Abstract:Several human studies indicate that mobile phone specific electromagnetic fields may cause cancer in humans but the underlying molecular mechanisms are currently not known. Studies concerning chromosomal damage (which is causally related to cancer induction) are controversial and those addressing this issue in mobile phone users are based on the use of questionnaires to assess the exposure. We realized the first human intervention trial in which chromosomal damage and acute toxic effects were studied under controlled conditions. The participants were exposed via headsets at one randomly assigned side of the head to low and high doses of a UMTS signal (n = 20, to 0.1 W/kg and n = 21 to 1.6 W/kg Specific Absorption Rate) for 2 h on 5 consecutive days. Before and three weeks after the exposure, buccal cells were collected from both cheeks and micronuclei (MN, which are formed as a consequence of structural and numerical chromosomal aberrations) and other nuclear anomalies reflecting mitotic disturbance and acute cytotoxic effects were scored. We found no evidence for induction of MN and of nuclear buds which are caused by gene amplifications, but a significant increase of binucleated cells which are formed as a consequence of disturbed cell divisions, and of karyolitic cells, which are indicative for cell death. No such effects were seen in cells from the less exposed side. Our findings indicate that mobile phone specific high frequency electromagnetic fields do not cause acute chromosomal damage in oral mucosa cells under the present experimental conditions. However, we found clear evidence for disturbance of the cell cycle and cytotoxicity. These effects may play a causal role in the induction of adverse long term health effects in humans.
Human population studies with the exfoliated buccal micronucleus assay: statistical and epidemiological issues

Abstract:Objectives: Micronucleus (MN) assay in buccal exfoliated cells is a minimally invasive method for monitoring genetic damage in human populations. Statistical and epidemiological issues related to the design and analysis of studies based on this biomarker are addressed. Methods: A systematic review of recent literature on the buccal MN assay has been carried out to provide a state-of-the-art evaluation of how critical topics such as control for confounding, sample size and statistical power, number of cells scored, endpoint selection, and statistical modelling, are considered. In addition, a meta-analysis has been performed to estimate the impact of most common confounders on MN frequency, and to provide a baseline value of MN frequency in the control population. Results: A total number of 63 eligible studies were included in the analysis. Age (98.4%), gender (85.7%), and smoking habit (90.5%) were the most commonly studied confounders. Univariate statistics were estimated in most studies while multivariate analysis was applied only in the 47.6%. Baseline MN frequency in controls was 1.10/1000 cells (95% confidence interval 0.70-1.72), and the relative increment in subjects exposed to genotoxic agents or affected by disease correlated with similar observations in lymphocytes (R(2)=0.74). A minimum number of 4000 cells is recommended to reduce the variability of the MN mean estimates, in contrast with the current practice of scoring only 2000 cells (81% of studies). Poisson or Negative Binomial are the preferred statistical models when more than 2000 cells are scored. Studies scoring smaller numbers of cells should consider the use of statistical models taking into account the excess of zeros, e.g., the Zero Inflated Poisson (ZIP) models. Conclusions: The quality of papers published on the buccal MN assay can be substantially improved, with better consideration of basic issues such as power analysis, control for confounding, choice of the statistical model, and the number of cells to be scored.

Abstract:BACKGROUND: The radiation released at the time of dental panoramic radiographs causes genotoxic and cytotoxic effects on epithelial cells. OBJECTIVE: This research aimed to evaluate the changes in the frequencies of micronucleated cells in patients' buccal epithelial cells following dental panoramic radiography. METHODS: 74 patients were recruited for the study who were advised for panoramic radiographs. Using a wooden spatula, the buccal epithelial cells were scraped from both cheeks before to panoramic radiation exposure and ten days after the panoramic radiation exposure. Giemsa stain was used to stain the cells, and 500 cells were scored on a slide to determine the frequency of micronuclei. To determine the difference between the frequency of micronuclei before and after radiation exposure, a paired t-test was used in the statistical analysis. RESULTS: The proportion of micronuclei cells was 0.11% before radiation exposure and 0.57% following radiation exposure after 10 days. A statistically significant increase in the frequencies of micronuclei was noted after radiation exposure values. CONCLUSION: This study revealed the genotoxicity of epithelial cells with dental panoramic radiation exposure. It is advised to reduce the use of such radiographs and to use only when there is no other diagnostic tool that is helpful or when absolutely essential.

