditions to study the changes in each physiological trait due to drought stress. All genotypes showed a high significant genetic variation in all the physio-morphological traits scored under drought stress. High phenotypic and genotypic correlations were found among all seedling morphological traits. Among the studied indices, the drought tolerance index (DTI) had the highest phenotypic and genotypic correlations with all tolerance and recovery traits. The broad-sense heritability (H 2 ) estimates were high for morphological traits (83.85–92.27), while the physiological traits ranged from 96.41 to 98.68 under the control conditions and from 97.13 to 99.99 under drought stress. The averages of the physiological traits (proteins, amino acids, proline, glucose, fructose, and total soluble carbohydrates) denoted under drought stress were higher than those recorded under well-watered conditions except for proteins. In this regard, amino acids, glucose, and total soluble carbohydrates had a significant correlation with all morphological traits. The selection for drought tolerance revealed 10 tolerant genotypes from different countries (8 genotypes from Egypt, one from Morocco, and one from the United States). These selected genotypes were screened for the presence of nine specific TaDREB1 alleles. Six primers were polymorphic among the selected genotypes. Genetic diversity among the selected genotypes was investigated using 21,450 SNP markers. The results of the study shed light on the different mechanisms for drought tolerance that wheat plants use to tolerate and survive under drought stress. The genetic analysis performed in this study suggested the most suitable genotypes for selective breeding at the seedling stage under water deficit.

Abstract:Introduction: Wheat ( Triticum aestivum L.) is among the world's most important staple food crops. In the current climate change scenario, a better understanding of wheat response mechanisms to water stress could help to enhance its productivity in arid ecosystems. Methods: In this study, water relations, gas exchange, membrane integrity, agronomic traits and molecular analysis were evaluated in six wheat genotypes (D117, Syndiouk, Tunisian durum7 (Td7), Utique, Mahmoudi AG3 and BT) subjected to drought-stress. Results and discussion: For all the studied genotypes, drought stress altered leaf area, chlorophyll content, stomatal density, photosynthetic rate and water-use efficiency, while the relative water content at turgor loss point (RWC0) remained stable. Changes in osmotic potential at turgor loss point (Ψπ 0 ), bulk modulus of elasticity (Ɛmax) and stomatal regulation, differed greatly among the studied genotypes. For the drought-sensitive genotypes AG3 and BT, no significant changes were observed in Ψπ 0 , whereas the stomatal conductance (gs) and transpiration rate (E) decreased under stress conditions. These two varieties avoided turgor loss during drought treatment through an accurate stomatal control, resulting in a significant reduction in yield components. On the contrary, for Syndiouk, D117, Td7 and Utique genotypes, a solute accumulation and an increase in cell wall rigidity were the main mechanisms developed during drought stress. These mechanisms were efficient in enhancing soil water uptake, limiting leaf water loss and protecting cells membranes against leakage induced by oxidative damages. Furthermore, leaf soluble sugars accumulation was the major component of osmotic adjustment in drought-stressed wheat plants. The transcriptional analysis of genes involved in the final step of the ABA biosynthesis (AAO) and in the synthesis of an aquaporin (PIP2:1) revealed distinct responses to drought stress among the selected genotypes. In the resistant genotypes, PIP2:1 was significantly upregulated whereas in the sensitive ones, its expression showed only a slight induction. Conversely, the sensitive genotypes exhibited higher levels of AAO gene expression compared to the resistant genotypes. Our results suggest that drought tolerance in wheat is regulated by the interaction between the dynamics of leaf water status and stomatal behavior. Based on our findings, Syndiouk, D117, Utique and Td7, could be used in breeding programs for developing high-yielding and drought-tolerant wheat varieties.

Abstract:Independent and combined drought and heat stress negatively affect wheat yield and physiology. The present study was aimed to quantify effects of Drought [D], heat [H] and combined heat and drought [HD] during reproductive stage on wheat yield and to identify physio-biochemical traits which were strongly associated with improved yield and tolerance of wheat under stressful conditions. One hundred and eight elite diverse wheat genotypes were exposed to [H], [D] and [HD] treatments from heading till maturity. Grain yield was reduced by 56.47%, 53.05% and 44.66% under [HD], [H] and [D] treatment, respectively. The [HD] treatment affects the grain yield by reducing metabolism and mobilization of reserves to developing grains and leaves. Disintegration of membrane structure, chlorophyll and protein molecules was higher under [H] stress than [D] stress while water status of genotypes and sink strength was more affected by [D] than [H] stress. Multivariate analysis showed a strong correlation of chlorophyll content before and after anthesis, water-soluble carbohydrates (WSC), proline content (PC) and all other studies agronomic and physiological traits with grain yield while days to anthesis (DTA) and days to maturity (DTM) were negatively associated with grain yield under stress showing advantage of early maturity during stress. Traits having a major contribution in the first two principal components under different stress treatments may lead to improved varieties with heat and drought stress tolerance. To best of over knowledge, the present study is the first detailed study which used physiological and biochemical traits to explain the variation in grain yield and related traits in diverse wheat germplasm.
