Abstract:Understanding how and when key novel adaptations evolved is a central goal of evolutionary biology. Within the immigrans-tripunctata radiation of Drosophila, many mushroom-feeding species are tolerant of host toxins, such as cyclopeptides, that are lethal to nearly all other eukaryotes. In this study, we used phylogenetic and functional approaches to investigate the evolution of cyclopeptide tolerance in the immigrans-tripunctata radiation of Drosophila. First, we inferred the evolutionary relationships among 48 species in this radiation using 978 single copy orthologs. Our results resolved previous incongruities within species groups across the phylogeny. Second, we expanded on previous studies of toxin tolerance by assaying 16 of these species for tolerance to α-amanitin and found that six of them could develop on diet with toxin. Finally, we asked how α-amanitin tolerance might have evolved across the immigrans-tripunctata radiation, and inferred that toxin tolerance was ancestral in mushroom-feeding Drosophila and subsequently lost multiple times. Our findings expand our understanding of toxin tolerance across the immigrans-tripunctata radiation and emphasize the uniqueness of toxin tolerance in this adaptive radiation and the complexity of biochemical adaptations.
Cis- and trans-acting variants contribute to survivorship in a naïve Drosophila melanogaster population exposed to ryanoid insecticides

Abstract:Insecticide resistance is a paradigm of microevolution, and insecticides are responsible for the strongest cases of recent selection in the genome of Drosophila melanogaster Here we use a naïve population and a novel insecticide class to examine the ab initio genetic architecture of a potential selective response. Genome-wide association studies (GWAS) of chlorantraniliprole susceptibility reveal variation in a gene of major effect, Stretchin Myosin light chain kinase (Strn-Mlck), which we validate with linkage mapping and transgenic manipulation of gene expression. We propose that allelic variation in Strn-Mlck alters sensitivity to the calcium depletion attributable to chlorantraniliprole's mode of action. GWAS also reveal a network of genes involved in neuromuscular biology. In contrast, phenotype to transcriptome associations identify differences in constitutive levels of multiple transcripts regulated by cnc, the homolog of mammalian Nrf2. This suggests that genetic variation acts in trans to regulate multiple metabolic enzymes in this pathway. The most outstanding association is with the transcription level of Cyp12d1 which is also affected in cis by copy number variation. Transgenic overexpression of Cyp12d1 reduces susceptibility to both chlorantraniliprole and the closely related insecticide cyantraniliprole. This systems genetics study reveals multiple allelic variants segregating at intermediate frequency in a population that is completely naïve to this new insecticide chemistry and it foreshadows a selective response among natural populations to these chemicals.
A potential role for the gut microbiota in the specialisation of Drosophila sechellia to its toxic host noni (Morinda citrifolia)

Abstract:Adaptation to a novel food source can have significant evolutionary advantages. The fruit fly, Drosophila sechellia, is a specialist of the toxic plant noni (Morinda citrifolia). Little is known as to how D. sechellia has become resistant to the toxins in the fruit - comprised predominantly of octanoic acid - but to date, the behavioural preferences for the fruit and genetic architecture underlying them, have been well studied. Here, we examine whether the gut microbiota could have played a role in adaptation to the fruit. In the first series of experiments, we examine the gut microbiota of wild-type, laboratory reared flies and characterise the gut microbiota when reared on the natural host plant, versus a standard Drosophila diet. We show a rapid transition in the core bacterial diversity and abundance within this species and discover sole precedence of Lactobacillus plantarum when reared on M. citrifolia. We also discover that flies reared on a laboratory diet are more likely to carry bacterial pathogens such as Bacillus cereus, although their function in Drosophila is unknown. Flies reared on a laboratory diet have a significantly reduced weight but with no impact on the risk of death before adulthood, when compared to the wild noni diet. In the second series of experiments, we examine the potential role of the gut microbiota in adaptation to octanoic acid resistance in this species and its sister species, Drosophila melanogaster, to which the fruit is usually fatal. We use a combination of methods to analyse resistance to octanoic acid by conducting life history analysis, behavioural assays and bacterial analysis in both D. sechellia and D. melanogaster. We find that by creating experimental evolution lines of D. melanogaster supplemented with gut microbiota from D. sechellia, we can decrease D. melanogaster aversion to octanoic acid, with the flies even preferring to feed on food supplemented with the acid. We suggest this represents the first step in the evolutionary and ecological specialisation of D. sechellia to its toxic host plant, and that the gut microbiota, Lactobacillus plantarum in particular, may have played a key role in host specialisation.
The Power of Drosophila Genetics in Studying Insect Toxicology and Chemical Ecology