Abstract:Abstract The aim of this study was to investigate if age and body mass of humans have an impact on the DNA-damaging properties of high-frequency mobile phone-specific electromagnetic fields (HF-EMF, 1950 MHz, universal mobile telecommunications system, UMTS signal) and if this form of radiation has an impact on the genotoxic effects of occupationally relevant exposures. Pooled peripheral blood mononuclear cells (PBMC) from three groups [young normal weight, young obese (YO), and older age normal weight individuals] were exposed to different doses of HF-EMF (0.25, 0.5, and 1.0 W/kg specific absorption rate—SAR) and simultaneously or sequentially to different chemicals which cause DNA damage (CrO3, NiCl2, benzo[a]pyrene diol epoxide—BPDE, and 4-nitroquinoline 1-oxide—4NQO) via different molecular mechanisms. We found no difference in regard to the background values in the three groups but a significant increase of DNA damage (81% without and 36% with serum) in cells from old participants after radiation with 1.0 W/kg SAR 16 h. In combined treatment experiments we found no impact of the UMTS signal on chemically induced DNA damage in the different groups in general. However, a moderate decrease of DNA damage was seen in simultaneous treatment experiments with BPDE and 1.0 W/kg SAR in the YO group (decline 18%). Taken together our findings indicate that HF-EMF cause DNA damage in PBMC from older subjects (69.1 years). Furthermore, they show that the radiation does not increase induction of DNA damage by occupationally relevant chemicals.

Abstract:To investigate the effect of microwave emitted by mobile phones on the rat central nervous system (CNS), in vitro cultured cortical neuronal cells and in vivo rat’s brain were exposed to the electromagnetic waves emitted by a microwave transmitter that mimics the working frequency of mobile phones. Trypan blue staining and terminal deoxynucleotidy transferase-mediated dUTP nick-end labeling (TUNEL) were used to determine the survival state of neuronal cells while immunohistochemistry method was used to determine the expression level of Bcl-2 and Bax. Buccal mucosa cell damage in individuals following dental X-ray examinations

Abstract:The aim of the present study was to monitor genotoxic and cytotoxic effect of X-ray on exfoliated buccal mucosa cells and investigate the association between the effects and the accumulated absorbed doses of oral mucosa. 98 participants’ buccal mucosa cells were collected before and 10 days after different series of dental radiographs performed. Cytological preparations were successively dyed with the methods of Feulgen and fast-green, and analyzed under a light microscope. Micronuclei (MN)and other cells were scored. Accumulated absorbed dose of buccal mucosa was estimated with the method of anthropomorphic phantom and dosimeter chips. The dose rang was 0.18–3.54 mGy. A significant difference in the rate of MN cell was found before and after X-ray examinations ( P = 0.008) as well as in the rates of Pyknotic (p < 0.001) and Karyolytic cell (p = 0.0021). When only the patients whose mucosa absorbed dose is lower than 1 mGy was analyzed, significant differences were not found except for Karyolytic cells (p = 0.0313). There was a correlation between the accumulated does and the change rate (ρ = 0.25, p = 0.0118). The frequency of micronuclei cells in buccal mucosa may be increased when a series of dental radiographs including a CBCT examination was performed.

