Abstract:Sugars, produced through photosynthesis, are at the core of all organic compounds synthesized and used for plant growth and response to environmental changes. Their production, transport, and utilization are highly regulated and integrated throughout the plant life cycle. The maintenance of sugar partitioning between the different subcellular compartments and between cells is important in adjusting the photosynthesis performance and response to abiotic constraints. Here, we investigated, in Arabidopsis, the consequences of the disruption of four genes coding for SWEET sugar transporters (SWEET11, SWEET12, SWEET16, and SWEET17) on plant photosynthesis and response to drought. Our results show that mutations in both SWEET11 and SWEET12 genes lead to an increase of cytosolic sugars in mesophyll cells and phloem parenchyma cells, which impacts several photosynthesis-related parameters. Further, our results suggest that in the swt11swt12 double mutant, the sucrose-induced feedback mechanism on stomatal closure is poorly efficient. On the other hand, changes in fructose partitioning in mesophyll and vascular cells, measured in the swt16swt17 double mutant, positively impact gas exchanges, probably through an increased starch synthesis together with higher vacuolar sugar storage. Finally, we propose that the impaired sugar partitioning, rather than the total amount of sugar observed in the quadruple mutant, is responsible for the enhanced sensitivity upon drought. This work highlights the importance of considering SWEET-mediated sugar partitioning rather than global sugar content in photosynthesis performance and plant response to drought. Such knowledge will pave the way to design new strategies to maintain plant productivity in a challenging environment.

Abstract:Plant SWEETs (sugars will eventually be exported transporters) play a role in plant growth and plant response to biotic and abiotic stresses. In the present study, DsSWEET12 from Dianthus spiculifolius was identified and characterized. Real-time quantitative PCR analysis revealed that DsSWEET12 expression was induced by sucrose starvation, mannitol, and hydrogen peroxide. Colocalization experiment showed that the DsSWEET12-GFP fusion protein was localized to the plasma membrane, which was labeled with FM4-64 dye, in Arabidopsis and suspension cells of D. spiculifolius. Compared to wild type plants, transgenic Arabidopsis seedlings overexpressing DsSWEET12 have longer roots and have a greater fresh weight, which depends on sucrose content. Furthermore, a relative root length analysis showed that transgenic Arabidopsis showed higher tolerance to osmotic and oxidative stresses. Finally, a sugar content analysis showed that the sucrose content in transgenic Arabidopsis was less than that in the wild type, while fructose and glucose contents were higher than those in the wild type. Taken together, our results suggest that DsSWEET12 plays an important role in seedling growth and plant response to osmotic and oxidative stress in Arabidopsis by influencing sugar metabolism.

Abstract:Sugars Will Eventually be Exported Transporters (SWEETs) are the most recently discovered family of plant sugar transporters. By acting as uniporters, SWEETs facilitate the diffusion of sugars across cell membranes and play an important role in various physiological processes such as abiotic stress adaptation. AtSWEET17, a vacuolar fructose facilitator, was shown to be involved in the modulation of the root system during drought. In addition, previous studies have shown that overexpression of an apple homolog leads to increased drought tolerance in tomato plants. Therefore, SWEET17 might be a molecular element involved in the plant ́s drought response. However, the role and function of SWEET17 in aboveground tissues of Arabidopsis under drought stress remains elusive. By combining gene expression analysis and stem architecture with the sugar profiles of different aboveground tissues, we uncovered a putative role of SWEET17 in carbohydrate supply and thus cauline branch elongation, especially during periods of carbon limitation, as occurs under drought stress. Thus, SWEET17 seems to be involved in maintaining efficient plant reproduction under drought-stress conditions.

