Abstract:Habitat simplification and intensive use of pesticides are main drivers of global arthropod declines and are, thus, decreasing natural pest control. Organic farming, complex landscapes, and local vineyard management practices such as implementation of flower-rich cover-crop mixtures may be a promising approach to enhance predator abundance and, therefore, natural pest control. We examined the effect of organic versus integrated management, cover-crop diversity in the vineyard inter-rows, and landscape composition on the natural pest control of Lobesia botrana eggs and pupae. Predation of L. botrana pupae was reduced by organic farming and species-poor cover-crops by about 10%. Predation rates of L. botrana eggs did not differ significantly in any of the studied management options. Dominant predators were earwigs (Forficulidae), bush crickets (Tettigoniidae), and ants (Formicidae). Negative effects of organic viticulture are most likely related to the negative nontarget effects on arthropods related to the frequent sulfur and copper applications in combination with the avoidance of strongly damaging insecticides by integrated winegrowers. While a 10% difference in predation rates on a single pest stage is unlikely to have strong practical implications, our results show that the assumed effectiveness of environmentally friendly agriculture needs to be evaluated for specific crops and regions.

Effects of fungicide application on physiological and molecular responses of grapevine (Vitis vinifera L.): a comparison between copper and sulfur fungicides applied alone and in combination with novel fungicides

Abstract:Background: Chemical products against fungi and oomycetes pose serious environmental issues. In the last decade, the use of less impacting active ingredients was encouraged to reduce chemical inputs in viticulture. In this study, the effect of different antifungal compounds on grapevine agronomic, physiological, and molecular responses in the vineyard was evaluated in addition to protection against powdery and downy mildews. Results: In 2 years and in two Vitis vinifera cultivars (Nebbiolo and Arneis), a conventional crop protection approach, based on traditional fungicides (sulfur and copper), was compared to combined strategies. A well-known resistance inducer (potassium phosphonate), Bacillus pumilus strain QST 2808 and calcium oxide, both active ingredients whose biological interaction with grapevine is poorly characterized, were applied in the combined strategies in association with chemical fungicides. Despite a genotype effect occurred, all treatments optimally controlled powdery and downy mildews, with minimal variations in physiological and molecular responses. Gas exchange, chlorophyll content and photosystem II efficiency increased in treated plants at the end of season, along with a slight improvement in the agronomic performances, and an activation of molecular defense processes linked to stilbene and jasmonate pathways. Conclusion: The disease control strategies based on potassium phosphonate, Bacillus pumilus strain QST 2808 or calcium oxide combined with traditional chemical compounds did not cause severe limitations in plant ecophysiology, grape quality, and productive yields. The combination of potassium phosphonate and calcium oxide with traditional fungicides can represent a valuable strategy for reducing copper and sulfur inputs in the vineyards, including those organically managed. © 2023 The Authors. Organic management promotes natural pest control through altered plant resistance to insects

Abstract:Reduced insect pest populations found on long-term organic farms have mostly been attributed to increased biodiversity and abundance of beneficial predators, as well as to changes in plant nutrient content. However, the role of plant resistance has largely been ignored. Here, we determine whether host plant resistance mediates decreased pest populations in organic systems and identify potential underpinning mechanisms. We demonstrate that fewer numbers of leafhoppers (Circulifer tenellus) settle on tomatoes (Solanum lycopersicum) grown using organic management as compared to conventional. We present multiple lines of evidence, including rhizosphere soil microbiome sequencing, chemical analysis and transgenic approaches, to demonstrate that changes in leafhopper settling between organically and conventionally grown tomatoes are dependent on salicylic acid accumulation in plants and mediated by rhizosphere microbial communities. These results suggest that organically managed soils and microbial communities may play an unappreciated role in reducing plant attractiveness to pests by increasing plant resistance.

