Abstract:Diatom responses to ocean acidification have been documented with variable and controversial results. We grew the coastal diatom Thalassiosira weissflogii under 410 (LC, pH 8.13) vs 1000 μatm (HC, pH 7.83) pCO2 and at different levels of light (80, 140, 220 μmol photons m-2 s-1), and found that light level alters physiological responses to OA. CO2 concentrating mechanisms (CCMs) were down-regulated in the HC-grown cells across all the light levels, as reflected by lowered activity of the periplasmic carbonic anhydrase and decreased photosynthetic affinity for CO2 or dissolved inorganic carbon. The specific growth rate was, however, enhanced significantly by 9.2% only at the limiting low light level. These results indicate that rather than CO2 "fertilization", the energy saved from down-regulation of CCMs promoted the growth rate of the diatom when light availability is low, in parallel with enhanced respiration under OA to cope with the acidic stress by providing extra energy.
Decreased photosynthesis and growth with reduced respiration in the model diatom Phaeodactylum tricornutum grown under elevated CO2 over 1800 generations

Abstract:Studies on the long-term responses of marine phytoplankton to ongoing ocean acidification (OA) are appearing rapidly in the literature. However, only a few of these have investigated diatoms, which is disproportionate to their contribution to global primary production. Here we show that a population of the model diatom Phaeodactylum tricornutum, after growing under elevated CO2 (1000 μatm, HCL, pHT : 7.70) for 1860 generations, showed significant differences in photosynthesis and growth from a population maintained in ambient CO2 and then transferred to elevated CO2 for 20 generations (HC). The HCL population had lower mitochondrial respiration, than did the control population maintained in ambient CO2 (400 μatm, LCL, pHT : 8.02) for 1860 generations. Although the cells had higher respiratory carbon loss within 20 generations under the elevated CO2 , being consistent to previous findings, they downregulated their respiration to sustain their growth in longer duration under the OA condition. Responses of phytoplankton to OA may depend on the timescale for which they are exposed due to fluctuations in physiological traits over time. This study provides the first evidence that populations of the model species, P. tricornutum, differ phenotypically from each other after having been grown for differing spans of time under OA conditions, suggesting that long-term changes should be measured to understand responses of primary producers to OA, especially in waters with diatom-dominated phytoplankton assemblages.

Abstract:Ocean acidification and warming are two major environmental stressors; however, the generality of how warming will alter growth responses of phytoplankton to ocean acidification is less known. Here, enhancement of growth by high CO2 (HC) in Phaeodactylum tricornutum and Thalassiosira weissflogii was most prominent at optimum temperature. The extent to which growth rates in HC cultures were raised compared to low CO2 (LC) cultures tended to decrease with increasing or decreasing temperature, compared to the optimum. Further mechanistic studies in P. tricornutum revealed that cellular carbon and nitrogen content, superoxide dismutase activity, and respiration were generally higher in HC than those in LC at high and low temperatures, whereas PSII photochemical parameters were generally lower in HC than in LC at high and low temperatures. These results indicate that HC-grown cells needed to invest more energy and materials to maintain intracellular homeostasis and repair damage induced by the unsuitable temperatures.

Photosynthetic responses of the marine diatom Thalassiosira pseudonana to CO2-induced seawater acidification

Abstract:Ocean acidification due to atmospheric CO2 rise is expected to influence marine phytoplankton. Diatoms are responsible for about 40% of the total primary production in the ocean. In order to investigate the physiological response of marine diatom Thalassiosira pseudonana to ocean acidification, we grew the cells under ambient CO2 level (380 µatm) versus the elevated CO2 level (800 µatm) at a light level of 180 µmol m−2 s−1 for 30 generations. The intracellular carbonic anhydrase activity was suppressed and the photosynthetic affinity for CO2 was lowered in the high CO2-grown cells, reflecting a downregulation of the CO2 concentrating mechanism (CCM). PSI activity was enhanced to support an increase in ATP synthesis by cyclic electron transfer as required for transport of inorganic carbon and regulation of intracellular pH. The energetic benefit from the downregulation of CCM to growth as reported in other diatom species was not observed here in T. pseudonana.

