Abstract:A signal-amplified electrochemiluminescent (ECL) sensor chip was developed for sensitive analysis of procalcitonin (PCT). Herein, we first prepared a self-enhanced luminophore, which enhanced ECL responses through intramolecular reactions. Second, Au-Pd bimetallic nanocrystals and mixed-valence Ce-based metal-organic frameworks (MOFs) were introduced as co-reaction promoters to facilitate the reduction of dissolved O-2. Based on the synergistic catalysis of Au and Pd, the spontaneous cyclic reaction of Ce(III)/Ce(IV), and the high electrochemical active surface area of Ce(III, IV) MOF, a large number of superoxide anion radicals (O-2(center dot-)) and hydroxyl radicals (OH center dot) were produced. Therefore, the luminescence efficiency of N-(aminobutyl)-N-(ethylisoluminol)-dissolved O-2 (ABEI-O-2) systems were greatly improved, providing a new prospect for the application of dissolved O-2 in ECL analysis. In addition, the affinity peptide ligands were used for the directional connection of antibodies to provide protection for the bioactivity of the proposed sensor. Finally, the microfluidic technology was applied to ECL analysis to integrate the three-electrode detection system into the self-assembled microfluidic chip, which realized the automation and portability of the detection process. The developed sensor showed high sensitivity for PCT detection with a detection limit of 3.46 fg/mL, which possessed positive significance for the clinical diagnosis of sepsis.
Fiber-shaped Organic Electrochemical Transistors for Biochemical Detections with High Sensitivity and Stability

Abstract:Precise and continuous monitoring of biochemicals by biosensors assists to understand physiological functions for various diagnostics and therapeutic applications. For implanted biosensors, small size and flexibility are essential for minimizing tissue damage and achieving accurate detection. However, the active surface area of sensor decreases as the sensor becomes smaller,which will increase the impedance and decrease the signal to noise ratio, resulting in a poor detection limit. Taking advantages of local amplification effect, organic electrochemical transistors(OECTs) constitute promising candidates for high-sensitive monitoring. However, their detections in deep tissues are rarely reported. Herein, we report a family of implantable, fiber-shaped all-in-one OECTs based on carbon nanotube fibers for versatile biochemical detection including H 2 O 2 , glucose, dopamine and glutamate. These fiber-shaped OECTs demonstrated high sensitivity, dynamical stability in physiological environment and antiinterference capability. After implantation in mouse brain, 7-day dopamine monitoring in vivo was realized for the first time.These fiber-shaped OECTs could be great additions to the "life science" tool box and represent promising avenue for biomedical monitoring.
Blending Enzyme Immobilization Enabled High Performance Glucose Sensor Based on an n-channel Organic Electrochemical Transistor

Abstract:Organic electrochemical transistors (OECTs) have emerged as an advantageous choice for constructing biosensors due to their remarkable water compatibility, low operating voltage, and inherent amplification capability. However, the current research on bio-sensing based on OECTs predominantly employs p-type material PEDOT:PSS as the channel material. Nevertheless, the utilization of a single material and its depleting characteristics impose significant limitations on device miniaturization and integration applications. We present a high-sensitivity glucose sensor based on n-channel accumulation-type OECT through immobilization the glucose oxidase on Pt gate electrode with an blending approach. The glucose oxidase directly mixed with bovine serum albumin, chitosan and immolizied on the gate electode, demonstrating a stable and sensitive response to the glucose. The modified n-channel organic electrochemical transistor demonstrates a highly sensitive response to glucose across a concentration range from M to mM, with a quantified stable sensitivity over 2.69/mmol by normalizing the current change with respect to concentration. The device also exhibits selectivity towards glucose (compared with high concentrations of lactic acid, different concentration gradients of ascorbic acid, and uric acid), rendering it suitable for noninvasive glucose detection in body fluid like sweat, saliva etc. This flexible and ensitive electrochemical transistor sensor holds immense potential for the development of potable healthcare biosensing.

Surface plasmon resonance enhanced real-time photoelectrochemical protein sensing by gold nanoparticle-decorated TiO₂ nanowires.

