Abstract:This study focuses on creating new forms of biomimetic nanofiber composites by combining copolymerizing and electrospinning approaches in the field of nanomedicine. The process involved utilizing the melt polymerization of proline (Pr) and hydroxyl proline (Hyp) to synthesize polymers based on Pr (PPE) and Hyp (PHPE). These polymers were then used in a grafting copolymerization process with chitosan (CS) to produce PHPC (1560 ± 81.08 KDa). A novel electrospun nanofiber scaffold was then produced using PHPC and/or CS, hyaluronic acid, polyvinyl alcohol, and naringenin (NR) as a loading drug. Finally, Mouse Dermal Fibroblast (MDF) cells were introduced to the wound dressing and assessed their therapeutic potential for wound healing in rats. The scaffolds were characterized by FTIR, NMR, DSC, and SEM analysis, which confirmed the amino acid grafting, loading drug, and porous and nanofibrous structures (>225 nm). The results showed that the PHPC-based scaffolds were more effective for swelling/absorption of wound secretions, had more elasticity/elongation, faster drug release, more MDF-cytocompatibility, and antibacterial activity against multidrug-resistant S. aureus compared to CS-based scaffolds. The in vivo studies showed that NR in combination with MDF can accelerate cell migration/proliferation, and remodeling phases of wound healing in both PHPC/CS-based scaffolds. Moreover, PHPC-based scaffolds promote collagen content, and better wound contraction, epithelialization, and neovascularization than CS-based, showing potential as wound-dressing.

Abstract:A highly porous nanofibrous network that can functionalize antibacterial and therapeutic agents can be considered a suitable option for skin wound healing. In this study, α-tricalcium phosphate (α-TCP)/nitrogen-doped carbon quantum dots (N-CQDs) nanocomposite was synthesized and then applied to the fabrication of novel chitosan (CS)/silk fibroin (SF)/N-CQDs/α-TCP wound dressing via electrospinning system. The prepared nanomaterials were well characterized using X-ray diffraction, Fourier-transform infrared, scanning and transmission electron microscopes analyses, and antibacterial assay. Furthermore, nanofibers were evaluated regarding their physical properties, such as tensile behavior, water uptake capacity, and water contact angle. The results reveal that CS/SF/N-CQDs/α-TCP showed lower MIC values against E. coli and S. aureus (1.45 ± 0.26 mg/mL and 1.59 ± 0.12 mg/mL) compared to other synthesized materials. Also, in-vitro investigations were performed, and the MTT assay on the HFF cell line revealed that CS/SF/N-CQDs/α-TCP nanofiber could possess good biocompatibility. Interestingly, the scratch test proved that faster cell migration and proliferation occurred in the presence of CS/SF/N-CQDs/α-TCP 73.23 ± 2.71 %). Finally, we examined the wound healing ability of CS/SF/N-CQDs/α-TCP nanofiber using an animal model. The results confirmed that produced nanofiber could efficiently promote wound closure by 96.73 ± 1.25 % in 12 days. Histopathological analyses verified accelerated re-epithelization and well-structured epidermis in CS/SF/N-CQDs/α-TCP nanofiber-treated group. Based on our findings, the CS/SF/N-CQDs/α-TCP nanofiber with excellent antimicrobial properties is highly suitable for wound healing and skin tissue regeneration applications.
Investigation of a novel bilayered PCL/PVA electrospun nanofiber incorporated Chitosan-LL37 and Chitosan-VEGF nanoparticles as an advanced antibacterial cell growth-promoting wound dressing

