Abstract:Abstract Necrotic enteritis (NE) is being considered as one of the most important intestinal diseases in the recent poultry production systems, which causes huge economic losses globally. NE is caused by Clostridium perfringens , a pathogenic bacterium, and normal resident of the intestinal microflora of healthy broiler chickens. Gastrointestinal tract (GIT) of broiler chicken is considered as the most integral part of pathogen’s entrance, their production and disease prevention. Interaction between C. perfringens and other pathogens such as Escherichia coli and Salmonella present in the small intestine may contribute to the development of NE in broiler chickens. The antibiotic therapy was used to treat the NE; however European Union has imposed a strict ban due to the negative implications of drug resistance. Moreover, antibiotic growth promoters cause adverse effects on human health as results of withdrawal of antibiotic residues in the chicken meat. After restriction on use of antibiotics, numerous studies have been carried out to investigate the alternatives to antibiotics for controlling NE. Thus, possible alternatives to prevent NE are bio-therapeutic agents (Probiotics), prebiotics, organic acids and essential oils which help in nutrients digestion, immunity enhancement and overall broiler performance. Recently, probiotics are extensively used alternatives to antibiotics for improving host health status and making them efficient in production. The aim of review is to describe a replacement to antibiotics by using different microbial strains as probiotics such as bacteria and yeasts etc. having bacteriostatic properties which inhibit growth of pathogens and neutralize the toxins by different modes of action.

Abstract:Necrotic enteritis (NE) in poultry is an economically important disease caused by Clostridium perfringens type A bacteria. A global trend on restricting the use of antibiotics as feed supplements in food animal production has caused a spike in the NE incidences in chickens, particularly in broiler populations. Amongst several non-antibiotic strategies for NE control tried so far, probiotics seem to offer promising avenues. The current review focuses on studies that have evaluated probiotic effects on C. perfringens growth and NE development. Several probiotic species, including Lactobacillus, Enterococcus, Bacillus, and Bacteroides bacteria as well as some yeast species have been tested in chickens against C. perfringens and NE development. These findings have shown to improve bird performance, reduce C. perfringens colonization and NE-associated pathology. The underlying probiotic mechanisms of NE control suggest that probiotics can help maintain a healthy gut microbial balance by modifying its composition, improve mucosal integrity by upregulating expression of tight-junction proteins, and modulate immune responses by downregulating expression of inflammatory cytokines. Collectively, these studies indicate that probiotics can offer a promising platform for NE control and that more investigations are needed to study whether these experimental probiotics can effectively prevent NE in commercial poultry operational settings.

Abstract:In commercial poultry production, chickens are reared under intensive conditions, which may allow infections to spread quickly. Antibiotics are used at sub-therapeutic doses in livestock and poultry feed to prevent diseases and improve productivity. However, restrictions on the use of antibiotics at sub-therapeutic concentrations in livestock feed due to growing concerns of antimicrobial resistance (AMR), together with antibiotic residues in meat and eggs has prompted poultry researchers and feed producers to look for viable alternatives. Thus, there is increasing interest in developing natural alternatives to in-feed antibiotics to improve chicken productivity and health. Probiotics, specifically from the genus Bacillus have proven to be effective due to their spore-forming capabilities. Furthermore, their ability to withstand heat during feed processing and be stored for a long time without losing viability as well as their potential to function in the acidic medium of the chicken gut, provide them with several advantages over conventional probiotics. Several studies regarding the antimicrobial and antioxidant activities of Bacillus probiotics and their positive impact in chicken nutrition have been documented. Therefore, the present review shields light on the positive effect of Bacillus probiotics as alternatives to in-feed antibiotics on growth performance, serum chemistry, antioxidant status, intestinal histomorphology and lesion scores of disease-challenged broiler chickens and the mechanisms by which they exert their actions. It is concluded that Bacillus probiotics supplementation improve growth, health and productive indices of disease-challenged broiler chickens and can be a good alternative to in-feed antibiotics. However, more studies are required on the effect of Bacillus probiotics supplementation in broiler chickens to maximize productivity and achieve the ultimate goal of stopping the usage of antibiotics at sub-therapeutic doses in broiler chicken feed to enhance performance.