Abstract:Insect toxicology and chemical ecology are inherently interconnected disciplines, both dedicated to unraveling the intricate relationships between insects and the diverse array of chemical compounds that pervade their surroundings. Drosophila melanogaster , owing to its genetic and physiological similarities to other insects, serves as a robust model system in the study of insect toxicology. Moreover, state-of-the-art techniques in Drosophila neurobiology have extensively probed the chemosensory system of insects, providing significant insights into their adaptation to chemical environments. In this review, we emphasize the advancements achieved through the application of Drosophila genetics in investigations spanning both of these fields, significantly enhancing our understanding of the mode of action and resistance mechanisms of insecticides, as well as unraveling the molecular and cellular mechanisms underlying insect chemosensation and associated behaviors. The profound insights derived through this tiny fly not only enrich our understanding of the broader world of insects but also hold the potential to develop more effective and sustainable strategies for pest management.
Mutational Analysis of Aspergillus fumigatus Volatile Oxylipins in a Drosophila Eclosion Assay

Abstract:Aspergillus fumigatus is a ubiquitous opportunistic pathogen. We have previously reported that volatile organic compounds (VOCs) produced by A. fumigatus cause delays in metamorphosis, morphological abnormalities, and death in a Drosophila melanogaster eclosion model. Here, we developed A. fumigatus deletion mutants with blocked oxylipin biosynthesis pathways (∆ppoABC) and then exposed the third instar larvae of D. melanogaster to a shared atmosphere with either A. fumigatus wild-type or oxylipin mutant cultures for 15 days. Fly larvae exposed to VOCs from wild-type A. fumigatus strains exhibited delays in metamorphosis and toxicity, while larvae exposed to VOCs from the ∆ppoABC mutant displayed fewer morphogenic delays and higher eclosion rates than the controls. In general, when fungi were pre-grown at 37 °C, the effects of the VOCs they produced were more pronounced than when they were pre-grown at 25 °C. GC–MS analysis revealed that the wild-type A. fumigatus Af293 produced more abundant VOCs at higher concentrations than the oxylipin-deficient strain Af293∆ppoABC did. The major VOCs detected from wild-type Af293 and its triple mutant included isopentyl alcohol, isobutyl alcohol, 2-methylbutanal, acetoin, and 1-octen-3-ol. Unexpectedly, compared to wild-type flies, the eclosion tests yielded far fewer differences in metamorphosis or viability when flies with immune-deficient genotypes were exposed to VOCs from either wild-type or ∆ppoABC oxylipin mutants. In particular, the toxigenic effects of Aspergillus VOCs were not observed in mutant flies deficient in the Toll (spz6) pathway. These data indicate that the innate immune system of Drosophila mediates the toxicity of fungal volatiles, especially via the Toll pathway.

Abstract:Abstract Periodic food shortage is a common ecological stressor for animals, likely to drive physiological and metabolic adaptations to alleviate its consequences, particularly for juveniles that have no option but to continue to grow and develop despite undernutrition. Here we study changes in metabolism associated with adaptation to nutrient shortage, evolved by replicate Drosophila melanogaster populations maintained on a nutrient-poor larval diet for over 240 generations. In a factorial metabolomics experiment we showed that both phenotypic plasticity and genetically-based adaptation to the poor diet involved wide-ranging changes in metabolite abundance; however, the plastic response did not predict the evolutionary change. Compared to nonadapted larvae exposed to the poor diet for the first time, the adapted larvae showed lower levels of multiple free amino acids in their tissues—and yet they grew faster. By quantifying accumulation of the nitrogen stable isotope 15N we show that adaptation to the poor diet led to an increased use of amino acids for energy generation. This apparent “waste” of scarce amino acids likely results from the trade-off between acquisition of dietary amino acids and carbohydrates observed in these populations. The three branched-chain amino acids (leucine, isoleucine, and valine) showed a unique pattern of depletion in adapted larvae raised on the poor diet. A diet supplementation experiment demonstrated that these amino acids are limiting for growth on the poor diet, suggesting that their low levels resulted from their expeditious use for protein synthesis. These results demonstrate that selection driven by nutrient shortage not only promotes improved acquisition of limiting nutrients, but also has wide-ranging effects on how the nutrients are used. They also show that the abundance of free amino acids in the tissues does not, in general, reflect the nutritional condition and growth potential of an animal.