AtSWEET11 and AtSWEET12 in Arabidopsis act synergistically to perform distinct physiological roles, particularly in apoplasmic phloem loading, seed filling, and sugar level alteration at the site of pathogen infection. Plasma membrane-localized AtSWEET11 and AtSWEET12 transporters exclusively facilitate sucrose transport along the concentration gradient. This article examines the sucrose binding pocket of AtSWEET11 and AtSWEET12 using docking studies, and how they act synergistically in various functions throughout plant development and during abiotic and biotic stresses. Further, we highlight the phylogenetic and the in-silico analyses of AtSWEET11 and AtSWEET12 orthologs from 39 economically important plant species that could provide new platforms for future studies on sugar allocation mechanisms across the different plant families. In-depth understanding of these transporters and their molecular regulatory mechanisms could be harnessed for crop improvement and crop protection.

Abstract:The sugar will eventually be exported transporter (SWEET) members in Arabidopsis, AtSWEET11 and AtSWEET12 are the important sucrose efflux transporters that act synergistically to perform distinct physiological roles. These two transporters are involved in apoplasmic phloem loading, seed filling, and sugar level alteration at the site of pathogen infection. Here, we performed the structural analysis of the sucrose binding pocket of AtSWEET11 and AtSWEET12 using molecular docking followed by rigorous molecular dynamics (MD) simulations. We observed that the sucrose molecule binds inside the central cavity and in the middle of the transmembrane (TM) region of AtSWEET11 and AtSWEET12, that allows the alternate access to the sucrose molecule from either side of the membrane during transport. Both AtSWEET11 and AtSWEET12, shares the similar amino acid residues that interact with sucrose molecule. Further, to achieve more insights on the role of these two transporters in other plant species, we did the phylogenetic and the in‐silico analyses of AtSWEET11 and AtSWEET12 orthologs from 39 economically important plants. We reported the extensive information on the gene structure, protein domain and cis‐acting regulatory elements of AtSWEET11 and AtSWEET12 orthologs from different plants. The cis‐elements analysis indicates the involvement of AtSWEET11 and AtSWEET12 orthologs in plant development and also during abiotic and biotic stresses. Both in silico and in planta expression analysis indicated AtSWEET11 and AtSWEET12 are well‐expressed in the Arabidopsis leaf tissues. However, the orthologs of AtSWEET11 and AtSWEET12 showed the differential expression pattern with high or no transcript expression in the leaf tissues of different plants. Overall, these results offer the new insights into the functions and regulation of AtSWEET11 and AtSWEET12 orthologs from different plant species. This might be helpful in conducting the future studies to understand the role of these two crucial transporters in Arabidopsis and other crop plants.

Abstract:Sugars, which are critical osmotic compounds and signalling molecules in plants, and Sugars Will Eventually be Exported Transporters (SWEETs), which constitute a novel family of sugar transporters, play central roles in plant responses to multiple abiotic stresses. In the present study, a member of the SWEET gene family from cucumber (Cucumis sativus L.), CsSWEET2, was identified and characterized. Histochemical analysis of β-glucuronidase expression in transgenic Arabidopsis plants showed that CsSWEET2 is highly expressed in the leaves; subcellular localization indicated that CsSWEET2 proteins are localized in the plasma membrane and endoplasmic reticulum. Heterologous expression assays in yeast demonstrated that CsSWEET2 encodes an energy-independent hexose/H+ uniporter that can complement both glucose and fructose transport deficiencies. Compared with wild-type Arabidopsis plants, transgenic Arabidopsis plants overexpressing CsSWEET2 had much lower relative electrolyte leakage levels and were much more resistant to cold stress. Sugar content analysis showed that glucose and fructose levels in the transgenic Arabidopsis plants were significantly higher than those in the wild-type plants. Taken together, our results suggest that, by mediating sugar metabolism and compartmentation, CsSWEET2 plays a vital role in improving plant cold tolerance.