Abstract:Recently developed photoelectrochemical (PEC) sensing systems represent a unique potential detection method for real-time analysis of chemical/biological molecules, while the low absorption of TiO2 nanomaterials in the visible wavelength region and the slow surface charge transfer efficiency limit the ultimate sensitivity. Here we develop a gold nanoparticle-decorated TiO2 nanowire sensor for PEC detection of protein binding. The direct attachment of Au nanoparticles to TiO2 nanowires offers strong surface plasmon resonance for electrochemical field effect amplification, yielding a similar to 100% increase of photocurrent density. In addition, the surface functionalization of gold nanoparticles allows for direct capturing of target proteins near the Au/TiO2 interface and thus substantially enhances the capability of attenuation of energy coupling between Au and TiO2, leading to much-improved sensor performance. As a proof of concept, cholera toxin subunit B has been robustly detected by the TiO2-Au nanowire sensor functionalized with ganglioside GM!, with a high sensitivity of 0.167 nM and excellent selectivity. Furthermore, the real-time feature of photoelectrochemical sensing enables direct measurement of binding kinetics between cholera toxin subunit B and GM!, yielding association and disassociation rate constants and an equilibrium constant K-d of 4.17 nM. This surface plasmon resonance-enhanced real-time, photoelectrochemical sensing design may lead to exciting biodetection capabilities with high sensitivity and real-time kinetic studies.
Interfacing aptamers, nanoparticles and graphene in a hierarchical structure for highly selective detection of biomolecules in OECT devices

Abstract:Abstract In several biomedical applications, the detection of biomarkers demands high sensitivity, selectivity and easy-to-use devices. Organic electrochemical transistors (OECTs) represent a promising class of devices combining a minimal invasiveness and good signal transduction. However, OECTs lack of intrinsic selectivity that should be implemented by specific approaches to make them well suitable for biomedical applications. Here, we report on a biosensor in which selectivity and a high sensitivity are achieved by interfacing, in an OECT architecture, a novel gate electrode based on aptamers, Au nanoparticles and graphene hierarchically organized to optimize the final response. The fabricated biosensor performs state of the art limit of detection monitoring biomolecules, such as thrombin-with a limit of detection in the picomolar range (≤ 5 pM) and a very good selectivity even in presence of supraphysiological concentrations of Bovine Serum Albumin (BSA-1mM). These accomplishments are the final result of the gate hierarchic structure that reduces sterich indrance that could contrast the recognition events and minimizes false positive, because of the low affinity of graphene towards the physiological environment. Since our approach can be easily applied to a large variety of different biomarkers, we envisage a relevant potential for a large series of different biomedical applications.

Abstract:Cell surface glycans play critical roles in diverse biological processes, such as cell-cell communication, immunity, infection, development, and differentiation. Their expressions are closely related to cancer growth and metastasis. This work demonstrates an organic electrochemical transistor (OECT)-based biosensor for the detection of glycan expression on living cancer cells. Herein, mannose on human breast cancer cells (MCF-7) as the target glycan model, poly dimethyl diallyl ammonium chloride-multiwall carbon nanotubes (PDDA-MWCNTs) as the loading interface, concanavalin A (Con A) with active mannose binding sites, aptamer and horseradish peroxidase co-immobilized gold nanoparticles (HRP-aptamer-Au NPs) as specific nanoprobes are used to fabricate the OECT biosensor. In this strategy, PDDA-MWCNT interfaces can enhance the loading of Con A, and the target cells can be captured through Con A via active mannose binding sites. Thus, the expression of cell surface can be reflected by the amount of cells captured on the gate. Specific nanoprobes are introduced to the captured cells to produce an OECT signal because of the reduction of hydrogen peroxide catalyzed by HRP conjugated on Au nanoparticles, while the aptamer on nanoprobes can selectively recognize the MCF-7 cells. It is reasonable that more target cells are captured on the gate electrode, more HRP-nanoprobes are loaded thus a larger signal response. The device shows an obvious response to MCF-7 cells down to 10 cells/μL and can be used to selectively monitor the change of mannose expression on cell surfaces upon a treatment with the N-glycan inhibitor. The OECT-based biosensor is promising for the analysis of glycan expressions on the surfaces of different types of cells.

