Abstract:Ulcerative colitis has been consistently associated with gut microbiota imbalance and disturbed immune system. Emerging research suggests a protective function of polyphenols on prevention and treatment of ulcerative colitis, yet underlying mechanisms remain unclear. Fu brick tea, a postfermented tea, contains abundant polyphenols with anti-inflammatory and antioxidant properties. In the present study, we found that prophylactic supplementation of polyphenols extracted from Fu brick tea (FBTP) dose-dependently alleviated colitis symptoms, immune cells infiltration, and pro-inflammatory cytokines secretion in mice suffering dextran sulfate sodium induced murine colitis. FBTP substantially reshaped gut microbiota and promoted microbial transformation of tryptophan into indole-3-acetic acid (I3A), thereafter leading to aryl hydrocarbon receptor (AHR)-mediated protection from colitis through enhanced expressions of IL-22 and tight junction proteins (i.e., ZO-1, occluding and claudin-1) in colon. Multiomics integration analyses revealed strong connections between I3A, tryptophan-metabolizing bacteria, AHR activity, and pathological phenotypes of colitis. Notably, FBTP failed to significantly alleviate colitis symptoms in the absence of gut microbiota, while intragastric administration of I3A could imitate benefits of FBTP on colitis alleviation and intestinal epithelial homeostasis through a direct enhancement in AHR activity in microbiota-depleted mice. These findings further determine the key role of gut microbiota controlled I3A-AHR signaling in mediating the FBTP on colitis alleviation. This study provides the first data proposing the FBTP as a natural prebiotic for colitis alleviation through the gut microbiota-dependent modulation of the AHR pathway. Most importantly, we also identified I3A as a key microbial metabolite targeted by FBTP for exhibiting health-promoting effects.
Effects of Fu brick tea polysaccharides on gut microbiota and fecal metabolites of HFD/STZ-induced type 2 diabetes rats

Abstract:The prevalence of type 2 diabetes mellitus (T2DM) has dramatically increased globally, and the antidiabetic effects and underlying mechanisms of the polysaccharides extracted from Fu brick tea (FBTP) were investigated in high-fat diet (HFD)/streptozotocin (STZ)-induced T2DM rats. Administration of FBTP at 200 and 400 mg per kg bw significantly relieved dyslipidemia (i.e. TC, TG, LDL-C and HDL-C), insulin resistance (IR) and pancreas oxidative stress (i.e. CAT and GSH-Px) in T2DM rats. Mechanistically, FBTP rescued the HFD/STZ-induced alterations in the abundance of Bacteroidota, Actinobacteriota, Proteobacteria and Firmicutes. At the genus level, FBTP notably increased the abundance of Ruminococcus, Lactobacillus and Lachnospiraece_NK4A136_group, but reduced the population of Prevotella and Faecalibaculum in T2DM rats. FBTP also significantly elevated colonic short-chain fatty acid (SCFAs) levels. Moreover, apparent changes in amino acid absorption and metabolism were observed upon FBTP intervention. These findings suggested that FBTP might alleviate T2DM by reshaping the gut microbiota and regulating intestinal metabolites.

Abstract:Liubao tea, a traditional dark tea, has gained widespread recognition for various health benefits. In this study, the effects of Liubao tea extract (LTE) on obesity-related hyperlipidemia and the potential mechanism involved were explored. Anti-obesity compounds such as tricetin, isovitexin, tiliroside, etc. in LTE were identified. In high-fat diet mouse models, LTE effectively reduced tissues, organs, and body weight growth, and restored abnormal serum lipid levels. LTE could reverse adipocyte enlargement, lipid accumulation, and hepatic microstructure abnormalities. Notably, LTE reshaped gut microbiota by boosting beneficial bacteria (e.g., Bacteroides, Akkermansia, Psychrobacter) and suppressing harmful bacteria (e.g., Dubosiella, Faecalibaculum). Spearman correlation analysis unveiled significant associations between serum lipid levels, weight gain, LTE dosage, and gut microbiota, underlining the modulatory effects of LTE on metabolic disorders via the regulation of intestinal microbiota. Collectively, LTE could serve as a potential therapy for obesity-related hyperlipidemia prevention.