Abstract:Background Light is a key regulatory factor for photosynthesis and metabolism of microalgae. The diatom Phaeodactylum tricornutum is capable of exhibiting metabolic flexibility in response to light fluctuations. However, the metabolic switching and underlying molecular mechanisms upon illumination transitions remain poorly understood for this industrially relevant marine alga. To address these, the physiochemical and molecular responses of P. tricornutum upon high light (HL) and recovery (HLR) were probed. Results Upon HL, P. tricornutum exhibited quick responses, including decreases in cell division, major light harvesting pigments (e.g., chlorophyll a , β -carotene, and fucoxanthin), chloroplastidic membrane lipids (e.g., monogalactosyldiacylglycerol, digalactosyldiacylglycerol, and sulfoquinovosyldiacylglycerol), and long-chain polyunsaturated fatty acids (e.g., C20:5), as well as increases in carbohydrates and neutral lipids particularly triacylglycerol. During HLR stage when the stress was removed, these physiochemical phenotypes were generally recovered, indicative of a rapid and reversible changes of P. tricornutum to cope with illumination transitions for survival and growth. Through the integrated analysis with time-resolved transcriptomics, we revealed the transcriptional control of photosynthesis and carbon metabolism in P. tricornutum responding to HL, which could be reversed more or less during the HLR stage. Furthermore, we highlighted key enzymes involved in carotenoid biosynthesis and lipid metabolism of P. tricornutum and identified monooxygenases putatively responsible for catalyzing the ketolation step towards fucoxanthin synthesis from neoxanthin. Conclusions The detailed profiling of physiochemical and transcriptional responses of P. tricornutum to HL-HLR treatments advances our understanding on the adaption of the alga to illumination transitions and provides new insights into engineering of the alga for improved production of value-added carotenoids and lipids.
Two Southern Ocean diatoms are more sensitive to ocean acidification and changes in irradiance than the prymnesiophyte Phaeocystis antarctica

Abstract:To better understand the impact of ocean acidification (OA) and changes in light availability on Southern Ocean phytoplankton physiology, we investigated the effects of pCO2 (380 and 800 µatm) in combination with low and high irradiance (20 or 50 and 200 µmol photons m-2 s-1 ) on growth, particulate organic carbon (POC) fixation and photophysiology in the three ecologically relevant species Chaetoceros debilis, Fragilariopsis kerguelensis and Phaeocystis antarctica. Irrespective of the light scenario, neither growth nor POC per cell was stimulated by OA in any of the tested species and the two diatoms even displayed negative responses in growth (e.g. C. debilis) or POC content (e.g. F. kerguelensis) under OA in conjunction with high light. For both diatoms, also maximum quantum yields of photosystem II (Fv /Fm ) were decreased under these conditions, indicating lowered photochemical efficiencies. To counteract the negative effects by OA and high light, the two diatoms showed diverging photoacclimation strategies. While cellular chlorophyll a (Chl a) and fucoxanthin contents were enhanced in C. debilis to potentially maximize light absorption, F. kerguelensis exhibited reduced Chl a per cell, increased disconnection of antennae from photosystem II reaction centers and strongly lowered absolute electron transport rates (ETR). The decline in ETRs in F. kerguelensis might be explained in terms of different species-specific strategies for tuning the available flux of adenosine triphosphate and nicotinamide adenine dinucleotide phosphate. Overall, our results revealed that P. antarctica was more tolerant to OA and changes in irradiance than the two diatoms, which may have important implications for biogeochemical cycling.
Physiological and biochemical responses of Thalassiosira weissflogii (diatom) to seawater acidification and alkalization