Abstract:Affinity-based organic electrochemical transistor (OECT) sensors offer an attractive approach to point-of-care diagnostics due to their extreme sensitivity and easy operation; however, their application in the real world is frequently challenged by the poor storage stability of antibody proteins and the interference from biofouling in complex biofluids. In this work, we developed an antibody-free and antifouling OECT biosensor to detect C-reactive protein (CRP) at ultra-high specificity and sensitivity. The key to this novel biosensor is the gate coated by phosphorylcholine-functionalized poly (3,4-ethylene dioxythiophene) (PEDOT-PC), which possesses large capacitance and low impedance, prevents biofouling of bovine serum albumin (BSA) and the fetal bovine serum (FBS), and interacts specifically with CRP molecules in the presence of calcium ions. This PEDOT-PC-gated OECT biosensor demonstrated exceptional sensitivity when detecting the CRP molecules at 10 pg/mL, while significantly depressing the signal from the nonspecific binding. This indicates that this biosensor could detect the CRP molecules directly without nonspecific binding blocking, the usual process for the earlier transistor sensors before detection. We envision that this PEDOT-PC-gated OECT biosensor platform may offer a potentially valuable tool for point-of-care diagnostics as it alleviates concerns about poor antibody stability and BSA blocking inconstancy.

Abstract:In this study, a novel organic photophotochemical transistor (OPECT) biosensing platform was proposed for dual-mode detection of CEA. The dual-mode detecting system is achieved benefit from the exceptional photoelectric performance of MIL-53(Fe) -NH 2 @ZnIn 2 S 4 (MNZ) and the peroxidase enzyme (POD) activity of ZIF-67/Cu 0.76 Co 2.24 O 4 (ZIF-67/CuCoO). Ab2- ZIF-67/CuCoO probe was immobilized on a 96-well plate by enzyme-linked immunosorbent assay, which accelerated the oxidation of 3,3,5,5-tetramethylbenzidine (TMB) by hydrogen peroxide and thus successfully realized visual detection of CEA. Additionally, in OPECT biosensing mode, the oxidized form of TMB (oxTMB·) serves as a consumptive agents of the electron donor AA, which cause the photocurrent change of MNZ heterojunction, leading to a decrease in the channel current of poly (ethylenedioxythiophene): poly (styrene sulfonate) (PEDOT: PSS) organic transistor. The integration of nanobiocatalysts and the OPECT system demonstrates excellent detection performance for CEA, with a detection limit as low as 3.24 fg mL −1 and expanded their prospective applications for clinical detection of nucleic acids, proteins, and other tumor markers.

Abstract:High‐performance flexible electrochemical transistors based on nanoporous conjugated polymer aerogels are developed by synergistic assembly and crosslinking of conjugated polymer, cellulose nanofiber, and polyorganosiloxane. They show enhanced ion penetration/transport and transconductance and can be used for highly sensitive glucose sensors with an ultralow detection limit, electrophysiological signal monitoring, and high‐performance artificial synapses. Organic electrochemical transistors (OECTs) possess low operating voltage and excellent amplification capability and show promising applications in biosensors and flexible electronics. However, the active layers of OECTs are usually dense films, which show limited ion penetration/transport, resulting in limited performances of OECTs. Here, unprecedented high‐performance flexible multifunctional OECTs based on nanoporous (mainly 2–60 nm), high‐specific‐surface‐area (255–281 m2 g−1), and flexible conjugated polymer aerogel films are developed. The nanoporous structures effectively facilitate ion penetration/transport, leading to significantly enhanced transconductance (48.5–53.5 mS) and on/off ratio (6.4 × 104) of the OECTs compared with those of dense devices. The resulting OECT‐based glucose sensors exhibit an ultralow detection limit of 1 pm, approximately two to three orders of magnitude lower than those of the previously reported OECT glucose sensors, and an ultrabroad detection range of 1 pm–5 m. They can detect trace amounts of glucose in sweat, serum, saliva, and urine in real time. Moreover, the OECTs can be used for high‐performance flexible artificial synapses and electrocardiogram monitoring. This work provides a powerful strategy toward highly sensitive biosensors, artificial synapses, and electrophysiological signal monitoring.

