Abstract:We studied changes in body composition in great knots, Calidris tenuirostris, before and after a migratory flight of 5,400 km from northwest Australia to eastern China. We also took premigratory birds into captivity and fasted them down to their equivalent arrival mass after migration to compare organ changes and nutrient use in a low-energy-turnover fast with a high-energy-turnover fast (migratory flight). Migrated birds were as economical as any fasting animal measured yet at conserving protein: their estimated relative protein contribution (RPC) to the energy used was 4.0%. Fasted birds had an estimated RPC of 6.8% and, consequently, a much lower lean mass and higher fat content for an equivalent body mass than migrated birds. Lean tissue was catabolized from most organs in both groups, except the brain. Furthermore, a principal components biplot showed that individuals were grouped primarily on the basis of overall organ fat or lean tissue content rather than by the size of specific organs. This indicates that organ changes during migratory flight are similar to those of a low-energy fast, although the length of the fast in this study probably accentuated organ reductions in some functional groups. Whether the metabolic characteristics of a flying migratory fast follow the three-phase model described in many inactive fasting animals is unclear. We have some evidence for skeletal fat being catabolized without phase 3 of a fast having been reached.

Abstract:Long-distance migration in birds is a complex syndrome that involves high energy costs and, in some species, substantial physiological re-organisation. Such flexible migratory phenotypes are commonly associated with bird species flying non-stop across vast ecological barriers, where there are few opportunities to stop and refuel en route . Prior to making migratory flights, some species have been found to atrophy organs that are not required (e.g., digestive organs) and grow those associated with powering flight (pectora muscles and heart), presumably to optimise costs. However, most studies of this flexibility have required sacrificing study animals and this has limited our capacity to measure individual variation and its potential consequences. Here we investigate the behavioural and, indirectly, physiological adaptation of an arctic breeding long-distance migrant the light-bellied brent goose Branta bernicla hrota , during spring staging in southwest Iceland. We use a sequential sampling approach to record behavioural observations and conduct stable isotope analysis of faecal samples from uniquely marked individuals to assess protein catabolism. Individuals showed a three-phase fuel deposition process, with initial slow intake rates followed by hyperphagia and then a period of inactivity immediately prior to migratory departure (despite multiple days with favourable wind conditions). The C:N ratio and δ 15 N values in faeces were significantly linked to fat deposition during the latter stages and suggests catabolism (reorganisation of proteins) occurring prior to departure. Our results suggest a strategic delay in migratory departure to enable reorganisation into a flying phenotype and that the extent of this varies among individuals.

Abstract:ABSTRACT Migratory birds are physiologically specialized to accumulate massive fat stores (up to 50–60% of body mass), and to transport and oxidize fatty acids at very high rates to sustain flight for many hours or days. Target gene, protein and enzyme analyses and recent -omic studies of bird flight muscles confirm that high capacities for fatty acid uptake, cytosolic transport, and oxidation are consistent features that make fat-fueled migration possible. Augmented circulatory transport by lipoproteins is suggested by field data but has not been experimentally verified. Migratory bats have high aerobic capacity and fatty acid oxidation potential; however, endurance flight fueled by adipose-stored fat has not been demonstrated. Patterns of fattening and expression of muscle fatty acid transporters are inconsistent, and bats may partially fuel migratory flight with ingested nutrients. Changes in energy intake, digestive capacity, liver lipid metabolism and body temperature regulation may contribute to migratory fattening. Although control of appetite is similar in birds and mammals, neuroendocrine mechanisms regulating seasonal changes in fuel store set-points in migrants remain poorly understood. Triacylglycerol of birds and bats contains mostly 16 and 18 carbon fatty acids with variable amounts of 18:2n-6 and 18:3n-3 depending on diet. Unsaturation of fat converges near 70% during migration, and unsaturated fatty acids are preferentially mobilized and oxidized, making them good fuel. Twenty and 22 carbon n-3 and n-6 polyunsaturated fatty acids (PUFA) may affect membrane function and peroxisome proliferator-activated receptor signaling. However, evidence for dietary PUFA as doping agents in migratory birds is equivocal and requires further study.

