Abstract:We investigated the antiproliferative effect of genistein, and its antileukemia effect in combination with cytosine arabinoside (ara-C) in acute myeloid leukemia (AML). Optimal dosage of genistein as single agent and in combination with ara-C was first determined in vitro. Genistein demonstrated a dose- and time-dependent inhibition of cell proliferation, induction of apoptosis, and cell-cycle arrest at G(2)/M phase. Gene-expression profiles revealed mitogen-activated protein kinase (MAPK) signaling as one of the most affected biological pathways. Phosphatidylinositol 3 kinase, protein kinase A, protein kinase C, MAPK kinase 4, KIT, PIM1, and transforming growth factor-beta receptor 1, were significantly downregulated by genistein. To test whether genistein could augment the antiproliferation activity of ara-C, two groups of severe combined immunodeficient mice were inoculated with NB4 and HL-60 cells, respectively, followed by treatment with either genistein or combination of genistein and ara-C. The combination treatment significantly inhibited tumor growth, and improved survival of NB4 (p = 0.0031) and HL-60 (p = 0.0007) xenograft mice. Our present study highlighted the schedule-dependent synergistic antileukemia effect of genistein with chemotherapy in both in vitro and in vivo models. This novel combination could potentially be a promising regimen for treatment of AML.
Sinomenine Enhanced Aclarubicin-Induced Apoptosis by Blocking Nf-Kappa B Pathway in Hl-60 Cells

Abstract:Sinomenine (SN) a component from Sinomenium acutum, widely used as an anti-inflammatory agent in Chinese traditional medicine, was reported to have anti-tumor activities. This study aims to examine the anti-leukemic activity of SN with or without aclarubicin (Acla) on leukemia cells and its molecular mechanism. HL-60 cells were treated with SN (5 to 20 ng/ml) with or without Acla (0.05 to 0.25 mu g/ml). To study apoptosis, the cells were stained with Annexin V - propidium iodide (PI) and assessed by flow cytometry. To investigate the molecular mechanism, Caspase 3, Caspase 9 and cyclooxygenase-2 (Cox-2) were assessed by Western blot, NF-kB activity and prostaglandin E2 (PGE2) were tested by ELISA. Exposure to Acla (even at 0.25 mu g/ml) alone or SN alone (20 ng/ml) for 20 h did not induce significant apoptosis. However, SN (5 to 20 ng/ml) promoted apoptosis induced by 0.1 mu g/ml Acla in a dose-dependent manner. Increased Caspase 3 and 9 protein expression correlated positively with SN concentration when combined with 0.1 mu g/ml Acla. In addition, SN significantly inhibited Acla-induced NF-kB activation, Cox-2 gene expression and PGE2 production. In summary, SN most likely enhances apoptosis by suppressing NF-kB activation, and Cox-2 and PGE2 expression. In the future, this natural agent may provide a new avenue for anti-leukemia treatment.
The Basic Research of the Combinatorial Therapy of ABT-199 and Homoharringtonine on Acute Myeloid Leukemia.

Abstract:BACKGROUND:Existing research shows that ABT-199, as a first-line drug, have been widely used in hematological malignancies, especially in leukemia, but the clinical efficacy of single drug therapy was limited part of the reason was that BCL-2 inhibitors failure to target other anti-apoptotic BCL-2 family proteins, such as MCL-1. In this case, combination therapy may be a promising way to overcome this obstacle. Here, we investigate the preclinical efficacy of a new strategy combining ABT-199 with homoharringtonine (HHT), a selective inhibitor of MCL-1 may be a promising approach for AML treatment as these two molecules are important in apoptosis.METHODS:A Cell Counting Kit-8 (CCK8) assay and flow cytometry were used to determine the half-maximal inhibitory concentration (IC50) value and cell apoptosis rate, respectively. The flow cytometry results showed that combined treatment with HHT and ABT-199 caused apoptosis in AML patient samples (n=5) but had no effect on normal healthy donor samples (n=11). Furthermore, we used a Western blot assay to explore the mechanism underlying the efficacy of HHT combined with ABT-199. Finally, antileukemic activity was further evaluated in vivo xenograft model.RESULTS:Our results indicated that ABT-199 combined with HHT significantly inhibited cell growth and promoted apoptosis in both AML cell lines and primary AML tumors in a dose- and time-dependent manner. Moreover, HHT combined with ABT-199 suppressed AML cell growth and progression in vivo xenograft model.CONCLUSIONS:Our research found that HHT combined with ABT-199 exerted its anti-leukemia effect by inducing apoptosis through the treatment of AML in vitro and in vivo.
[Effects of Berbamine on K562 Cells and Its Mechanisms in Vitro and in Vivo].

