Abstract:The inhibitory effects of zinc oxide nanoparticles (ZnO NPs) and impacts of N-acyl-homoserine lactone (AHL)-based quorum sensing (QS) on biological nitrogen removal (BNR) performance have been well-investigated. However, the effects of ammonia nitrogen (NH4+-N) concentrations on NP toxicity and AHL regulation have seldom been addressed yet. This study consulted on the impacts of ZnO NPs on BNR systems when high NH4+-N concentration was available. The synergistic toxic effects of high-strength NH4+-N (200 mg/L) and ZnO NPs resulted in decreased ammonia oxidation rates and dropped the nitrogen removal efficiencies by 17.5% ± 0.2%. The increased extracellular polymeric substances (EPS) production was observed in response to the high NH4+-N and ZnO NP stress, which indicated the defense mechanism against the toxic effects in the BNR systems was stimulated. Furthermore, the regulatory effects of exogenous N-decanoyl-homoserine lactone (C10-HSL)-mediated QS system on NP-stressed BNR systems were revealed to improve the BNR performance under different NH4+-N concentrations. The C10-HSL regulated the intracellular reactive oxygen species levels, denitrification functional enzyme activities, and antioxidant enzyme activities, respectively. This probably synergistically enhanced the defense mechanism against NP toxicity. However, compared to the low NH4+-N concentration of 60 mg/L, the efficacy of C10-HSL was inhibited at high NH4+-N levels of 200 mg/L. The findings provided the significant application potential of QS system for BNR when facing toxic compound shock threats.
Pluripotency of endogenous AHL-mediated quorum sensing in adaptation and recovery of biological nitrogen removal system under ZnO nanoparticle long-term exposure

Abstract:The impacts of quorum sensing (QS) on nanoparticle (NP)-stressed biological nitrogen removal (BNR) system have seldom been addressed yet. In this study, the contributions of endogenous N -acyl-homoserine lactone (AHL)-based QS regulation to the BNR system's adaptation to the zinc oxide (ZnO) NP stress and its recovery potential were systematically investigated. Although 1 mg/L ZnO NPs exerted little impact on the BNR system, chronic exposure to 10 mg/L ones depressed the system's BNR performance which irreversibly impaired the nitrification process even when the system entered the recovery period with no NP added anymore. Meanwhile, ZnO NPs exhibited hormesis effects on the production of AHLs and extracellular polymeric substance (EPS), and activities of superoxide dismutase and catalase. During the ZnO NP exposure period, C 4 -HSL, C 6 -HSL, and C 10 -HSL were discovered to be positively associated with nitrogen removal efficiency, tightly-bound EPS production, and antioxidase activities. Besides, the shifts of Nitrospira , Dechloromonas , Aeromonas , Acinetobacter , Delftia , and Bosea were expected to determine the AHL's dynamic distribution. During the system's recovery stage, Dechloromonas replaced Candidatus_Competibacter as the dominant denitrification-related genus. Dechloromonas abundance elevated with the increased contents of C 4 -HSL in the aqueous and EPS phases and C 10 -HSL in EPS and sludge phases, and were expected to promote the activities of BNR-related and antioxidant enzymes, and the EPS production to assist in the recovery of the impaired system's BNR performance. The QS-related BNR genera exhibited higher resilience to ZnO NPs than quorum quenching-related ones, indicating their critical role in nitrogen removal in the restored system. This work provided an insight into the potential pluripotency of AHL-based QS regulation on the ZnO NP-stressed BNR system's adaptation and recovery.

Abstract:The effects and mechanisms of three N -acyl-homoserine lactones (AHLs) (C 4 -HSL, C 6 -HSL, and C 10 -HSL) on responses of biological nitrogen removal (BNR) systems to zinc oxide nanoparticle (NP) shock were investigated. All three AHLs improved the NP-impaired ammonia oxidation rates by up to 50.0 % but inhibited the denitrification process via regulating nitrogen metabolism-related enzyme activities. C 4 -HSL accelerated the catalase activity by 13.2 %, while C 6 -HSL and C 10 -HSL promoted the superoxide dismutase activity by 26.6 % and 18.4 %, respectively, to reduce reactive oxygen species levels. Besides, the enhancements of tryptophan protein and humic acid levels in tightly-bound extracellular polymeric substance by AHLs were vital for NP toxicity attenuation. The metabonomic analysis demonstrated that all three AHLs up-regulated the levels of lipid- and antioxidation-related metabolites to advance the system's resistance to NP shock. The "dual character" of AHLs emphasized the concernment of legitimately employing AHLs to alleviate NP stress for BNR systems.