Abstract:Polysaccharides from Fuzhuan brick tea (FBTPS), one of most important bioactive components in tea, showed various health-promoting functions. Our previous work demonstrated that the crude FBTPS (CFBTPS) could modulate the gut microbiota. However, which purified fraction in CFBTPS contributing to the modulation of gut microbiota remains unclear. Thus, the fermentation characteristics and probiotic activity of a purified fraction (FBTPS-2-1) of CFBTPS were evaluated in this work. The results showed that gut microbiota could utilize FBTPS-2-1 to produce short-chain fatty acids including acetic, propionic, n-butyric and n-valeric acids. FBTPS-2-1 could modulate the structure and metabolic pathways of gut microbiota. FBTPS-2-1 could increase the health-promoting gut microbiota such as Prevotellaceae and Bifidobacteriaceae, and decreased the harmful bacteria such as Enterobacteriaceae and Fusobacteriaceae. The results of metagenomics showed that Prevotella copri and Megamonas funiformis were the dominant bacteria after fermentation of FBTPS-2-1. Furthermore, FBTPS-2-1 could regulate the biosynthesis and metabolism pathways of gut microbiota. Thus, the enrichment of food with FBTPS-2-1 is expected as a potential strategy for promoting human health due to modulation of gut microbiota.

Fu brick tea protects against high-fat diet-induced obesity phenotypes via promoting adipose browning and thermogenesis in association with gut microbiota

Abstract:Fu brick tea is one of the most famous microbially fermented teas that has received considerable attention owing to its promising anti-obesity capacity; however, the underlying mechanisms of its action remain largely unexplored. Herein, an eight-week oral administration of Fu brick tea aqueous extract (FTE) was observed to dose-dependently reduce body weight and abnormal fat accumulation for inguinal white adipose tissue, stimulate beige-fat development and thermogenesis in the brown adipose tissue of mice fed with a high-fat diet (HFD) (p < 0.05). FTE ameliorated HFD-induced gut dysbacteriosis and improved the microbiome ecology such that it exhibited an increased capacity to reduce the host adiposity, abnormal glycometabolism, and hepatic steatosis. FTE increased the abundance of beneficial bacteria strains, e.g., Lactobacillus, Roseburia, Bifidobacterium, Akkermansia, Parabacteroides, and Prevotella, accompanied with the improved production of short-chain fatty acids (p < 0.05). Moreover, the PICRUSt pathway analysis revealed that FTE upregulated genes enriched in pathways of the carbohydrate metabolism, signaling molecules and immune system. As a rising star of post-fermented teas with the low cost, high accessibility and confirmed health benefits, our findings indicate the beneficial impacts of Fu brick tea on the promotion of adipose browning and thermogenesis in association with gut microbiota reconstructions, paving the way to restrict obesity and its associated metabolic diseases.

Abstract:Fu brick tea (FBT), a post-fermented dark tea, is highly esteemed for its abundant nutritional and medicinal values. Fu brick polysaccharides (FBTPs) are acidic heteropolysaccharides primarily composed of galactose and galacturonic acid, which are crucial components of FBT. FBTPs exhibit multiple bioactivities, including immunomodulatory, antioxidant, anti-inflammatory, regulatory effects on intestinal microbiota, anti-obesity, among others. Owing to their significant marketing potential and promising development prospects, FBTPs have attracted considerable attention from researchers worldwide. However, the specific mechanisms and underlying structure-function relationships of FBTPs are not well understood. Consequently, this review aims to provide comprehensive and cutting-edge information on the extraction, purification, structural characteristics, and biological activities of FBTPs, with an emphasis on exploring how their structural characteristics influence biological activities and therapeutic potential. We found that different materials and extraction techniques could result in differences in the structure-activity relationship of FBTPs. Furthermore, monosaccharide composition and molecular weight could also significantly impact the bioactivities of FBTPs, such as lipid-lowering effects and immunomodulatory activity. This review would further facilitate the applications of FBTPs as therapeutic agents and functional foods, thereby laying a solid foundation for their further development and utilization.