Abstract:To elucidate the antileukemi effects of berbamine and the possible molecular mechanisms in vitro and in vivo, MTT method was used to examine the effect of berbamine on K562 cell growth. The apoptosis rate was measured by flow cytometry. The mRNA expression level of BCR/ABL gene (semiquantity value) was determined by RT-PCR and the BCR/ABL protein (P210) level was detected by Western blot. The K562-bearing mice were used to reveal the therapeutic effect in vivo. The results showed that a significant time- and concentration-dependent inhibition of cell growth was found in the cells treated with berbamine. After the cells were exposed to 8.0 microg/ml berbamine for 24, 48 and 72 hours, the percentage of growth inhibition of K562 cells progressively increased by (26.63 +/- 3.57)%, (61.84 +/- 4.74)%, (75.32 +/- 1.95)%, respectively (compared with control, P < 0.01). The IC(50) (72 hours) value was 5.227 +/- 1.307 microg/ml. The apoptosis rate of K562 cells treated with 8.0 microg/ml berbamine for 24 and 72 hours increased from (29.20 +/- 3.82)% to (61.77 +/- 4.35)% (P < 0.01). Berbamine down-regulated the expression levels of bcr/abl gene and P210 in K562 cells in a time- and concentration-dependent manner. The bcr/abl expression decreased from (1.38 +/- 0.02) to (0.97 +/- 0.01) after exposure of the cells to 8.0 microg/ml berbamine for 0 and 72 hours (P < 0.01). When the cells were treated with 4.0 - 16.0 microg/ml berbamine for 24 hours, the level of P210 decreased from (0.95 +/- 0.03) to (0.63 +/- 0.01) (P < 0.01). In vivo, after treatment for 4 weeks, the tumor weight of berbamine-treated group was also lower than that of untreated group [(1.46 +/- 0.43) g vs (2.90 +/- 0.94) g, P < 0.01] and the inhibition rate was 49.66%, moreover, berbamine down-regulated the expression level of bcr/abl gene of tumor cells. It is concluded that berbamine can obviously inhibit the cell proliferation and induce apoptosis in K562 cell lines in a time- and concentration-dependent manner in vitro. The mechanisms of berbamine-induced apoptosis may be involved in down-regulation of bcr/abl gene expression and P210 level. In vivo, berbamine can aslo display a better antileukemic effect and down-regulate expression of bcr/abl gene. Berbamine extracted from Chinese herb may be a promising candidate of new drug for clinical anticancer treatment, especially for bcr-abl(+) diseases.
Homoharringtonine targets Smad3 and TGF-β pathway to inhibit the proliferation of acute myeloid leukemia cells.

Abstract:Homoharringtonine (HHT) has long and widely been used in China for the treatment of acute myeloid leukemia (AML), the clinical therapeutic effect is significant but the working mechanism is poorly understood. The purpose of this study is to screen the possible target for HHT with virtual screening and verify the findings by cell experiments. Software including Autodock, Python, and MGL tools were used, with HHT being the ligand and proteins from PI3K-Akt pathway, Jak-stat pathway, TGF-beta pathway and NK-kappa B pathway as the receptors. Human AML cell lines including U937, KG-1, THP-1 were cultured and used as the experiment cell lines. MTT assay was used for proliferation detection, flowcytometry was used to detect apoptosis and cell cycle arrest upon HHT functioning, western blotting was used to detect the protein level changes, viral shRNA transfection was used to suppress the expression level of the target protein candidate, and viral mRNA transfection was used for overexpression. Virtual screening revealed that smad3 from TGF-beta pathway might be the candidate for HHT binding. In AML cell line U937 and KG-1, HHT can induce the Ser423/425 phosphorylation of smad3, and this phosphorylation can subsequently activate the TGF-beta pathway, causing cell cycle arrest at G1 phase in U937 cells and apoptosis in KG-1 cells, knockdown of smad3 can impair the sensitivity of U937 cell to HHT, and over-expression of smad3 can re-establish the sensitivity in both cell lines. We conclude that smad3 is the probable target protein of HHT and plays an important role in the functioning mechanism of HHT.
Scutellarein Selectively Targets Multiple Myeloma Cells by Increasing Mitochondrial Superoxide Production and Activating Intrinsic Apoptosis Pathway.