Abstract:Chronic wounds have become a growing concern as they can have a profound impact on individuals, potentially resulting in mortality. It is crucial to prevent and manage bacterial infections, particularly drug-resistant ones. Antimicrobial peptides, such as LL-37, can firmly eliminate pathogens. Additionally, the process of angiogenesis, facilitated by growth factors like VEGF, is essential for tissue repair and wound healing. To enhance the stability and bioavailability of therapeutic agents, targeted delivery strategies utilizing Chitosan-based carriers have been employed. Electrospun biopolymers in advanced wound dressings have revolutionized wound care by providing a more effective and efficient solution for promoting tissue regeneration and speeding up the healing process. The present investigation utilized Chitosan nanoparticles to encapsulate the recombinant LL37 peptide and VEGF. An in-depth investigation was carried out to analyze the biophysical and morphological traits of the LL37-CSNPs and VEGF-CSNPs. The first support layer consisted of PCL electrospun nanofiber, followed by the electrospinning of PVA/CsLL37, PVA/CsVEGF, and PVA/CsLL37/CsVEGF onto the PCL layer. An in vitro examination assessed the fabricated nanofibers' morphological, mechanical, and biological characteristics. The antimicrobial effects were tested on methicillin-resistant Staphylococcus aureus (MRSA). The in vivo experiments assessed the antibacterial and wound-healing capabilities of the nanofibers. The findings validated the continuous release of LL37 and VEGF. The composite material PCL/PVA/CsLL37/CsVEGF demonstrated potent bactericidal and antioxidant characteristics. The cytotoxic assay demonstrated the biocompatibility of the fabricated nano mats and their potential to accelerate fibroblast cell proliferation. The efficacy of PVA/CsLL37/CsVEGF in promoting wound healing was confirmed through an in vivo wound healing assay. Furthermore, the histological analysis provided evidence of faster epidermal formation and improved antibacterial activity in wounds covered with PVA/CsLL37/CsVEGF. Adding LL37 and VEGF to the composite material improves the immune response and promotes blood vessel formation, accelerating wound healing and decreasing inflammation.

Abstract:Recently, the design of polymer nanofibers using the electrospinning process has attracted much interest. Particularly the use of natural polymers has promoted many advantages in their biomedical applications. However, the combination of multiple natural polymers remains a great challenge in terms of electrospun production and applied performance. From this perspective, the current investigation highlights the study of the preparation of electrospun nanomaterial scaffolds based on combined natural polymers for improved wound healing performance. First, we have synthesized a crosslinked polymer by reacting microcrystalline cellulose (MC) and chitosan (CS) biopolymer via the intermediate of citric acid as a crosslinking agent. Then a natural propolis biomolecule was incorporated into the polymer network. Different MC/CS blend ratios of 90/10 and 70/30 were then used and various machine parameters were optimized to obtain nanofiber scaffolds with excellent strength and structures. SEM, IR, physicochemical, mechanical, and morpho-logical characterization were then performed. SEM evaluation revealed homogeneous and bead-free nanofibrous structures, with well-defined morphology and a random deposition that could accurately mimic the extracellular matrix of native skin. The calculated average nanofiber diameters for the MC/CS blend ratios at 90/10 and 70/30 were 431.4 and 441.2 nm, respectively. The results showed that when the chitosan amount increased, larger nanofibers with narrow diameter distribution appeared. The prepared nanomaterials had a significant and close water vapor permeability of about 1735.12 and 1698.52 g per m per day for the two blend ratios of 90/10 and 70/30, respectively. The examination of swelling behavior revealed a noteworthy enhancement in hydrophilicity, a necessary attribute for improved healing efficacy. FT-IR analysis confirmed the success and the stability of the chemical crosslinking reaction between the two biopolymers before nanofiber conception. Excellent mechanical properties were acquired, based on the chitosan content. Both developed nanofiber scaffolds exhibited high tensile strength and Young's modulus values. The incorporation of 30% chitosan versus 10% results in an increase in tensile strength of 11% and 14% in Young's modulus. Therefore, we could adjust the different mechanical properties simply by varying the mixing rate of the electrospun polymers. Using epithelial HepG2 cells, viability and kinetic cell adhesion assays were assessed to obtain biological evaluation. No cytotoxicity was observed and good cytocompatibility was confirmed. Functionalized nanofiber biomaterials with different MC/CS ratios substantiated significant bactericidal effectiveness against Gram-positive and Gram-negative bacterial culture strains. The novel functional electrospun wound dressing scaffold demonstrated effective and promising biomedical performance, healing both acute and chronic wounds.