Abstract:Biological control of crop pests is a major ecosystem service affected by several variables acting at multiple spatial scales. Among these variables, heterogeneity at the habitat and landscape scales are known key drivers of trophic interactions and pest density in agroecosystems. However, studies that try to disentangle their relative effects in perennial cropping systems are scarce and nothing is known about their impacts on insect pest density and pesticide applications.We examined the effect of heterogeneity at these two scales on grape moths, one of the most damaging insect pests in European vineyards, and their biological control in 20 vineyards during three consecutive years. We used local vegetation management and the proportion of semi-natural habitats in the surrounding landscape as proxies of heterogeneity at the habitat and landscape scales. Grape moth density was measured over time, as well as biological control services provided by different groups: birds, invertebrate predators, parasitoids and entomopathogenic fungi.Over the 3 years, grape moth density was mainly determined by local heterogeneity, with significantly fewer larvae of the first generation established in vineyards with full compared to partial grass cover.Despite these effects, biological control of grape moths was not primarily affected by local vegetation management but by landscape heterogeneity, and the direction of this effect varied over time. Notably, predation by birds increased with landscape heterogeneity in spring, depending on local vegetation management, while attacks by pathogenic fungi decreased with landscape heterogeneity during winter.Synthesis and applications. Our results suggest that bottom-up processes related to habitat heterogeneity drive grape moth occurrence much more than top-down processes. These results have important implications for the ecological intensification of vineyard landscapes. We found that maintaining full grass cover within vineyards reduced grape moth density to a level below common intervention thresholds. Landscape heterogeneity in the close vicinity of vineyards contributed to improved biological pest control by birds, but depended on local vegetation management. Moreover, opposing effects of landscape management on biological pest control services over time revealed that strategies based only on manipulating landscape heterogeneity might not be the optimal option to limit grape moth density in vineyards.

Abstract:In agricultural landscapes, the clearing of semi‐natural habitats (SNH) and the intensive use of pesticides contribute to declines of biodiversity, including crop pollinators. Using yellow pan traps to sample wild bees, we investigated how SNH in the surrounding landscape, organic and conventional management, and the reduction of fungicides affect wild bee diversity in 32 vineyards in southwest Germany. Our study underlines that the local provision of diverse vegetation in vineyards and the landscape‐scale provision of suitable SNH are key factors for wild bee conservation in viticulture. In agricultural landscapes, the removal of semi‐natural habitats (SNH) and the intensive use of pesticides contribute to declines of biodiversity, including crop pollinators such as bees (Hymenoptera: Apoidea). However, effects of pesticide use and landscape characteristics on bees have rarely been studied together. In this study, we investigated how SNH in the surrounding landscape, organic and conventional management, and the reduction of fungicides affect wild bee diversity in 32 vineyards in southwest Germany. We used yellow pan traps to sample bees in a crossed design of management (organic vs. conventional) and fungicide use (reduced in fungus‐resistant grape varieties vs. regular) along a gradient with increasing proportions of SNH in the surrounding landscape. Higher proportions of SNH influenced species composition of bees and increased the abundance and richness of above‐ground‐nesting species. Organic vineyards had a 49% higher abundance of bees compared to conventional vineyards. The reduction of fungicides did not affect bee diversity nor abundance. The absence of a response to fungicide intensity suggests that the benefit of organically managed vineyards to wild bees was through differences in their vegetation management, which is in line with the positive response of bees to SNH in the surrounding landscape. Synthesis and applications: Our study underlines that the local provision of diverse vegetation in vineyards and the landscape‐scale provision of suitable SNH are key factors for wild bee conservation in viticulture.

Abstract:In viticulture, an intensive pesticide schedule is often required to control fungal and oomycete diseases and meet production standards. Accordingly, a reduction of synthetic fungicides and the application of environmentally-friendly crop protection alternatives are becoming increasingly important for a more sustainable viticulture. Even though the adoption of resistant/tolerant varieties is still limited in some traditional grape growing regions, the application of biological and/or physical tools, can in part substitute synthetic fungicides applications, and the use of decision support systems can help in reducing chemical treatments. This chapter presents the state of the art in grapevine protection to reduce pesticide use in viticulture and explores various alternative sustainable management strategies aimed to effectively control grapevine diseases triggered by fungi and oomycetes in nurseries, young and mature vineyards.