Abstract:Sucrose is essential for plants for several reasons: It is a source of energy, a signaling molecule, and a source of carbon skeletons. Sucrose phosphate synthase (SPS) catalyzes the conversion of uridine diphosphate glucose and fructose-6-phosphate to sucrose-6-phosphate, which is rapidly dephosphorylated by sucrose phosphatase. SPS is critical in the accumulation of sucrose because it catalyzes an irreversible reaction. In Arabidopsis thaliana, SPSs form a gene family of four members, whose specific functions are not clear yet. In the present work, the role of SPSA2 was investigated in Arabidopsis under both control and drought stress conditions. In seeds and seedlings, major phenotypic traits were not different in wild-type compared with spsa2 knockout plants. By contrast, 35-day-old plants showed some differences in metabolites and enzyme activities even under control conditions. In response to drought, SPSA2 was transcriptionally activated, and the divergences between the two genotypes were higher, with spsa2 showing reduced proline accumulation and increased lipid peroxidation. Total soluble sugars and fructose concentrations were about halved compared with wild-type plants, and the plastid component of the oxidative pentose phosphate pathway was activated. Unlike previous reports, our results support the involvement of SPSA2 in both carbon partitioning and drought response.

Abstract:Abstract Background Floral transition initiates reproductive development of plants and occurs in response to environmental and endogenous signals. In Arabidopsis thaliana , this process is accelerated by several environmental cues, including exposure to long days. The photoperiod-dependent promotion of flowering involves the transcriptional induction of FLOWERING LOCUS T ( FT ) in the phloem of the leaf. FT encodes a mobile protein that is transported from the leaves to the shoot apical meristem, where it forms part of a regulatory complex that induces flowering. Whether FT also has biological functions in leaves of wild-type plants remains unclear. Results In order to address this issue, we first studied the leaf transcriptomic changes associated with FT overexpression in the companion cells of the phloem. We found that FT induces the transcription of SWEET10 , which encodes a bidirectional sucrose transporter, specifically in the leaf veins. Moreover, SWEET10 is transcriptionally activated by long photoperiods, and this activation depends on FT and one of its earliest target genes SUPPRESSOR OF CONSTANS OVEREXPRESSION 1 ( SOC1 ). The ectopic expression of SWEET10 causes early flowering and leads to higher levels of transcription of flowering-time related genes in the shoot apex. Conclusions Collectively, our results suggest that the FT-signaling pathway activates the transcription of a sucrose uptake/efflux carrier during floral transition, indicating that it alters the metabolism of flowering plants as well as reprogramming the transcription of floral regulators in the shoot meristem.

Abstract:Crop yield depends on efficient allocation of sucrose from leaves to seeds. In Arabidopsis, phloem loading is mediated by a combination of SWEET sucrose effluxers and subsequent uptake by SUT1/SUC2 sucrose/H+ symporters. ZmSUT1 is essential for carbon allocation in maize, but the relative contribution to apoplasmic phloem loading and retrieval of sucrose leaking from the translocation path is not known. We therefor tested whether SWEETs are important for phloem loading in maize. Here we identified three leaf-expressed SWEET sucrose transporters as key components of apoplasmic phloem loading in Zea mays L. Notably, ZmSWEET13 paralogs (a, b, c) are among the highest expressed genes in the leaf vasculature. Genome-edited triple knock-out mutants are severely stunted. Photosynthesis of mutants was impaired and leaves accumulated starch and soluble sugars. RNA-seq revealed profound transcriptional deregulation of genes associated with the photosynthetic apparatus and carbohydrate metabolism. GWAS analyses may indicate that variability in ZmSWEET13s is correlated with agronomical traits, specifically flowering time and leaf angle. This work provides support for cooperation of three ZmSWEET13s with ZmSUT1 in phloem loading in Zea mays L. Our study highlights these three ZmSWEET13 sucrose transporters as possible candidates for the engineering of crop yield.

