Abstract:Pogostemon cablin Benth. (patchouli) is an important herb which possesses many therapeutic properties and is widely used in the fragrance industries. In traditional medicinal practices, it is used to treat colds, headaches, fever, nausea, vomiting, diarrhea, abdominal pain, insect and snake bites. In aromatherapy, patchouli oil is used to relieve depression, stress, calm nerves, control appetite and to improve sexual interest. Till now more than 140 compounds, including terpenoids, phytosterols, flavonoids, organic acids, lignins, alkaloids, glycosides, alcohols, aldehydes have been isolated and identified from patchouli. The main phytochemical compounds are patchouli alcohol, α-patchoulene, β-patchoulene, α-bulnesene, seychellene, norpatchoulenol, pogostone, eugenol and pogostol. Modern studies have revealed several biological activities such as antioxidant, analgesic, anti-inflammatory, antiplatelet, antithrombotic, aphrodisiac, antidepressant, antimutagenic, antiemetic, fibrinolytic and cytotoxic activities. However, some of the traditional uses need to be verified and may require standardizing and authenticating the bioactivity of purified compounds through scientific methods. The aim of the present review is to provide comprehensive knowledge on the phytochemistry and pharmacological activities of essential oil and different plant extracts of patchouli based on the available scientific literature. This information will provide a potential guide in exploring the use of main active compounds of patchouli in various medical fields.

A Comparative Study on Chemical Composition, Pharmacological Potential and Toxicity of Pogostemon cablin Linn., (Patchouli) Flower and Leaf Essential Oil

Abstract:Pogostemon cablin Linn., commonly known as patchouli is an aromatic plant with immense use in aroma and perfumery. The purpose of this study was to examine and compare the chemical composition, antimicrobial activity, and various biological activities of Pogostemon cablin Linn flower and leaf essential oils. The compositional analysis was carried out using GC/MS, while standard biological experiments were used to determine the pharmacological potential of both the essential oils. Patchouli alcohol (27.52%, 44.52%) was found to be the major compound followed by caryophyllene (18.23%, 12.86%) for both flowers and leaves essential oil (EO) respectively. P. cablin flower (PCFEO) and leaf essential oil (PCLEO) exhibited weak to strong antimicrobial activities against selected microbial strains. The mitotic index for flower and leaf EO was 14.38% and 15.20% respectively. PCFEO and PCLEO had chromosomal aberrations of 9.0% and 9.60%, respectively. Both PCFEO and PCLEO had substantial antioxidant activity, with PCLEO exhibiting the highest antioxidant activity (IC 50 = 1.31 μg/mL). Highest anti-inflammatory activity was exhibited by PCLEO (IC 50 = 4.49 μg/mL). Similarly, the anti-diabetic ability of PCLEO (IC 50 = 14.19 μg/mL) was greater than the PCFEO (IC 50 = 15.60 μg/mL). The PCLEO (IC 50 = 7.42 μg/mL) and PCFEO (IC 50 = 12.67 μg/mL) both exhibited better anticholinesterase activity than galanthamine (IC 50 = 22.16 μg/mL). Better tyrosinase inhibitory activity was shown by PCLEO with IC 50 = 4.03 μg/mL than PCFEO IC 50 = 4.44 μg/mL. Apart from their antimicrobial activity, the PCLEO was found to outperform both PCFEO as well as the standards in their respective assays. Thus, the present study affirmed the wide application aspects of patchouli essential oil apart from its aromatic application.

Abstract:Abstract Patchouli (“Guanghuoxiang”) or scientifically known as Pogostemon cablin Benth, belonging to the family Lamiaceae, has been used in traditional Chinse medicine (TCM) since the time of the Eastern Han dynasty. In TCM theory, patchouli can treat colds, nausea, fever, headache, and diarrhea. Various bioactive compounds have been identified in patchouli, including terpenoids, phytosterols, flavonoids, organic acids, lignins, glycosides, alcohols, pyrone, and aldehydes. Among the numerous compounds, patchouli alcohol, β-patchoulene, patchoulene epoxide, pogostone, and pachypodol are of great importance. The pharmacological impacts of these compounds include anti-peptic ulcer effect, antimicrobial effect, anti-oxidative effect, anti-inflammatory effect, effect on ischemia/reperfusion injury, analgesic effect, antitumor effect, antidiabetic effect, anti-hypertensive effect, immunoregulatory effect, and others.For this review, we examined publications from the previous five years collected from PubMed, Web of Science, Springer, and the Chinese National Knowledge Infrastructure databases. This review summarizes the recent progress in phytochemistry, pharmacology, and mechanisms of action and provides a reference for future studies focused on clinical applications of this important plant extract.