Abstract:Arbuscular mycorrhizal fungi (AMF) form symbioses with the majority of plant species and can provide multiple benefits to the host plant. In agro-ecosystems, the abundance and community structure of AMF are affected by agricultural management practices. This review describes and discusses current knowledge on the effects of inorganic and organic chemical pesticides on AMF in the conflicting area between agricultural use and environmental concerns. Variable effects have been reported following chemical pesticide use, ranging from neutral to positive and negative. Moreover, a species-specific reaction has been documented. The reported effects of pesticides on arbuscular mycorrhizal symbiosis are very diverse, and even when the same substance is investigated, the results are often contradictory. These effects depend on many parameters, such as the active substance, the mode of action, the mode of application and the dosage. In the field, determinants such as the physico-chemical behavior of the active substances, the soil type and other soil microorganisms contribute to the fate of pesticides and thus the amount of active substances to which AMF are exposed. This review highlights that the fate of AMF following pesticide use needs to be addressed in a broader agro-ecosystem context. © 2018 The Authors. Abstract:Growers of organic tree fruit face challenges in controlling some pests more easily suppressed by broad-spectrum insecticides in conventionally managed orchards. In recent decades, there has been a move towards organically growing varieties normally reliant on synthetic chemical pesticides (e.g., Gala), often to meet retailer/consumer demands. This inevitably makes crop protection in organic orchards more challenging, as modern varieties can be less tolerant to pests. In addition, there have been substantial reductions in plant protection product (PPP) approvals, resulting in fewer chemical options available for integrated pest management (IPM)-maintained orchards. Conversely, the organic management of fruit tree pests involves many practices that could be successfully implemented in conventionally grown crops, but which are currently not. These practices could also be more widely used in IPM-maintained orchards, alleviating the reliance on broad-spectrum PPP. In this review, we evaluate organic practices, with a focus on those that could be incorporated into conventional apple and pear production. The topics cover cultural control, biological control, physical and pest modifications. While the pests discussed mainly affect European species, many of the methods could be used to target other global pests for more environmentally sustainable practices.

Remifentanil for Fetal Immobilization and Maternal Sedation During Fetoscopic Surgery: A Randomized, Double-Blind Comparison with Diazepam

Abstract:Obstetric endoscopy procedures are routinely performed at our institution to treat selected complications of monochorionic twin gestation. We perform these procedures under combined spinal epidural anesthesia plus maternal sedation. In the absence of general anesthesia, fetal immobilization is not achieved. We hypothesized that remifentanil would induce adequate maternal sedation and provide fetal immobilization, which is equal or superior to that induced by diazepam. Fifty-four second trimester pregnant women were included in this randomized, double-blind trial. After combined spinal epidural anesthesia, maternal sedation was initiated using either incremental doses of diazepam or a continuous infusion of remifentanil. Maternal sedation, hemodynamics, side effects, and fetal hemodynamics and immobilization were evaluated before, during, and for 60 min after surgery. Remifentanil produced adequate maternal sedation with mild but clinically irrelevant respiratory depression (respiratory rate 13 ± 4 breaths/min and Pco2 38.6 ± 4 mm Hg at 40 min of surgery), whereas diazepam resulted in a more pronounced maternal sedation but no respiratory depression (respiratory rate 18 ± 3 breaths/min and Pco2 32.7 ± 3 mm Hg at 40 min of surgery). Compared with diazepam, fetal immobilization with remifentanil occurred faster and was more pronounced, resulting in improved surgical conditions; the number of gross body and limb movements was 12 ± 4 (diazepam) versus 2 ± 1 (remifentanil) at 40 min of surgery. Because of this, the mean (range) duration of surgery was significantly shorter in the remifentanil-treated patients, 60 (54–71) min versus 80 (60–90) min in the diazepam group. We conclude that remifentanil produces improved fetal immobilization with good maternal sedation and only minimal effects on maternal respiration.

Abstract:Ecological intensification of agro-ecosystems, based on the optimization of ecological functions such as biological pest control, to replace agrochemical inputs is a promising route to reduce the ecological footprint of agriculture while maintaining commodity production. However, the performance of organic farming, often considered as a prototype of ecological intensification, in terms of pest control remains largely unknown. Here, using two distinct meta-analyses, we demonstrate that, compared to conventional cropping systems, (i) organic farming promotes overall biological pest control potential, (ii) organic farming has higher levels of overall pest infestations but (iii) that this effect strongly depends on the pest type. Our study shows that there are lower levels of pathogen infestation, similar levels of animal pest infestation and much higher levels of weed infestation in organic than in conventional systems. This study provides evidence that organic farming can enhance pest control and suggests that organic farming offers a way to reduce the use of synthetic pesticide for the management of animal pests and pathogens without increasing their levels of infestation.

