Abstract:Platelets have the functions of promoting blood coagulation and accelerating hemostasis, playing an important role in human body. It is of great medical significance to realize clinical rapid micro-detection of platelets by spectral analysis, which is the development direction of clinical detection in the future. However, due to the problem of unobvious characteristic of platelet absorption spectrum, the results of modeling and analysis cannot meet the clinical accuracy requirements. In order to improve the analysis accuracy, based on the "M+N" theory, this paper comprehensively considers the influence of the concentrations of measured component platelet and non-measured component hemoglobin on modeling analysis, and uses the method of selecting training set based on the concentration distribution of two components. At the same time, considering the characteristic of the linear model, the samples at both ends of the concentration of two components are selected as the training set, and the cubic term fitting method is combined to model and predict the concentration of platelet. The following experiments were designed: the training sets were selected by four different methods and used for modeling to predict the platelet concentration, and compared the modeling results of different methods. Through the modeling and prediction of 222 samples, the result showed that the method of selecting the training set with the concentration distribution of two components could effectively improve the prediction accuracy of the established model, and got a better model with better performance, the correlation coefficient Rc reached 0.63, which was 24.98% higher than the result of full modeling for all samples, and RMSE decreased by 10.02%. Considering the influence of non-measured components in modeling is of great significance to improve the prediction accuracy of measured components, and selecting samples from both ends of the concentration values of two components as the training set can further improve the performance and accuracy of the model.
The Clinical Application of Leukocyte VCS Parameters in the Detection of Blood Bacterial Infection

Abstract:Objective To investigate the clinical application of VCS parameters of leukocyte in the detection of blood bacterial infection, Methods The subjects consisted of 120 patients with blood bacterial infection,69 non-infectious fever patients and 67 health controls.The VCS parameters of neutrophil and lymphocyte were examined with Coulter LH 750 hematology analyzer.The parameters examined including mean channel of neutrophil volume(MNV),neutrophil volume distribution width (NDW) ,mean channel of neutrophil conductivity (MNC),mean channel of neutrophil scatter (MNS),mean channel of lymphocyte volme(MLV),lymphocyte volume distfibufion width (LDW),mean channel of lymphocyte conductivity (MLC) and mean channel of lymphocyte scatter (MLS).Additionally,120 blood bacterial infection patients were grouped according to WBC count(WBC≤10×109/L group and WBC＞10×109/L group),neutrophii rate(≥85%group and＜85%group)and bacterial stain(Gram positive bacteria group and Gram negative bacteria group).VCS parameters among these groups were compared.Results The results of blood infection group were as follows:MNV 156±15,NDW 23.31±3.72,MNS 137±7,MLV 87±12,LDW 17.50±3.38.MLC 110±5 and MLS 69±12.The results of non-infectious fever group were as follows:MNV 151±8,NDW 21.33 ±2.62,MNS 132±10,MLV 91±4.LDW 15.78±1.96.MLC 117±4 and MLS 62±6.The results of control group were as follows:MNV145 ±5.NDW 18.43±0.93.MNS 143 ±4,MLV 84±2,LDW 13.30±0.76.MLC 108±1 and MLS 62±2.There were significant diffierences among these three groups (F value were 19.295,26.272,32.767,6.226,31.016,23.739 and 12.662 respectively,P＜0.05 or P＜0.01).In the infection group.the MNV and NDW were 152 ±16 and 22.19±3.45 respectively for WBC≤10×109/L group.159±12 and 25.29±3.43 respectively for WBC＞10×109/L group.They were both significantly different compared with control group (F valRe were 21.575 and 40.856 respectively,P＜0.01).Also in the infection group.the MNV and NDW were 159±12 and 24.88 ±3.74 respectively for neutrophil rate≥85%group.151±16 and 21.68±2.29 respectively for neutrophil rate＜85%group.They were both significantly different compared with control group(F value were 23.76 and 43.22 respectively,P＜0.01).The MNV and NDW were 157±15 and 24.25±3.39 respectively in those cases with gram-negative bacteremia,153±14 and 21.51±3.78 respectively in those cases with gram-positive bacteremia.They were both signifieanfly difierent compared with control group (F value were 18.74 and 37.47 respectively,P＜0.01).With a cut-off value of 20.50 for the NDW,a sensitivity of 76.7%and specificity of 98.3% were achieyed in diagnosing blood hacterial infection.Conclusion The VCS parameters can reflect the morphologic change of leukocyte in blood bacterial infection.Additionally.the NDW can detect blood bacterial infection more sensitively and specifically.
Enhancing Model Robustness Through Different Optimization Methods and 1-D CNN to Eliminate the Variations in Size and Detection Position for Apple SSC Determination

