Abstract:Soil compaction easily causes root hypoxia stress, resulting in poor root growth and the absorption of soil water and nutrients. We hypothesized that aerated irrigation (AI) could enhance nutrient uptake and utilization, thus unlocking the high yield potential by increasing soil aeration and root morphology indicators compared with that in the non-aeration treatment. A greenhouse experiment was conducted to investigate the effect of soil aeration and root morphology on the yield of greenhouse cucumbers. The dissolved oxygen (DO) in irrigation water at 10 mg L−1 (A1), 20 mg L−1 (A2), and non-aeration treatment (A0) were applied via a subsurface drip irrigation system. The soil respiration rates, DO in soil water, root morphology, and crop yield were measured. The results showed that AI could significantly improve the soil respiration rate, DO in soil water, and root morphology compared with non-aeration treatment. The A2 significantly increased soil respiration rate by 11.63% and 11.93%, respectively, compared with the A1 and A0 treatments. Under A1 and A2, the DO in soil water increased by 20.01% and 18.02%, respectively, compared with the A0. Moreover, during the flowering and fruit set, the mature, and the late stages, the root surface area, root volume, root tip number, root forks, and root dry weight in the A2 treatment significantly increased than that in the A0 treatment. The soil respiration rate, DO in soil water, root length, and root forks were the main indexes correlated to the yield, respectively. The DO in soil water and root forks number significantly influenced the yield. The cucumber yield and economic benefits in A2 peaked at 53.04 t ha−1 and 3.95 × 104 USD ha−1, increased by 7.86% and 7.30% compared with that in the A0 treatment, respectively (p < 0.05). The results could provide technical support and scientific knowledge for regulating greenhouse cucumbers under AI.

Aerated Irrigation Promotes Soil Respiration and Microorganism Abundance around Tomato Rhizosphere

Abstract:Aerated irrigation (AI), by which aerated water is supplied to soil through subsurface drip irrigation (SDI), has shown high yield potential and application prospects. This study evaluated the impact of AI on soil aeration, respiration components, and microorganism abundance to determine the relationships between these parameters. Four treatments (W1AI, W2AI, W1CK, and W2CK) combining AI and nonaerated SDI (CK) and two irrigation levels (W1 and W2, with crop-pan coefficients of 0.6 and 1.0) were conducted in a greenhouse during two tomato (Lycopersicon esculentum Mill.) growing seasons in April to July in 2015 and 2016. The results showed that AI improved soil aeration conditions, with soil oxygen concentration and air-filled porosity significantly increasing by 5.86 and 5.16%, respectively, compared with CK. The W1AI treatment increased soil, microbial, and root respiration as well as bacteria, actinomycetes, and fungi abundance by 18.8, 10.0, 23.4, 30.1, 29.4, and 30.1%, respectively, compared with W1CK, and W2AI significantly increased these by 24.2, 15.3, 28.7, 39.2, 39.3, and 37.1%, respectively, compared with W2CK. Moreover, the abundance of bacteria, actinomycetes, and fungi with W2AI significantly increased by 26.9, 27.1, and 24.8%, respectively, compared with W1AI. Soil oxygen concentration (under controlled soil temperature) and air-filled porosity positively correlated with soil respiration and microorganism abundance. Mean fruit yield with AI was 22.4% higher than CK. Aerated irrigation therefore stimulated soil respiration and microbial proliferation by improving soil aeration conditions. Enhanced soil aeration, respiration, and microorganism abundance under AI provide economic benefit by improving fruit yield.