Abstract:The highly heterogeneous nature of Botrytis cinerea, provides adaptive benefits to variable environmental regimes. Disentangling pathogen population structure in anthropogenic agroecosystems is crucial to designing more effective management schemes. Herein, we studied how evolutionary forces exerted in different farming systems, in terms of agrochemicals-input, shape B. cinerea populations. In total, 360 B. cinerea isolates were collected from conventional and organic, strawberry and tomato farms in Cyprus and Greece. The occurrence and frequency of sensitivities to seven botryticides were estimated. Results highlighted widespread fungicide resistance in conventional farms since only 15.5 % of the isolates were sensitive. A considerable frequency of fungicide-resistant isolates was also detected in the organic farms (14.9%). High resistance frequencies were observed for boscalid (67.7%), pyraclostrobin (67.3%), cyprodinil (65.9%), and thiophanate-methyl (61.4%) in conventional farms, while high levels of multiple fungicide resistance were also evident. Furthermore, B. cinerea isolates were genotyped using a set of seven microsatellite markers (SSRs). Index of association analyses (Ia and rBarD) suggests asexual reproduction of the populations, even though the mating-type idiomorphs were equally distributed, indicating frequency-dependent selection. Fungicide resistance was correlated with farming systems across countries and crops, while SSRs were able to detect population structure associated with resistance to thiophanate-methyl, pyraclostrobin, boscalid, and cyprodinil. The expected heterozygosity in organic farms was significantly higher than in conventional; suggesting the absence of selective pressure that may change the allelic abundance in organic farms. However, genetic variance among strawberry and tomato populations was high, ranking host specificity higher than other selection forces studied.

Abstract:Viticulture worldwide is challenged by grapevine trunk diseases (GTDs). Involvement of arthropods in the dissemination process of GTD pathogens, notably esca pathogens, is indicated after detection of associated pathogens on arthropod exoskeletons, and demonstration of transmission under artificial conditions. The present study is the first to quantify spore loads via qPCR of the esca-relevant pathogen Phaeomoniella chlamydospora on arthropods collected in German vineyards, i.e., European earwigs (Forficula auricularia), ants (Formicidae), and two species of jumping spiders (Marpissa muscosa and Synageles venator). Quantification of spore loads showed acquisition on exoskeletons, but most arthropods carried only low amounts. The mycobiome on earwig exoskeletons was described for the first time to reveal involvement of earwigs in the dispersal of GTDs in general. Metabarcoding data support the potential risk of earwigs as vectors for predominantly Pa. chlamydospora and possibly Eutypa lata (causative agent of Eutypa dieback), as respective operational taxonomical unit (OTU) assigned genera had relative abundances of 6.6% and 2.8% in total reads, even though with great variation between samples. Seven further GTD-related genera were present at a very low level. As various factors influence the successful transmission of GTD pathogens, we hypothesize that arthropods might irregularly act as direct vectors. Our results highlight the importance of minimizing and protecting pruning wounds in the field.

Abstract:Herbicides are increasingly applied in vineyards worldwide. However, not much is known on potential side effects on soil organisms or on the nutrition of grapevines (Vitis vinifera). In an experimental vineyard in Austria, we examined the impacts of three within-row herbicide treatments (active ingredients: flazasulfuron, glufosinate, glyphosate) and mechanical weeding on grapevine root mycorrhization; soil microorganisms; earthworms; and nutrient concentration in grapevine roots, leaves, xylem sap and grape juice. The three herbicides reduced grapevine root mycorrhization on average by 53% compared to mechanical weeding. Soil microorganisms (total colony-forming units, CFU) were significantly affected by herbicides with highest CFUs under glufosinate and lowest under glyphosate. Earthworms (surface casting activity, density, biomass, reproduction) or litter decomposition in soil were unaffected by herbicides. Herbicides altered nutrient composition in grapevine roots, leaves, grape juice and xylem sap that was collected 11 months after herbicide application. Xylem sap under herbicide treatments also contained on average 70% more bacteria than under mechanical weeding; however, due to high variability, this was not statistically significant. We conclude that interdisciplinary approaches should receive more attention when assessing ecological effects of herbicides in vineyard ecosystems.

