Abstract:The influence of light regulation on fungal growth and enzyme production was tested on endophytic isolates of Fusarium proliferatum (CCH) , Colletotrichum boninense (PL1, PL9, OL2), Colletotrichum gloeosporiodes (OL3) and Colletotrichum siamense (PL3). The isolates were treated with blue, red, green, and yellow light, while white fluorescent light (12 h light/12 h dark photoperiod) and 24 h dark conditions were applied as control. Results revealed that coloured light treatments induced formation of circadian rings, while exposure to white light and dark conditions showed less pronounced circadian rings. Growth and sporulation of endophytes were not significantly influenced by light. By contrast, enzyme production was affected by coloured light treatments, notably with red (amylase), blue (cellulase) and yellow (cellulase, xylanase, L-asparaginase) light, resulting in lower enzyme levels for certain isolates. Under control conditions, enzyme production was relatively higher for amylase, cellulase, xylanase (for cultures incubated in the dark), and for L-asparaginase (for cultures incubated in white fluorescent light). Among the endophytic isolates, F. proliferatum (CCH) showed better response to coloured light treatment as higher sporulation and enzyme production was detected, although growth was significantly suppressed. On the contrary, C. gloeosporiodes (OL3) showed better growth but significantly lower enzyme production and sporulation when treated with the various coloured light. This study revealed that coloured light may have the potential to manipulate growth, sporulation and enzyme production in certain fungal species as strategies for fungal control or for harnessing of valuable enzymes.

Characterization of putative antimicrobial compounds produced by endolichenic Fusarium solani exposed to light treatments

Abstract:The endolichenic Fusarium solani (EF5), known to show induced metabolite production when exposed to red and green lights, was selected for characterization of their putative light-regulated bioactive compounds. To achieve this, fractionation was first performed for crude extracts from cultures of F. solani (EF5) incubated in green, red, white-fluorescent light and dark conditions. The extract yielded 12 (dark condition) to 15 (exposed to green, red, and white-fluorescent lights) fractions, and each of the fractions was tested for antimicrobial activities. The fraction (fraction 5) that showed the most promising antimicrobial activity was then subjected to high-performance liquid chromatography (HPLC) and liquid chromatography–mass spectrometry (LC-MS) to identify the bioactive compounds. Results revealed detection of two new metabolites from endolichenic F. solani , putatively identified as 8-deoxyjavanicin and fusolanone A, which are known to have antimicrobial properties. This study revealed that red and green lights trigger the production of 8-deoxyjavanicin and fusolanone A, which likely contributed to the antimicrobial properties demonstrated by endolichenic F. solani .

The development and nutritional quality of Lyophyllum decastes affected by monochromatic or mixed light provided by light-emitting diode

Abstract:Edible fungi has certain photo-sensitivity during the mushroom emergence stage, but there has been few relevant studies on the responses of Lyophyllum decastes to different light quality. L. decastes were planted in growth chambers with different light qualities that were, respectively, white light (CK), monochromatic red light (R), monochromatic blue light (B), mixed red and blue light (RB), and the mixture of far-red and blue light (FrB). The photo-sensitivity of L. decastes was investigated by analyzing the growth characteristics, nutritional quality, extracellular enzymes as well as the light photoreceptor genes in mushroom exposed to different light treatments. The results showed that R led to mycelium degeneration, fungal skin inactivation and failure of primordial formation in L. decastes. The stipe length, stipe diameter, pileus diameter and the weight of fruiting bodies exposed to RB significantly increased by 8.0, 28.7, 18.3, and 58.2% respectively, compared to the control (p < 0.05). B significantly decreased the stipe length and the weight of fruiting body, with a decrease of 8.5 and 20.2% respectively, compared to the control (p < 0.05). Increased color indicators and deepened simulated color were detected in L. decastes pileus treated with B and FrB in relative to the control. Meanwhile, the expression levels of blue photoreceptor genes such as WC-1, WC-2 and Cry-DASH were significantly up-regulated in mushroom exposed to B and FrB (p < 0.05). Additionally, the contents of crude protein and crude polysaccharide in pileus treated with RB were, respectively, increased by 26.5 and 9.4% compared to the control, while those in stipes increased by 5.3 and 58.8%, respectively. Meanwhile, the activities of extracellular enzyme such as cellulase, hemicellulase, laccase, manganese peroxidase, lignin peroxidase and amylase were significant up-regulated in mushroom subjected to RB (p < 0.05), which may promote the degradation of the culture materials. On the whole, the largest volume and weight as well as the highest contents of nutrients were all detected in L. decastes treated with RB. The study provided a theoretical basis for the regulation of light environment in the industrial production of high quality L. decastes.
Light Irradiation Coupled with Exogenous Metal Ions to Enhance Exopolysaccharide Synthesis from Agaricus Sinodeliciosus ZJU-TP-08 in Liquid Fermentation