Abstract:<p>Soil aeration can enhance soil enzyme activity and plant growth. These effectsmay be caused by variations in soil microbial diversity and plant root development. However, little is known about the responses of soil microbial composition, structure and diversity and plant root development to artificial soil aeration. The aims of this study were to examine the composition, structure and diversity of rhizosphere bacterial communities and tomato root morphology under 4 aeration volumes in combination with drip tubing placed at 2 different soil depths. The aeration volumes (V) were 0, 0.5, 1, and 1.5 times (CK, V1, V2, and V3, respectively) the standard aeration volume. The drip irrigation tube burial depths were 15 (D15) and 40 cm (D40). The results demonstrated that soil aeration had a significant influence on root performance (root morphology and activity) and soil bacterial diversity in the tomato root zone. In comparison to the CK treatment, V3 soil aeration increased root length, surface area, tip number and activity by 7.9%, 6.3%, 14.5% and 7.0%, respectively. The ACE, Chao index, and Shannon diversity increased by 5.3%, 4.7% and 3.5%, respectively. The abundance of Acidobacteria increased and that of Gammaproteobacteria decreased in response to the soil aeration treatments; conversely, Geobacteraceae and Halanaerobiaceae were eliminated. Moreover, different sub‐surface tubing placement depths changed the rhizosphere bacterial community. The results indicate that soil aeration ameliorates hypoxic conditions and provides benefits to soil bacterial communities and root morphology.</p><p>This article is protected by copyright. Effects of copper based foliar fertilizer and controlled release fertilizer on growth and leaf protective enzyme activities of pepper

Abstract:Objectives]According to characteristics of fertilizer loving, fertilizer tolerance and strict requirement for water, the objective of this experiment was to study effects of copper-based foliar fertilizers ( CBFF ) and controlled-release fertilizers ( CRF) on yield, quality, copper partitioning and activities of protective enzymes in plants of pepper, and to provide a theoretical basis for high-yielding and cultivation of pepper.[Methods]With the cultivar, Chifeng Niujiaowang as the test material, a pot experimental design was a factorial arrangement in a split-plot with two replicates.The main plots were CKW ( sprayed with water ) and CBFF ( copper-based foliar fertilizers), and the sub-plots were NF (no fertilizer), CCF (common compound fertilizers), CRF (controlled-release fertilizers) and CRF +W ( controlled-release fertilizers with water retaining agent ) .There were eight treatments as the combination both foliar fertilizer and soil fertilizer.Pepper fruits were harvested and weighted based on pepper maturation.Fresh leaves and a portion of fruits were collected and calculated in the fruit maturation period, and the concentrations of Cu, Fe and Zn in the plants were determine by using atomic absorption spectrometry.[Results]In 2011 and 2012 pepper growing seasons, the different interactions of CBFF and CRF have significant effects on the yield, and the optimal combination of fertilization for the yield is CBFF+CRF+W. Under the optimal application condition, the yields are significantly increased by 24.1%and 29.0%, respectively in the first year (2011) and the second year (2012) compared to CKW+CRF+W.In growing seasons, compared to the spraying clear water, the foliar spraying copper fertilizer significantly increases copper concentration in the pepper plants.Compared with the utilization of CRF+W, the Cu concentration of pepper fruits is increased by 103.9% with CBFF.Meanwhile, the active oxygen that is produced by copper based foliar fertilizers leads to increase the superoxide dismutase ( SOD) , peroxidase ( POD) activities, while the application of CBFF decreases the catalase (CAT) activity.By applying the CRF+W, the SOD activity of the spraying CBFF is increased by 26.6%.In the treatment with CBFF+CRF, higher POD activity and lower CAT activity are observed in leaves of pepper.Considering a variation in the content in pepper fruits, the treatments of CBFF+CRF and CBFF+CRF+W increase the soluble protein and vitamin C contents by a range from 12.8%-178.4%.[Conclusions] These results indicate that the appropriate combined fertilization management can increase micronutrients contents in peppers and improve the yield and nutrition quality of pepper.
Effects of Aerated Drip Irrigation on the Soil Nitrogen Distribution, Crop Growth, and Yield of Chili Peppers

Abstract:In order to study the soil nitrogen (N) distribution pattern in the root zone of chili peppers under aerated drip irrigation (ADI) conditions and analyze the relationship between soil N distribution and crop growth, two irrigation methods (conventional drip irrigation and ADI) and three N levels (0, 140, and 210 kg hm−2) were set up in this experiment. Soil samples were collected by the soil auger method at the end of different reproductive periods, and the uniformity coefficient of soil N in the spatial distribution was calculated by the method of Christiansen's coefficient. The growth status and soil-related indices of pepper were determined at each sampling period, and the relationships between soil N distribution and chili pepper growth were obtained based on principal component analysis (PCA). The results showed that the spatial content of soil nitrate-N (NO3−-N) fluctuated little during the whole reproductive period of chili peppers under ADI conditions, and the coefficient of uniformity of soil NO3−-N content distribution increased by 5.29~37.63% compared with that of conventional drip irrigation. The aerated treatment increased the root length and surface area of chili peppers. In addition, the ADI treatments increased the plant height, stem diameter, root vigor, and leaf chlorophyll content to some extent compared with the nonaerated treatment. The results of PCA showed that the yield of chili peppers was positively correlated with the uniformity coefficient of soil NO3−-N, root vigor, and root length. ADI can significantly improve the distribution uniformity of soil NO3−-N and enhance the absorption and utilization of N by the root system, which in turn is conducive to the growth of the crop, the formation of yields, and the improvement of fruit quality.