Abstract:Plant growth and development rely on the transport and use of sugars produced during photosynthesis. Sugars have a dual function as nutrients and signal molecules in the cell. Many factors maintaining sugar homeostasis and signaling are now identified, but our understanding of the mechanisms involved in coordinating intracellular and intercellular sugar translocation is still limited. We also know little about the interplay between sugar transport and signaling and the formation of the vascular system, which controls long‐distance sugar translocation. Sugar signaling has been proposed to play a role; however, evidence to support this hypothesis is still limited. Here, we exploited recent transcriptomics datasets produced in aerial organs of Arabidopsis to identify genes coding for sugar transporters or signaling components expressed in the vascular cells. We identified genes belonging to sugar transport and signaling for which no information is available regarding a role in vasculature development. In addition, the transcriptomics datasets obtained from sugar‐treated Arabidopsis seedlings were used to assess the sugar‐responsiveness of known genes involved in vascular differentiation. Interestingly, several key regulators of vascular development were found to be regulated by either sucrose or glucose. Especially CLE41, which controls the procambial cell fate, was oppositely regulated by sucrose or glucose in these datasets. Even if more experimental data are necessary to confirm these findings, this survey supports a link between sugar transport/signaling and vascular system development.This article is protected by copyright. Abstract:The Sugars Will Eventually be Exported Transporters (SWEET) family is a class of sugar transporters that play key roles in phloem loading, seed filling, pollen development and the stress response in plants. Here, a total of 18 JcSWEET genes were identified in physic nut (Jatropha curcas L.) and classified into four clades by phylogenetic analysis. These JcSWEET genes share similar gene structures, and alternative splicing of messenger RNAs was observed for five of the JcSWEET genes. Three (JcSWEET1/4/5) of the JcSWEETs were found to possess transport activity for hexose molecules in yeast. Real-time quantitative PCR analysis of JcSWEETs in different tissues under normal growth conditions and abiotic stresses revealed that most are tissue-specifically expressed, and 12 JcSWEETs responded to either drought or salinity. The JcSWEET16 gene responded to drought and salinity stress in leaves, and the protein it encodes is localized in both the plasma membrane and the vacuolar membrane. The overexpression of JcSWEET16 in Arabidopsis thaliana modified the flowering time and saline tolerance levels but not the drought tolerance of the transgenic plants. Together, these results provide insights into the characteristics of SWEET genes in physic nut and could serve as a basis for cloning and further functional analysis of these genes.
Impact of Drought on Soluble Sugars and Free Proline Content in Selected Arabidopsis Mutants

Abstract:Water shortage is an increasing problem affecting crop yield. Accumulation of compatible osmolytes is a typical plant response to overcome water stress. Sucrose synthase 1 (SUS1), and glucan, water dikinase 2 (GWD2) and δ1-pyrroline-5-carboxylate synthetase 1 (P5CS1) are members of small protein families whose role in the response of Arabidopsis thaliana plants to mild osmotic stress has been studied in this work. Comparative analysis between wild-type and single loss-of-function T-DNA plants at increasing times following exposure to drought showed no differences in the content of water-insoluble carbohydrate (i.e., transitory starch and cell wall carbohydrates) and in the total amount of amino acids. On the contrary, water-soluble sugars and proline contents were significantly reduced compared to wild-type plants regardless of the metabolic pathway affected by the mutation. The present results contribute to assigning a physiological role to GWD2, the least studied member of the GWD family; strengthening the involvement of SUS1 in the response to osmotic stress; showing a greater contribution of soluble sugars than proline in osmotic adjustment of Arabidopsis in response to drought. Finally, an interaction between proline and soluble sugars emerged, albeit its nature remains speculative and further investigations will be required for complete comprehension.