Abstract:Ear, Nose &Throat Journal, Ahead of Print. Objectives: The use of mobile phone is increasing around the world. Although it is beneficial in terms of communication, the electromagnetic radiations emitted by mobile phones may cause undesirable biological effects on the human body. In practical use, the tissue with which mobile phones come into most and are closest is the parotid gland. This study investigated the effects of the 1800 MHz electromagnetic field created by a generator on the parotid gland in rats. Methods: A total of 21 Sprague–Dawley Albino rats were included in the study. The rats were randomly divided into three equal groups. To simulate a mobile phone in conversation mode, the first study group was exposed to an 1800-MHz electromagnetic field for 6 hours a day for 30 days, and the second study group was exposed to an 1800-MHz electromagnetic field for 12 hours a day for 30 days. After 30 days, rats were sacrificed, and histopathological and immunohistochemical methods were used to evaluate the effects on the parotid gland. The total antioxidant level and the total oxidant level were measured biochemically in homogenized parotid tissue. Results: Histopathological results showed an increase in degeneration in rats exposed to electromagnetic fields for 6 and 12 hours a day, and immunohistochemical analysis showed an increase in the apoptotic index in both study groups (P = .001, P < .001). Intranuclear inclusions was observed during histopathological examination performed by electron microscopy. Conclusions: This study observed that the 1800 MHz electromagnetic field caused undesirable adverse histopathological and biochemical effects on the parotid gland of rats. Histopathological and biochemical findings were detected with increasing contact and exposure time. This study will lead to other studies on this topic and contribute to the literature by completing other studies.

Abstract:Reducing the interference between ultra wideband (UWB) and existing wireless communication systems has become of importance recently. In this study, we propose an equal 1 to 4 power divider (PD) which enables the suppression of three different frequencies, 3.95, 5.2 and 5.8 GHz, in the UWB frequency band by means of U-shaped defected ground structure (DGS). In this respect, it is essential to provide steep band-rejection with high Q-factor. The authors have used a novel method to attain this goal using U-shaped DGS. Studies show that there is a relation between the line width and Q-factor of a given band-notched structure. Therefore, instead of using a 50-50-50 ohm configuration, a 50-100-50 ohm transformation is used which has a narrower line width in the middle of the PD providing a steeper suppression with a high Q-factor. The measured Q-factor compared to similar structures is higher having a value of 27.5 with more than 30 dB suppression.

Abstract:Objectives: To evaluate chromosomal damage and cytotoxicity in exfoliated buccal mucosa cells, cells of the tongue and epithelial gingival cells from adults following CBCT scan and to compare the sensitivity of the different exfoliated cells to a same dosage of ionizing radiation. Methods: The study included 46 healthy participants (median age 27 years; age range 23-42 years) who had a CBCT scan. Exfoliated mucosa cells were collected immediately before the CBCT scan and 10 days after. Cells were centrifuged, fixed in the fluid of methanol : glacial acetic acid (3 : 1) and stained using the method of Schiff's reagent and fast green. One observer analyzed all the slides. For interobserver variances, a second observer scored 16 slides chosen from all the subjects. The same set of 16 slides were analyzed once again a month later for intraobserver variances. Results: There is no significant differences for micronucleated cells before and after a CBCT scan in exfoliated buccal mucosa cells (p = 0.476), cells of the tongue (p = 0.884) and epithelial gingival cells (p = 0.362). The frequencies of pyknosis cell and karyolysis cell had significantly increased after CBCT scan in the three groups. No significant difference was found among the three kinds of mucosa cells (p = 0.557). The interobserver (p = 0.624) and intraobserver (p = 0.193) variances were not significant. Conclusions: A CBCT scan may induce cytotoxicity but not chromosomal damage in the oral mucosa cells, including buccal mucosa cells, cells of the tongue or epithelial gingival cells. The sensitivity of the different exfoliated cells to the same dosage of radiation had no statistically significant difference.