Abstract:Herein we report a high-gain signal-on poly(3,4-ethylenedioxythiophene):poly(styrene sulfonate) (PEDOT:PSS) organic electrochemical transistor (OECT) biosensing using an accumulation-mode PEDOT:PSS OECT modulated by a light-fueled MXene/MOFs/NiO Schottky heterojunction. In such a system, the MXene/MOFs/NiO Schottky heterojunction exhibited superior gating effect, as it not only enabled the fast-directional charge transfer but also guaranteed the maximal accessibility of the electrolyte to topped 2D MXene with large surface area. In linkage with a bi-enzyme cascade system, the quinone derivatives produced by the cascade reaction of alkaline phosphatase (ALP) and tyrosinase (TYR) could serve as effective electron acceptors for the representative Ti3C2/PCN-224/NiO heterojunction, underpinning an innovative method for sensitive detection of ALP activity with a low detection limit of 0.001 U L−1. Remarkably, the as-developed system demonstrated a remarkable current gain as high as near 104, which to our knowledge is the highest one among existing OECT biosensory devices. This work represents a generic protocol to develop the novel signal-on PEDOT:PSS OECT platform towards biochemical detection and beyond.

Abstract:Metal-organic framework (MOF) derivatives with unique physicochemical and electronic properties have seen a tremendous growth in diverse applications. Organic optobioelectronics have long been pursued in modern electronics for next-generation bio-relevant implementations. The intersection of these two disciplines could be an appealing way to pursue better performance of materials and devices. Herein this work reports the exploration of MOF derivatives and its ionic modulation for gating organic photoelectrochemical transistor (OPECT) biosensing. In the representative system of poly(ethylenedioxythiophene):poly(styrene sulfonate) (PEDOT:PSS) gated by zeolitic-imidazolate-framework (ZIF)-8-derived CdxZn1-xS, a high current gain could be achieved at zero gate bias. In connection to a CuO nanoparticle-labeled sandwich immunoassay, acidolysis-triggered Cu2+-induced ionic modulation of the system results into a good performance toward human IgG with a low limit of detection of 0.003 pg/mL. This work features the MOF derivative-gated organic electronics and is expected to inspire more interest to explore various MOF derivative electronics with unknown possibilities, considering the diversity of MOF derivatives.

Monose-modified organic electrochemical transistors for cell surface glycan analysis via competitive recognition to enzyme-labeled lectin

Abstract:A competitive strategy for glycan determination on cell surface with organic electrochemical transistors (OECTs) has been developed. The carboxylic multi-wall carbon nanotubes were firstly immobilized on the gate interface to cross-link the specific monose with adipic dihydrazide as the linker, which could then competitively recognize horseradish peroxidase (HRP)–labeled lectin with the target monose on the cell surface. The HRP captured on the gate interface through the affinity of lectin to monose finally catalyzed the reduction of hydrogen peroxide to produce the output current signal for detection of cell surface monose under the optimal gate voltage of 0.9 V. Using mannose and galactose groups as the target models, HRP-labeled concanavalin A and peanut agglutinin were used to competitively recognize these groups on both cell surface and gate interface, respectively. The amounts of mannose and galactose on HeLa cells were measured to be 3.41 × 10 8 and 2.92 × 10 8 molecules per cell, respectively. The changes of the mannose and galactose expressions upon external stimulation were also observed with the proposed biosensors, which showed consistent results with flow cytometric analysis, indicating that the OECT-based biosensor is suitable for analysis of different glycan expressions on cell surface. Abstract:This study aimed to expand extant Alzheimer's disease (AD) research on cluster size and switching strategies in semantic verbal fluency (SVF). First, it addressed a significant shortcoming in research, that is, the use of a single semantic category (i.e., Animal). Second, it examined subjects of lower education levels, as research to date has primarily focussed on tertiary education. Subjects were 26 mild to moderate AD patients and 26 healthy elderly, 65 years old and over, with less than 12 years of schooling. The study examined the cluster size and switching variables, together with the number of subcategories, in two semantic categories (i.e., Animal and Supermarket). Furthermore, it investigated the error patterns (i.e., repetitions and categorical errors) across three semantic categories (i.e., Animal, Supermarket and Furniture). The findings provide preliminary support for category-specific effects on qualitative aspects of SVF highlighting the need to incorporate multiple semantic categories to research and clinical practice. This paper drew on preliminary analyses undertaken during the first author's PhD on ‘Language use in dementia and normal ageing', and the results were reported as an appendix body of the thesis. The PhD was funded by the Australian Postgraduate Award. Sensitive organic electrochemical transistor biosensors: Comparing single and dual gate functionalization and different COOH-functionalized bioreceptor layers