Abstract:Migrating birds fly non-stop for hours or even for days. They rely mainly on fat as fuel complemented by a certain amount of protein. Studies on homing pigeons and birds flying in a wind-tunnel suggest that the shares of fat and protein on total energy expenditure vary with flight duration and body fat stores. Also, flight behaviour, such as descending flight, is expected to affect metabolism. However, studies on free flying migrant birds under natural conditions are lacking.

Abstract:Migratory birds undergo physiological and behavioral changes to fuel their high energy demanding migratory flights. They increase their food intake as a part of the preparation for migration which results in increase in their body mass. Fat, carbohydrate and protein acquired from food are stored mainly in the adipose tissue (triglycerides), muscle and liver (glycogen) and body organs (protein) in migratory birds. These stored foods act as fuels to support birds’ migratory flights. Dietary carbohydrates and lipids not only provide energy for migration but also help in fattening as carbohydrates can be converted into fat and lipids which can be stored. Lipolysis of adipose-stored fats leads to the production of triglycerides, fatty acids and glycerol, which provide energy for migration. Fats are depleted after long migratory flights and replenished during refueling at the stopover sites. Being chemically reduced and hydrophobic in nature, fat releases more energy on oxidation as compared to carbohydrate and protein. Due to its high energy-yielding nature, the fat is the preferred fuel to support migration in birds. Migratory birds deposit fat and deplete it during the course of migration. Though, the stored fat acts as the primary source of energy, metabolism of body protein also provides energy for migratory flights. Uric acid in plasma is elevated when protein is catabolized. The metabolism of carbohydrate, stored as glycogen in liver and muscle in migratory birds, produces glucose which also fuels migration. Glucose in migratory birds is maintained at stable levels in plasma and it provides energy only for a flight of short period. Further, catabolism of carbohydrate and protein results in release of metabolic water which helps the migratory birds to maintain their water balance during long dehydrating flight conditions. Different levels of plasma metabolites in migratory birds act as significant indicators of their physiological and metabolic state. Plasma metabolites also give an idea of feeding, fasting and refueling during migration in birds. The available information is scanty and fragmented about how birds meet their migratory requirements and overcome the physiological challenges encountered during migration. The present review article, therefore, focuses on the biomolecules and their plasma biochemistry during migration in birds.

Abstract:The efficacy and tolerability of erythromycin acistrate (EA), a new erythromycin derivative, and enterocoated erythromycin base (EB) were studied in 183 outpatients belonging to the personnel of Helsinki University Central Hospital. The patients had acute respiratory tract infections. The dosage of EA was 400 mg tid and that of EB 500 mg tid, and the treatment period ranged from seven to 14 days. The cure rate of patients taking full courses of treatment was good in both treatment groups, 96% in the EA-(n = 81) and 93% in the EB-group (n = 73). EA caused statistically significantly (P less than 0.05) less gastrointestinal side effects than EB. These side effects were also milder in the EA- than in the EB-group. The treatment was discontinued in seven patients (7%) in the EA- and in 12 patients (13%) in the EB-group because of gastrointestinal side effects. Neither of the drugs caused clinically important elevations in liver enzymes, nor were changes observed in the laboratory safety parameters measured. EA seems thus to be as effective as and better tolerated than enterocoated EB.

Abstract:Series capacitors (SCs) in transmission lines increase the complexity of transmission line protection. In this paper, a wavelet based energy traveling wave protection and fault locating method for series compensated transmission lines is proposed. The method requires GPS synchronized measurements at both ends of the line. The paper first presents the propagation properties of transient currents with different frequency, and then a proper frequency band condition is generated to minimize the transient attenuation. The method uses energy traveling wave in certain frequency band extracted by wavelet transform, and the wavefront time difference of two measurement units at each end of transmission line is introduced for the purpose of executing tripping logic and computing fault location. Numerical simulations verify that the method provides a robust protection criterion and an accurate fault locating method, independently of type, fault impedances and locations along the line. The algorithm also works for different SCs locations, and it is not influenced by MOV/air gap operation.