[Effects of effective microorganisms on growth promotion and the rhizosphere eukaryotic community structure of pepper in Xinjiang, China]

Abstract:Effective microorganisms (EM) might alleviate deterioration of soil environmental quality and yield decline of pepper (Capsicum annuum) caused by continuous replanting and imbalanced fertilizer application in Xinjiang. We investigated the effects of applying EM microbial agent on the growth of pepper plants, yield, soil nutrient content, soil enzyme activity, and rhizosphere eukaryotic community. The results showed that the application of EM microbial agent increased plant height, stem diameter, leaf length, leaf width and root length by 22.6%, 35.3%, 33.3%, 29.7% and 15.1%, respectively. It also increased fruit width, individual fruit weight, and yield by 5.3%, 42.9%, and 74.7%, respectively. After the application of EM microbial agent, the levels of soil available nitrogen increased by 10.2% and 5.8% during the flowering and maturity stages, respectively. Similarly, available phosphorus increased by 10.4% and 13.4%, respectively. The soil sucrase activity was increased by 40.7%, 14.6%, and 9.3% during the seedling, flowering, and maturity stages, respectively. Urease activity was also increased by 7.9%, 10.2%, and 11.5%, respectively. Furthermore, the application of EM microbial agent increased soil peroxidase activity by 16.8% and 44.6% at flowering and maturity stages, respectively. The application of microbial agent significantly altered the β-diversity of the rhizosphere eukaryotic community in pepper plants. Specifically, microbial agent increased the relative abundances of populations belonging to Enchytraeus and Sminthurides genera, which could contribute to soil improvement and nutrient cycling. Compared to the CK, the relative abundance of pathogenic microorganisms including Olpidium and Aplanochytrium genera decreased by 98.0% and 89.3%, and the relative abundance of the Verticillium decreased to 0. These results demonstrated that EM microbial agent could increase soil nutrient content, enhance soil enzyme activity, and reduce soil pathogenic fungi in the pepper cultivation areas of Xinjiang, thus achieving beneficial effects on pepper growth and fruit yield.
Coupled treatment of wet-dry alternation irrigation and seedling-promoting fertilizer has a beneficial effect on the rhizosphere microbial community profile and regenerating physiology of ratoon rice

Abstract:Reasonable water and nitrogen management is a key measure to promote root function and improve the axillary bud germination rate and yield of ratoon rice. However, the effects of water and nitrogen management on root physiology and rhizosphere microorganisms in ratoon rice are largely unclear. This study focused on endogenous hormones in the roots and the rhizosphere microbial community structure of ratoon rice under different water and seedling-promoting fertilizer treatments. The endogenous hormones in the root-stem junctions and the rhizosphere bacterial community structure varied significantly between the wet-dry alternation irrigation coupled with the application of seedling-promoting nitrogen fertilizer (WDN), wet-dry alternation irrigation without nitrogen application (WD), and flood irrigation without nitrogen application (FI). Compared with WD and FI, WDN significantly increased auxin content by 8.13 and 4.83 times and by 14.52 and 14.50 times in the root-stem junctions at the 5th and 10th days after treatment, respectively, and gibberellin A 3 content by 16.70% and 29.74% at the 10th day, respectively. The WDN treatment also significantly increased the diversity and evenness indices of rhizosphere bacterial communities, as well as the relative abundances of plant growth-promoting rhizobacteria, including Brevibacillus , Paenibacillus , Rhizobium , and Clostridium . The white root number and the contribution of the white root weight to the total root weights of ratoon rice in the WDN treatment were significantly higher than those in the WD and FI treatments by 34.41% and 25.98% and by 220.51% and 190.16%, respectively. The results showed that the WDN could significantly increase the levels of endogenous hormones related to root primordium formation and root elongation and improve the population of bacteria related to nutrient cycling and growth promotion in the rhizosphere soil. These improvements significantly promoted the new roots and the growth of low nodal axillary buds, and increased the yield of ratooning rice. The results provide guidance for improving the rhizosphere cultivation of ratooning rice.