Abstract:In this study, a highly porous 3D‐printed wound healing core/shell scaffold was fabricated using poly‐lactic acid (PLA). The core of scaffold was composed of hyaluronic acid (HA), copper carbon dots (Cu‐CDs), Rosmarinic acid and chitosan hydrogel. Cu‐CDs were synthesized using ammonium hydrogen citrate under hydrothermal conditions. Formulation containing 1mg.mL‐1concentration of Cu‐CDs showed an excellent antibacterial activity against gram bacteria. At 0.25 mg.mL‐1 of Cu‐CDs concentration, scaffold had a good biocompatibility as confirmed by cytotoxicity assay on L929 fibroblast stem cells. In‐vivo wound healing experiments on groups of rats revealed that after 15 days of treatment, the optimal formulation of composite scaffold significantly improves the wound healing process compared to the PLA scaffold. This finding was confirmed by histological analysis and the relative expression of PDGF, TGF‐β and MMP‐1 genes. The biocompatible antibacterial CU‐CDS /PLA/HA/chitosan/ Rosmarinic acid nanocomposite is a promising wound healing scaffold which highly accelerates the process of skin regeneration.This article is protected by copyright. Design of 3D polycaprolactone/ε-polylysine-modified chitosan fibrous scaffolds with incorporation of bioactive factors for accelerating wound healing

Abstract:Electrospun nanofibrous scaffolds show great application potentials for wound healing owing to their effective simulation of extracellular matrix (ECM). Three-dimensional (3D) nanofibrous dressings exhibit relatively high specific surface areas, better mimicry of native ECM, adjustable hydrophilicity and breathability, good histocompatibility, enhanced wound healing, and reduced inflammation. In the present work, we designed the 3D polycaprolactone/ε-polylysine modified chitosan (PCL/PCS) nanofibrous scaffolds by an electrospinning and gas foaming process. Then, gelatin and heparin (Gel/Hep) were assembled onto the surface of PCL/PCS nanofibers by electrostatic adsorption, and vascular endothelial growth factors (VEGFs) were also synchronously incorporated into Gel/Hep layer to form a multifunctional 3D nanofibrous scaffold (PCL/PCS@Gel/Hep+VEGF) for accelerating wound healing. The as-fabricated 3D PCL/PCS@GEL/Hep+VEGF nanofibrous scaffold showed excellent antibacterial ability, hemocompatibility and biocompatibility in vitro and wound healing ability in vivo. Immunological analysis showed that the as-fabricated nanofibrous scaffold inhibited inflammation at the wound sites while promoting angiogenesis during the wound healing process. STATEMENT OF SIGNIFICANCE: The electrospun 3D fibrous scaffolds using polycaprolactone/ε-polylysine modified chitosan (PCL/PCS) have been fabricated as backbone for mimicking the extracellular matrix (ECM). Gelatin and heparin (Gel/Hep) were wrapped onto the surface of PCL/PCS fibers by electrostatic adsorption and vascular endothelial growth factors (VEGFs) were also synchronously incorporated into surface Gel/Hep layer to form multifunctional 3D fibrous scaffolds. The as-fabricated multifunctional 3D fibrous scaffolds with good antibacterial ability and biocompatibility have been used as dressings for accelerating wound healing by inhibiting inflammation at the wound sites while promoting angiogenesis during the wound healing process.
Curcumin‐Loaded Co‐Axial Electrospun Chitosan/Polyvinyl Alcohol/Calcium Chloride Nanofibrous Membranes for Wound Healing Enhancement