Abstract:To promote Agaricus sinodeliciosus var. Chaidam ZJU-TP-08 growth and metabolites accumulation, a novel integrated strategy was developed by adopting high levels of metal ions coupled with light treatment. The results revealed that yellow and blue light could significantly promote biomass and exopolysaccharides production, respectively. Furthermore, the yellow–blue light shift strategy could stimulate exopolysaccharides formation. Ca2+ ions coupled with blue light mostly promoted exopolysaccharides production related to oxidative stress, which was 42.00% and 58.26% higher than that of Ca2+ ions coupled with the non-light and dark cultivation without Ca2+ ions in 5-L bioreactor. RNA-seq was performed to uncover the underlined molecular mechanism regulated by light-induced gene expressions in exopolysaccharides biosynthesis and oxidative stress. The findings of this work provide valuable insights into adopting metal ions coupled with the light-assisted method for the macrofungus submerged fermentation for exopolysaccharides production.
Light and phytochrome PHY control the production of edible fungus Flammulina filiformis by regulating the morphogenesis of fruiting bodies and l-lysine accumulation

Abstract:Flammulina filiformis , a representative umbelliferous fungus, has a long stipe and high l -lysine content, thus is widely cultivated and consumed. Currently, there is a lack of theoretical guidance on how to better use light to cultivate edible fungi without photosynthesis such as F. filiformis in industrialized cultivation. Previous studies have found that blue light can affect the yield and l -lysine content of F. filiformis . The primary focus of this work was the phytochrome PHY in the light signaling pathway and its role in F. filiformis production. Unlike plants in which the expression of PHY was activated by only red light, it was found that different visible lights (including red, blue, green, and white light) can stimulate the up-regulation of FfPhy transcript levels. Throughout the developmental stages of F. filiformis , the transcript level of FfPhy was significantly up-regulated during the formation of fruiting body and in the stipe in the elongation stage. Further, FfPhy knockdown strain showed the markedly shorter stipe length than WT, resulting in a significantly reduced yield. RNA-Seq analysis showed that the most genes in MAPK signaling pathway and its downstream regulatory processes, mainly focusing on cell division and cell wall remodeling, were down-regulated after FfPhy knockdown. It suggested that FfPhy regulates the fruiting body elongation through acting on cell division and cell wall remodeling, thereby affecting the morphological development of the stipe rather than the pileus. Interestingly, FfPhy knockdown also inhibits the accumulation of l -lysine content by promoting l -lysine degradation instead of inhibiting l -lysine biosynthesis, indicating that its influence extends to metabolic processes related to l -lysine metabolism. These findings provide new insights into photobiological effect of FfPhy in macrofungus F. filiformis , and have potential guiding significance for cultivation and breeding to increase mushroom yield and l -lysine content.

Different wavelengths of LED irradiation promote secondary metabolite production in Pycnoporus sanguineus for antioxidant and immunomodulatory applications