Abstract:Objectives: Scutellarein is a flavonoid monomer found in traditional Chinese medicine such as Scutellaria barbata. This study aimed to investigate the cytotoxic effect of scutellarein treatment on multiple myeloma (MM) cells. Methods: circulating B lymphocytes (CBL) isolated from healthy donors’ peripheral blood served as control for MM.1R and IM-9 MM cells. CLB and MM cells were treated with various concentrations of scutellarein before their cell viability and apoptosis being evaluated. Nude mice burdened with MM xenograft tumor were intravenously injected with different concentrations of scutellarein, and their tumor burden change were monitored. Apoptosis of MM cells or CBL after scutellarein treatment was assayed by measuring caspase-3, -8 and -9 activities. FADD or APAF1 gene knockdown in MM cells was achieved by lentiviral transfection. Amount of Cytochrome C in cytosol or mitochondria as well as that of Bax and Bcl-2 protein were evaluated by Western blot. Mitochondria-induced apoptosis was assayed by measuring mitochondrial membrane potential change. Production of general reactive oxygen species and mitochondrial superoxide in MM or CBL was detected after scutellarein treatment, which was reduced by MitoTEMPO or apocynin treatment, respectively. Results: Scutellarein treatment showed potent cytotoxicity on MM cells but not on viable CBL, and intravenous injection of scutellarein significantly reduced MM xenograft tumor burden in nude mice. Scutellarein treatment in MM cells activated the mitochondrial-mediated intrinsic apoptosis pathway by increasing the production of mitochondrial superoxide, which was reduced to ROS by NADPH, but this effect was weakened in healthy CBL. Co-treatment with scutellarein synergized with bortezomib in inducing apoptosis in MM cells in vitro and in reducing tumor volume in MM xenografted nude mice. Conclusions: Scutellarein induced mitochondrial-mediated intrinsic apoptosis selectively on malignant cells comparing to healthy cells.
Beta-aescin: A Potent Natural Inhibitor of Proliferation and Inducer of Apoptosis in Human Chronic Myeloid Leukemia K562 Cells in Vitro

Abstract:Beta-aescin, a natural triterpenoid saponin isolated from the seed of Chinese horse chestnut (Aesculus chinensis), is known to generate a wide variety of biochemical and pharmacological effects. In the present study, the authors investigated the anti-proliferative and apoptotic effects of beta-aescin in human chronic myeloid leukemia K562 cell line in vitro. The anti-proliferative effects were detected by CFU-K562 colony formation and cell viability assay. The apoptotic effects were analysed by morphological analysis, annexin V assay, DNA fragmentation assay and flow cytometry DNA content analysis. The results showed that beta-aescin exhibited potent dose- and time-dependent anti-proliferative effects in K562 cells. Morphological evidence of apoptosis, a significant increase of annexin V+ and PI− cells (early apoptotic) and apoptotic DNA fragmentation, were observed in cells treated with beta-aescin. Flow cytometry analysis revealed that beta-aescin could lead to an accumulation of sub G1 population in K562 cells, and suggesting a potential G1 phase accumulation in cell cycle profile of K562 cells. Our findings revealed that beta-aescin is a potent natural inhibitor of proliferation and inducer of apoptosis in K562 cells, and beta-aescin may be a candidate lead compound to explore potential antileukemia drugs.
Influence of CDK4/6 on proliferation and apoptosis of acute leukemia cells

