Abstract:Understanding the functions of biomolecules at the single-molecule level is crucial due to their important and diverse roles in cell regulation. Recently, nanotweezers made of dual carbon nanoelectrodes have been developed for single-cell biopsies by applying a high alternating voltage. However, high electric voltage can induce Joule heating, water electrolysis, and other side effects on cell activity, which may be unfavorable for cellular applications. Here, we report a low-voltage nanotweezer for trapping of single DNA molecules using etching-engineered nanoelectrodes which effectively reduce the minimum trapping voltage by six times. Meanwhile, the low-voltage nanotweezer displays an improved trapping stiffness. Based on the finite element method simulations, we attribute the mechanism for the low-voltage nanotweezers to the increase in spatial heterogeneity and nonuniformity of electric field by etching of quartz near the nanoelectrodes. This work opens a new dimension for noninvasive single-molecule manipulation in solution and potential applications in single-cell biopsies.

Abstract:The new Cerec 3D design software for inlays and partial and full crowns simplifies work when producing several restorations in one session. Quite significant progress has been achieved, in that the entire row of teeth of a quadrant can be acquired completely and displayed by successively overlapping optical impressions. The digital working model of a quadrant in which all preparations are acquired is the result. The restorations can be designed individually and inserted virtually. Thanks to virtual insertion, the proximal contacts to neighboring restorations can be designed perfectly and all restorations finally designed, milled, and inserted in one sitting. This method provides a significant rationalization effect.
Rapid Trapping and Label-free Characterization of Single Nanoscale Extracellular Vesicles and Nanoparticles in Solution

Abstract:Achieving high-throughput, comprehensive analysis of single nanoparticles to determine their size, shape, and composition is essential for understanding particle heterogeneity with applications ranging from drug delivery to environmental monitoring. Existing techniques are hindered by low throughput, lengthy trapping times, irreversible particle adsorption, or limited characterization capabilities. Here, we introduce Interferometric Electrohydrodynamic Tweezers (IET), an integrated platform that rapidly traps single nanoparticles in parallel within three seconds. IET enables label-free characterization of particle size and shape via interferometric imaging and identifies molecular composition through Raman spectroscopy, all without the need for fluorescent labeling. We demonstrate the platform's capabilities by trapping and imaging colloidal polymer beads, nanoscale extracellular vesicles (EVs), and newly discovered extracellular nanoparticles known as supermeres. By monitoring their interferometric contrast images while trapped, we accurately determine the sizes of EVs and supermeres. Our IET represents a powerful optofluidics platform for comprehensive characterization of nanoscale objects, opening new avenues in nanomedicine, environmental monitoring, and beyond.

Abstract:The single-molecule technique for investigation of an unlabeled protein in solution is very attractive but with great challenges. Nanopore sensing as a label-free tool can be used for collecting the structural information of individual proteins, but currently offers only limited capabilities due to the fast translocation of the target. Here, a reliable and facile method is developed to convert the silicon nitride nanopore to a stable nanonet platform for single-entity sensing by electrophoretic or electroosmotic trapping. A nanonet is fabricated based on a material reorganization process caused by electron-beam and light-irradiation treatment. Using protein molecules as a model, it is revealed that the solid-state nanonet can produce collision and trapping flipping signals of the protein, which provides more structural information than traditional nanopore sensing. More importantly, thanks to the excellent stability of the solid-state silicon nitride nanonet, it is demonstrated that the ultraviolet-light-irradiation-induced structural-change process of an individual protein can be captured. The developed nanonet supplies a robust platform for single-entity studies but is not limited to proteins.

Abstract:We demonstrate the trapping of single 20 and 40 nm polystyrene spheres at the cleaved end of a fiber optic with a double nanohole aperture in gold and without any microscope optics. An optical transmission increase of 15% indicates a trapping event for the 40 nm particle, and the jump is 2% for the 20 nm particle. This modular technique can be used to replace those used with current optical trapping setups that require complicated free space optics and frequent calibration, with one that is modular and requires no free space optics. This simple arrangement with the potential for fiber translation is of interest for future biosensor and optical nano-pipette devices.