Abstract:Iodine deficiency was investigated in 575 women from 12 villages of Pupaman, Si Chomphu and Amphoe Chum Phae in Khon Kaen province in a project survey under Thai-German cooperation entitled "Promotion of Health and Nutrition Status of Rural Women in Northeast Thailand". The prevalence of goiter (grades 1A, 1B, 2 and 3) were 71.6 per cent in Pupaman, 58.4 per cent in Amphoe Si Chomphu and 35.1 per cent in Amphoe Chum Phae. However, the percentage of urine iodine deficiency of the women from these districts, is reversely with the palpation grading of thyroid gland. The high prevalence of goiter in Pupaman might occur due to goitrogenic effects, although the urine iodine concentration was lower than other districts. Further study about the determination of thiocyanate should be suggested in the high prevalence of goiter.

Abstract:Objective To evaluate the genetic damage and cellular death in exfoliated buccal mucosa cells from patients undergoing dental Oral and maxillofacial radiology.Methods Orthodontics (14 male and 10 male) before and 7-10 days after they underwent different series of X-ray examinations.Cytological preparations were successively dyed with methods of Feulgen and fast-green,and analyzed under a fluorescence microscope.Micronucleated cells and other type of cells (basal cells,binucleated cells,condensed chromatin cells,karyorrhectic cells,pyknotic cells,karyolytic cells and cells with nuclear buds) were scored in 1000 cells.Results There was no statistically significant difference (P ＞0.05) between the frequency of micronucleated cells before and after dental X-ray exposures.The frequency of micronucleus was not significantly different between the patients aged under 18 and those over 18 years old.Large dose exposure was related to increased karyolytic cells (P =0.03).Conclusions X-ray dose was directly related to increased cell apotosis.The patients younger than 18 years old were not more susceptible to X-ray than those over 18.

Abstract:This study aimed to evaluate all studies which used the micronucleus assay using oral cells in the attempt to understand whether such technique is efficient in evaluating genotoxicity in gas station attendants. Full manuscripts from 16 studies were carefully selected by the authors. Our results demonstrate that continuous exposure to derivatives of petroleum may lead to genotoxic effects since all studies demonstrated positive findings (16 out of 16) and 11 of them had a strong or moderate final rating. In summary, our results reveal that gas station attendants are occupationally exposed to genotoxic agents and that the micronucleus assay in oral mucosa is indeed an effective method to evaluate genotoxicity in this specific case. Such findings are very important for protecting these professionals who are continuously exposed to chemicals for long periods.

Micronucleus evaluation of exfoliated buccal epithelial cells using liquid-based cytology preparation

Abstract:Objective: Micronuclei (MN) are biomarkers that can be applied to buccal epithelial cells to assess populations occupationally exposed to potentially carcinogenic agents. Liquid-based cytology (LBC) is a way to improve and refine the results obtained by this test. Study design: Exfoliated buccal cells were collected from 40 subjects (20 construction workers from the Barretos Cancer Hospital and 20 administrative staff from the same institution). LBC and three stains (Feulgen/fast green, Papanicolaou and Giemsa) were used to prepare the slides. Student's t test was applied for statistical comparisons of the data. A p value of <0.05 was considered statistically significant. Results: Regardless of the stain employed, the frequency of MN was greater in the case group (Feulgen/fast green: 5.15; Papanicolaou: 29; Giemsa: 26) than in the control group (Feulgen/fast green: 2.30; Papanicolaou: 17; Giemsa: 15). Conclusions: Using LBC to prepare slides and evaluate the frequency of MN potentially serves as a screening option for more comprehensive studies of cancer risk among populations occupationally exposed to potentially carcinogenic agents. In addition, the residual fluid enables the preparation of slides for DNA-specific stains that can be compared to those with Papanicolaou stain.
Mobile telephony radiation exerts genotoxic action and significantly enhances the effects of gamma radiation in human cell