Abstract:To facilitate rapid wound healing, a nanofibrous membrane consisting of chitosan (CS), polyvinyl alcohol (PVA), calcium chloride (CC), and CCM was generated as a core‐shell structure (PVA/CCM@CS/PVA/CC) utilizing a coaxial electrospinning process. The nanofiber had smooth surfaces, a uniform shape, and an average diameter of 301.55 ± 76.77 nm. Nanofibrous membranes significantly enhanced wound healing in mice compared to control samples. Nanofibrous membranes were developed by combining chitosan (CS), polyvinyl alcohol (PVA), and calcium chloride (CC) to incorporate curcumin (CCM) through a co‐axial electro‐spinning method to improve CCM bioavailability. The optimal parameters were PVA/CS ratio of 7:3 v/v, PVA/CCM ratio of 8:2 v/v, CC amount of 0.5 g, fiber collection distance of 15 cm, voltage value of 18 kV, and injection flow of 0.1 mL/h (both core and shell layer). The prepared PVA/CCM@CS/PVA/CC nanofiber showed smooth surfaces and uniform bead‐free morphology with core‐shell structure. The average fiber diameter was 301.55 ± 76.77 nm with a narrow distribution. Using the Korsmeyer‐Peppas kinetic release model, the nanofiber membrane effectively released ∼85% CCM at pH 7.4 over 96 h via Fickian diffusion transport mechanism. In vitro antibacterial tests demonstrated that the membrane was efficient against both Gram‐positive (Staphylococcus aureus) and Gram‐negative (Pseudomonas aeruginosa) bacteria. When compared to free CCM, the membrane improved cell survival due to its nontoxic and cytobiocompatible features. In vivo tests found that the membrane significantly improved healing in incised‐wound rats, reducing inflammation, fibroblast proliferation, and collagen deposition. These findings indicate that the novel PVA/CCM@CS/PVA/CC nanofiber membrane, which has numerous biological functions, could be a promising candidate for wound dressing therapeutic applications.

Lung Inflammation Associated With Clinical Acute Necrotizing Pancreatitis in Dogs

Abstract:Although dogs with acute necrotizing pancreatitis (ANP) can develop respiratory complications, there are no data describing lung injury in clinical cases of ANP in dogs. Therefore, we conducted a study to characterize lung injury and determine if pulmonary intravascular macrophages (PIMs) are induced in dogs with ANP (n = 21) compared with control dogs (n = 6). Two pathologists independently graded histologic sections of pancreas from clinical cases to characterize the severity of ANP (total scores of 3–10) compared with controls showing histologically normal pancreas (total scores of 0). Based on histological grading, lungs from dogs with ANP showed inflammation (median score, 1.5; range, 0–3), but the scores did not differ statistically from the control lungs (median score, 0.5; range, 0–2). A grid intersects-counting method showed an increase in the numbers of MAC387-positive alveolar septal mononuclear phagocyte profiles in lungs of dogs with ANP (ratio median, 0.0243; range, 0.0093–0.0734, with 2 outliers at 0.1523 and 0.1978) compared with controls (ratio median, 0.0019; range, 0.0017–0.0031; P < .0001). Only dogs with ANP showed labeling for von Willebrand factor in alveolar septal capillary endothelial cells, septal inflammatory cells, and alveolar macrophages. Toll-like receptor 4 and interleukin 6 were variably expressed in alveolar macrophages and septal inflammatory cells in lungs from both ANP and control dogs. Inducible nitric oxide synthase was detected in alveolar macrophages of dogs with ANP only. These data show that dogs with ANP have lung inflammation, including the recruitment of PIMs and expression of inflammatory mediators.