Abstract:We developed new measurement configurations based on organic electrochemical transistors (OECTs). Three types of COOH-functionalized bioreceptor layers were deposited on indium tin oxide (ITO) electrodes on poly(ethylene terephthalate) (PET) substrates and their performance was tested using single gate functionalization organic electrochemical transistor (S-OECT) and dual gate functionalization organic electrochemical transistor (D-OECT) configurations. The three layers included one p-type semiconductor, one insulator, and one self-assembled layer, and the dual gates were connected in series through buffer solutions, so the solution-electrode interfaces had the opposite polarities. We investigated the sensitivities of these systems using the human IgG antigen-human IgG antibody receptor pair for main experiments, and drifts of antibody-functionalized gates without analytes as control experiments. Drifts without analyte can obscure the real sensitivity. We show that the D-OECT has the capability to cancel the drifts, and is also beneficial for showing the sensitivity more exactly. This configuration has the ability to increase the accuracy of antibody-antigen interaction detection, and further decrease or eliminate the effect of ions in the buffer solution. We also prove that the D-OECT can work well with different bioreceptor materials, which indicates that the system can be further applied to different conditions.
MXene@MnIn2S4‐Gated Organic Photoelectrochemical Transistors with Nanozyme‐Mediated Multiple Quenching Effects for Ultrasensitive Detection of Okadaic Acid

Abstract:The organic photoelectrochemical transistor (OECT) is rationally integrated with a heterojunction of MXene‐MnIn2S4 with efficient light‐electric conversion. The Faradaic regime of the OECT can thus be broadened towards zero gate bias under light illumination. Amperometric output with enhanced signal resolution can be obtained with nanozymatic biocatalytic precipitation. Organic optoelectronics have attracted widespread interdisciplinary research interest but lags far behind in the application in marine environmental detection. The organic photoelectrochemical transistor (OPECT) shows promise as a powerful tool for comprehensive monitoring and early warning of marine conditions, which can be further enhanced by the valuable signal amplification strategy of nanozyme‐mediated catalytic precipitation. Herein, OPECT technology is integrated with nanozyme‐mediated catalytic precipitation for the first time, establishing an ultrasensitive detection platform for okadaic acid (OA). Specifically, MXene@MnIn2S4 (MXMnIS) hybrid composed of Schottky‐junction is synthesized via a hydrothermal method, which can efficiently modulate the device with high current gain. Linking with a sandwich immunoassay, the Ru‐C3N4 nanozyme with peroxidase‐mimicking activity can catalyze the oxidation of 4‐chloro‐1‐naphthol (4‐CN) to form an insoluble precipitate on the electrode surface, resulting in a decrease in the photocurrent and altering the transistor response. Importantly, the proposed OPECT biosensor presented an excellent sensitivity and a low detection limit (32.5 pM), fully satisfying the fundamental requirements for the quantitative detection of intracellular and extracellular OA in the practical culture media of Prorocentrum lima at different growth stages. This OPECT platform based on the nanozyme‐mediated quenching effect is significant for effectively monitoring the safety of the marine ecological environment and food safety.