Abstract:Migratory songbirds undertake challenging journeys to reach their breeding grounds each spring. They accomplish these nonstop flapping feats of endurance through a suite of physiological changes, including the development of substantial fat stores and flight muscle hypertrophy and an increased capacity for fat catabolism. In addition, migratory birds may show large reductions in organ masses during flight, including the flight muscle and liver, which they must rapidly rebuild during their migratory stopover before replenishing their fat stores. However, the molecular basis of this capacity for rapid tissue remodeling and energetic output has not been thoroughly investigated. We performed RNA-sequencing analysis of the liver and pectoralis flight muscle of captive white-throated sparrows in experimentally photostimulated migratory and nonmigratory condition to explore the mechanisms of seasonal change to metabolism and tissue mass regulation that may facilitate these migratory journeys. Based on transcriptional changes, we propose that tissue-specific adjustments in preparation for migration may alleviate the damaging effects of long-duration activity, including a potential increase to the inflammatory response in the muscle. Furthermore, we hypothesize that seasonal hypertrophy balances satellite cell recruitment and apoptosis, while little evidence appeared in the transcriptome to support myostatin-, insulin-like growth factor 1-, and mammalian target of rapamycin-mediated pathways for muscle growth. These findings can encourage more targeted molecular studies on the unique integration of pathways that we find in the development of the migratory endurance phenotype in songbirds.NEW & NOTEWORTHY Migratory songbirds undergo significant physiological changes to accomplish their impressive migratory journeys. However, we have a limited understanding of the regulatory mechanisms underlying these changes. Here, we explore the transcriptomic changes to the flight muscle and liver of white-throated sparrows as they develop the migratory condition. We use these patterns to develop hypotheses about metabolic flexibility and tissue restructuring in preparation for migration.

Abstract:Birds remodel their flight muscle metabolism prior to migration to meet the physiological demands of migratory flight, including increases in both oxidative capacity and defence against reactive oxygen species. The degree of plasticity mediated by changes in these mitochondrial properties is poorly understood but may be explained by two non-mutually exclusive hypotheses: variation in mitochondrial quantity or individual mitochondrial function. We tested these hypotheses using yellow-rumped warblers (Setophaga coronata), a Nearctic songbird which biannually migrates two to five thousand kilometres. We predicted higher flight muscle mitochondrial abundance and substrate oxidative capacity and decreased reactive oxygen species emission in migratory warblers captured during autumn migration compared to a short-day photoperiod-induced non-migratory phenotype. We assessed mitochondrial abundance via citrate synthase activity and assessed isolated mitochondrial function using high-resolution fluororespirometry. We found 60% higher tissue citrate synthase activity in the migratory phenotype, indicating higher mitochondrial abundance. We also found 70% higher state 3 respiration (expressed per unit citrate synthase) in mitochondria from migratory warblers when oxidizing palmitoyl-carnitine, but similar H2O2 emission rates between phenotypes. By contrast, non-phosphorylating respiration was higher and H2O2 emission rates were lower in the migratory phenotype. However, flux through electron transport system complexes I-IV, II-IV and IV were similar between phenotypes. In support of our hypotheses, these data suggest that flight muscle mitochondrial abundance and function are seasonally remodeled in migratory songbirds to increase tissue oxidative capacity without increasing reactive oxygen species formation.

Abstract:Migratory birds excel in phenotypic flexibility, adapting physiologically as their life histories and environments require. Discerning the metabolic processes underlying migrants' physiology, an emergent property of multiple continuous and dynamic organism–environment interactions, is therefore challenging, particularly under natural conditions. Accordingly, analyses of snapshot‐sampled serum‐circulating metabolites, versatile and readily applicable for migrating birds, have increasingly become the method of choice for such physiologic inference. However, the atemporal nature of single sampling might obscure the links between observed metabolite concentrations and the processes producing them, necessitating an analytical decoupling of focal processes from their broader biochemical background. In the present study, we examined how variation in combined fat and muscle fuel stores, traits pivotal in migratory context, relates to the serum‐circulating metabolomes of spring‐migrating Eurasian blackcaps stopping‐over. Our analyses accounted for potential spatiotemporal influences in the form of time past night's fasting and random local conditions across three sites within the Negev Desert. We shifted the focus from compound‐level analysis of preselected metabolites towards the level of inclusive metabolome, quantifying serum‐circulating lipophilic and polar molecules via UHPLC–MS/MS untargeted metabolomic technique. Our results indicated a general relationship between fuel stores and the metabolome, comprising 16 326 lipophilic and 6923 polar compounds, among which 918 and 44 were annotated, respectively. By applying generalized latent‐variable linear modeling (GLLVM) upon concentrations of annotated metabolites, we identified several candidate biomarkers, some novel in migratory context, notably the fuel‐associated increase in serum ceramides likely derived from circulating very low‐density lipoproteins (VLDLs). Relying on estimated metabolite links with fuel and foraging time and on modeled residual covariations among metabolites, we demonstrate fuel–metabolite associations generally consistent with higher fat‐ and lower protein mobilization in birds having greater stores and with decreased fuel utilization as ingested nutrients accumulate over time, thus introducing a novel approach for the physiological study of migrating birds.