Abstract:I previously reported chromosomal damage in human peripheral blood lymphocytes (HPBLs) induced by: a) mobile telephony (MT) electromagnetic fields (EMFs)/electromagnetic radiation (EMR), b) a high caffeine dose, and c) the combination of the two stressors. HPBLs from the same subjects exposed to gamma radiation at doses 0.1, 0.3, or 0.5 Gy, displayed more aberrations than those exposed to MT EMFs or the high caffeine dose in a dose-dependent manner. When the cells exposed to these gamma radiation doses were pre-exposed to a single 15-min MT EMF exposure, the number of aberrations increased significantly more than the sum number of aberrations induced by the individual stressors in all subjects. Thus, MT EMF exposure at a power density ~136 times below the latest International Commission on Non- Ionizing Radiation Protection (ICNIRP) exposure limit, apart from the fact that it is genotoxic by itself, significantly enhanced the genotoxic action of gamma radiation. Since gamma radiation at similar doses is applied for diagnostic and therapeutic purposes, people should be aware of the increased risk during treatment periods. Comparison of the genotoxic action between MT EMF and gamma radiation shows that the ICNIRP limits are, at least, ~4.5×10<sup>4</sup> times less stringent than the limits for gamma radiation.

Abstract:There has been growing concern about the possibility of adverse health effects resulting from exposure to radiofrequency radiations (RFR), such as those emitted by wireless communication devices. Since the introduction of mobile phones many studies have been conducted regarding alleged health effects but there is still some uncertainty and no definitive conclusions have been reached so far. Although thermal effects are well understood they are not of great concern as they are unlikely to result from the typical low-level RFR exposures. Concern rests essentially with the possibility that RFR-exposure may induce non-thermal and/or long-term health effects such as an increased cancer risk. Consequently, possible genetic effects have often been studied but with mixed results. In this paper we review the data on alleged RFR-induced genetic effects from in vitro and in vivo investigations as well as from human cytogenetic biomonitoring surveys. Attention is also paid to combined exposures of RFR with chemical or physical agents. Again, however, no entirely consistent picture emerges. Many of the positive studies may well be due to thermal exposures, but a few studies suggest that biological effects can be seen at low levels of exposure. Overall, however, the evidence for low-level genotoxic effects is very weak.
Evaluation of micronuclear frequencies in both circulating lymphocytes and buccal epithelial cells of patients with oral lichen planus and oral lichenoid contact reactions

Abstract:Objective: The aim of this study was to evaluate the frequency of micronuclei (MNs) in both circulating lymphocytes and buccal epithelial cells of patients with oral lichenoid contact reactions (OLCRs) or with oral lichen planus (OLP) and compare their MN scores with those of healthy controls (HCs). Material and methods: The study group included 21 patients (mean age 51.3 ± 12.4; 6 males, 15 females) with OLCRs and 22 patients (mean age 47.6 ± 14.4; 4 males, 18 females) with OLP who were clinically diagnosed and histopathologically confirmed according to WHO diagnostic criteria (WHO Collaborating Centre for Oral Precancerous Lesions, 1978). All patients with OLCR demonstrated contact allergy to tested dental materials when evaluated by skin patch testing according to International Contact Dermatitis Research Group (ICDRG), while all OLP patients tested negative to patch testing. Seventeen individuals with no oral mucosal disorders (mean age 51.7 ± 11.3; 8 males, 9 females) were recruited to constitute the healthy control group. Results: Most OLP and OLCR lesions were of reticular type (83%), and OLP lesions were distributed bilaterally on the buccal mucosa (90.5%). The medians of MN frequencies in buccal epithelial cells in OLP and OLCR groups were significantly higher when compared with HC group (P < 0.001). [Correction added on 30 May 2014, after first online publication: in the results, 2nd sentence, the word 'lymphocytes' has been removed.] There was no significant difference between OLP group (14.5 range 3-95) and OLCR group (16.0 range 3-93) in terms of median MN frequencies in buccal epithelial cells (P = 0.724) nor in peripheral lymphocytes between OLP group (2.0 range 0-7) and OLCR group (1.0 range 0-6) (P = 0.92). [Correction added on 30 May 2014, after first online publication: (P = 0.92) was wrongly placed after 'peripheral lymphocytes' and has now been shifted to the end of the last sentence.] Conclusions: Micronuclei scores do not distinguish OLP from OLCR when using buccal smears. OLP and OLCR both demonstrated significantly higher MN frequencies in buccal cells, compared with healthy controls. MN assessment in both buccal epithelial cells and circulating lymphocytes may serve as a potential biomarker tool for evaluating any cancer risk in OLP and OLCR. Abstract:With the rapid development of communication technology, there is a widespread use of mobile telephone, whose effect on human has been getting more and more attention. The electromagnetic wave can penetrate biosystems and interacts with biological tissue at different biological levels. Although many researchers have been studying biological effects of electromagnetic radiation for years, their results are not consistent and need to be confirmed by reliable and repeatable experiments. Since the energy of electromagnetic radiation of mobile phone is much low and the organism has an adaptability to external stimulation, short-time radiation may not induce obvious effect on organism. To avoid other inductive factors in the environment, we used in vitro cell culture technology and exposure experiment to study the activating effect of 900 MHz electromagnetic fields (EMF) on NIH/3T3 cells. NIH/3T3 cells are sensitive to sarcoma virus focus formation and leukaemia virus propagation, and therefore are often used for DNA transformation studies. The NIH/3T3 cells were divided into four groups, the control group with sham exposure and the three exposure groups exposed to 10W/m2, 50W/m2 and 90W/m2 916MHz EMF respectively. The exposure lasted 2 hours a day. After exposure we found the cells morphology changed and had the characteristics of epithelial cells. The Soft agar assay showed that all the exposure groups formed colonies in soft agar while the control group did not form colonies. The results in this study suggest that 916MHz electromagnetic waves can promote malignant transformation of NIH/3T3 cells and induce cellular canceration .
Time trend in incidence of malignant neoplasms of the central nervous system in relation to mobile phone use among young people in Japan