Abstract:Pycnoporus sanguineus is a fungus of the phylum Basidiomycota that has many applications in traditional medicine, modern pharmaceuticals, and agricultural industries. Light plays an essential role in the metabolism, growth, and development of fungi. This study evaluated the mycelial growth and antioxidant and anti-inflammatory activities in P. sanguineus fermentation broth (PFB) cultured under different wavelengths of LED irradiation or in the dark. Compared to the dark cultures, the dry weight of mycelia in red- and yellow-light cultures decreased by 37 and 35% and the yields of pigments increased by 30.92 ± 2.18 mg and 31.75 ± 3.06 mg, respectively. Compared with the dark culture, the DPPH free radical scavenging ability, ABTS+ free radical scavenging capacity, and reducing power of yellow-light cultures increased significantly, and their total phenolic content peaked at 180.0 ± 8.34 μg/mL. However, the reducing power in blue-light cultures was significantly reduced, though the total phenol content did not vary with that of dark cultures. In LPS- and IFN-γ-stimulated RAW 264.7 cells, nitrite release was significantly reduced in the red and yellow light-irradiated PFB compared with the dark culture. In the dark, yellow-, and green-light cultures, TNF-α production in the inflamed RAW 264.7 cells was inhibited by 62, 46, and 14%, respectively. With red-, blue-, and white-light irradiation, TNF-α production was significantly enhanced. Based on these results, we propose that by adjusting the wavelength of the light source during culture, one can effectively modulate the growth, development, and metabolism of P. sanguineus.

Abstract:Light is an important factor for the growth of many forms of life, including mushrooms. This paper highlights the effects of the different wavelengths (red, 650 nm; blue, 450 nm; green, 525 nm) of light emitting diodes (LEDs) on the growth, biomass production and antioxidant properties of Ganoderma lucidum. G. lucidum is a white-rot, wood-degrading fungus in the Basiodiomycota that typically grows on logs. Mycelia were grown on coconut water agar (CWA) solid media for analysis of mycelial colony diameter as well as coconut water (CW) submerged culture for analysis of mycelial biomass weight. Both set-ups were incubated in variously-colored LED chambers. The DPPH radical scavenging activity and total phenolic content of the harvested mycelia were also determined. After three days of incubation, G. lucidum mycelia under red LED recorded the highest mean mycelial colony diameter of 72.50 mm. In terms of mycelia biomass production, G. lucidum mycelia exposed in red LED and dark produced the heaviest weight. On the other hand, mycelia grown under green LED had the highest radical scavenging activity of 66.49% while those harvested from red LED showed the highest total phenolic content of 81.29 mg GAE / g of sample. Our results demonstrate that LED color influences the mycelial growth, biomass production and antioxidant activities of G. lucidum.
Differential Influence of Supplements on Lignocellulolytic Enzyme Activities, Growth and Production of Pleurotus Ostreatus

Abstract:Lignocellulolytic enzymes are extracellular biocatalysts secreted by filamentous fungi and are involved in the breakdown of recalcitrant lignocellulose plant materials into useful products necessary for fungal growth and development. Even though several studies on filamentous fungi have reported the impact of different substrates on lignocellulolytic enzymes, there is limited information on how mushroom supplements affect secretion of the enzymes, growth, and yield of Pleurotus ostreatus when using the alkaline treatment method. In this study, we investigated the influence of cornmeal (T1) and coffee grounds (T2) supplements on lignocellulosic enzymes at different growth stages and the production of P. ostreatus. We found that lignolytic enzyme activity was significantly higher in the control (CK) and T2 during mycelial stages, while CK had the lowest hydrolytic enzyme activity during primordia and fruiting. Unlike T1 which had the best biological efficiency, T2 exhibited significantly higher levels of lignolytic enzymes during mycelial stage, whereas CMCase and xylanase activities were higher in supplemented treatments than in the control during primordia and fruiting. Taken together our results demonstrated that cornmeal and coffee ground supplements reduce mycelium growth rate, enhance the production of hydrolytic enzymes during fruiting, and remarkably increase the yield and protein content of P. ostreatus.
Light quality regulation of growth and endogenous IAA metabolism of Ganoderma lucidum mycelium

Abstract:To study the effect and mechanism of the light quality acting on Ganoderma lucidum, and provide a theoretical basis for G. lucidum mycelium cultivation, we focused on growth and endogenous IAA metabolism of G. lucidum mycelium under different light-emitting diode (LED) condition. The growth index, endogenous levels of IAA and Enzymes related to IAA metabolism and Polysaccharides content were investigated in different growth periods. Results showed that blue light irradiation was the best from the viewpoint of steady growth and polysaccharides accumulation, red light irradiation improved endogenous IAA level and promoted growth of mycelium in early stage of cultivation, green light irradiation decreased growth rate and fresh weight of mycelium, but increased drying rate. Enzymes related to IAA metabolism also significantly influenced by light quality. The activity of indole acetic acid oxidase (IAAO), peroxidase (POD) and tryptophan synthetase with blue light irradiation were showed high level in early time, but decreased later, and the IAA content was consistently at lower level than that in other treatments, while mycelium irradiated with yellow light showed the highest activity of both IAAO and tryptophan synthetase, and medium level of IAA content. In conclusion, the light quality affects growth and regulation of the level of endogenous IAA of G. lucidum mycelium.