Abstract:The nanocomposite membrane incorporating a bio‐active rosmarinic acid‐hydroxyapatite hybrid (HAP‐RA) at 1 wt.% demonstrated enhanced hydrophilicity, water uptake, biodegradability, and mechanical properties, and showed excellent antibacterial activity against Staphylococcus aureus and Escherichia coli. Various wound dressings have been developed so far for wound healing, but most of them are ineffective in properly reestablishing the skin's structure, which increases infection risks and dehydration. Electrospun membranes are particularly interesting for wound dressing applications because they mimic the extracellular matrix of healthy skin. In this study, a potential wound healing platform capable of inducing synergistic antibacterial and antioxidation activities was developed by incorporating bio‐active rosmarinic acid‐hydroxyapatite hybrid (HAP‐RA) with different contents (0.5, 1, and 1.5 wt.%) into the electrospun polyamide 6 (PA6) nanofibers. Then, polyethylene glycol (PEG) was introduced to the nanofibrous composite to improve the biocompatibility and biodegradability of the dressing. The results indicated that the hydrophilicity, water uptake, biodegradability, and mechanical properties of the obtained PA6/PEG/HAP‐RA nanofibrous composite enhanced at 1 wt.% of HAP‐RA. The nanofibrous composite had excellent antibacterial activity. The antioxidation potential of the samples was assessed in vitro. The MTT assay performed on the L929 cell line confirmed the positive effects of the nanofibrous scaffold on cell viability and proliferation. According to the results, the PA6/PEG/HAP‐RA nanofibrous composite showed the desirable physiochemical and biological properties besides antibacterial and antioxidative capabilities, making it a promising candidate for further studies in wound healing applications.

Abstract:Bioresponsive nanodrug delivery systems have excellent potential in tissue engineering applications. Poly-anionic and poly-cationic biopolymers have provided a superior platform for designing pH-sensitive drug delivery systems. In this regard, hyaluronic acid-chitosan-polyvinyl alcohol complex nanofibers with high quality and reproducibility were produced by optimizing the solution preparation process. In addition, the synthesized composite nanofiber, with 66.82 kN/mm toughness, 200% swelling ratio, and 60% porosity, exhibited excellent properties to meet the requirements of the ideal wound dressing. Green cross-linking with citric acid prevented the destruction of the nanofiber even after prolonged immersion in biological solutions. ζ potential studies demonstrated that the synthesized nanofiber has a negative surface charge (∼-30) at physiological pH. The pKa of the temporin-Ra peptide is about 10, and as a result the peptide molecules have a net positive charge in physiological conditions. Therefore, peptide molecules immobilized on the synthesized scaffold based on surface adsorption. In vivo evaluation has proven that the wound bed has an alkaline environment, facilitating peptide release from the nanofiber scaffold. Electrospun nanofibers can imitate the architecture of the extracellular matrix for accelerating wound healing. In vitro investigation showed better adhesion, proliferation, migration, and fibroblast cell growth on peptide-loaded nanofiber samples than other groups. In vivo studies on full-thickness wounds in the mouse model indicated that the designed nanofiber was gradually absorbed without causing dryness or infection. On day 6, the peptide-loaded nanofiber revealed 60% wound closure compared to the control group (17%). In addition, based on histological studies, the composite nanofiber demonstrated excellent tissue repair ability, hence these active nanofiber mats can be a good alternative to existing wound dressings. Gene expression studies show that the antimicrobial peptide promotes the inflammatory phase of wound healing in a shorter time frame by accelerating the tumor necrosis factor-α cytokine response.
Electrospun porous bilayer nano-fibrous fish collagen/PCL bio-composite scaffolds with covalently cross-linked chitooligosaccharides for full-thickness wound-healing applications

Abstract:The development of tissue-engineered biodegradable artificial tissue substitutes with extracellular matrix-mimicking properties that govern the interaction between the material and biological environment is of great interest in wound-healing applications. In the present study, novel bilayer nanofibrous scaffolds composed of fish collagen (FC) and poly(ε-caprolactone) (PCL) were fabricated using electrospinning, with the covalent attachment of chitooligosaccharides (COS) via carbodiimide chemistry. The architecture and fiber diameter of the non-cross-linked nanofibrous scaffolds remained consistent irrespective of the polymer ratio under different electrospinning conditions, but the fiber diameter changed after cross-linking in association with the FC content. Fourier-transform infrared spectroscopy analysis indicated that the blend of biomaterials was homogenous, with an increase in COS levels with increasing FC content in the nanofibrous scaffolds. Based on cytocompatibility analysis (i.e., the cellular response to the nanofibrous scaffolds and their interaction), the nanofibrous scaffolds with high FC content were functionally active in response to normal human dermal fibroblast‑neonatal (NHDF-neo) and HaCaT keratinocyte cells, leading to the generation of a very effective tissue-engineered implant for full-thickness wound-healing applications. In addition to these empirical results, an assessment of the hydrophilicity, swelling, and mechanical integrity of the proposed COS-containing FC-rich FC/PCL (FCP) nanofibrous scaffolds confirmed that they have significant potential for use as tissue-engineered skin implants for rapid skin regeneration.