Abstract:Aromatic plants are known for the production of phytochemicals like essential oils which have several biological activities and have convincing fragrances. A frequent growth in the worldwide market of essential oil has been observed as it holds various beneficial therapeutic properties, which make them useful in food, fragrances, and medicinal sectors. The source of the patchouli oil isPogostemon cablin Benth, belonging to Lamiaceae family. Due to its lifelong and preserving properties, the oil has huge importance in aromatherapy and perfumery. Patchouli oil is the blend of various sesquiterpenes in which patchoulol (tricyclic sesquiterpenes) is the major constituent and the primary reason for the traditional aroma of the oil. This review article gives an overview on the importance of patchouli oil, major components of the oil, techniques employed to extract oil, its biological activities, the methods to isolate patchoulol from oil, and utilization of waste generated after the extraction.

Abstract:Patchouli alcohol (PA), a tricyclic sesquiterpene, is one of the critical bioactive ingredients and is mainly isolated from aerial part of Pogostemon cablin (known as guanghuoxiang in China) belonging to Labiatae. So far, PA has been widely applied in perfume industries. This review was written with the use of reliable information published between 1974 and 2016 from libraries and electronic researches including NCKI, PubMed, Reaxys, ACS, ScienceDirect, Springer, and Wiley-Blackwell, aiming at presenting comprehensive outline of security, pharmacokinetics, and bioactivities of PA and at further providing a potential guide in exploring the PA and its use in various medical fields. We found that PA maybe was a low toxic drug that was acquired numerously through vegetable oil isolation and chemical synthesis and its stability and low water dissolution were improved in pharmaceutics. It also possessed specific pharmacokinetic characteristics, such as two-compartment open model, first-order kinetic elimination, and certain biometabolism and biotransformation process, and was shown to have multiple biological activities, that is, immunomodulatory, anti-inflammatory, antioxidative, antitumor, antimicrobial, insecticidal, antiatherogenic, antiemetic, whitening, and sedative activity. However, the systematic evaluations of preparation, pharmaceutics, toxicology, pharmacokinetics, and bioactivities underlying molecular mechanisms of action also required further investigation prior to practices of PA in clinic.

Abstract:Pogostemon cablin is an important aromatic medicinal herb widely used in the pharmaceutical and perfume industries. However, our understanding of the phytochemical compounds and metabolites within P. cablin remains limited. To our knowledge, no integrated studies have hitherto been conducted on the metabolites of the aerial parts of P. cablin . In this study, twenty-three volatile compounds from the aerial parts of P. cablin were identified by GC-MS, predominantly sesquiterpenes. Quantitative analysis showed the highest level of patchouli alcohol in leaves (24.89 mg/g), which was 9.12 and 6.69-fold higher than in stems and flowers. UHPLC-QTOFMS was used to analyze the non-volatile compounds of leaf, stem and flower tissues. The differences in metabolites between flower and leaf tissues were the largest. Based on 112, 77 and 83 differential metabolites between flower-leaf, flower-stem and leaf-stem, three tissue-specific biomarkers of metabolites were identified, and the differential metabolites were enriched in several KEGG pathways. Furthermore, labeling differential metabolites in the primary and secondary metabolic pathways showed that flowers accumulated more lipids and amino acids, including proline, lysine and tryptophan; the leaves accumulated higher levels of terpenoids, vitamins and flavonoids, and stems contained higher levels of carbohydrate compounds. Based on the role of acetyl coenzyme A, the distribution and possible exchange mechanism of metabolites in leaves, stems and flowers of P. cablin were mapped for the first time, laying the groundwork for future research on the metabolites in P. cablin and their regulatory role.