Abstract:Insecticides have become the primary selective force in many insect species; however, whether beneficial insects developed resistance remains unknown. We analyzed the sequences of hymenopteran GABAA receptor subunit gene Rdl ( resistance to dieldrin ), which encodes the target of cyclodiene and phenylpyrazole insecticides. The resistance-conferring A2 S mutations were found in seven parasitoid wasps and similar amino acid replacements at homologous sites have been identified in four of their resistant hosts. Abstract:Ionotropic γ-aminobutyric acid (GABA) receptors in insects, specifically those composed of the RDL (resistant to dieldrin) subunit, serve as important targets for commonly used synthetic insecticides. These insecticides belong to various chemical classes, such as phenylpyrazoles, cyclodienes, meta-diamides, and isoxazolines, with the latter two potentially binding to the transmembrane inter-subunit pocket. However, the specific amino acid residues that contribute to the high sensitivity of insect RDL receptors to these novel insecticides remain elusive. In this study, we investigated the susceptibility of seven distinct Drosophila melanogaster Rdl point mutants against four meta-diamide and isoxazoline insecticides: isocycloseram, fluxametamide, fluralaner, and broflanilide. Our findings indicate that, despite exhibiting increased sensitivity to fluralaner in vitro, the RdlI276C mutant showed resistance to isocycloseram and fluxametamide. Similarly, the double-points mutant RdlI276F+G279S also showed decreased sensitivity to the tested isoxazolines. On the other hand, the RdlG335M mutant displayed high levels of resistance to all tested insecticides. Molecular modeling and docking simulations further supported these findings, highlighting similar binding poses for these insecticides. In summary, our research provides robust in vivo evidence supporting the idea that the inter-subunit amino acids within transmembrane M1 and M3 domains form the binding site crucial for meta-diamide and isoxazoline insecticide interactions. This study highlights the complex interplay between mutations and insecticide susceptibility, paving the way for more targeted pest control strategies.

Abstract:Pathogens exert strong selection on hosts, that evolve and deploy different defensive strategies, namely minimizing pathogen exposure (avoidance), directly promoting pathogen elimination (resistance), and/or managing the deleterious effects of illness (disease tolerance). However, how the response to pathogens partitions across these processes has never been directly assessed in a single system, let alone in the context of known adaptive trajectories under controlled selection regimes. Here, an experimental evolution system composed of D. melanogaster and its natural pathogen P. entomophila is used to independently assess the role of behavioural traits, and of resistance and disease tolerance mechanisms on host evolution. We compare one replicate of a population adapted to oral infection with P. entomophila (BactOral) to a replicate of its control population to find no evidence for behavioural change but measurable differences in both resistance and disease tolerance. In BactOral, we identify a relative decrease in bacterial loads correlated with an increase in gut production of specific AMPs, but no differences in bacterial intake, in gut cell renewal rate, or in the rate of bacterial defecation, pointing to a strengthening in resistance. Additionally, we posit that disease tolerance also contributes to the adaptive response of the BactOral population through a tighter control of its immune response and of the deleterious effects of exposure. This study reveals a genetically complex and mechanistically multi-layered response, possibly reflecting the structure of adaptation to infection in natural populations.

Abstract:Disease resistance (defined as the host capacity to limit systemic infection intensity) and disease tolerance (defined as the host capacity to limit infection-induced damage) are two complementary defense strategies that help the hosts maximize their survival and fitness when infected with pathogens and parasites. In addition to the underlying physiological mechanisms, existing theory postulates that these two strategies differ in terms of the conditions under which each strategy evolves in the host populations, their evolutionary dynamics, and the ecological and epidemiological consequences of their evolution. Here we explored if one or both of these strategies evolve when host populations are subjected to selection for increased post-infection survival. We experimentally evolved Drosophila melanogaster populations, selecting for the flies that survived an infection with the entomopathogen Enterococcus faecalis. We found that the host populations evolved increased disease resistance in response to selection for increased survival. This was despite the physiological costs associated with increased resistance, expression of which varied with the phase of infection. We did not find evidence of any change in disease tolerance in the evolved host populations.