Comparative analysis of the plasma metabolome of migrating passerines during stopover: Novel insights into flight metabolism

Abstract:During long distance migration, many birds may experience periods of either prolonged fasting, during endurance flights, or extensive feeding during stopovers. It was previously shown that habitat selection during stopover can largely affect the migration outcome of an individual. Despite decades of research of the avian metabolism during stopover and migration, many questions have remained unanswered, as such research mainly focused on targeted metabolites and fat metabolism. Here, we examined the plasma-metabolome of migrating passerines prior to their crossing the Sahara Desert. Birds were sampled at two sites populated by Pistacia trees, bearing fat-rich fruits, and at an additional site dominated by blooming Eucalyptus trees. The blood samples were analyzed using both GC-MS and LC-MS, using an untargeted approach. We found that birds from one of the sites had a distinguish metabolic profile, suggesting recent landing. Examination of metabolic pathways activated during stopovers indicated a crucial role for cycling glucose through the Cori and Cahill cycles in resting and recovery processes. This novel perspective, conducted on free-ranging birds, suggests the evolution of avian insulin resistance due to factors such as endurance exercise, fasting, and a preference for fatty acid oxidation during migration, akin to cell trauma recovery. Additionally, we investigated inter-site variations in birds’ metabolic profiles. Significant variations were observed in both polar and lipophilic metabolites among the sites. Differences in polar metabolites were primarily attributed to variations in the physiological state of the birds between sites, while distinctions in the lipophilic profiles of rested birds were linked to variations in their primary food sources. This study underscores the challenge of interpreting commonly used indicators for assessing migrating birds’ physiological states and site quality, which are predominantly derived from lipid metabolism, in complex ecological systems.

Abstract:During migratory stopovers, birds must make decisions about when and where to travel and these decisions are likely contingent on their fuel stores, food availability, and antioxidant capacity as well as seasonal changes in key environmental factors. We conducted a field experiment on an offshore stopover site (Block Island, Rhode Island, United States: 41°130N, 71°330W) during autumn migration to test the hypothesis that birds with greater fuel stores and non-enzymatic antioxidant capacity have shorter stopovers than lean birds with low antioxidant capacity, and to determine the extent to which this depends on migration strategy. We used a 2 × 2 factorial field experiment (two levels each of available food and dietary polyphenols) with four species of songbirds kept in captivity for 3–5 days to produce experimental groups with different fuel stores and antioxidant capacity. We attached digital VHF transmitters to assess stopover duration and departure direction using automated telemetry. Non-enzymatic antioxidant capacity increased during refueling for Red-eyed Vireos ( Vireo olivaceus ) and Blackpoll Warblers ( Setophaga striata ) fed ad lib diets, and for ad lib fed Hermit Thrushes ( Catharus guttatus ) supplemented with polyphenols, but not for Yellow-rumped Warblers ( Setophaga coronata coronata ). Glutathione peroxidase (GPx) decreased during captivity and was influenced by dietary treatment only in Red-eyed Vireos. Oxidative damage decreased during captivity for all species except Yellow-rumped Warblers. Stopover duration was shorter for Vireos and Blackpolls fed ad lib as compared to those fed maintenance. Ad lib fed Hermit Thrushes supplemented with polyphenols had shorter stopovers than those fed ad lib , as did thrushes fed at maintenance and supplemented with polyphenols compared with those fed at maintenance alone. There was no influence of condition on stopover duration for Yellow-rumped Warblers. Departure direction was not strongly related to condition, and birds primarily reoriented north when departing Block Island. Thus, fat stores and oxidative status interacted to influence the time passerines spent on stopover, and condition-dependent departure decisions were related to a bird’s migration strategy. Therefore, seasonal variation in macro- and micro-nutrient resources available for refueling at stopover sites can affect body condition and antioxidant capacity and in turn influence the timing and success of migration.