Abstract:The visible/near-infrared (Vis/NIR) spectroscopy technique has been widely used for the online detection of soluble solids content (SSC) in apples. However, external factors such as sample size and detection position can cause spectral distortion, resulting in a decline in detection accuracy. In this study, we aimed to develop a more robust prediction model that can resist the impact of sample size and detection position on the model. Firstly, we collected and analyzed the transmission spectra of apple samples under different sizes and detection positions using a self-designed Vis/NIR spectroscopy online acquisition device. It was found that the effect of fruit size and detection position on Vis/NIR spectra was due to optical path difference. Thus, a diameter correction method was utilized to uniformly correct the obtained absorbance spectra. The performance of local models achieved better results after correction. And then, global models with various preprocessing methods were developed. To further improve the model performance, changeable size moving window (CSMW) and competitive adaptive reweighted sampling (CARS) were utilized to select the effective wavelengths. After that, one dimensional-convolutional neural network (1D-CNN) model was constructed, which outperformed the other models without any preprocessing and optimization methods, and the values of RCal2, RMSEC, RPre2, and RMSEP are 0.953, 0.254 %, 0.900, and 0.371 %, respectively. In this study, conventional PLSR modelling methods and deep 1D-CNN method were compared under the influence of the two external factors, and the result showed that 1D-CNN can serve as a more convenient alternative for apple online SSC determination, which could significantly reduce the complexity of the Vis/NIR spectroscopy modeling process.
Broadening the bands for improving the accuracy of noninvasive blood component analysis

Abstract:Signal to noise ratio (SNR) of the spectrum is the overriding factor that limits the accuracy of noninvasive blood component measurement based on spectral analysis. In this paper, we applied the multi-dimensional spectral measurement strategy of the "M+N" theory to the quantitative blood component analysis based on the dynamic spectrum (DS) theory, in the form of increasing the wavelength range of the collected spectrum, and discussed the effects of spectral difference coefficient on measurement accuracy, to investigate the effects of wavelength range on the accuracy. We collected photoplethysmogram (PPG) signals across a broad range (313-1388 nm) using a spectrometer system consisting of multiple light sources and spectrophotometers covering different wavelength range assembled on-site. Using DS data obtained with the optimizing differential extraction method, we established calibration models for hemoglobin concentration. The new calibration model showed that compared with the model used the DS under the generally used range (550-1050 nm), the root mean square error of calibration set (RMSEC) decreased from 10.15 4 g/L to 5.509 g/L, and the correlation coefficient of calibration set (Rc) increased from 0.655 to 0.912. This study shows that the acquisition of signals from a wider wavelength range can improve the accuracy of noninvasive blood component measurement.

Analysis of serum total bilirubin content based on dual-position joint spectrum of "M plus N" theory and the logarithmic method

Abstract:At present, in the chemical quantitative analysis of complex solutions based on spectroscopy, the accuracy of the content analysis of complex solution is difficult to meet the requirements due to the overlapping spectral lines, low signal-to-noise ratio, and scattering characteristics of various components of complex solution. In this paper, the logarithmic method is used to preprocess the spectral data in the spectral preprocessing stage, and the two-position and multi-mode joint spectral strategy of "M plus N" theory is applied to the quantitative analysis of serum components. The serum samples are illuminated by dual-position ultraviolet LED light source, and the two spectra of the vertical position and the coaxial facing position of the light source and the optical fiber are collected respectively. Then the partial least square (PLS) method was used to establish models to analyze the concentration of total bilirubin in serum by the spectrum of vertical position, the spectrum of coaxial facing position, and the spectrum of the combination of the former two. Among the experimental results, the model established by the combination of the two spectra collected by two positions has a good result. The correlation coefficient of all samples predicted by this model is 0.971223, and the root mean square error is 1.96645 μmol/L. This study shows that the method of the logarithmic, collecting spectra and analyzing the composition content of complex solutions by using the multi-location and multi-mode strategy of "M + N" theory can effectively improve the prediction accuracy of the model and has practical significance for the chemical quantitative analysis of complex solutions based on spectroscopy.
High-precision non-invasive RBC and HGB detection system based on spectral analysis