Abstract:Essential oils are gaining popularity for their use in treating depression, including that extracted from patchouli leaves and stems (Pogostemon cablin). Herein, we used patchouli oil (PO) containing a high amount of patchouli alcohol derived from P. cablin var. Tapak Tuan. The aim of this study was to investigate the antidepressant potential of PO, with a variety of patchouli alcohol concentrations obtained from a separation process using vacuum distillation with different temperature ranges. The initial patchouli oil (iPO) was traditionally distilled by a local farmer and further distilled using a rotary evaporator at temperature ranges of 115–160 °C (POF-1); 120–160 °C (POF-2), and 125–160 °C (POF-3), resulting in products with different patchouli alcohol concentrations. POF-3, with the highest patchouli alcohol content of 60.66% (based on gas chromatography-mass spectrometry), was used for cooling crystallization, resulting in 100% patchouli alcohol crystal (pPA). A tail suspension test (TST) was performed on a rat model to screen the antidepressant potential of iPO and its derivatives. The TST results revealed that POF-3 had the best antidepressant-like effect and was second only to the fluoxetine-based antidepressant, Kalxetin®, where both groups had significant reductions of immobility time post-treatment (p < 0.0001). Other than patchouli alcohol, POF-3 also contained ledol and trans-geraniol, which have been reported for their antidepressant-related activities. Brain dopamine levels increased significantly in the group treated with POF-3 (p < 0.05) as compared with the control group), suggesting its primary anti-depressant mechanism. These findings suggest the potential of vacuum-distilled patchouli oil in reducing depression via dopamine elevation.

Abstract:Pogostemon cablin (Blanco) Benth. is an important medicinal plant; however, its molecular biology remains largely unexplored. Here, we report the results of whole transcriptome-wide analyses of P. cablin using next-generation sequencing technology. In the present study, we generated 108,996 unigenes using RNA sequencing data. Among these unigenes, 65,536 were annotated in the NR database. There were 40,670 intact open reading frames (ORFs), which accounted for 69 % of the 58,820 predicted ORFs, demonstrating that our assembled transcriptome was of high quality. Gene expression levels in the leaf and stem were quantified, and 18,481 unigenes (17 %) were determined to be significantly differentially expressed. The patchouli alcohol (PA) and pogostone (PO) metabolic pathway of P. cablin was also reconstructed, and among the 17 putative homologs to PA and PO biosynthesis-related genes, seven were significantly upregulated in the stems, and three genes were significantly upregulated in the leaves. The correlation between the essential oil contents of P. cablin and the potentially pivotal genes for PA and PO production were identified. Considering that the contents of PA and PO in leaves and stems are markedly different in the same plant and we did not detect any pattern in the expression levels of candidate genes between leaves and stems, we speculate that after the biosynthesis, more PA probably accumulates in the leaves, and more PO in the stems of P. cablin. The transcriptome of P. cabin will facilitate further researches on the molecular system biology of the Pogostemon genus, as well as the in-depth studyies of PA and PO biosynthesis and its potential applications for genetic engineering.

Uncovering the pharmacological mechanisms of Patchouli essential oil for treating ulcerative colitis

Abstract:Ethnopharmacological relevance: Pogostemonis Herba has long been used in traditional Chinese medicine to treat inflammatory disorders. Patchouli essential oil (PEO) is the primary component of Pogostemonis Herba, and it has been suggested to offer curative potential when applied to treat ulcerative colitis (UC). However, the pharmacological mechanisms of PEO for treating UC remain to be clarified. Aim of the study: To elucidate the pharmacological mechanisms of PEO for treating UC. Methods and results: In the present study, transcriptomic and network pharmacology approaches were combined to clarify the mechanisms of PEO for treating UC. Our results reveal that rectal PEO administration in UC model mice significantly alleviated symptoms of UC. In addition, PEO effectively suppressed colonic inflammation and oxidative stress. Mechanistically, PEO can ameliorate UC mice by modulating gut microbiota, inhibiting inflammatory targets (OPTC, PTN, IFIT3, EGFR, and TLR4), and inhibiting the PI3K-AKT pathway. Next, the 11 potential bioactive components that play a role in PEO's anti-UC mechanism were identified, and the therapeutic efficacy of the pogostone (a bioactive component) in UC mice was partially validated. Conclusion: This study highlights the mechanisms through which PEO can treat UC, providing a rigorous scientific foundation for future efforts to develop and apply PEO for treating UC.
Si(004) measurements of wafer curvature due to the stress introduced by PECVD/HDP SiN films in wafer fabrication