These nanofibers possess unique three-dimensional (3D) interconnected porous structures that result in a higher surface-area-to-volume ratio and porosity. This study was carried out to prepare nanofibrous membranes by electrospinning a blend of PVA/chitosan polymeric solution functionalized with different ratios of copper oxide. Chitosan-stabilized CuO nanoparticles (CH-CuO NPs) were biosynthesized successfully utilizing chitosan as the capping and reducing agent. XRD analysis confirmed the monoclinic structure of CH-CuO NPs. In addition, the electrospun nanofibrous membranes were UV-crosslinked for a definite time. The membranes containing CH-CuO NPs were characterized by X-ray diffraction (XRD), scanning electron microscopy (SEM), differential scanning calorimetry (DSC), Fourier transform infrared (FTIR) spectroscopy, ultraviolet-visible (UV-vis) spectrophotometry, and dynamic light scattering (DLS). SEM results showed the nanosize of the fiber diameter in the range of 147-207 nm. The FTIR spectroscopy results indicated the successful incorporation of CH-CuO NPs into the PVA/chitosan nanofibrous membranes. DSC analysis proved the enhanced thermal stability of the nanofibrous membranes due to UV-crosslinking. Swelling and degradation tests were carried out to ensure membrane stability. Greater antimicrobial activity was observed in the nanoparticle-loaded membrane. An in vitro release study of Cu2+ ions from the membrane was carried out for 24 h. The cytotoxicity of CH-CuO NP-incorporated membranes was investigated to estimate the safe dose of nanoparticles. An in vivo test using the CH-CuO NP-loaded PVA/chitosan membrane was conducted on a mice model, in which wound healing occurred in approximately 12 days. These results confirmed that the biocompatible, nontoxic nanofibrous membranes are ideal for wound-dressing applications.
Capsaicin/silica-infused polygalacturonic acid/polyvinyl alcohol nano-matrix for enhanced wound healing in skin injuries

Abstract:Wound healing is a complex physiological process, demanding advanced strategies for efficient tissue regeneration. To address this, we developed a novel nanofibrous matrix composed of polygalacturonic acid (PGA), polyvinyl alcohol (PVA), capsaicin, and zinc-doped mesoporous silica (Zn/MCM-41). This copolymeric matrix offers enhanced mechanical stability, controlled drug release, and improved cellular adhesion and proliferation, leading to effective tissue regeneration. Infusing capsaicin/Zn-MCM-41 confers synergistic advantages, including accelerated wound closure, diminished inflammation, and enhanced tissue regeneration, culminating in superior wound healing outcomes. Comprehensive physicochemical characterization of the nanofiber was conducted, employing techniques such as EDS, EDX, XRD, FESEM, HRTEM, FTIR, BET, TGA, DSC and Zeta potential, confirming the successful synthesis of Zn/MCM-41 at the nanoscale, exhibiting uniform porosity, colloidal stability, and thermal resilience. In vivo quantitative assessment demonstrates a significant acceleration in wound healing facilitated by the composite nanofibers. Furthermore, the incorporation of capsaicin into Zn/MCM-41 augments the wound healing process, as corroborated by histological evaluations. In summary, our investigation introduces an advanced composite nanofiber formulation that promotes accelerated wound healing quantitatively through the synergistic amalgamation of PVA, PGA, capsaicin, and Zn/MCM-41. The demonstrated efficacy of the nanofibers underscores their potential in translational regenerative medicine and wound healing applications.