Abstract:Increasing atmospheric pCO2 leads to seawater acidification, which has attracted considerable attention due to its potential impact on the marine biological carbon pump and function of marine ecosystems. Alternatively, phytoplankton cells living in coastal waters might experience increased pH/decreased pCO2 (seawater alkalization) caused by metabolic activities of other photoautotrophs, or after microalgal blooms. Here we grew Thalassiosira weissflogii (diatom) at seven pCO2 levels, including habitat-related lowered levels (25, 50, 100, and 200 µatm) as well as present-day (400 µatm) and elevated (800 and 1600 µatm) levels. Effects of seawater acidification and alkalization on growth, photosynthesis, dark respiration, cell geometry, and biogenic silica content of T. weissflogii were investigated. Elevated pCO2 and associated seawater acidification had no detectable effects. However, the lowered pCO2 levels (25 ∼ 100 µatm), which might be experienced by coastal diatoms in post-bloom scenarios, significantly limited growth and photosynthesis of this species. In addition, seawater alkalization resulted in more silicified cells with higher dark respiration rates. Thus, a negative correlation of biogenic silica content and growth rate was evident over the pCO2 range tested here. Taken together, seawater alkalization, rather than acidification, could have stronger effects on the ballasting efficiency and carbon export of T. weissflogii.

Interactive Effects of Elevated CO2 Concentration and Light on the Picophytoplankton Synechococcus

Abstract:Synechococcus is a major contributor to the primary production in tropic and subtropical oceans worldwide. Responses of this picophytoplankton to changing light and CO 2 levels is of general concern to understand its ecophysiology in the context of ocean global changes. We grew Synechococcus sp. (WH7803), originally isolated from subtropic North Atlantic Ocean, under different PAR levels for about 15 generations and examined its growth, photochemical performance and the response of these parameters to elevated CO 2 (1,000 μatm). The specific growth rate increased from 6 μmol m –2 s –1 to reach a maximum (0.547 ± 0.026) at 25 μmol m –2 s –1 , and then became inhibited at PAR levels over 50 μmol m –2 s –1 , with light use efficiency (α) and photoinhibition coefficient (β) being 0.093 and 0.002, respectively. When the cells were grown at ambient and elevated CO 2 concentration (400 vs. 1,000 μatm), the high-CO 2 grown cells showed significantly enhanced rates of electron transport and quantum yield as well as significant increase in specific growth rate at the limiting and inhibiting PAR levels. While the electron transport rate significantly increased at the elevated CO 2 concentration under all tested light levels, the specific growth did not exhibit significant changes under the optimal growth light condition. Our results indicate that Synechococcus WH7803 grew faster under the ocean acidification (OA) treatment induced by CO 2 enrichment only under limiting and inhibiting light levels, indicating the interactive effects and implying that the picophytoplankton respond differentially at different depths while exposing changing light conditions.

Abstract:The yeast two-hybrid system is a powerful technique that detects interactions between two proteins and has been useful in identifying new binding partners. However, the system fails to detect protein-protein interactions that require the presence of additional components of a multisubunit complex. Here we demonstrate that the vector YIpDCE1 can be used to express elongins B and C in yeast, and that these proteins form a stable complex that interacts with the von Hippel-Lindau tumor-suppressor gene product (pVHL). Only when pVHL and elongins B and C (VBC) are present does an interaction with the cullin family member, hCUL-2, occur, forming the heterotetrameric pVHL/elongin BC/hCUL-2 complex. This system was then used to map the binding region of hCUL-2 for the VBC complex. The first amino-terminal 108 aa of hCUL-2 are necessary for interaction with the VBC complex. The elongin BC dimer acts as a bridge between pVHL and hCUL-2 because pVHL and hCUL-2 can form distinct complexes with elongins B and C. These results reveal a striking structural resemblance of pVHL/elongin BC/hCUL-2 complex with the E3-like ubiquitin ligase complex SKP1/Cullin/F-box protein with respect to protein composition and sites of interactions. Thus, it seems possible that pVHL/elongin BC/hCUL-2 complex will possess ubiquitin ligase activity targeting specific proteins for degradation by the proteasome.
Effect of mono- and dichromatic light quality on growth rates and photosynthetic performance of Synechococcus sp. PCC 7002.