Abstract:Refuelling by migratory birds before take-off on long flights is generally considered a two-phase process, with protein accumulation preceding rapid fat deposition. The first phase expresses the demands for a large digestive system for nutrient storage after shrinkage during previous flights, the second phase the demands for fat stores to fuel the subsequent flight. At the last staging site in northward migration, this process may include expression of selection pressures both en route to and after arrival at the breeding grounds, which remains unascertained. Here we investigated changes in body composition during refuelling of High Arctic breeding red knots (Calidris canutus piersmai) in the northern Yellow Sea, before their flight to the tundra. These red knots followed a three-phase fuel deposition pattern, with protein being stored in the first and last phases, and fat being deposited mainly in the second phase. Thus, they did not shrink nutritional organs before take-off, and even showed hypertrophy of the nutritional organs. These suggest the build up of strategic protein stores before departure to cope with a protein shortage upon arrival on the breeding grounds. Further comparative studies are warranted to examine the degree to which the deposition of stores by migrant birds generally reflects a balance between concurrent and upcoming environmental selection pressures.
Flying fast improves aerodynamic economy of heavier birds

Abstract:A paradox of avian long-distance migrations is that birds must greatly increase their body mass prior to departure, yet this is presumed to substantially increase their energy cost of flight. However, here we show that when homing pigeons flying in a flock are loaded with ventrally located weight, both their heart rate and estimated energy expenditure rise by a remarkably small amount. The net effect is that costs per unit time increase only slightly and per unit mass they decrease. We suggest that this is because these homing flights are relatively fast, and consequently flight costs associated with increases in body parasite drag dominate over those of weight support, leading to an improvement in mass-specific flight economy. We propose that the relatively small absolute aerodynamic penalty for carrying enlarged fuel stores and flight muscles during fast flight has helped to select for the evolution of long-distance migration.

Abstract:Abstract Methylmercury (MeHg) is a global pollutant that can cause metabolic disruptions in animals and thereby potentially compromise the energetic capacity of birds for long-distance migration, but its effects on avian lipid metabolism pathways that support endurance flight and stopover refueling have never been studied. We tested the effects of short-term (14-d), environmentally relevant (0.5 ppm) dietary MeHg exposure on lipid metabolism markers in the pectoralis and livers of yellow-rumped warblers ( Setophaga coronata ) that were found in a previous study to have poorer flight endurance in a wind tunnel than untreated conspecifics. Compared to controls, MeHg-exposed birds displayed lower muscle aerobic and fatty acid oxidation capacity, but similar muscle glycolytic capacity, fatty acid transporter expression, and PPAR expression. Livers of exposed birds indicated elevated energy costs, lower fatty acid uptake capacity, and lower PPAR- γ expression. The lower muscle oxidative enzyme capacity of exposed birds likely contributed to their weaker endurance in the prior study, while the metabolic changes observed in the liver have potential to inhibit lipogenesis and stopover refueling. Our findings provide concerning evidence that fatty acid catabolism, synthesis, and storage pathways in birds can be dysregulated by only brief exposure to MeHg, with potentially significant consequences for migratory performance.

Abstract:Seasonal bird migration may provide energy benefits associated with moving to areas with less physiologically challenging climates or increased food availability, but migratory movements themselves may carry high costs. However, time-dynamic energy profiles of free-living migrants-especially small-bodied songbirds-are challenging to measure. Here we quantify energy output and thermoregulatory costs in partially migratory common blackbirds using implanted heart rate and temperature loggers paired with automated radio telemetry and energetic modelling. Our results show that blackbirds save considerable energy in preparation for migration by decreasing heart rate and body temperature 28 days before departure, potentially dwarfing the energy costs of migratory flights. Yet, in warmer wintering areas, migrants do not appear to decrease total daily energy expenditure despite a substantially reduced cost of thermoregulation. These findings indicate differential metabolic programmes across different wintering strategies despite equivalent overall energy expenditure, suggesting that the maintenance of migration is associated with differences in energy allocation rather than with total energy expenditure.
Stopovers Serve Physiological Recovery in Migratory Songbirds