Abstract:Thin film deposition process invariably introduces compressive or tensile stress in the films. The stress in a film causes the wafer to warp whose curvature is estimated in a wafer fab using optical reflectance technique. Alternatively, the wafer curvature can also be measured using the high resolution XRD (HRXRD) Si(004) rocking curves. In this paper, the HRXRD technique was employed to evaluate the stress in PECVD and HDP SiN films deposited on blanket Si wafers (prime-grade material) which were subjected to different Xe ion-implant energy conditions. It will be shown that stress-matching in PECVD and HDP SiN can be achieved by tuning Xe implant energy. From the change in the width of Si(004) peak, a qualitative assessment regarding the quality of single crystal Si lattice will be presented which is not possible to measure with optical reflectance methods.
Molecular identification and expression of sesquiterpene pathway genes responsible for patchoulol biosynthesis and regulation in Pogostemon cablin

Abstract:Background: Many commercially important drug and flavor compounds are secondary metabolites of terpenoid origin. Pogostemon cablin, a commercially important industrial and medicinal crop, accumulates abundant patchouli oil comprised of more than 24 unique sesquiterpene compounds, with the most abundant being patchouli alcohol. Results: In this study, we analyzed the P. cablin transcriptome library, obtaining 74 terpenoid biosynthesis-related genes, and identified their expression patterns in leaves, stems, and flowers. These genes are members of 15 different families, and we detected all the enzymes involved in the sesquiterpenes pathway that are responsible for patchoulol biosynthesis. Sequence structure, homology, conserved domain properties, and phylogeny of certain identified genes were systematically investigated. Color complementation assay was used to verify the functional activity of the MEP pathway proteins. Exogenous hormone treatment revealed that patchoulol synthesis is induced by methyl jasmonate (MeJA). Quantitative reverse-transcription PCR analysis indicated that the MVA pathway genes (acetoacetyl-CoA thiolase, 3-hydroxy-3-methylglutaryl-coenzyme A reductase, mevalonate diphosphate decarboxylase, and farnesyl diphosphate synthase) participate in patchoulol biosynthesis and are mediated by MeJA. Conclusions: Taken together, this is the first report of integrated analysis of P. cablin MVA and MEP pathway related genes, providing a better understanding of terpenoid and/or patchoulol biosynthesis in P. cablin, and the basis for improving patchoulol production through genetic engineering.
Antimicrobial evaluation of the Patchouli (Pogostemon cablin Benth.) leaf essential oil combination with standard antimicrobial compounds

Abstract:Pogostemon cablin Benth's leaves, which belong to the Lamiaceae family, are used to make patchouli essential oil (PEO). Sesquiterpenes are the main compounds in essential oil. Antibacterial, antifungal, antiviral, antiemetic, and anti-inflammatory activities are just a few of the many pharmacological actions that are well-known. In this investigation, cefuroxime (CEF), moxifloxacin (MOX), clarithromycin (CLA), fluconazole (FLU), and terbinafine (TER) were combined with patchouli (Pogostemon cablin leaf essential oil). Gram-positive Staphylococcus aureus ATCC 6538, Gram-negative Escherichia coli ATCC 8739, and yeasts Candida albicans ATCC 10231, and C. tropicalis ATCC 750 were tested against all combinations. Drug interaction was given as fractional inhibitory concentrations (FIC) after combinations of the essential oils were tested for their minimum inhibitory concentrations (MIC). The checkerboard method was used to assess antimicrobial interactions, Fluconazole, terbinafine, cefuroxime, and clarithromycin combined with essential oil demonstrated an "additive effect" against Candida strains together with E. coli and S. aureus.