Abstract:Light represents a major carrier of information in nature. The molecular machineries translating its electromagnetic energy (photons) into the chemical language of cells transmit vital signals for adjustment of virtually every living organism to its habitat. Fungi react to illumination in various ways, and we found that they initiate considerable adaptations in their metabolic pathways upon growth in light or after perception of a light pulse. Alterations in response to light have predominantly been observed in carotenoid metabolism, polysaccharide and carbohydrate metabolism, fatty acid metabolism, nucleotide and nucleoside metabolism, and in regulation of production of secondary metabolites. Transcription of genes is initiated within minutes, abundance and activity of metabolic enzymes are adjusted, and subsequently, levels of metabolites are altered to cope with the harmful effects of light or to prepare for reproduction, which is dependent on light in many cases. This review aims to give an overview on metabolic pathways impacted by light and to illustrate the physiological significance of light for fungi. We provide a basis for assessment whether a given metabolic pathway might be subject to regulation by light and how these properties can be exploited for improvement of biotechnological processes.

Photoregulation of the biosynthetic activity of the edible medicinal mushroom Lentinula edodes in vitro

Abstract:The findings of the study demonstrate the impact of low-intensity laser and quasi-monochromatic light on the biosynthetic activity of the edible medicinal fungus L. edodes during submerged cultivation. An artificial lighting installation based on matrices of light-emitting diodes (LED) emitting light at 470 nm (blue), 530 nm (green), 650 nm (red), and argon gas laser (488 nm) was used. Irradiation with blue and red LED and laser led to a shortening of the lag phase by 2 days and an increase in the mycelial mass. Irradiation with laser light resulted in the highest mycelial mass yield (14.1 g/L) on the 8th day of cultivation. Irradiation in all used wavelength ranges caused an increase in the synthesis of both extracellular and intracellular polysaccharides. Laser light at 488 nm and LED at 470 nm proved to be the most effective. Irradiation with red, green, and blue laser light caused an increase in the total amount of fatty acids in the mycelial mass compared to the control. A significant distinction in qualitative composition was observed: short-chain acids C6‒C12 compounds were produced under red light irradiation, whereas long-chain C20‒C24 were formed under green light irradiation. The most significant changes in the aromatic profile of the mycelial mass and culture liquid were recorded upon irradiation with green light. The content of aromatic components increased 24.6 times in the mycelial mass and 38.5 times in the culture liquid. The results suggest the possibility of using low-intensity quasi-monochromatic light for targeted regulation of L. edodes biosynthetic activity.Graphical Abstract

Inhibition of Aspergillus Oryzae Mycelium Growth and Conidium Production by Irradiation with Light at Different Wavelengths and Intensities

Abstract:Aspergillus oryzae is a safe filamentous fungus widely used in the food, medicine, and feed industries, but there is currently not enough research on the light response of A. oryzae. In this study, 12 different light conditions were set and A. oryzae GDMCC 3.31 was continuously irradiated for 72 h to investigate the effect of light on mycelial growth and conidium production. Specifically, each light condition was the combination of one light wavelength (475, 520, or 630 nm) and one light intensity (20, 40, 60, or 80 mu mol photon m(-2) s(-1)). The results show that mycelium growth was inhibited significantly by green light (wavelength of 520 nm and intensities of 20 and 60 mu mol photon m(-2) s(-1)) and blue light (wavelength of 475 nm and intensity of 80 mu mol photon m(-2) s(-1)). The production of conidia was suppressed only by blue light (wavelength of 475 nm and intensities of 40, 60, and 80 mu mol photon m(-2) s(-1)), and those levels of inhibition increased when the intensity of blue light increased. When the strain was irradiated by blue light (80 mu mol photon m(-2) s(-1)), the number of conidia was 57.4% less than that of the darkness group. However, within our set range of light intensities, A. oryzae GDMCC 3.31 was insensitive to red light (wavelength of 630 nm) in terms of mycelium growth and conidium production. Moreover, interaction effects between light wavelength and intensity were found to exist in terms of colony diameter and the number of conidia. This research investigated the light response of A. oryzae, which may provide a new method to regulate mixed strains in fermented foods by light. IMPORTANCE Studies on the monochromatic light response of Aspergillus nidulans and Neurospora crassa have gone deep into the molecular mechanism. However, research methods for the light response of A. oryzae remain in the use of white light sources. In this study, we first demonstrated that A. oryzae GDMCC 3.31 was sensitive to light wavelength and intensity. We have observed that blue light inhibited its growth and sporulation and the inhibitory effect increased with intensity. This research not only adds new content to the study of the photoreaction of Aspergillus but also brings new possibilities for the use of light to regulate mixed strains and ultimately improve the flavor quality of fermented foods.
Co-cultured Production of Lignin-Modifying Enzymes with White-Rot Fungi