Abstract:Even though SWEETs (Sugars Will Eventually be Exported Transporters) have been found in every sequenced plant genome, a comprehensive understanding of their functionality is lacking. In this study, we focused on the SWEET family of barley (Hordeum vulgare). A radiotracer assay revealed that expressing HvSWEET11b in African clawed frog (Xenopus laevis) oocytes facilitated the bidirectional transfer of not just sucrose and glucose, but also cytokinin. Barley plants harboring a loss-of-function mutation of HvSWEET11b could not set viable grains, while the distribution of sucrose and cytokinin was altered in developing grains of plants in which the gene was knocked down. Sucrose allocation within transgenic grains was disrupted, which is consistent with the changes to the cytokinin gradient across grains, as visualized by magnetic resonance imaging and Fourier transform infrared spectroscopy microimaging. Decreasing HvSWEET11b expression in developing grains reduced overall grain size, sink strength, the number of endopolyploid endosperm cells, and the contents of starch and protein. The control exerted by HvSWEET11b over sugars and cytokinins likely predetermines their synergy, resulting in adjustments to the grain's biochemistry and transcriptome.

Abstract:Abstract Sugars Will Eventually be Exported Transporters (SWEETs) have important roles in numerous physiological mechanisms where sugar efflux is critical, including phloem loading, nectar secretion, seed nutrient filling, among other less expected functions. They mediate low affinity and high capacity transport, and in angiosperms this family is composed by 20 paralogs on average. As SWEETs facilitate the efflux of sugars, they are highly susceptible to hijacking by pathogens, making them central players in plant–pathogen interaction. For instance, several species from the Xanthomonas genus are able to upregulate the transcription of SWEET transporters in rice (Oryza sativa), upon the secretion of transcription-activator-like effectors. Other pathogens, such as Botrytis cinerea or Erysiphe necator, are also capable of increasing SWEET expression. However, the opposite behavior has been observed in some cases, as overexpression of the tonoplast AtSWEET2 during Pythium irregulare infection restricted sugar availability to the pathogen, rendering plants more resistant. Therefore, a clear-cut role for SWEET transporters during plant–pathogen interactions has so far been difficult to define, as the metabolic signatures and their regulatory nodes, which decide the susceptibility or resistance responses, remain poorly understood. This fuels the still ongoing scientific question: what roles can SWEETs play during plant–pathogen interaction? Likewise, the roles of SWEET transporters in response to abiotic stresses are little understood. Here, in addition to their relevance in biotic stress, we also provide a small glimpse of SWEETs importance during plant abiotic stress, and briefly debate their importance in the particular case of grapevine (Vitis vinifera) due to its socioeconomic impact.

Abstract:Sugar transport from the source leaf to the sink organ is critical for seed development and crop yield, as well as for responding to abiotic stress. SWEETs (sugar will eventually be exported transporters) mediate sugar efflux into the reproductive sink and are therefore considered key candidate proteins for sugar unloading during seed development. However, the specific mechanism underlying the sugar unloading to seeds in Camellia oleifera remains elusive. Here, we identified a SWEET gene named CoSWEET10, which belongs to Clade III and has high expression levels in the seeds of C. oleifera. CoSWEET10 is a plasma membrane-localized protein. The complementation assay of CoSWEET10 in SUSY7/ura3 and EBY.VW4000 yeast strains showed that CoSWEET10 has the ability to transport sucrose, glucose, and fructose. Through the C. oleifera seeds in vitro culture, we found that the expression of CoSWEET10 can be induced by hexose and sucrose, and especially glucose. By generating the restoration lines of CoSWEET10 in Arabidopsis atsweet10, we found that CoSWEET10 restored the seed defect phenotype of the mutant by regulating soluble sugar accumulation and increased plant drought tolerance. Collectively, our study demonstrates that CoSWEET10 plays a dual role in promoting seed development and enhancing plant drought resistance as a sucrose and hexose transporter.