Abstract:Objective To investigate the influence of cyclin-dependent kinase(CDK) 4/6 on the proliferation and apoptosis of acute leukemia cells.Methods The acute leukemia cells U937 and RS4;11 were treated by the inhibitor of CDK4/6 PD0332991 0.01,0.1,1,10 and 20μmol/L for 24 and 48hours.The cell proliferation was detected by CCK-8 assay.The U937(group U)and RS4;11(group R)cells were treated by PD0332991 1μmol/L and 5μmol/L,respectively.Normal cells were taken as the controls(group CU and group CR).The cell apoptosis and cell cycle were examined by flow cytometry and the mRNA expressions of Bax,Bcl-2,X-linked inhibitor of apoptosis protein(XIAP),CDK2,CDK6 and Caspase-3 were detected by RT-PCR.Results The rate of cell proliferation was decreased with the increase of the concentration of PD0332991(P<0.05),which was more obvious after 48hours treatment than that after 24hours treatment(P<0.05).Compared to groups of CU and CR,the rate of cell apoptosis and proportion of G0/G1 were increased,the proportion of S and G2/M was decreased,the mRNA expressions of Bax and Caspase-3 were upregulated and those of Bcl-2,XIAP,CDK2 and CDK6 were downregulated after 24 hours treatment in groups of U and R(P<0.05).Conclusion The inhibition of CDK4/6 can inhibit the proliferation and induce the apoptosis of acute leukemia cells,which may be related to the upregulation of Bax and Caspase-3 and downregulation of Bcl-2,XIAP,CDK2 and CDK6.
Scutellarin improves the radiosensitivity of non‐small cell lung cancer cells to iodine‐125 seeds via downregulating the AKT / mTOR pathway

Abstract:<section class="article-section__content"><h3 class="article-section__sub-title section1"> Background</h3><p>In our previous study, we indicated that scutellarin (SCU) induced an anticancer effect in A549 cells. However, whether SCU regulates the radiosensitivity of non-small cell lung cancer (NSCLC) and its related mechanism is still unclear.</p></section><section class="article-section__content"><h3 class="article-section__sub-title section1"> Methods</h3><p>In this study, we explored the anticancer effect induced by iodine-125 (<sup>125</sup>I) and SCU at a sensitizing concentration in A549 and H1975 cells. Cellular apoptosis and proliferation were detected by flow cytometry, Bcl-2/Bax expression level, cell cycle, CCK-8, and EdU staining. A tumor model using nude mice was also carried out to investigate the combined effect of <sup>125</sup>I and SCU in vivo. In addition, the expression level of AKT/mTOR pathway was detected to investigate whether it is linked to the anticancer effect of <sup>125</sup>I and SCU. </p></section><section class="article-section__content"><h3 class="article-section__sub-title section1"> Results</h3><p>SCU at a sensitizing concentration promoted the <sup>125</sup>I-induced apoptosis and antiproliferative effect in A549 and H1975 cells. Abstract:Patients with lung squamous cell carcinoma (SCC) still benefit few from new therapies of lung cancer, and seeking for potential effective therapies is imperative. Honokiol (HNK) is a natural compound isolated from the magnolia plant with numerous pharmacological activities and exerts anticancer activity. Here, we demonstrated that the effect of HNK in lung SCC cells in vitro and related signaling mechanisms. Cell viability of human lung SCC cell lines NCI-H520 and SK-MES-1 exposed to HNK were measured by CCK-8. Flow cytometry was applied to identify the cellular apoptosis with HNK exposure of SCC cells compared to lung adenocarcinoma cells. Fibroblast growth factor(FGF)-2 expression of both SCC cells with HNK exposure were explored by quantitative real-time PCR. Furthermore, the effect of HNK on FGFR1 and signal pathways related to proliferation, apoptosis and cell cycle were detected by western blot. In two subpopulations of human lung SCC cells, NCI-H520 and SK-MES-1, HNK exposure remarkably inhibited the cell proliferation in both time-and dose-dependent manners. Compared to A549 cells, a more significant cell apoptosis was observed in NCI-H520 cells after HNK incubation. Next, real-time PCR analysis indicated that HNK down-regulated the expression of FGF2 in both NCI-H520 cells and SK-MES-1 cells in dose dependence. Meanwhile, HNK also obviously inhibited FGFR1 in both cell lines. Moreover, HNK exposure activated PARP and caused caspase-3 cleavage, suppressed cyclineD1 and ERK expression. These results broaden our understanding of the mechanisms on HNK effects in lung SCC, and reinforce the possibility of its potential anticancer benefit on SCC patients.
Salvia Miltiorrhiza Injection Inhibited the Proliferation of AML Cells by Inducing Apoptosis through the p38MAPK Pathway