Abstract:Abstract Patchouli oil is derived from the patchouli plant, a member of the family Lamiaceae. Similar plants from this family include mint, sage and lavender. The Pogostemon cablin species of the plant is the most popular for cultivating essential oil due to the species’ outstanding therapeutic properties. The oil is obtained through an intricate steam distillation process. Initially used in South East Asia to mask the smell of body decomposition during funerals in the 17th century, and later as an insect repellent in the 1800s, patchouli leaves entered the European market in the 19th century. Nowadays, it is used across perfumery, aromatherapy, skincare and spiritual practices. Once a symbol of wealth, patchouli was so beloved by Tutankhamun that he was buried with 10 gallons of the oil in his tomb in ancient Egypt. Concerning dermatological conditions, patchouli was an early treatment for acne due to its anti-inflammatory and antibacterial properties. It has also shown benefits for dermatitis, eczema, dandruff and oily scalp conditions. As a cell rejuvenator, it can help reduce scar appearance and aid in wound healing. These effects are attributed to the flavonoids, mono- and sesquiterpenoids present in patchouli. In 2013, Yang et al. used molecular docking technology to investigate the antibacterial activity of patchouli oil in vitro (Yang X, Zhang X, Yang SP, Liu WQ. Evaluation of the antibacterial activity of patchouli oil. Iran J Pharm Res 2013; 12: 307–16). Twenty-six compounds in the oil displayed antibacterial activity. In further studies, molecular docking tools and viewer software were used for virtual screening of these compounds. The most significant finding was alpha-patchouli alcohol being a potential inhibitor of the cyclo-oxygenase 1 enzyme. Inhibiting this enzyme would reduce the prostaglandin production involved in the inflammatory process. Different studies have also shown a correlation with patchouli oil and the downregulation of matrix metalloproteinases (MMP-1 and MMP-3) involved in collagen degradation. Further experimental evidence has demonstrated positive results for ultraviolet-induced skin photoageing and the protective effects of P. cablin in mice. Researchers applied patchouli oil for 2 h before ultraviolet exposure to the dorsal skin of mice. Patchouli oil doses of 6 mg per mouse and 9 mg per mouse significantly suppressed skin wrinkle formation, improved skin elasticity, and increased collagen content by 21.9% and 26.3%, respectively. Patchouli oil maintains a prominent role in traditional Asian medicine, while in the West, there is minimal literature, indicating substantial potential for patchouli in dermatological practice. The promising evidence for photoageing necessitates continued investigation. In the cosmetic industry, patchouli remains a popular ingredient in beard oils and more, appreciated by many.

Abstract:Breast cancer is one of the highest known causes of death in Indonesia, especially for women. Pogostemon cablin Benth is one of the Aceh's endemic herbal plants that has been studied to have potential such as anti-inflammatory, antiproliferative, antioxidant, antimicrobial, and proapoptotic. Network pharmacology approach was conducted to explore and analyze the potential of P. cablin as an anti-breast cancer. P. cablin plant compounds were obtained from GCMS and breast cancer genes data were obtained from OMIM, GeneCard, and DisGeNet databases. Target proteins and pathways involved were identified using STRING-DB and Metascape. Network analysis was performed using Cytoscape. A total of 65 plant compounds with 554 target proteins and 1854 disease genes were obtained. Based on the results of combining protein targets using Venn diagrams, 138 overlapping proteins between drug compounds and breast cancer disease targets were identified. Based on KEGG and GO analysis, P. cablin is known to have potential in breast cancer treatment/therapeutic mechanisms. Based on the "compound-protein target-pathway" multi-target mechanism, pogostol; 1H-Cycloprop[e]azulen-7-ol, decahydro-1,1,7-trimethyl-4-methylene-, [1ar-(1aα,4aα,7β,7aβ,7bα)]-; 3-Hexen-1-ol, 2,5-dimethyl-, acetate, (Z)-, 5β,7βH,10α-Eudesm-11-en-1α-ol; Acetic acid, 3-hydroxy-6-isopropenyl-4,8a-dimethyl 1,2,3,5,6,7,8,8a-octahydronaphthalen-2-yl ester; and Humulenol-II interacts with proteins that play a significant role in breast cancer, which are MAPK1, EGFR, TNF, AKT1, and JAK2. Hence, it can be concluded that P. cablin has good potential to be a source of therapeutic treatment against breast cancer. However, it needs to be tested clinically to further determine the effects of this P. cablin compound.