Abstract:Synechococcus sp. PCC 7002 was grown to steady state in optically thin turbidostat cultures under conditions for which light quantity and quality was systematically varied by modulating the output of narrow-band LEDs. Cells were provided photons absorbed primarily by chlorophyll (680 nm) or phycocyanin (630 nm) as the organism was subjected to four distinct mono- and dichromatic regimes. During cultivation with dichromatic light, growth rates were generally proportional to the total incident irradiance at values <275 μmol photons m(-2) · s(-1) and were not affected by the ratio of 630:680 nm wavelengths. Notably, under monochromatic light conditions, cultures exhibited similar growth rates only when they were irradiated with 630 nm light; cultures irradiated with only 680 nm light grew at rates that were 60-70% of those under other light quality regimes at equivalent irradiances. The functionality of photosystem II and associated processes such as maximum rate of photosynthetic electron transport, rate of cyclic electron flow, and rate of dark respiration generally increased as a function of growth rate. Nonetheless, some of the photophysiological parameters measured here displayed distinct patterns with respect to growth rate of cultures adapted to a single wavelength including phycobiliprotein content, which increased under severely light-limited growth conditions. Additionally, the ratio of photosystem II to photosystem I increased ~40% over the range of growth rates, although cells grown with 680 nm light only had the highest ratios. These results suggest the presence of effective mechanisms which allow acclimation of Synechococcus sp. PCC 7002 acclimation to different irradiance conditions.
Mapping Radar Glacier Zones and Dry Snow Line in the Antarctic Peninsula Using Sentinel-1 Images

Abstract:Surface snowmelt causes changes in mass and energy balance, and endangers the stabilities of the ice shelves in the Antarctic Peninsula (AP). The dynamic changes of the snow and ice conditions in the AP were observed by Sentinel-1 images with a spatial resolution of 40 m in this study. Snowmelt detected by the special sensor microwave/imager (SSM/I) is used to study the relationship between summer snowmelt and winter synthetic aperture radar (SAR) backscatter. Radar glacier zones (RGZs) classifications were conducted based on their differences in liquid snow content, snow grain size, and the relative elevations. We developed a practical method based on the simulations of a microwave scattering model to classify RGZs by using Sentinel-1 images in the AP. The summer snowmelt detected by SSM/I and Sentinel-1 data are compared between 2014 and 2015. The SSM/I-derived melting days is used to validate the winter dry snow line (DSL). RGZs derived from Sentinel-1 images suggest that snowmelt expanded from inland of the Larsen C Ice Shelf to the coastal area, whereas an opposite direction was found in the George VI Ice Shelf. The long melting season in the grounding zone of the Larsen C Ice Shelf may result from the adiabatically-dried fohn winds on the east side of the AP. As the uppermost limit of summer snowmelt, DSL was mapped based on the winter Sentinel-1 mosaic of the AP. Compared with the SSM/I-derived melting days, the winter DSL mainly distributed in the areas melted for one to three days in summer. DSL elevations on the Palmer Land increased from south to north.
The Response of Three Southern Ocean Phytoplankton Species to Ocean Acidification and Light Availability: A Transcriptomic Study

Abstract:Ocean acidification (OA) and high light was found to negatively affect the Antarctic key species Phaeocystis antarctica, Fragilariopsis kerguelensis and Chaetoceros debilis. To unravel the underlying physiological response at the transcriptomic level, these species were grown under ambient and elevated pCO2 combined with low or high light. RNA sequencing revealed that the haptophyte was much more tolerant towards OA than the two diatoms as only these showed distinct OA-dependent gene regulation patterns. Under ambient pCO2, high light resulted in decreased glycolysis in P. antarctica. Contrastingly, upregulation of genes related to cell division and transcription as well as reduced expression of both cata- and anabolic carbon related pathways were seen in C. debilis. OA in combination with low light led to reduced respiration, but also surprisingly to higher expression of genes involved in light protection, transcription and translation in C. debilis. Though not affecting P. antarctica, OA combined with high light caused also photosensitivity in both diatoms. As additional response reallocation of carbon to lipids was found in C. debilis under these conditions. Overall, we conclude that P. antarctica is better adapted than the two diatoms to OA and high light.
Effects of photoperiod on the growth and physiological responses in Ulva prolifera under constant and diurnal temperature difference conditions