Abstract:We present a device that allows a user to trap a single nanoscale object in solution at ambient temperature, and then to position the trapped object with nanoscale resolution. This anti-Brownian electrophoretic trap (ABEL trap) works by monitoring the Brownian motion of the particle (via fluorescence microscopy), and then applying a feedback voltage to the solution so that the electrophoretic drift exactly cancels the Brownian motion. The ABEL trap works on any object that can be imaged optically and that acquires a charge in water. The ABEL trap is noninvasive, is gentle enough to handle biological molecules, and can trap objects far smaller than can be trapped with laser tweezers. Our proof-of-principle device can trap fluorescent polystyrene nanospheres with diameters down to 20nm.

Abstract:There is tantalizing evidence that proteins can be accurately and selectively manipulated by higher order electric field effects within microfluidic devices. The accurate and precise manipulation of proteins in these platforms promises to disrupt and revolutionize many fields, most notably analytical biochemistry. Several lines of experimental evidence suggest much higher forces are generated compared to those calculated from traditional theories and those higher forces arise from subtle structural features of the proteins providing selectivity. New theories reflect some of the experimental evidence in the magnitude of the force predicted and inclusion of subtle structural features absent in traditional continuum theory. Unfortunately, the experimental evidence is largely exploratory in nature and lacks one or more important elements that prevents a clear interpretation and comparison to not only the other existing data, but also quantitative comparison to the evolving theoretical descriptions. Here, a clear and interpretable experimental system is presented that quantitatively determines the dielectrophoretic susceptibility of unlabeled, unaggregated native‐structure protein molecules that are exposed to modest electric fields (105–106 V/m) for short periods of time (∼5 ms) without significant increases in local concentration. The platform uses sub‐nanogram quantities of protein, the probed volume upon determination is a few picoliters, and the total analysis time is 10 s.

Abstract:Single unmodified biomolecules in solution can be observed and characterized by interferometric imaging approaches; however, Rayleigh scattering limits this to larger proteins (typically >30 kDa). We observe real-time image tracking of unmodified proteins down to 14 kDa using interference imaging enhanced by surface plasmons launched at an aperture in a metal film. The larger proteins show slower diffusion, quantified by tracking. When the diffusing protein is finally trapped by the nanoaperture, we perform complementary power spectral density and noise amplitude analysis, which gives information about the protein. This approach allows for rapid protein characterization with minimal sample preparation and opens the door to characterizing protein interactions in real time.

Abstract:Solid-state nanopore has been proved as a versatile method for label-free single molecule detection with fairly high resolution. Protein detection using solid-state nanopores is generally challenging because its globular shape, compared with worm-shape DNA, leads to fast transit times which are confounded with limited bandwidth. One strategy is to use a comparable size solid-state nanopore to the size of protein; the confinement effect of a smaller nanopore achieves slow protein protein transit times and better resolution. However, certain proteins such as adenylate kinase (AdK) and dihydrofolate reductase (DHFR) stick to pores that are of similar diameters to the proteins, making it challenging to acquire sufficient translocation events prior to clogging. With larger pores, however, many events are missed due to fast protein translocation times. In this report, we overcome this issue by developing a setup for driving proteins by employing two driving forces, namely pressure-driven flow and an electrical potential across the pore. Using this method, we can tune protein transit times and maintain a high signal-to-noise, even for pores that are 1.5 times larger than the protein size. Control is highlighted by detecting the large conformational change of AdK upon binding a lock-substrate, as well as by detecting a smaller conformational change in DHFR upon binding the anti-cancer drug methotrexate.

Abstract:Trapping nanoscopic objects to observe their dynamic behaviour for extended periods of time is an ongoing quest. Particularly, sub-100nm transparent objects are hard to catch and most techniques rely on immobilisation or transient diffusion through a confocal laser focus. We present an Anti-Brownian ELectrokinetic trap (pioneered by A. E. Cohen and W. E. Moerner) to hold nanoparticles and individual FoF1-ATP synthase proteins in solution. We are interested in the conformational dynamics of this membrane-bound rotary motor protein that we monitor using single-molecule FRET. The ABELtrap is an active feedback system cancelling the nano-object's Brownian motion by applying an electric field. We show how the induced electrokinetic forces confine the motion of nanoparticles and proteoliposomes to the centre of the trap.