Abstract:Poultry production has undergone a substantial increase compared to the livestock industries since 1970. However, the industry worldwide is now facing challenges with the removal of in-feed antibiotics completely or gradually, as the once well-controlled poultry diseases have re-emerged to cause tremendous loss of production. Necrotic enteritis (NE) is one of the most important diseases which costs the industry over two billion dollars annually. In this paper, we review the progress on the etiology of NE and its control through dietary modifications, pre- and probiotics, short chain fatty acids, and vaccination. The other likely measures resulted in the most advances in the toxin characterization are also discussed. Vaccine strategies may have greater potential for the control of NE mainly due to clearer etiology of NE having been elucidated in recent years with the identification of necrotic enteritis toxin B-like (NetB) toxin. Therefore, the use of alternatives to in-feed antibiotics with a better understanding of the relationship between nutrition and NE, and limiting exposure to infectious agents through biosecurity and vaccination, might be a tool to reduce the incidence of NE and to improve gut health in the absence of in-feed antibiotics. More importantly, the combinations of different measures may achieve greater protection of birds against the disease. Among all the alternatives investigated, prebiotics, organic acids and vaccination have shown improved gastrointestinal health and thus, have potential for the control of NE.

Abstract:This experiment was conducted to study the effects of probiotics( PRO) and cello-oligosaccharide( COS) on nutrient digestion,mRNA expression of intestinal mucosa amino acid transporters and nitrogen emission of broilers under heat stress. Nine hundred 21-day-old Ross 308 broilers w ere randomly allocated to 5groups: thermoneutralzone( HN) group,heat stress( HS) group,probiotics( HS+0.15%PRO) group,cellooligosaccharide( HS+0.15%COS) group and synbiotics( HS +SYN,SYN = 0.15%PRO +0.15%COS) group.Birds w ere either raised in a thermoneutral zone( 22 ℃) in HN group,and the others w ere subjected to cyclic heat stress by exposing to 33 ℃ from 08: 00 to 18: 00 and 22 ℃ from 18: 00 to next day 08: 00. Raising experiment period w as 21 days. The results show ed as follow s: 1) compared w ith birds kept in HN group,average daily gain and average daily feed intake of broilers subjected to heat stress w ere significantly reduced 10.65%( P0.05) and 8.04%( P0.05),respectively. Average daily gain,average daily feed intake and feed to gain ratio of broilers subjected to heat stress w ith diet added probiotics or cello-oligosaccharide individually w ere not significantly different( P0.05). Apparent digestibility of crude protein and crude fiber in HS group w as significantly decreased( P0.05),nitrogen intake and nitrogen retention w ere significantly decreased( P0.05),nitrogen excretion and nitrogen excretion rate w ere significantly increased( P0.05),ammonia nitrogen,uric acid contents in excreta and ammonia concentration in chicken house w ere significantly increased( P0.05),the activity of urease in excreta w as significantly increased( P0.05),and mRNA expression of the basic amino acid transporter( rBAT),y+L amino acid transporter-2( y+LAT2),cationic amino acid transporter-1( CAT1) and cationic amino acid transporter-4( CAT4) in intestinal mucosa was significantly down-regulated( P0.05). 2) Compared with HS group,apparent digestibility of crude protein was significantly increased in HS+PRO and HS+COS groups( P0.05),nitrogen excretion and nitrogen excretion rate in HS+PRO and HS+COS groups w ere significantly reduced( P0.05),w hile nitrogen retention w as significantly increased( P0.05). Compared w ith HS group,excreta ammonia nitrogen content in HS+PRO group w as significantly low-ered( P0.05),and ammonia concentration in chicken house and activity of urease in excreta of HS +COS group w ere significantly reduced( P0.05). 3) As compared w ith HS group,apparent digestibility of crude protein and crude fiber w ere significantly increased( P0.05) in HS+SYN group. Nitrogen excretion and nitrogen excretion rate in HS+SYN group w ere significantly reduced( P0.05),w hile nitrogen retention w as significantly increased( P0.05). The results indicate that heat stress decrease nutrient digestion and increase nitrogen emission of broilers,and dietary addition of probiotics and cello-oligosaccharide are effective in partially ameliorating these changes. The combination of probiotics and cello-oligosaccharide is superior to individually addition.