Abstract:Microplastics (MPs) produced by the decomposition of plastics exist persistently, interfering with soil fertility and plant nutrition. Both arbuscular mycorrhizal (AM) fungi and earthworms are beneficial in terrestrial ecosystems, but their interactions under MPs contamination are unclear so far. Here, the influences of inoculating earthworms ( Eisenia fetida ) on indigenous AM fungi and pepper ( Capsicum annuum L.) growth were investigated in a vegetable soil treated with 0.1% low-density polyethylene (LDPE), while the specific interactions of earthworm and AM fungus ( Funneliformis caledonium ) under LDPE contamination were further resolved in another experiment using sterilized soil. Inoculation of earthworms shifted soil AM fungal community structure, replacing the predominant genus Glomus by Paraglomus , and increased the abundance, diversity (i.e., Shannon) index, and root colonization rate of AM fungi by 108, 34.6 and 45.0%, respectively. Earthworms also significantly decreased soil pH, and significantly increased soil alkaline phosphatase (ALP) activity, shoot biomass and fruit yield of pepper by 394, 82.8 and 188%, respectively. In the sterilized soil, both E. fetida and F. caledonium improved pepper growth, while the latter noticeably increased phosphorus (P) translocation efficiency from root to shoot, and the combination induced the highest soil ALP activity and pepper fruit yield. Furthermore, the significantly interactive effects between earthworm and AM fungus were observed in soil pH and available P concentration, as well as in shoot P concentration and fruit yield of pepper. This study revealed the interaction between earthworms and AM fungi under MPs contamination conditions for the first time, indicating that earthworms could facilitate vegetable growth via enhancing the propagation and P-promoting function of AM fungi in LDPE-contaminated soils. Contrasting effect of irrigation practices on the cotton rhizosphere microbiota and soil functionality in fields

Abstract:Drip irrigation under plastic film mulch is a common agricultural practice used to conserve water. However, compared to traditional flood irrigation with film mulch, this practice limit cotton root development from early flowering stage and may cause premature senescence in cotton. Changes of root will consequently shape the composition and activity of rhizosphere microbial communities, however, the effect of this farming practice on cotton rhizosphere microbiota remains poorly understood. This study investigated rhizosphere bacteria and soil functionality in response to different irrigation practices —including how changes in rhizosphere bacterial diversity alter soil nutrient cycling. Drip irrigation under plastic film mulch was shown to enhance bacterial diversity by lowering the salinity and increasing the soil moisture. However, the reduced root biomass and soluble sugar content of roots decreased potential copiotrophic taxa, such as Bacteroidetes , Firmicutes , and Gamma- proteobacteria , and increased potential oligotrophic taxa, such as Actinobacteria , Acidobacteria , and Armatimonadetes . A core network module was strongly correlated with the functional potential of soil. This module not only contained most of the keystone taxa but also comprised taxa belonging to Planctomycetaceae , Gemmatimonadaceae , Nitrosomonadaceae , and Rhodospirillaceae that were positively associated with functional genes involved in nutrient cycling. Drip irrigation significantly decreased the richness of the core module and reduced the functional potential of soil in the rhizosphere. Overall, this study provides evidence that drip irrigation under plastic film mulch alters the core bacterial network module and suppresses soil nutrient cycling.