Abstract:To monitor conformational changes of individual membrane transporters in liposomes in real time, we attach two fluorophores to selected domains of a protein. Sequential distance changes between the dyes are recorded and analyzed by Forster resonance energy transfer (FRET). Using freely diffusing membrane proteins reconstituted in liposomes, observation times are limited by Brownian motion through the confocal detection volume. A. E. Cohen and W. E. Moerner have invented and built microfluidic devices to actively counteract Brownian motion of single nanoparticles in electrokinetic traps (ABELtrap). Here we present a version of an ABELtrap with a laser focus pattern generated by electro-optical beam deflectors and controlled by a programmable FPGA. This ABELtrap could hold single fluorescent nanobeads for more than 100 seconds, increasing the observation times of a single particle by more than a factor of 1000. Conformational changes of single FRET-labeled membrane enzymes FoF1-ATP synthase can be detected in the ABELtrap.

Abstract:Abstract Heterogeneous nanoscale extracellular vesicles (EVs) are of significant interest for disease detection, monitoring, and therapeutics. However, trapping these nano-sized EVs using optical tweezers has been challenging due to their small size. Plasmon-enhanced optical trapping offers a solution. Nevertheless, existing plasmonic tweezers have limited throughput and can take tens of minutes for trapping for low particle concentrations. Here, we present an innovative approach called geometry-induced electrohydrodynamic tweezers (GET) that overcomes these limitations. GET generates multiple electrohydrodynamic potentials, allowing parallel transport and trapping of single EVs within seconds. By integrating nanoscale plasmonic cavities at the center of each GET trap, single EVs can be placed near plasmonic cavities, enabling instant plasmon-enhanced optical trapping upon laser illumination without detrimental heating effects. These non-invasive scalable hybrid nanotweezers open new horizons for high-throughput tether-free plasmon-enhanced single EV trapping and spectroscopy. Other potential areas of impact include nanoplastics characterization, and scalable hybrid integration for quantum photonics.

Abstract:Accurate detection and characterization of nanoparticles within confined spaces is crucial for applications ranging from nanofluidics to biotechnology. We present a novel approach that combines interferometric scattering (iSCAT) detection with trapping by dielectrophoresis (DEP) to achieve label-free detection of nanoparticles that are trapped and/or actuated between nano-gap electrodes. DEP utilizes the interaction between the induced dipole of the particle and the applied electric field to create a trapping potential. We demonstrate our method by trapping and label-free detection of down to 20 nm polystyrene nanoparticles. Additionally, we demonstrate that the signal-to-noise ratio of our detection can be boosted up to 20-fold by periodic actuation of the nanoparticle in the trap. This is done by a digital lock-in detection scheme on the modulated scattering signal. Our method holds promise for various applications, including assembly of nanoparticles, single-particle property analysis, and nanofluidic devices.

Electromagnetic manipulation of sub-500 Da biomolecules

Abstract:The manipulation of nanoscale matter has the potential to revolutionize a variety of fields across nanoscience and technology. Here, we demonstrate experimentally and characterize numerically a device that combines the benefits of dielectrophoresis (DEP) - long-range and strong trapping forces - with those of plasmonic tweezers - high sensitivities - to achieve a remarkable efficiency in the trapping and sensing of metallic nanoparticles and biomolecules. In particular, we show the DEP trapping and surface enhanced Raman scattering characterization of bovine serum albumin and Rhodamine B, thus extending the applications of tweezing devices to molecules having masses of only a few hundreds of Da. This range covers virtually any molecule relevant for life, from tiny oligopeptides to large proteins. This pushes our manipulation capabilities deep into the realms of efficient single-molecule biosensing and quantum science, providing a powerful platform to probe matter at the nanoscale.

Effect of sperm immobilisation and demembranation on the oocyte activation rate in the mouse

Abstract:To analyse the effect of the state of the sperm plasma membrane on oocyte activation rate following intracytoplasmic sperm injection (ICSI), three types of human and mouse spermatozoa (intact, immobilised and Triton X-100 treated) were individually injected into mouse oocytes. At 30, 60 and 120 min after injection, maternal chromosomes and sperm nuclei within oocytes were examined. Following human sperm injection, the fastest and the most efficient oocyte activation and sperm head decondensation occurred when the spermatozoa were treated with Triton X-100. Intact spermatozoa were the least effective in activating oocytes. Thus, the rate of mouse oocyte activation following human sperm injection is greatly influenced by the state of the sperm plasma membrane during injection. When mouse spermatozoa were injected into mouse oocytes, the rates of oocyte activation and sperm head decondensation within activated oocytes were the same irrespective of the type of sperm treatment prior to injection. We witnessed that live human spermatozoa injected into moue oocytes often kept moving very actively within the ooplasm for more than 60 min, whereas motile mouse spermatozoa usually became immotile within 20 min after injection into the ooplasm. In 0.002% Triton X-100 solution, mouse spermatozoa are immobilised faster than human spermatozoa. These facts seem to suggest that human sperm plasma membranes are physically and biochemically more stable than those of mouse spermatozoa. Perhaps the physical and chemical properties of the sperm plasma membrane vary from species to species. For those species whose spermatozoa have ‘stable’ plasma membranes, prior removal or ‘damage’ of sperm plasma membranes would increase the success rate of ICSI.