Abstract:The current study investigated the efficacy of a probiotic mixture on ameliorating heat stress-induced impairment of intestinal microflora, morphology, and barrier integrity in broilers. The probiotic mixture contained Bacillus licheniformis, Bacillus subtilis, and Lactobacillus plantarum. Three hundred sixty 21-d-old Ross 308 male broilers were allocated in 4 experimental treatments, each of which was replicated 6 times with 15 broilers per replicate. A 2 × 2 factorial design was used in the study, and the main factors were composed of diet (basal diet or addition of 1.5 g/kg of probiotic mixture) and temperature (thermoneutral zone or heat stress). From d 22 to 42, birds were either raised in a thermoneutral zone (22°C) or subjected to cyclic heat stress by exposing them to 33°C for 10 h (from 0800 to 1800) and 22°C from 1800 to 0800. Compared with birds kept in the thermoneutral zone, birds subjected to heat stress had reduced ADG and ADFI; lower viable counts of Lactobacillus and Bifidobacterium and increased viable counts of coliforms and Clostridium in small intestinal contents; shorter jejunal villus height, deeper crypt depth, and lower ratio of villus height to crypt depth; decreased jejunal transepithelial electrical resistance and a higher level of jejunal paracellular permeability of fluorescein isothiocyanate dextran 4 kDa; and downregulated protein levels of occludin and zonula occludens-1 (P < 0.05). Supplemental probiotics increased (P < 0.05) small intestinal Lactobacillus and Bifidobacterium, jejunal villus height, protein level of occludin, and decreased (P < 0.05) feed to gain ratio and small intestinal coliforms. These results indicate that dietary addition of probiotic mixture was effective in partially ameliorating intestinal barrier function. But no temperature × diet interaction was observed in the present study, revealing that the supplemented probiotics had the same effect at both temperatures.

As compared with HS group,viable counts of Lactobacillus,Bifidobacterium,Escherichia coli and Clostridium in cecal contents in HS+PRO group were not significantly different( P>0. 05 ),the viable counts of Lactobacillus in cecal contents was significantly in-creased( P<0.05),and Escherichia coli and Clostridium in cecal contents in HS+COS group were significant-ly decreased( P<0.05). 3)As compared with HS group,feed to gain ratio in HS+SYN group was significant-ly decreased by 4.21%(P<0.05);the viable counts of Lactobacillus and Bifidobacterium in cecal contents were significantly increased(P<0.05),and Escherichia coli and Clostridium in cecal contents were signifi-cantly decreased( P<0.05);villus height and villus height to crypt depth ratio were significantly increased (P<0.05);TER value was significantly increased(P<0.05)and the flux of FD4 was significantly reduced ( P<0.05). These results indicate that additions of probiotics and cello-oligosaccharide are effective in partially ameliorating intestinal barrier function. The combination of probiotics and cello-oligosaccharid has better effect on ameliorating intestinal barrier function than supplementing them alone.
Comparative efficacy of postbiotic, probiotic, and antibiotic against necrotic enteritis in broiler chickens

Abstract:Prevention of necrotic enteritis (NE), caused by Clostridium perfringens (C. perfringens), is one of the most important goals to improve the profitability of broiler chickens. This work aimed to compare the efficacy of 2 antibiotic alternatives including a postbiotic (dry feed additive and aqueous nonviable Lactobacillus (L.) species fermentation) and a probiotic (dry feed additive and aqueous Bacillus (B.) subtilis and B. lischeniformis mixture) with an antibiotic (amoxicillin in water) against NE. Four hundred, day-old broiler chicks were divided into 8 equal groups (Gs), n = 50 each (5 replicates; 10 each). Chickens of G1 (postbiotic dry-feed additive), G2 (postbiotic and antibiotic in drinking water), G3 (postbiotic dry and aqueous), G4 (probiotic dry-feed additive), G5 (probiotic and antibiotic in drinking water), G6 (probiotic dry and aqueous), and G7 (nontreated) were orally inoculated with a toxigenic C. perfringens type A on the d 19 to 21 of age and predisposed with 3X coccidial vaccine for induction of NE. However, chickens of G8 were kept nontreated or challenged. The severity of NE signs was markedly decreased in G3 in comparison with other challenged treatment groups, and the mortality rates were 22%, 10%, 16%, 22%, 12%, 20%, and 36% in Gs 1, 2, 3, 4, 5, 6, and 7, respectively. The best significant (P ≤ 0.05) feed conversion ratio was detected in G3 (1.51), G6 (1.54), and G2 and G8 (1.61). In addition, the European production efficiency factor was significantly (P ≤ 0.05) improved in G3 (279.33) and G2 (266.67), but it was decreased in G7 (177.33) when compared with G8 (339.33). An improvement in intestinal and hepatic pathology and liver function tests, as well as a significant (P ≤ 0.05) decrease in bacterial counts were observed in Gs 2, 5, 3, 6, 1, and 4, respectively in comparison with G7. Immunologically, the highest significant (P ≤ 0.05) hemagglutination inhibition antibody titers for Newcastle disease virus vaccine were in Gs 1 and 3 (6.4 log2). In conclusion, the combined feed and water postbiotic treatment demonstrated promising results in ameliorating the severity of NE and improving the hepatic and the immune status of broiler chickens when compared with the commonly used probiotic and antibiotic.
Necrotic enteritis and antibiotic-free production of broiler chickens: Challenges in testing and using alternative products