High Concentrations of Dissolved Organic Nitrogen and N-nitrosodimethylamine Precursors in Effluent from Biological Nutrient Removal Process with Low Dissolved Oxygen Conditions.

Abstract:The presence of dissolved organic nitrogen (DON) in biological nutrient removal (BNR) effluent has led to increased concern about its adverse effects on wastewater discharge and reuse applications. Previous studies have demonstrated efficient biological inorganic nitrogen removal in BNR under low dissolved oxygen (DO) conditions; however, information on DON is scarce. This study investigated low-DO effects on DON and N-nitrosodimethylamine (NDMA) precursor concentrations in BNR effluents. Identical BNR reactors consisting of an external real-time DO intelligent control system were operated at three different DO concentrations (0.3, 1.0, and 4.0 mgO2/L). Surprisingly, significantly higher values of effluent DON (p<0.05, t-test) and NDMA precursors (p<0.01, t-test) were observed at lower DO levels. Ultrahigh-resolution mass spectrometry analysis showed that molecules produced by microbes at low-DO levels exhibited high proteins/amino sugars-like and low normal oxidation state of carbon characteristics, which possibly acted critical roles in NDMA formation. Furthermore, path analysis by partial least-squares path modeling suggested that NDMA formation potential had strong associations with microbe-DON network stability of microbe-DON co-occurrence interactions (r=0.979, p<0.01). These results highlight the necessity of reconsidering the feasibility of BNR systems operating at low-DO concentrations considering the adverse effects of DON on wastewater discharge and reuse applications.
Effect of dissolved oxygen on high C/N wastewater treatment in moving bed biofilm reactors based on heterotrophic nitrification and aerobic denitrification: Nitrogen removal performance and potential mechanisms

Abstract:In this study, it was investigated the nitrogen removal (NR) performance and potential mechanism for high C/N wastewater treatment under different dissolved oxygen (DO) concentrations. The results showed that DO concentration significantly affected the removal efficiency of total nitrogen (TN). When the initial DO increased from 0.5 to 1.5 mg/L, TN removal efficiency significantly increased from 65 % to 85 %. However, a further DO increase did not promote TN removal, and the NR was only 80 % with an initial DO concentration of 3.5 mg/L. The effect of DO concentration on NR was influenced by the combined action of functional bacteria and electron flow. Excessive DO concentration did not positively affect NR efficiency but promoted electron utilization and respiratory proliferation. When the DO concentration was 1.5 mg/L, more electrons generated by sodium acetate metabolism were transferred to the aerobic denitrification process, compared to when the DO concentration was 3.5 mg/L.

Abstract:The unique properties and growing usage of zinc oxide nanoparticles increase their release in municipal wastewater treatment plants. Therefore, these nanoparticles, by interacting with microorganisms, can fail the suitable functioning of biological systems in treatment plants. For this reason, research into the toxicity of ZnO is urgent. In the present study, the toxicity mechanism of ZnO-NPs towards microbial communities central to granular activated sludge (GAS) performance was assessed over 120-day exposure. The results demonstrate that the biotoxicity of ZnO-NPs is dependent upon its dosage, exposure time, and the extent of reactive oxygen species (ROS) production. Furthermore, GAS performance and the extracellular polymeric substances (EPS) content were significantly reduced at 50 mg/L ZnO-NPs. This exposure led to decreases in the activity of ammonia monooxygenase (25.2%) and nitrate reductase (11.9%) activity. The Field emission scanning electron microscopy images confirmed that ZnO-NPs were able to disrupt the cell membrane integrity and lead to cell/bacterial death via intracellular ROS generation which was confirmed by the Confocal Laser Scanning Microscopy analysis. After exposure to the NPs, the bacterial community composition shifted to one dominated by Gram-positive bacteria. The results of this study could help to develop environmental standards and regulations for NPs applications and emissions.