Abstract:Non-invasive blood composition analysis based on dynamic spectrum (DS) theory has gained significant attention due to its non-invasive, simple, and fast performance. However, most of the multi-wavelength photoplethysmography (PPG) detection devices used to obtain DS are composed of halogen light sources and spectrometers and cannot detect effective PPG signals in the visible light short band (400-620 nm), which limits the detection accuracy of blood components with significant absorption spectral differences in that band. Therefore, this paper designs a multi-wavelength spectral acquisition system that can measure high signal-to-noise ratio (SNR > 65 dB) PGG signals at wavelengths of 405, 430, 450, 505, 520, and 570 nm and combines this system with a halogen lamp spectrometer acquisition system for non-invasive blood component detection. Furthermore, this paper collects the DS of 272 subjects with the combined system and establishes a predictive model for DS with the content of red blood cell (RBC) and hemoglobin (HGB) components. The results show that, compared with the halogen lamp spectrometer acquisition system, the correlation coefficient (Rp) of RBC and HGB prediction model established by the combined system has increased by 0.0619 and 0.0489, respectively, and the root mean square error (RMSE) has decreased by 0.08 1e12/L and 0.85 g/L, which confirm the feasibility of the designed multi-wavelength spectrum acquisition system to enhance the accuracy of blood component detection.
Improvement of NIR Models for Quality Parameters of Leech and Earthworm Medicines Using Outlier Multiple Diagnoses

Abstract:Leeches and earthworms are the main ingredients of Shuxuetong injection compositions, which are natural biomedicines. Near infrared (NIR) diffuse reflection spectroscopy has been used for quality assurance of Chinese medicines. In the present work, NIR spectroscopy was proposed as a rapid and nondestructive technique to assess the moisture content (MC), soluble solid content (SSC) and hypoxanthine content (HXC) of leeches and earthworms. This study goal was to improve NIR models for accurate quality control of leech and earthworm using outlier multiple diagnoses (OMD). OMD was composed of four outlier detection methods: spectrum outlier diagnostic (MD), leverage diagnostic (LD), principal component scores diagnostic (PCSD) and factor loading diagnostic (FLD). Conventional outlier diagnoses (MD, LD) and OMD were compared, and the best NIR models were those based on OMD. The correlation coefficients ([Formula: see text]) for leech were 0.9779, 0.9616 and 0.9406 for MC, SSC and HXC, respectively. The values of relative standard error of prediction (RSEP) for leech were 2.3%, 5.1% and 9.0% for MC, SSC and HXC, respectively. The values of [Formula: see text] for earthworm were 0.9478, 0.9991 and 0.9605 for MC, SSC and HXC, respectively. The values of RSEP for earthworm were 8.8%, 2.4% and 12% for MC, SSC and HXC, respectively. The performance of the NIR models was certainly improved by OMD.
Accurate Leukocyte Detection Based on Deformable-DETR and Multi-Level Feature Fusion for Aiding Diagnosis of Blood Diseases

Abstract:In standard hospital blood tests, the traditional process requires doctors to manually isolate leukocytes from microscopic images of patients' blood using microscopes. These isolated leukocytes are then categorized via automatic leukocyte classifiers to determine the proportion and volume of different types of leukocytes present in the blood samples, aiding disease diagnosis. This methodology is not only time-consuming and labor-intensive, but it also has a high propensity for errors due to factors such as image quality and environmental conditions, which could potentially lead to incorrect subsequent classifications and misdiagnosis. To address these issues, this paper proposes an innovative method of leukocyte detection: the Multi-level Feature Fusion and Deformable Self-attention DETR (MFDS-DETR). To tackle the issue of leukocyte scale disparity, we designed the High-level Screening-feature Fusion Pyramid (HS-FPN), enabling multi-level fusion. This model uses high-level features as weights to filter low-level feature information via a channel attention module and then merges the screened information with the high-level features, thus enhancing the model's feature expression capability. Further, we address the issue of leukocyte feature scarcity by incorporating a multi-scale deformable self-attention module in the encoder and using the self-attention and cross-deformable attention mechanisms in the decoder, which aids in the extraction of the global features of the leukocyte feature maps. The effectiveness, superiority, and generalizability of the proposed MFDS-DETR method are confirmed through comparisons with other cutting-edge leukocyte detection models using the private WBCDD, public LISC and BCCD datasets. Rapid monitoring approaches for concentration process of lanqin oral solution by near-infrared spectroscopy and chemometric models