Abstract:Co-cultivation was a potential strategy in lignocellulolytic biodegradation with producing high activity enzymes due to their synergistic action. The objective of this study was to investigate the rarely understood effects of co-culturing of two white-rot fungi on lignin-modifying enzymes (LMEs) production. Six species, Bjerkandera adusta, Phlebia radiata, Pleurotus ostreatus, Dichomitus squalens, Hypoxylon fragiforme and Pleurotus eryngii, were cultured in pairs to study the production of LMEs. The paired hyphal interaction observed showed that P. eryngii is not suitable for co-growth. The use of agar plates containing dye RBBR showed elevated decolourisation at the confrontation zone between mycelia. Laccase was significantly stimulated only in the co-culture of P. radiata with D. squalens under submerged cultivation; the highest value was measured after 4 days of incubation (120 U mg−1). The improved productions of MnP and LiP were simultaneously observed at the co-culture of P. ostreatus and P. radiata (MnP = 800 nkat L−1 after 4 days of incubation; LiP = 60 nkat L−1 after 7 days of incubation), though it was not a good producer of laccase. P. ostreatus appeared to possess specific potential to be used in co-cultured production of LMEs. The phenotype of LMEs production was not only dependent on the species used but also regulated by different nutritions available in the culture medium. The present data will provide evidence for illustrating the regulatory roles of C/N on LMEs production under the co-cultures’ circumstances.
The influence of monochromatic lights on flavonoid production by the fungus Sanghuangporus vaninii: Modeling of kinetic profiles and expression levels of important genes in flavonoid synthesis

Abstract:<p>Four different monochromatic lights (blue, red, yellow and green) were used to study their separate influence on the growth, morphology and flavonoid accumulation by <em>S. vaninii</em> compared to under white. The results showed that the accumulation rate of total flavonoids under blue light was faster, while the total flavonoid accumulation rate under red light was slower than under white light, and other monochromatic lights had little effect on the synthesis of total flavonoids. The kinetic model parameters and fitting values are basically consistent with the experimental values. Through the development of kinetic model and model validation, the maximum flavonoid concentration under blue light increased by 25.2 % compared with that under white. The relative expression of phenylalanine ammonia lyase (PAL), 4-coumaric acid coenzyme A ligase (4CL) and flavonoid-3', 5'-hydroxylase (F3', 5'H) genes were significantly up-regulated under blue light. The influence of lights on the fungal flavonoid production was in part in line with the key genes expression involved.</p>

Red and far-red light improve the antagonistic ability of Trichoderma guizhouense against phytopathogenic fungi by promoting phytochrome-dependent aerial hyphal growth