Abstract:Grapevine downy mildew (GDM) is one of the most serious diseases of grapevines. With limitations in the use of copper-based products imposed for organic agriculture by the European Union, research for alternatives is encouraged. The aim of this research was to follow a 2-year trial to evaluate the control of GDM using some alternative compounds, and to determine their effects on shoot growth, plant photosynthesis, and grape quality and quantity. Under low disease pressure, Bordeaux mixture, copper hydroxide, laminarin combined with low copper, and 0.5 and 0.8% chitosan had the lowest GDM incidence, reduced on leaves by 96, 95, 75, 56, and 81%, respectively, compared with the untreated control in the last survey. With high disease pressure, Bordeaux mixture, laminarin combined with Saccharomyces extracts, and 0.5 and 0.8% chitosan had the lowest GDM incidence, reduced on grape by 86, 37, 66, and 75%, respectively, compared with the untreated control in the survey of mid-July. Chitosan at 0.8% lowered net photosynthesis, due to reduced stomatal conductance, leaf area, and dry weight, with no negative effects observed on the quantity of the grape berries and the quality parameters of their juice. Among the alternatives to copper, chitosan provided the best GDM protection and reduced the vigor of the vegetation, inducing physiological changes without negative effects on grape production.

Abstract:Fertilizer practices can significantly impact the fruit quality and microbial diversity of the orchards. The fungi on the surface of fruits are essential for fruit storability and safety. However, it is not clear whether fertilization affects the fungal diversity and community structure on the surface of grape berries. Here, grape quality and the fungal diversity on the surface of grapes harvested from three fertilizer treatments were analyzed shortly after grape picking (T0) and following 8 days of storage (T1). The study involved three treatments: (1) common chemical fertilizer for 2 years (CH); (2) increased organic fertilizer and reduced chemical fertilizer for 1 year (A.O); and (3) increased organic fertilizer and reduced chemical fertilizer for 2 years (B.O). The application of increased organic fertilizer and reduced chemical fertilizer increased the soluble solids content (SSC) of the grape berries and decreased the pH of the grape juice. A total of 827,947 high-quality fungal sequences were recovered and assigned to 527 operational taxonomic units. Members of the Ascomycota phylum were dominant in all samples and accounted for 94.41% of the total number of detected sequences, followed by the Basidiomycota (5.05%), and unidentified fungi (0.54%). Alpha and beta diversity analyses revealed significantly different fungal populations in the three fertilizer treatments over the two time periods. The fungal diversity and richness on the grape berry surface in the B.O and A.O treatments were higher than those in the CH treatment. Among the detected fungi, the B.O treatments were mainly Pichia , Aureobasidium , and Candida genera, while the CH treatments were Botrytis , Aspergillus , and Penicillium . Moreover, significant differences were revealed between the two assessment times (T0 and T1). The samples from the T0 timepoint had higher fungal richness and diversity than the samples from T1 timepoint. Increasing organic fertilizer usage in grape management could improve grape quality and went on to increase the fungal diversity, as well as the relative abundance (RA) of beneficial fungi on grape berry surfaces. The correlation analysis suggested that the pH of the grape juice was significantly negatively correlated with fungal diversity parameters.

Abstract:We propose a general framework for smooth regression of a functional response on one or multiple functional predictors. Using the mixed model representation of penalized regression expands the scope of function on function regression to many realistic scenarios. In particular, the approach can accommodate a densely or sparsely sampled functional response as well as multiple functional predictors that are observed: 1) on the same or different domains than the functional response; 2) on a dense or sparse grid; and 3) with or without noise. It also allows for seamless integration of continuous or categorical covariates and provides approximate confidence intervals as a by-product of the mixed model inference. The proposed methods are accompanied by easy to use and robust software implemented in the pffr function of the R package refund. Methodological developments are general, but were inspired by and applied to a Diffusion Tensor Imaging (DTI) brain tractography dataset.