Abstract:Dietary fiber is a carbohydrate polymer with ten or more monomeric units that are resistant to digestion by human digestive enzymes, and it has gained widespread attention due to its significant role in health improvement through regulating gut microbiota. In this review, we summarized the interaction between dietary fiber, gut microbiota, and obesity, and the beneficial effects of dietary fiber on obesity through the modulation of microbiota, such as modifying selective microbial composition, producing starch-degrading enzymes, improving gut barrier function, reducing the inflammatory response, reducing trimethylamine N-oxide, and promoting the production of gut microbial metabolites (e.g., short chain fatty acids, bile acids, ferulic acid, and succinate). In addition, factors affecting the gut microbiota composition and metabolites by dietary fiber (length of the chain, monosaccharide composition, glycosidic bonds) were also concluded. Moreover, strategies for enhancing the biological activity of dietary fiber (fermentation technology, ultrasonic modification, nanotechnology, and microfluidization) were subsequently discussed. This review may provide clues for deeply exploring the structure-activity relationship between dietary fiber and antiobesity properties by targeting specific gut microbiota.
Pomegranate Fruit Pulp Polyphenols Reduce Diet-Induced Obesity with Modulation of Gut Microbiota in Mice.

Abstract:BACKGROUND Pomegranate is a rich source of polyphenols and has been used as a dietary supplement and pharmaceutical ingredient. This study aimed to investigate the pomegranate fruit pulp polyphenols (PFP) with regard to their anti-obesity activity and gut microbiota-modulating effect in mice. Thirty-six 4-week-old specific pathogen-free C57BL/6J mice (weight: 17.7-20.8 g) were randomly divided into three groups and fed with low-fat diet (10% fat energy), high-fat diet (HFD) (45% fat energy), or HFD supplemented with PFP by intragastric administration for 14 weeks. The obesity-related clinical indicators were investigated, and the composition of fecal microbiota was analyzed by 16S rRNA sequencing. RESULTS Our results showed that PFP treatment reduced HFD-induced body weight gain by 35.23% (P < 0.05), steatosis scores by 50% (P < 0.05) and insulin resistance by 56.84% (P < 0.05), compared with the mice fed HFD alone. Moreover, compared with the mice in the HFD group, PFP supplement changed the composition of the gut microbiota, and enriched Akkermansia muciniphila, Parabacteroides distasonis, Bacteroides acidifaciens, Mucispirillum schaedleri and Lachnospiraceae bacterium 28-4, which were negatively correlated with physical biomarkers, including body weight, glucose, triglycerides and total cholesterol. CONCLUSION PFP alleviated HFD-induced obesity, insulin resistance and hepatic steatosis in mice, and the changes in the gut microbiota might be one of the potential mechanisms through which PFP improved obesity and obesity-related disorders, eventually benefiting the recipient. Abstract:Fu-brick tea (FBT) is predominately fermented by Eurotium Cristatum, FBT polysaccharides (FTPs) and Eurotium Cristatum extracellular polysaccharides (ECPs) are the main active substances in FBT and Eurotium Cristatum, respectively. FTPs was shown to exhibit higher levels of uronic acids, proteins, and polyphenols as compared to ECPs (p < 0.05), contributing to the superior antioxidant activity observed in FTPs. Additionally, FTPs had better water solubility and thermal stability than ECPs. Interestingly, in vitro digestive simulation revealed that FTPs and ECPs resist digestion in the stomach and small intestine. Excitingly, utilizing in vitro fermentation with feces from healthy individuals and type 2 diabetes mellitus (T2DM) patients demonstrated that FTPs and ECPs promote the production of SCFAs. Still, FTPs resulted in greater SCFAs contents than ECPs (p < 0.05). Moreover, FTPs and ECPs fermentation by T2DM patients' fecal microbiota affected different metabolomic pathways. Our findings suggested that FTPs holds great promise for application in functional foods.