Abstract:Qualitative and quantitative detection methods based on near-infrared spectroscopy (NIRs) have been proposed in the process analysis of traditional Chinese medicine in recent years. In this study, rapid monitoring methods were developed for quality control of concentration process of lanqin oral solution (LOS). Partial least squares regression (PLSR) method was adopted to construct quantitative models for epigoitrin, geniposide, baicalin, berberine hydrochloride and density. Simultaneously, the genetic algorithm joint extreme learning machine (GA-ELM) was first applied in qualitative analysis of NIRs to distinguish end point of concentration process. Results of PLSR models were satisfactory with the relative standard error of calibration valued at 3.80%, 3.75%, 3.79%, 11.5% and 1.22% for epigoitrin, geniposide, baicalin, berberine hydrochloride and density respectively, and the residual predictive deviation values were higher than 3. For qualitative analysis, the GA-ELM model obtained 100% prediction accuracy. The PLSR quantitative models and the end point discrimination model constructed by GA-ELM correspond with the requirements of practical applications. An Optimizing Dynamic Spectrum Differential Extraction Method for Noninvasive Blood Component Analysis

Abstract:Dynamic spectra (DS) can greatly reduce the influence of individual differences and the measurement environment by extracting the absorbance of pulsating blood at multiple wavelengths, and it is expected to achieve noninvasive detection of blood components. Extracting high-quality DS is the prerequisite for improving detection accuracy. This paper proposed an optimizing differential extraction method in view of the deficiency of existing extraction methods. In the proposed method, the sub-dynamic spectrum (sDS) is composed by sequentially extracting the absolute differences of two sample points corresponding to the height of the half peak on the two sides of the lowest point in each period of the logarithm photoplethysmography signal. The study was based on clinical trial data from 231 volunteers. Single-trial extraction method, original differential extraction method, and optimizing differential extraction method were used to extract DS from the volunteers’ experimental data. Partial least squares regression (PLSR) and radial basis function (RBF) neural network were used for modeling. According to the effect of PLSR modeling, by extracting DS using the proposed method, the correlation coefficient of prediction set ( R p ) has been improved by 17.33% and the root mean square error of prediction set has been reduced by 7.10% compared with the original differential extraction method. Compared with the single-trial extraction method, the correlation coefficient of calibration set ( R c ) has increased from 0.747659 to 0.8244, with an increase of 10.26%, while the correlation coefficient of prediction set ( R p ) decreased slightly by 3.22%, much lower than the increase of correction set. The result of the RBF neural network modeling also shows that the accuracy of the optimizing differential method is better than the other two methods both in calibration set and prediction set. In general, the optimizing differential extraction method improves the data utilization and credibility compared with the existing extraction methods, and the modeling effect is better than the other two methods.

Spectral Analysis Methods Based on Background Subtraction and Curvature Calculation Used in the Detection or Quantification of Hemolysis and Icterus in Blood-derived Clinical Samples

Abstract:Objective We aimed to find new methods to detect and quantify hemolysis and icterus which may cause assay biases. These methods need to determine each of these interferents in the presence of various other interferents. They also need to have less stringent requirements in development and implementation than those conventional analyzers currently must satisfy. Design and methods We developed two spectral analysis methods that obtain absorption signals of interest by background subtraction or by calculating the spectral curvatures near the peaks of interest. We optimized and tested the performance of these methods using a plasma sample set with permutations of the levels of hemolysis, icterus, and lipemia (using 510 samples in total). Results The processed signals correlated well with concentrations of hemoglobin and bilirubin, indicators of hemolysis and icterus, respectively. Through iterations of randomly splitting the samples for calibration and testing, the two new methods performed as well as those used on conventional analyzers. We demonstrated that the two methods can lessen the application requirements of 1) prior knowledge of the absorption spectra of individual interferents, 2) calibration over a wide concentration range for each interferent, and 3) the need for full-range spectrophotometers spanning most of the ultraviolet/visible spectrum. We also proposed a hardware setup to detect and quantify hemolysis or icterus with a camera and two optical filters. Conclusions This work indicates that new methods of spectral analysis can reduce practical constraints in the development of interference screening systems. These methods could also benefit other assays that rely on reading spectral signals.
A deep learning model for detection of leukocytes under various interference factors