Abstract:Photoperiod and temperature are two main factors in the growth of macroalgae, and changes in photoperiod and diurnal temperature difference exist in natural condition. In order to study the effects of photoperiod and diurnal temperature difference on the growth of green algae Ulva prolifera, we cultured this species under three light/dark cycles (light: dark = 10:14, 12:12 and 16:08) with constant (22 °C for light and dark period, noted as 22-22 °C) and diurnal temperature difference (22 °C and 16 °C for light and dark period, respectively, noted as 22-16 °C) conditions. The results showed that: 1) Compared with 10:14 light/dark cycle, the growth of U. prolifera under 12:12 light/dark cycle was significantly enhanced by 39% and 16% for 22-22 °C and 22-16 °C treatments, respectively, while the increase proportion decreased when the daylength increase from 12 h to 16 h. 2) The enhancement in growth induced by diurnal temperature difference was observed under 10:14 light/dark cycle, but not for 12:12 and 16:08 light/dark cycle treatments. 3) The Chl a content and photosynthetic rate increased under short light period and 22-22 °C conditions, while under 22-16 °C conditions, higher photosynthetic rate was observed under 12:12 light/dark cycle and no significant difference in Chl a content was observed. 4) Under 22-22 °C conditions, compared with 10:14 (L:D) treatment, the expression levels of proteins in light-harvesting complexes, PSII and carbon fixation were down regulated, while the photorespiration and pentose phosphate pathway (PPP) were up regulated by 16:08 light dark cycle. Then we speculate that the higher photosynthetic rate may be one compensation mechanism in short photoperiod, and under long light period condition the up regulations of photorespiration and PPP can be in charge of the decrease in enhancement growth induced by longer daylength.

Abstract:Abstract Although the adverse impacts of ocean acidification (OA) on marine calcifiers have been investigated substantially, the anti-stress abilities regulated by increased light availability are unclear. Herein, the interactive effects of three light levels combined with two p CO 2 concentrations on the physiological acclimation of the calcifying macroalga Halimeda opuntia were investigated using a p CO 2 –light coupling experiment. The results indicate that OA exhibits an adverse role in influencing algal growth, calcification, photosynthesis and other physiological performances in H. opuntia. The relative growth rate in elevated p CO 2 significantly declined by 13.14%–41.29%, while net calcification rates decreased by nearly three-fold under OA. Notably, increased light availability could enhance stress resistance by the accumulation of soluble organic molecules, especially soluble carbohydrate, soluble protein and free amino acids, and in combination with metabolic enzyme-driven activities alleviated OA stress. Carotenoid content in low light conditions accumulated remarkably and rapid light curves for relative electron transport rate was significantly enhanced by increasing light intensities, indicating that this new organization of the photosynthetic machinery in H. opuntia accommodated light variations and elevated p CO 2 conditions. Taken together, the results describe stress resistance by the enhancement of metabolic performance in marine calcifiers to mitigate OA stress. Abstract:Abstract Unlike in terrestrial and freshwater ecosystems, light fields in oceans fluctuate due to both horizontal current and vertical mixing. Diatoms thrive and dominate the phytoplankton community in these fluctuating light fields. However, the molecular mechanisms that regulate diatom acclimation and adaptation to light fluctuations are poorly understood. Here, we performed transcriptome sequencing, metabolome profiling, and 13C-tracer labeling on the model diatom Phaeodactylum tricornutum. The diatom acclimated to constant light conditions was transferred to six different light conditions, including constant light (CL5d), short-term (1 h) high light (sHL1h), and short-term (1 h) and long-term (5 days) mild or severe light fluctuation conditions (mFL1h, sFL1h, mFL5d, and sFL5d) that mimicked land and ocean light levels. We identified 2,673 transcripts (25% of the total expressed genes) expressed differentially under different fluctuating light regimes. We also identified 497 transcription factors, 228 not reported previously, which exhibited higher expression under light fluctuations, including 7 with a light-sensitive PAS domain (Per-period circadian protein, Arnt-aryl hydrocarbon receptor nuclear translocator protein, Sim-single-minded protein) and 10 predicted to regulate genes related to light-harvesting complex proteins. Our data showed that prolonged preconditioning in severe light fluctuation enhanced photosynthesis in P. tricornutum under this condition, as evidenced by increased oxygen evolution accompanied by the upregulation of Rubisco and light-harvesting proteins. Furthermore, severe light fluctuation diverted the metabolic flux of assimilated carbon preferentially toward fatty acid storage over sugar and protein. Our results suggest that P. tricornutum use a series of complex and different responsive schemes in photosynthesis and carbon metabolism to optimize their growth under mild and severe light fluctuations. These insights underscore the importance of using more intense conditions when investigating the resilience of phytoplankton to light fluctuations.