Abstract:Fu Brick tea belongs to fermented dark tea, which is one of the six categories of tea. Fu Brick tea has been reported to reduce adiposity and has beneficial effects for hypercholesterolemia and cardiovascular disease. Theabrownin (TB) is one of the pigments with the most abundant content in Fu Brick tea. TB has also been reported to have lipid-lowering effects, but its mechanism remains unclear. We found that TB could effectively reduce the insulin resistance and fat deposition induced by high fat diet (HFD), decrease inflammation in liver, improve intestinal integrity, and reduce endotoxin in circulation. Further studies showed that TB increased the abundance of Verrucomicrobiota and reduced the abundance of Firmicutes and Desulfobacterota in the intestinal tract of obese mice. The alteration of gut microbiota is closely linked with the metabolic phenotype after TB treatment through correlation analysis. Moreover, TB changed the gut microbial metabolites including L-ornithine, α-ketoglutarate, and glutamine, which also been found to be upregulated in liver after TB intervention. In vitro, L-ornithine, α-ketoglutarate, or glutamine significantly reduced lipopolysaccharide (LPS)-induced inflammation in macrophage. Therefore, our results suggest that TB can reduce adiposity, systemic insulin resistance, and liver inflammation induced by HFD through altering gut microbiota and improving intestinal tight junction integrity. Microbial metabolites might also play a role in ameliorating the HFD-induced phenotype by TB.

Abstract:The effect of Fuzhuan brick tea (FBT) on metabolism in obese mice is mediated by regulation of N-methyltransferase by aryl hydrocarbon receptor. The expression of the phosphatidylethanolamine N-methyltransferase gene is regulated by many transcription factors, and those specific to this effect need further investigation. Experimental animal studies have been designed to observe the effects of a single drug or the sequential effects of drugs. A washout period should be included if different drugs (e.g., antibiotics and FBT) are given to avoid or reduce additive effects or synergy. Currently, most experimental studies performed in mice used only male animals. However, experience has revealed that the results of using only male mice are very likely to have sex differences.
Fuzhuan brick tea ameliorates hepatic steatosis and steatohepatitis through gut microbiota-derived aryl hydrocarbon receptor ligands in high-fat diet-induced obese mice

Abstract:High-fat diet (HFD) induced obesity and its associated conditions, such as hepatic steatosis and steatohepatitis, are major health concerns worldwide. Previous studies have reported the excellent efficiency of Fuzhuan brick tea (FBT) in attenuating HFD-induced obesity and metabolic disorders. In this study, we investigated the effects of FBT on hepatic steatosis and simple steatohepatitis in HFD-induced obese mice, as well as the metabolic function of the gut microbiome using metagenomics and metabolomics. The results showed that FBT ameliorated dyslipidemia, hepatic steatosis and steatohepatitis in HFD-induced obese mice by normalizing the gut microbiota structure and tryptophan metabolism. FBT increased the cecal abundance of aryl hydrocarbon receptor (AhR)-ligand producing bacteria such as Lactobacillus_reuteri and Lactobacillus_johnsonii, at the expense of AhR-ligand consuming bacteria, such as Faecalibaculum_rodentium and Escherichia_coli, and elevated the cecal contents of AhR-ligands such as IAA, IPA, and KYNA. Furthermore, FBT regulated the expressions of AhR and its targeted lipometabolic genes such as Pemt, Fasn, and SREBP-1c, as well as other inflammatory genes including TNF-α, IL-6, and IL-1β in the liver of mice. Overall, these findings highlight the beneficial effects of FBT on obesity-related hepatic steatosis and steatohepatitis via microbiota-derived AhR signaling.
Anti-obesity and Hypolipidemic Effects of Fuzhuan Brick Tea Water Extract in High-Fat Diet-Induced Obese Rats.