Rhizosphere Ventilation Effects on Root Development and Bacterial Diversity of Peanut in Compacted Soil

Abstract:Soil compaction is one of the crucial factors that restrains the root respiration, energy metabolism and growth of peanut (Arachis hypogaea L.) due to hypoxia, which can be alleviated by ventilation. We therefore carried out a pot experiment with three treatments: no ventilation control (CK), (2) ventilation volumes at 1.2 (T1), and 1.5 (T2) times of the standard ventilation volume (2.02 L/pot). Compared to no-ventilation in compacted soil, ventilation T1 significantly increased total root length, root surface area, root volume and tips at the peanut anthesis stage (62 days after sowing), while T2 showed a negative impact on the above-mentioned root morphological characteristics. At the podding stage (S2, 95 days after sowing), both ventilation treatments improved root morphology, especially under T1. Compared to CK, both ventilation T1 and T2 decreased the activities of enzymes involving the anaerobic respiration, including root lactate dehydrogenase, pyruvate decarboxylase and alcohol dehydrogenase. The activities of antioxidant enzymes of root superoxide dismutase, peroxidase and catalase also decreased at S1, while superoxide dismutase and peroxidase significantly increased under T1 at S2. The ventilation of compacted soil changed soil nitrogen-fixing bacterial communities, with highest bacterial alpha diversity indices under T1. The Pearson correlation analyses indicated a positive relationship between the relative abundance of Bradyrhizobiaceae and root activity, and between unclassified_family of Rhizobiales and the root surface area, while Enterobacteriaceae had a negative impact on the root nodule number. The Pearson correlation test showed that the root surface, tips and activity positively correlated with root superoxide dismutase and peroxidase activities. These results demonstrate that soil ventilation could enhance plant root growth, the diversity and function of soil nitrogen-fixing bacterial communities. The generated results from this present study could serve as important evidence in alleviating soil hypoxia caused by compaction.

Abstract:Plant root‐associated microbial communities profoundly affect plant nutrition and productivity. Although elevated atmospheric CO2 and warming affect above‐ and belowground plant processes, it remains unclear how root‐associated microbial communities respond to elevated CO2 and warming. In this study, an open‐air field experiment was conducted to assay the interactive effects of elevated CO2 (500 ppm) and warming (+2 °C) on the root‐associated microbiota and soil enzyme activities in a rice‐wheat rotation ecosystem. The results revealed that elevated CO2 significantly increased rhizosphere soil organic carbon (SOC) and total nitrogen contents. In addition, glucosidase, β‐xylosidase, and phosphatase activities significantly increased. The richness and Shannon diversity indices were significantly higher in rhizosphere soil than in root endosphere. Elevated CO2 and warming significantly impacted the rhizosphere soil microbiota and altered their composition by changing the relative abundance of some specific groups. Soil pH, SOC, and available potassium content significantly altered the dominant bacterial phyla in the rhizosphere. SOC affected root endophytic bacterial phyla. Bacterial and fungal genera were significantly correlated with soil variables in the rhizosphere than in the root endosphere. These results indicate that microbial communities in the rhizosphere are more sensitive to elevated CO2 and warming than those in the root endosphere.

Effects of Irrigation Using Activated Brackish Water on the Bacterial Community Structure of Rhizosphere Soil

Abstract:The technology for the activation of irrigation brackish water was successful at improving the soil environment and crop yields. However, very few studies have examined how activated brackish water irrigation affects the rhizosphere bacterial communities and network patterns. By combining 16S rRNA high-throughput sequencing, we evaluated the diversity and composition of the rhizosphere bacterial community after being subjected to different types of irrigation with activated brackish water. We also analyzed the correlation and co-occurrence networks among the bacterial diversity, composition, and rhizosphere soil properties. The results showed that compared with brackish water irrigation, the salt content of activated irrigation water significantly decreased by 9.35%, 9.83%, and 12.95%, respectively. Irrigation with different types of activated brackish water had no significant ( p > 0.05) effect on the diversity of the rhizosphere bacterial community, but it significantly changed its community composition, which was primarily dominated by soil nutrient indicators. The soil total nitrogen (TN) showed a significant ( p < 0.01) negative correlation with the Chao1 index. Additionally, the changes in bacterial communities under different types of activated brackish irrigation water mainly occurred at the genus level. We showed that the rhizosphere soil that had been irrigated with oxygenated brackish water and magnetized brackish water better supported the reproduction of some soil-borne pathogens. Magnetization-oxygenation coupling treatment could significantly reduce the colonization of soil-borne pathogens of the rhizosphere soil, while also favoring the function of functional bacteria involved in soil nutrient transformation. This study highlights the main factors affecting the rhizosphere soil bacterial community structure by activated brackish water irrigation, while also providing new technical support for brackish water irrigation.