Abstract:The aim of this study was to examine whether incidence of malignant neoplasms of the central nervous system from 1993 to 2010 has increased among young people in Japan, and whether the increase could be explained by increase in mobile phone use. Joinpoint regression analysis of incidence data was performed. Subsequently, the expected incidence rate was calculated assuming that the relative risk was 1.4 for those who used mobile phones more than 1640 h cumulatively. Annual percent change was 3.9% (95% confidence interval [CI], 1.6-6.3) for men in their 20s from 1993 to 2010, 12.3% (95% CI, 3.3-22.1) for women in their 20s from 2002 to 2010, 2.7% (95% CI, 1.3-4.1) for men in their 30s from 1993 to 2010, and 3.0% (95% CI, 1.4-4.7) for women in their 30s from 1993 to 2010. Change in incidence rates from 1993 to 2010 was 0.92 per 100,000 people for men in their 20s, 0.83 for women in their 20s, 0.89 for men in their 30s, and 0.74 for women in their 30s. Change in expected incidence rates from 1993 to 2010 was 0.08 per 100,000 people for men in their 20s, 0.03 for women in their 20s, 0.15 for men in their 30s, and 0.05 for women in their 30s. Patterns in sex-, age-, and period-specific incidence increases are inconsistent with sex-, age-, and period-specific prevalence trends, suggesting the overall incidence increase cannot be explained by heavy mobile phone use. Abstract:Objective To evaluate the frequency of micronucleated (MN) cells in different sites of normal oral mucosa.Methods The smears of oral exfoliative cells were made from buccal mucosa,the dorsum and ventrum of the tongue from 90 normal individuals.The number of MN cells was counted.Results The frequency of MN cells in buccal mucosa was higher than both in the dorsum and ventrum of the tongue.There was a significant increase of MN cells in smokers than in nonsmokers (P 0.001 ).The consumption of tobacco and the period of smoking (year) had statisitically significant correlation with the mean percentage of MN cells.Conclusion Buccal mucosa exposes to all kinds of stimulates more directly than the ventrum of the tongue.Tobacco has an important effects on carcinogenesis.