Abstract:The purpose of this study was to explore the antitumor effect and mechanism of Salvia miltiorrhiza injection (SMI) on acute myeloid leukemia (AML) cells in vitro and in vivo. Bioinformatics was used to detect c-Myc mRNA expression in AML patients in the Oncomine database. qRT‒PCR and western blotting were used to detect the mRNA and protein expression of c-Myc and HOXA5 in clinical samples. Different concentrations (6.25, 12.5, 25, 50 and 100 μg/mL) of SMI were added to KG1a and HL60 cells for 24, 48 and 72 h to determine the IC 50 value of SMI. A CCK-8 assay was used to detect the effects of different concentrations of SMI and different treatment times on the proliferation of KG1a and HL60 cells. The indicated concentrations of SMI and SB203580 were used to treat KG1a and HL60 cells. The cell cycle distribution was determined by flow cytometry. The percentage of apoptotic cells was detected by Hoechst 33258 staining and flow cytometry. qRT‒PCR was performed to detect the mRNA expression of p38, c-Myc and HOXA5 in KG1a and HL60 cells. Western blotting was used to detect the protein expression of p38, p-p38, c-Myc, HOXA5, cCaspase 3 and cPARP in KG1a and HL60 cells. AutoDock software was used to analyze the molecular docking of the three main active components of SMI with c-Myc. AutoDock analysis revealed that the binding effect of molecular leisure was evaluated by binding energy, and a binding energy <−5 kcal/mol was considered good. SMI decreased the mRNA and protein expression of c-Myc and HOXA5. SMI significantly inhibited the proliferative activity of KG1a and HL60 cells and induced their apoptosis. However, SMI had no significant effect on the cell cycle distribution of KG1a and HL60 cells. With increasing SMI concentrations, the p-p38/p38 ratio increased, while the protein expression of c-Myc and HOXA5 decreased, and the protein expression of cCaspase and cPARP increased. However, SB203580 intervention in addition to SMI reversed these changes. Tanshinone IIA, cryptanshinone and salvianolic acid B can bind to multiple sites of c-Myc. In summary, SMI could be used for the treatment of acute leukemia, and its mechanism may be related to activation of the p38MAPK signaling pathway.

Pretreatment of Umbilical Cord Derived MSCs with IFN-γ and TNF-α Enhances the Tumor-Suppressive Effect on Acute Myeloid Leukemia.

Abstract:Currently, the standard therapeutic approach of AML consists of chemotherapy and allogeneic hematopoietic stem cell transplantation (HSCT). However, these strategies are usually associated with adverse side effects and high risk of relapse following HSCT. Thus, it is imperative to find an alternative way against AML progression. Here, we showed that treatment with umbilical cord-derived mesenchymal stem cells (UC-MSCs) could efficiently induce apoptosis in both primary AML patient-derived leukemic cells and AML cell lines. Mechanistically, tumor necrosis factor-alpha-related apoptosis-inducing ligand (TRAIL) in UC-MSCs mediated the proapoptotic effect in AML cells. Besides, indoleamine 2,3-dioxygenase (IDO) secreted by UC-MSCs blocked the cell cycle progression and inhibited the proliferation of AML cells. Importantly, we found that incubation of UC-MSCs with IFN-gamma and TNF-alpha could upregulate the expression of TRAIL and IDO, resulting in an intensive pro-apoptotic efficacy. UC-MSCs pre-treatment could not only relieve the AML burden but also eliminate AML cells in a xenograft AML model. Our findings have shed light on an effective pre-activated approach to aggravating the anti-leukemia effect of MSC. Furthermore, a novel and safe stem cell-based therapy approach for AML treatment.
Research on joint application of chrysanthemum morifolium and aclarubicin in promoting HL-60 cells apoptosis

Abstract:Objective To explore the effects of combined use of chrysanthemum morifolium and aclarubicin on inducing apoptosis of leukemia cells( HL-60). Results Low-concentration aclarubicin had little effect on apoptotic rate of HL-60cells,as well as caspase 3,caspase 9,COX-2,NF-κB expressions on HL-60 cells. Combined treatment of chrysanthemum morifolium and lower-concentration aclarubicin can enhance the apoptotic rate,expression of caspase 3 and caspase 9,down-regulate COX-2 expression. This effect was highly correlated with the concentration of chrysanthemum morifolium. NF-κB activity was obviously suppressed by combination of chrysanthemum morifolium and aclarubicin. Conclusion Chrysanthemum morifolium can enhance the aclarubicin-mediated apoptotic effect on HL-60 cells,up-regulate expression of caspase 3 and caspase 9,down-regulate COX-2 expression in HL-60 cells via suppressing activity of NF-κB.