Abstract:The Rhodopseudomonas palustris ( R. palustris ) PSB06 can promote crop growth, as it maybe regulates microbial communities in plant root soil, soil physicochemical properties, thus creating a favorable habitat for the crop growth. However, there are few studies on the yields and rhizosphere microbial community of R. palustris PSB06 agent. In the study, the high-throughput sequencing was used to study the changes of rhizosphere soil bacterial community after PSB06 treatment. The results indicated R. palustris PSB06 agent increased the pepper yield by 33.45% when compared to control group, with better effect than other treatments. And it also significantly increased soil nitrogen concentration. R. palustris PSB06 agent had improved pepper rhizosphere bacterial α diversity and changed the community structure. Acidobacteria, Proteobacteria, Actinomycetes and Firmicutes were dominant phyla in all the pepper rhizosphere soil samples. The results showed that soil bacterial community were significantly positively correlated with pH ( R = 0.8537, P = 0.001) and total nitrogen ( R = 0.4347, P = 0.003). The nine significantly enriched OTU in R.palustris PSB06 treatment (PB) group belong to Nitrososphaera (OTU_109, OTU_14, OTU_18, OTU_8), Lysobacter (OTU_2115, OTU_13), Arenimonas (OTU_26), Luteimonas (OTU_49), and Ramlibacter (OTU_70) were significantly positively correlated with the total yield of pepper ( R > 0.5, P < 0.05). Overall, our results provide a theoretical basis for studying the microbial regulation of R.palustris PSB06 on rhizosphere soil.
Root exudation under maize/soybean intercropping system mediates the arbuscular mycorrhizal fungi diversity and improves the plant growth

Abstract:Introduction: Maize/soybean intercropping is a common cropping practice in Chinese agriculture, known to boost crop yield and enhance soil fertility. However, the role of below-ground interactions, particularly root exudates, in maintaining intercropping advantages in soybean/maize intercropping systems remains unclear. Methods: This study aimed to investigate the differences in root exudates between intercropping and monocropping systems through two pot experiments using metabolomics methods. Multiple omics analyses were conducted to explore correlations between differential metabolites and the community of Arbuscular Mycorrhizal Fungi (AMF), shedding light on the mechanisms underlying the dominance of intercropping from the perspective of root exudates-soil microorganism interactions. Results and discussion: The study revealed that intercropping significantly increased the types and contents of root exudates, lowered soil pH, increased the availability of nutrients like available nitrogen (AN) and available phosphorus (AP), and enhanced AMF colonization, resulting in improving the community composition of AMF. Besides, root exudates in intercropping systems differed significantly from those in monocropping, with 41 and 39 differential metabolites identified in the root exudates of soybean/maize, predominantly amino acids and organic acids. The total amount of amino acids in the root exudates of soybean intercropping was 3.61 times higher than in monocropping. Additionally, the addition of root exudates significantly improved the growth of soybean/maize and AMF colonization, with the mycorrhizal colonization rate in intercropping increased by 105.99% and 111.18% compared to monocropping, respectively. The identified metabolic pathways associated with root exudates were closely linked to plant growth, soil fertility improvement, and the formation of AMF. Correlation analysis revealed a significant relationship (P < 0.05) between certain metabolites such as tartaric acid, oxalic acid, malic acid, aspartic acid, alanine, and the AMF community. Notably, the photosynthetic carbon fixation pathway involving aspartic acid showed a strong association with the function of Glomus_f_Glomerace, the dominant genus of AMF. A combined analysis of metabolomics and high throughput sequencing revealed that the root exudates of soybean/maize intercropping have direct or indirect connections with AMF and soil nutrients. Conclusion: This suggests that the increased root exudates of the soybean/maize intercropping system mediate an improvement in AMF community composition, thereby influencing soil fertility and maintaining the advantage of intercropping.