Abstract:The global trend towards raising broiler chickens without the use of in-feed antibiotics (IFAs) means that there is an ongoing need to develop alternative treatments capable of delivering the benefits that IFAs previously provided. IFAs supported the productivity performance of chickens and played a key role in maintaining their health. Necrotic enteritis (NE) is an important disease of broilers that affects health, productivity, and welfare, and was previously well controlled by IFAs. However, with the reduction in IFA use, NE is resurgent in some countries. Vaccines and various feed additives, including pre-, pro-, and postbiotics, phytobiotics, fatty acids, and phage therapies have been introduced as alternative methods of NE control. While some of these feed additives have specific activity against the NE pathogen, Clostridium perfringens, most have the more general goal of reinforcing gut health. Extensive reviews of the effects of many of these feed additives on gut health have been published recently. Hence, rather than cover previously well reviewed areas of research this review focuses on the challenges and pitfalls in undertaking experimental assessment of alternative NE treatments and translating laboratory research to real world commercial production settings. The review is based on the author's particular experience, reading, thoughts, and analysis of the available information and inevitably presents a particular understanding that is likely to be at odds with others thinking on these issues. It is put forward to stimulate thinking and discussion on the issues covered.

Abstract:The gastrointestinal tract (GIT) provides the biological environment for nutrient digestion and absorption, and protection from pathogens and toxins. Broilers are fast growing because of the great potential of intestinal epithelia for nutrient absorption, and efficient conversion of nutrient to muscle. Physiologically, reactive oxygen species (ROS) and reactive nitrogen species (RNS) are generated by GIT epithelial cells either from oxygen metabolism or by enteric commensal bacteria and regulate gut health. However, increased production of ROS elevates free radical production and antioxidant insults resulting in oxidative stress. Oxidative stress in poultry GIT is derived from nutritional, environmental heat stress, and pathological factors, which alters overall performance as well as meat and egg quality. Supplementation of exogenous vitamins, antioxidants, and plant extract having antioxidant properties scavenge ROS and are beneficial in mitigating oxidative stress in the GIT. This review highlights the involvement of oxidative stress in the gastrointestinal functionality of poultry and potential intervention strategies to maintain redox balance in the GIT.

Abstract:Background Chicken meat is one of the most consumed meats worldwide and poultry production is increasing at an exponential rate. Reducing antibiotic usage has resulted in the recurrence of subclinical necrotic enteritis again and influenced global poultry production. Probiotics are potential antibiotic substitutes that can be used to prevent subclinical necrotic enteriti. However, the precise mechanism of action of probiotics and information on which gut microbes confer this efficacy remain elusive. Methods and results The subclinical necrotic enteritis animal model was used to reveal the mechanism underlying the effect of probiotics on intestinal health through RNA sequencing and 16S rDNA amplicon sequencing. Bacillus licheniformis H2 feeding significantly reduced the relative abundance of Clostridium perfringens in the ileum and markedly ameliorated the pathological damage in the ileum and liver. In addition, oral administration of B. licheniformis H2 contributed to the enhancement of the intestinal barrier function and epithelial renewal, reducing energy consumption, and improving enteral nutrition absorption. Probiotic B. licheniformis H2 also ameliorated the inflammatory response and increased the immunity of subclinical necrotic enteritis infected broilers. Finally, B. licheniformis H2 feeding regulated liver gene expression to suppress immune response and promoted growth and metabolism depending on the gut microbiota. Conclusions These results indicated the mechanism of probiotic action of B. licheniformis H2 in maintaining intestinal health and thus promoting growth and B. licheniformis H2 may serve as an antibiotic substitute to prevent subclinical necrotic enteritis in poultry farming.