Abstract:PURPOSE:FLT3 mutations occurred in approximately one third of patients with acute myeloid leukemia (AML). FLT3-ITD mutations caused the constitutive activation of the RAS/MAPK signaling pathway. Ribosomal S6 Kinases (RSKs) were serine/threonine kinases that function downstream of the Ras/Raf/MEK/ERK signaling pathway. However, roles and mechanisms of RSKs inhibitor LJH-685, and combinational effects of LJH-685 and FLT3 inhibitor FF-10101 on AML cells were till unclear.METHODS:Cell viability assay, CFSE assay, RT-qPCR, Colony formation assay, PI stain, Annexin-V/7-AAD double stain, Western blot, and Xenogeneic transplantation methods were used to used to investigate roles and mechanisms of LJH-685 in the leukemogenesis of AML.RESULTS:LJH-685 inhibited the proliferation and clone formation of AML cells, caused cell cycle arrest and induced the apoptosis of AML cells via inhibiting the RSK-YB-1 signaling pathway. MV4-11 and MOLM-13 cells carrying FLT3-ITD mutations were more sensitive to LJH-685 than that of other AML cell lines. Further studies suggested that LJH-685 combined with Daunorubicin or FF- 10101 synergistically inhibited the cell viability, promoted the apoptosis and caused cycle arrest of AML cells carrying FLT3-ITD mutations. Moreover, in vivo experiments also indicated that LJH-685 combined with FF-10101 or Daunorubicin prolonged the survival time of NSG mice and reduced the leukemogenesis of AML.CONCLUSION:Thus, these observations demonstrated combination of RSK inhibitor LJH-685 and FLT3 inhibitor FF-10101 showed synergism anti-leukemia effects in AML cell lines with FLT3-ITD mutations via inhibiting MAPK-RSKs-YB-1 pathway and provided new targets for therapeutic intervention especially for AML with FLT3-ITD mutations and Daunorubicin-resistant AML.

Abstract:Purpose: To investigate the effect and mechanism of action of Lithospermum erythrorhizon root extract (LR) in childhood acute leukemia. Methods: Human leukemic lymphoblast (CCRF-CEM cell line) cells were treated with LR (2, 4, and 8 mg/mL). Cell viability, cell apoptosis and cell cycle were measured using 3-(4,5-dimethyfthiazol-2-thiazolyl)-2,5-diphenyltetrazolium bromide (MTT) assay and flow cytometty, respectively. The levels of cell-cycle-related proteins including cyclin D1, cyclin El, cyclin-dependent kinase 2 (CDK2), CDK4 and CDK6, as well as cleaved caspase 3, cleaved caspase 9 and Bcl-2-associated x (Bax). Also determined were B-cell lymphoma-2 (Bcl-2), E-cadherin, N-cadherin, vimentin, zonula occludens 1 (ZO-1), matrix metalloproteinase-2 (MMP-2) and MMP-9. Cell migration and invasion were assessed using scratch and Transwell assays. Finally, phosphoinositide 3-kinase (PI3K), phosphorylated serine/threonine kinase (p-AKT), total AKT (t-AKT), mammalian target of rapamycin (mTOR), and phosphorylated mTOR (p-m TOR) were measured using western blotting. Results: Lithospermum erythrorhizon root extract not only dose-dependently inhibited cell viability, induced G1 phase accumulation, and downregulated CDK2, CDK4, CDK6, cyclin D1, and cyclin E1, but also elevated apoptosis, cleaved caspase 3, cleaved caspase 9, and Bax and decreased Bcl-2 expression levels. In addition, Lithospermum erythrorhizon root extract suppressed migration and invasion of CCRF-CEM cells, downregulated N-cadherin, vimentin, MMP-2, and MMP-9, and upregulated E-cadherin and ZO-1. Moreover, Lithospermum erythrorhizon root extracts dosedependently inhibited PI3K, p-AKT (ser473)/t-AKT, p-mTOR (ser2448)/mTOR, and p-mTOR (ser2481)/mTOR. Conclusion: The findings provide a potential therapeutic approach to the treatment of childhood acute leukemia.
Effect of scutellarin on BV-2 microglial-mediated apoptosis in PC12 cells via JAK2/STAT3 signalling pathway