Abstract:Defenses conferred by microbial symbionts play a vital role in the health and fitness of their animal hosts. An important outstanding question in the study of defensive symbiosis is what determines long term stability and effectiveness against diverse natural enemies. In this study, we combine genome and transcriptome sequencing, symbiont transfection and parasite protection experiments, and toxin activity assays to examine the evolution of the defensive symbiosis between Drosophila flies and their vertically transmitted Spiroplasma bacterial symbionts, focusing in particular on ribosome-inactivating proteins (RIPs), symbiont-encoded toxins that have been implicated in protection against both parasitic wasps and nematodes. Although many strains of Spiroplasma, including the male-killing symbiont (sMel) of Drosophila melanogaster, protect against parasitic wasps, only the strain (sNeo) that infects the mycophagous fly Drosophila neotestacea appears to protect against parasitic nematodes. We find that RIP repertoire is a major differentiating factor between strains that do and do not offer nematode protection, and that sMel RIPs do not show activity against nematode ribosomes in vivo. We also discovered a strain of Spiroplasma infecting a mycophagous phorid fly, Megaselia nigra. Although both the host and its Spiroplasma are distantly related to D. neotestacea and its symbiont, genome sequencing revealed that the M. nigra symbiont encodes abundant and diverse RIPs, including plasmid-encoded toxins that are closely related to the RIPs in sNeo. Our results suggest that distantly related Spiroplasma RIP toxins may perform specialized functions with regard to parasite specificity and suggest an important role for horizontal gene transfer in the emergence of novel defensive phenotypes.

Convergent resistance to GABA receptor neurotoxins through plant-insect coevolution

Abstract:The molecular mechanisms of coevolution between plants and insects remain elusive. GABA receptors are targets of many neurotoxic terpenoids, which represent the most diverse array of natural products known. Over deep evolutionary time, as plant terpene synthases diversified in plants, so did plant terpenoid defence repertoires. Here we show that herbivorous insects and their predators evolved convergent amino acid changing substitutions in duplicated copies of the Resistance to dieldrin (Rdl) gene that encodes the GABA receptor, and that the evolution of duplicated Rdl and terpenoid-resistant GABA receptors is associated with the diversification of moths and butterflies. These same substitutions also evolved in pests exposed to synthetic insecticides that target the GABA receptor. We used in vivo genome editing in Drosophila melanogaster to evaluate the fitness effects of each putative resistance mutation and found that pleiotropy both facilitates and constrains the evolution of GABA receptor resistance. The same genetic changes that confer resistance to terpenoids across 300 Myr of insect evolution have re-evolved in response to synthetic analogues over one human lifespan.
Additive and mostly adaptive plastic responses of gene expression to multiple stress in Tribolium castaneum

Abstract:Gene expression is known to be highly responsive to the environment and important for adjustment of metabolism but there is also growing evidence that differences in gene regulation contribute to species divergence and differences among locally adapted populations. However, most studies so far investigated populations when divergence had already occurred. Selection acting on expression levels at the onset of adaptation to an environmental change has not been characterized. Understanding the mechanisms is further complicated by the fact that environmental change is often multivariate, meaning that organisms are exposed to multiple stressors simultaneously with potentially interactive effects. Here we use a novel approach by combining fitness and whole-transcriptome data in a large-scale experiment to investigate responses to drought, heat and their combination in Tribolium castaneum. We found that fitness was reduced by both stressors and their combined effect was almost additive. Expression data showed that stressor responses were acting independently and did not interfere physiologically. Since we measured expression and fitness within the same individuals, we were able to estimate selection on gene expression levels. We found that variation in fitness can be attributed to gene expression variation and that selection pressures were environment dependent and opposite between control and stress conditions. We could further show that plastic responses of expression were largely adaptive, i.e. in the direction that should increase fitness.During environmental change organisms are exposed to several stressors simultaneously to which they can show plastic and evolutionary responses. Gene regulation can play a crucial role in both processes. Measuring gene expression gives us detailed insights into underlying molecular pathways, relative importance of different stressors and how they may interfere with each other in combination. However, it is challenging to understand how plastic responses are ultimately linked to fitness and adaptation. We combined measurements of a fitness component (offspring number) with gene expression data to study responses of the model organism T. castaneum (red flour beetle) to single and joint effects of drought and heat. Higher temperatures resulted in a stronger reduction of offspring number and a larger number of differently expressed genes compared to reduced humidity. We could show that stressors affected different physiological processes and consequently responses did not show much modification in combination. Combining fitness and transcriptomic data of the same individuals enabled us to estimate selection on gene expression levels. We found that selection under control conditions (to which the beetles are adapted) was in general opposite to selection under the new, stressful conditions and that large parts of the response to heat were adaptive.