Abstract:To evaluate the efficacy of synbiotic during a necrotic enteritis (NE) infection, a total of 360 day-old chicks were randomly assigned into 4 experimental groups in a 2 × 2 factorial setup: control, challenge, synbiotic (1 g/kg), and challenge + synbiotic, with 6 replicates. NE was induced by gavaging 1 × 104Eimeria maxima oocysts and 1 × 108 CFU/mL of Clostridium perfringens on d 14 (D14) and D19, 20, and 21, respectively. At D35, the NE challenge decreased the BW gain (P < 0.001) and increased feed conversion ratio (P = 0.03), whereas synbiotic supplementation decreased the feed intake (P = 0.04). At D21, NE challenge increased gut permeability (P < 0.001), decreased regulatory T cells (Tregs) in the cecal tonsil (CT) (P = 0.02), increased Tregs in the spleen (P = 0.02), decreased nitric oxide (NO) production in the spleen (P = 0.04) and decreased IL-10 expression in CT (P = 0.02), whereas synbiotic supplementation increased CD4+:CD8+ T cells in the spleen (P < 0.001) and decreased interferon (IFN)-γ expression in the jejunum (P = 0.07), however, synbiotic supplementation during NE challenge decreased mid-gut lesion score (P < 0.001), increased CD4+:CD8+ T cells in CT and decreased IgA production in bile (P < 0.001), compared to the control group. At D28, synbiotic supplementation decreased CD4+:CD8+ T cells in CT (P < 0.001), whereas synbiotic supplementation during NE challenge decreased Tregs in CT (P < 0.001) and increased NO production in the spleen (P = 0.04), compared to the control group. At D35, the NE challenge decreased CD4+:CD8+ T cells in the spleen (P = 0.03), decreased IgA production in bile (P = 0.02), decreased IL-10 expression in CT (P = 0.04), and decreased IL-10 (P = 0.009), IFN-γ (P = 0.03) and inducible nitric oxide synthase (P = 0.02) expression in the jejunum, whereas synbiotic supplementation increased Tregs in the spleen (P = 0.04), compared to control group. Synbiotic supplementation during the NE challenge decreased both IL-1β (P = 0.02) and IFN-γ (P = 0.001) expression in CT, compared to the control group. It can be concluded that synbiotic supplementation increases production performance by decreasing mid-gut lesions and enhancing protective immunity against NE, and efficiency of synbiotic could be improved by blending additional probiotics and prebiotics.

Abstract:SUMMARYUsing antibiotics as growth promotants has been banned due to the increasing problem of antibiotic resistance and the presence of antibiotic residues in poultry products, which pose a risk to human health. Therefore, probiotics could be utilised as a promising alternative to antibiotic growth promoters, because of the increased requirement for antibiotic-free and organic poultry products. Probiotics are viable micro-organisms used as feed additives. Scientists have paid much attention to the benefits of probiotics due to their ability to preserve a healthy intestinal microbial balance. Probiotic supplementation could enhance the gut structure integrity, physiological function, intestinal immunity and health. Probiotics are also used to enhance the antioxidative status via activation of antioxidative enzymes and to reduce lipid peroxidation. Moreover, dietary probiotic micro-organisms are able to stimulate humoral- and cell-mediated immunity and regulate pro- and anti-inflammatory cytokines in the host. These physiological and immunological benefits of probiotics are translated into various improvements, such as in growth performance, laying performance, egg quality, meat quality and meat safety. Interestingly, probiotics have positive impacts on alleviating heat stress in poultry. The current review is an effort to shed light on recent findings related to the benefits of dietary probiotics on gut development, nutrient absorption, gut microbiota, antioxidative properties, immune responses and productive performance in poultry.