Abstract:Previous studies have shown that scutellarin inhibits the excessive activation of microglia, reduces neuronal apoptosis, and exerts neuroprotective effects. However, whether scutellarin regulates activated microglia-mediated neuronal apoptosis and its mechanisms remains unclear. This study aimed to investigate whether scutellarin can attenuate PC12 cell apoptosis induced by activated microglia via the JAK2/STAT3 signalling pathway. Microglia were cultured in oxygen–glucose deprivation (OGD) medium, which acted as a conditioning medium (CM) to activate PC12 cells, to investigate the expression of apoptosis and JAK2/STAT3 signalling-related proteins. We observed that PC12 cells apoptosis in CM was significantly increased, the expression and fluorescence intensity of the pro-apoptotic protein Bax and apoptosis-related protein cleaved caspase-3 were increased, and expression of the anti-apoptotic protein B-cell lymphoma-2 (Bcl-2) was decreased. Phosphorylation levels and fluorescence intensity of the JAK2/STAT3 signalling pathway-related proteins JAK2 and STAT3 decreased. After treatment with scutellarin, PC12 cells apoptosis as well as cleaved caspase-3 and Bax protein expression and fluorescence intensity decreased. The expression and fluorescence intensity of Bcl-2, phosphorylated JAK2, and STAT3 increased. AG490, a specific inhibitor of the JAK2/STAT3 signalling pathway, was used. Our findings suggest that AG490 attenuates the effects of scutellarin. Our study revealed that scutellarin inhibited OGD-activated microglia-mediated PC12 cells apoptosis which was regulated via the JAK2/STAT3 signalling pathway.

Abstract:Objective:To explore the machanism of apoptosis induced by Ara-C in HL-60 cells.Methods:Cell viability was determined by trypan blue exclusion,the growth inhibition was analyzed by MTT assay;cell apoptosis by morphology and flow cytometry;Bcl-2 and caspase-3 by western blot.Results:Ara-C(1-100μM)inhibited cellular proliferation and induced apoptosis and reducde the levels of Bcl-2 proteins and activated caspase-3 in a dose and time dependent fashion in HL-60 cells.PD98059(25μM)(a inhibitor of MEK)enhanced apoptosis and activity of caspase-3 induced by Ara-C without altering Bcl-2 protein.However,SB203580(20μM)(a inhibitor of P38MAPK)reduced the apoptosis and activity of caspase-3 induced by Ara-C without altering Bcl-2 protein.SP600125(a inhibitor of JNK)did not affected effects of Ara-C.Conclusion:Ara-C downregulates Bcl-2 and activates caspase-3 during apoptosis in HL-60 cells;a inhibitor of MEK enhances apoptosis induced by Ara-C.However,a inhibitor of P38MAPK reduces the effects of Ara-C in HL-60 cells.

Abstract:Background: T-LAK cell-Originated Protein Kinase (TOPK) belongs to the serine/threonine protein kinase family. It is highly expressed in RPMI7951 melanoma cells. Scutellarin (SCU) is an active ingredient extracted from Erigeron breviscapus (Vant.) Hand.-Mazz. Its main physiological functions are related to its anti-inflammatory and antitumour activities.Methods: The relationship between SCU and TOPK was assessed by molecular docking, an in vitro binding assay and an in vitro kinase assay. The effect of SCU on RPMI7951 cells was detected by MTS and soft agar assays. TOPK knockdown was induced by lentiviral infection. The TOPK downstream signalling pathway was detected by western blot and immunohistochemical analyses in vitro and in vivo.Results: SCU was found to directly bind with TOPK and inhibit TOPK activity in vitro. SCU inhibited the proliferation and colony formation of RPMI7951 cells in a dose-dependent manner. Silencing TOPK decreased the sensitivity of colon cancer cells to SCU. SCU inhibited the phosphorylation levels of Extracellular Regulated protein Kinases 1/2 (ERK1/2) and histone H3 in a time-and dose-dependent manner in RPMI7951 cells. In addition, SCU inhibited the growth of xenograft tumours of RPMI7951 cells and decreased the phosphorylation levels of extracellular regulated protein kinases 1/2 and histone H3 in vivo.Conclusion: The results showed that SCU exerts promising antitumour effects on human RPMI7951 cells by inhibiting the activity of TOPK.