Abstract:Continuous cropping obstacles have become a serious factor restricting sustainable development in modern agriculture, while companion planting is one of the most common and effective methods for solving this problem. Here, we monitored the effects of companion planting on soil fertility and the microbial community distribution pattern in pepper monoculture and companion plantings. Soil microbial communities were analyzed using high-throughput sequencing technology. Companion plants included garlic (T1), oat (T2), cabbage (T3), celery (T4), and white clover (T5). The results showed that compared with the monoculture system, companion planting significantly increased the activities of soil urease (except for T5) and sucrase, but decreased catalase activity. In addition, T2 significantly improved microbial diversity (Shannon index), while T1 resulted in a decrease of bacterial OTUs and an increase of fungal OTUs. Companion planting also significantly changed soil microbial community structures and compositions. Correlation analysis showed that soil enzyme activities were closely correlated with bacterial and fungal community structures. Moreover, the companion system weakened the complexity of microbial networks. These findings indicated that companion plants can provide nutrition to microbes and weaken the competition among them, which offers a theoretical basis and data for further research into methods for reducing continuous cropping obstacles in agriculture.

[Response Characteristics of Soil Fungal Community Structure to Long-term Continuous Cropping of Pepper]

Abstract:This study aimed to clarify the effect of long-term continuous cropping of pepper on soil fungal community structure, reveal the mechanism of continuous cropping obstacles, and provide a theoretical basis for the ecological safety and sustainable development of pepper industry. We took the pepper continuous cropping soil in the vegetable greenhouse planting base of Tongren City as the research object. The diversity and community structure of fungi in farmland soil were analyzed using Illumina MiSeq high-throughput sequencing, the responses of soil physio-chemical properties and fungal community characteristics to long-term continuous pepper cropping were discussed, and the relationships between the characteristics of fungal community structure and environmental factors were determined using CCA and correlation network analysis. The results showed that with the extension of pepper continuous cropping years, the soil pH value and organic matter (OM) content decreased, total phosphorus (TP) and available phosphorus (AP) contents increased, hydrolyzed nitrogen (AN) and available potassium (AK) contents decreased first and then increased, and total nitrogen (TN) and total potassium (TK) contents did not change significantly. Long-term continuous cropping decreased the Chao1 index and observed species index and decreased the Shannon index and Simpson index. The change in continuous cropping years had a significant effect on the relative abundance of soil fungal dominant flora. At the phylum level, the relative abundance of Mortierellomycota decreased with the extension of pepper continuous cropping years, the relative abundance of Ascomycota increased first and then decreased, and the relative abundance of Basidiomycota decreased first and then increased. At the genus level, with the increasing of pepper continuous cropping years, the relative abundance of Fusarium increased, and the relative abundance of Mortierella and Penicillium decreased. In addition, long-term continuous cropping simplified the soil fungal symbiosis network. CCA analysis indicated that pH, OM, TN, AN, AP, and AK were the driving factors of soil fungal community structure, and correlation network analysis showed that pH, OM, TN, TP, TK, AN, AP, and AK were the driving factors of soil fungal community structure, including Fusarium, Lophotrichus, Penicillium, Mortierella, Botryotrichum, Staphylotrichum, Plectosphaerella, and Acremonium. In conclusion, continuous cropping changed the soil physical and chemical properties, affected the diversity and community structure of the soil fungal community, changed the interaction between microorganisms, and destroyed the microecological balance of the soil, which might explain obstacles associated with continuous cropped pepper.
Multi-factor correlation analysis of the effect of root-promoting practices on tobacco rhizosphere microecology in growth stages

Abstract:Some agronomic practices not only promote the development of crop roots and increase overall plant performance but also affect colonisation by rhizosphere microorganisms. However, the composition and temporal dynamics of the tobacco rhizosphere microbiota under different root-promoting practices are poorly understood. Here, we characterised the tobacco rhizosphere microbiota at the knee-high, vigorous growing, and maturity stages under the application of potassium fulvic acid (PFA), γ-Polyglutamic acid (PGA), soymilk root irrigation (SRI), and conventional fertilization (CK) and its correlation with root characteristics and soil nutrients. The results showed that three root-promoting practices notably improved the dry and fresh root weights. Total nitrogen and phosphorus, available phosphorus and potassium, and organic matter contents in the rhizosphere markedly increased at the vigorous growing stage. The rhizosphere microbiota was changed through root-promoting practices. However, with tobacco growth, the change of rhizosphere microbiota showed a pattern of slow first and then fast and the microbiota of different treatments gradually approached. SRI reduced plant-pathogenic fungi but increased chemoheterotrophic and phototrophic bacteria, and arbuscular mycorrhizal fungi. PFA and PGA markedly increased arbuscular mycorrhizal and ectomycorrhizal fungi at the knee-high stage, which benefitted tobacco nutrient absorption. The correlation between rhizosphere microorganisms and environmental factors varied at different growth stages. Notably, the rhizosphere microbiota was more sensitive to environmental factors at the vigorous growing stage, and the interactions were more complex than in other stages. Furthermore, a variance partitioning analysis showed that the influence of root-soil interaction on the rhizosphere microbiota increased with tobacco growth. Overall, all three root-promoting practices could improve root characteristics, rhizosphere nutrient, and rhizosphere microbiota to varying degrees and increase the tobacco biomass, among which PGA had the most obvious effect and most suitable for tobacco cultivation. Our findings revealed the role of root-promoting practices in shaping the rhizosphere microbiota during plant growth and elucidated the assembly patterns and environmental drivers of crop rhizosphere microbiota driven by the application of root-promoting practices in agricultural production.