Abstract:The effects of ocean acidification and solar radiation on marine organisms have received increasing attention. Coccolithophores are a major producer of dimethylsulfoniopropionate (DMSP), which is a precursor of dimethylsulfide (DMS), a volatile biogenic active gas related to climate. Here, we investigated the individual and combined effects of elevated CO2 and ultraviolet radiation (UVR) on growth, DMS, and DMSP production of Emiliania huxleyi. Elevated CO2 (1000 μatm, HC) decreased the cell concentration, DMS, and particulate DMSP (DMSPp) concentrations by 17%, 20%, and 13%, respectively, compared with ambient CO2 (400 μatm, LC) in the semi-continuous culture. The addition of UVA to photosynthetically active radiation (PAR) increased cell concentration of E. huxleyi by 16% on day 4, which may be due to the photorepair effects induced by UVA, and the effect was time-dependent. PAR + UVA and PAR + UVA + UVB exposure decreased cellular DMS by 25%-56%, and increased cellular DMSPp by 60%-130% compared with PAR on days 3-4. Cellular DMSPp followed the order: PAR + UVA > PAR + UVA + UVB > PAR, and HC had no significant effects on cellular DMSPp compared with LC in the combined experiment. These results aid our understanding of the effects of ocean acidification and UV radiation on the production of methyl sulfur compounds in the ocean.

Abstract:Ocean acidification (OA) caused by rising atmospheric CO 2 concentration and solar ultraviolet radiation (UVR) resulting from ozone depletion may affect marine organisms, but little is known regarding how unicellular Rhodosorus sp. SCSIO-45730, an excellent species resource containing various biological-active compounds, responds to OA and UVR. Therefore, we conducted a factorial coupling experiment to unravel the combined effects of OA and UVR on the growth, photosynthetic performances, biochemical compositions and enzyme activities of Rhodosorus sp. SCSIO-45730, which were exposed to two levels of CO 2 (LC, 400 μatm, current CO 2 level; HC, 1000 μatm, future CO 2 level) and three levels of UVR (photosynthetically active radiation (PAR), PAR plus UVA, PAR plus UVB) treatments in all combinations, respectively. Compared to LC treatment, HC stimulated the relative growth rate (RGR) due to higher optimum and effective quantum yields, photosynthetic efficiency, maximum electron transport rates and photosynthetic pigments contents regardless of UVR. However, the presence of UVA had no significant effect but UVB markedly reduced the RGR. Additionally, higher carbohydrate content and lower protein and lipid contents were observed when Rhodosorus sp. SCSIO-45730 was cultured under HC due to the ample HCO3− applications and active stimulation of metabolic enzymes of carbonic anhydrase and nitrate reductase, thus resulting in higher TC/TN. OA also triggered the production of reactive oxygen species (ROS), and the increase of ROS coincided approximately with superoxide dismutase and catalase activities, as well as phenols contents. However, UVR induced photochemical inhibition and damaged macromolecules, making algal cells need more energy for self-protection. Generally, these results revealed that OA counteracted UVR-related inhibition on Rhodosorus sp. SCSIO-45730, adding our understanding of the red algae responding to future global climate changes.