Abstract:Chronic skin wounds are characterised by a non-healing process that makes necessary the application of wound dressings on the damaged area to promote and facilitate the recovery of skin’s physiological integrity. The aim of the present work is to develop a bioactive dressing that, once applied on the injured tissue, would exert antibacterial activity and promote adhesion and proliferation of fibroblasts. Nanofibres consisting of poly(lactic acid) (PLA) and essential oils (EOs) were electrospun and coated with a medium molecular weight chitosan (CS). Black pepper essential oil (BP-EO) or limonene (L), well-known for their antibacterial properties, were added to the PLA/acetone solution before electrospinning; phase separation phenomena occurred due to the poor solubility of the EOs in the PLA solution and led to fibres having surface nano-pores. The porous electrospun fibres were coated with CS to produce hydrophilic membranes that were easy to handle, biocompatible, and suited to promote cellular proliferation. The fibrous scaffolds were tested in terms of mechanical resistance, wettability, antibacterial activity, in-vitro cytotoxicity, and ability to promote fibroblasts’ adhesion and proliferation. The results obtained proved that the CS coating improved the hydrophilicity of the fibrous mats, enhanced EO’s antibacterial potential, and promoted cell adhesion and proliferation.

Abstract:Electrospinning of a heterogeneous solution is difficult to continue because the required process parameters are different for multiple phases. In this study, nanofibrous mats were successfully prepared from a heterogeneous blend of solid cellulose nanocrystals (CNC) and hydroxyapatite nanoparticles (HAp) in a solution mixture of chitosan and gelatin using an electrospinning technique. HAp and CNC were used as filler materials in the nanofibrous mats. Gelatin and chitosan polymer chains in the mats were crosslinked using glutaraldehyde. The fiber diameter was noticed to decrease from around 86 to 43 nm with the increase of electrical conductivity of the spinning solution from 890 to 1166 μS cm-1 and after crosslinking a significant variation in fibers' diameter was noticed. The elemental analysis data showed that around 85% of the HAp used in the spinning solution was passed through the nozzle and the rest of the portion remained settled in the spinning syringe. In the XRD study, the crystallinity of chitosan, HAp and CNC was not observed in the non-crosslinked and crosslinked mats. The TGA analysis showed that the crosslinked mat has no weight retention at 500 °C which is due to its complete amorphous nature. The mats showed single-phase transition temperatures in DSC analysis which proves that no segregation of materials was present in the electrospun fibers. FTIR analysis of the mats showed a new peak at 1205 cm-1 which suggests the Michael addition type reactions to be happened between chitosan and gelatin. Cytotoxicity analysis of the mats on the vero-cell line showed around 95% of cell viability. The prepared mats were applied as wound dressings on a mice model experiment and 50% faster healing of wounds on the mice was noticed for the non-crosslinked mats than the control one.

Abstract:Abstract Nanotechnology had been widely developed for medical application in various forms, either nanoparticles or nanofibers. One of the most attractive use is nanofibers as a wound dressing. An ideal wound dressing should have characteristics of easily sterilized, non-toxic, biodegradable, good oxygen circulation, and anti-microbial performance. Those characteristics could be obtained by using a biomaterial composite which had a good biocompatibility on human body tissues, such as chitosan and hydroxyapatite (HA). Chitosan had an anti-microbial property, while HA could increase the cell generation rate. Therefore, this work would synthesize a chitosan/HA nanofiber as a potential wound dressing via electrospinning method. HA was firstly prepared by electrolysis using alternating electrode configuration. The dope solution of chitosan/HA for electrospinning was prepared by adding polyvinyl alcohol (PVA) as a coadjutant polymer. The morphology of the obtained nanofibers was investigated by SEM. It was revealed that the nanofibers had an average diameter of 261.87 nm which was spun each other forming many pores which was good for oxygen circulation. Therefore, chitosan/HA nanofibers could be a promising material for wound dressing application.
Synergistic antibacterial and wound healing effects of chitosan nanofibers with ZnO nanoparticles and dual antibiotics