Abstract:The majority of pathologies in poultry are linked to intestinal chronic inflammation due to a disbalance of the gut microbiota. Thus, a healthy microbiota drives the gut integrity, and the gut’s biological and metabolic functionalities, including efficacious use of nutrition, but also immunity, and neuroendocrine systems. However, many external factors are disturbing a stable, healthy gut microbiota. Heat stress, dysbiosis, leaky gut syndrome, and mycotoxins are the main “secret killers” in poultry that lead to chronic oxidative stress and inflammation, which in turn impact the health and animal performance. Additionally, chronic stress in poultry is linked with the emergence of antimicrobial resistance (AMR), which the WHO has recently identified to be among the most important problems threatening human health globally that increased the demand for safe antimicrobials to treat the collateral damages resulting from dysbiosis. Several alternative feed additives such as probiotics, prebiotics, fatty acids, and amino acids have been described to restore intestinal microbiota. Additionally, some phytogenic substances have anti-inflammatory and antioxidant activities. These natural products are also capable to modulate gut microbiota in a symbiotic equilibrium, thereby enabling the intestinal tract to withstand both infectious and non-infectious stressors. Nevertheless, several challenges, such as the bioavailability, rate of absorption, quality inconsistency, public acceptance, and cost-effective delivery methods, make the feasibility and application of phytogenic substances on a commercial scale complicated. In this review, the main drivers of chronic inflammation in poultry have been discussed. Additionally, the potential use of alternatives to antibiotics to restore the gastrointestinal microbiota in poultry and the possibilities for overcoming breakdowns in poultry farming were highlighted
Supplementation of Postbiotic RI11 Improves Antioxidant Enzymes Activity, Upregulated Gut Barrier Genes and Reduced Cytokines, Acute Phase Proteins and HSP70 Gene Expression Levels in Heat-Stressed Broilers

Abstract:Abstract Background To alleviate the adverse impacts of stressful environmental conditions on poultry and promoting the animal's health and growth performance, antibiotics have been added to poultry diets as growth promoters. Nevertheless, improper and overuse of antibiotics as feed additives have resulted the emergence of antibiotic-resistant bacteria and increased the levels of antibiotic residues in animal products, which have disastrous effects on the health of both animals and humans. Postbiotics produced from probiotic Lactobacillus plantarum have been the recent research of interest as dietary additives for livestock and potential alternatives to antibiotics. However, there is very scarce of study has considered the effect of postbiotics on broilers under heat stress. The aim of this work was to evaluate the impacts of feeding different levels of postbiotic RI11 on antioxidant enzyme activity, physiological stress indicators, cytokines and gut barrier genes expression in broilers under heat stress. Materials and Methods A total of 252 male Cobb 500 were fed with 1 of 7 diets: NC (negative control, 0.0% RI11) basal diet; OTC (positive control) NC + 0.02% (w/w) oxytetracycline; AA (antioxidant control) NC + 0.02% (w/w) ascorbic acid. Four further groups were NC + 0.2, 0.4, 0.6 and 0.8% postbiotic RI11 (v/w) of the respective levels. Results Supplementation of different levels of postbiotic RI11 increased plasma glutathione peroxidase, catalase and glutathione enzymes activity. Postbiotic RI11 groups upregulated the mRNA expression of interleukin 10 and downregulated of interleukin 8, tumor necrosis factor-alpha, heat shock protein 70 and alpha 1- acid glycoprotein levels compared to the NC and OTC groups. Feeding various doges of postbiotic RI11 improved the integrity of the intestinal barrier by the upregulation of zonula occludens-1 and mucin2 mRNA expressions. However, no difference was observed in claudin1, ceruloplasmin, interleukin 6, interleukin 2 and interferon expression, but downregulation for occludin expression as compared with the NC group. Supplementation of postbiotic RI11 in different levels quadratically increased the plasma glutathione peroxidase, catalase and glutathione, interleukin 10, mucin2 and zonula occludens-1 mRNA expression, and reduced plasma interleukin 8, tumor necrosis factor-alpha, alpha 1- acid glycoprotein and heat shock protein 70 mRNA expression. Supplementation of postbiotic RI11 at level 0.6% was sufficient to achieve the improvement in health of broiler chickens under heat stress as compared to other levels. Conclusions The results suggested that postbiotic produced from L. plantarum RI11 particularly at level 0.6% (v/w) could be used as an alternative to antibiotics and natural sources of antioxidants in the poultry industry.