Abstract:Light as a source of information regulates morphological and physiological processes of fungi, including development, primary and secondary metabolism, or the circadian rhythm. Light signaling in fungi depends on photoreceptors and downstream components that amplify the signal to govern the expression of an array of genes. Here, we investigated the effects of red and far-red light in the mycoparasite Trichoderma guizhouense on its mycoparasitic potential. We show that the invasion strategy of T . guizhouense depends on the attacked species and that red and far-red light increased aerial hyphal growth and led to faster overgrowth or invasion of the colonies. Molecular experiments and transcriptome analyses revealed that red and far-red light are sensed by phytochrome FPH1 and further transmitted by the downstream MAPK HOG pathway and the bZIP transcription factor ATF1. Overexpression of the red- and far-red light-induced fluffy gene fluG in the dark resulted in abundant aerial hyphae formation and thereby improvement of its antagonistic ability against phytopathogenic fungi. Hence, light-induced fluG expression is important for the mycoparasitic interaction. The increased aggressiveness of fluG -overexpressing strains was phenocopied by four random mutants obtained after UV mutagenesis. Therefore, aerial hyphae formation appears to be a trait for the antagonistic potential of T . guizhouense . Light controls a large proportion of the genome. While Trichoderma species encode the red-light receptor phytochrome, a detailed study of red-light responses and signaling was lacking. Here, we show that the mycoparasitic fungus Trichoderma guizhouense uses distinct strategies to attack phytopathogenic fungi. It can overgrow or invade the colonies of the attacked species. Red and far-red light promoted aerial hyphal growth and thereby increased both, overgrowth or invasion of the host colonies. The effect required the phytochrome-signaling pathway. We further show that screening for mutants producing more aerial hyphae proves to be an easy way to develop strains with improved biocontrol performance.

Abstract:SummaryPhytochromes (Phy) in filamentous fungi are Group VIII histidine kinases that share a unique N‐terminal photosensory core, but their functions are largely unknown. Here we show that Beauveria bassiana Phy (Bbphy) is functionally vital for growth, conidiation and multistress tolerance of the fungal entomopathogen lacking sexual stage. Colony growth of ΔBbphy was significantly slower in a nutrition‐rich medium but faster in several minimal media. Conidial yield of ΔBbphy in the rich medium increased at the fitted rate of 3.4 × 107 conidia h−1 white light in the light/dark cycles of 0:24 to 16:8 h, decreased greatly in the short‐, long‐ and full‐day cycles of red/far‐red light, but was unaffected under full‐day blue light. Moreover, ΔBbphy showed higher osmosensitivity, increased antioxidant capability, and decreased conidial thermotolerance and UV‐B resistance, accompanied with downregulation of Hog1 phosphorylation and of four Hog1‐related genes under osmotic stress, and upregulation of five superoxide dismutases and four catalases under oxidative stress. All the changes were restored by the gene complementation. Taken together, Bbphy controls conidiation by responding to daylight length and red/far‐red light and regulates multistress responses perhaps because of an involvement in Hog1 pathway. Our findings highlight diverse functions of Bbphy in B. bassiana.
Effect of different light of LED light quality on growth and antioxidant enzyme activities of Ganoderma lucidum

Abstract:Objective: To study the effect of light quality on growth, antioxidant enzyme activities of Ganoderma lucidum mycelium. Method: G. lucidum mycelium was cultured under different light qualities by light emitting diodes(LED). The growth G. lucidum mycelium was observed and antioxidant enzyme activities was determined in different growth periods. Result: Under the red LED, the blue LED and dark condition(CK), the mycelium grew faster than that under other light qualities. The white LED resulted in a largest increase in the amount of the mycelium and always kept the activities of CAT high level. Major fluctuations of POD activities emerged under the green LED, while enhanced severely in the late phase. Under the yellow LED, the activities of SOD appeared high level. However, SOD activities on dark(CK) raised obviously in late period. At the late stage, the content of mycelium polysaccharides was significant higher than that under the blue LED. Conclusion: The light quality could influence the growth and metabolism of G. lucidum mycelium.
Effect of LED Blue Light on Penicillium digitatum and Penicillium italicum Strains

Abstract:Studies on the antimicrobial properties of light have considerably increased due in part to the development of resistance to actual control methods. This study investigates the potential of light-emitting diodes (LED) blue light for controlling Penicillium digitatum and Penicillium italicum. These fungi are the most devastating postharvest pathogens of citrus fruit and cause important losses due to contaminations and the development of resistant strains against fungicides. The effect of different periods and quantum fluxes, delaying light application on the growth and morphology of P. digitatum strains resistant and sensitive to fungicides, and P. italicum cultured at 20°C was examined. Results showed that blue light controls the growth of all strains and that its efficacy increases with the quantum flux. Spore germination was always avoided by exposing the cultures to high quantum flux (700 μmol m(-2) s(-1) ) for 18 h. Continuous light had an important impact on the fungus morphology and a fungicidal effect when applied at a lower quantum flux (120 μmol m(-2) s(-1) ) to a growing fungus. Sensitivity to light increased with mycelium age. Results show that blue light may be a tool for P. digitatum and P. italicum infection prevention during handling of citrus fruits.
Intensity of Supplemental Greenhouse Lighting Affects Strawberry Plant Growth, Anthracnose Infection Response, and Colletotrichum Pathogen Development in Culture