Natural and Artificial Selection for Parasitoid Resistance in Drosophila melanogaster Leave Different Genetic Signatures

Abstract:Adaptation of complex traits depends on standing genetic variation at multiple loci. The allelic variants that have positive fitness effects, however, can differ depending on the genetic background and the selective pressure. Previously, we interrogated the Drosophila melanogaster genome at the population level for polymorphic positions and identified 215 single nucleotide polymorphisms (SNPs) that had significantly changed in frequency after experimental evolution for increased parasitoid resistance. In the current study, we follow up on 11 of these SNPs as putative targets of the experimental selection process (Jalvingh et al., 2014). We study the patterns of genetic variation for these SNPs in several European field populations. Furthermore, we associate the genetic variation of these SNPs to variation in resistance against the parasitoid Asobara tabida, by determining the individual phenotype and SNP genotype for 144 individuals from four Selection lines and four non-selected Control lines and for 400 individuals from 12 Field lines that differ in parasitoid resistance. For the Selection lines we additionally monitored the changes in allele frequencies throughout the five generations of experimental selection. For three genes, mbl (Zn-finger protein), mthl4 (G-protein coupled receptor) and CG17287 (protein-cysteine S-palmitoyltransferase) individual SNP genotypes were significantly associated with resistance level in the Selection and Control lines. Additionally, the minor allele in mbl and mthl4 were consistently and gradually favored throughout the five generations of experimental evolution. However, none of these alleles did appear to be associated to high resistance in the Field lines. We suggest that, within field populations, selection for parasitoid resistance is a gradual process that involves co-adapted gene complexes. Fast artificial selection, however, enforces the sudden cumulating of particular alleles that confer high resistance (genetic sweep). We discuss our findings in the context of local adaptation.

Fungal metabolic plasticity and sexual development mediate induced resistance to arthropod fungivory

Abstract:Prey organisms do not tolerate predator attack passively but react with a multitude of inducible defensive strategies. Although inducible defence strategies are well known in plants attacked by herbivorous insects, induced resistance of fungi against fungivorous animals is largely unknown. Resistance to fungivory is thought to be mediated by chemical properties of fungal tissue, i.e. by production of toxic secondary metabolites. However, whether fungi change their secondary metabolite composition to increase resistance against arthropod fungivory is unknown. We demonstrate that grazing by a soil arthropod, Folsomia candida , on the filamentous fungus Aspergillus nidulans induces a phenotype that repels future fungivores and retards fungivore growth. Arthropod-exposed colonies produced significantly higher amounts of toxic secondary metabolites and invested more in sexual reproduction relative to unchallenged fungi. Compared with vegetative tissue and asexual conidiospores, sexual fruiting bodies turned out to be highly resistant against fungivory in facultative sexual A. nidulans . This indicates that fungivore grazing triggers co-regulated allocation of resources to sexual reproduction and chemical defence in A. nidulans . Plastic investment in facultative sex and chemical defence may have evolved as a fungal strategy to escape from predation.

However, it's unclear whether adaptation to combined stress can occur without compromising viability and thermal plasticity. We tested thermotolerance and thermal plasticity in larval Drosophila melanogaster selected under different temperatures (18°C, 25°C, 28°C) and diets (standard, diluted, low protein:carbohydrate). Basal cold tolerance was affected by both low protein concentration and temperature; larvae evolved higher basal cold tolerance on diluted and low protein:carbohydrate diets at 18°C and 28°C. Hardening increased cold tolerance for most lines, except those selected at 18°C and 28°C on low protein:carbohydrate diets and 25°C on standard diets. Basal heat tolerance was affected by selection temperature, with higher tolerance in 25°C selection lines. An interaction between selection temperature, selection diet and hardening treatment affected plasticity in heat tolerance, with most selection lines showing reduced heat tolerance upon hardening, except for lines selected at 25°C on low protein diets and at 28°C on standard diets. Our results indicate that adaptation to combined stress allows basal cold tolerance and plasticity in cold tolerance to evolve independently, but not heat tolerance, highlighting the increased vulnerability of ectotherms to long-term climate change.