Abstract:Background Fuzhuan brick tea is a kind of microbial fermented tea, which has received increasing attention in recent years owing to its benefits for human health. In this study, the anti-obesity and hypolipidemic effects of Fuzhuan brick tea water extracts (FTEs) were investigated. Results FTEs consisted of 204.07 +/- 3.38 mg g1 polyphenol, 109.20 +/- 1.36 mg g1 flavonoids, and others. The FTEs significantly suppressed the increase of body weight and accumulation of adipose tissue, and reduced the level of serum triacylglycerol, total cholesterol and low-density lipoprotein (LDL) cholesterol in obese rats fed a high-fat diet. Moreover, FTEs attenuated the gene expression of sterol regulatory element binding protein-1c, fatty acid synthase and CCAAT/enhancer binding protein , which is related to lipogenic metabolism. In contrast, the gene expressions of enzymes involved in energy expenditure and lipodieresis including hepatic peroxisome proliferator-activated receptor , carnitine palmitoyltransferase 1a and LDL receptor gene expression were increased by FTE treatment. Conclusions Our study demonstrates that FTEs have anti-obesity and hypolipidemic functions, suggesting that it might be effective for treatment of obesity and hyperlipemia. Abstract:Fu brick tea (FBT) and its extracts have good lipid-lowering effects and have been used in the treatment of obesity in previous studies. Unfortunately, the therapeutic effect of FBT on nonalcoholic fatty liver disease (NAFLD) has not been thoroughly studied. In this study, we explored the mechanism by which FBT alleviates NAFLD from the perspective of the gut microbiota and liver metabolites. The results showed that FBT could reduce the body weight, liver weight and abdominal fat of NAFLD mice, and improve liver pathological morphology, liver lipid deposition, blood lipids and liver function. Moreover, FBT improved the diversity of the gut microbiota and changed the profile of liver metabolism in NAFLD mice. Further studies showed that FBT could ameliorate the cecum barrier, and regulate the effects of factors related to lipid synthesis in the cecum and liver of NAFLD mice. In conclusion, the present study confirmed that FBT can alleviate high fat induced NAFLD by regulating the homeostasis of the gut microbiota and liver metabolites.

Abstract:As living standards advance, an escalating emphasis is placed on health, particularly in relation to prevalent chronic metabolic disorders. It is necessary to explore safe and effective functional foods or drugs. Fu brick tea (FBT) is a kind of dark tea fermented by fungi. The extracts are rich in compounds that can effectively relieve metabolic diseases such as hyperglycemia and hyperlipidemia, protect the liver, improve human immunity, enhance antioxidant activity, and regulate intestinal flora. This paper summarizes the biological activities and mechanisms of the extracts, polysaccharides, and small molecular compounds of FBT, which provides a certain theoretical basis for the rational, systematic, comprehensive development and utilization of the FBT resources. It is expected to develop and apply these active substances in health care products and natural medicines and provide more beneficial and diversified FBT products for human beings.
Moringa-Fu brick tea extract attenuated high-fat diet-induced obesity via modulating bile acid metabolism and gut microbiota in rats

Abstract:Obesity is regarded to be correlated with disorders of lipid metabolism, bile acid metabolism and dysbiosis of the gut microbiota. Moringa-Fu brick tea (MFBT), a functional food, has a significant anti-obesity effect. However, the effectiveness and mechanisms of MFBT in preventing obesity require further investigation. Here, our results show that moringa-Fu brick tea extract (MFTE) displays anti-obesity effects by reducing high-fat diet-induced weight gain and adipose tissue accumulation in rats, and attenuating visceral adiposity index, oxidative stress and inflammation. In addition, MFTE could regulate gut microbiota diversity, reduce the relative abundance of microbioa associated with bile salt hydrolase (BSH) activity, such as Blautia , Ruminococcaceae , Coprococcus and Roseburia . Moreover, MFTE could improve the expression of the main rate-limiting enzymes CYP7A1 and CYP27A1 in the process of bile acid synthesis, downregulate the expression of intestinal FXR protein, promote the serum bile acid level, and reduce the accumulation of cholesterol in serum and liver. Taken together, these results suggested that MFBT could serve as a functional food for prevention of obesity and hyperlipidemia in association with regulating bile acid metabolism and gut microbiota.