Abstract:Ethnopharmacological relevance Scutellaria baicalensis Georgi, a widely used Chinese medicinal herb, has shown effectiveness against lung cancer. Scutellarein, a key component of Scutellaria baicalensis , also demonstrates anticancer properties in lung cancer. However, the underlying mechanisms have not yet been clarified. Aim of the study This study aimed to investigate the effects of scutellarein in the treatment of NSCLC and its underlying mechanisms. Methods This study explored the effects of scutellarein on non–small cell lung cancer (NSCLC) and its mechanisms. A Lewis lung cancer mouse model was established to assess scutellarein's anticancer activity in vivo . Additionally, the compound's effects on cell proliferation, colony formation, migration, and apoptosis were evaluated in vitro using A549 and H1299 lung cancer cells. Metabolomics analysis was conducted to identify changes in cellular metabolism due to scutellarein, while molecular docking and western blotting techniques were employed to elucidate the molecular mechanisms of its anti-lung cancer effects. Results Scutellarein significantly inhibited lung cancer xenograft tumor growth. In vitro studies showed that scutellarein suppressed migration and colony formation in A549 and H1299 cells, induced cell cycle arrest, and triggered cell apoptosis. Notably, scutellarein profoundly altered amino acid metabolism, particularly affecting glutamine metabolites. It affected key glutamine transporters ASCT2 and LAT1, as well as glutaminase GLS1, leading to their reduced expression. Conclusion Scutellarein effectively inhibits lung cancer growth both in vivo and in vitro by inducing cell apoptosis and downregulating the glutamine metabolic pathway.

Abstract:Background : Scutellaria barbata D. Don (SB), commonly known as Ban Zhi Lian and firstly documented by Shigong Chen, is a dried whole plant that has been studied for its therapeutic effects on breast cancer, colon cancer, and prostate cancer. Among its various compounds, scutellarin (SCU) has been demonstrated with anti-tumor effects. Purpose : This study aimed to evaluate the effects of SB water extract (SBW) and scutellarin on breast cancer stem cells (BCSCs), and to investigate their potential therapeutic effects on breast tumors in mice. Methods : BCSCs were enriched from human breast cancer cells (MDA-MB-231 and MDA-MB-361) and their characteristics were analyzed. The effects of varying concentrations of SBW and scutellarin on cell viability, proliferation, self-renewal, and migration abilities were studied, along with the underlying mechanisms. The in vivo anti-tumor effects of scutellarin were further evaluated in SCID/NOD mice. Firstly, mice were inoculated with naïve BCSCs and subjected to treatment with scutellarin or vehicle. Secondly, BCSCs were pre-treated with scutellarin or vehicle prior to inoculation into mice. Results : The derived BCSCs expressed CD44, CD133 and ALDH1, but not CD24, indicating that BCSCs have been successfully induced from both MDA-MB-231 and MDA-MB-361 cells. Both SBW and scutellarin reduced the viability, proliferation, sphere and colony formation, and migration of BCSCs. In mice with tumors derived from naïve BCSCs, scutellarin significantly reduced tumor growth, expression of proliferative (Ki67) and stem cell markers (CD44), and lung metastasis. In addition, pre-treatment with scutellarin also slowed tumor growth. Western blot results suggested the involvement of Wnt/β-catenin, NF-κB, and PTEN/Akt/mTOR signaling pathways underlying the inhibitory effects of scutellarin. Conclusion : Our study demonstrated for the first time that both SB water extract and scutellarin could reduce the proliferation and migration of BCSCs in vitro. Scutellarin was shown to possess novel inhibitory activities in BCSCs progression. These findings suggest that Scutellaria barbata water extract, in particular, scutellarin, may have potential to be further developed as an adjuvant therapy for reducing breast cancer recurrence.