Regulation of membrane fixation and energy production/conversion for adaptation and recovery of ZnO nanoparticle impacted Nitrosomonas europaea

Abstract:The ZnO nanoparticle (NP) effects on typical ammonia-oxidizing bacteria, Nitrosomonas europaea in a chemostat bioreactor, and the cells' toxicity adaptation and recovery potentials were explored. Hardly any inhibition was observed when the NP concentration was high up to 10 mg/L. The cells exposed to 50 mg/L ZnO NPs displayed time-dependent impairment and recovery potentials in terms of cell density, membrane integrity, nitrite production rate, and ammonia monooxygenase activity. The 6-h NP stress impaired cells were nearly completely restored during a 12-h recovery incubation, while the longer exposure time would cause irretrievable cell damage. Microarray analysis further indicated the transcriptional adaptation of N. europaea to NP stress. The regulations of genes encoding for membrane permeability or osmoprotectant, membrane integrity preservation, and inorganic ion transport during NP exposure and cell recovery revealed the importance of membrane fixation and the associated metabolisms for cells' self-protection and the following recovery from NP stress. The oxidative phosphorylation, carbon assimilation, and tricarboxylic acid (TCA) cycling pathways involved in the cells' antitoxicity activities and would promote the energy production/conversion efficiency for cell recovery. The heavy metal resistance, histidine metabolism, toxin-antitoxin defense, glycolysis, and sulfate reduction pathways were also suggested to participate in the cell detoxication and recovery processes. All these findings provided valuable insights into the mechanisms of cell-mediated ZnO NP cytotoxicity and their potential impacts on wastewater nitrogen removal system.
Transient Disturbance of Engineered ZnO Nanoparticles Enhances the Resistance and Resilience of Anammox Process in Wastewater Treatment

Abstract:The increasing use of engineered nanoparticles (NPs) in consumer and industrial products raises concerns about their environmental impacts, but their potential influence on anaerobic ammonium oxidation (anammox) process in wastewater treatment remains unknown. In this study, the response of granule-based anammox reactor to different loads of ZnONPs was investigated. The introduction of 1-5mgL-1 ZnONPs did not affect reactor performance, but 90% of the nitrogen removal capacity was deprived by a shock of 10mgL-1 ZnONPs within 3days. Anammox activity was significantly inhibited, but no significant stimulation of intracellular reactive oxygen species (ROS) production or extracellular lactate dehydrogenase (LDH) activity was observed. The inhibition was thus mainly due to the accumulation of toxic Zn(II) ions in anammox biomass. However, the resistance and resilience of this anammox reactor to ZnONPs were enhanced by intermittent perturbations in the mode of "shock-recovery". The up-regulated abundance of Zn(II)-exporter ZntA might contribute to the enhanced resistance. In addition, these repeated transient disturbances improved the functional specificity of the anammox community despite the reduction of its diversity. Overall, these results may provide useful references for evaluating and controlling the risk of NPs to anammox process.

Abstract:The main purposes of this study were to investigate the effects of different dissolved oxygen (DO) conditions (0.3 and 1.0 mg/L) on the microbial interactions in the partial nitrification/anammox (PN/A) process. The results indicated that the high concentration of DO (1.0 mg/L) significantly inhibited the activity of anammox bacteria (AnAOB) while enhanced the activity of nitrite oxidation bacteria (NOB) in the PN/A reactor. 1.0 mg/L DO promoted the abundance of NOB (such as Nitrospira ) and some heterotrophic microorganisms (e.g., SBR1031 and OLB13), while decreased ammonia oxidizing bacteria (AOB, e.g., Nitrosomonas ) and AnAOB (Ca. Brocadia and Ca. Kuenenia ). 1.0 mg/L DO concentration driven a larger and more complex network with increased percentage of positive links, which might be a strategy for microbes to resist the adverse environment. Furthermore, the subnetworks of anammox bacteria and Nitrosomonas became smaller and simpler, whilst the subnetworks of heterotrophic bacteria (SBR1031) were more complex and connected. The decreased activity and abundance of AnAOB, together with the shifts of microbial interactions might result in the deterioration of nitrogen removal under 1.0 mg/L DO. This study provides a relatively comprehensive perspective for us to understand the effects of DO on the microbial interactions in the PN/A system.