Abstract:Abstract The accurate detection of leukocytes is the basis for the diagnosis of blood system diseases. However, diagnosing leukocyte disorders by doctors is time-consuming and requires extensive experience. Automated detection methods with high accuracy can improve detection efficiency and provide recommendations to inexperienced doctors. Current methods and instruments either fail to automate the identification process fully or have low performance and need suitable leukocyte data sets for further study. To improve the current status, we need to develop more intelligent strategies. This paper investigates fulfilling high-performance automatic detection for leukocytes using a deep learning-based method. We established a new dataset more suitable for leukocyte detection, containing 6273 images (8595 leukocytes) and considering nine common clinical interference factors. Based on the dataset, the performance evaluation of six mainstream detection models is carried out, and a more robust ensemble model is proposed. The mean of average precision (mAP) @IoU = 0.50:0.95 and mean of average recall (mAR)@IoU = 0.50:0.95 of the ensemble model on the test set are 0.853 and 0.922, respectively. The detection performance of poor-quality images is robust. For the first time, it is found that the ensemble model yields an accuracy of 98.84% for detecting incomplete leukocytes. In addition, we also compared the test results of different models and found multiple identical false detections of the models, then provided correct suggestions for the clinic.

Abstract:Background The differential of leukocytes functions as the first indicator in clinical examinations. However, microscopic examinations suffered from key limitations of low throughputs in classifying leukocytes while commercially available hematology analyzers failed to provide quantitative accuracies in leukocyte differentials. Methodology A home‐developed imaging and impedance flow cytometry of microfluidics was used to capture fluorescent images and impedance variations of single cells travelling through constrictional microchannels. Convolutional and recurrent neural networks were adopted for data processing and feature extractions, which were then fused by a support vector machine to realize the four‐part differential of leukocytes. Results The classification accuracies of the four‐part leukocyte differential were quantified as 95.4% based on fluorescent images plus the convolutional neural network, 90.3% based on impedance variations plus the recurrent neural network and 99.3% on the basis of fluorescent images, impedance variations and deep neural networks. Conclusion Based on single‐cell fluorescent imaging and impedance variations coupled with deep neural networks, the four‐part leukocyte differential can be realized with an almost 100% accuracy. Abstract:Because of the bulk, complexity, calibration requirements, and need for operator training, most current flow-based blood counting devices are not appropriate for field use. Standard imaging methods could be much more compact, inexpensive, and with minimal calibration requirements. However, due to the diffraction limit, imaging lacks the nanometer precision required to measure red blood cell volumes. To address this challenge, we utilize Mie scattering, which can measure nanometer-scale morphological information from cells, in a dark-field imaging geometry. The approach consists of a custom-built dark-field scattering microscope with symmetrically oblique illumination at a precisely defined angle to record wide-field images of diluted and sphered blood samples. Scattering intensities of each cell under three wavelengths are obtained by segmenting images via digital image processing. These scattering intensities are then used to determine size and hemoglobin information via Mie theory and machine learning. Validation on 90 clinical blood samples confirmed the ability to obtain mean corpuscular volume (MCV), mean corpuscular hemoglobin concentration (MCHC), and red cell distribution width (RDW) with high accuracy. Simulations based on historical data suggest that an instrument with the accuracy achieved in this study could be used for widespread anemia screening.