Abstract:Sugars are the principal carbon and energy sources and serve as osmotic regulators and radical scavengers, thus playing an important role in plant responses to drought. Sugar transporters steering the distribution of sugar are vital players involved in tolerance to drought. Sugars Will Eventually be Exported Transporters (SWEETs) facilitate both the influx and efflux of mono- and/or disaccharides and control both inter and intracellular distribution of sugars. PagSWEET17a in Populus alba × P. glandulosa is one of four orthologous genes of AtSWEET17 in Arabidopsis. Unlike AtSWEET17, which is a vacuolar localized transporter, PagSWEET17a is localized to the endoplasmic reticulum (ER). Here, the role of PagSWEET17a in poplar responses to drought stress was investigated. PagSWEET17a was specifically expressed in cambium cells in younger root but mainly located in phloem fibers, and xylem vessels and fiber cells in the root undergoing secondary growth. Loss of PagSWEET17a inhibited the growth of roots in poplar seedlings and led to a decreased tolerance to drought. Analysis of sugar profiles revealed that accumulations of predominant sugars were significantly suppressed in both knockout (KO) mutant lines under drought. PagSWEET17a might contribute to poplar tolerance to drought by promoting drought-induced root expansion and diminishing oxidative damages caused by the stress.

Abstract:The main phloem loader in potato, sucrose transporter StSUT1, is co-expressed with two members of the SWEET gene family: StSWEET11b, a clade III member of SWEET carriers assumed to be involved in sucrose efflux, and StSWEET1g, a clade I member involved in glucose efflux into the apoplast that physically interacts with StSUT1. We investigated the functionality of SWEET carriers via uptake experiments with fluorescent glucose or sucrose analogs. Inhibition or overexpression of StSWEET1g/SlSWEET1e affected tuberization and flowering in transgenic potato plants. Isolation of the apoplasmic fluid by vacuum infiltration centrifugation revealed changes in the apoplasmic hexose composition and mono-to-disaccharide ratio, affecting sink strength. Down-regulation of StSWEET1g expression affected the expression of SP6A, a tuberigen, and miR172 under LD conditions, leading to early flowering and tuberization. A systematic screen for StSWEET1g-interacting protein partners revealed several proteins affecting cell wall integrity and strengthening. StSWEET1g and the main interaction partners were strongly down-regulated during tuber development. We discuss whether StSWEET1g activity might be linked to cell wall remodeling during tuber development and the switch from apoplasmic to symplasmic phloem unloading.

Abstract:Abstract Galactose is an abundant and essential sugar used for the biosynthesis of many macromolecules in different organisms, including plants. Galactose metabolism is tightly and finely controlled, since excess galactose and its derivatives are inhibitory to plant growth. In Arabidopsis (Arabidopsis thaliana), root growth and pollen germination are strongly inhibited by excess galactose. However, the mechanism of galactose-induced inhibition during pollen germination remains obscure. In this study, we characterized a plasma membrane-localized transporter, Arabidopsis Sugars Will Eventually be Exported Transporter 5, that transports glucose and galactose. SWEET5 protein levels started to accumulate at the tricellular stage of pollen development and peaked in mature pollen, before rapidly declining after pollen germinated. SWEET5 levels are responsible for the dosage-dependent sensitivity to galactose, and galactokinase is essential for these inhibitory effects during pollen germination. However, sugar measurement results indicate that galactose flux dynamics and sugar metabolism, rather than the steady-state galactose level, may explain phenotypic differences between sweet5 and Col-0 in galactose inhibition of pollen germination.

Abstract:The intercellular transport of sugars, nutrients, and small molecules is essential for plant growth, development, and adaptation to environmental changes. Various stresses are known to affect the cell-to-cell molecular trafficking modulated by plasmodesmal permeability. However, the mechanisms of plasmodesmata modification and molecules involved in the phloem unloading process under stress are still not well understood. Here, we show that heat stress reduces the root meristem size and inhibits phloem unloading by inducing callose accumulation at plasmodesmata that connect the sieve element and phloem pole pericycle. Furthermore, we identify the loss-of-function of CALLOSE SYNTHASE 8 (CalS8), which is expressed specifically in the phloem pole pericycle, decreasing the plasmodesmal callose deposition at the interface between the sieve element and phloem pole pericycle and alleviating the suppression at root meristem size by heat stress. Our studies indicate the involvement of callose in the interaction between root meristem growth and heat stress and show that CalS8 negatively regulates the thermotolerance of Arabidopsis roots.