Abstract:Several pests and diseases have grapevine as their favourite host and the vineyard as preferred environment, so an intensive pesticide schedule is usually required to meet qualitative and quantitative production standards. The need to prevent the negative impact of synthetic chemical pesticides on human health and the environment and the consumer expectations in term of chemical residues in food stimulated the research of innovative tools and methods for sustainable pest management. The research project PURE (www.pure-ipm.eu) was a Europe-wide framework, which demonstrated that several solutions are now available for the growers and evaluated several new alternatives that are under development or almost ready for being applied in practice. Although the use of resistant/tolerant varieties is not yet feasible in several traditional grape growing areas, at least part of the synthetic chemical pesticides can be substituted with biocontrol agents to control pests and pathogens and/or pheromone mating disruption, or the number of treatments can be reduced by the use of decision support systems, which identify the optimal timing for the applications. This review presents the state of the art and the perspectives in the field of grapevine protection tools and strategies.

Abstract:The bryostatins are a unique family of cancer chemotherapeutic candidates isolated from marine bryozoa. While their molecular mode of action is not known, these macrolactones exhibit high affinities for protein kinase C (PKC) isozymes, compete for the phorbol ester binding site on PKC, and stimulate kinase activity in vitro and in vivo. Unlike the phorbol esters, they do not act as tumor promoters. Despite promising biological properties, the supply of these compounds is limited by the difficulty of their isolation from natural sources and their synthetic complexity. A new class of bryostatin analogues which retain the putative recognition domain of the bryostatins but are simplified through deletions and modifications in the C1—C14 spacer domain have been designed using computer models. A convergent synthesis has been realized for the production, in gram quantities, of these recognition and spacer domains whose coupling allows for the generation of a range of analogues. The final closure process involves a novel macrotransacetalization reaction which proceeds with complete stereoselectivity. The solution structures of two synthetic analogues were determined by NMR spectroscopy and found to be very similar to the previously reported structures of bryostatins 1 and 10. In addition, these structures appear to indicate that the stereochemistry of the C3 hydroxyl group plays a significant role in the conformation of the macrolactone. All analogues bound strongly to a mixture of PKC isozymes, and several exhibited significant levels of in vitro growth inhibitory activity against human cancer cell lines. Taken together, this work provides important steps toward the development and understanding of simplified, synthetically accessible analogues of the bryostatins as potential chemotherapeutic agents. © 1999 John Wiley & Sons, Inc. Natural vegetation benefits synergistic control of the three main insect and pathogen pests of a fruit crop in southern Africa

Abstract:Summary Most studies of the potential for natural habitat to improve agricultural productivity have been conducted in transformed, temperate regions, but little is known of the importance of agroecosystem services in biodiverse developing countries. Natural vegetation may promote the density and/or diversity of natural enemies of crop pests, but the strength of the effect varies, and few studies directly measure concurrent impacts on pest density. Considering multiple pest species within the same agroecosystem may help explain why some pests are more affected than others by landscape complexity. Here, we investigated multiple pest species (three species of Tephritidae fruit fly, leaf galling flies and pathogenic fungi Fusarium spp.) and their enemies in cultivated mango Mangifera indica , in north‐eastern South Africa. The density of generalist Tephritidae fruit flies increased with distance from natural vegetation during harvesting months, and predation rate of pupae sharply decreased from ~50% at the edge with natural vegetation to 0% at 250 m into the crop. Parasitism rates of the cryptic, gall‐forming fly increased with proximity to natural vegetation, but pest density was unrelated to distance from natural vegetation. Incidence of the fungal pathogen disease increased with distance from natural vegetation, possibly due to decreased predation of commensal mites. Although the relationship with distance to natural vegetation was significant for all species considered, the strength of this relationship varied across pest species and type of natural enemy studied, suggesting the benefits of natural vegetation depend on each natural enemy species' ability to disperse into the agricultural environment. Synthesis and applications . Our results suggest that natural vegetation is a net source of natural enemies in a region of South Africa that still contains much of its natural biodiversity. However, the decline in natural enemies, and increase in pests, with distance from natural habitat indicates that this biocontrol is limited by natural enemy dispersal. In landscapes like these that are still dominated by natural habitat, conservation biocontrol can still be improved by management aimed at providing corridors of key plants and habitat elements into the crops, to facilitate natural enemy dispersal. Our results suggest that natural vegetation is a net source of natural enemies in a region of South Africa that still contains much of its natural biodiversity.