Abstract:Oxygen-based membrane biofilm reactor (O 2 -MBfR) can achieve bubble-free efficient linear alkylbenzene sulfonate (LAS) mineralization. However, effluent microbially derived dissolved organic nitrogen (mDON) reduces recycled water quality and induces water eutrophication. How dissolved oxygen (DO) impacts the dynamics of mDON during LAS biodegradation is unknown. Here, we explored the fate of mDON releasing, molecular characteristics and formation metabolisms in the O 2 -MBfR responding to DO dynamics using Fourier transform ion cyclotron resonance mass spectrometry and metagenomic analyses. Results showed that DO presents a negative impact (λ = -0.242, P < 0.001) on mDON production, but can positively influence mDON reduction through influencing total organic nitrogen (TDN) removal. DO of 0.3 mg/L led to the lowest concentration of mDON with the least number of organic molecular formulas and high proportion of stable N2– and N3-containing organics. Much lower (0.0 mg/L) or higher (0.6 mg/L) DO concentrations favored dissimilatory-based ammonia formation and assimilatory nitrogen fixation that enhancing mDON formation. Firmocutes were the dominant bacteria that contributing to mDON formation. TDN (λ = 0.844, P < 0.001) and pH (λ = 0.248, P < 0.01) directly affected the effluent mDON concentration. The relatively low effluent mDON under anoxic condition suggested that DO controlling can be an effective strategy to minimize bioavailable mDON discharges in the O 2 -MBfR, which showed great implication potential for high-quality reclaimed water production and eutrophication control.

Abstract:The nitrogen transformation performances and greenhouse gas nitrous oxide (N2O) emissions in a sequencing batch reactor under chronic exposure to zinc oxide nanoparticles (ZnO NPs) were quantified and the system's self-recovery potentials were assessed. ZnO NPs posed a dose-dependent depression effect on the removal efficiencies of ammonia nitrogen (NH4+-N) and total nitrogen (TN), and the N2O emissions. The suppressed N2O emissions had a positive relationship with the activity ratios of nitrite/NO reductases and N2O reductase, and were expected to be caused by the inhibited heterotrophic denitrification process. The inhibition of glucose metabolism key enzymes and electron transport chain activities would be responsible for the heterotrophic denitrification performances deterioration. Furthermore, the removal efficiencies of NH4+-N and TN were recovered to control levels through the nitrite-shunt. However, the N2O emission increased significantly above the control during the recovery period mainly due to the irreversibility of the depressed nitrite oxidation activities.

Abstract:For many engineered nanoparticles, the primary pathway of release into the environment is via sewage and industrial wastewater discharges. In this work, the removal of uncoated ZnO nanoparticles (ZnO NPs) during simulated wastewater treatment processes and its impact on treatment performance were examined. Simulated primary clarification removed the majority (about 70%) of the dosed ZnO NPs. During simulated sequencing batch reactor (SBR) processes, ZnO NPs were completely removed in each cycle throughout the 11-day experimental duration (two cycles per day). Continuous input of ZnO NPs into the wastewater (at concentrations up to 5 mg L-1) did not reduce chemical oxygen demand (COD) removal. NH4+-N removal was reduced at a dosing concentration of 5 mg L-1 ZnO NPs per cycle. Inhibition of respiration of nitrifying microorganisms by ZnO NPs corroborated the reduction of NH4+-N removal. These results indicate that if the wastewater is treated, the release of ZnO NPs into receiving water bodies would be minimal and ZnO NPs would mainly accumulate in biosolids. Uncoated ZnO NPs in wastewater at very high concentrations may have some adverse effects on activated sludge process.
Response of Anammox Granules to ZnO Nanoparticles at Ambient Temperature