Abstract:One concern that has been considered potentially fatal is bacterial infection. In addition to the development of biocompatible antibacterial dressings, the screening and combination of new antibiotics effective against antibiotic resistance are crucial. In this study, designing hemostasis electrospun composite nanofibers containing chitosan (CS), polyvinyl pyrrolidone (PVP) and Gelatin (G) as the major components of hydrogel and natural nanofibrillated sodium alginate (SA)/polyvinyl alcohol (PVA) and ZnO nanoparticles (ZnONPs) combination as the nanofiller ingredient, has been investigated which demonstrated significant potential for accelerating wound healing. The hydrogels were developed for the delivery of the amikacin and cefepime antibiotics, along with zinc oxide nanoparticles that were applied to an electrospun layer. Amikacin is a highly effective aminoglycoside antibiotic, particularly for hospital-acquired infections, but its use is limited due to its toxicity. By utilizing it in low concentrations in the form of nanofibers and combining it with cefepime, which exhibits synergistic effects, enhanced efficacy against bacterial pathogens is achieved while potentially minimizing cytotoxicity compared to individual antibiotics. This dressing demonstrated efficient drug release, flexibility, and good swelling properties, indicating its suitable mechanical properties for therapeutic applications. After applying the biocompatible hydrogel to wounds, a significant acceleration in wound closure was observed within 14 days compared to the control group. Furthermore, the notable antibiotic and anti-inflammatory properties underscore its effectiveness in wound healing, making it a promising candidate for medical applications.

Abstract:Advanced therapeutic dressings are the current research interest that achieves rapid and complete wound healing. The healing of skin wounds demonstrates a remarkable cellular function process that is distinct in nature and involves the interaction of various cells, growth factors, and cytokines. In this study, nanofibrous scaffolds were prepared using poly(ε-caprolactone) and polylactic acid as mat scaffolds. Also, copper nanoparticles (CuNPs) and curcumin were added to fabricate a hybrid nanocomposite scaffold using the electrospinning technique. The fabricated scaffolds were analyzed by scanning electron microscopy (SEM), transmission electron microscopy (TEM), tensile analysis, porosity, and water vapor transmission rate. SEM analysis proved the nanostructured fibers have suitable porosity without any beads. The presence of CuNPs in the PCL-Cu nanofibrous mat was confirmed by TEM analysis. The tensile test confirmed an increase and decrease in the elongation ratio by CuNPs and curcumin addition, respectively. The biocompatibility and cell attachment to the nanofibers were proved by MTT and DAPI staining which proved a significantly positive effect of curcumin in the cell growth. The scaffold can hinder both the gram-negative and gram-positive bacteria through direct contact with them. This research study showed that the addition of CuNPs and curcumin in the hybrid nanocomposite scaffold compensates for each other's shortcomings and has the potential to be used in wound healing applications.

Abstract:The protein-polysaccharide nanofibers have attracted intensive attention in promoting wound healing, due to their components and nanoscale fibrous structure that mimics the native extracellular matrix (ECM). For the full-thickness wounds, in addition to promoting healing, hemostatic property and antibacterial activity are also of critical importance. However, currently, protein-polysaccharide-based nanofiber membranes exhibit poor mechanical properties, lack inherent hemostatic and antibacterial capabilities, as well as the ability to promote tissue repair. In this study, we developed composited membranes, which were composed of collagen (Col) and chitosan (Chs), through solvent alteration and post-processing, the membranes showed enhanced stability under physiological conditions, proper hydrophilic performance and improved mechanical property. Appropriated porosity and water vapor transmission rate, which benefit to wound healing, were detected among all the membranes except for Col membrane. Aimed at wound dressing, hemocompatibility, antibacterial activity and cell proliferation of the electrospun membranes were evaluated. The results indicated that the Col/Chs composited membranes exhibited superior blood clotting capacity, and the membranes with Chs exceeding 60% possessed sufficient antibacterial activity. Moreover, compared with Chs nanofibers, significant increase in cell grow was detected in Col/Chs (1:3) membrane. Taken together, the electrospun membrane with multiple properties favorable to wound healing, superior blood coagulation, sufficient antibacterial performance and promoting cell proliferation property make it favorable candidate for full-thickness skin wound healing.