Tea (Camellia sinensis L.) flower polysaccharide attenuates metabolic syndrome in high-fat diet induced mice in association with modulation of gut microbiota

Abstract:There is a growing body of evidence suggesting that dietary polysaccharides play a crucial role in preventing metabolic syndrome (MetS) through their interaction with gut microbes. Tea (Camellia sinensis L.) flower polysacchride (TFPS) is a novel functional compound known for its diverse beneficial effects in both vivo and vitro. To further investigate the effects of TFPS on MetS and gut microbiota, and the possible association between gut microbiota and their activities, this study was carried out on mice that were fed a high-fat diet (HFD) and given oral TFPS at a dose of 400 and 800 mg/kg·body weight (BW)/d, respectively. TFPS treatment significantly mitigated HFD-induced MetS, evidenced by reductions in body weight, fat accumulation, plasma levels of total cholesterol (TC), triglycerides (TG), low-density lipoprotein cholesterol (LDL-C), and pro-inflammatory cytokines tumor necrosis factor α (TNF-α), interleukin 6 (IL-6), and IL-1β, along with an increase in plasma IL-10 levels. Furthermore, TFPS induced alterations in the diversity and composition of HFD-induced gut microbiota. Specifically, TFPS influenced the relative abundance of 11 genera, including Lactobacillus and Lactococcus, which showed strong correlations with metabolic improvements and likely contributed to the amelioration of MetS. In conclusion, TFPS exhibits promising prebiotic properties in preventing MetS and regulating gut microbiota.

Abstract:Both tea polyphenols (TP) and Lactobacillus rhamnosus have been shown to alleviate obesity, and regulate lipid metabolism. However, the combined effects and their underlying mechanisms of action remain elusive. In the present work, the effects of TP, Lactobacillus rhamnosus R5 (R5), and TP+R5 on blood lipids and the gut microbiota of mice fed with a high-fat diet were compared. Results showed that the combination of TP and R5 effectively increased the serum levels of high-density lipoprotein cholesterol (HDL-C), and decreased total cholesterol (TC) levels, low-density lipoprotein cholesterol (LDL-C) levels, and the atherosclerosis index (AI) in mice fed with a high-fat diet. The combination treatment resulted in a modification of the structure of the gut microbiota in mice, as evidenced by a decrease in the F/B ratio, and an increase in the abundance of beneficial genera, such as Akkermansia muciniphila, Faecalibaculum rodentium, and Ruminococcus_uncultured bacterium. Additionally, the contents of SCFAs (acetic, propionic, and butyric acids) in faeces also increased. These could provide new ideas for anti-obesity methods, and a theoretical basis for the development of foods combining probiotics and tea polyphenols.

Fu Brick Tea as a Staple Food Supplement Attenuates High Fat Diet Induced Obesity in Mice

Abstract:Fu brick tea (FBT), a product of microbial fermentation from primary dark tea, also known as raw material tea (RMT), has been extensively studied for its functional properties. However, its potential as a staple food supplement for weight loss remains poorly understood. This study compared the weight loss effects of orlistat, traditional plain noodles (NN), and noodles supplemented with varying amounts of RMT (RMTN) and FBT (FBTN), with the aim to elucidate their lipid-reducing effects and underlying mechanisms. Experimental trials on high fat diet fed mice revealed significant weight loss, lipid-lowering, and hypoglycemic effects upon supplementation with orlistat, RMTN, and FBTN. Moreover, supplementation with orlistat, RMTN, and FBTN effectively restored serum and liver-related index levels, mitigating high-fat diet-induced dyslipidemia. Additionally, these supplements ameliorated liver and kidney damage by inhibiting oxidative stress and inflammatory responses. Furthermore, orlistat, RMTN, and FBTN exert their anti-obesity effects primarily by modulating genes associated with lipid metabolism and inflammatory responses and through regulation of the composition and structure of the gut microbiota. Importantly, FBTN demonstrated a significantly stronger lipid-lowering effect compared to RMTN, particularly at higher tea addition ratios. In contrast, NN supplementation exhibited minimal to no weight loss effects. Based on these findings, it could be inferred that FBT holds promise as a staple food supplement to ameliorate high-fat diet-induced obesity and its associated health conditions.