Abstract:The effects of ZnO nanoparticles on anammox granules at ambient temperature were investigated. The presence of ZnO nanoparticles was capable of inhibiting the anammox granules, and the inhibitory effects would increase as the added concentration increased (24 h exposure to ZnO NPs at 5, 50 and 150 mg/L reduced the anammox activity by 18.4%, 35.5% and 50.4%, respectively). The content of extracellular polymeric substances (EPS) was also observed to be decreased by ZnO NPs, which led to the reduction of their protective capability to the inner microorganisms of the anammox granules. The nanotoxicity was attributed to both nanoparticles and released zinc cations, which penetrated into the granules and increase the reactive oxygen species (ROS) production significantly, resulting in the decrease of viability of anammox bacteria. These results provide important information for the integration of anammox into mainstream treatment with presence of ZnO NPs.
Effects of nano metal oxide particles on denitrifying phosphorus removal system: Potential stress mechanism and recovery strategy

Abstract:The accumulation of nano metal oxide particles (NMOPs) in municipal sewage treatment systems harms the microbial community and its metabolism in activated sludge system, resulting in the degradation of its pollutants removal performance. In this work, the stress effect of NMOPs on the denitrifying phosphorus removal system was systematically investigated in terms of pollutants removal performance, key enzyme activities, microbial community diversity and abundances, and intracellular metabolites. Among the ZnO NPs, TiO2 NPs, CeO2 NPs, and CuO NPs, the ZnO NPs showed the most significant impacts with the chemical oxygen demand, total phosphorus, and nitrate nitrogen removal ratio decreased from above 90 % to 66.50 %, 49.13 %, and 57.11 %, respectively. The addition of surfactants and chelating agents could relieve the toxic effect of NMOPs on the denitrifying phosphorus removal system, and the chelating agents were more effective than surfactants in performance recovery. After adding ethylene diamine tetra acetic acid, the removal ratio of chemical oxygen demand, total phosphorus, and nitrate nitrogen under ZnO NPs stress was restored to 87.31 %, 88.79 %, and 90.35 %, respectively. The study provides valuable knowledge to better understand the impacts and stress mechanism of NMOPs on activated sludge systems and provides a solution to recover the nutrients removal performance of denitrifying phosphorus removal system under NMOPs stress.

Abstract:The effects of zinc oxide nanoparticles (nZnO) were examined for nitrifying bacterial enrichments by measuring substrate (ammonia) specific oxygen uptake rates (sOUR) in conjunction with the transcript level of functional genes involved in nitrification quantified by reverse transcriptase-quantitative polymerase chain reaction. Samples from nitrifying bioreactor were exposed in batch vessels to nZnO (1, 5 and 10 mg/L) for either 3 or 6 h. There was considerable increase in sOUR-based nitrification inhibition with increasing dosages of nZnO. At 10 mg/L nZnO, the inhibition was about 35% and 50% for 3 and 6 h exposure, respectively. As the nZnO dosage was increased, the transcript levels of amoA, hao, and nirK for 6 h exposure samples were decreased which corresponded well with sOUR data. In addition, an increase in intracellular reactive oxygen species content was observed for the cells of nitrifying bacteria when the total nZnO concentrations were 5 and 10 mg/L. The agreement between the relative expression of functional genes and sOUR shown in this study reveals the effectiveness of using transcriptional responses in conjunction with physiological activity for better understanding the impact of inhibitory compounds such as nanoparticles on nitrification process.

Abstract:This work compared the efficiencies and internal reasons for aniline removal in a bioaugmented sequencing batch reactor at elevated dissolved oxygen (DO) concentrations. Nearly complete aniline removal was achieved while medium DO of 2.65 mg/L was optimal for subsequent nitrogen removal via heterotrophic nitrification-aerobic denitrification. Apart from the highest bacterial diversity richness, favorable DO condition largely enriched putatively aniline degrader, nitrifiers and denitrifiers. Further evidence from qPCR confirmed that moderate DO significantly stimulated the functional genes including aniline degrading gene tad, nitrifying genes amoA, hao, and denitrifying genes nirS and napA, respectively. This study indicates that the significant enrichment of key microorganisms and effective functional genes under optimal DO is the inner mechanism for reliable aniline degradation and subsequent nitrogen removal in the activated sludge reactor.