Abstract:The fungal endophyte Phomopsis liquidambari can improve nitrification rates and alter the abundance and composition of ammonia-oxidizers in the soil rhizosphere of rice. Aerenchyma is related to oxygen transport efficiency and contributes to the enhanced rhizospheric nitrification under flooding conditions. However, whether and how P. liquidambari affects aerenchyma formation is largely unknown. We therefore conducted pot and hydroponic experiments to investigate the changes of aerenchyma area, ethylene and indole-3-acetic acid (IAA) levels in rice with or without P. liquidambari infection. Our results showed that the larger aerenchyma area in rice roots with P. liquidambari inoculation was associated with markedly up-regulated expression of genes related to aerenchyma formation. Meanwhile, P. liquidambari inoculation substantially elevated root porosity (POR) and radial oxygen loss (ROL), leading to the enhancement of oxidation-reduction potential (ORP) under pot condition. Besides, P. liquidambari significantly increased IAA and ethylene levels in rice by stimulating the expression of genes involved in auxin and ethylene biosyntheses. Furthermore, auxin that partly acting upstream of ethylene signalling played an essential role in P. liquidambari-promoted aerenchyma formation. These results verified the direct contribution of P. liquidambari in promoting aerenchyma formation via the accumulation of IAA and ethylene in rice roots, which provides a constructive suggestion for improving hypoxia tolerance through plant-endophyte interactions.

Abstract:As essential approaches for conservation agricultural practices, straw residue retention and crop rotation have been widely used in the Mollisols of Northeast China. Soil organic carbon, root development and microbial community are important indicators representing soil, crop and microbiota, respectively, and these factors work together to influence soil fertility and crop productivity. Studying their changes and interactions under different conservation practices is crucial to provide a theoretical basis for developing rational agricultural practices. The experiment in this study was conducted using the conventional practice (continuous maize without straw retention, C) and three conservation practices, namely, continuous maize with straw mulching (CS), maize–peanut rotation (R), and maize–peanut rotation with straw mulching (RS). Straw mulching (CS) significantly increased soil total organic carbon (TOC), active organic carbon (AOC), and microbial biomass carbon (MBC), but did not promote maize yield. Maize–peanut rotation (R and RS) significantly increased dissolved organic carbon (DOC) in the rhizosphere by promoting root growth, and maize yield (increased by 10.2%). For the microbial community structure, PERMANOVA and PCoA indicated that the bacterial community differed significantly between rhizosphere soil and bulk soil, but the fungal community shifted more under different agricultural practices. The correlation analysis indicated that the rotation system promoted the association between the soil DOC and the microbial community (especially the bacterial community), and straw mulching enhanced the connection between the soil TOC and the fungal community. Some plant growth–promoting rhizobacteria (including Bacillus , Streptomyces , Rhizobium , and Pseudomonas ) were enriched in the rhizosphere soil and were increased in the rotation system (R and RS), which might be due to an increase in the soil rhizosphere DOC level. These beneficial microbes had significantly negative correlations with several fungal groups (such as Mycosphaerella , Penicillium , Paraphoma and Torula ) that were classified as plant pathotrophs by FUNGuild. These results indicated that ensuring plant root development and improving root–bacteria interactions are of great importance to guarantee crop yield when implementing conservation tillage practices.