Abstract:AbstractMigrating birds perform extreme endurance exercise when flying. This shifts the balance between the production of reactive oxygen species and the antioxidant defense system toward the former, potentially generating oxidative damages. In between migratory flights, birds make stopovers, where besides accumulating fuel (mainly fats), they are assumed to rest and recover from the strenuous flight. We performed a series of studies on both temporarily caged (northern wheatears) and free-flying (northern wheatears and European robins) migrants to investigate whether migrants recover during stopover by decreasing the amount of oxidative lipid damage (malondialdehyde [MDA]) and/or increasing the total nonenzymatic antioxidant capacity (AOX). In caged wheatears, MDA decreased within a single day. These birds were able to simultaneously accumulate considerable amounts of fuel. Also, in the free-flying wheatears, there was a decrease in MDA during stopover; however, this process seemed incompatible with refueling. The reason for this difference could relate to constraints in the wild that are absent in caged birds, such as food limitation/composition and locomotor activity. In the robins, there was a near significant decrease in MDA concentration in relation to how long the birds were already at stopover, suggesting that this species also physiologically recovers during stopover. AOX did not change during stopover in either of the wheatear studies. For the robins, however, uric acid-corrected AOX declined during stopover. Our results show that during stopover, migrating birds rapidly reduce oxidative lipid damage, thereby likely recovering their physiological state. In addition to the commonly accepted function of refueling, stopovers thus probably serve physiological recovery.
Differential changes in the morphology and fuel loads of obligatory and partial migrant passerines over half a century in Britain

Abstract:Migratory distances and stopover locations are changing for many passerines in response to climate change. Morphological changes have been linked to rising global temperatures in both migrants and residents, but the implications of these changes on fuel loads, and associated flight ranges are little studied. Wing length and body mass changes between 1964 and 2020 were calculated for 15 migrant and partially migrant passerines in Britain. Changes in fuel load and lean body mass were also estimated and used to predict flight ranges. Twelve of the species have undergone morphological changes and eight species, estimated fuel load changes. Nine species were estimated to have reduced flight ranges, indicating that the morphological changes have not compensated fully for the reduction in flight range experienced since 1964. Partial migrants showed greater decreases in flight ranges than did full migrants, which may indicate greater behavioural plasticity in the former. Those species which do not adapt morphologically or behaviourally may be unable to complete long migrations, resulting in restriction to sub-optimal breeding/wintering habitats, or a need for a sooner first stop and more stops en route. This highlights the importance of conserving migratory stopover sites, particularly in the Mediterranean and North Africa that immediately precede major geographical barriers, as-well-as breeding and wintering grounds.

Abstract:Dietary micronutrients have the ability to strongly influence animal physiology and ecology. For songbirds, dietary polyunsaturated fatty acids (PUFAs) and antioxidants are hypothesized to be particularly important micronutrients because of their influence on an individual's capacity for aerobic metabolism and recovery from extended bouts of exercise. However, the influence of specific fatty acids and hydrophilic antioxidants on whole‐animal performance remains largely untested. We used diet manipulations to directly test the effects of dietary PUFA, specifically linoleic acid (18:2n6), and anthocyanins, a hydrophilic antioxidant, on basal metabolic rate (BMR), peak metabolic rate (PMR), and rates of fat catabolism, lean catabolism, and energy expenditure during sustained flight in a wind tunnel in European starlings (Sturnus vulgaris). BMR, PMR, energy expenditure, and fat metabolism decreased and lean catabolism increased over the course of the experiment in birds fed a high (32%) 18:2n6 diet, while birds fed a low (13%) 18:2n6 diet exhibited the reverse pattern. Additionally, energy expenditure, fat catabolism, and flight duration were all subject to diet‐specific effects of whole‐body fat content. Dietary antioxidants and diet‐related differences in tissue fatty acid composition were not directly related to any measure of whole‐animal performance. Together, these results suggest that the effect of dietary 18:2n6 on performance was most likely the result of the signaling properties of 18:2n6. This implies that dietary PUFA influence the energetic capabilities of songbirds and could strongly influence songbird ecology, given their availability in terrestrial systems.