Abstract:Traditional chemical pesticides pose potential threats to human health, the environment, and food safety, and there is an urgent need to develop botanical pesticides that are easily degradable, renewable, and environmentally compatible. This research serves to detect the lethal impacts of Amanita pantherina(DC.:Fr) Schrmm.(Agaricales, Amanitaceae, Amanita), Amanita virgineoides Bas (Agaricales, Amanitaceae, Amanita), Coprinus comatus (O.F.Müll.) Pers. (Agaricales, Psathyrellaceae, Coprinus), Pycnoporus cinnabarinus(Jacq.:Fr) Karst (Polyporales, Polyporaceae, Polyporus) and Phallus rubicundus (Bosc) Fr. (Phallales, Phallaceae, Phallus) on Drosophila melanogaster(Diptera, Drosophilidae, Drosophila), including their effects on lifespan, fecundity, offspring growth and developmental characteristics, antioxidant enzyme activity, peroxide content, and the gene transcription associated with signaling pathways and lifespan of D. melanogaster. The results demonstrated that they all produced lethal effects on D. melanogaster. Female flies were more sensitive to the addition of macrofungi to their diet and have a shorter survival time than male flies. The toxic activity of A. pantherina-supplemented diet was the strongest, so that the D. melanogaster in this group had no offspring. The macrofungal-supplemented diets were able to significantly reduce the activity of antioxidant enzymes, accumulate peroxidation products, up-regulatd the transcription of genes related to signaling pathways, inhibit the expression of longevity genes, reduce the lifespan and fertility of D. melanogaster. Consequently, we hypothetically suggest that medicinal C. comatus, P. cinnabarinus and P. rubicundus hold the potential to be developed into an environmentally friendly biopesticide for fly killing.

Abstract:Abstract Insecticide resistance is considered a classic model of microevolution, where a strong selective agent is applied to a large natural population, resulting in a change in frequency of alleles that confer resistance. While many insecticide resistance variants have been characterized at the gene level, they are typically single genes of large effect identified in highly resistant pest species. In contrast, multiple variants have been implicated in DDT resistance in Drosophila melanogaster; however, only the Cyp6g1 locus has previously been shown to be relevant to field populations. Here we use genome-wide association studies (GWAS) to identify DDT-associated polygenes and use selective sweep analyses to assess their adaptive significance. We identify and verify two candidate DDT resistance loci. A largely uncharacterized gene, CG10737, has a function in muscles that ameliorates the effects of DDT, while a putative detoxifying P450, Cyp6w1, shows compelling evidence of positive selection.

Abstract:Resistance to medical triazoles in Aspergillus fumigatus is an emerging problem for patients at risk of aspergillus diseases. There are currently two presumed routes for medical triazole-resistance selection: (i) through selection pressure of medical triazoles when treating patients and (ii) through selection pressure from non-medical sterol-biosynthesis-inhibiting (SI) triazole fungicides which are used in the environment. Previous studies have suggested that SI fungicides can induce cross-resistance to medical triazoles. Therefore, to assess the potential of selection of resistance to medical triazoles in the environment, we assessed cross-resistance to three medical triazoles in lineages of A. fumigatus from previous work where we applied an experimental evolution approach with one of five different SI fungicides to select for resistance. In our evolved lines we found widespread cross-resistance indicating that resistance to medical triazoles rapidly arises through selection pressure of SI fungicides. All evolved lineages showed similar evolutionary dynamics to SI fungicides and medical triazoles, which suggests that the mutations inducing resistance to both SI fungicides and medical triazoles are likely to be the same. Whole-genome sequencing revealed that a variety of mutations were putatively involved in the resistance mechanism, some of which are in known target genes.

Abstract:Actinobacteria produce antibacterial and antifungal specialized metabolites. Many insects harbour actinobacteria on their bodies or in their nests and use these metabolites for protection. However, some actinobacteria produce metabolites that are toxic to insects and the evolutionary relevance of this toxicity is unknown. Here we explore chemical interactions between streptomycetes and the fruit fly Drosophila melanogaster. We find that many streptomycetes produce specialized metabolites that have potent larvicidal effects against the fly; larvae that ingest spores of these species die. The mechanism of toxicity is specific to the bacterium's chemical arsenal: cosmomycin D producing bacteria induce a cell death-like response in the larval digestive tract; avermectin producing bacteria induce paralysis. Furthermore, low concentrations of volatile terpenes like 2-methylisoborneol that are produced by streptomycetes attract fruit flies such that they preferentially deposit their eggs on contaminated food sources. The resulting larvae are killed during growth and development. The phenomenon of volatile-mediated attraction and specialized metabolite toxicity suggests that some streptomycetes pose an evolutionary risk to insects in nature.