Abstract:The effect of supplemental lighting on strawberry growth and anthracnose disease response of three strawberry ( Fragaria × ananassa ) cultivars was evaluated in two greenhouse trials, and the effect on strawberry anthracnose pathogens ( Colletotrichum sp.) was evaluated in the laboratory. The objective of the greenhouse trials was to determine the effect of various intensities of the red and blue light emitting diode (LED) light treatment on strawberry plant vigor, injury, and disease development. In these trials, the duration of supplemental light treatments was split into two 4-hour periods: dawn and dusk. The intensity of the red and blue LED bulbs was set using an adjustable dial at 1 or 3 in trial 1 and at 1, 5, or 10 in trial 2. Illuminance and photosynthetic photon flux densities of the light treatments ranged from lows of 402 lx and 5 μmol⋅m –2 ⋅s –1 (blue LED 1) to highs of 575 lx and 25 μmol⋅m –2 ⋅s –1 (red LED 1 + blue LED 3) in trial 1, and from lows of 4213 lx and 81 μmol⋅m –2 ⋅s –1 (red LED 1) to highs of 7051 lx (red LED 5) and 194 μmol⋅m –2 ⋅s –1 (red LED 10) in trial 2. Lower light intensities in trial 1 resulted in no significant differences as a result of light treatments in relative chlorophyll content, plant vigor ratings, or disease severity ratings (DSRs). However, plant injury ratings were significantly greater in plants in the wide-spectrum fluorescent (WSF) plus ultraviolet B (UVB) light treatment compared with the other treatments. Under the higher light intensities in trial 2, there were more significant effects among light treatments. Relative chlorophyll content of plants in the WSF + UVB, WSF, and red LED 1 treatments was significantly greater than that of plants in the red LED 10 treatment; however, plants in the red LED 10 treatment had the greatest injury ratings. Detached leaves from plants in the red 5 LED and red 10 LED treatments inoculated with Colletotrichum gloeosporioides received the greatest DSRs, and leaves from plants in the red LED 1 and WSF treatments received the lowest DSRs. In the laboratory, five days of exposure to supplemental lights did not prevent the growth of isolates of three species of Colletotrichum pathogens even though the intensity of the LED lights was set at their highest intensity. However, growth of isolates exposed to the WSF + UVB light treatment was slowed.

Abstract:We report the long-term survival and functional outcome of the Birmingham Hip Resurfacing (BHR) in patients aged < 50 years at operation, and explore the factors affecting survival. Between 1997 and 2006, a total of 447 BHRs were implanted in 393 patients (mean age 41.5 years (14.9 to 49.9)) by one designing surgeon. The mean follow-up was 10.1 years (5.2 to 14.7), with no loss to follow-up. In all, 16 hips (3.6%) in 15 patients were revised, giving an overall cumulative survival of 96.3% (95% confidence interval (CI) 93.7 to 98.3) at ten years and 94.1% (95% CI 84.9 to 97.3) at 14 years. Using aseptic revision as the endpoint, the survival for men with primary osteoarthritis (n = 195) was 100% (95% CI 100 to 100) at both ten years and 14 years, and in women with primary osteoarthritis (n = 109) it was 96.1% (95% CI 90.1 to 99.9) at ten years and 91.2% (95% CI 68.6 to 98.7) at 14 years. Female gender (p = 0.047) and decreasing femoral head size (p = 0.044) were significantly associated with an increased risk of revision. The median Oxford hip score (OHS, modified as a percentage with 100% indicating worst outcome) at last follow-up was 4.2% (46 of 48; interquartile range (IQR) 0% to 24%) and the median University of California, Los Angeles (UCLA) score was 6.0 (IQR 5 to 8). Men had significantly better OHS (p = 0.02) and UCLA scores (p = 0.01) than women. The BHR provides excellent survival and functional results in men into the second decade, with good results achieved in appropriately selected women.