Abstract:Patchouli extracts and oils extracted from Pogostemon cablin are essential raw material for the perfume and cosmetics industries, in addition to being used as a natural additive for food flavoring. Steam distillation is a standard method used for plant extraction. However, this method causes thermal degradation of some essential components of the oil. In this study, patchouli was extracted with supercritical carbon dioxide (SC-CO2) under different conditions of pressure (10–30 MPa) and temperature (40–80 °C). The chemical components of the crude extracted oil and the functional group were characterized using gas chromatography-mass spectrometry (GC-MS) and Fourier Transform Infrared Spectroscopy (FT-IR). The extraction with supercritical carbon dioxide was shown to provide a higher yield (12.41%) at a pressure of 20 MPa and a temperature of 80 °C. Patchouli alcohol, Azulene, δ-Guaiene, and Seychellene are the main bioactive compounds that GC-MS results have identified. FTIR spectra showed alcohol, aldehyde, and aromatic ring bond stretching peaks. Extraction of patchouli with supercritical carbon dioxide provided a higher yield and a better quality of the crude patchouli oil.

Abstract:Pogocablanols A–G ( 1 – 7 ), seven new compounds with an unprecedented sesquiterpenoid skeleton, were isolated from the aerial parts of Pogostemon cablin . This novel type of sesquiterpenoids, named pogocablane-type sesquiterpenoids, possesses a bicyclic nonisoprenoid system, including a five-membered carbocycle with a methyl group, a six-membered carbocycle with two methyl groups, and a methylene connecting the two carbocycles. Particularly, three isomers have an interesting 1-oxaspiro[4,4]nonane moiety. Their structures and absolute configurations were determined using extensive spectroscopic evidence, single-crystal X-ray diffraction, and electronic circular dichroism calculations. Pogocablanols A–D ( 1 – 4 ) exhibited a concentration-dependent inhibitory effect on rat jejunum smooth muscle contraction induced by acetylcholine, with E max values varying from 48.87 ± 4.01% to 69.02 ± 7.54%. Preliminary structure–activity relationship analysis suggested that the oxaspirocycle and the pyran ring in pogocablanols are important structural features for the relaxing effects on jejunum smooth muscle. This study offered crucial support for more investigation and development of P. cablin by showing that these pogocablane-type sesquiterpenoids had the potential to be applied in pharmaceutical and functional food formulations. Effect of patchouli (Pogostemon cablin) essential oil on in vitro and in vivo leukocytes behavior in acute inflammatory response

Abstract:Purpose: The aim of this study was to evaluate the effect of Pogostemon cablin essential oil (PEO) on leukocyte behavior in the inflammatory response. Methods and results: PEO was analyzed using Gas Chromatography/Mass Spectrometry (GC/SM) and Nuclear Magnetic Resonance Spectroscopy (NMR) methods and showed predominance of patchoulol (38.50%), α-bulnesene (20.37%), α-guaiene (12.31%), seychellene (8.33%) and α-patchoulene (4.91%). PEO at concentrations of 1, 3, 10, 30, 60 and 90μg/ml reduced the in vitro neutrophil chemotaxis toward fMLP, and at concentrations of 3 and 10μg/ml, increased the phagocytic activity of neutrophils. Topical application of PEO in high concentrations promoted an increase of ear edema and myeloperoxidase (MPO) activity. However, the oral treatment with 100, 200 and 300mg/kg reduced leukocyte recruitment, nitric oxide (NO) production, and rolling and adherent leukocyte number in the microcirculation. Conclusion: PEO affects the leukocyte behavior, and the mechanism proposed of PEO seems to be, at least in part, involving the participation of NO and pro-inflammatory cytokines.