Abstract:Abstract Many marine organisms are exposed to decreasing O 2 levels due to warming-induced expansion of hypoxic zones and ocean deoxygenation (DeO 2 ). Nevertheless, effects of DeO 2 on phytoplankton have been neglected due to technical bottlenecks on examining O 2 effects on O 2 -producing organisms. Here we show that lowered O 2 levels increased primary productivity of a coastal phytoplankton assemblage, and enhanced photosynthesis and growth in the coastal diatom Thalassiosira weissflogii . Mechanistically, reduced O 2 suppressed mitochondrial respiration and photorespiration of T. weissflogii , but increased the efficiency of their CO 2 concentrating mechanisms (CCMs), effective quantum yield and improved light use efficiency, which was apparent under both ambient and elevated CO 2 concentrations leading to ocean acidification (OA). While the elevated CO 2 treatment partially counteracted the effect of low O 2 in terms of CCMs activity, reduced levels of O 2 still strongly enhanced phytoplankton primary productivity. This implies that decreased availability of O 2 with progressive DeO 2 could boost re-oxygenation by diatom-dominated phytoplankton communities, especially in hypoxic areas, with potentially profound consequences for marine ecosystem services in coastal and pelagic oceans.

Abstract:Coastal waters undergo dynamic changes in seawater carbonate chemistry due to natural and anthropogenic factors. Despite this, our current understanding of how coastal phytoplankton respond to fluctuating pH is limited. In the present study, we investigated the physiological responses of two coastal diatoms Thalassiosira pseudonana and Thalassiosira weissflogii to seawater acidification and diurnally fluctuating pH under natural solar irradiance. Seawater acidification did not significantly impact the growth, maximum and effective quantum yield of PSII, and photosynthetic rates of the two species. However, it did increase the maximum relative electron transport rate of T. weissflogii by 11%. Overall, fluctuating pH had neutral or positive effects on both species. It enhanced the light-saturated photosynthetic rate of T. weissflogii by 20% compared to cells grown under seawater acidification condition. Results from the short-term pH exposure experiment revealed that the photosynthetic rates of both species remained unaffected by acute pH changes, indicating their tolerance to varying pH. Nevertheless, it is crucial to consider dynamic pH when predicting changes in primary production in coastal waters, given the interplay of various environmental drivers.

Abstract:In order to understand how darkness/irradiance and low nighttime temperature might alter physiology of Ulva prolifera under lower salinity conditions, we analyzed the growth rates, water content, superoxide dismutase (SOD) activity, total soluble proteins (SPs) and carbohydrates content at the end of dark and light period under three temperature levels (25-25 °C treatment: 25 °C for day and night; 15-15 °C treatment: 15 °C for day and night; 25-15 °C treatment: 25 °C for day with 15 °C for night) and two salinity conditions (15, 25), meanwhile, the pigment content (chlorophyll a and b), chlorophyll fluorescence and photosynthetic oxygen evolution also were determined during light phase. We found that the U. prolifera showed higher growth rate and SOD activity during dark phase at 25 °C, but this dark-induced increase could not be observed at 15 °C. The reasons for this increase varied, however, maybe not included water content and SPs for no significant difference in water content observed under all the treatments, as well as lower SPs content for dark period aside that at 15 °C and salinity 15. Compared to other two temperature treatments, the thalli grown at 25-15 °C showed higher growth rate and the photosynthetic oxygen evolution rate in light phase under salinity 15 conditions, although the maximum relative electron transport rate (rETRmax) showed higher value under 25 °C treatment. These results indicate that the darkness and the lower nighttime temperature maybe responsible reason for the rapid growth of these green tide algae.