Abstract:Anti-Brownian traps confine single particles in free solution by closed-loop feedback forces that directly counteract Brownian motion. Extended-duration measurements on trapped objects allow detailed characterization of photophysical and transport properties as well as observation of infrequent or rare dynamics. However, this approach has been generally limited to particles that can be tracked by fluorescence emission. Here we present the Interferometric Scattering Anti-Brownian ELectrokinetic (ISABEL) trap, which uses interferometric scattering rather than fluorescence to monitor particle position. By decoupling the ability to track (and therefore trap) a particle from collection of its spectroscopic data, the ISABEL trap enables confinement and extended study of single particles that do not fluoresce, only weakly fluoresce, or exhibit intermittent fluorescence or photobleaching. This new technique significantly expands the range of nanoscale objects that may be investigated at the single-particle level in free solution.

Abstract:Proteins are structurally dynamic macromolecules, and it is challenging to quantify the conformational properties of their native state in solution. Nanopores can be efficient tools to study proteins in a solution environment. In this method, an electric field induces electrophoretic and/or electro-osmotic transport of protein molecules through a nanopore slightly larger than the protein molecule. High-bandwidth ion current measurement is used to detect the transit of each protein molecule. First, our measurements reveal a correlation between the mean current blockade amplitude and the radius of gyration for each protein. Next, we find a correlation between the shape of the current signal amplitude distributions and the protein fluctuation as obtained from molecular dynamics simulations. Further, the magnitude of the structural fluctuations, as probed by experiments and simulations, correlates with the ratio of α-helix to β-sheet content. Abstract:Plasmonic nanostructures can overcome Abbe's diffraction limit to generate strong gradient fields, enabling efficient optical trapping of nano-sized particles. However, it remains challenging to achieve stable trapping with low incident laser intensity. Here, we demonstrate a Fano resonance-assisted plasmonic optical tweezers (FAPOT), for single nanoparticle trapping in an array of asymmetrical split nano-apertures, milled on a 50 nm gold thin film. Stable trapping is achieved by tuning the trapping wavelength and varying the incident trapping laser intensity. A very large normalized trap stiffness of 8.65 fN/nm/mW for 20 nm polystyrene particles at a near-resonance trapping wavelength of 930 nm was achieved. We show that trap stiffness on resonance is enhanced by a factor of 63 compared to off-resonance conditions. This can be attributed to the ultra-small mode volume, which enables large near-field strengths and a cavity Purcell effect contribution. These results should facilitate strong trapping with low incident trapping laser intensity, thereby providing new options for studying transition paths of single molecules, such as proteins, DNA, or viruses.

Abstract:Protein analysis has relied on electrophoresis, mass spectroscopy and immunoassay, which separate, detect and identify proteins based on the size, charge, mobility and binding to antibodies. However, measuring these properties at the single molecule level has not been possible. We tether a protein to a surface with a flexible polymer, oscillate the protein with an alternating electric field, and image the protein oscillation with a near field imaging method, from which we determine the size, charge, mobility the protein. We also measure binding of antibodies to single proteins and ligand binding-induced conformational changes in single proteins. We anticipate that this capability will provide a new way for protein analysis, and a single molecule immunoassay for disease biomarker detection.

Abstract:Nanohole optical tweezers have been used by several groups to trap and analyze proteins. In this work, we demonstrate that it is possible to create high-performance double nanohole (DNH) substrates for trapping proteins without the need for any top-down approaches (such as electron microscopy or focused-ion beam milling). Using polarization analysis, we identify DNHs as well as determine their orientation and then use them for trapping. We are also able to identify other hole configurations, such as single, trimers and other clusters. We explore changing the substrate from glass to polyvinyl chloride to enhance trapping ability, showing 7 times lower minimum trapping power, which we believe is due to reduced surface repulsion. Finally, we present tape exfoliation as a means to expose DNHs without damaging sonication or chemical methods. Overall, these approaches make high quality optical trapping using DNH structures accessible to a broad scientific community.