Abstract:The environmental impacts of the use of engineered nanoparticles (NPs) remain unclear and have attracted increasing concern worldwide. Considering that NPs eventually end up in wastewater treatment systems, the potential impact of ZnO and TiO2 NPs on the activated sludge was investigated using laboratory-scale sequencing batch reactors (SBRs). Short-term (24 h) exposure to 1, 10 and 100 mg/L shock loads of NPs reduced the oxygen uptake rate of the activated sludge by 3.55%-12.51% compared with the controls. In our experiment, the toxicities of TiO2 NPs were higher than those of ZnO NPs as reflected in the inhibition of oxygen utilization in the activated sludge. However, both the short-term (24 h) and long-term (21 days) exposure to ZnO and TiO2 NPs did not adversely affect the pollutant removal of the SBRs. Furthermore, the polymerase chain reaction-denaturing gel gradient electrophoresis revealed that the microbial community did not significantly vary after the short-term exposure (24 h) to 1, 10 and 100 mg/L shock loads of NPs; however, the cluster analysis in our experiment revealed that the slight difference caused by the NPs largely depended on exposure time rather than on NP type and NP concentration. The long-term exposure (13 days) to 10 mg/L shock load of ZnO or TiO2 NPs caused no substantial microbial community shifts in the activated sludge. Limited dissolved oxygen facilitated nitrogen removal at biocathode during the hydrogenotrophic denitrification process using bioelectrochemical system

Abstract:Effects of limited dissolved oxygen (DO) on hydrogenotrophic denitrification at biocathode was investigated using bioelectrochemical system. It was found that total nitrogen removal increased by 5.9%, as DO reached about 0.24 mg/L with the cathodic chamber unplugged (group R_Exposure). With the presence of limited DO, not only the nitrogen metabolic pathway was influenced, but the composition of microbial communities of ammonia-oxidizing bacteria and nitrite-oxidizing bacteria were enriched accordingly. After metagenomic analysis, enriched genes in R_Exposure were found to be associated with nearly each of nitrogen removal steps as denitrification, nitrification, DNRA, nitrate assimilation and even nitrogen fixation. Moreover, genes encoding both Complexes III and IV constituted the electron transfer chain were significantly enriched, indicating that more electrons would be orientated to the reduction of NO2--N, NO-N and oxygen. Therefore, enhanced nitrogen removal could be attained through the co-respiration of nitrate and oxygen by means of NH4+-N oxidation.
A family with massive acute vitamin D intoxication

Abstract:Acute massive vitamin D overdosage occurred in a family after eating food cooked in a nut oil containing 5 million units of vitamin D3/ml. The plasma vitamin D was 55 and 60 i.u./ml in the father and mother respectively, and 9·6 i.u./ml in their 11-month-old infant (normal range, 0-1·6 i.u./ml). All the family presented with symptoms of hypercalcaemia and the infant responded quickly to prednisone. After steroids had failed to control the hypercalcaemia in the parents, neutral phosphate was successful, although necessary for 9 months. Before phosphate therapy it was shown that both parents were in strongly negative calcium balance, indicating that the vitamin D was mobilizing calcium from bone. Eleven years later all 3 patients are well but a renal biopsy in one of them shows persistent nephrocalcinosis.
Recovery profile of anaerobic ammonium oxidation (anammox) bacteria inhibited by ZnO nanoparticles

Abstract:The potential effects of nanoparticles (NPs) on biological treatment processes have become significant due to their increasing industrial applications. The purpose of this research was to investigate the self-recovery ability of anammox bacteria following acute ZnO NPs toxicity. In this context, a 2-liter lab-scale anammox reactor was operated for 550 days to enrich the biomass required to the batch exposure tests. Anammox culture was firstly exposed to four different doses of ZnO NPs (50, 75, 100 and 200 mg/L) for 24 h. Then, the ZnO NPs were removed and self-recovery performance of the anammox bacteria was assessed by evaluating the nitrogen removal capacities for 72 h. Besides the nitrogen removal performance, extracellular polymeric substances (EPS) production was also detected to deeply understand the response of the enriched anammox culture against ZnO NPs exposure. The results revealed that sudden and high load of ZnO NPs (100 and 200 mg/L) resulted in persistent impairment to the nitrogen removal performance of the enriched anammox culture. However, relatively lower doses (50 and 75 mg/L) caused deceleration of the nitrogen removal performance during the recovery period. In addition, EPS content in the reactor decreased along with escalating load of ZnO NPs.