Abstract:Enzyme-like nanomaterials (nanozyme) has been used to generate amplified response signals in biosensors instead of the bio-enzyme, on account of their higher stability and comparable catalytical activity. Especially the metal-organic framework (MOF) materials with ultra-high porosity and high specific surface area, which is more suitable for deriving superior nanozyme. In order to deal with the increasing demand of healthcare diagnostics and biomedical research, here we propose a novel approach for ultrasensitive photoelectrochemical (PEC) immunoassay. The bio-inspired MOF-derived nanozyme Hemin/BSA@ZIF-8 was introduced into the target biorecognition and sensing processes to accelerate the oxidization of substrate to produce insoluble precipitate, which will impede the electron transfer and lead to a significant reduction in photocurrent. The proposed PEC sensor exhibits a wide linear range of 0.1 pg mL −1 ~ 100 ng mL −1 , and a low detection limit of 11.9 fg mL −1 for cardiac troponin I (cTnI) detection. This work brings a new insight of the bio-inspired MOF nanozyme-mediated PEC immunoassay designing, and offers a novel sensing strategy for accurate human biomarker analysis.

Abstract:An ultra-sensitive ratiometric electrochemical aptasensor was constructed based on metal-organic frameworks (MOFs) and bimetallic oxides for the detection of the human epidermal growth factor receptor 2 (HER2), a breast cancer marker. The aluminum metal-organic framework (Al-MOF) and cerium-metal-organic framework (Ce-MOF) have higher specific surface area, which is conducive to load more aptamers or complementary DNA (cDNA), and realize the amplification of internal reference signal Fc. Furthermore, nanoflower-like bismuth copper oxide (Bi2CuO4) with abundant active sites was introduced to modify more aptamers on its surface, which were then fixed to the glassy carbon electrode (GCE) to amplify the detection signal. The quantitative detection of HER2 was achieved by differential pulse voltammetry (DPV), cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). The materials were characterized by scanning electron microscope, transmission electron microscope, Zeta potential analyzer, X-ray diffraction and X-ray photoelectron spectroscopy. The ratiometric electrochemical aptasensor based on nanomaterial and chain displacement signal amplification technology could discern HER2 in a very wide range (0.001-20.0 ng/mL) with an extremely low detection limit (0.049 pg/mL) and has demonstrated good performance in clinical serum analysis. This strategy also provides a feasible idea for sensitive analysis of other clinical tumor markers.

Abstract:A palladium nanoparticle decorated carbon nanotube was designed as a label for preparation of a highly sensitive disposable immunosensor. The immunosensor was constructed by assembling the capture antibody on gold nanoparticles decorated graphene nanosheets modified screen printed carbon working electrode. With a sandwich immunoassay mode, the palladium nanoparticle decorated carbon nanotubes were captured to the immunocomplex and showed strong electrocatalytic activity toward oxygen reduction. The use of carbon nanotube carrier offered a high amount of palladium nanoparticles on each immunoconjugate, hence amplified the detectable signal from the electro-reaction of dissolved oxygen. The graphene nanosheets and gold nanoparticles improved the electronic conductivity and the hydrophilicity of electrode surface for immobilization of the capture antibody, respectively. Under optimal conditions, a linear detection range from 50pg/mL to 10ng/mL and a limit of detection of 44pg/mL (0.3pM) were achieved for human IgG. Using dissolved oxygen as a signal reporter, the detection process avoided deoxygenation. The immunosensor showed acceptable stability, precision and accuracy, indicating potential applications in clinical diagnostics.

Abstract:We demonstrate a biosensor structure consisting of an IGZO (Indium-Gallium-Zinc-Oxide) TFT (thin film transistor) and an extended sensing pad. The TFT acts as the sensing and readout device, while the sensing pad ensures the isolation of biological solution from the transistor channel layer, and meanwhile increases the sensing area. The biosensor is functionalized by first applying ZnO nanorods to increase the surface area for attracting electrical charges of EGFR (epidermal growth factor receptor) antibodies. The device is able to selectively detect 36.2 fM of EGFR in the total protein solution of 0.1 ng/ml extracted from squamous cell carcinoma (SCC). Furthermore, the conjugation duration of the functionalized device with EGFR can be limited to 3 min, implying that the biosensor has the advantage for real-time detection.