Here, we proposed a general strategy to unravel leukocytes heterogeneity and screen differentiated metabolites as biomarker candidates for leukocyte subtypes using the label-free mass cytometry (CyESI-MS) combined with a homemade data processing workflow. Taking leukemia cells as an example, metabolic fingerprints of single leukemia cells were obtained from 472 HL-60, 416 THP-1, 313 U937, 356 Jurkat, and 366 Ramos cells, with throughput up to 40 cells/min. Five leukemia subtypes were clearly distinguished by unsupervised learning tSNE analysis of the single-cell metabolic fingerprints. Cell discrimination in the mixed leukemia cell samples was also realized by supervised learning of the single-cell metabolic fingerprints with high recovery and good repetition (98.31 +/- 0.24%, -102.35 +/- 4.82%). Statistical analysis and metabolite assignment were carried out to screen characteristic metabolites for discrimination and 36 metabolites with significant differences were annotated. Then, differentiated metabolites for pairwise discrimination of five leukemia subtypes were further selected as biomarker candidates. Furthermore, discriminating cultured leukemia cells from human normal leukocytes, separated from fresh human peripheral blood, was performed based on single-cell metabolic fingerprints as well as the proposed biomarker candidates, unveiling the potential of this strategy in clinical research. This work makes efforts to realize high-throughput single-leukocyte metabolic analysis and metabolite-based discrimination of leukocytes. It is expected to be a powerful means for the clinical molecular diagnosis of hematological diseases.

Abstract:In zinc smelting solution, because the concentration of zinc is too high, the spectral signals of trace copper are masked by the spectral signals of zinc, and their spectral signals overlap, which makes it difficult to detect the concentration of trace copper. To solve this problem, a spectrophotometric method based on integrated and partition modeling is proposed. Firstly, the derivative spectra based on continuous wavelet transform are used to preprocess the spectral signal and highlight the spectral peak of copper. Then, the interval partition modeling is used to select the optimal characteristic interval of copper according to the root mean square error of prediction, and the wavelength points of the absorbance matrix are selected by correlation-coefficient threshold to improve the sensitivity and linearity of copper ions. Finally, the partial least squares integrated modeling based on the Adaboost algorithm is established by using the selected wavelength to realize the concentration detection of trace copper in the zinc liquid. Comparing the proposed method with existing regression methods, the results showed that this method can not only reduce the complexity of wavelength screening, but can also ensure the stability of detection performance. The predicted root mean square error of copper was 0.0307, the correlation coefficient was 0.9978, and the average relative error of prediction was 3.14%, which effectively realized the detection of trace copper under the background of high-concentration zinc liquid.

Abstract:Lymphocyte subsets are the predictors of disease diagnosis, treatment, and prognosis. Determination of lymphocyte subsets is usually carried out by flow cytometry. Despite recent advances in flow cytometry analysis, most flow cytometry data can be challenging with manual gating, which is labor-intensive, time-consuming, and error-prone. This study aimed to develop an automated method to identify lymphocyte subsets. We propose a knowledge-driven combined with data-driven method which can gate automatically to achieve subset identification. To improve accuracy and stability, we have implemented a Loop Adjustment Gating to optimize the gating result of the lymphocyte population. Furthermore, we have incorporated an anomaly detection mechanism to issue warnings for samples that might not have been successfully analyzed, ensuring the quality of the results. The evaluation showed a 99.2 % correlation between our method results and manual analysis with a dataset of 2,000 individual cases from lymphocyte subset assays. Our proposed method attained 97.7 % accuracy for all cases and 100 % for the high-confidence cases. With our automated method, 99.1 % of manual labor can be saved when reviewing only the low-confidence cases, while the average turnaround time required is only 29 s, reducing by 83.7 %. Our proposed method can achieve high accuracy in flow cytometry data from lymphocyte subset assays. Additionally, it can save manual labor and reduce the turnaround time, making it have the potential for application in the laboratory.

Abstract:The accuracy of near-infrared quantitative models for traditional Chinese medicine is restricted by the matrix, low-concentration markers, and overlapping spectral bands. Competitive adaptive reweighted sampling, a recently developed algorithm, selects an optimal combination of multicomponent spectral data. In this article, the performance of this method was evaluated through the analysis of traditional Chinese medicine. The near-infrared spectra of the pharmaceutics were obtained and the concentration of puerarin was determined. After optimization of spectral pretreatment methods, competitive adaptive reweighted sampling was performed in each dataset to select key wavenumbers. Sixty-eight, thirty, and eight variables were selected for the raw material, intermediate product, and final product, respectively. Partial least squares models were constructed based on the selected variables. Enhanced accuracy was obtained using the competitive adaptive reweighted sampling-coupled models. The results indicated that competitive adaptive reweighted sampling improved the accuracy and simplified calibration while offering a new approach for the rapid and nondestructive analysis of traditional Chinese medicine.