Abstract:Ethnopharmacological relevance Plants of the Podocarpus species belong to the Podocarpaceae family and are largely distributed in the southern hemisphere. Beside the commercially and ecologically valuable, plants of the Podocarpus species are also used in traditional medicines in some countries for treating asthma, fever, venereal diseases, eye diseases, etc Aim of the study In recent decades, the identities and pharmacological activities of phytochemicals extracted from Podocarpus plants have been widely studied. However, there have been no comprehensive and systematic reviews. This article aims to systematically review the latest research on the putative mechanisms underlying pharmacological actions of phytochemicals from the Podocarpus species, as well as to lay a foundation for promoting the development of plant resources from this genus, further drug research, and product development. Materials and methods A comprehensive search of PubMed, Google Scholar, Web of Science, Elsevier and CNKI databases was conducted using the keywords " Podocarpus ", "traditional usage", "phytochemistry", "pharmacology", "nagilactone", etc . Related papers published among July 1964 to February 2023 were collected to summarize the research progress. All plant names were determined through the "The Plant List" ( http://www.theplantlist.org/ ). Results To date, 262 chemical constituents have been isolated and identified from 26 Podocarpus plants; among these, norditerpene bilactone is the main pharmacologically active component. Norditerpene bilactones are reported to have anti-cancer, anti-inflammatory, antioxidant, antibacterial, anti-tyrosinase, neuroprotective, anti-plasmodial, anti-mutagenic, and anti-atherosclerotic properties as well as other pharmacological activities, which support its traditional uses. Conclusion Extensive studies on phytochemistry and pharmacology of Podocarpus species lead to discovery of a series of hopeful leading compounds with unique chemical structure, especially the nor- and bis-norditerpenoid dilactones with four isoprene units. These compounds have been proved to possess various pharmacological activities. This review will provide a reference for further research and promote the idea of combining modern research with traditional medicinal applications of Podocarpus plants.

Abstract:<h3 class="Heading">Background</h3><p class="Para"><em class="EmphasisTypeItalic">Pogostemon cablin</em> (Blanco) Benth. (Patchouli) is an important aromatic and medicinal plant and widely used in traditional Chinese medicine as well as in the perfume industry. Patchoulol is the primary bioactive component in <em class="EmphasisTypeItalic">P. cablin</em>, its biosynthesis has attracted widespread interests. Previous studies have surveyed the putative genes involved in patchoulol biosynthesis using next-generation sequencing method; however, technical limitations generated by short-read sequencing restrict the yield of full-length genes. Additionally, little is known about the expression pattern of genes especially patchoulol biosynthesis related genes in response to methyl jasmonate (MeJA). Our understanding of patchoulol biosynthetic pathway still remained largely incomplete to date.</p><h3 class="Heading">Results</h3><p class="Para">In this study, we analyzed the morphological character and volatile chemical compounds of <em class="EmphasisTypeItalic">P. cablin cv.</em> '<em class="EmphasisTypeItalic">Zhanxiang</em>', and 39 volatile chemical components were detected in the patchouli leaf using GC-MS, most of which were sesquiterpenes. Furthermore, high-quality RNA isolated from leaves and stems of <em class="EmphasisTypeItalic">P. cablin</em> were used to generate the first full-length transcriptome of <em class="EmphasisTypeItalic">P. cablin</em> using PacBio isoform sequencing (Iso-Seq). In total, 9.7 Gb clean data and 82,335 full-length UniTransModels were captured. 102 transcripts were annotated as 16 encoding enzymes involved in patchouli alcohol biosynthesis. Accorded with the uptrend of patchoulol content, the vast majority of genes related to the patchoulol biosynthesis were up-regulated after MeJA treatment, indicating that MeJA led to an increasing synthesis of patchoulol through activating the expression level of genes involved in biosynthesis pathway of patchoulol. Moreover, expression pattern analysis also revealed that transcription factors participated in JA regulation of patchoulol biosynthesis were differentially expressed.</p><h3 class="Heading">Conclusions</h3><p class="Para">The current study comprehensively reported the morphological specificity, volatile chemical compositions and transcriptome characterization of the Chinese-cultivated <em class="EmphasisTypeItalic">P. cablin cv.</em> '<em class="EmphasisTypeItalic">Zhanxiang</em>', these results contribute to our better understanding of the physiological and molecular features of patchouli, especially the molecular mechanism of biosynthesis of patchoulol. Our full-length transcriptome data also provides a valuable genetic resource for further studies in patchouli.</p>