Abstract:For many years, poultry production has relied on the use of antibiotics for disease treatment, prevention, and growth promotion among other perceived benefits. However, the excessive use of antibiotics especially when administered at sub-therapeutic levels is now facing growing consumer dissatisfaction due to the emergence of antibiotic resistance. Stakeholders in the animal feed industry are presently preoccupied with the need to develop alternative substances that will substitute synthetic antibiotics to ensure the safety of meat and other animal products. Among the substitutes for antibiotics, probiotics have appeared to be a viable option. In this review, positive attributes of supplementing probiotics in poultry are discussed. A literature search was performed with an electronic database such as Science Direct, Google Scholar, JURN, Directory of Open Access Journals, and ResearchGate. In the search process, the keywords were "Probiotic", "Gut modulation", "immune system", and "poultry". Searched articles were evaluated and sorted according to publication dates, and the levels of evidence were varying with probiotic microorganism species used, level of supplantation, and the method used for supplementary. Literature search results show that probiotics have numerous benefits on gut health and gut morphometrics and immune status of poultry through various mechanisms. Knowledge of these could enable an increase in the utilization of probiotics in poultry farming thereby reducing the challenges associated with the use of antibiotics.
The Effect of Clostridium butyricum on Gut Microbiota, Immune Response and Intestinal Barrier Function During the Development of Necrotic Enteritis in Chickens

Abstract:Necrotic enteritis (NE) causes huge economic losses to the poultry industry. Probiotics are used as potential alternatives to antibiotics to prevent NE. It is known that Clostridium butyricum can act as a probiotic that can prevent infection. However, whether or not it exerts a beneficial effect on NE in chickens remains elusive. Therefore, we investigated the impact of C. butyricum on immune response and intestinal microbiota during the development of NE in chickens, including experimental stages with basal diets, high-fishmeal-supplementation diets, and Clostridium perfringens challenge. Chickens were divided into two groups from day 1 to day 20: one group had its diet supplemented with C. butyricum supplementation and one did not. At day 20, the chickens were divided into four groups: C. perfringens challenged and unchallenged chickens with and without C. butyricum supplementation. All groups were fed a basal diet for 13 days and thereafter a basal diet with 50% fishmeal from day 14 to 24. Chickens were infected with C. perfringens from day 21 to 23. At days 13, 20 and 24, samples were collected for analysis of the relative expression of immune response and intestinal mucosa barrier-related genes and intestinal microbes. The results show that C. butyricum can inhibit the increase in IL-17A gene expression and the reduction in Claudin-1 gene induced-expression caused by C. perfringens challenge. Moreover, C. butyricum was found to increase the expression of anti-inflammatory IL-10 in infected chickens. Although C. butyricum was found to have a significant beneficial effect on the structure of intestinal bacteria in the basal diet groups and decrease the abundance of C. perfringens in the gut, it did not significantly affect the occurrence of intestinal lesions and did not significantly correct the shift in gut bacterial composition post C. perfringens infection. In conclusion, although C. butyricum promotes the expression of anti-inflammatory and tight junction protein genes and inhibits pro-inflammatory genes in C. perfringens-challenged chickens, it is not adequate to improve the structure of intestinal microbiota in NE chickens. Therefore, more effective schemes of C. butyricum supplementation to prevent and treat NE in chickens need to be identified.

Abstract:Along with banning antibiotics, necrotic enteritis (NE), especially subclinical enteritis (SNE), poses a significant threat to the chicken industry; however, probiotics are a potentially promising intervention. We aimed to investigate the beneficial effects of Bacillus subtilis DSM29784 (BS) on the treatment of Clostridium perfringens (CP)-induced SNE in broilers. A total of 360 1-day-old broiler chicks were divided into three treatment groups, namely control (Ctr), SNE, and BS treatment (BST) groups, all of which were fed with a basal died for 21days, and then from day 22 onward, only the BST group had a BS supplemented diet (1×10 9 colony-forming units BS/kg). On day 15, all chicks, except the Ctr group, were challenged with a 20-fold dose coccidiosis vaccine and 1ml CP (2×10 8 ) on days 18–21 for SNE induction. Beneficial effects were observed on growth performance in BST compared to SNE broilers. BST treatment alleviated intestinal lesions and increased the villus height/crypt depth ratio. Further, BST broilers showed increased maltase activity in the duodenum compared with SNE chicks, and a significantly decreased caspase-3 protein expression in the jejunum mucosa. Moreover, an increased abundance of Ruminococcaceae and Bifidobacterium beneficial gut bacteria and an altered gut metabolome were observed. Taken together, we demonstrate that the manipulation of microbial gut composition using probiotics may be a promising prevention strategy for SNE by improving the composition and metabolism of the intestinal microbiota, intestinal structure, and reducing inflammation and apoptosis. Hence, BS potentially has active ingredients that may be used as antibiotic substitutes and effectively reduces the economic losses caused by SNE. The findings of this study provide a scientific foundation for BS application in broiler feed in the future.
